Abstract

The study investigated the use of patch testing with sodium tetrachloropalladate (Na2PdCl4) and palladium chloride (PdCl2) to detect palladium sensitization in patients. Results showed a higher proportion of positive reactions to Na2PdCl4 compared to PdCl2, with a significant number of clinically relevant reactions. The study suggests that Na2PdCl4 at 3% pet. is more suitable for detecting palladium sensitization due to its increased concentration of palladium and diagnostic performance. The findings align with previous studies indicating the higher sensitivity of Na2PdCl4 compared to PdCl2 in detecting palladium sensitization. [Extracted from the article]

Abstract

Cytokines were the first modern immunotherapies to produce durable responses in patients with advanced cancer, but they have only modest efficacy and limited tolerability.sup.1,2. In an effort to identify alternative cytokine pathways for immunotherapy, we found that components of the interleukin-18 (IL-18) pathway are upregulated on tumour-infiltrating lymphocytes, suggesting that IL-18 therapy could enhance anti-tumour immunity. However, recombinant IL-18 previously did not demonstrate efficacy in clinical trials.sup.3. Here we show that IL-18BP, a high-affinity IL-18 decoy receptor, is frequently upregulated in diverse human and mouse tumours and limits the anti-tumour activity of IL-18 in mice. Using directed evolution, we engineered a 'decoy-resistant' IL-18 (DR-18) that maintains signalling potential but is impervious to inhibition by IL-18BP. Unlike wild-type IL-18, DR-18 exerted potent anti-tumour effects in mouse tumour models by promoting the development of poly-functional effector CD8.sup.+ T cells, decreasing the prevalence of exhausted CD8.sup.+ T cells that express the transcriptional regulator of exhaustion TOX, and expanding the pool of stem-like TCF1.sup.+ precursor CD8.sup.+ T cells. DR-18 also enhanced the activity and maturation of natural killer cells to effectively treat anti-PD-1 resistant tumours that have lost surface expression of major histocompatibility complex class I molecules. These results highlight the potential of the IL-18 pathway for immunotherapeutic intervention and implicate IL-18BP as a major therapeutic barrier. Abstract

This report offers a consensus opinion on the diagnosis, epidemiology, treatment, and prevention of leptospirosis in dogs, an important zoonosis. Clinical signs of leptospirosis in dogs relate to development of renal disease, hepatic disease, uveitis, and pulmonary hemorrhage. Disease may follow periods of high rainfall, and can occur in dogs roaming in proximity to water sources, farm animals, or wildlife, or dogs residing in suburban environments. Diagnosis is based on acute and convalescent phase antibody titers by the microscopic agglutination test (MAT), with or without use of polymerase chain reaction assays. There is considerable interlaboratory variation in MAT results, and the MAT does not accurately predict the infecting serogroup. The recommended treatment for optimal clearance of the organism from renal tubules is doxycycline, 5 mg/kg p.o. q12h, for 14 days. Annual vaccination can prevent leptospirosis caused by serovars included in the vaccine and is recommended for dogs at risk of infection.

Abstract

Clinical success in T cell therapy has stimulated widespread efforts to increase safety and potency and to extend this technology to solid tumors. Yet progress in cell therapy remains restricted by the limited payload capacity, specificity of target cell transduction, and transgenic gene expression efficiency of applied viral vectors. This renders complex reprogramming or direct in vivo applications difficult. Here, we developed a synergistic combination of trimeric adapter constructs enabling T cell-directed transduction by the human adenoviral vector serotype C5 in vitro and in vivo. Rationally chosen binding partners showed receptor-specific transduction of otherwise non-susceptible human T cells by exploiting activation stimuli. This platform remains compatible with high-capacity vectors for up to 37 kb DNA delivery, increasing payload capacity and safety because of the removal of all viral genes. Together, these findings provide a tool for targeted delivery of large payloads in T cells as a potential avenue to overcome current limitations of T cell therapy.

41. Monitoring coastal land-water topography – Very high resolution satellite data analytics reveal data in space and time

Abstract

Coastal morphology is key to modern understanding of threats and nature risks to coastal communities. A quarter of the world’s beaches are eroding due to natural or man-made activities. Dynamics of coastal morphology occurs in space and time (4D), making it difficult to measure using standard survey approaches. We introduce a technique that uses multispectral satellite data and satellite lidar to provide frequent spatial observations of coastal zones. Thanks to international cooperation through the Horizon 2020 program, the analytical processes are based on a highly automatic inversion of the radiative transfer equation (RTE), which enables to model shallow water depth without the access to local survey data. The workflow is installed on AWS cloud environment with directly linked to the satellite data archive. Leveraging these Copernicus satellite data, the solution can generate high frequent and high-resolution shallow water bathymetric data continuously and for the past decade. In combination with land topographic models derived from stereo satellite imagery and verification with bathy-topo data from a space-born green lidar system (IceSat-2 Atlas), this allows for verification and control of vertical accuracies.This solution allows for a rapid and scalable approach to generate shallow water bathymetric and topographic surfaces from small sites to a national level. It serves as a crucial dataset to manage coastal resilience strategies and reduce disaster risk. This will be demonstrated in pilot sites from the southern Baltic to the Caribbean where we can quantify and visualise the changes of the coastal morphology in 4D., The 28th IUGG General Assembly (IUGG2023) (Berlin 2023)
