17. Phytoalexin-deficient mutants of Arabidopsis reveal that PAD4 encodes a regulatory factor and that four PAD genes contribute to downy mildew resistance

Abstract

We are working to determine the role of the Arabidopsis phytoalexin, camalexin, in protecting the plant from pathogen attack by isolating phytoalexin-deficient (pad) mutants in the accession Columbia (Col-0) and examining their response to pathogens. Mutations in PAD1, PAD2, and PAD4 caused enhanced susceptibility to the bacterial pathogen Pseudomonas syringae pv. maculicola strain ES4326 (PsmES4326), while mutations in PAD3 or PAD5 did not. Camalexin was not detected in any of the double mutants pad1-1 pad2-1, pad1 pad3-1 or pad2-1 pad3-1. Growth of PsmES4326 in pad1-1 pad2-1 was greater than that in pad1-1 or pad2-1 plants, while growth in pad1-1 pad3-1 and pad2-1 pad3-1 plants was similar to that in pad1-1 and pad2-1 plants, respectively. The pad4-1 mutation caused reduced camalexin synthesis in response to PsmES4326 infection, but not in response to Cochliobolus carbonum infection, indicating that PAD4 has a regulatory function. PAD1, PAD2, PAD3 and PAD4 are all required for resistance to the eukaryotic biotroph Peronospora parasitica. The pad4-1 mutation caused the most dramatic change, exhibiting full susceptibility to four of six Col-incompatible parasite isolates. Interestingly, each combination of double mutants between pad1-1, pad2-1 and pad3-1 exhibited additive shifts to moderate or full susceptibility to most of the isolates.

Abstract

Genotypic identification and pathogenicity characterization are performed on Burkholderia cepacia complex isolates from the rhizosphere of onion and organic soils in Michigan. The studies have shown that multiple Burkholderia cepacia complex species have colonized the onion rhizosphere and have the potential to cause sour skin rot disease of onion.

Abstract

Lignin is hypothesized to be a factor in the ability of cucurbits to resist fungal infection. To determine if lignin was present around sites of infection, susceptible cucumber plants and cucumber plants with systemic induced resistance were inoculated with Colletotnchum lagenanum, and the ultrastructure of plant and pathogen was examined 24 to 72 hours later. Conidia produced germ tubes with appressoria within 24 hours of inoculation on both control and induced plants. Penetration of plant epidermal cell walls occurred by 48 hours, and papillae formed at the site of penetration. Both elemental bromine and potassium permanganate were used to histochemically stain for lignin. Potassium permanganate, used together with energy dispersive X-ray microanalysis (EDS), indicated the presence of lignin in papillae and in electron-dense areas of plant cell walls directly beneath the appressoria. Using EDS, silicon was shown to be localized in the electron-dense areas of cucumber leaf cells. This is the first report of silicon in induced cucumber plants. 47. Late blight (<TOGGLE>Phytophthora infestans</TOGGLE> (Mont) De Bary) development from potato seed-pieces treated with fungicides

Abstract

Fungicides were applied as seed-piece treatments to control potato late blight, caused by Phytophthora infestans, US8, A2 biotype in controlled environment and field experiments. Efficacy of seed treatments for controlling late blight was examined under three disease development regimes simulated by artifical inoculation; (a) seed-borne infection, (b) transmission of infection resulting from spread during the seed-cutting operation, and (c) infection of foliage by aerial inoculation. Emergence of plants from the seed-borne infection was uniformly low (&lt;40%) in controlled environment and field experiments. In controlled environment experiments some of the plants that emerged from fungicide-treated seed-pieces were infected with late blight. Following exposure of tuber surfaces to P infestans, emergence rates from seed-pieces treated with formulated products that included mancozeb in the formulation were comparable to the untreated and non-inoculated control in controlled environment and field experiments. Plants that emerged from non-inoculated seed-pieces treated with fungicides that contained active ingredients known to be effective against foliar late blight had lower percentage foliar infection after inoculation than the untreated control. Leaves close to the base of the stem had fewer infections than leaves attached at the mid region of the main stem, 14 days after inoculation, in some of the controlled environment studies. In contrast, field experiments conducted under conditions conducive to late blight development showed that none of the seed treatments applied to late blight-free seed-pieces delayed the onset and severity of late blight infection. In potato production areas at risk of early season late blight, seed treatments applied to healthy seed may confer limited protection against late blight between planting and the first scheduled applications of prophylactic foliar fungicides. © 1999 Society of Chemical Industry
