The integration of Artificial Intelligence (AI), particularly Large Language Model (LLM)-based systems, in education has shown promise in enhancing teaching and learning experiences. However, the advent of Multimodal Large Language Models (MLLMs) like GPT-4 with vision (GPT-4V), capable of processing multimodal data including text, sound, and visual inputs, opens a new era of enriched, personalized, and interactive learning landscapes in education. Grounded in theory of multimedia learning, this paper explores the transformative role of MLLMs in central aspects of science education by presenting exemplary innovative learning scenarios. Possible applications for MLLMs could range from content creation to tailored support for learning, fostering competencies in scientific practices, and providing assessment and feedback. These scenarios are not limited to text-based and uni-modal formats but can be multimodal, increasing thus personalization, accessibility, and potential learning effectiveness. Besides many opportunities, challenges such as data protection and ethical considerations become more salient, calling for robust frameworks to ensure responsible integration. This paper underscores the necessity for a balanced approach in implementing MLLMs, where the technology complements rather than supplants the educator's role, ensuring thus an effective and ethical use of AI in science education. It calls for further research to explore the nuanced implications of MLLMs on the evolving role of educators and to extend the discourse beyond science education to other disciplines. 8. Using HeuristicsMiner to Analyze Problem-Solving Processes: Exemplary Use Case of a Productive-Failure Study

- Abstract
This paper presents a fine-grained process analysis of 22 students in a classroom-based learning setting. The students engaged (and failed) in problem-solving attempts prior to instruction (i.e., the Productive-Failure approach). We used the HeuristicsMiner algorithm to analyze the data of a quasi-experimental study. The applied algorithm allowed us to investigate temporally structured think-aloud data, to outline productive and unproductive problem-solving strategies. Our analyses and findings demonstrated that HeuristicsMiner enables researchers to effectively mine problem-solving processes and sequences, even for smaller sample sizes, which cannot be done with traditional coding-and-counting strategies. The limitations of the algorithm, as well as further implications for educational research and practice, are also discussed.

Oligodendrogliomas are defined at the molecular level by the presence of an IDH mutation and codeletion of chromosomal arms 1p and 19q. In the past, case reports and small studies described gliomas with sarcomatous features arising from oligodendrogliomas, so called oligosarcomas. Here, we report a series of 24 IDH-mutant oligosarcomas from 23 patients forming a distinct methylation class. The tumors were recurrences from prior oligodendrogliomas or developed de novo. Precursor tumors of 12 oligosarcomas were histologically and molecularly indistinguishable from conventional oligodendrogliomas. Oligosarcoma tumor cells were embedded in a dense network of reticulin fibers, frequently showing p53 accumulation, positivity for SMA and CALD1, loss of OLIG2 and gain of H3K27 trimethylation (H3K27me3) as compared to primary lesions. In 5 oligosarcomas no 1p/19q codeletion was detectable, although it was present in the primary lesions. Copy number neutral LOH was determined as underlying mechanism. Oligosarcomas harbored an increased chromosomal copy number variation load with frequent CDKN2A/B deletions. Proteomic profiling demonstrated oligosarcomas to be highly distinct from conventional CNS WHO grade 3 oligodendrogliomas with consistent evidence for a smooth muscle differentiation. Expression of several tumor suppressors was reduced with NF1 being lost frequently. In contrast, oncogenic YAP1 was aberrantly overexpressed in oligosarcomas. Panel sequencing revealed mutations in NF1 and TP53 along with IDH1/2 and TERT promoter mutations. Survival of patients was significantly poorer for oligosarcomas as first recurrence than for grade 3 oligodendrogliomas as first recurrence. These results establish oligosarcomas as a distinct group of IDH-mutant gliomas differing from conventional oligodendrogliomas on the histologic, epigenetic, proteomic, molecular and clinical level. The diagnosis can be based on the combined presence of (a) sarcomatous histology, (b) IDH-mutation and (c) TERT promoter mutation and/or 1p/19q codeletion, or, in unresolved cases, on its characteristic DNA methylation profile.

Glioblastoma IDH-wildtype presents with a wide histological spectrum. Some features are so distinctive that they are considered as separate histological variants or patterns for the purpose of classification. However, these usually lack defined (epi-)genetic alterations or profiles correlating with this histology. Here, we describe a molecular subtype with overlap to the unique histological pattern of glioblastoma with primitive neuronal component. Our cohort consists of 63 IDH-wildtype glioblastomas that harbor a characteristic DNA methylation profile. Median age at diagnosis was 59.5 years. Copy-number variations and genetic sequencing revealed frequent alterations in TP53, RB1 and PTEN, with fewer gains of chromosome 7 and homozygous CDKN2A/B deletions than usually described for IDH-wildtype glioblastoma. Gains of chromosome 1 were detected in more than half of the cases. A poorly differentiated phenotype with frequent absence of GFAP expression, high proliferation index and strong staining for p53 and TTF1 often caused misleading histological classification as carcinoma metastasis or primitive neuroectodermal tumor. Clinically, many patients presented with leptomeningeal dissemination and spinal metastasis. Outcome was poor with a median overall survival of only 12 months. Overall, we describe a new molecular subtype of IDH-wildtype glioblastoma with a distinct histological appearance and genetic signature.

Clear cell meningioma represents an uncommon variant of meningioma that typically affects children and young adults. Although an enrichment of loss-of-function mutations in the SMARCE1 gene has been reported for this subtype, comprehensive molecular investigations are lacking. Here we describe a molecularly distinct subset of tumors (n = 31), initially identified through genome-wide DNA methylation screening among a cohort of 3093 meningiomas, of which most were diagnosed histologically as clear cell meningioma. This cohort was further supplemented by an additional 11 histologically diagnosed clear cell meningiomas for analysis (n = 42). Targeted DNA sequencing revealed SMARCE1 mutations in 33/34 analyzed samples, accompanied by a nuclear loss of expression determined via immunohistochemistry and a decreased SMARCE1 transcript expression in the tumor cells. Analysis of time to progression or recurrence of patients within the clear cell meningioma group (n = 14) in comparison to those with meningioma WHO grade 2 (n = 220) revealed a similar outcome and support the assignment of WHO grade 2 to these tumors. Our findings indicate the existence of a highly distinct epigenetic signature of clear cell meningiomas, separate from all other variants of meningiomas, with recurrent mutations in the SMARCE1 gene. This suggests that these tumors may arise from a different precursor cell population than the broad spectrum of the other meningioma subtypes.

25. Productive versus Vicarious Failure: Do Students Need to Fail Themselves in Order to Learn?

- Abstract
Productive Failure is an instructional approach in which students engage in problem-solving attempts prior to instruction. The Productive Failure approach has been found to be more supportive for conceptual knowledge acquisition during subsequent instruction compared to a direct-instruction condition in which students receive instruction first followed by problem solving. Failing at problem-solving attempts prior to instruction is considered to prepare students to better understand the concepts underlying the problem. However, it is an open question whether students need to fail themselves to be effectively prepared for subsequent instruction. We therefore compared the effectiveness of two alternative preparatory activities: Students either engaged in problem-solving attempts themselves or were shown problem-solving attempts of another student. This experimental variation allows to show the necessity of own failure for the effectiveness of the PF approach. The results indicated that the two conditions were equally effective at preparing students for subsequent instruction.

28. Examining Positive and Negative Interdependence in an Elementary School CSCL Setting

- Abstract
Social interdependence is a key concept in CSCL research. However, investigations of students' positive and negative interdependence during collaborative activities have often relied on self-report, rather than dialogue analysis. Bringing together politeness and social interdependence theory, we assessed "dialogue indicators" of positive and negative interdependence from behavioral data (namely, face-saving and face-threatening dialogue moves) and compared the results with those of self-report scales. We analyzed a data set of 30 elementary students learning fractions with an intelligent tutoring system (ITS). Our initial analyses focus on the link between use of language that is face-saving (e.g., marking identity with statements such as "we are great") or face-threatening (e.g., insulting), and students' preferences to collaborate and compete. We found only non-significant correlations between these two broad categories, but found significant correlations between dialogue indicators, such as the use of identity markers and joking, that suggest directions for subsequent studies.

29. Preparatory Effects of Problem Solving versus Studying Examples Prior to Instruction

The Productive Failure (PF) approach prompts students to attempt to solve a problem prior to instruction -- at which point they typically fail. Yet, research on PF shows that students who are involved in problem solving prior to instruction gain more conceptual knowledge from the subsequent instruction compared to students who receive the instruction first. So far, there is no conclusive evidence, however, that the beneficial effects of PF are explained by the attempt to generate "one's own" solutions prior to instruction. The literature on example-based learning suggests that observing someone else engaging in problem-solving attempts may be an equally effective means to prepare students for instruction. In an experimental study, we compared a PF condition, in which students were actively engaged in problem solving prior to instruction, to two example conditions, in which students either observed the complete "problem-solving-and-failing" process of another student engaging in PF or looked at the outcome of this process (i.e., another student's failed solution attempts). Rather than worked examples of the correct solution procedure, the students observed examples of failed solution attempts. We found that students' own problem solving was not superior to the two example conditions. In fact, students who observed the complete PF process even outperformed students who engaged in PF themselves. Additional analyses revealed that the students' prior knowledge moderated this effect: While students who observed the complete PF process were able to take advantage of their prior knowledge to gain more conceptual knowledge from the subsequent instruction, prior knowledge did not affect students' post-test performance in the PF condition.

ABSTRACT Purpose Methods Results Conclusion Microfocused ultrasound with real‐time visualization (MFU‐V) is often used for noninvasive skin lifting, by precisely targeting dermal and subcutaneous tissues to create thermal coagulation points (TCPs). These TCPs denature collagen and initiate a transient inflammatory response, ultimately attracting dermal fibroblasts and inducing efficient neocollagenesis and extracellular matrix (ECM) remodeling, yielding to MFU‐V's desired skin‐lifting effects. The current study investigates MFU‐V's underlying mode of action based on the histological progression of TCPs in the skin, providing new insight into the technology's regenerative effect.Following standard triple‐depth MFU‐V treatment, in vivo skin samples were assessed using histology and immunohistochemistry to evaluate TCPs, heat shock protein (HSP47), and elastin expression in fibroblasts.MFU‐V treatment induced elongated, flame‐like TCPs with denatured collagen at focal depths of 1.5, 3.0, and 4.5 mm within the skin—each corresponding to its respective transducer depth. Time‐dependent progression of TCPs showed significantly increased scores of fibroblasts and mature collagen along with recruitment of HSP47‐positive fibroblasts to TCP areas on Day 90. Collagen formation and later maturation were visualized. Newly synthesized elastin significantly increased in the TCP area on Day 90 compared to Day 14.This work provides histological evidence of stimulation and regeneration of newly synthesized elastin fibers after TCP induction. MFU‐V‐generated TCPs triggered the body's own healing cascade of collagen denaturation, transient inflammation, proliferation, and tissue remodeling, resulting in attraction of HSP47‐positive fibroblasts to the TCP sites, and new collagen and elastin fiber regeneration by fibroblasts. Besides the well‐described neocollagenesis, this study demonstrates that MFU‐V treatment induces elastin neogenesis that may result not only in skin lifting but also in improved skin elasticity, providing an overall regenerative effect. We also explored whether the perceived model-observer similarity had an impact on the effectiveness of observing examples of failure. The results showed that observing examples effectively prepares students for learning from instruction. However, observing the model's problem-solving-and-failing process did not prepare students more effectively than merely looking at the outcome. Studying examples were more effective if model-observer similarity was high.

47. Influence of Eddy Current Losses in Nonsuperconducting Layers of HTS in Superconducting Electrical Machines

- Abstract
Numerical analysis of high-temperature superconductors (HTS) commonly addresses ac losses in the superconducting ReBCO layer. However, alternating magnetic fields also induce eddy current losses in the metallic nonsuperconducting (non-SC) substrate, silver, and copper layers of the HTS, which cannot be overlooked for certain operating temperatures and frequencies. Previously, non-SC losses have been estimated numerically for simple geometries at 77 K. Using a multilayer H-A-formulated model, this article estimates the eddy current losses for a full-scale 2.5-MW SC aviation motor for frequencies between 50 and 1000 Hz and temperatures between 25 and 60 K. Furthermore, a sensitivity study is presented to show how ac losses are impacted by specific design parameters, such as the copper purity, angle of the external field, tapes per turn, tape height, and copper layer thickness. The results show that eddy current losses are negligible for high HTS temperatures and low frequencies. However, at low temperatures and high frequencies, these losses represent a significant share of the total, meaning that while they can be neglected for low-speed machinery, they must be considered for power-dense machinery, particularly if the planned cooling method involves liquid hydrogen temperatures.
