Abstract The present study entails the systematic development and validation of a stability-indicating RP-HPLC method for the analysis of sitagliptin and ertugliflozin in a fixed-dose combination. Analytical quality by design (AQbD) concepts were used to define critical method variables, employing Pareto risk assessment and a Placket-Burman screening design, preceded by a Box-Behnken design with response surface analysis to optimise critical method parameters such as % acetonitrile (X1), buffer pH (X2) and column oven temperature (X3). Multiple response optimisation (Derringer’s desirability) of variables was accomplished by studying critical analytical attributes, such as resolution, retention time and theoretical plates. The title analytes were separated effectively on a PRONTOSIL C18 column at 37 °C using a mobile phase of acetonitrile:acetate buffer, pH 4.4 (36:64 percent v/v), pumped at a flow rate of 1 mL/min, and UV detection at 225 nm. Linearity was observed over a concentration range of 25-150 µg/mL and 3.75-22.5 µg/mL at retention times of 2.82 and 3.92 min for sitagliptin and ertugliflozin, respectively. The method obeyed all validation parameters of the ICH Q2(R1) guidelines. The proposed robust method allows the study of the selected drugs in pharmaceutical dosage forms as well as in drug stability studies under various stress conditions.

Popular IoT devices such as smartphones have dramatically enhance the network connection that has posed new unique security threats that calls for more developed security systems to protect connected systems. Thus, this research aims at comparing the performance of different machine learning algorithms that are K-Nearest Neighbors (KNN), Support Vector Machines (SVM), Random Forest (RF), and Deep Neural Networks (DNN) to address the anomalies identification and IoT security improvement. These algorithms were used in the study to assess the IoT traffic and attack data set. This is due to the fact that results which were obtained proved that DNNs had the highest level of accuracy of about 97. 5%, RF with 93, Breast with 90 and GYN with 87. 2%, SVM with 89. 7% and K-Nearest Neighbors with 85%. 4%. While the DNN had a better accuracy than the other models, the model was more computationally intensive compared to the other one; RF is a good trade-off between accuracy and time. KNN while being computationally cheap had the lowest accuracy and higher FPR. Comparison of the proposed work with already published literature also validates that as the present day algorithms provide rudimentary level of security, this study reveals that enhancing and combining these techniques is essential to enhance real-time detection and robustness of the systems. The intricate nature of remote sensing image dehazing poses a formidable challenge due to its multifaceted characteristics. Considered as a preliminary step for advanced remote sensing image tasks, haze removal becomes crucial. A novel approach is introduced with the objective of dehazing an image employing an encoder-decoder architecture embedded in a generative adversarial network (GAN). This innovative model systematically captures low-frequency information in the initial phase and subsequently assimilates high)frequency details from the remote sensing image. Incorporating a skip connection within the network serves the purpose of preventing information loss. To enhance the learning capability and assimilate more valuable insights, an additional component, the multi-scale attention module, is introduced. Drawing inspiration from multi-scale networks, an enhanced module is meticulously designed and incorporated at the network’s conclusion. This augmentation methodology aims to further enhance the dehazing capabilities by assimilating context information across various scales. The material for fine-tuning the dehazing algorithm has been obtained from the RICE-I dataset that serves as the testing ground for a comprehensive comparison between our proposed method and other two alternative approaches. The experimental results distinctly showcase the superior efficacy of our method, both in qualitative and quantitative terms. Our proposed methodology performed better with respect to contemporary dehazing techniques in terms of PSNR and SSIM although it requires longer simulation times. So, it could be concluded that we contributed a more comprehensive RS picture dehazing methodology to the existing dehazing methodology literature. [ABSTRACT FROM AUTHOR]

Ten natural dye yielding and two mordant plants were biochemically analyzed. Though natural dyes are widely used, information about the active principles responsible for dyeing is hardly available. In the present experiment, total chlorophyll, carotinoids, tannins, phenolics, flavonoids and curcumin were determined among the dye yielding plants, while K, S, P, Ca, Mg, Mn, Zn, Fe, Cu and Co were determined in the case of mordant plants. In Bixa orellana, used for yellow dyeing, the carotinoid content was 163.11 mg g-1 and in Clerodendrum chinense and Datura stramonium, which were used for green colouring, total chlorophyll content of 10.29 mg and 11.83 mg g-1 was recorded. Curcumin content responsible for orange colouring in Curcuma domestica was 27.7 mg g-1 while flavonoid content in Solanum nigrum and Terminalia chebula, which were used for brown, brown to black dyes was 24.89 and 21.73 mg g-1. Among the plants used for dyeing different colours, Punica granatum and Parkia timoriana were found to contain higher amounts of total phenols and bound phenols by recording 681.2 mg g-1 and 287.6 mg g-1 total phenols and 151.6 mg g-1 and 130.2mg g-1 bound phenols, while higher amounts of orthodihydric phenols and tannins were recorded in Punica granatum and Strobilanthes flaccidifolius by recording 20.11mg g-1 and 9.54mg g-1 orthodihydric phenols and 675.57mg g-1 and 648.12 mg g-1 tannins, respectively. In case of the plants used as mordant, higher contents of Ca, Mg, K, Zn, Fe and Mn were detected in Achyranthes aspera, while higher amounts of P, Fe and Cu were recorded in Garcinia xanthochymus.

Cancer is the uncontrolled proliferation of abnormal cells in the body. There is a foreseeable need for an effective anti-carcinogenic drug. In this regard, zerumbone (ZER) is identified as one such therapeutic herbal compound that has been shown to enhance the anticancer activity of cisplatin (CIS), with negligible side effects. Yet, the fundamental mechanisms of co-treatment of ZER and CIS on Hepatocellular carcinoma remain indefinable. The current study is endeavored to evaluate the anti-cancer effect of the individual and co-treatment of ZER, CIS and its combination on Diethyl nitrosamine induced hepatic cancer in wild-type zebra fish (Danio Rerio) models. Our careful analysis on treated and untreated fishes shows that CIS + ZER combination group restricted further progression of hepatocellular carcinoma cells significantly, which concludes that co-treatment of ZER with CIS was therapeutically effective for treating human HCC cancer cells which were induced into zebra fish.

Oxidative stress is considered as a major risk factor that contributes to age-related increase in lipid peroxidation and declined antioxidants in the central nervous system during aging. Grape seed extract, one of the bioflavonoid, is widely used for its medicinal properties. In the present study, we evaluated the role of grape seed extract on lipid peroxidation and antioxidant status in discrete regions of the central nervous system of young and aged rats. Male albino rats of Wistar strain were divided into four groups: Group I-control young rats, Group II-young rats treated with grape seed extract (100 mg/kg body weight) for 30 days, Group III-aged control rats and Group IV-aged rats supplemented with grape seed extract (100 mg/kg body weight) for 30 days. Age-associated increase in lipid peroxidation was observed in the spinal cord, cerebral cortex, striatum and the hippocampus regions of aged rats (Group III). Activities of antioxidant enzymes like superoxide dismutase, catalase, glutathione peroxidase and levels of non-enzymic antioxidants like reduced glutathione, Vitamin C and Vitamin E were found to be significantly decreased in all the brain regions studied in aged rats when compared to young rats. However, normalized lipid peroxidation and antioxidant defenses were reported in the grape seed extract-supplemented aged rats. These findings demonstrated that grape seed extract enhanced the antioxidant status and decreased the incidence of free radical-induced lipid peroxidation in the central nervous system of aged rats.

The free radical theory of cell aging may have significant relevance in the pathogenesis of a number of age-related neurological disorders. A large body of experimental evidence supports the existence of a relationship between genomic instability, DNA damage and aging. The age-associated accumulation of oxidative DNA damage is well documented in central nervous system. The decline of mitochondrial respiratory function and loss of normal cellular homeostasis as consequences of excessive accumulation of endogenous oxidative damages to DNA have long been indicated in the aging process. In the present study, age-associated alterations in the content of DNA and accumulation of oxidative DNA damage products such as 8-OHdG and DNA protein cross-links are mainly focused. In parallel, we have also investigated the salubrious effect of l-carnitine against oxidative DNA damage as it possesses energy and antioxidant improving properties. Our results thus reveal that L-carnitine has inhibiting effect on the accumulation of age-related oxidative DNA damage in various brain regions, viz. cerebral cortex, striatum and hippocampus.

Mitochondria link the energy -- releasing activities of electron transport and proton pumping with the energy conserving process of oxidative phosphorylation to form ATP. A declined mitochondrial performance has been generally observed during aging. In the present investigation, the activities of tricarboxylic acid cycle enzymes such as isocitrate, alpha-ketoglutarate, succinate and malate dehydrogenases and electron transport complexes I-IV were measured in mitochondria isolated from brain regions like cortex, striatum and hippocampus of young and aged rats before and after L-Carnitine supplementation. All the three brain regions of aged rats showed decreased activities of isocitrate, alpha-ketoglutarate and succinate dehydrogenases, complexes I and IV when compared to control young rats. Striatum seems to be the most susceptible region when compared to hippocampus and cortex. L-Carnitine supplementation to aged rats reversed the activities of these enzymes to near normal whereas treatment to young rats did not show any significant alterations. These results confirm that L-Carnitine can alleviate the age-associated decline in the metabolic efficiency of mitochondria in all three brain regions under investigation.
