Abstract

Optical properties of polymer capped gold nanoparticles of various sizes (diameter 3-6 nm) have been studied. We present a new scheme to extract size dependent variation of total dielectric function of gold nanoparticles from measured UV-Vis absorption data. The new scheme can also be used, in principle, for other related systems as well. We show how quantum effect, surface atomic co - ordination and polymer - nanoparticle interface morphology leads to a systematic variation in inter band part of the dielectric function of gold nanoparticles, obtained from the analysis using our new scheme. Careful analysis enables identification of the possible changes to the electronic band structure in such nanoparticles., Comment: 13 pages,7 figures, 1 table

22. Insulin Signalling: Essential Role of a 222 Da Molecular Mediator, Co-Insulin (Co-Ins)

Abstract

An attempt has been made to probe into the molecular basis of insulin resistance. The target was to look into the existence of a molecule that could interfere with insulin receptor-mediated molecular signaling. Human plasma was chromatographed on an affinity column of insulin receptor (IR) Sepharose, and the IR bound fraction was collected and subjected to analysis. In the process, what was revealed was a small molecule that bound to insulin and not to insulin receptor. This 222 Da molecule has been designated as Co-Insulin (Co-Ins). Its structural identity is 3-(2-amino-5-(aminomethyl)-2-hydroxy-1,3-dioxolan-4-yl)-2,3-dihydroxypropanal. Molecular modeling studies show that Co-Ins binds to a specified site on the A-chain of insulin. An interaction between the A and B chains of insulin ensues, leading to the binding of insulin B-chain with the α-subunit of IR. The ensuing interaction between IRα and IRβ leads to auto-tyrosine phosphorylation of IRβ and tyrosine phosphorylation of the IRS family consisting of at least four proteins. From this point onwards, insulin action proceeds in two directions; ‘Immediate’ and ‘Delayed.’ ‘Immediate’ is represented by the hypoglycemic response, already known to be mediated by Glut-4, apparently influenced by the binding of one of the 4 tyrosine-phosphorylated IRS molecules. ‘Delayed’ is represented by the hitherto unknown nuclear entry of the tyrosine-phosphorylated 43 kDa and 49 kDa fragments of IRβ, mediated by yet another tyrosine-phosphorylated IRS. Indirect evidence indicates that this IRS recognizes nuclear localization signals on the IRβ fragments. The striking observation made in these studies is that Co-Ins has no independent role to perform other than binding to its specified site on insulin and initiating the signaling events. The study opens up a hitherto unrecognized question: are there similar small molecules that mediate the actions of other peptide hormones in the mammalian system?

33. COVID-19 Omicron variant-inhibitory in silico evidence for phytocompounds of an ancient Ayurvedic formulation

Abstract

The emerging SARS-COV-2 variants and the global COVID-19 outbreak underscore the urgent need for prophylactics and improved antivirals. Scientists are racing to develop vaccines and antiviral drugs. While vaccinations will be the primary pandemic prophylactic and, appropriate antiviral medications will be needed as a treatment strategy. The variant strains of the virus spawned the second and third waves in several countries, and it affected universally. Antiviral drugs with novel mechanisms of action are still needed to combat emerging SARS-CoV-2 mutations. The present research confirmed in silico the prospective efficacy of an ancient Ayurvedic formulation made for prophylactic and treatment material against symptoms similar to the COVID-19 Wuhan_Hu_1st strain, which had been provided with similar in silico evidence in our earlier report. Clinical studies with safe chemical cocktails may have prospects for countries where the practice of Ayurveda is not legal.

Abstract

Head and neck malignancies have always been challenging for the clinician, both with regards to locoregional control and distant metastasis. Aggressive approaches translate to an acceptable locoregional control, but distant failures pose a dilemma. Newer, sophisticated, imaging modalities have helped in early diagnosis of solitary metastasis, and in turn have opened up an array of interventional procedures, which to some extent improve the disease-free survival and quality of life, as was seen in the present case of locoregionally controlled advanced hypopharyngeal malignancy who presented with solitary distant metastasis. Still, diligent care needs to be taken not to aggravate the scenario with these interventions.
