The capability to selectively and precisely modulate neural activities represents a powerful tool for neuroscience research and clinical therapeutics. Traditional electrical stimulations associate with bulky and tethered implants, and optogenetic methods rely on genetic modification for cell targeting. Here, we report an optoelectronic, non-genetic strategy for exciting and inhibiting neural activities, accomplished by bioresorbable, thin-film silicon (Si) diodes. Under illumination, these devices establish polarity-dependent, positive or negative voltages at the semiconductor/solution interface. Such photovoltaic signals enable deterministic depolarization and hyperpolarization of cultured neurons, upregulating and downregulating intracellular calcium dynamics in vitro. Furthermore, flexible, thin-film Si based devices mounted on the nerve tissue selectively activate and silence in vivo activities, both in the peripheral nerve and the brain. Finally, these Si membranes naturally dissolve within the animal body. Such a Si-based material and device platform offers broad potential for biomedical applications.

Dysregulation of ion and potential homeostasis in the scala media is the most prevalent cause of hearing loss in mammals. However, it is not well understood how the development and function of the stria vascularis regulates this fluid homeostasis in the scala media. From a mouse genetic screen, we characterize a mouse line, named 299, that displays profound hearing impairment. Histology suggests that 299 mutant mice carry a severe, congenital structural defect of the stria vascularis. The in vivo recording of 299 mice using double-barreled electrodes shows that endocochlear potential is abolished and potassium concentration is reduced to ∼20 mM in the scala media, a stark contrast to the +80 mV endocochlear potential and the 150 mM potassium concentration present in healthy control mice. Genomic analysis revealed a roughly 7-kb-long, interspersed nuclear element (LINE-1 or L1) retrotransposon insertion on chromosome 11. Strikingly, the deletion of this L1 retrotransposon insertion from chromosome 11 restored the hearing of 299 mutant mice. In summary, we characterize a mouse model that enables the study of stria vascularis development and fluid homeostasis in the scala media.

Neural activities can be modulated by leveraging light-responsive nanomaterials as interfaces for exerting photothermal, photoelectrochemical or photocapacitive effects on neurons or neural tissues. Here we show that bioresorbable thin-film monocrystalline silicon pn diodes can be used to optoelectronically excite or inhibit neural activities by establishing polarity-dependent positive or negative photovoltages at the semiconductor/solution interface. Under laser illumination, the silicon-diode optoelectronic interfaces allowed for the deterministic depolarization or hyperpolarization of cultured neurons as well as the upregulated or downregulated intracellular calcium dynamics. The optoelectronic interfaces can also be mounted on nerve tissue to activate or silence neural activities in peripheral and central nervous tissues, as we show in mice with exposed sciatic nerves and somatosensory cortices. Bioresorbable silicon-based optoelectronic thin films that selectively excite or inhibit neural tissue may find advantageous biomedical applicability.

Significance Some animals have evolved an adaptive ability for vocalizing and hearing ultrasonic frequencies that are inaudible to humans (>20 kHz). For decades, it has been postulated that animals hear ultrasonic frequencies with cochlear hair cells using an identical set of mechanotransduction molecules in the hair bundles for hearing audible frequencies. Here, we show that mice lacking mechanosensitive PIEZO2 ion channels have difficulty hearing at ultrasonic frequencies but remain remarkably sensitive to audible frequencies. Thus, animals may use a partially different mechanism for sensing ultrasonic sound emissions., Ultrasonic hearing and vocalization are the physiological mechanisms controlling echolocation used in hunting and navigation by microbats and bottleneck dolphins and for social communication by mice and rats. The molecular and cellular basis for ultrasonic hearing is as yet unknown. Here, we show that knockout of the mechanosensitive ion channel PIEZO2 in cochlea disrupts ultrasonic- but not low-frequency hearing in mice, as shown by audiometry and acoustically associative freezing behavior. Deletion of Piezo2 in outer hair cells (OHCs) specifically abolishes associative learning in mice during hearing exposure at ultrasonic frequencies. Ex vivo cochlear Ca2+ imaging has revealed that ultrasonic transduction requires both PIEZO2 and the hair-cell mechanotransduction channel. The present study demonstrates that OHCs serve as effector cells, combining with PIEZO2 as an essential molecule for ultrasonic hearing in mice.

A biodegradable and self-electrified conduit device is achieved to offer postoperative electrical cues for nerve regeneration., Peripheral nerve regeneration remains one of the greatest challenges in regenerative medicine. Deprivation of sensory and/or motor functions often occurs with severe injuries even treated by the most advanced microsurgical intervention. Although electrical stimulation represents an essential nonpharmacological therapy that proved to be beneficial for nerve regeneration, the postoperative delivery at surgical sites remains daunting. Here, a fully biodegradable, self-electrified, and miniaturized device composed of dissolvable galvanic cells on a biodegradable scaffold is achieved, which can offer both structural guidance and electrical cues for peripheral nerve regeneration. The electroactive device can provide sustained electrical stimuli beyond intraoperative window, which can promote calcium activity, repopulation of Schwann cells, and neurotrophic factors. Successful motor functional recovery is accomplished with the electroactive device in behaving rodent models. The presented materials options and device schemes provide important insights into self-powered electronic medicine that can be critical for various types of tissue regeneration and functional restoration.

Frameshift mutation caused by small insertions/deletions (indels) often generate truncated and non-functional proteins, which underlies 22% inherited Mendelian disorders in humans. However, there is no efficient in vivo gene therapy strategies available to date, especially in postmitotic systems. Here, we leveraged the non-homologous end joining (NHEJ) mediated non-random editing profiles to compensate the frameshift mutation in a USH1F mouse model – av3j. After treatment by the selected gRNA, about 50% editing products showed reading-frame restoration, and more than 70% targeted hair cells recovered mechanotransduction. In vivo treatment ameliorated the hearing and balance symptoms in homozygous mutant mice. Furthermore, a scale-up analysis of 114 gRNAs targeting 40 frameshift deafness mutations reveals that 65% loci have at least one gRNA with predicted therapeutic potential. Together, our study demonstrates that the NHEJ-mediated frame restoration is a simple and highly efficient therapeutic strategy for small-indel induced frameshift mutations.

Automatic extraction of Chinese synonyms plays an important role in information retrieval and semantic resource construction. Based on the analyzing and comparing the different technologies of synonyms extraction, this paper proposes multi-strategy method including literal similarity algorithm, pattern matching algorithm and PageRank algorithm to extraction Chinese synonyms from encyclopedia resource. The method is language-independent and supports any domain. In addition, it can be able to extract synonyms in various types of expressions. The experiment result shows that the method is feasible and practical.
