15. Integrating Urban Schools in a Modern Context: Roadblocks and Challenges with the Use of Magnet Schools

Abstract

It has been 50 years since magnet schools were first used to address segregation in public schools. The context of magnet schools has changed dramatically during this time, raising questions about the role of magnet schools in a modern context. This study utilized data from a large urban district with more than 100 magnet schools to assess integration from magnet schools in a modern context. The results point to integration in magnet schools, but this integration comes at the cost of segregating the traditional public schools in the district. Interdistrict transfers could improve the integrative capacity of magnet schools.

16. Career and Technical Education Credentials in Virginia High Schools: Trends in Attainment and College Enrollment Outcomes. REL 2022-132

Abstract

In Virginia, there has been a long-term effort to increase the number of graduates who earn career and technical education (CTE) credentials. These CTE credentials are intended to provide high school graduates with additional preparation for college and careers. In 2013, the Virginia Board of Education added a CTE credential requirement to the Standard diploma for students who entered grade 9 for the first time in 2013 or later. Graduates can complete this requirement by passing an approved assessment and do not have to take any CTE courses. The CTE credential requirement was added to the Standard diploma because graduates earning this diploma are less likely to enroll in, persist in, or complete college than graduates who earn Virginia's other main diploma, the Advanced Studies diploma. In addition, this new policy is part of the state's efforts to eliminate racial and socioeconomic inequities in Virginia public schools. Graduates who are Black or Hispanic, who are English learner students, who are in economically disadvantaged circumstances, or who receive special education services are more likely than other groups of graduates to earn the Standard diploma rather than the Advanced Studies diploma. At the request of Virginia CTE leaders, the Regional Educational Laboratory Appalachia conducted a descriptive study of attainment rates of CTE credentials, completion rates of CTE programs of study, and college enrollment rates for Standard diploma graduates from 2011 to 2017, the years before and after the policy change. Education stakeholders in Virginia and elsewhere can use the results of this study to inform their CTE policies. From 2011 to 2017, the percentage of Standard diploma graduates who earned at least one CTE credential increased from 23 percent to 91 percent. A similar increase occurred among Advanced Studies diploma graduates, even though the CTE credential requirement applied only to Standard diploma graduates. The attainment rates of CTE credentials increased for all groups of Standard diploma graduates, including groups based on demographic characteristics, federal program participation, and academic achievement. While the percentages of Standard diploma graduates who earned a CTE credential increased consistently from 2011 to 2017, their college enrollment rates dropped. The percentage of Standard diploma graduates completing a CTE program of study, which requires taking CTE courses that are not required to earn a credential but may still be helpful for later student outcomes, decreased in 2016 and 2017. The study findings suggest a need to examine workforce outcomes for Standard diploma graduates to fully understand whether this policy is meeting its intended goals. In addition, the findings suggest a need to consider other methods to address outcomes for Virginia's Standard diploma graduates, such as support for implementing practices with rigorous evidence of effectiveness for improving college and career outcomes. Regional Educational Laboratory Appalachia researchers developed the Cost-Feasibility Analysis (CFA) Toolkit to help education leaders estimate whether implementing supplemental online programs is affordable given a school or district's available resources. The CFA Toolkit guides users through a four-stage process that yields cost information that can support decision-making about implementing such a program. The toolkit includes guidance, helpful resources, and an Excel-based cost-estimation tool that supports users with planning (stage 1), collecting data (stage 2), estimating program costs (stage 3), and determining the feasibility of implementing the supplemental online program (stage 4).

Abstract

Antibias training is increasingly identified as a strategy to reduce maternal health disparities. Evidence to guide this work is limited. We conducted a community-guided scoping review to characterize new antibias research. Four of 508 projects met our criteria: US-based, publicly funded, initiated from January 1, 2018 to June 30, 2022, and featuring an intervention to reduce bias or racism in maternal health care providers. Training was embedded in multicomponent interventions in 3 projects, limiting its evaluation as a stand-alone intervention. Major public funders have sponsored few projects to advance antibias training research in maternal health. More support is needed to develop a rigorous and scalable evidence base.

19. Virginia High School Graduates' Career and Technical Education Credentials: Top Credentials over Time and across Student Groups. REL 2021-063

Abstract

In Virginia, all high school students can earn either a Standard diploma or an Advanced Studies diploma, the latter being a college preparatory diploma. Starting in 2017, the Virginia Department of Education (VDOE) began requiring students graduating with the Standard diploma to earn a career and technical education (CTE) credential to encourage them to pursue opportunities that enhance their career readiness. This is likely to be particularly important for students graduating with the Standard diploma, as they have been shown to have limited success in postsecondary education. This study examined the CTE credentials Virginia high school graduates most commonly earned from 2011 through 2017. The five most commonly earned CTE credentials in Virginia remained the same during this time period, but the percentage of students earning the Workplace Readiness Skills (WRS) and W!se Financial Literacy Certification credentials increased. Both of these credentials cover broad skills relevant to a wide range of jobs, as opposed to a specific occupation or industry. Although the new CTE requirement applies only to Standard diploma graduates, there were few differences in the top 10 credentials by diploma type, both in terms of which credentials were most common as well as the rates at which students earned these credentials. Regardless of diploma type, in 2017, 9 of the top 10 credentials were broad credentials that were not narrowly aligned to a specific occupation or industry. This study also looked at the top 10 credentials earned by 2017 Standard diploma graduates across a variety of student subgroups, including English learner students, economically disadvantaged students, students with disabilities, and racial/ethnic subgroups. English learner students and students with disabilities earned the top 10 credentials at lower rates than other Standard diploma graduates. Student credential-earning rates differed the most by geographic region, both in terms of which credentials appeared in the top 10 and the percentage of students earning the top 10 credentials. This study highlights the need for additional analyses to help CTE stakeholders and policymakers understand the value of different types of CTE credentials. In particular, Virginia and other states might explore the relative value of broad CTE credentials that apply to a wide range of jobs and have become increasingly prevalent in Virginia compared with CTE credentials that are more narrowly aligned with a specific occupation or industry.

Abstract

BackgroundGenetic mutations underlying familial Alzheimer's disease (AD) were identified decades ago, but the field is still in search of transformative therapies for patients. While mouse models based on overexpression of mutated transgenes have yielded key insights in mechanisms of disease, those models are subject to artifacts, including random genetic integration of the transgene, ectopic expression and non-physiological protein levels. The genetic engineering of novel mouse models using knock-in approaches addresses some of those limitations. With mounting evidence of the role played by microglia in AD, high-dimensional approaches to phenotype microglia in those models are critical to refine our understanding of the immune response in the brain.MethodsWe engineered a novel App knock-in mouse model (AppSAA) using homologous recombination to introduce three disease-causing coding mutations (Swedish, Arctic and Austrian) to the mouse App gene. Amyloid-β pathology, neurodegeneration, glial responses, brain metabolism and behavioral phenotypes were characterized in heterozygous and homozygous AppSAA mice at different ages in brain and/ or biofluids. Wild type littermate mice were used as experimental controls. We used in situ imaging technologies to define the whole-brain distribution of amyloid plaques and compare it to other AD mouse models and human brain pathology. To further explore the microglial response to AD relevant pathology, we isolated microglia with fibrillar Aβ content from the brain and performed transcriptomics and metabolomics analyses and in vivo brain imaging to measure energy metabolism and microglial response. Finally, we also characterized the mice in various behavioral assays.ResultsLeveraging multi-omics approaches, we discovered profound alteration of diverse lipids and metabolites as well as an exacerbated disease-associated transcriptomic response in microglia with high intracellular Aβ content. The AppSAA knock-in mouse model recapitulates key pathological features of AD such as a progressive accumulation of parenchymal amyloid plaques and vascular amyloid deposits, altered astroglial and microglial responses and elevation of CSF markers of neurodegeneration. Those observations were associated with increased TSPO and FDG-PET brain signals and a hyperactivity phenotype as the animals aged.DiscussionOur findings demonstrate that fibrillar Aβ in microglia is associated with lipid dyshomeostasis consistent with lysosomal dysfunction and foam cell phenotypes as well as profound immuno-metabolic perturbations, opening new avenues to further investigate metabolic pathways at play in microglia responding to AD-relevant pathogenesis. The in-depth characterization of pathological hallmarks of AD in this novel and open-access mouse model should serve as a resource for the scientific community to investigate disease-relevant biology.

22. Assessing the Alignment between West Virginia's High School Career and Technical Education Programs and the Labor Market. REL 2020-019

Abstract

To help students leave high school on a path toward success in the labor market, education policymakers and practitioners often focus on improving career and technical education (CTE) opportunities in high school. Understanding the alignment between high school CTE programs and the labor market is an important step in this process. To support CTE improvement efforts, this study quantitatively assessed the alignment between West Virginia's high school CTE programs and the labor market, with a focus on alignment to regional high-demand occupations that require moderate occupational preparation. These "high-demand study occupations" are the 20 occupations in each region of West Virginia with the largest number of long-term projected employment openings from 2014 to 2024 that require more than a high school diploma (for example, a license or work-related experience) but less than a bachelor's degree. The study found that 70 percent of West Virginia's long-term projected employment openings typically require some occupational preparation beyond a high school diploma but less than a bachelor's degree. Further, 93 percent of the regional long-term projected employment openings in high-demand study occupations were served by at least one aligned CTE program in the same region. However, students in only 53 percent of the state's CTE programs were in a program that aligned to at least one high-demand study occupation within their region. West Virginia stakeholders can use the findings to improve their CTE system's alignment and better prepare students for a postsecondary career. This study also serves as an example for policymakers and practitioners in other states who are interested in quantifying their CTE system's alignment in order to make data-informed decisions. Abstract

Here we report the generation of a multimodal cell census and atlas of the mammalian primary motor cortex as the initial product of the BRAIN Initiative Cell Census Network (BICCN). This was achieved by coordinated large-scale analyses of single-cell transcriptomes, chromatin accessibility, DNA methylomes, spatially resolved single-cell transcriptomes, morphological and electrophysiological properties and cellular resolution input-output mapping, integrated through cross-modal computational analysis. Our results advance the collective knowledge and understanding of brain cell-type organization1-5. First, our study reveals a unified molecular genetic landscape of cortical cell types that integrates their transcriptome, open chromatin and DNA methylation maps. Second, cross-species analysis achieves a consensus taxonomy of transcriptomic types and their hierarchical organization that is conserved from mouse to marmoset and human. Third, in situ single-cell transcriptomics provides a spatially resolved cell-type atlas of the motor cortex. Fourth, cross-modal analysis provides compelling evidence for the transcriptomic, epigenomic and gene regulatory basis of neuronal phenotypes such as their physiological and anatomical properties, demonstrating the biological validity and genomic underpinning of neuron types. We further present an extensive genetic toolset for targeting glutamatergic neuron types towards linking their molecular and developmental identity to their circuit function. Together, our results establish a unifying and mechanistic framework of neuronal cell-type organization that integrates multi-layered molecular genetic and spatial information with multi-faceted phenotypic properties.

Abstract

The spike protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is critical for virus infection through the engagement of the human ACE2 protein1 and is a major antibody target. Here we show that chronic infection with SARS-CoV-2 leads to viral evolution and reduced sensitivity to neutralizing antibodies in an immunosuppressed individual treated with convalescent plasma, by generating whole-genome ultra-deep sequences for 23 time points that span 101 days and using in vitro techniques to characterize the mutations revealed by sequencing. There was little change in the overall structure of the viral population after two courses of remdesivir during the first 57 days. However, after convalescent plasma therapy, we observed large, dynamic shifts in the viral population, with the emergence of a dominant viral strain that contained a substitution (D796H) in the S2 subunit and a deletion (ΔH69/ΔV70) in the S1 N-terminal domain of the spike protein. As passively transferred serum antibodies diminished, viruses with the escape genotype were reduced in frequency, before returning during a final, unsuccessful course of convalescent plasma treatment. In vitro, the spike double mutant bearing both ΔH69/ΔV70 and D796H conferred modestly decreased sensitivity to convalescent plasma, while maintaining infectivity levels that were similar to the wild-type virus.The spike substitution mutant D796H appeared to be the main contributor to the decreased susceptibility to neutralizing antibodies, but this mutation resulted in an infectivity defect. The spike deletion mutant ΔH69/ΔV70 had a twofold higher level of infectivity than wild-type SARS-CoV-2, possibly compensating for the reduced infectivity of the D796H mutation. These data reveal strong selection on SARS-CoV-2 during convalescent plasma therapy, which is associated with the emergence of viral variants that show evidence of reduced susceptibility to neutralizing antibodies in immunosuppressed individuals.

34. Adding It Up: How the Florida College and Career Readiness Initiative Impacted Developmental Education

Abstract

The Florida College and Career Readiness Initiative (FCCRI) was a statewide policy requiring college readiness testing and participation in college readiness courses for high school students. We used regression discontinuity to compare outcomes for students scoring just above and below test score cutoffs for assignment to FCCRI. We also examined impacts for students from a wider range of academic performance by using a before-after regression analysis to compare outcomes for targeted students before and after their schools implemented the FCCRI. The FCCRI increased the likelihood of enrolling in nondevelopmental courses for some targeted students, although results differ by academic performance. However, smaller differences in the likelihood of passing nondevelopmental courses suggest that some students were not prepared for these courses.

Florida enacted legislation in 2008 for a statewide program known as the Florida College and Career Readiness Initiative (FCCRI), which was intended to reduce the need for postsecondary remediation. The FCCRI consisted of testing grade 11 students to determine their college readiness and offering math and English college readiness and success (CRS) courses in grade 12 for students who did not test college-ready the year before. FCCRI is based on the theory that providing college readiness testing and CRS courses in high school may raise students' awareness of their academic deficiencies and motivate them to further develop college-level skills in their senior year. We found considerable variation across districts and schools in the initiative's implementation and level of compliance with state requirements for participation. We estimated program impacts using two different methods. First, we used a regression discontinuity design to compare outcomes for students scoring just above and below test score cutoffs for assignment to the FCCRI. Among the two cohorts of students required to participate in the FCCRI, we found little to no effect on short-term outcomes including high school graduation, college enrollment, and enrolling in or passing nondevelopmental courses. There is little evidence for improved enrollment or pass rates in for-credit coursework among the highest- and lowest-performing targeted students. However, enrollment and pass rates in transition and degree credit courses were similar for students on the margins of assignment to college readiness courses in either subject, indicating that students just below college-ready were able to "catch up" by the time they enrolled in college. There were also few differences between marginal targeted and non-targeted students on longer-term outcomes including persistence, transfer, nondevelopmental enrollment and pass, and degree completion rates. Second, we examined the effect of offering the FCCRI to students from a wider range of academic performance levels by using regression analysis to compare student outcomes for targeted students in schools before and after the schools implemented the FCCRI. As with the regression discontinuity analyses, we found that the FCCRI had little to no effect on most short-term student outcomes. We did find that the treatment group was more likely to both take and pass nondevelopmental courses in math and English. Although the average effects were small, the magnitudes of these effects were quite large for some portions of the achievement distribution, with effects of up to 10.7 percentage points for the treatment group. Finally, we estimated the cost for the FCCRI's ongoing implementation at $57 per targeted student in the 2014/2015 school year, with about 63 percent of per-student program costs incurred at the school level ($36), 33 percent at the district level ($19), and 3 percent at the state level ($2). We also found that while net costs exceeded benefits for the FCCRI as a whole, program effects and costs varied both within and between districts.

Abstract

A major sex difference in Alzheimer's disease (AD) is that men with the disease die earlier than do women. In aging and preclinical AD, men also show more cognitive deficits. Here, we show that the X chromosome affects AD-related vulnerability in mice expressing the human amyloid precursor protein (hAPP), a model of AD. XY-hAPP mice genetically modified to develop testicles or ovaries showed worse mortality and deficits than did XX-hAPP mice with either gonad, indicating a sex chromosome effect. To dissect whether the absence of a second X chromosome or the presence of a Y chromosome conferred a disadvantage on male mice, we varied sex chromosome dosage. With or without a Y chromosome, hAPP mice with one X chromosome showed worse mortality and deficits than did those with two X chromosomes. Thus, adding a second X chromosome conferred resilience to XY males and XO females. In addition, the Y chromosome, its sex-determining region Y gene (Sry), or testicular development modified mortality in hAPP mice with one X chromosome such that XY males with testicles survived longer than did XY or XO females with ovaries. Furthermore, a second X chromosome conferred resilience potentially through the candidate gene Kdm6a, which does not undergo X-linked inactivation. In humans, genetic variation in KDM6A was linked to higher brain expression and associated with less cognitive decline in aging and preclinical AD, suggesting its relevance to human brain health. Our study suggests a potential role for sex chromosomes in modulating disease vulnerability related to AD.

38. Pricing and funding with case mix models

Abstract

The aim of this workshop is to describe and explore current case mix costing systems in NZ and their use in funding. The workshop is designed to range from specifics (a cost system), through the process of compiling national case weights and their role in activity based funding, to a general perspective on these were used inside a District Health Board. The first speaker will describe the essential features of the Waitemata District cost system and how it relates to the National Common Costing Standards. The second speaker will explain the national system for calculating case weights and pricing them together with the determination of some specific price adjusters (e.g. the tertiary adjuster). Our third speaker will discuss what is activity based funding and how case mix fits into this together with some of the advantages of case mix in funding and decision making. Our fourth speaker will provide a historical perspective of how case mix was used in internal funding allocations and evaluation in a tertiary DHB. All four panellists have extensive experience in the New Zealand health sector and a detailed knowledge of case mix and funding at national and District levels. With the end of the population based funding model and the new health body, Te Whatu Ora, this workshop provides an essential foundation for considering different funding models for the New Zealand health sector. Consequently, it should be of interest to managers and clinicians in the healthcare sector, and especially for financial teams, business analysts and those providing consulting services to the sector. Abstract

School choice has emerged as the linchpin of President Trump's urban education reform plan, but it remains unclear how school choice policies will shape the educational experiences of the most underserved student groups, particularly English learners (ELs). Using quantitative data from one large urban school district, we examine EL participation in a system of school choice. Specifically, we investigate the extent to which never, current, and former ELs enroll in a nonzoned school. We find significant differences in the likelihood that students across these groups engage in school choice, raising important questions about whether school choice reforms are accessible to current ELs.
