The objective of this study was to determine the anatomical morphology and histology of the intestineof water monitor (Varanussalvator). Intestine organ from an adult male water monito was used in thisstudy. Perfusion was performed to flow the 4% paraformaldehyde fixative solution through the heart.Intestine was collected, then divided into six sections, followed with dipping them into 4% paraformaldehydesolution for four days, continued histological preparations and stained with haematoxylin eosin (HE). Theanatomical morphology observation was performed before the intestine was processed into histologicalpreparations by observing the outer shape of the intestine and measuring it’slength. Morphologicalobservation was made †after processing theintestinal tissue by observing each portion of the intestinalwall structure. The results showed that transverse fold were found on the outer and inner surfaces of theintestine section I-IV, while in the V-VI part of the intestine, the folds were not observed anymore. Theintestine wall consisted of four layers, such as mucosa, submucosa, tunica muscularis, and serosa. Thevilli on intestine section I-IV were relatively longer and higher, while the intestine vili in section V-VI wererelatively short. The entire mucosa in water monitor was coated with absorptive columnar epitheliumwith goblet cells. Based on the result study, it can be concluded that the water monitor has more efficientdigestive process which can be seen morphologically from the relatively short intestinal, has transversefolds on the surface of the small intestine, and does not have cecum. The intestinal wall consists of layersi.e : mucosa, submucosa, tunica muscularis, and serosa.

The objective of this study was to determine the carbohydrate content of salivary glands lingual,sublingual, and mandibular glands of water monitor (Varanus salvator). This studywas used the lingualglands, sublingual, and mandibularof an adult male water monitor. Salivary glands were fixed by soakingin a solution of 4% paraformaldehyde for four days. Then, the tissuewas processed into routine histologicalpreparations. Salivary gland tissue was stained with alcianblue staining (AB) pH 2.5 for detecting acidiccarbohydrates and was stained with periodic acid Schiff (PAS) for detecting neutral carbohydrates. Theresults showed that the lingual and sublingual glands showed a positive reaction to AB pH 2.5 stainingand PAS with the intensity varied from weak (+) to strong intensity (+++) scattered in the cytoplasm of thecell. In the cytoplasm of cells of the mandibular glands showed negative reaction (-) to the AB pH 2.5staining but reacted positively to PAS with the intensity varied from weak (+) to strong (+++). Based onthe result of the study it can be concluded that lingual and sublingual glands of Water monitor containacidic and neutral carbohydrates while mandibular glands only contain neutral carbohydrates.

The waste problem in Aceh Besar District has been going on for quite some time now, which root cause can be tracked back to the lack of waste collection and treatment facilities compared to its residents’ waste growth. Therefore, the amount of untreated waste continues to increase. Meunasah Papeun Village as one of the villages in Aceh Besar District also faces the same problem, where the facilities and capacity of the Village Waste Management Unit (VWM) cannot keep up with the growth of residents' waste. Untreated waste ends up in illegal dumps or is burned by residents. This has reduced the comfort and quality of the village environment. In this Community Development Program (CDP), an approach was taken to reduce the waste that ended up in landfills through education and training on recycling and composting. The process sorted waste into three categories: organic, non-organic and residual waste. Some organic waste was composted and the rest became residue. Non-organic waste was sorted into recyclable and residual groups. This CDP was carried out in collaboration with Bank Sampah USK (BSU) to collect residents' recyclable waste. The residual was collected by VWM to be sent into the landfill. The residents who participated in these activities were able to cut their waste from a third to more than half. The extra money from recycling that was deposited into BSU was an additional advantage.

The presence of glycoconjugates in caput, corpus, and cauda epididymis of the Javan muntjacs was performed using lectin histochemical method. The epididymis organs were obtained from two adult male Javan muntjacs in hard antler stage and then immediately fixed in Bouin’s solution and processed in histological manner. Six types of biotinylated lectins namely Peanut agglutinin (PNA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), and Ulex europaeus agglutinin I (UEA I) were applied to detect the specific glycoconjugates in the epididymal tissue of muntjacs. The result showed the presence of glycoconjugates were detected by five types of lectins except UEA I in caput, corpus, and cauda epididymis with specific distribution and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, N-acetyl-galactosamine, N-acetyl-glucosamine, and sialic acid were stained with various intensity in epithelial cells, basal cells, stereocilia, spermatozoa, and stroma of caput, corpus, and cauda epididymis. The presence of sialic acid on the surface of spermatozoa detected by WGA are thought to be play a role on sperm protection during their maturation and transit along epididymal lumen. In conclusion, the difference of glycoconjugates distribution pattern and lectin binding in the epididymal caput, corpus, and cauda of the Javan muntjac indicates the regional differences in epididymal function in spermatozoa maturation.

This study was conducted to describe the stages of seminiferous epithelium (SE), determine the relative frequency of the stages, and identify the steps of spermatid development during spermatogenesis in the testicular tissue of Aceh bull. Seven pairs of the testicular organs of Aceh bull (Bos indicus) were used and then processed in a histological manner for staining using haematoxylin and eosin (H&E) and periodic acid-Schiff-haematoxylin (PAS-H). The stages of seminiferous tubules were examined using a tubular morphology method while spermatid development was observed based on the acrosome formation during spermatid development. Eight stages (stages I to VIII) of SE were found in the testicular seminiferous tubules of Aceh bull. Furthermore, the percentage of the relative frequency of each stage was 25.48, 15.38, 12.92, 4.74, 14.97, 10.69, 10.74, and 5.08%, respectively, with the relative frequency of premeiotic, meiotic, and postmeiotic phases being 53.78, 4.74, and 41.48%, respectively. Spermatid development from round to elongated spermatids occurred in 14 steps. Steps 1 to 7 were observed in stage I, steps 8 and 9 in stage II, steps 10 and 11 in stage III, step 12 in stage IV, step 13 in stages V and VI, and step 14 in stages VII and VIII. These findings can be used as a basis for further studies, particularly in evaluating the abnormality of the cellular composition of the seminiferous tubule in each stage of spermatogenesis and also in determining daily sperm production in Aceh bull.

This study aimed to analyze the muscle microstructure of longissimus dorsi Brahman cross (BX) cattle slaughtered at the slaughterhouse of North Aceh District. The meat samples for the study were taken from three male BX cattle aged 18-30 months with body weights ranging from 400 kg to 500 kg and the average body condition scores (BCS) of 3-4. The muscle used was the longissimus dorsi muscle. The mean (±SE) diameter and cross-sectional area (CSA) of muscle fibers and the percentage of fast fibers in the longissimus dorsi BX muscle were 71.69±1.23 µm, 4263.43±305.69 µm2 , and 48.17±8.81%, respectively. This study concluded that the longissimus dorsi muscle of the BX has relatively large muscle fibers, some of which in each fascicle are composed of fast muscle (fast fiber/type II).

The Aceh cattle are local Indonesian beef cattle that are farmed in Aceh Province. This type of cattle is one of the sources of meat for the Aceh people. This study aims to analyze the quality of two primal cuts (longissimus lumborum and semitendinosus muscle) from Aceh cattle based on the muscle microstructure characteristics and MSTN gene expression. This study used a sample of longissimus lumborum and semitendinosus muscles from 18 adult male Aceh cattle with the age of 2–2.5 years and a BCS of 3.24. Muscle samples were obtained shortly after the cattle were slaughtered in slaughterhouses in Banda Aceh and Aceh Besar districts. Muscle microstructure analysis was performed using the HE, Masson’s trichrome, and immunohistochemistry staining methods, while the MSTN gene expression analysis was performed using the qPCR method. The analysis of the physical quality of meat includes pH, meat color, fat color, cooking loss, water holding capacity, and WBSF value. The results showed that the area of LL muscle fibers was smaller than that of ST with relatively the same diameter. Both muscles were dominated by fast fibers with a percentage of 82.37% (LL muscle) and 91.80% (ST muscle). The area and composition of the type of muscle fibers are the main factors that influence the tenderness of Aceh beef. A higher distribution of collagen was found in ST muscles than in LL muscles. MSTN gene expression in both muscle types was relatively the same. Aceh cattle have large muscle fibers and are dominated by fast fibers with a high percentage, resulting in a low level of the tenderness of Aceh beef. However, the level of tenderness of Aceh beef is still in accordance with the cooking preparation of original and favorite cuisine of Aceh people.

This study aims to evaluate the economic potential of Aceh cattle from the perspective of stock farmers, traders, and consumers as the three main players in the business. This study was conducted from June to August 2018 in Banda Aceh City and Aceh Besar District, Aceh Province, Indonesia. It used related literature and a survey method that involved interviews, observations, and questionnaires from 9 stock farmers, 23 traders, and 22 consumers to obtain data. The data presented descriptively and analyzed using SWOT (strengths, weaknesses, opportunities, and threats) analysis. The results show that the strength factors of the Aceh beef business are high selling prices, easy maintenance, the good taste of meat, and that do not shrink quickly when cooked. Meanwhile, the weakness factors are the limitation of the availability of seeds, traditional methods of assessing carcass weight during the buying and selling process, and the low productivity of the cattle compared with other breeds. The factors that become opportunities include the high demand for the beef, on both weekdays and religious holidays, the selling price tends to increase each year, and the government's commitment to increase local cattle productivity through various programs. However, there are certain threats to the development of these cattle, and they include the high price of calves, competition with other more productive breeds, the existence of other cheaper sources of animal protein, and a decrease in consumer purchasing power. The difference between the value of the strength and weakness factors was 0.12, which signified a positive result, while that between the opportunity and threat factors was −0.29, which signified a negative result. Furthermore, the coordinate value from the Cartesian field shows that the development of these cattle is in quadrant IV, which means that the prospect for development is still quite high, regardless of the threat. In conclusion, Aceh cattle have a high economic potential to be developed, which needs to be strengthened through the rebranding program while at the same time maintaining the sustainability of the Aceh cattle breed in Indonesia. Meanwhile, the government is expected to supervise and regulate the trade of these cattle to provide beef at an affordable price for consumers.

The aim of this research was to determine the anatomy and histology of pancreas of water monitor lizard (Varanus salvator). Two pancreas were used in this research (a male and a female). Water monitor lizard was euthanized using chloroform and subsequently necropsied to collect the pancreas. The pancreas was washed in 0.9 % NaCl solution. Anatomical observations were carried out including location, shape, and color of pancreas, also the length and weight measurements. Then, pancreas was fixed with 4% paraformaldehyde for 7 days to proceed to histological preparation and hematoxylin eosin (HE) staining. The results of the anatomical observations showed that pancreas of water monitor lizard have two lobes, dorsal lobe (splenic lobe) and ventral lobe (duodenal lobe). Dorsal lobe which was in oval form adhered with the spleen, and ventral lobe was located in the cranial of intestines with the form of long spherical. Both of pancreas lobes were white-yellow color with the length of 1.45±0.64 cm (dorsal lobe) and 7.00±0.42 cm (ventral lobe). Weight of dorsal lobe was 0.15±0.01 g and ventral lobe was 6.35±2.30 g. The histological observations found the acinar cells, ducts, and blood vessels in both of pancreas lobes. Additionally, the similar structure of Langerhans islet only found at the ventral lobe of pancreas. In conclusion, pancreas of water monitor lizard consists of two lobes which located differently. Ventral lobe is larger than the dorsal lobe. Both lobes have similar histological structure except the Langerhans islets are only appeared in the ventral lobe.

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 µg/ml of concentration for PNA lectins and 15µg/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis. Stages of the seminiferous epithelium of the testis of the wild Javan muntjac (Muntiacus muntjak muntjak) in hard antler period were characterized based on the tubular morphology method. The number and the relative frequencies of seminiferous epithelium stages and the morphometry of germinal cell nuclei were identified microscopically. We identified eight stages of seminiferous epithelium in testicular tissue of the Javan muntjac and found that the relative frequencies of stages I to VIII were 14.87, 15.12, 17.75, 6.87, 7.37, 12.37, 13, and 12.62%, respectively. The diameter of the nuclei of germinal cells varied in each stage of seminiferous epithelium. Diplotene-stage primary spermatocytes had prominent and large nuclei ~8.97 ± 1.0 μm in stages III and IV. Pachytene primary spermatocytes appeared in most stages, except stage IV, whereas leptotene- and diplotene-stage primary spermatocytes were found in stages I and II, and III and IV, respectively. Round spermatids were observed in stages IV to VIII and in stage I but were absent in stages II and III, while elongated spermatids were observed in all stages except stage I. Our findings show that the stages of seminiferous epithelium in the Javan muntjac are similar to those found in neotropical cervids, small ruminants, and other domestic animals.

The objective of this study was to determine the correlation between spermatozoa morphometry and testosterone concentration in the epididymal caput, corpus, and cauda of aceh cattle. The epididymal sperm swab slides were stained using Williams's staining to examined the morphology and morphometry of spermatozoa whereas the level of testosterone from epididymal caput, corpus, and cauda extract was measured using enzyme-linked immunosorbent assay (ELISA). The data of sperm morphology were presented descriptively whereas the data of sperm morphometry and testosterone concentration were analyzed using one way ANOVA. Pearson correlation test was conducted to know the correlation between the total length of spermatozoa (TLS) and testosterone concentration in three parts of epididymis. The result showed that the sperm morphology in caput, corpus, and cauda of epididymis was not different statistically. The TLS of aceh cattle at caput, corpus, and cauda epididymis were 69,70±0,8 µm, 70,90±0.39 µm, and 72,98±0.74 µm respectively and statistically different (p0.05). The positive correlation (r= 0.413, p= 0.416) was found between TLS and testosterone concentration at caput whereas in corpus and cauda those correlation were negative and not different (corpus: r= -0.638, p= 0.172 and cauda: r= -0.719, p= 0,108). In conclusion, the longest-sized spermatozoa found in the cauda epididymis are mature spermatozoa where the maturation process optimally takes place in caput epididymis. Testosterone found in the epididymis of aceh cow can be used as an alternative source of testosterone.

Penelitian ini bertujuan mengetahui distribusi glycoconjugate yang terekspresi pada sel epitelium oviduk kancil (Tragulus javanicus). Dalam penelitian ini digunakan satu oviduk kancil yang berasal dari satu ekor kancil b etina dewasa berumur lebih dari satu tahun. Sampel difiksasi dengan larutan Bouin dan diproses menurut standar histologi sampai menjadi blok parafin dan dipotong dengan ketebalan 5 µm. Jenis lektin yang digunakan adalah biotinylated (Con A, PNA, RCA, UEA I, dan WGA) dengan dosis masing-masing sebanyak 15 µg/ml. Hasil penelitian diketahui bahwa glycoconjugate dengan residu gula galaktosa, glukosa, manosa, N-asetilgalaktosamin, N-asetilglukosamin, fukosa, dan asam sialat ditemukan pada bagian apikal sel epitel dan di dalam sitoplasma. Glycoconjugate dengan residu gula N-asetilgalaktosamin merupakan glycoconjugate yang paling banyak ditemukan di bagian apikal sel epitel dan di dalam sitoplasma dibandingkan dengan glycoconjugate dengan residu gula lainnya.

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak epididimis (EE) terhadap peningkatan kualitas spermatozoa kambing jantan lokal. Dalam penelitian ini digunakan 12 ekor kambing jantan lokal, berumur 1,5 tahun dengan bobot badan 10-15 kg dan dibagi atas empat kelompok (K0, KP1, KP2, dan KP3). Kelompok K0, hanya diinjeksi dengan NaCl fisiologis sedangkan kelompok KP1, KP2, dan KP3 diinjeksi EE masing-masing 1, 2, dan 3 ml/ekor selama 13 hari berturut-turut. Pada hari ke-14, dilakukan pengambilan semen kambing dengan elektroejakulator dan selanjutnya dilakukan pemeriksaan kualitas spermatozoa. Hasil penelitian menunjukkan bahwa pemberian EE dengan dosis 1 dan 3 ml/ekor EE selama 13 hari berturut-turut menyebabkan peningkatan kualitas spermatozoa dibanding kelompok kontrol. Berdasarkan hal tersebut dapat disimpulkan bahwa EE berpotensi meningkatkan kualitas spermatozoa pada kambing jantan lokal.

The data available on the female reproductive organ of mouse deer (Tragulus javanicus) is still very limited. A study was therefore conducted to investigate the distribution and the concentration of carbohydrate residues during the development of ovary follicles. An ovary at luteal phase was used in this study. Thin sections of the ovary were prepared occording to the standard methods and they were then histochemically stained with flourecnece-labelled lectins such as peanut agglutinin (PNA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), Winged bean agglutinin (WGA) and Ulex europaeus agglutinin (UEA). The result showed that changes in the distribution and the concentration of carbohydrate occured during the development of the follicle. During the preantral stage, the cytoplasm of oosit contained carbohydrate with the residues of glucosa dan mannosa. Zona pelusida contained carbohydrates with residues of glucosa, mannosa, galactosa dan N-asetylgalactosamine, whereas extracellular matrix contained carbohydrate with the residues of glucosa dan mannosa. In the antral follicle, the cyitoplasm of oocytes contained carbohydarte with the residues of galactosa dan N-asetylgalactosamine, whereas its zona pelusida, extracellular matrix and follicular fluid contained carbohydarte with the residues of fucosa, N-asetylglucosamin and cyalic acid. Diffrences in the types and the distribution pattern of carbohydrates were observed in this study, both in preantral and antral follicles.
