Abstract

Biogeographic variations in the gut microbiota are pivotal to understanding the global pattern of host–microbiota interactions in prevalent lifestyle-related diseases. Pakistani adults, having an exceptionally high prevalence of type 2 diabetes mellitus (T2D), are one of the most understudied populations in microbiota research to date. The aim of the present study is to examine the gut microbiota across individuals from Pakistan and other populations of non-industrialized and industrialized lifestyles with a focus on T2D. The fecal samples from 94 urban-dwelling Pakistani adults with and without T2D were profiled by bacterial 16S ribosomal RNA gene and fungal internal transcribed spacer (ITS) region amplicon sequencing and eubacterial qPCR, and plasma samples quantified for circulating levels of lipopolysaccharide-binding protein (LBP) and the activation ability of Toll-like receptor (TLR)-signaling. Publicly available datasets generated with comparable molecular methods were retrieved for comparative analysis of the bacterial microbiota. Overall, urbanized Pakistanis’ gut microbiota was similar to that of transitional or non-industrialized populations, depleted in Akkermansiaceae and enriched in Prevotellaceae (dominated by the non-Westernized clades of Prevotella copri). The relatively high proportion of Atopobiaceae appeared to be a unique characteristic of the Pakistani gut microbiota. The Pakistanis with T2D had elevated levels of LBP and TLR-signaling in circulation as well as gut microbial signatures atypical of other populations, e.g., increased relative abundance of Libanicoccus/Parolsenella, limiting the inter-population extrapolation of gut microbiota-based classifiers for T2D. Taken together, our findings call for a more global representation of understudied populations to extend the applicability of microbiota-based diagnostics and therapeutics.

Abstract

Featured Application: The potential use of bacteria and their enzymes for the degradation of plastic waste and the recovery of value-added products considering bio-up recycling for the circular economy. A polyethylene terephthalate (PET)-degrading bacterium identified as Stenotrophomonas maltophilia PRS8 was isolated from the soil of a landfill. The degradation of the PET bottle flakes and the PET prepared as a powder were assessed using live cells, an extracellular medium, or a purified cutinase-like enzyme. These treated polymers were analyzed using Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The depolymerization products, identified using HPLC and LC-MS, were terephthalic acid (TPA), mono(2-hydroxyethyl)-TPA (MHET), and bis(2-hydroxyethyl)-TPA (BHET). Several physicochemical factors were optimized for a better cutinase-like enzyme production by using unique single-factor and multi-factor statistical models (the Plackett–Burman design and the central composite design software). The enzyme was purified for homogeneity through column chromatography using Sephadex G-100 resin. The molecular weight of the enzyme was approximately 58 kDa. The specific activity on para nitrophenyl butyrate was estimated at 450.58 U/mg, with a purification of 6.39 times and a yield of 48.64%. The enzyme was stable at various temperatures (30–40 °C) and pH levels (8.0–10.0). The enzyme activity was significantly improved by the surfactants (Triton X-100 and Tween-40), organic solvent (formaldehyde), and metals (NiCl2 and Na2SO4). The extracellular medium containing the cutinase-type enzyme showed a depolymerization yield of the PET powder comparable to that of Idonella skaiensis IsPETase and significantly higher than that of Humicola insolens thermostable HiCut (HiC) cutinase. This study suggests that S. maltophilia PRS8 is able to degrade PET at a mesophilic temperature and could be further explored for the sustainable management of plastic waste. [ABSTRACT FROM AUTHOR]

Abstract

β-glucosidases are found in all domains of life and has the ability to hydrolyse the glycosidic bond between carbohydrates and non-carbohydrates residues making it a potent enzyme from an industrial and agricultural perspective. The aim of the current study was isolation, screening, and identification of bacterial strains having maximum β-glucosidase activity from soil sample of Jacobabad, Pakistan. The isolates were screened primarily, and efficient isolates were confirmed for β-glucosidase production through secondary screening. Among the different isolated strains, 1j2 was selected as the best β-glucosidase producer. The isolate showed 99% similarity with Bacillus stercoris after 16S rRNA sequencing. Different culture conditions were optimized for effective β-glucosidase production. Enhanced level of β-glucosidase was achieved at 40°C, pH 8 at 150 rpm in a β-glucosidase production medium supplemented with a combination of agricultural residues of wheat bran 0.25% (w/v) and orange peel 0.25% (w/v), using 5% inoculum. Bacillus stercoris produced a higher yield (2.89U/mg) of β-glucosidase under optimized conditions. Partial purification of β-glucosidase was achieved after 80% ammonium sulfate precipitation. The enzyme was purified and had shown maximum specific activity of 53.2 U/mg with molecular weight of approximately 36 kDa. In all the tested metals Cu2+ was observed to have inhibitory effect while Ca2+ had not affected the enzyme activity significantly. While organic solvents and surfactants had significantly reduced the β-glucosidase activity. Based on the afore mentioned abilities of purified β-glucosidase from Bacillus stercoris strain it could be a promising player for the industrial applications of lignocellulose hydrolysis found in agricultural residues for the cheap production of biofuels.
