3. Chemical Constituents from Memecylon Scutellatum with Nitric Oxide Inhibitory Activity.

Abstract

Objectives: Memecylon scutellatum (Lour.) Hook. & Arn. (Melastomataceae) is a relatively lesser-known species within this genus. This study aims to explore its chemical structure profile and evaluate its ability to inhibit NO production. Methods: The chemical structures of MS were elucidated by comparing their NMR data, and all compounds were evaluated for NO inhibition using the Griess assay. Results: One new compound namely, memeloside (1) and fourteen known compounds, 1- O -(β -D-glucopyranosyl)-2-[2-methoxy-4-(ω -hydroxypropyl)-phenoxy]-propan-3-ol (2), (2 R)- O -[4′-(3″-hydroxypropyl)-2′,6′dimethoxyphenyl]-3- O - β -D-glucopyranosyl- sn -glycerol (3), schizandriside (4), 2-butanone 4-glucopyranoside 6′- O -gallate (5), epicatechin (6), epigallocatechin (7), epigallocatechin gallate (8), myricitrin (9), benzyl O - α -L-rhamnopyranosyl-(1→6)- β -D-glucopyranoside (10), benzyl glucopyranoside (11), 3,4,5-trimethoxyphenyl- β -D-glucopyranoside (12), icariside B1 (13), gallic acid (14), and (7 R,8 R)- threo -7,9,9ʹ-trihydroxy-3,3ʹ-dimethoxy-8- O -4ʹ-neolignan 4- O - β -D-glucopyranoside (15) were isolated from the leaves of M. scutellatum. Compounds 4 and 5 significantly exhibited inhibition of NO production, with IC50 values of 88.92 ± 1.42 µM and 17.14 ± 0.98 µM, respectively. Conclusions: One new compound, named memeloside (1) and fourteen known compounds isolated from the leaves of M. scutellatum. Compounds 4 and 5 demonstrated significant inhibition of NO production, with IC50 values of 88.92 ± 1.42 µM and 17.14 ± 0.98 µM, respectively. One new bithiophene derivative, 5-(but-3-en-1-yn-1-yl)-5′-(methoxymethyl)-2,2′-bithiophene (1), along with twelve known compounds, senecioester (2), tiglinsaureester (3), 5-acetoxymethyl-2′-(but-3-en-1-yn-1-yl)-2,5′-bithiophene (4), 5-(4-isovaleroyloxybut-1-ynyl)-2,2′-bithiophene (5), 5-hydroxymethyl-(2,5′:2′,5′′)-terthienyl tiglate (6), 5-hydroxymethyl-(2,5′:2′,5′′)-terthienyl agelate (7), 5- hydroxymethyl-2,5′:2′,5′′-terthiophene dimethylacrylate (8), 5-methoxymethyl-2,2′:5′,2′′-terthiophene (9), α-terthiophene (10), 1,3,8,9-tetrahydroxycoumestan 3-sulfate (11), demethylwedelolactone (12), and wedelolactone (13) were isolated from the methanol extract of aerial parts of Eclipta prostrata (L.) L. All isolated compounds were evaluated for the protective ability on the HepG2 cells. At the concentration of 100 μM, compounds 11-13 showed the highest hepatoprotective effects, with HepG2 cell viability ranging from 38.68% to 48.54%. Abstract

One new indol, N-methoxymethyltryptophol (1), one new phenolic, (2 R)-2-(4-hydroxyphenyl)ethyl 2-hydroxy-3-phenylpropanoate (2) and fifteen known compounds (3–17) were isolated from the methanol extract of the fermentation of marine microalgae Aurantiochytrium sp. SC145. Their structures were elucidated by 1D-, 2D-NMR spectroscopic analysis, HR-ESI-MS, quantum chemical calculation methods and by comparing their NMR data with those reported in the literature. All compounds were evaluated for their antimicrobial activities against microorganisms. Compounds 2, 3 and 11 significantly exhibited antimicrobial activities on all tested Gram-(+), Gram-(-) bacteria and the yeast C. albicans with MIC values ranging from 32 to 256 µg/mL. Abstract

A new iridoid glucoside, moridoside (1), and nine known compounds, asperulosidic acid (2), 6-O-epi-acetylscandoside (3), geniposidic acid (4), 2-hydroxymethylanthraquinone (5), 2-hydroxymethyl-3-hydroxyanthraquinone (6), damnacanthol (7), lucidine-ω-methyl ether (8), 2-hydroxy-1-methoxyanthraquinone (9), and 3,8-dihydroxy-1,2-dimethoxyanthraquinone (10) were isolated from the methanol extract of Morinda officinalis How. roots. Their structural identification was carried out based on the spectroscopic evidence. All compounds were evaluated for their nitric oxide (NO) production inhibitory activities in LPS-stimulated RAW264.7 macrophages. Compounds 5-7 significantly inhibited the production of NO with IC50 values of 28.4, 33.6, and 30.5 μM, respectively.

Abstract

Propolis from stingless bees in Vietnam hasbeen used in traditional medicine to improve health and prevent diseases.However, there have been few investigations on the chemical constituents andbioactivity of Vietnamese stingless bee propolis. This work studied thechemical composition of the stingless bee propolis collected from the nests of Lisotrigonafurva in Binhdinh province, Vietnam using combined chromatographic methods.The chemical structures of isolated compounds were determined by MS, NMRspectral analysis. Several xanthones including cochichinone A, cochichinone I,cochichinone J, cratoxylumxanthone B, α-mangostanin and pruniflorone S wereisolated from the ethyl acetate extract of Lisotrigona furva propolis.This is the first report on the chemical constituents of propolis from Lisotrigonaspecies. Cratoxylum cochinchinense (Hypericaceae) is the plant sourcefor the stingless bee Lisotrigona furva propolis. Two new prenylated flavonoids, 4´-methyl-8-prenyltaxifolin (1) and 6,8-diprenyl-4´-methyl-naringenin (2) and a new geranylated stilbene, 4′-deprenyl-4-methoxymappain (3) together with eight known flavonoids (4–11) were isolated from the fruits of Macaranga balansae Gagnep. Abstract

Two new lignans, palatiferin A ( 1) and palatiferin B ( 2), were isolated from the roots of PSEUDERANTHEMUM PALATIFERUM, together with five known triterpenes, epifriedelanol ( 3), lupeol ( 4), lupenone ( 5), betulin ( 6), pomolic acid ( 7), and a dipeptide asperglaucide ( 8). Their structures were established from 2D NMR and mass spectroscopy. The absolute configuration of 1 and 2 was proposed based on the comparison of their optical rotation activities with those of compounds with similar structures such as wodeshiol and paulownin. The new lignans, palatiferin A ( 1) and palatiferin B ( 2) exhibited a moderate cytotoxicity against KB and HepG2 cell lines. However, betulin and lupeol, two abundant compounds from the roots of P. PALATIFERUM, showed cytotoxic and antimicrobial activities.

Abstract

Cleistanone (1) was isolated from the leaves of Cleistanthus indochinensis and features a new type of triterpenoid carbon skeleton. Its structure was confirmed by X-ray diffraction analysis. A plausible biosynthetic pathway was proposed for 1 and is thought to proceed through a Wagner–Meerwein rearrangement. Cleistanone (1) was found to show cytotoxic activity against human oral epidermoid carcinoma KB cells.
