Abstract

Ossification of the posterior longitudinal ligament of the spine (OPLL) has been mapped to a predisposing locus on chromosome 6p close to the HLA complex. A genetic-linkage study has been carried out in 91 affected sib pairs from 53 Japanese families. OPPLL is a common disorder among Japanese people and all over Asia.

Abstract

The crude PG fraction extracted under dissociative conditions was subjected to DEAE-Sephacel ion-exchange chromatography, gel-filtration on Sepharose CL-4B, and Octyl-Sepharose CL-4B chromatography, and decorin was obtained. The amino-terminal amino acid sequence of decorin was found to be identical to those of human bone PG-II and human fetal membrane PG-II. The analyses of the GAG chains using two-dimensional electrophoresis on cellulose acetate membranes, affinity high-performance liquid chromatography (HPLC) with a hydroxyapatite column and gel-filtration HPLC after chondroitinase digestion, showed the presence of dermatan sulfate (DS) chains and chondroitin sulfate (CS). Abstract

Three types of proteoglycans (PG-I, II and III) present in the human yellow ligament extracellular matrix were separated using a combination of CsCl isopycnic density gradient centrifugation, DEAE-Sephacel ion-exchange chromatography, gel-filtration on Sepharose CL-4B and CL-2B, and Octyl-Sepharose CL-4B chromatography. Proteoglycan-I was a high-molecular-weight proteoglycan, which was excluded by Sepharose CL-4B, and its molecular weight, estimated using sodium dedecyl sulfate-polyacrylamide gel electrophoresis, was over 350,000. The glycosaminoglycan chain of PG-I was composed of chondroitin 6-sulfate, and PG-I appeared to be an aggrecan-type proteoglycan. Proteoglycan-II and III were low-molecular-weight proteoglycans with molecular weights of 190,000 and 105,000, respectively. The core protein sizes of PG-II and III were similar, 44,000, but anti-human decorin antibody reacted with PG-III, not with PG-II. The glycosaminoglycan chain of PG-II contained dermatan sulfate and chondroitin 6-sulfate, whereas that of PG-III contained the former only. In the light of these results, PG-II and III were suggested to be biglycan- and decorin-type proteoglycans, respectively. The major proteoglycan in the human yellow ligament extracellular matrix was PG-III.

Abstract

One type of large proteoglycan and three types of small proteoglycans (decorin, decorin-subtype, and biglycan) were purified by chromatography, and a -elastin was isolated by alkali treatment from human yellow ligaments taken at the time of operation. The interaction of the proteoglycans with immobilized a -elastin on a sensor was analyzed by surface plasmon resonance, and we confirmed that each of the small proteoglycans exhibited a specific binding to a -elastin. The binding sites of small proteoglycans were contained in the protein cores. In addition, the differences in the interaction of the small proteoglycans with a -elastin of normal and ossified ligaments were compared. The interactions of the small proteoglycans with a -elastin of the ossified ligaments were lower than those with a -elastin of the normal ligaments. Abstract

The extent and distribution of linkage disequilibrium (LD) in humans is a current topic especially for gene mapping of complex diseases. Akaike's information criterion (AIC) was applied to estimate LD and compared with other standard LD measures, D' and r². By comparison of an independent model (IM; linkage equilibrium) and a dependent model (DM; linkage disequilibrium), the parsimonious model is the one with the smaller AIC score. Therefore, the extent of LD by AIC is expressed as AIC(IM) — AIC(DM)(AIC(LD)). A total of 39 singlenucleotide polymorphisms on a 1.6-Mb region of chromosome 21q22 were identified, and genotyped in 192 Japanese individuals. All possible pairs were analyzed to estimate LD and the analyses were compared. AIC(LD) became highly positive as the D' value increased and was negative at D' values of around 0.2. Because a negative value of AIC(LD) implies linkage equilibrium, D' values below 0.2 should be regarded as linkage equilibrium. The LD estimate by AIC yielded results similar to those obtained by r², indicating that AIC(LD) would be useful for fine gene mapping. [ABSTRACT FROM AUTHOR]

46. Spinous ProcessSplitting Laminoplasty Using Hydroxyapatite Spinous Process Spacer

Abstract

Between February 1986 and August 1989, 45 patients with cervical myelopathy were treated by spinous process-splitting laminoplasty. Hydroxyapatite intraspinous spacers were used to maintain the enlargement of the cervical spinal canal. The shape of this spacer is trapezoidal. After sagittal splitting of the spinous process, the spacer was inserted between the two haives and affixed with the wire. Histologic study showed there was good fusion between the spacer and bone. In all cases, good enlargement of the cervical spinal canal was achieved. Spacer displacement, wire breakage, and postoperative infection were not seen. There was no postoperative neurologic deterioration. Computed tomography showed that the width of the cervical spinal canal was maintained.
