1. Genetic Variation and Heritability of Sensory and Artisan Bread Traits in a Set of SRW Wheat Breeding Lines

Abstract

Focus on local food production and supply chains has heightened in recent years, as evidenced and amplified by the COVID-19 pandemic. This study aimed to assess the suitability of soft red winter (SRW) wheat breeding lines for local artisan bakers interested in locally sourced, strong gluten wheat for bread. Seventy-six genotyped SRW wheat breeding lines were milled into whole wheat flour and baked into small loaves. Bread aroma, flavor, and texture were evaluated by a sensory panel, and bread quality traits, including sedimentation volume, dough extensibility, and loaf volume, were measured to estimate heritability. SE-HPLC was performed on white flour, and breeding lines were characterized for different protein fraction ratios. Heritability of loaf volume was moderately high (h2 = 0.68), while heritability of sedimentation volume, a much easier trait to measure, was slightly lower (h2 = 0.55). Certain protein fraction ratios strongly related to loaf volume had high heritability (h2 = 0.7). Even though only a moderate heritability estimate of dough extensibility was found in our study, high positive correlations were found between this parameter and sedimentation volume (r = 0.6) and loaf volume (r = 0.53). This low-input and highly repeatable parameter could be useful to estimate dough functionality characteristics. Flavor and texture heritability estimates ranged from 0.16 to 0.37, and the heritability estimate of aroma was not significantly different from zero. However, the sensorial characteristics were significantly correlated with each other, suggesting that we might be able to select indirectly for aroma by selecting for flavor or texture characteristics. From a genome-wide association study (GWAS), we identified six SNPs (single nucleotide polymorphisms) associated with loaf volume that could be useful in breeding for this trait. Producing high-quality strong gluten flour in our high rainfall environment is a challenge, but it provides local growers and end users with a value-added opportunity.

Abstract

The possibility that N fertilizer increases soil organic matter (SOM) mineralization and, as a result, reduces SOM stocks has led to a great debate about the long-term sustainability of maize-based agroecosystems as well as the best method to estimate fertilizer N use efficiency (FNUE). Much of this debate is because synthetic N fertilizer can positively or negatively affect SOM mineralization via several direct and indirect pathways. Here, we test a series of hypotheses to determine the direction, magnitude, and mechanism of N fertilizer effect on SOM mineralization and discuss the implications for methods to estimate FNUE. We measured the effect of synthetic N fertilizer on SOM mineralization via gross ammonification at two long-term experiments in central and southern Iowa, USA with replicated plots of continuous maize that received one of three “historical” N fertilizer rates (zero, moderate or high) from 1999 to 2014. In 2015, prior to our measurements, we split the historical N fertilizer rate plots into two subplots that received either the site-specific agronomic optimum N rate or zero N fertilizer. At the onset of rapid maize N uptake, N fertilizer reduced gross ammonification by 13–21% (2–5 kg NH4-N ha−1 d−1). A companion laboratory experiment rejected the hypothesis that differences in net primary productivity between fertilized and unfertilized treatments explained the negative effect of N fertilizer on SOM mineralization. Moreover, the NH4+ pool size was negatively correlated with the gross ammonification rate (r2 = 0.85, p < 0.001). Thus, we conclude that NH4+-N fertilizer had a direct suppressive effect on SOM mineralization. These results demonstrate that the direct effect of N fertilizer on microbial activity can exceed the indirect effects of N fertilizer via large changes in NPP that alter organic matter inputs, soil temperature and moisture content. The magnitude of this effect and specificity to NH4+-N has significant implications for fertilizer management as well as the measurement and modeling of agroecosystem N dynamics including FNUE.

Abstract

Potential carbon mineralization (Cmin) is a commonly used indicator of soil health, with greater Cmin values interpreted as healthier soil. While Cmin values are typically greater in agricultural soils managed with minimal physical disturbance, the mechanisms driving the increases remain poorly understood. This study assessed bacterial and archaeal community structure and potential microbial drivers of Cmin in soils maintained under various degrees of physical disturbance. Potential carbon mineralization, 16S rRNA sequences, and soil characterization data were collected as part of the North American Project to Evaluate Soil Health Measurements (NAPESHM). Results showed that type of cropping system, intensity of physical disturbance, and soil pH influenced microbial sensitivity to physical disturbance. Furthermore, 28% of amplicon sequence variants (ASVs), which were important in modeling Cmin, were enriched under soils managed with minimal physical disturbance. Sequences identified as enriched under minimal disturbance and important for modeling Cmin, were linked to organisms which could produce extracellular polymeric substances and contained metabolic strategies suited for tolerating environmental stressors. Understanding how physical disturbance shapes microbial communities across climates and inherent soil properties and drives changes in Cmin provides the context necessary to evaluate management impacts on standardized measures of soil microbial activity. Abstract

Nitrogen fertilization is critical to optimize short-term crop yield, but its long-term effect on soil organic C (SOC) is uncertain. Here, we clarify the impact of N fertilization on SOC in typical maize-based (Zea mays L.) Midwest U.S. cropping systems by accounting for site-to-site variability in maize yield response to N fertilization. Within continuous maize and maize-soybean [Glycine max (L.) Merr.] systems at four Iowa locations, we evaluated changes in surface SOC over 14 to 16 years across a range of N fertilizer rates empirically determined to be insufficient, optimum, or excessive for maximum maize yield. Soil organic C balances were negative where no N was applied but neutral (maize-soybean) or positive (continuous maize) at the agronomic optimum N rate (AONR). For continuous maize, the rate of SOC storage increased with increasing N rate, reaching a maximum at the AONR and decreasing above the AONR. Greater SOC storage in the optimally fertilized continuous maize system than in the optimally fertilized maize-soybean system was attributed to greater crop residue production and greater SOC storage efficiency in the continuous maize system. Mean annual crop residue production at the AONR was 22% greater in the continuous maize system than in the maize-soybean system and the rate of SOC storage per unit residue C input was 58% greater in the monocrop system. Our results demonstrate that agronomic optimum N fertilization is critical to maintain or increase SOC of Midwest U.S. cropland.

Abstract

Cumulative crop recovery of synthetic fertiliser nitrogen (N) over several cropping seasons (legacy effect) generally receives limited attention. The increment in crop N uptake after the first-season uptake from fertiliser can be expressed as a fraction (∆RE) of the annual N application rate. This study aims to quantify ∆RE using data from nine long-term experiments (LTEs). As such, ∆RE is the difference between first season (RE1st) and long-term (RELT) recovery of synthetic fertiliser N. In this study, RE1st was assessed either by the 15N isotope method or by a zero-N subplot freshly superimposed on a long-term fertilised LTE treatment plot. RELT was calculated by comparing N uptake in the total aboveground crop biomass between a long-term fertilised and long-term control (zero-N) treatment. Using a mixed linear effect model, the effects of climate, crop type, experiment duration, average N rate, and soil clay content on ∆RE were evaluated. Because the experimental setup required for the calculation of ∆RE is relatively rare, only nine suitable LTEs were found. Across these nine LTEs in Europe and North America, the mean ∆RE was 24.4% (±12.0%, 95% CI) of annual N application, with higher values for winter wheat than for maize. This result shows that fertiliser-N retained in the soil and stubble may contribute substantially to crop N uptake in subsequent years. Our results suggest that an initial recovery of 43.8% (±11%, 95% CI) of N application may increase to around 66.0% (±15%, 95% CI) on average over time. Furthermore, we found that ∆RE was not clearly related to long-term changes in topsoil total N stock. Our findings show that the—often used—first-year recovery of synthetic fertiliser N application does not express the full effect of fertiliser application on crop nutrition. The fertiliser contribution to soil N supply should be accounted for when exploring future scenarios on N cycling, including crop N requirements and N balance schemes. Highlights: Nine long-term cereal experiments in Europe and USA were analysed for long-term crop N recovery of synthetic N fertiliser. On average, and with application rates between 34 and 269 kg N/ha, crop N recovery increased from 43.8% in the first season to 66.0% in the long term. Delta recovery was larger for winter wheat than maize. Observed increases in crop N uptake were not explained by proportionate increases in topsoil total N stock.
