Today's high-power laser systems are capable of reaching photon intensities up to 1022 W cm-2, generating plasmas when interacting with material. The high intensity and ultrashort laser pulse duration (fs) make direct observation of plasma dynamics a challenging task. In the field of laser-plasma physics and especially for the acceleration of ions, the spatio-temporal intensity distribution is one of the most critical aspects. We describe a novel method based on a single-shot (i.e. single laser pulse) chirped probing scheme, taking nine sequential frames at frame rates up to THz. This technique, to which we refer as temporally resolved intensity contouring (TRIC) enables single-shot measurement of laser-plasma dynamics. Using TRIC, we demonstrate the reconstruction of the complete spatio-temporal intensity distribution of a high-power laser pulse in the focal plane at full pulse energy with sub-picosecond resolution.

The shape of a wave carries all information about the spatial and temporal structure of its source, given that the medium and its properties are known. Most modern imaging methods seek to utilize this nature of waves originating from Huygens' principle. We discuss the retrieval of the complete kinetic energy distribution from the acoustic trace that is recorded when a short ion bunch deposits its energy in water. This novel method, which we refer to as Ion-Bunch Energy Acoustic Tracing (I-BEAT), is a refinement of the ionoacoustic approach. With its capability of completely monitoring a single, focused proton bunch with prompt readout and high repetition rate, I-BEAT is a promising approach to meet future requirements of experiments and applications in the field of laser-based ion acceleration. We demonstrate its functionality at two laser-driven ion sources for quantitative online determination of the kinetic energy distribution in the focus of single proton bunches.

To ensure the safe and efficient application of ocean alkalinity enhancement (OAE), it is crucial to investigate its impacts on the carbonate system. While modeling studies reported a sequestration potential of 3–30 Gt carbon dioxide (CO2) per year (Oschlies et al., 2023), there has been a lack of empirical data to support the applicability of this technology in natural environments. Recent studies have described the effect of runaway carbonate precipitation in the context of OAE, showing that calcium carbonate (CaCO3) formation was triggered if certain Ωaragonite saturation thresholds were exceeded. This effect could potentially lead to a net loss of the initially added alkalinity, counteracting the whole concept of OAE. The related precipitation can adversely affect the carbon storage capacity and may in some cases result in CO2 emissions. Experiments at the Espeland marine biological station (Bergen, Norway) were conducted to systematically study the chemical consequences of OAE deployment. The experiments lasted for 20–25 d to monitor the temporal development of carbonate chemistry parameters after alkalinity addition and the subsequent triggered carbonate precipitation process. Identified uniform patterns before and during the triggered runaway process can be described by empirical functional relationships. For approaches equilibrated to the CO2 concentration of the atmosphere, total alkalinity (TA) levels of up to 6500 µmolkg-1 remained stable without loss of total alkalinity (TA) for up to 20 d. Higher implemented TA levels, up to 11 200 µmolkg-1 , triggered runaway carbonate formation. Once triggered, the loss of alkalinity continued until the Ωaragonite values leveled out at 5.8–6.0, still resulting in a net gain of 3600–4850 µmolkg-1 in TA. The non- CO2 -equilibrated approaches, however, only remained stable for TA additions of up to 1000 µmolkg-1. The systematic behavior of treatments exceeding this level allows us to predict the duration of transient stability and the quantity of TA loss after this period. Once triggered, the TA loss continued in the non- CO2 -equilibrated approaches until Ωaragonite values of 2.5–5.0 were reached, in this case resulting in a net loss of TA. To prevent a net loss of TA, treated water must be diluted below the time-dependent critical levels of TA and Ωaragonite within the identified transient stability duration. Identified stability and loss patterns of added TA depend on local environmental conditions impacting the carbonate system, such as salinity, temperature, biological activity, and particle abundance. Incorporating such stability and loss patterns into ocean biogeochemical models, which are capable of resolving dilution processes of treated and untreated water parcels, would, from a geochemical perspective, facilitate the prediction of safe local application levels of OAE. This approach would also allow an accurate determination of the fate of added alkalinity and a more realistic carbon storage potential estimation compared to the assessments that neglect carbonate system responses to OAE. The Brāhmī leaf in Sanskrit and Uigur (TT VIII H) from the Berlin Turfan collection, edited by A. von Gabain in 1954, suggests an Indian origin, although this cannot be definitively proven in its current form. The fragment appears to be a commentary on the Agraprajñaptisūtra, the sūtra that declares the triratna (Buddha, Dharma, Saṃgha) as the best. The preserved part is about the question of its origin or occasion (utpatti). The present new edition includes an introduction on the Agraprajñaptisūtra (I), the text, translation, and comments (III), along with the description of the leaf, characteristic usage of the Uigur Brāhmī script and thoughts on dating (II) and three Appendices (IV) on the Agraprajñaptisūtra, the *Ekāgrasūtra in Uigur sources, and the interpretation of etadagrikeṣu vyākr̥teṣu. We report on a target system supporting automated positioning of nano-targets with a precision resolution of in three dimensions. It relies on a confocal distance sensor and a microscope. The system has been commissioned to position nanometer targets with 1Â Hz repetition rate. Integrating our prototype into the table-top ATLAS 300 TW-laser system at the Laboratory for Extreme Photonics in Garching, we demonstrate the operation of a 0.5Â Hz laser-driven proton source with a shot-to-shot variation of the maximum energy about 27% for a level of confidence of 0.95. The reason of laser shooting experiments operated at 0.5Â Hz rather than 1Â Hz is because the synchronization between the nano-foil target positioning system and the laser trigger needs to improve.

Acting through a combination of direct and indirect pathogen clearance mechanisms, blood-derived antimicrobial compounds (AMCs) play a pivotal role in innate immunity, safeguarding the host against invading microorganisms. Besides their antimicrobial activity, some AMCs can neutralize endotoxins, preventing their interaction with immune cells and avoiding an excessive inflammatory response. In this study, we aimed to investigate the influence of unfractionated heparin, a polyanionic drug clinically used as anticoagulant, on the endotoxin-neutralizing and antibacterial activity of blood-derived AMCs. Serum samples from healthy donors were pre-incubated with increasing concentrations of heparin for different time periods and tested against pathogenic bacteria (Acinetobacter baumannii, Enterococcus faecium, Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa, Staphylococcus aureus) and endotoxins from E. coli, K. pneumoniae, and P. aeruginosa. Heparin dose-dependently decreased the activity of bloodderived AMCs. Consequently, pre-incubation with heparin led to increased activity of LPS and higher values of the pro-inflammatory cytokines tumor necrosis factor a (TNF-a) and interleukin 6 (IL-6). Accordingly, higher concentrations of A. baumannii, E. coli, K. pneumoniae, and P. aeruginosa were observed as well. These findings underscore the neutralizing effect of unfractionated heparin on blood-derived AMCs in vitro and may lead to alternative affinity techniques for isolating and characterizing novel AMCs with the potential for clinical translation. Introduction: When the kidneys or liver fail, toxic metabolites accumulate in the patient's blood, causing cardiovascular and neurotoxic complications and increased mortality. Conventional membrane-based extracorporeal blood purification procedures cannot remove these toxins efficiently. The aim of this in vitro study was to determine whether commercial hemoperfusion adsorbers are suitable for removing protein-bound retention solutes from human plasma and whole blood as well as to compare the removal to conventional hemodialysis. Methods: For in vitro testing of the removal of protein-bound substances, whole blood and plasma were spiked with uremic retention solutes (homocysteine, hippuric acid, indoxyl sulfate, 3-carboxy-4-methyl-5-propyl-2-furanpropionic acid) and the toxins of liver failure (bilirubin, cholic acid, tryptophan, phenol). Subsequently, the protein binding of each retention solute was determined. The adsorption characteristics of the hemoperfusion adsorbers, Jafron HA and Biosky MG, both approved for the adsorption of protein-bound uremic retention solutes and Cytosorb, an adsorber recommended for adsorption of cytokines, were tested by incubating them in spiked whole blood or plasma for 1 h. Subsequently, the adsorption characteristics of the adsorbers were tested in a dynamic system. For this purpose, a 6-h in vitro hemoperfusion treatment was compared with an equally long in vitro hemodialysis treatment. Results: Hippuric acid, homocysteine, indoxyl sulfate, and tryptophan were most effectively removed by hemodialysis. Bilirubin and cholic acid were removed best by hemoperfusion with Cytosorb. A treatment with Jafron HA and Biosky MG showed similar results for the adsorption of the tested retention solutes and were best for removing phenol. 3-Carboxy-4-methyl-5-propyl-2-furanpropionic acid could not be removed with any treatment method. Discussion/Conclusion: A combination of hemodialysis with hemoperfusion seems promising to improve the removal of some toxic metabolites in extracorporeal therapies. Aims Catheter ablation (CA) of post-ablation left atrial tachycardias (LATs) can be challenging. So far, pulsed field ablation (PFA) has not been compared to standard point-by-point radiofrequency current (RFC) energy for LAT ablation. To compare efficacy of PFA vs. RFC in patients undergoing CA for LAT. Methods and results Consecutive patients undergoing LAT-CA were prospectively enrolled (09/2021–02/2023). After electro-anatomical high-density mapping, ablation with either a pentaspline PFA catheter or RFC was performed. Patients were matched 1:1. Ablation was performed at the assumed critical isthmus site with additional ablation, if necessary. Right atrial tachycardia (RAT) was ablated with RFC. Acute and chronic success were assessed. Fifty-six patients (n = 28 each group, age 70 ± 9 years, 75% male) were enrolled. A total of 77 AT (n = 67 LAT, n = 10 RAT; 77% macroreentries) occurred with n = 32 LAT in the PFA group and n = 35 LAT in the RFC group. Of all LAT, 94% (PFA group) vs. 91% (RFC group) successfully terminated to sinus rhythm or another AT during ablation (P = 1.0). Procedure times were shorter (PFA: 121 ± 41 vs. RFC: 190 ± 44 min, P < 0.0001) and fluoroscopy times longer in the PFA group (PFA: 15 ± 9 vs. RFC: 11 ± 6 min, P = 0.04). There were no major complications. After one-year follow-up, estimated arrhythmia free survival was 63% (PFA group) and 87% (RFC group), [hazard ratio 2.91 (95% CI: 1.11–7.65), P = 0.0473]. Conclusion Pulsed field ablation of post-ablation LAT using a pentaspline catheter is feasible, safe, and faster but less effective compared to standard RFC ablation after one year of follow-up. The development of carbon dioxide removal methods, coupled with decreased CO2 emissions, is fundamental to achieving the targets outlined in the Paris Agreement limiting global warming to 1.5 °C. Here we are investigating the importance of the organic carbon feedstock to support silicate mineral weathering in small-scale flow through bioreactors and subsequent CO2 sequestration. Here, we combine two bacteria and two fungi, widely reported for their weathering potential, in simple flow through bioreactors (columns) consisting of forsterite and widely available, cheap organic carbon sources (wheat straw, bio-waste digestate of pig manure and biowaste, and manure compost), over six weeks. Compared to their corresponding abiotic controls, the inoculated straw and digestate columns release more total alkalinity (~2 times more) and produce greater dissolved and solid inorganic carbon (29% for straw and 13% for digestate), suggesting an increase in CO2 sequestration because of bio-enhanced silicate weathering. Microbial biomass is higher in the straw columns compared to the digestate and manure compost columns, with a phospholipid fatty acid derived total microbial biomass 10 x greater than the other biotic columns. Scanning Electron Microscopy imaging shows the most extensive colonisation and biofilm formation on the mineral surfaces in the straw columns. The biotic straw and digestate columns sequester 50 and 14 mg C more than their abiotic controls respectively, while there is no difference in the manure columns. The selection of organic carbon sources to support microbial communities in the flow through bioreactors controlls the silicate weathering rates and CO2 sequestration. [ABSTRACT FROM AUTHOR]

Ocean alkalinity enhancement (OAE) is considered for the long-term removal of gigatons of carbon dioxide (CO2) from the atmosphere to achieve our climate goals. Little is known, however, about the ecosystem-level changes in biogeochemical functioning that may result from the chemical sequestration of CO2 in seawater, and how stable the sequestration is. We studied these two aspects in natural plankton communities under carbonate-based, CO2-equilibrated OAE in the nutrient-poor North Atlantic. During a month-long mesocosm experiment, the majority of biogeochemical pools, including inorganic nutrients, particulate organic carbon and phosphorus as well as biogenic silica, remained unaltered across all OAE levels of up to a doubling of ambient alkalinity (+2400 µeq kg-1). Noticeable exceptions were a minor decrease in particulate organic nitrogen and an increase in the carbon to nitrogen ratio (C:N) of particulate organic matter in response to OAE. Thus, in our nitrogen limited system, nitrogen turnover processes appear more susceptible than those of other elements leading to decreased food quality and increased organic carbon storage. However, alkalinity and chemical CO2 sequestration were not stable at all levels of OAE. Two weeks after alkalinity addition, we measured a loss of added alkalinity and of the initially stored CO2 in the mesocosm where alkalinity was highest (+2400 µeq kg-1, Ωaragonite ~10). The loss rate accelerated over time. Additional tests showed that such secondary precipitation can be initiated by particles acting as precipitation nuclei and that this process can occur even at lower levels of OAE. In conclusion, on the one hand, our study under carbonate-based OAE where the carbon is already sequestered, the risk of major and sustained impacts on biogeochemical functioning may be low in the nutrient-poor ocean. On the other hand, the durability of carbon sequestration using OAE could be constrained by alkalinity loss in supersaturated waters with precipitation nuclei present. Our study provides evaluation of ecosystem impacts of an idealised OAE deployment for monitoring, reporting and verification (MRV) in an oligotrophic system. Whether biogeochemical functioning is resilient to more technically simple and economically more viable approaches that induce stronger water chemistry perturbations remains to be seen. Groundwater level (GWL) dynamics result from a complex interplay between groundwater systems and the Earth system. This study aims to identify common hydrogeological patterns and to gain a deeper understanding of the underlying similarities and their link to physiographic, climatic, and anthropogenic controls of groundwater in coastal regions. The most striking aspects of GWL dynamics and their controls were identified through a combination of statistical metrics, calculated from about 8000 groundwater hydrographs, pattern recognition using clustering algorithms, classification using random forest, and SHapley Additive exPlanations (SHAPs). Hydrogeological similarity was defined by four clusters representing distinct patterns of GWL dynamics. These clusters can be observed globally across different continents and climate zones but simultaneously vary regionally and locally, suggesting a complicated interplay of controlling factors. The main controls differentiating GWL dynamics were identified, but we also provide evidence for the currently limited ability to explain GWL dynamics on large spatial scales, which we attribute mainly to uncertainties in the explanatory data. Finally, this study provides guidance for systematic and holistic groundwater monitoring and modeling and motivates a consideration of the different aspects of GWL dynamics, for example, when predicting climate-induced GWL changes, and the use of explainable machine learning techniques to deal with GWL complexity – especially when information on potential controls is limited or needs to be verified.
