We have studied the alpha decays of 214Po into 210Pb and of 212Po into 208Pb tagged by the coincidence with the preceding beta decays from 214Bi and 212Bi, respectively. The employed 222Rn, 232Th, and 220Rn sources were sealed inside quartz vials and inserted in the Counting Test Facility at the underground Gran Sasso National Laboratory in Italy. We find that the mean lifetime of 214Po is (236.00 +- 0.42(stat) +- 0.15(syst)) \mu s and that of 212Po is (425.1 +- 0.9(stat) +- 1.2(syst)) ns. Our results, obtained from data with signal-to-background ratio larger than 1000, reduce the overall uncertainties and are compatible with previous measurements., Comment: RevTex, 11 pages, 5 figures, 3 tables. Borexino was the first experiment to detect solar neutrinos in real-time in the sub-MeV region. In order to achieve high precision in the determination of neutrino rates, the detector design includes an internal and an external calibration system. This paper describes both calibration systems and the calibration campaigns that were carried out in the period between 2008 and 2011. We discuss some of the results and show that the calibration procedures preserved the radiopurity of the scintillator. The calibrations provided a detailed understanding of the detector response and led to a significant reduction of the systematic uncertainties in the Borexino measurements.

Borexino is a large-volume liquid scintillator detector installed in the underground halls of the Laboratori Nazionali del Gran Sasso in Italy. After several years of construction, data taking started in May 2007. The Borexino phase I ended after about three years of data taking. Borexino provided the first real time measurement of the $^{7}$Be solar neutrino interaction rate with accuracy better than 5% and confirmed the absence of its day-night asymmetry with 1.4% precision. This latter Borexino results alone rejects the LOW region of solar neutrino oscillation parameters at more than 8.5 $\sigma$ C.L. Combined with the other solar neutrino data, Borexino measurements isolate the MSW-LMA solution of neutrino oscillations without assuming CPT invariance in the neutrino sector. Borexino has also directly observed solar neutrinos in the 1.0-1.5 MeV energy range, leading to the first direct evidence of the $pep$ solar neutrino signal and the strongest constraint of the CNO solar neutrino flux up to date. We have measured the muon flux at the underground Gran Sasso National Laboratory (3800 m w.e.) to be (3.41 \pm 0.01) \times 10-4m-2s-1 using four years of Borexino data. A modulation of this signal is observed with a period of (366\pm3) days and a relative amplitude of (1.29 \pm 0.07)%. The measured phase is (179 \pm 6) days, corresponding to a maximum on the 28th of June. Using the most complete atmospheric data models available, muon rate fluctuations are shown to be positively correlated with atmospheric temperature, with an effective coefficient {\alpha}T = 0.93 \pm 0.04. Borexino, a liquid scintillator detector at LNGS, is designed for the detection of neutrinos and antineutrinos from the Sun, supernovae, nuclear reactors, and the Earth. The feeble nature of these signals requires a strong suppression of backgrounds below a few MeV. Very low intrinsic radiogenic contamination of all detector components needs to be accompanied by the efficient identification of muons and of muon-induced backgrounds. Muons produce unstable nuclei by spallation processes along their trajectory through the detector whose decays can mimic the expected signals; for isotopes with half-lives longer than a few seconds, the dead time induced by a muon-related veto becomes unacceptably long, unless its application can be restricted to a sub-volume along the muon track. Consequently, not only the identification of muons with very high efficiency but also a precise reconstruction of their tracks is of primary importance for the physics program of the experiment. The Borexino inner detector is surrounded by an outer water-Cherenkov detector that plays a fundamental role in accomplishing this task. The detector design principles and their implementation are described. The strategies adopted to identify muons are reviewed and their efficiency is evaluated. The overall muon veto efficiency is found to be 99.992% or better. Ad-hoc track reconstruction algorithms developed are presented. Their performance is tested against muon events of known direction such as those from the CNGS neutrino beam, test tracks available from a dedicated External Muon Tracker and cosmic muons whose angular distribution reflects the local overburden profile. The achieved angular resolution is 3-5 deg and the lateral resolution is 35-50 cm, depending on the impact parameter of the crossing muon. Background: Wiping is an effective and established method for decontaminating surfaces, it involves applying liquid while simultaneously removing soils. Chlorine dioxide is a high-level disinfectant that can be applied to surfaces, including those of medical devices, with the aid of mechanical action or wiping. The European Standard (EN) 16615 evaluates the microbiocidal efficacy of a chemical disinfectant when it is applied with mechanical action, i.e., when applied by wiping. It has been suggested that manual methods employing human intervention can lead to variations in a validated process. One such assumption is that variable wiping force that exists between users may influence the effectiveness of the process. Method: The average wiping force of individuals with varying physical characteristics was investigated. A bespoke testing apparatus was employed in a series of EN 16615 four-field tests, with the aim to investigate if variable wiping force between individuals impacts the microbiocidal efficacy of a chlorine dioxide-based wipe disinfectant if tested as applied as in practice. Results: Individuals applied variable forces when wiping. Physical attributes such as hand dominance and body build were shown to influence the force applied. The average wiping force calculated from this pool of participants was found to be lower than the stated range given by the EN 16615. The pre-impregnated chlorine dioxide-based wipe achieved biocidal efficacy in the series of EN 16615 tests. No trend relating to the efficacy of the product and the different forces applied could be drawn. Conclusion: The data demonstrates that individuals apply different forces to a surface when wiping. However, the variability expressed when wiping has no impact on the microbiocidal efficacy of a manually applied chlorine dioxide-based disinfectant. [ABSTRACT FROM AUTHOR]

11. Langmuir Wave Generation Through A Neutrino Beam Instability

A standard version of a kinetic instability for the generation of Langmuir waves by a beam of electrons is adapted to describe the analogous instability due to a beam of neutrinos. The interaction between a Langmuir wave and a neutrino is treated in the one-loop approximation to lowest order in an expansion in $1/M_W^2$ in the standard electroweak model. It is shown that this kinetic instability is far too weak to occur in a suggested application to the reheating of the plasma behind a stalled shock in a type II supernova (SN). This theory is also used to test the validity of a previous analysis of a reactive neutrino beam instability and various shortcomings of this theory are noted. In particular, it is noted that relativistic plasma effects have a significant effect on the calculated growth rates, and that any theoretical description of neutrino-plasma interactions must be based directly on the electroweak theory. The basic scalings discussed in this paper suggest that a more complete investigation of neutrino-plasma processes should be undertaken to look for an efficient process capable of driving the stalled shock of a type II SN., Comment: 23 pages, incl. 5 postscript figures

A role for dynamin in clathrin-mediated endocytosis is now well established. However, mammals express three closely related, tissue-specific dynamin isoforms, each with multiple splice variants. Thus, an important question is whether these isoforms and splice variants function in vesicle formation from distinct intracellular organelles. There are conflicting data as to a role for dynamin-2 in vesicle budding from the TGN. To resolve this issue, we compared the effects of overexpression of dominant-negative mutants of dynamin-1 (the neuronal isoform) and dynamin-2 (the ubiquitously expressed isoform) on endocytic and biosynthetic membrane trafficking in HeLa cells and polarized MDCK cells. Both dyn1(K44A) and dyn2(K44A) were potent inhibitors of receptor-mediated endocytosis; however neither mutant directly affected other membrane trafficking events, including transport mediated by four distinct classes of vesicles budding from the TGN. Dyn2(K44A) more potently inhibited receptor-mediated endocytosis than dyn1(K44A) in HeLa cells and at the basolateral surface of MDCK cells. In contrast, dyn1(K44A) more potently inhibited endocytosis at the apical surface of MDCK cells. The two dynamin isoforms have redundant functions in endocytic vesicle formation, but can be targeted to and function differentially at subdomains of the plasma membrane.

Mitosis in human cells is initiated by the protein kinase Cdc2-cyclin B1, which is activated at the end of G2 by dephosphorylation of two inhibitory residues, Thr14 and Tyr15. The G2 arrest that occurs after DNA damage is due in part to stabilization of phosphorylation at these sites. We explored the possibility that entry into mitosis is also regulated by the subcellular location of Cdc2-cyclin B1, which is suddenly imported into the nucleus at the end of G2. We measured the timing of mitosis in HeLa cells expressing a constitutively nuclear cyclin B1 mutant. Parallel studies were performed with cells expressing Cdc2AF, a Cdc2 mutant that cannot be phosphorylated at inhibitory sites. Whereas nuclear cyclin B1 and Cdc2AF each had little effect under normal growth conditions, together they induced a striking premature mitotic phenotype. Nuclear targeting of cyclin B1 was particularly effective in cells arrested in G2 by DNA damage, where it greatly reduced the damage-induced G2 arrest. Expression of nuclear cyclin B1 and Cdc2AF also resulted in significant defects in the exit from mitosis. Thus, nuclear targeting of cyclin B1 and dephosphorylation of Cdc2 both contribute to the control of mitotic entry and exit in human cells.

Background: Antipsychotics are perceived to be over-used in the management of behavioural problems in people with an intellectual disability (ID). Published guidelines have set good practice standards for the use of these drugs for behavioural indications. We sought to identify the range of indications for which antipsychotic drugs are prescribed in people with ID and to audit clinical practice against the standards. Method: Data were collected from the clinical records of individuals with ID who were under the care of mental health services in the UK, and prescribed an antipsychotic drug. Results: The sample comprised 2319 patients from 39 clinical services. Twenty-seven per cent of the patients had a diagnosis of a psychotic illness (ICD-10 F20-29) and 27% an affective illness (ICD-10 F30-39). The proportion who did not have a psychiatric diagnosis ranged from 6% of those with borderline/mild ID to 21% of those with severe/profound ID. Overall, the most common indications for prescribing an antipsychotic drug were comorbid psychotic illness, anxiety and agitation, and a range of behavioural disturbances. The prevalence of use of antipsychotic drugs to manage challenging behaviour in the absence of concomitant mental illness increased with the severity of ID and accounted for almost half of prescriptions in those with severe/profound ID. Adherence to the audit standards related to documentation of clinical indications and review of efficacy was high. Side effect monitoring was less assiduous. Conclusions: In clinical practice, most prescriptions for antipsychotic drugs in people with ID are consistent with the evidence base and the overall quality of prescribing practice, as measured against recognised standards, is good, although in some patients potentially remedial side effects may not be detected and treated.

- Abstract
Objective: To document the recovery of vascular services in Europe following the first COVID-19 pandemic peak. Methods: An online structured vascular service survey with repeated data entry between 23 March and 9 August 2020 was carried out. Unit level data were collected using repeated questionnaires addressing modifications to vascular services during the first peak (March – May 2020, “period 1”), and then again between May and June (“period 2”) and June and July 2020 (“period 3”). The duration of each period was similar. From 2 June, as reductions in cases began to be reported, centres were first asked if they were in a region still affected by rising cases, or if they had passed the peak of the first wave. These centres were asked additional questions about adaptations made to their standard pathways to permit elective surgery to resume. Results: The impact of the pandemic continued to be felt well after countries’ first peak was thought to have passed in 2020. Aneurysm screening had not returned to normal in 21.7% of centres. Carotid surgery was still offered on a case by case basis in 33.8% of centres, and only 52.9% of centres had returned to their normal aneurysm threshold for surgery. Half of centres (49.4%) believed their management of lower limb ischaemia continued to be negatively affected by the pandemic. Reduced operating theatre capacity continued in 45.5% of centres. Twenty per cent of responding centres documented a backlog of at least 20 aortic repairs. At least one negative swab and 14 days of isolation were the most common strategies used for permitting safe elective surgery to recommence. Conclusion: Centres reported a broad return of services approaching pre-pandemic “normal” by July 2020. Many introduced protocols to manage peri-operative COVID-19 risk. Backlogs in cases were reported for all major vascular surgeries.

- Abstract
Background Although patients with Alzheimer’s disease and other cognitive-related neurodegenerative disorders may benefit from early detection, development of a reliable diagnostic test has remained elusive. The penetration of digital voice-recording technologies and multiple cognitive processes deployed when constructing spoken responses might offer an opportunity to predict cognitive status. Objective To determine whether cognitive status might be predicted from voice recordings of neuropsychological testing Design Comparison of acoustic and (para)linguistic variables from low-quality automated transcriptions of neuropsychological testing (n = 200) versus variables from high-quality manual transcriptions (n = 127). We trained a logistic regression classifier to predict cognitive status, which was tested against actual diagnoses. Setting Observational cohort study. Participants 146 participants in the Framingham Heart Study. Measurements Acoustic and either paralinguistic variables (e.g., speaking time) from automated transcriptions or linguistic variables (e.g., phrase complexity) from manual transcriptions. Results Models based on demographic features alone were not robust (area under the receiver-operator characteristic curve [AUROC] 0.60). Addition of clinical and standard acoustic features boosted the AUROC to 0.81. Additional inclusion of transcription-related features yielded an AUROC of 0.90. Conclusions The use of voice-based digital biomarkers derived from automated processing methods, combined with standard patient screening, might constitute a scalable way to enable early detection of dementia.

Background Fibrosis is a hallmark of heart disease, particularly following myocardial infarction (MI) and in heart failure. We previously identified a key role for extracellular matrix protein (ECM1) in wound healing post-MI but the cellular origin and mechanism of ECM1 remained elusive. Here, we investigate the spatiotemporal cellular origin of ECM1 in healthy and diseased human and mouse hearts, and ECM1 dependent human cardiac fibroblast (HuCFb) signalling mechanisms. Methods ECM1 specific analysis of existing single-cell/-nuclei RNA sequencing data was conducted. ECM1 expression was assessed in non-failing (NF), ischaemic (ICM) and dilated (DCM) failing human heart tissue via immunoblotting (n=8), immunohistochemistry (ICM; n=8, NF; n=4) and mRNA in-situ hybridisation (n=3). HuCFbs were treated with recombinant ECM1 and assessed via phosphoproteomics (n=6), wound healing assay (n=4), MTT assay (n=8), qPCR (n=6). Results ECM1 originates from fibroblasts, macrophages/monocytes (MΦ/Mo), and pericytes/vascular cells in uninjured human and mouse hearts. In mouse hearts post-MI, ECM1 originates predominantly from M1MΦ/Mo at day-3, and myofibroblasts and M2MΦ’s at day-7, and expression correlated with cell-cell communication, collagen organisation, inflammation, adhesion, and migration. Fittingly, ECM1 expression was upregulated in human ICM (p=0.048) and DCM (p=0.027), localised interstitially to fibrotic, inflammatory, and peri-vascular areas. Variability of rainfall in East Africa has major impacts on lives and livelihoods. From floods to droughts, this variability is important on short daily time‐scales to longer decadal time‐scales, as is apparent from the devastating effects of droughts in East Africa over recent decades. Past studies have highlighted the Congo airmass in enhancing East African rainfall. Our detailed analysis of the feature shows that days with a westerly moisture flow, bringing the Congo airmass, enhance rainfall by up to 100% above the daily mean, depending on the time of year. Conversely, there is a suppression of rainfall on days with a strong easterly flow. Days with a westerly moisture flux are in a minority in all seasons but we show that long rains with more westerly days are wetter, and that during the most‐recent decade which has had more frequent droughts (associated with the “Eastern African climate paradox”), there has been few days with such westerlies. We also investigate the influence of the Madden–Julian Oscillation (MJO) and tropical cyclones, and their interaction with the westerly flow. We show that days of westerly moisture flux are more likely during phases 3 and 4 of the MJO and when there are one or more tropical cyclones present. In addition, tropical cyclones are more likely to form during these phases of the MJO, and more likely to be coincident with westerlies when forming to the east of Madagascar. Overall, our analysis brings together many different processes that have been discussed in the literature but not yet considered in complete combination. The results demonstrate the importance of the Congo airmass on daily to climate time‐scales, and in doing so offers useful angles of investigation for future studies into prediction of East African rainfall.
