Abstract Background Myo-Inositol Phosphate Synthase (MIP) catalyzes the conversion of glucose 6- phosphate into inositol phosphate, an essential nutrient and cell signaling molecule. Data obtained, first in bovine brain and later in plants, established MIP expression in organelles and in extracellular environments. A physiological role for secreted MIP has remained elusive since its first detection in intercellular space. To provide further insight into the role of MIP in intercellular milieus, we tested the hypothesis that MIP may function as a growth factor, synthesizing inositol phosphate in intercellular locations requiring, but lacking ability to produce or transport adequate quantities of the cell–cell communicator. This idea was experimentally challenged, utilizing a Saccharomyces cerevisiae inositol auxotroph with no MIP enzyme, permeable membranes with a 0.4 µm pore size, and cellular supernatants as external sources of inositol isolated from S. cerevisiae cells containing either wild-type enzyme (Wt-MIP), no MIP enzyme, auxotroph (Aux), or a green fluorescent protein (GFP) tagged reporter enzyme (MIP- GFP) in co- culturing experiments. Results Resulting cell densities and microscopic studies with corroborating biochemical and molecular analyses, documented sustained growth of Aux cells in cellular supernatant, concomitant with the uptakeof MIP, detected as MIP-GFP reporter enzyme. These findings revealed previously unknown functions, suggesting that the enzyme can: (1) move into and out of intercellular space, (2) traverse cell walls, and (3) act as a growth factor to promote cellular proliferation of an inositol requiring cell. Conclusions Co-culturing experiments, designed to test a probable function for MIP secreted in extracellular vesicles, uncovered previously unknown functions for the enzyme and advanced current knowledge concerning spatial control of inositol phosphate biosynthesis. Most importantly, resulting data identified an extracellular vesicle (a non-viral vector) that is capable of synthesizing and transporting inositol phosphate, a biological activity that can be used to enhance specificity of current inositol phosphate therapeutics.

In response to COVID-19, educators rapidly pivoted to new and innovative ways of delivering lecture material. The ability to host synchronous lectures on platforms like Zoom gave students continued access to classroom material in the face of an ongoing pandemic. The purpose of this study was to investigate the differences in exam scores between students attending a physiology class (PHSL3051) synchronously via Zoom or asynchronously by viewing recorded lectures posted after class. Students in PHSL3051 were evaluated with four unit exams and one cumulative final exam. Although pooled analysis of all students showed that synchronous lecture viewing positively predicted exam scores, this positive association was even larger when the data were analyzed by gender and ethnicity. For female-identified students and students of color (SOC), attending lectures synchronously was associated with average scores on every unit exam that were higher by 2.7-7.4 percentage points. Moreover, the greater a student's synchronous participation in the course throughout the semester, the better that student's performance on the cumulative final exam was likely to be. These data highlight the need to better understand how different groups of undergraduate students select and respond to different assessment methods used in the same course, which may have long-term effects on their overall performance at 4-year institutions.

Kedelai merupakan komoditas pertanian yang sangat penting di Indonesia. Namun, produksi kedelai nasional belum dapat memenuhi kebutuhan kedelai nasional. Upaya yang dapat dilakukan adalah dengan menerapkan pola tanam tumpangsari kedelai dengan jagung. Penelitian ini bertujuan untuk mencari genotip kedelai yang mampu beradaptasi pada pertanaman tumpangsari dengan jagung pola 3:1 serta menghitung produktivitas penggunaan lahan setiap genotip pada pertanaman tumpangsari dengan jagung pola 3:1. Penelitian ini menggunakan metode eksperimen Rancangan Acak Kelompok (RAK). 16 genotip kedelai digunakan sebagai perlakuan, dan diulang sebanyak dua kali. Untuk melihat respons genotip pada pertanaman tumpangsari dilakukan dengan uji Least Significant Increase (LSI) pada taraf signifikansi 5%. Pendugaan produktivitas lahan pada setiap genotip dihitung berdasarkan nilai Nisbah Kesetaraan Lahan. Hasil penelitian ini menunjukkan genotip kedelai yang memperlihatkan respons paling baik pada pertanaman tumpangsari kedelai jagung dibandingkan kultivar cek untuk tinggi tanaman yaitu genotip BTN 5 dan JT 3, karakter jumlah cabang produktif yaitu genotip BTN 5, karakter luas daun dan indeks luas daun yaitu genotip JT 3, karakter sudut daun yaitu genotip CK 6, karakter bobot per plot yaitu genotip KBI 2, dan Nisbah Kesetaraan Lahan yaitu genotip CK 6 dan KBI 2.Genotip BTN 1, BTN 2, BTN 5, CK 15, CK 6, JT 3, KA 6, KA 7, KBI 2, KH 8, Cikuray dan Malikka memiliki nilai NKL lebih besar dari 1,0. Soybean is one of important agricultural commodity in Indonesia. However, the national soybean production is not sufficient to meet the needs of national soybean. Effort to do is by applying an intercropping soybean with corn. The objectives of the research was to find soybean genotypes that can adapt to intercropped plantation with corn in pattern of 3 : 1 as well as the productivity of the land use of each genotype in intercropping with maize in 3 : 1 pattern. The research used an experimental method randomized block design (RBD), 16 soybean genotypes as treatments, and repeated twice. To see the response of genotype in intercropping planting was done by using Least Significant Increase (LSI) at the level of significant 5%. Estimation of land productivity on each genotype was calculated with Land Equation Ratio value. The results showed that the best response of soybean genotype compared checks cultivar for plant height were genotype BTN 5 and JT 3, character number of productive branches was genotype BTN 5, the character of leaf area and leaf area index was genotype JT 3 , leaf angle character was genotype CK 6, characters of weights per plot was KBI 2 genotype and Land Equation Ratio value were CK 6 and KBI 2. Genotypes of BTN 1, BTN 2, BTN 5, CK 15, CK 6, JT 3, KA 6, KA 7, KBI 2, KH 8, Cikuray and Malikka had Land Equivalent Ratio values greater than 1.0.

Abstract Background Assisted living (AL) is an increasingly common residential setting for persons with dementia; yet concerns exist about sub-optimal care of this population in AL given its lower levels of staffing and services. Our objectives were to (i) examine associations between AL setting (dementia care vs. other), COVID-19 pandemic waves, and prevalent antipsychotic, antidepressant, anti-dementia, benzodiazepine, and anticonvulsant drug use among residents with dementia/cognitive impairment, and (ii) explore associations between resident and home characteristics and prevalent medication use. Methods We conducted a population-based, repeated cross-sectional study using linked clinical and health administrative databases for all publicly funded AL homes in Alberta, Canada, examined between January 2018 - December 2021. The quarterly proportion of residents dispensed a study medication was examined for each setting and period (pandemic vs. comparable historical [2018/2019 combined]) focusing on four pandemic waves (March-May 2020, September 2020-February 2021, March-May 2021, September-December 2021). Log-binomial GEE models estimated prevalence ratios (PR) for period (pandemic vs. historical periods), setting (dementia care vs. other) and period-setting interactions, adjusting for resident (age, sex) and home (COVID-19 cases, health region, ownership) characteristics. Results On March 1, 2020, there were 2,779 dementia care and 3,013 other AL residents (mean age 83, 69% female) with dementia/cognitive impairment. Antipsychotic use increased during waves 2–4 in both settings, but this was more pronounced in dementia care than other AL during waves 3 and 4 (e.g., adjusted [adj]PR 1.20, 95% CI 1.14–1.27 vs. adjPR 1.09, 95% CI 1.02–1.17, interaction p = 0.023, wave 3). Both settings showed a statistically significant but modest increase in antidepressant use and decrease in benzodiazepine use. For dementia care AL residents only, there was a statistically significant increase in gabapentinoid use during several waves (e.g., adjPR 1.32, 95% CI 1.10–1.59, wave 3). Other than a modest decrease in prevalent anti-dementia drug use for both settings in wave 2, no other significant pandemic effects were observed. Conclusions The persistence of the pandemic-associated increase in antipsychotic and antidepressant use in AL residents coupled with a greater increase in antipsychotic and gabapentinoid use for dementia care settings raises concerns about the attendant risks for residents with cognitive impairment.

Abstract Background After settling in the United States (US), immigrants often accumulate obesity and cardiovascular risk factors. As mood is often associated with health behaviors in the US population, mood may be an important mediating factor in immigrant populations. Methods The Healthy Immigrant Community (HIC) study, set in southeast Minnesota, enrolled 475 adult participants in a weight loss intervention designed to reduce cardiovascular risk. Baseline questionnaires assessed mood, nutrition, physical activity, self-efficacy for healthy eating and physical activity, social support, and cohesion. A single-item mood rating of poor or fair was considered “negative”, while ratings of good, very good, or excellent were considered “positive”. Results Hispanic/Latino (n = 268) and Somali (n = 181) adults enrolled in HIC completed baseline measures and were included in this analysis. Participants endorsing negative mood compared to positive mood had lower healthy eating scores (p = 0.02), lower physical activity levels (p = 0.03), lower confidence in eating a healthy diet (p = 0.001), and felt less of a sense of belonging to their community (p = 0.01). One of the biological parameters of air quality can be seen from the presence of microorganisms such as bacteria. The presence of airborne microorganisms, especially in enclosed spaces can affect human health, one of which can cause respiratory infections. On the other hand, people often use air freshener products. Therefore, this research was conducted with the aim of analyzing the potential of air freshener based on siwalan (Borasus flabellifer) coir extract and lemongrass as antibacterial against airborne bacteria. This research is an experimental research with a pre test-post test design. The data obtained is in the form of the value of the Total Plate Count of indoor air bacteria before and after treatment using air freshener based on coir extract of siwalan and lemongrass. The results showed that both formulations (P1 and P2) of air freshener based on coir extract and lemongrass were able to reduce the number of airborne bacterial colonies in the room, with a significance value of 0.000

Abstract Background Neurodevelopmental disorders (NDDs) and/or associated multiple congenital abnormalities (MCAs) represent a genetically heterogeneous group of conditions with an adverse prognosis for the quality of intellectual and social abilities and common daily functioning. The rapid development of exome sequencing (ES) techniques, together with trio-based analysis, nowadays leads to up to 50% diagnostic yield. Therefore, it is considered as the state-of-the-art approach in these diagnoses. Results In our study, we present the results of ES in a cohort of 85 families with 90 children with severe NDDs and MCAs. The interconnection of the in-house bioinformatic pipeline and a unique algorithm for variant prioritization resulted in a diagnostic yield of up to 48.9% (44/90), including rare and novel causative variants (41/90) and intragenic copy-number variations (CNVs) (3/90). Of the total number of 47 causative variants, 53.2% (25/47) were novel, highlighting the clinical benefit of ES for unexplained NDDs. Moreover, trio-based ES was verified as a reliable tool for the detection of rare CNVs, ranging from intragenic exon deletions (GRIN2A, ZC4H2 genes) to a 6-Mb duplication. The functional analysis using PANTHER Gene Ontology confirmed the involvement of genes with causative variants in a wide spectrum of developmental processes and molecular pathways, which form essential structural and functional components of the central nervous system. Conclusion Taken together, we present one of the first ES studies of this scale from the central European region. Based on the high diagnostic yield for paediatric NDDs in this study, 48.9%, we confirm trio-based ES as an effective and reliable first-tier diagnostic test in the genetic evaluation of children with NDDs.

Background and Objectives: Kidney disease (KD) is a common complication of diabetes mellitus (DM) associated with adverse outcomes of renal failure, cardiovascular disease, and mortality. The aim of this study was to determine the prevalence and awareness of the KD among the DM type 2 (T2DM) patients. Materials and Methods: This cross-sectional study was conducted at the University Hospital of Split between November and December of 2023 during an open call for DM patients. For each participant, blood and urine samples, along with relevant medical information, were collected, and adherence to the Mediterranean diet (MeDi) was assessed using the Mediterranean Diet Service Score (MDSS). Furthermore, blood pressure was measured, along with body composition and anthropometric parameters. Results: Of 252 T2DM patients with a median age of 67 years (IQR: 60–73), 130 (51.6%) were women. The median duration of T2DM was 10 years (IQR: 6–20). Despite the fact that 80.95% of total participants reported receiving dietary guidelines from any source, only 53.2% reported adhering to the suggested instructions, while according to the MDSS, only 7.2% adhered to the MeDi. The median body mass index was 27.6 kg/m2 (24.2–31), with 70.1% of participants overweight or obese. Only 6% of participants believed they had KD, but after blood and urine sample analysis, 31% were found to have KD. Conclusions: This study highlights a significant gap in awareness of KD, low adherence to MeDi, and a high prevalence of obesity among T2DM patients. Due to the increasing number of T2DM patients, it is crucial to improve healthy lifestyle education and make modifications within this group, as well as perform regular screening for KD and medical check-ups.

This article presents a mixed-methods research protocol suitable for studying everyday mobility and its effects on life chances for a wide range of populations. We argue that it can address, among other issues, two important methodological challenges in the study of mobility: addressing pre-reflexivity and habits, as well as differentiating between voluntary and forced movements. The research protocol combines a problem-centered interview, two weeks of GPS tracking, and a subsequent mobility interview. It has been shaped by its application to more than 60 socially disadvantaged residents of rural peripheries as part of an international research project. It contributes to existing qualitative methods of spatial analysis because its design incorporates not only theoretical considerations but also insights of real-world application from conducting it in varied contexts and with different populations, which are typically underrepresented in more elaborate research designs. Nevertheless, due to its thematic openness, the design is not only interesting for mobility research, but also for other research focusing on spatial phenomena at the individual level. We provide a detailed account of the method’s implementation, contextualize it in relation to previous approaches, and discuss the analytical potential of the data obtained. We suggest that by triangulating movement data with interview data, perceptions and meanings of everyday mobility can be uncovered that would otherwise remain at a pre-reflexive level.

The concept of Smart Cities integrates innovative technologies to improve citizens’ quality of life in towns and cities worldwide. Crisis management is a separate section directly managed by the leadership of municipalities, cities and counties in cooperation between police, fire and municipal police to ensure the safety of residents and safety in public spaces. The purpose of this study is to investigate to which extent publicly available information related to the field of crisis management is unavailable to residents in municipalities, towns and cities through online information systems. The primary aim is to provide suggestions for a general information system structure and content that would highlight and satisfy the need to address the crisis management issue, especially in providing immediate information to the population through an innovative online form. The achievement of this goal is methodologically based on qualitative research analysing and comparing the information published for residents through Smart City information systems in selected towns and municipalities. Document analysis or conceptual design was applied, and evaluation criteria for objective assessment of Smart City information systems were appropriately determined. The comparative analysis based on this set of criteria enabled the development of the proposals of information systems’ content that can be used to keep the information systems for Smart Cities in cities, municipalities and regions, actual and beneficial. From the available resources, two main modules that focused either on citizens or cities were synthesised. Moreover, SWOT analysis or the Smart Regions Rapid Response structure was derived. Acquired results outline generic structures and contents that support the development of the concept of Smart Cities and can be suitably implemented for the development of the modification of information systems containing relevant information for residents, cities and municipalities, focusing on citizen safety.

Background: Arterial hypertension (AH) is a significant risk factor for cardiovascular disease and is associated with increased arterial stiffness, particularly as measured by pulse wave velocity (PWV). This study aims to explore the relationships between age groups, antihypertensive and new oral antidiabetic drugs, body composition, and arterial stiffness parameters in hypertensive patients. Methods: A single-center cross-sectional study was conducted including 584 participants who underwent 24 h ambulatory blood pressure monitoring (including central blood pressure (BP) and PWV measurement), body composition analysis, and provided medical history and current pharmacotherapy data. Results: The study found that PWV was significantly higher in patients with poorly regulated BP in those aged 65 years and older. Significant PWV predictors included systolic BP, heart rate, peripheral mean arterial pressure, peripheral pulse pressure, augmentation index, calcium channel blockers, moxonidine, sodium–glucose co-transporter 2 inhibitors, urapidil, and statin prescription. Also, statistically significant negative correlations were found between PWV and visceral fat level, fat-free mass, and the percentage of muscle mass. Conclusions: The findings suggest that arterial stiffness is interconnected with peripheral and central blood pressure parameters, body composition parameters, and prescribed hypertensive and new antidiabetic drugs.

Abstract Background Morphological and traditional genetic studies of the young Pliocene genus Hyles have led to the understanding that despite its importance for taxonomy, phenotypic similarity of wing patterns does not correlate with phylogenetic relationship. To gain insights into various aspects of speciation in the Spurge Hawkmoth (Hyles euphorbiae), we assembled a chromosome-level genome and investigated some of its characteristics. Results The genome of a male H. euphorbiae was sequenced using PacBio and Hi-C data, yielding a 504 Mb assembly (scaffold N50 of 18.2 Mb) with 99.9% of data represented by the 29 largest scaffolds forming the haploid chromosome set. Consistent with this, FISH analysis of the karyotype revealed n = 29 chromosomes and a WZ/ZZ (female/male) sex chromosome system. Estimates of chromosome length based on the karyotype image provided an additional quality metric of assembled chromosome size. Rescaffolding the published male H. vespertilio genome resulted in a high-quality assembly (651 Mb, scaffold N50 of 22 Mb) with 98% of sequence data in the 29 chromosomes. The larger genome size of H. vespertilio (average 1C DNA value of 562 Mb) was accompanied by a proportional increase in repeats from 45% in H. euphorbiae (measured as 472 Mb) to almost 55% in H. vespertilio. Several wing pattern genes were found on the same chromosomes in the two species, with varying amounts and positions of repetitive elements and inversions possibly corrupting their function. Conclusions Our two-fold comparative genomics approach revealed high gene synteny of the Hyles genomes to other Sphingidae and high correspondence to intact Merian elements, the ancestral linkage groups of Lepidoptera, with the exception of three simple fusion events. We propose a standardized approach for genome taxonomy using nucleotide homology via scaffold chaining as the primary tool combined with Oxford plots based on Merian elements to infer and visualize directionality of chromosomal rearrangements. The identification of wing pattern genes promises future understanding of the evolution of forewing patterns in the genus Hyles, although further sequencing data from more individuals are needed. The genomic data obtained provide additional reliable references for further comparative studies in hawkmoths (Sphingidae).

Lack of improved varieties and soil fertility degradation are among the major factors that limit durum wheat (Triticum turgidum L.) production in Ethiopia. The objectives of this study were to determine the response of durum wheat cultivars (i) to nitrogen, phosphorus, sulphur, and boron (NPSB) fertilizer blend; (ii) rust infection; and (iii) partial budget analysis using NPSB fertilizer blend. Penelitian ini dilakukan untuk memberikan pemahaman serta menyampaikan gambaran tentang proses penerimaan diri pada anak yang mengalami kematian orang tua secara mendadak. Penelitian ini menggunakan metode kualitatif studi kasus instrumental beserta model analisis data miles and huberman. Teknik pemantapan kredibilitas yang digunakan adalah metode membercheck. Partisipan yang masuk kriteria dalam penelitian ini yaitu anak yang memiliki pengalaman dalam menghadapi kematian kedua orangtua yang meninggal secara mendadak dan mengalami peristiwa tersebut pada usia anak-anak (6-12 tahun). Hasil penelitian menunjukkan bahwa setiap anak sebagai individu yang menghadapi peristiwa kematian kedua orangtua memiliki tahapan penerimaannya tersendiri, meskipun dinamika yang dilalui sesuai dengan yang disampaikan oleh Kubler-Ross. Ditemukan bahwa terdapat respon yang terjadi akibat kematian orang tua pada remaja dan dewasa muda yang berjenis kelamin laki-laki. Respon remaja laki-laki yaitu grieving akibat kematian orang tua dan pada dewasa muda akan merasakan kesedihan yang lebih intens, dan tekanan pribadi yang lebih besar. Namun ada perbedaan dengan remaja laki-laki di panti asuhan yang memiliki resiliensi dan dapat mengurangi rasa cemas dan depresi akibat kematian orang tua.

Abstract Introduction The antifibrotic drug nintedanib is used for the treatment of idiopathic pulmonary fibrosis (IPF). We analysed the effect of nintedanib on antifibrotic treatment outcome in real-world cohorts of Czech EMPIRE registry. Patients/methods Data of 611 Czech IPF subjects, 430 (70%) treated with nintedanib (NIN group), 181 (30%) with no-antifibrotic treatment (NAF group) were analysed. The influence of nintedanib on overall survival (OS), pulmonary function parameters as forced vital capacity (FVC) and diffusing lung capacity for carbon monoxide (DLCO), as well as GAP score (gender, age, physiology) and and CPI (composite physiological index) were investigated. Listeria monocytogenes is an important pathogen responsible for listeriosis, a serious foodborne illness associated with high mortality rates. Therefore, L. monocytogenes is considered a challenge for the food industry due to the ability of some strains to persist in food-associated environments. Biofilm production is presumed to contribute to increased L. monocytogenes resistance and persistence. The aims of this study were to (1) assess the biofilm formation of L. monocytogenes isolates from a meat processing facility and sheep farm previously characterized and subjected to whole-genome sequencing and (2) perform a comparative genomic analysis to compare the biofilm formation and the presence of a known set of biofilm-associated genes and related resistance or persistence markers. Among the 37 L. monocytogenes isolates of 15 sequence types and four serogroups involved in this study, 14%, 62%, and 24% resulted in the formation of weak, moderate, and strong biofilm, respectively. Increased biofilm-forming ability was associated with the presence of the stress survival islet 1 (SSI-1), inlL, and the truncated inlA genes. Combining the phenotypic and genotypic data may contribute to understanding the relationships between biofilm-associated genes and L. monocytogenes biofilm-forming ability, enabling improvement in the control of this foodborne pathogen.

The aim of this cross-sectional study was to evaluate the differences in the levels of advanced glycation end products (AGE) between patients with chronic kidney disease (CKD) and kidney transplant recipients (KTRs) and to investigate the risk factors for the AGE levels in each group of these patients. There were 217 participants total, of which 99 (45.6%) were KTRs and 118 (54.4%) had CKD. Data on the levels of AGE, body mass composition, anthropometric parameters, central and peripheral blood pressure, and clinical and laboratory parameters were gathered for each study participant. The AGE values of the CKD and KTRs groups did not differ from one another. In both groups, a lower estimated glomerular filtration rate, male sex, and older age were positive predictors for increased AGE values. Furthermore, higher levels of AGE were linked to lower central systolic blood pressure (cSBP) in the CKD group, whilst, in the KTRs group, higher levels of AGE were linked to a shorter time since kidney transplantation (KTx), more years of dialysis prior to KTx, lower levels of trunk visceral fat, the presence of arterial hypertension, and the absence of prescriptions for the antihypertensive medications urapidil and angiotensin II receptor blockers. Further studies are needed to better understand the above associations. Consequently, a personalised multidisciplinary approach to assess the cardiovascular as well as dietary and lifestyle risk factors to reduce the AGE levels in both KTRs and CKD patients may be implemented.

Bcr-Abl is an oncoprotein with aberrant tyrosine kinase activity involved in the progression of chronic myeloid leukemia (CML) and has been targeted by inhibitors such as imatinib and nilotinib. However, despite their efficacy in the treatment of CML, a mechanism of resistance to these drugs associated with mutations in the kinase region has emerged. Therefore, in this work, we report the synthesis of 14 new 2,6,9-trisubstituted purines designed from our previous Bcr-Abl inhibitors. Here, we highlight 11b, which showed higher potency against Bcr-Abl (IC50 = 0.015 μM) than imatinib and nilotinib and exerted the most potent antiproliferative properties on three CML cells harboring the Bcr-Abl rearrangement (GI50 = 0.7–1.3 μM). In addition, these purines were able to inhibit the growth of KCL22 cell lines expressing Bcr-AblT315I, Bcr-AblE255K, and Bcr-AblY253H point mutants in micromolar concentrations. Imatinib and nilotinib were ineffective in inhibiting the growth of KCL22 cells expressing Bcr-AblT315I (GI50 > 20 μM) compared to 11b–f (GI50 = 6.4–11.5 μM). Molecular docking studies explained the structure–activity relationship of these purines in Bcr-AblWT and Bcr-AblT315I. Finally, cell cycle cytometry assays and immunodetection showed that 11b arrested the cells in G1 phase, and that 11b downregulated the protein levels downstream of Bcr-Abl in these cells.

AbstractPurpose This study aimed to assess the quality of the lucite cone applicator (LCA), the standard applicator for superficial hyperthermia at the Erasmus MC Cancer Institute, using the most recent quality assurance guidelines, thus verifying their feasibility.Materials and methods The assessment was conducted on each of the six LCAs available for clinical treatments. The temperature distribution was evaluated using an infrared camera across different layers of a fat-muscle mimicking phantom. The maximum temperature increase, thermal effective penetration depth (TEPD), and thermal effective field size (TEFS) were used as quality metrics. The experimental results were validated through comparison with simulated results, using a canonical phantom model and a realistic phantom model segmented from CT imaging.Results A maximum temperature increase above 6 °C at 2 cm depth in the fat-muscle phantom for all the experiments was found. A mean negative difference between simulated and experimental data was of 1.3 °C when using the canonical phantom model. This value decreased to a mean negative difference of 0.4 °C when using the realistic model. Simulated and measured TEPD showed good agreement for both in silico scenarios, while discrepancies were present for TEFS.Conclusions The LCAs passed all QA guidelines requirements for superficial hyperthermia delivery when used singularly or in an array configuration. A further characterization of parameters such as antenna efficiency and heat transfer coefficients would be beneficial for translating experimental results to simulated values. Implementing the QA guidelines was time-consuming and demanding, requiring careful preparation and correct setup of antenna elements.

AbstractBackground Phantoms accurately mimicking the electromagnetic and thermal properties of human tissues are essential for the development, characterization, and quality assurance (QA) of clinically used equipment for Hyperthermia Treatment (HT). Currently, a viable recipe for a fat equivalent phantom is not available, mainly due to challenges in the fabrication process and fast deterioration.Materials and methods We propose to employ a glycerol-in-oil emulsion stabilized with ethylcellulose to develop a fat-mimicking material. The dielectric, rheological, and thermal properties of the phantom have been assessed by state-of-the-art measurement techniques. The full-size phantom was then verified in compliance with QA guidelines for superficial HT, both numerically and experimentally, considering the properties variability.Results Dielectric and thermal properties were proven equivalent to fat tissue, with an acceptable variability, in the 8 MHz to 1 GHz range. The rheology measurements highlighted enhanced mechanical stability over a large temperature range. Both numerical and experimental evaluations proved the suitability of the phantom for QA procedures. The impact of the dielectric property variations on the temperature distribution has been numerically proven to be limited (around 5%), even if higher for capacitive devices (up to 20%).Conclusions The proposed fat-mimicking phantom is a good candidate for hyperthermia technology assessment processes, adequately representing both dielectric and thermal properties of the human fat tissue while maintaining structural stability even at elevated temperatures. However, further experimental investigations on capacitive heating devices are necessary to better assess the impact of the low electrical conductivity values on the thermal distribution.

Context: Mealtimes in long-term care (LTC) settings play a pivotal role in the daily lives of residents. The COVID-19 pandemic and the required precautionary infection control mandates influenced many aspects of resident care within LTC homes, including mealtimes. Limited research has been conducted on how mealtimes in LTC were affected during the pandemic from staff perspectives. Objective: To understand the experiences of LTC staff on providing mealtimes during the pandemic. Methods: Semi-structured telephone interviews were conducted with 22 staff involved with mealtimes between February and April 2021. Transcripts were analysed using interpretive description. Findings: Three themes emerged from the analysis: (1) recognizing the influence of homes’ contextual factors. Home size, availability of resources, staffing levels and resident care needs influenced mealtime practices during the pandemic; (2) perceiving a compromised mealtime experience for residents and staff. Staff were frustrated and described residents as being dissatisfied with mealtime and pandemic-initiated practices as they were task-focused and socially isolating and (3) prioritizing mealtimes while trying to stay afloat. An ‘all hands-on deck’ approach, maintaining connections and being adaptive were strategies identified to mitigate the negative impact of the mandates on mealtimes during the pandemic. Limitations: Perspectives were primarily from nutrition and food service personnel. Implications: Overly restrictive public health measures resulted in mealtime practices that prioritized tasks and safety over residents’ quality of life. Learning from this pandemic experience, homes can protect the relational mealtime experience for residents by fostering teamwork, open and frequent communication and being flexible and adaptive.

The stable transformation of common bean is a challenging and time-consuming process. Although CRISPR/Cas9 has revolutionized gene editing with its high efficiency and specificity, the performance of the system can be affected by multiple factors, such as sgRNA specificity and effectiveness, and the choice of promoter used to drive Cas9 expression. The use of a hairy root transformation system to initially check the efficiency of sgRNAs and the impact of different promoters could speed up this process and increase the chances of success. We initially tested three different transformation methods to induce hairy roots and selected a preferred method suitable for a variety of different common bean genotypes. This method involved inoculating a severed radicle with Rhizobium rhizogenes K599 and was fast, had a high transformation frequency of 42-48%, and resulted in numerous hairy roots. This method was further used for the transformation of explants using R. rhizogenes harboring different CRISPR/Cas9 constructs and evaluated the on-target activity of sgRNAs targeting raffinose family oligosaccharides biosynthetic genes and the impact of different promoters driving Cas9 on the gene editing efficiency. Additionally, we evaluated the reliability of the in silico tools, CRISPOR, CRISPR RGEN, and inDelphi to predict the sgRNA efficiencies and resulting mutations. Our results showed that the hairy root transformation system allows for rapid evaluation of multiple sgRNAs and promoters. We also identified several highly efficient sgRNAs that induced frameshift mutations at rates of up to 70% when a parsley ubiquitin promoter was driving Cas9 expression, providing valuable information for the selection of the most effective sgRNAs and promoters for future transformation experiments. Although most of the computational models used to predict the sgRNA efficiency did not match the in planta results, the Lindel model proved to be the most reliable for P. vulgaris, accurately predicting the sgRNA efficiency and the type of induced mutation in most hairy roots. Furthermore, the inDelphi algorithm could correctly predict deletions and single nucleotide insertions resulting from DNA double-strand breaks in common bean. These results offer promising implications for enhancing precise editing in plants because they provide the possibility of predicting repair outcomes.

Polycystic ovary syndrome (PCOS) is an endocrine and metabolic disorder affecting women of reproductive age. Research has shown that epigenetic alterations such as DNA methylation may play a role in the development and progression of abnormal ovarian function and metabolic disorders in PCOS. Studies have identified specific genes (related with insulin signaling and steroid hormone metabolism) that are methylated in women with PCOS. DNA methylation appears to respond to various interventions aimed at altering health and lifestyle factors. We tested the efficacy of a mindfulness-based stress reduction program (MBSR) in PCOS patients. We examined its effects on anthropometric measurements, mental health and wellbeing, and alterations in DNA methylation in peripheral blood. MBSR was associated with a reduction in body mass index, waist circumference and blood glucose level, an improvement in subjectively perceived general health, emotional role limitation, and levels of pain, as well as mindfulness-like traits. MBSR reduced the expression of anxious symptomatology and subjectively perceived stress. Methylation changes were observed in four genes: COMT, FST, FKBP51, and MAOA. We conclude that MBSR may be a useful supplementary therapy to mitigate the deleterious effects of PCOS on mental health.

Jerusalem artichoke is crop with the possibility of a wide using. The main nutrient of this plant is a non-starch polysaccharide inulin, which is considered like prebiotic. Young rabbits after weaning lose an immunity obtained from colostrum; are stressed by weaning; pre-moult; the permanent incisors are growing and a starch digestion by an enzyme amylase is not fully developed. The aim of this study was to evaluate the effect of the dietary supplement of dried Jerusalem artichoke tuber and tops (the sources of non-starch saccharide) on growth performance and health status of broiler rabbits after weaning in view of prices and efficiency of feeding mixtures. The test was realized through experiments with a different percentage of Jerusalem artichoke tuber (25%, 10%) and top (30%, 20%, 10%). Every experiment was composed of 2 groups – the control group without a supplement of Jerusalem artichoke and an experimental group with a different percentage of Jerusalem artichoke tuber and top. Background A necessary precondition for a successful microwave hyperthermia (HT) treatment delivered by phased arrays is the ability of the HT applicator to selectively raise the temperature of the entire tumor volume. SAR-based treatment plan (HTP) optimization methods exploit the correlation between specific absorption rate (SAR) and temperature increase in order to determine the set of steering parameters for optimal focusing, while allowing for lower model complexity. Several cost functions have been suggested in the past for this optimization problem. However, their correlation with high and homogeneous tumor temperatures remains sub-optimal in many cases. Previously, we proposed the hot-to-cold spot quotient (HCQ) as a novel cost function for SAR-based HTP optimization and showed its potential to address these issues.Materials and methods In this work, we validate the HCQ on a standard ESHO patient repository within single and multi-frequency contexts. We verify its correlation with clinical SAR and temperature indexes, and compare it to HTPs obtained using a commonly accepted cost-function for SAR-based HTP (hot-spot to target quotient, HTQ).Results and discussion The results show that low HCQ values produce better SAR (TC50, TC75) and temperature metrics (T50, T90) than HTQ in most patient models and frequency settings. For the deep-seated tumors, the correlation between the clinical indicators and 1/HCQ is more favorable than the correlation exhibited by 1/HTQ.Conclusion The validation confirms the ability of HCQ to promote target coverage and hot-spot suppression in SAR-based HTP optimization, resulting in higher SAR and temperature indexes for deep-seated tumors.

Summary: Background: People with Down syndrome (DS) show clinical signs of accelerated ageing. Causative mechanisms remain unknown and hypotheses range from the (essentially untreatable) amplified-chromosomal-instability explanation, to potential actions of individual supernumerary chromosome-21 genes. The latter explanation could open a route to therapeutic amelioration if the specific over-acting genes could be identified and their action toned-down. Methods: Biological age was estimated through patterns of sugar molecules attached to plasma immunoglobulin-G (IgG-glycans, an established “biological-ageing-clock”) in n = 246 individuals with DS from three European populations, clinically characterised for the presence of co-morbidities, and compared to n = 256 age-, sex- and demography-matched healthy controls. Isogenic human induced pluripotent stem cell (hiPSCs) models of full and partial trisomy-21 with CRISPR-Cas9 gene editing and two kinase inhibitors were studied prior and after differentiation to cerebral organoids. Findings: Biological age in adults with DS is (on average) 18.4–19.1 years older than in chronological-age-matched controls independent of co-morbidities, and this shift remains constant throughout lifespan. Changes are detectable from early childhood, and do not require a supernumerary chromosome, but are seen in segmental duplication of only 31 genes, along with increased DNA damage and decreased levels of LaminB1 in nucleated blood cells. We demonstrate that these cell-autonomous phenotypes can be gene-dose-modelled and pharmacologically corrected in hiPSCs and derived cerebral organoids. Using isogenic hiPSC models we show that chromosome-21 gene DYRK1A overdose is sufficient and necessary to cause excess unrepaired DNA damage. Interpretation: Explanation of hitherto observed accelerated ageing in DS as a developmental progeroid syndrome driven by DYRK1A overdose provides a target for early pharmacological preventative intervention strategies. BackgroundThe review addresses the knowledge gap concerning the diagnostic value and clinical utility of tumor-educated platelets (TEPs) in adult patients with lung cancer.MethodsWe searched twelve databases: PubMed, CENTRAL, EMBASE, CINAHL, MEDLINE, Scopus, ProQuest, MedRxiv, BioRxiv, SSRN, Clinicaltrials.gov, and CNKI up to 24 March 2023, to include any diagnostic study regarding TEPs and LC. TEPs diagnostic value was evaluated from pooled sensitivity and specificity, positive likelihood ratio (PLR), negative likelihood ratio (NLR), diagnostic odds ratio (DOR), and the area under the curve (AUC). QUADAS 2 was used to assess the risk of bias. Heterogeneity analysis was assessed using the receiver operating characteristic (ROC) plane, Galbraith plot, bivariate boxplot, sensitivity analysis, and meta-regression. TEPs clinical utility was evaluated from Fagan’s nomogram.Results44 reports from 10 studies, including 7,858 events and 6,632 controls, were analyzed. The pooled sensitivity, specificity, PLR, NLR, and DOR were 0.80 (95% CI 0.79–0.80), 0.69 (95% CI 0.69–0.70), 2.92 (95% CI 2.50–3.41), 0.26 (95% CI 0.21–0.32), and 12.1 (95% CI 8.61–16.76), respectively. In addition, the AUC of the Summary ROC curve was 0.85 (95% CI: 0.81-0.88). The overall risk of bias was low. Heterogeneity may result from cancer stage, cancer control, measuring equipment, and RNA types across studies. There was no apparent publication bias (p=0.29) with significant positive (79%) and negative (22%) post-test probability, according to Deeks funnel plot asymmetry test and Fagan’s nomogram.ConclusionTEPs could be a moderately effective candidate biomarker for LC diagnosis.

Text-based chatbots are implemented in the financial sector to enhance the relationship between the customer and services provided by the sector, and also to address external challenges and customer requirements. The chatbot technology in the financial sector serves to examine customers' frequently asked questions and the representation of the process using machine learning. In light of this, this study presents a comprehensive systematic literature review of articles focused on text-based chatbots in the financial sector. It describes the understanding of chatbots in the financial sector in terms of implementation, adoption intention, attitude toward use and acceptance; it also describes how people experience chatbots, specifically in terms of perception, expectation and trust, as well as how they are engaging and emotionally motivated; management of the security and privacy vulnerabilities of the chatbots; and identifies the potential strategies that can hinder the efficient, successful evolution of chatbots in the financial sector. Finally, the main findings regarding the use of text chatbots in the financial sector are presented; additionally, the open issues in current research are highlighted and a number of research opportunities that can be pursued in the future are suggested.

Abstract Culicoides biting midges (Diptera: Ceratopogonidae) are the major vectors of bluetongue, Schmallenberg, and African horse sickness viruses. This study was conducted to survey Culicoides species in different parts of Ethiopia and to develop habitat suitability for the major Culicoides species in Ethiopia. Culicoides traps were set in different parts of the country from December 2018 to April 2021 using UV light Onderstepoort traps and the collected Culicoides were sorted to species level. To develop the species distribution model for the two predominant Culicoides species, namely Culicoides imicola and C. kingi, an ensemble modeling technique was used with the Biomod2 package of R software. KAPPA True skill statistics (TSS) and ROC curve were used to evaluate the accuracy of species distribution models. In the ensemble modeling, models which score TSS values greater than 0.8 were considered. Negative binomialregression models were used to evaluate the relationship between C. imicola and C. kingi catch and various environmental and climatic factors. During the study period, a total of 9148 Culicoides were collected from 66 trapping sites. Of the total 9148, 8576 of them belongs to seven species and the remaining 572 Culicoides were unidentified. The predominant species was C. imicola (52.8%), followed by C. kingi (23.6%). The abundance of these two species was highly influenced by the agro-ecological zone of the capture sites and the proximity of the capture sites to livestock farms. Climatic variables such as mean annual minimum and maximum temperature and mean annual rainfall were found to influence the catch of C. imicola at the different study sites. The ensemble model performed very well for both species with KAPPA (0.9), TSS (0.98), and ROC (0.999) for C. imicola and KAPPA (0.889), TSS (0.999), and ROC (0.999) for C. kingi. Culicoides imicola has a larger suitability range compared to C. kingi. The Great Rift Valley in Ethiopia, the southern and eastern parts of the country, and the areas along the Blue Nile and Lake Tana basins in northern Ethiopia were particularly suitable for C. imicola. High suitability for C. kingi was found in central Ethiopia and the Southern Nations, Nationalities and Peoples Region (SNNPR). The habitat suitability model developed here could help researchers better understand where the above vector-borne diseases are likely to occur and target surveillance to high-risk areas.

Accumulating evidence indicates that most primary Wharton’s jelly mesenchymal stem cells (WJ-MSCs) therapeutic potential is due to their paracrine activity, i.e., their ability to modulate their microenvironment by releasing bioactive molecules and factors collectively known as secretome. These bioactive molecules and factors can either be released directly into the surrounding microenvironment or can be embedded within the membrane-bound extracellular bioactive nano-sized (usually 30–150 nm) messenger particles or vesicles of endosomal origin with specific route of biogenesis, known as exosomes or carried by relatively larger particles (100 nm–1 μm) formed by outward blebbing of plasma membrane called microvesicles (MVs); exosomes and MVs are collectively known as extracellular vesicles (EVs). The bioactive molecules and factors found in secretome are of various types, including cytokines, chemokines, cytoskeletal proteins, integrins, growth factors, angiogenic mediators, hormones, metabolites, and regulatory nucleic acid molecules. As expected, the secretome performs different biological functions, such as immunomodulation, tissue replenishment, cellular homeostasis, besides possessing anti-inflammatory and anti-fibrotic effects. This review highlights the current advances in research on the WJ-MSCs’ secretome and its prospective clinical applications.

Abstract Background Many regulatory circuits in plants contain steps of targeted proteolysis, with the ubiquitin proteasome system (UPS) as the mediator of these proteolytic events. In order to decrease ubiquitin-dependent proteolysis, we inducibly expressed a ubiquitin variant with Arg at position 48 instead of Lys (ubK48R). This variant acts as an inhibitor of proteolysis via the UPS, and allowed us to uncover processes that are particularly sensitive to UPS perturbation. Results Expression of ubK48R during germination leads to seedling death. We analyzed the seedling transcriptome, proteome and metabolome 24 h post ubK48R induction and confirmed defects in chloroplast development. We found that mutations in single genes can suppress seedling lethality, indicating that a single process in seedlings is critically sensitive to decreased performance of the UPS. Suppressor mutations in phototropin 2 (PHOT2) suggest that a contribution of PHOT2 to chloroplast protection is compromised by proteolysis inhibition. Conclusions Overall, the results reveal protein turnover as an integral part of a signal transduction chain that protects chloroplasts during development.

On a daily basis, a wide range of materials including inorganic nanoparticles (NPs) inadvertently find their way into the environment. Meanwhile, intentionally used NPs, such as the new generation of nanofertilizers (NFs) are designed to enhance agronomic production. However, their physicochemical properties and not-so-well understood effects raise potential risks to the plant reproductive cycle, specifically pollen development, a subject largely absent in academic research. Even slight contamination, deformation, or aberration of pollen could have enormous impacts on the ecosystem. Thus, our objective was to evaluate the influence of various metal-based NPs on sunflower pollen morphology and its yield. Nano-formulations were applied during the 2019–2021 agronomic seasons on two sunflower hybrids, Neostar and Edison, in Dolná Malanta, near Nitra, Slovak Republic. Pollen morphology findings indicated that conventional ZnSO4 had the most positive impact on the size of pollen grains compared to ZnO-NPs, Fe3O4-NPs, and the NP-free control. Gold-NPs on SiO2 mesoporous silica (AuSi-NPs) showed a statistically insignificant impact, while the use of TiO2-NPs in agriculture remained a topic of debate. Surprisingly, pollen characteristics did not fully correspond to crop yields. Despite causing a reduction in pollen grain size, the TiO2-NPs consistently showed the highest yield compared to other variants. Employing low concentrations of NFs did not notably alter pollen morphology, reinforcing our commitment to eco-friendly, precise, and sustainable agriculture.

Abstract Antennas are the building block of radiative hyperthermia (HT) applicators. This study proposes a compact UWB antenna specifically tailored to meet the requirements for deep HT array applicators. The proposed Open Ridged‐Waveguide (ORWG) antenna, which is an adaptation of a double‐ridged horn antenna, operates over the frequency band of 400–800 MHz. It was experimentally assessed as a single element. The quality metrics considered were reflection coefficient, penetration depth, effective field size (EFS), and mutual coupling. The design shows a 75.5% fractional bandwidth with a reflection coefficient measured to be below −10 dB from 367 up to 820 MHz. The EFS is greater than the physical dimensions of the 3‐by‐4 cm aperture. The mutual coupling between two adjacent elements in the array, measured in a flat phantom arrangement, is lower than −30 dB throughout the entire band. The antenna's performance was further tested in two deep HT scenarios in order to assess the mutual coupling and focussing abilities while in the array configuration. To this end, phased array applicators consisting of 10 and 16 ORWG antennas were simulated in CST, and the results are presented for a homogeneous cylindrical muscle phantom and a realistic patient model, respectively. The good agreement between the simulated and measured results suggests that the antenna can be successfully used for HT.

The sigma H (σΗ) and sigma E (σE) subunits of Corynebacterium glutamicum RNA polymerase belong to Group 4 of sigma factors, also called extracytoplasmic function (ECF) sigma factors. Genes of the C. glutamicum σΗ regulon that are involved in heat and oxidative stress response have already been defined, whereas the genes of the σE regulon, which is involved in cell surface stress response, have not been explored until now. Using the C. glutamicum RES167 strain and its derivative C. glutamicum ΔcseE with a deletion in the anti-σΕ gene, differential gene expression was analyzed by RNA sequencing. We found 296 upregulated and 398 downregulated genes in C. glutamicum ΔcseE compared to C. glutamicum RES167. To confirm the functional link between σΕ and the corresponding promoters, we tested selected promoters using the in vivo two-plasmid system with gfpuv as a reporter gene and by in vitro transcription. Analyses with RNAP+σΗ and RNAP+σΕ, which were previously shown to recognize similar promoters, proved that the σΗ and σE regulons significantly overlap. The σE-controlled genes were found to be involved for example in protein quality control (dnaK, dnaJ2, clpB, and clpC), the regulation of Clp proteases (clgR), and membrane integrity maintenance. The single-promoter analyses with σΗ and σΕ revealed that there are two groups of promoters: those which are exclusively σΗ-specific, and the other group of promoters, which are σΗ/σE-dependent. No exclusively σE-dependent promoter was detected. We defined the consensus sequences of exclusively σΗ-regulated promotors to be −35 GGAAt and − 10 GTT and σΗ/σE-regulated promoters to be −35 GGAAC and − 10 cGTT. Fifteen genes were found to belong to the σΗ/σΕ regulon. Homology modeling showed that there is a specific interaction between Met170 in σΗ and the nucleotides −31 and − 30 within the non-coding strand (AT or CT) of the σΗ-dependent promoters. In σE, Arg185 was found to interact with the nucleotides GA at the same positions in the σE-dependent promoters.

Untuk itu perlu dibahas untuk diketahui perluasan makna dalam pemberian kepemilikan HMSRS, peralihan hak serta penjaminan yang diberikan kepada orang asing. Metode penelitian yang digunakan yaitu penelitian hukum normatif dengan pendekatan undang-undang dan konseptual. Penelitian ini mempunyai isu hukum yaitu akibat hukum diberikan hak milik dan peralihan atas HMSRS kepada orang asing. Hasil penelitian ini, yaitu bahwa dengan diberikannya HMSRS kepada Orang Asing, dapat mengurangi kesejahteraan rakyat dalam mendapatkan tempat tinggal. The purpose of this research is to develop mobile learning-based e-comic social studies teaching materials to pique the interest of fifth-graders in elementary school. The students in this study are from the fifth grade at Riyadh El Jannah Islamic School. The ADDIE model is used to conduct research development or research and development in this study. However, it only reaches the development stage due to circumstances that prevent it from completing all of the stages. This research used a questionnaire for expert evaluation as well as interviews with teachers and students. This study employed descriptive quantitative and descriptive qualitative data analysis techniques. The quantitative descriptive method is used to process expert data and student response questionnaires, while the qualitative descriptive method is used to describe corrective comments and validators' inputs. The feasibility test for mobile learning-based e-comic teaching materials received an average percentage of 93.78 percent, making it a worthy category. The final scores for product trials on teacher and student responses were 92 percent and 94.57 percent, respectively. Based on the findings of the data analysis, it can be concluded that the developed mobile learning-based e-comic teaching materials are suitable for classroom use
