Despite high cure rates in pediatric patients with acute leukemia, development of resistance limits the efficacy of antileukemic therapy.Tris(1,10-phenanthroline)tris(thiocyanato-κN)lanthanum(III) (KP772) is an experimental antineoplastic agent to which multidrug-resistant cell models have shown hypersensitivity. Antiproliferative and apoptotic activities of KP772 were tested in leukemia, lymphoma and solid tumor cell lines as well as primary leukemia cells (isolated from the bone marrow of a child with acute myeloid leukemia (AML). The ability to overcome drug resistances was investigated in doxorubicin- and vincristine-resistant cell lines. Real-time PCR was used to gain insight into the mechanism of apoptosis induction. KP772 inhibited proliferation and induced apoptosis in various leukemia and lymphoma cell lines in a concentration-dependent manner (LC50 = 1–2.5 µM). Primary AML cells were also sensitive to KP772, whereas daunorubicin showed no significant effect. KP772 induces apoptosis independently of Bcl-2, Smac, and the CD95 receptor and is also effective in caspase 3-deficient MCF7 cells, indicating that apoptosis is partly triggered independently of caspase 3. mRNA expression profiling revealed an upregulation of the BH3-only Bcl-2 protein Harakiri in the course of KP772-induced apoptosis. Remarkably, KP772 overcame drug resistance to doxorubicin and vincristine in vitro, and the apoptotic effect in resistant cells was even superior to that in non-resistant parental cells. In combination with vincristine, doxorubicin and cytarabine, synergistic effects were observed in BJAB cells. The cytotoxic potency in vitro/ex vivo and the remarkable ability to overcome multidrug resistance propose KP772 as a promising candidate drug for antileukemic therapy, especially of drug-refractory malignancies.Graphic abstract

The neuronal bHLH transcription factors Neurod2 and Neurod6 are predominantly expressed in postmitotic pyramidal neurons of the developing neocortex and have previ- ously been shown to promote pyramidal neuron survival, differentiation, migration, and cortical connectivity. The closely related factor Neurod1 is normally only transiently expressed by intermediate precursor cells of the subventricular zone. In Neurod2/6 double-deficient mice, however, Neurod1-expression is ectopically maintained in post- mitotic pyramidal neurons of the cortical plate, thereby partially compensating for the loss of Neurod2/6. In conditional Neurod1/2/6 triple-deficient mice selectively lacking the ectopic up-regulation of Neurod1, neuronal survival, differentiation, migration, and connectivity are significantly stronger disturbed. I aimed to further understand the functions of NeuroD-family bHLH proteins in cortex development, with a mechanistic focus on the maintenance of pyramidal neuron survival and the establishment of different pyramidal neuron identities during earlier cortex development. In Neurod1/2/6-deficient mice, I identified an unprecedentedly strong wave of apop- totic cell death that was restricted temporarily to E14/15 and spatially to the dorso- medial cortex. Probably as a consequence, Sox5-positive pyramidal neurons of the neocortical layer 6 are lost, and the Tbr2-positive progenitor pool in the subventricular and intermediate zones is expanded. Remaining pyramidal neurons show immature molecular identities and mostly miss axonal connectivity at birth. Artificial expression of Neurod2 or Neurod6 in a subset of postmitotic pyramidal neurons was sufficient to overcome those structural defects, showing that the underlying mechanisms are of cell- intrinsic nature. Initially following a candidate-based approach, I investigated known inhibitors of apoptosis, neurotrophin signaling, and cell cycle control, but I could not identify a mechanism explaining the strong phenotype in Neurod1/2/6-deficient mice. To gain further mechanistic insight, I performed an unbiased comparable transcrip- tome analysis of the central neocortex at E13, just before the manifestation of obvious structural abnormalities in Neurod1/2/6-deficient mice. The most interesting finding was a nearly 4-fold up-regulation of Harakiri (Hrk), a small BH3-only pro-apoptotic protein of the Bcl2-family. In-situ hybridization confirmed robustly increased Hrk- expression at E13–E16. In-utero electroporation of Hrk into the developing cortex of wild-type mice was sufficient to kill cortical neurons. Electroporation of Hrk-shRNA into Neurod1/2/6-deficient mice prevented apoptosis of electroporated cells. As NeuroD-proteins are transcriptional activators, I postulated a transcriptional repressor downstream of Neurod1/2/6 to normally prevent Hrk-triggered apoptosis. The strongest down-regulated genes in Neurod1/2/6-deficient mice were Bhlhb5 and Prdm8, which are known to interact and form a repressors complex in cortical neurons. Electroporation of Bhlhb5 and Prdm8 into the cerebral cortex of Neurod1/2/6- deficient mice could rescue apoptosis, confirming their role in the control of cell death downstream of Neurod1/2/6., Die neuronalen bHLH-Transkriptionsfaktoren Neurod2 und Neurod6 werden vor- wiegend in postmitotischen pyramidalen Neuronen des sich entwickelnden Neokortex exprimiert und haben bereits gezeigt, dass sie das Überleben, die Differenzierung, die Migration und die kortikale Konnektivität pyramidaler Neuronen fördern. Der eng ver- wandte Faktor Neurod1 wird normalerweise nur vorübergehend von intermediären Vor- läuferzellen in der subventrikulären Zone exprimiert. In Neurod2/6-doppeldeffizienten Mäusen wird die Neurod1-Expression jedoch ektopisch in postmitotischen Pyramiden- neuronen der kortikalen Platte aufrechterhalten, wodurch der Verlust von Neurod2/6 teilweise ausgeglichen wird. In konditionalen Neurod1/2/6-Dreifachmäusen, denen selektiv die ektopische Hochregulierung von Neurod1 fehlt, sind das neuronale Über- leben, die Differenzierung, die Migration und die Konnektivität deutlich stärker gestört. Mein Ziel war es, die Funktionen der bHLH-Proteine der NeuroD-Familie in der Kortex- entwicklung zu verstehen, wobei ich mich mechanistisch auf die Aufrechterhaltung des Überlebens von Pyramidenneuronen und die Etablierung verschiedener Identitäten von Pyramidenneuronen während der frühen Kortexentwicklung konzentrierte. Bei Neurod1/2/6-defizienten Mäusen konnte ich eine beispiellos starke Welle des apoptotischen Zelltods feststellen, die zeitlich auf E14/15 und räumlich auf den dorso- medialen Kortex beschränkt war. Wahrscheinlich als Folge davon gehen Sox5-positive Pyramidenneuronen der neokortikalen Schicht 6 verloren, und der Tbr2-positive Vorläuferpool in den subventrikulären und intermediären Zonen ist vermehrt. Die molekulare Identität der verbleibenden Pyramidenneuronen ist unreif und ihnen fehlt bei der Geburt meist die axonale Konnektivität. Die künstliche Expression von Neurod2 oder Neurod6 in einer Untergruppe postmitotischer Pyramidenneuronen reichte aus, um diese strukturellen Defekte zu beheben, was zeigt, dass die zugrunde liegenden Mechanismen zelleigener Natur sind. Zunächst untersuchte ich im Rahmen eines kandidatenbasierten Ansatzes bekannte Inhibitoren der Apoptose, der Neurotrophin- Signalübertragung und der Zellzykluskontrolle, konnte aber keinen Mechanismus identifizieren, der den starken Phänotyp in Neurod1/2/6-defizienten Mäusen erklärt. Um weitere Einblicke in die Mechanismen zu gewinnen, führte ich eine unvorein- genommene, vergleichbare Transkriptomanalyse des zentralen Neokortex bei E13 durch, also kurz vor dem Auftreten offensichtlicher struktureller Anomalien bei Neurod1/2/6-defizienten Mäusen. Der interessanteste Befund war eine fast 4-fache Hochregulierung von Harakiri (Hrk), einem kleinen, nur BH3 enthaltenden pro- apoptotischen Protein der Bcl2-Familie. Die In-situ-Hybridisierung bestätigte eine stark erhöhte Hrk-Expression bei E13-E16. Die inutero-Elektroporation von Hrk in den sich entwickelnden Kortex von Wildtyp-Mäusen war ausreichend, um kortikale Neuronen abzutöten. Die Elektroporation von Hrk-shRNA in Neurod1/2/6-defizienten Mäusen verhinderte die Apoptose der elektroporierten Zellen. Da NeuroD-Proteine Transkriptionsaktivatoren sind, postulierte ich einen Tran- skriptionsrepressor stromabwärts von Neurod1/2/6, der normalerweise die durch Hrk ausgelöste Apoptose verhindert. Die am stärksten herunterregulierten Gene in Neurod1/2/6-defizienten Mäusen waren Bhlhb5 und Prdm8, von denen bekannt ist, dass sie interagieren und einen Repressor-Komplex in kortikalen Neuronen bilden. Die Elektroporation von Bhlhb5 und Prdm8 in die Großhirnrinde von Neurod1/2/6- defizienten Mäusen konnte die Apoptose retten, was ihre Rolle bei der Kontrolle des Zelltods stromabwärts von Neurod1/2/6 bestätigt.

La lengua española actual cuenta con una locución verbal nacida a principios del siglo xx (hacerse el jaraquiri), cuyo componente nominal es una palabra de origen japonés, jaraquiri. El presente artículo pretende realizar, por primera vez en nuestro idioma, un estudio exhaustivo sobre esta construcción, propia de la lengua española y de otras lenguas próximas: sus características morfológicas, combinatorias, de uso, dialectales e históricas The current Spanish language has a verbal phrase born at the beginning of the xx century (hacerse el jaraquiri), whose nominal component is a Japanese loanword, jaraquiri. This article intends to carry out, for the first time in our language, an exhaustive study on this construction, which is not only typical of Spanish, but also of other languages: its morphology, its combinatory, its use, and its dialectal and historical characteristics.

Розглянуто дві камерні опери угорського композитора Петера Етвеша, «Харакірі» та «Радамес», які представляють ранню оперну творчість автора. Здійснено інтонаційно-драматургічний аналіз обох театральних творів та проаналізовано умови та історію їх створення. З’ясовано вплив різновидів японського традиційного театру, з якими угорський композитор ознайомився під час поїздки в Японію: но, бунраку, кабукі. Досліджено особливості використання японських традицій на різних рівнях в обох творах. У камерній опері «Харакірі» виявлено не лише японську традиційну ритуальну тематику, а й застосування характерного японського вокалу та використання традиційного духового інструменту – сакухачі. Натомість, у камерній опері «Радамес» відзначено елементи японського лялькового театру бунраку та його застосування не на тематичному, а на технічному рівні, згідно з яким одним актором керують троє режисерів. Крім японських традицій, виявлено також вплив театру абсурду. У результаті аналізу музики, розгляду умов створення, ознайомлення з літературними першоджерелами та композиційними методами створення даних камерних опер з’ясовано їх вагомий вплив на створення подальших, вже повномасштабних театральних композицій Петера Етвеша. У статті також розглянуто варіанти сучасних інтерпретацій проаналізованих композицій., In the article two chamber operas of the Hungarian composer, Peter Eötvös, «Harakiri» and «Radames» are considered. They represent the early period of the operatic creativity of the author. The conditions and the history of creation the operas, as well as the intonational-dramaturgic analysis of the theatrical words were made. The influence of varieties of the Japanese traditional theater, with which the composer was familiarized during his trip to Japan, was clarified: noh, bunraku, kabuki. The use of Japanese traditions at different levels was revealed in both cases. The chamber opera "Harakiri" revealed not only Japanese traditional ritual themes, but also the use of characteristic Japanese vocals and traditional wind instruments - shakuhachi. In the case of the chamber opera «Radames», the use of the Japanese puppet theater, the bunraku, was revealed, not at the thematic, but at the technical level, which is primarily manifested in the reception of the management of one actor by three directors at once. Thus, the influence of the theater of the absurd on the given theatrical work of the Hungarian composer was also revealed. As a result of the analysis of music, writing process conditions, literary sources, as well as compositional methods concerning these two chamber operas, their significance in creating the following already full-scale theatrical compositions by the Hungarian composer, Peter Eötvös, was clarified. The article also considers variants of modern interpretations of the analyzed compositions., Рассмотрены две камерные оперы венгерского композитора Петера Этвеша – «Харакири» и «Радамес», которые представляют ранний период оперного творчества автора. Осуществлен интонационно-драматургический анализ данных театральных произведений, а также проанализированы условия и история их написания. Выяснено влияние разновидностей японского традиционного театра, с которым композитор ознакомился во время поездки в Японию: но, бунраку, кабуки. Выявлено использование японских традиций на разных уровнях в обеих случаях. В камерной опере «Харакири» выявлено не только японскую традиционную ритуальную тематику, но также использование характерного японского вокала и традиционного духового инструмента – сакухачи. В случае камерной оперы «Радамес» отмечено использование японского кукольного театра, бунраку, не на тематическом, а на техническом уровне, что прежде всего проявляется в приеме управления одним актером сразу тремя режиссерами. Таким образом, выявлено также влияние театра абсурда на данное театральное произведение венгерского композитора. В результате анализа музыки, условий написания, литературных первоисточников, а также композиционных методов относительно данных двух камерных опер выяснено их значение в создании следующих, уже полномасштабных театральных композиций Петера Этвеша. В статье также рассмотрены варианты современных интерпретаций проанализированных композиций.

In malignen Zellen ist Apoptose häufig dysreguliert. Entscheidend bei der Regulierung der Apoptoseinduktion ist die Bcl-2-Proteinfamilie, deren Vertreter mittels Bcl-2-homologen Domänen (BH 1-4) interagieren. Die Proteinfamilie umfasst antiapoptotische Proteine wie Mcl-1 und proapoptotische wie Bax und Bak. Harakiri (Hrk) gehört zur proapoptotischen Untergruppe der lediglich die BH3-Domäne aufweisenden BH3 only-Proteine, die Bax und Bak durch Hemmung der antiapoptotischen Proteine aktivieren. Da die Expression von Hrk in verschiedenen Neoplasien erniedrigt ist, sollte in dieser Arbeit untersucht werden, welche Expressionsmuster von Bax und Bak Neoplasien einer Tumortherapie mittels Substitution von Hrk zugänglich machen. Um die durch Hrk aktivierten Signalwege zu untersuchen, wurde ein rekombinanter adenoviraler Vektor (Ad-Hrk) zur regulierbaren Überexpression von Hrk hergestellt. Mit dem Vektor transfizierte Hrk-negative HCT 116-Coloncarcinomzellen zeigen unter sog. On-Bedingungen eine deutliche Hrk-Expression. Die durchflusszytometrische Bestimmung der Rate hypodiploider Zellen erwies, dass die ektope Expression von Hrk in Wildtyp-HCT-116-Zellen Apoptose induziert. Um eine Abhängigkeit Hrk-induzierter Apoptose von Bax und Bak zu adressieren, wurde Hrk zusätzlich in HCT-116-Zellen mit spezifischem Verlust von Bax, Bak oder Bax und Bak überexprimiert. Im Gegensatz zu Bax-profizienten Zellen erwiesen sich Bax- defiziente HCT-116-Zellen unabhängig vom Bak-Expressionsstatus als resistent gegenüber Hrk. Hrk-Expression führte in Bax-profizienten Zellen, nicht aber in Bax-defizienten Zellen zum Verlust des mitochondrialen Membranpotentials, zur Caspase 3-Aktivierung und zur DNA-Fragmentierung. Die Hrk-Resistenz Bax- negativer/Bak-positiver HCT-116-Zellen konnte interessanterweise durch siRNA- vermittelte Herunterregulation des Bak-Inhibitors Mcl 1 überwunden werden. Die Ergebnisse zeigen, dass eine erniedrigte Bax-Expression in Kombination mit Mcl 1-Expression ein Resistenzfaktor gegenüber Hrk-induzierter Apoptose ist. Folglich kommen nur Bax exprimierende oder Mcl-1-negative Neoplasien für eine Hrk substituierende Tumortherapie in Frage. Da der Verlust von Bax und die Expression von Mcl-1 in Tumorzellen jedoch häufig vorkommen, ist vermutlich selbst in solchen Neoplasien ein Teil der Zellen resistent gegenüber Hrk- Substitution. Daher sollten entsprechende klinische Studien Kombinationstherapien mit einem Pharmakon nutzen, das diese Zellen mittels Inhibition von Mcl-1 sensibilisieren kann., Apoptosis is frequently dysregulated in malignant neoplasia. A crucial role in the regulation of apoptosis induction is played by the Bcl-2 family of proteins, which interact via Bcl-2 homology domains (BH1-4). The protein family comprises antiapoptotic proteins, such as Mcl-1, as well as proapoptotic proteins, such as Bax and Bak. Harakiri (Hrk) belongs to the subgroup of proapoptotic proteins which exhibit only the BH3 domain and are therefore called BH3-only proteins. These proteins activate Bax and Bak by inhibiting the antiapoptotic Bcl-2 family members. As the expression of hrk is suppressed in different types of neoplasia, Hrk is a potential target of a therapy that focusses on inducing apoptosis in tumour cells. The aim of this study is to determine which expression patterns of Bax and Bak in tumours are suitable for Hrk functionally substituting tumour therapies. To investigate the signaling pathways downstream of Hrk, a recombinant adenoviral vector system (Ad-Hrk) for controlled overexpression of the protein was established. HCT 116 colon carcinoma cells transfected with Ad-Hrk show a distinct expression of Hrk if incubated under so-called On-conditions. Flow cytometric analysis of the rate of hypodiploid cells revealed induction of apoptosis in wild type HCT 116 cells after ectopic expression of Hrk. To address a potential dependency of Hrk-induced apoptosis from Bax and Bak, Hrk was overexpressed also in HCT 116 cells with specific loss of Bax, Bak or both. In contrast to Bax-proficient cells, Bax-deficient HCT 116 cells turned out to be resistant to Hrk. This was irrespective of the expression status of Bak. In Bax proficient, but not in Bax deficient cells, expression of Hrk lead to loss of the mitochondrial membrane potential, activation of Caspase 3 and DNA fragmentation. Interestingly, the resistance of Bax negative/Bak positive HCT 116 cells towards Hrk was overcome by siRNA-mediated downregulation of the Bak inhibitor Mcl-1. The results demonstrate that a reduced expression of Bax in combination with the expression of Mcl 1 is a resistance factor against Hrk induced apoptosis. Hence only Bax expressing or Mcl-1 negative neoplasms are suitable for an Hrk substituting tumour therapy. As the loss of Bax and the expression of Mcl 1 are frequently found in tumour cells, presumably even in such neoplasms a part of the cells is resistant towards substitution of Hrk. Therefore accordant clinical studies ought to be based on combination therapies with drugs which can sensitize these cells by inhibition of Mcl 1.

The aim of this diploma thesis is to employ a theatrological approach to the work of one of the most original and influential Czech pantomime troupes of all time, entitled Pantomima Alfreda Jarryho (the Alfred Jarry Pantomime, AJP). The company was established in 1966 in Prague by two young mime artists Boris Hybner and Ctibor Turba and existed as late as the political liquidation of the troupe in 1972 that came due to the process of the so-called normalization period in the former Czechoslovakia. The thesis consists of four major parts that are divided into several subsections. The first part deals with the historical contexts of the art of mime, both international and domestic. The AJP troupe represents the second generation of Czech pantomime that refused the style of the so-called modern pantomime expressed in the international context by Marcel Marceau and in the Czech context by Ladislav Fialka at the Theatre Na zábradlí (Theatre on the Balustrade). Since there is no monograph strictly based on this theatre, although it played a major role among the newly established small theatres in the sixties (and had an important influence on the poetics of the AJP troupe), we briefly introduce the poetics of both the pantomime company led by Fialka and the drama company led by the director Jan Grossman,...

A case of suicide by harakiri is described. The position of the subject, the absence of the shirt and the abdominal L-shaped cut agreed well with the formal procedure of harakiri. The characteristics of the stab wounds present on the right-hand side of the neck confirmed the assumption of self-infliction and excluded, from a legal point of view, murder by consent.
