Abstract

Background: Human constitution, the fundamental basis of oriental medicine, is categorized into different patterns for a particular disease according to the physical, physiological, and clinical characteristics of the individuals. Obesity, a condition of metabolic disorder, is classified according to six patterns in oriental medicine, as follows: spleen deficiency syndrome, phlegm fluid syndrome, yang deficiency syndrome (YDS), food accumulation syndrome (FAS), liver depression syndrome (LDS), and blood stasis syndrome. In oriental medicine, identification of the disease pattern for individual obese patients is performed on the basis of differentiation in obesity syndrome index and, accordingly, personalized treatment is provided to the patients. The aim of the current study was to understand the obesity patterns in oriental medicine from the genomic point of view via determining the gene expression signature of obese patients using peripheral blood mononuclear cells as the samples.Methods: The study was conducted in 23 South Korean obese subjects (19 female and four male) with BMI ≥25 kg/m(2). Identification of oriental obesity pattern was based on the software-guided evaluation of the responses of the subjects to a questionnaire developed by the Korean Institute of Oriental Medicine. The expression profiles of genes were determined using DNA microarray and the level of transcription of genes of interest was further evaluated using quantitative real-time PCR (qRT-PCR).Results and Conclusion: Gene clustering analysis of the microarray data from the FAS, LDS, and YDS subjects exhibited disease pattern-specific upregulation of expression of several genes in a particular cluster. Further analysis of transcription of selected genes using qRT-PCR led to identification of specific genes, including prostaglandin endoperoxide synthase 2, G0/G1 switch 2, carcinoembryonic antigen-related cell adhesion molecule 3, cystein-serine-rich nuclear protein 1, and interleukin 8 receptor, alpha which were highly expressed in LDS obesity constitution. Our current study can be considered as a valuable contribution to the understanding of possible explanation for obesity pattern differentiation in oriental medicine. Further studies can address a novel possibility that the genomic and oriental empirical approaches can be combined and implemented in systematic and synergistic development of personalized medicine. 49. ESL learner's self-efficacy and language anxiety in computer-networked interaction

Abstract

This study investigated how two different interaction modes, computernetworked versus face-to-face modes, would affect ESL learners self-perceptions of efficacy and anxiety about learning English. From a social constructivist perspective, it was hypothesized that language learners self-perceptions of efficacy, pertinent emotional states, and corresponding performance would vary according to different learning contexts. Data were collected over one semester from two sections of ESL classes taught by one teacher using both quantitative and qualitative methods. As to the quantitative methods, four self-efficacy and four anxiety self-report questionnaires were administered throughout the semester to measure students emotional states in three different contexts, general, oral and computer discussions. In addition, their oral and computer discussions were recorded six times in the semester and analyzed. For qualitative analysis, students were interviewed and class sessions were observed. Correlation analysis identified varying levels of strength of relationship between self-efficacy and anxiety scores in each context because of different characteristics of the two constructs. In addition, the results of repeated measures contrasts suggested the potential for computer discussion to increase the selfefficacy of students for their oral discussion. According to the analysis of interview data, the participants culture group influenced the emotional experiences differently in the two different contexts of oral and computer discussions. The analysis of discussion data revealed increasing levels of participation in the computer discussion over time by the students who were considered reticent in class although perhaps at the expense of language quality. Most importantly, the analysis of interview data endorsed positive effects of computer discussion on English language learning in terms of students positive attitudes and favorable feelings for the learning context. Finally, the results of comparison between the ESL in this study and EFL students from Cheng (1998) provided a piece of evidence about how the learners self-perceptions were embedded in their real life and larger social contexts.

50. Metagenomics Analysis Reveals Unique Gut Microbiota Signature of Slow-Transit Constipation.

Abstract

Introduction: Altered gut microbiota may play a role in slow-transit constipation (STC). We conducted a study of gut microbiota composition and functionality in STC using metagenomic analyses., Methods: We assembled a clinical cohort of 24 patients with STC physiology age- and sex-matched to 24 controls. We performed shotgun metagenomic sequencing followed by prediction of metabolite composition from functional profiles., Results: In a middle-aged (mean 55.3 years), predominantly female cohort, there were no significant differences in α-diversity indices, but permutational multivariate analysis of variance analysis showed significant between-group differences (R 2 = 0.050, P < 0.001) between STC patients and controls. Gordonibacter pamelaeae , Bifidobacterium longum , Firmicutes bacterium co-abundance gene group 94, and Anaerotruncus colihominis were more abundant in STC, whereas Coprococcus comes and Roseburia intestinalis were more abundant in controls. Gut-derived metabolites varying in STC relative to controls were related to bile acid and cholesterol metabolism., Discussion: We found a unique metagenomic and metabolomic signature of STC., (Copyright © 2024 The Author(s).
