Parecoxib plays an important role in inhibition of human cancer. However, the effect of parecoxib on esophageal squamous cell carcinoma (ESCC) is still not well known. The purpose of this study was to investigate the effect of parecoxib on ESCC and its underlying mechanism.RNA-sequence analysis was performed to identify functional alterations and mechanisms. Cell cycle, proliferation, invasion, and migration were assessed using flow cytometry, CCK-8 assay, colony formation, transwell, and wound healing assays. Extracellular matrix (ECM) degradation was detected by substrate gel zymography and 3D cell culture assay. Western blotting was used to detect parecoxib-dependent mechanisms involving cell cycle, proliferation, invasion, and migration. Tumor formation in vivo was detected by mouse assay.Functional experiments indicated that parecoxib induced ESCC cell cycle arrest in G2 phase, and inhibited cell proliferation, invasion, and migration in vitro. Western blotting revealed that parecoxib downregulated the phosphorylation levels of AKT and PDK1, as well as the expression of the mutant p53, cyclin B1, and CDK1, while upregulating p21waf1. Parecoxib inhibited matrix metalloproteinase-2 (MMP2) secretion and invadopodia formation, which were related to ECM degradation. Furthermore, we found that parecoxib suppressed ESCC growth in heterotopic tumor models.Parecoxib inhibits ESCC progression, including cell cycle, proliferation, invasion, and migration, via the PDK1-AKT signaling pathway.

Background As a class of the opioid receptors, the kappa opioid receptor (KOR) has been verified to be a potential biomarker and therapeutic target for human malignant tumors. However, a thorough understanding of whether KOR affects progression of esophageal squamous cell carcinoma (ESCC) is still lacking. This study focused on exploring the effect of knocking down KOR in ESCC and its underlying mechanism. Methods Bioinformatics analysis was used to compare the different expression level of OPRK1 (KOR gene) in tumor and adjacent normal tissues, and predict the relationship between KOR expression and overall survival. RNA-sequence analysis was performed to detect the altered functions and mechanisms after down regulating KOR. The in vitro and in vivo assays were used to detect the effects of down-regulated KOR on cell proliferation, migration and invasion. Substrate gel zymography and 3D cell culture assays were used to find the effect of KOR knockdown on the degradation of extracellular matrix (ECM), and immunefluorescence was performed to detect the altered cytoskeleton. Western blotting and immunohistochemistry were used to explore the underlying mechanism pathway. Results Bioinformatics analysis revealed that the expression of OPRK1 was lower in tumor tissue than that in adjacent normal tissues, and lowered expression of KOR was associated with poorer overall survival. The in vitro assays demonstrated that down-regulation of KOR enhanced ESCC proliferation, metastasis and invasion. Western blotting revealed that down-regulation of KOR could activate PDK1-AKT signaling pathway, which actively regulated the cancer progression. Down-regulation of KOR enhanced the formation of invadopodia, secretion of matrix metalloproteinase-2 (MMP2) and rearrangement of cytoskeleton, which were positively related with the invasion of ESCC. KOR knockdown enhanced the tumor invasion and elevated the AKT phosphorylation in nude mice. The AKT kinase inhibition could reverse the effect of down-regulation of KOR. Conclusion KOR might act as a tumor suppressor in ESCC and down-regulation of KOR could enhance the ESCC tumor phenotype. Graphical abstract

Background The growing burden of end-stage renal disease (ESRD) has been a great challenge to the health care system of China. However, the exact epidemiological data for ESRD in China remain unclear. We aimed to investigate the epidemiology of ESRD treated by renal replacement therapy (RRT) in Nanjing based on analysing ten-year data of Nanjing three million insurance covered population. Methods Using the electronic registry system of Urban Employee Basic Medical Insurance (UEBMI), we included all subjects insured by UEBMI in Nanjing from 2005 to 2014 and identified subjects who developed ESRD and started RRT in this cohort. Results The UEBMI population in Nanjing increased from 1,301,882 in 2005 to 2,921,065 in 2014, among which a total of 5,840 subjects developed ESRD and received RRT. Over the 10-year period, the adjusted incidence rates of RRT in the UEBMI cohort gradually decreased from 289.3pmp in 2005 to 218.8pmp in 2014. However, the adjusted prevalence rate increased steadily from 891.7pmp in 2005 to 1,228.6pmp in 2014. The adjusted annual mortality rate decreased from 138.4 per 1000 patient-years in 2005 to 97.8 per 1000 patient-years in 2014. The long-term survival rate fluctuated over the past decade, with the 1-year survival rate ranging from 85.1% to 91.7%, the 3-year survival rate from 69.9% to 78.3% and the 5-year survival rate from 58% to 65.4%. Conclusion Nanjing is facing an increasing burden of ESRD with its improvement of medical reform. The ten-year complete registry data on RRT in urban employees in Nanjing provided a unique opportunity to understand the real threat of ESRD confronting China during its process of health care transition.

Background There are limited data on the trends of incidence or prevalence of end stage renal disease (ESRD) in China. To assist in future planning for the ESRD program, the trends of incidence, prevalence and health care costs were analyzed and forecasted to the year 2025 by modeling of historical data from 2004 through 2014. Methods Nanjing urban employee basic medical insurance (NJUEBMI) data were obtained from the Nanjing Medical Insurance Information System from 2004 to 2014. The time series forecasting system in SAS 9.4 was used. Each variable was independently forecasted by the fittest model, which was selected automatically or manually. Results The forecasting models demonstrated mean percent errors of −2.49 to 5.62 %, relative to the observed values. The R-square values for the forecasting models ranged from 0.756 to 0.997. On the basis of trends in the historical data, the models projected that the average annual increase in the NJUEBMI population was 4.77 %, with forecasted values of 5,029,270 in 2025 (95 % CI, 4,960,423-5,098,117). The incidence and prevalence of ESRD were projected to increase by 1.19 and 1.95 % annually and were expected to reach 250.5 pmp (95 % CI, 247.7–253.3) and 1505 pmp(95 % CI, 1450–1560) by 2025. Additionally, the costs associated with ESRD were forecasted to increase at a growth rate of 5.80 % for healthcare costs and 7.25‰ for per capita medical expenses, with forecasted values of ¥600.3 million ($92.4 million) (95 % CI, 541.8–658.9) and ¥99.0 thousand ($15.2 thousand) (95 % CI, 98.6–99.3), respectively, by 2025. The incidence and prevalence of kidney transplantation were projected to decrease by 6.58 and 9.79 % annually. Conclusions These projections suggest that the incidence, prevalence, healthcare costs, and per capita medical expenses of ESRD would increase in the NJUEBMI population. They provide a basis for discussing the trends of ESRD in China and facing the challenges from the ESRD program. In this investigation, the concentration levels of hair elements of calcium, iron, and zinc were measured in pregnant women from Tianjin metropolis, China. The subjects were 93 cases of pregnant women who had been suffering from calcium, iron, or zinc deficiency judged by blood tests at the mid-term of the second trimester or early in the third trimester. Of these 93 cases, 82 subjects had their hair element levels measured when the blood tests were conducted. Then, they were supplied with mineral element nutrients of gluconic acidic zinc (noted as Zn-nutrient), gluconic acidic calcium (Ca-nutrient), or/and ferrous sulfate (Fe-nutrient) which were correspondent to the deficient element(s) for more than 2 mo before 84 subjects returned to hospital for further diagnoses and had their hair element levels measured for the second time. Finally, in the third trimester or near-parturient phase, 13 subjects had their hair element levels measured again. Except for the deficiencies of calcium, iron, or/and zinc, these subjects were all healthy without symptoms of any diseases. The concentrations of hair Ca, Fe, and Zn were measured by X-ray fluorescence (XRF) spectrometry. These concentrations of the three hair elements measured at three different times were statistically analyzed. From the analyses, it was clear that hair concentrations of Ca, Fe, and Zn could reflect the effects of supplementation. Also, the mutual resistant effects among Ca-, Fe-, and Zn-nutrients were revealed. However, by appropriate combination, the mutual resistant effects could be depressed and mutual promotional effects might be enhanced. Finally, it could be concluded that mineral element deficiencies might be convalesced by adequate compensations of mineral element nutrients.

In this paper, a new method of quantitative description of earthquake precursors is proposed; by this method, the precursory pattern of time series can be quantitatively described with a two-dimensional matrix. On this basis, a method of automatic recognition or automatic acquirement of precursory pattern, called simply the AA method, is put forward. Then, taking North China region as an example, various seismological precursors such as the frequency, energy, b-value, etc. and various nonlinear parameter precursors such as the capacity dimension, information dimension, correlation dimension, Hurst index and its difference, etc. are analyzed and the 8 time series so obtained are recognized automatically using the proposed precursory pattern and AA method. Besides, C-method tests and very rigorous HF (history and future) tests are made. The result shows that the R-value of prediction efficacy assessment is fairly high.

The preliminary result of this work was obtained in 1992. At Consulting Meeting for Seismic Trends of China in Jan. 1993, we handed in a report and predicted that “Until now, the average magnitude curve\(\bar M\) haven’t gotten risen again after a fall, and the value ofH is under 0.4, so no alarm is made by each of these two. We predicted that, seismicity of the Chinese mainland in 1993 may be at the same level as in 1991–1992, or a little higher than it, but mustn’t be very high; this is to say that no earthquake withMs⩾7.2 will occur in the Chinese mainland in 1993.”

In this paper, four pattern recognition methods are set forth. Based on plane projection of samples and analysis of typical samples along with the few pattern recognition methods, the PTR algorithm for recognizing various structure samples is proposed. Also two examples are given and these show the PTR algorithm is effective.

Near-infrared spectroscopy (NIRs) is a novel measuring technology in neuroscience. It can be used to non-invasively measure the relative concentrations of oxy-hemoglobin (HbO2) and deoxy-hemoglobin (HbR) which can reflect the information related to cerebral blood volume and cerebral oxygen saturation. Therefore, it has the potential for noninvasive monitoring of cerebra ischemia. However, it still lacks reliable physiological information in cerebra ischemia, and its system volume is usually bulky. In this study, a portable, wireless multi-channel near-infrared spectroscopy for monitoring middle cerebral artery occlusion is proposed. We attempt to miniaturize NIRs system, and find meaningful information related to cerebra ischemia from the relative concentrations of HbO2 and HbR measured by using the proposed NIRs. We also hope the proposed NIRs system can provide a system prototype for monitoring middle cerebral artery occlusion in the future.

In this paper, the information function of sample distribution that showing the distribution situation of samples is proposed, then two plane projection methods that projecting samples in a multi-dimensional space to a plane is also proposed. Based on various data structures encountered in seismology, four typical models of sample structures are established in this paper, as well the best classifying methods of various typical samples are obtained.

A pulse oximeter is a noninvasive instrument used in clinical medicine to monitor arterial oxygen saturation. Fingertip-type pulse oximeters are popular, but their inconvenience for long-term monitoring in daily life means that other types of wearable pulse oximeters, such as a ring-type reflection pulse oximeter, needs to be developed. However, designing a ring-type pulse oximeter for measuring the oxygen saturation is difficult due to the complex tissue architecture of the finger base. This study used human tissue simulations to evaluate the practicability of a ring-type reflection pulse oximeter design. Moreover, given that the collection of diffusely reflected light can be enhanced by using a parabolic reflector, the efficacy of a ring-type reflection pulse oximeter with a parabolic reflector was also evaluated.

The hair and serum levels of calcium, iron, copper, and zinc levels were measured in a group of 70 healthy pregnant women and in 66 age-matched healthy controls living in the Tianjin city of the People’s Republic of China. The study subjects were classified into three subgroups according to gestational age. The hair concentrations were measured by X-ray fluorescence spectrometry, whereas those in sera were established by atomic absorption spectrometry. The hair concentrations of Ca, Fe, Cu, and Zn in the three groups of gravida were lower or significantly lower than those in controls. In sera, the differences did not show statistical significance in most cases. A deficiency of calcium was observed in subjects in the last trimester of gestation. This reinforces the importance of supplementation with calcium during pregnancy.

This article describes a study where the recovery of nasopharyngeal cancer (NPC) patients was traced by trace elements (TEs) in hair of these patients. These patients whose concentrations of TEs in hair were studied are volunteers who were divided into three groups B, C, and D. Group B was made up of volunteers who had just been diagnosed as having NPC, group C and group D were made up of volunteers who had been diagnosed as having NPC after 3 and 6 mo, respectively, of treatment. For comparison, a control group, namely group A, which was made up of volunteers from healthy persons was added to this study. By implementation of statistic pattern recognition methods, it has been found that the concentrations of TEs in hair can remarkably reflect different recovery phases of NPC patients.

This article describes a study where the trace elements (TEs) of four groups of volunteers were analyzed. The volunteers were divided into four groups A, B, C, and D. Group A was made up of healthy subjects, group B was made up of volunteers who had just been diagnosed as having naso-pharyngeal cancer (NPC), group C was made up of volunteers who had been diagnosed as having NPC after 3 mo of treatment, and group D was made up of volunteers who had been diagnosed as having NPC after 6 mo of treatment. In all groups, 11 trace elements in hair were analyzed. Our study shows that the concentrations of zinc (Zn), copper (Cu), manganese (Mn), and cobalt (Co) in group B are less than that of group A, whereas the concentration of titanium (Ti) in group B is higher. Our results also show that the difference in the level of TEs between group A and the other groups is dramatically decreased as the time of the treatment is increased. This may be a reflection of successful treatment.
