The development of T follicular helper (Tfh) cells is an ongoing process resulting in the formation of various Tfh subsets. Despite advancements, the precise impact of T cell receptor (TCR) stimulation on this process remains incompletely understood. This study explores how TCR-CD3 signaling strength influences naive CD4
+ T cell differentiation into Tfh-like cells and the concurrent expression of interleukin-21 (IL-21), interleukin-4 (IL-4), and interferon-gamma (IFN-γ). Strong TCR-CD3 stimulation induces proliferation and increased IL-21 expression in Tfh-like cells, which exhibit a characteristic phenotype expressing CXCR5 and PD1. The coexpression of IL-4 and IFN-γ in IL-21-producing Tfh-like cells is controlled by the strength TCR-CD3 stimulation; low stimulation favors IL-4, while strong stimulation enhances IFN-γ secretion. Exogenous addition of the effector cytokines IL-21 and IL-4 further modulate cytokine coexpression. These findings highlight the intricate regulatory mechanisms governing cytokine production and plasticity in Tfh-like cells, providing insights into B cell response modulation. Cellular therapies with CD4+ T regulatory cells (Tregs) hold promise of efficacious treatment for the variety of autoimmune and allergic diseases as well as posttransplant complications. Nevertheless, current manufacturing of Tregs as a cellular medicinal product varies between different laboratories, which in turn hampers precise comparisons of the results between the studies performed. While the number of clinical trials testing Tregs is already substantial, it seems to be crucial to provide some standardized characteristics of Treg products in order to minimize the problem. We have previously developed reporting guidelines called minimum information about tolerogenic antigen-presenting cells, which allows the comparison between different preparations of tolerance-inducing antigen-presenting cells. Having this experience, here we describe another minimum information about Tregs (MITREG). It is important to note that MITREG does not dictate how investigators should generate or characterize Tregs, but it does require investigators to report their Treg data in a consistent and transparent manner. We hope this will, therefore, be a useful tool facilitating standardized reporting on the manufacturing of Tregs, either for research purposes or for clinical application. This way MITREG might also be an important step toward more standardized and reproducible testing of the Tregs preparations in clinical applications.

Background and purpose: There are concerns for safety regarding SARS‐CoV‐2 vaccines for patients with autoimmune neuromuscular disease. We compared daily functioning using disease‐specific patient‐reported outcome measures (PROMs) before and after SARS‐CoV‐2 vaccinations. Methods: In this substudy of a prospective observational cohort study (Target‐to‐B!), patients with myasthenia gravis (MG), chronic inflammatory demyelinating polyneuropathy (CIDP), multifocal motor neuropathy (MMN), and idiopathic inflammatory myopathy (IIM) vaccinated against SARS‐CoV‐2 were included. Surveys of daily functioning (Myasthenia Gravis Activities of Daily Living, Inflammatory Rasch‐Built Overall Disability Scale, Multifocal Motor Neuropathy Rasch‐Built Overall Disability Scale, and Health Assessment Questionnaire–Disability Index) were sent before first vaccination and every 60 days thereafter for up to 12 months. Regression models were constructed to assess differences in PROM scores related to vaccination, compared to scores unrelated to vaccination. We also assessed the proportion of patients with deterioration of at least the minimal clinically important difference (MCID) between before first vaccination and 60 days thereafter. Results: We included 325 patients (median age = 59 years, interquartile range = 47–67, 156 [48%] female sex), of whom 137 (42%) had MG, 79 (24%) had CIDP, 43 (13%) had MMN, and 66 (20%) had IIM. PROM scores related to vaccination did not differ from scores unrelated to vaccination. In paired PROMs, MCID for deterioration was observed in three of 49 (6%) MG patients, of whom none reported a treatment change. In CIDP, MCID for deterioration was observed in eight of 29 patients (28%), of whom two of eight (25%) reported a treatment change. Conclusions: SARS‐CoV‐2 vaccination had no effect on daily functioning in patients with autoimmune neuromuscular diseases, confirming its safety in these patients. [ABSTRACT FROM AUTHOR]

- Abstract
Background:Messenger RNA (mRNA) vaccines provide robust protection against SARS-CoV-2 in healthy individuals. However, immunity after vaccination of patients with multiple sclerosis (MS) treated with ocrelizumab (OCR), a B cell-depleting anti-CD20 monoclonal antibody, is not yet fully understood. Methods:In this study, deep immune profiling techniques were employed to investigate the immune response induced by SARS-CoV-2 mRNA vaccines in untreated patients with MS (n=21), OCR-treated patients with MS (n=57) and healthy individuals (n=30). Results:Among OCR-treated patients with MS, 63% did not produce detectable levels of antibodies (non-seroconverted), and those who did have lower spike receptor-binding domain-specific IgG responses compared with healthy individuals and untreated patients with MS. Before vaccination, no discernible immunological differences were observed between non-seroconverted and seroconverted OCR-treated patients with MS. However, non-seroconverted patients received overall more OCR infusions, had shorter intervals since their last OCR infusion and displayed higher OCR serum concentrations at the time of their initial vaccination. Following two vaccinations, non-seroconverted patients displayed smaller B cell compartments but instead exhibited more robust activation of general CD4+ and CD8+ T cell compartments, as indicated by upregulation of CD38 and HLA-DR surface expression, when compared with seroconverted patients. Conclusion:These findings highlight the importance of optimising treatment regimens when scheduling SARS-CoV-2 vaccination for OCR-treated patients with MS to maximise their humoral and cellular immune responses. - Abstract
SARS-CoV-2-specific CD8+ T cells recognize conserved viral peptides and in the absence of cross-reactive antibodies form an important line of protection against emerging viral variants as they ameliorate disease severity. SARS-CoV-2 mRNA vaccines induce robust spike-specific antibody and T cell responses in healthy individuals, but their effectiveness in patients with chronic immune-mediated inflammatory disorders (IMIDs) is less well defined. These patients are often treated with systemic immunosuppressants, which may negatively affect vaccine-induced immunity. Indeed, TNF inhibitor (TNFi)-treated inflammatory bowel disease (IBD) patients display reduced ability to maintain SARS-CoV-2 antibody responses post-vaccination, yet the effects on CD8+ T cells remain unclear. Here, we analyzed the impact of IBD and TNFi treatment on mRNA-1273 vaccine-induced CD8+ T cell responses compared to healthy controls in SARS-CoV-2 experienced and inexperienced patients. CD8+ T cells were analyzed for their ability to recognize 32 SARS-CoV-2-specific epitopes, restricted by 10 common HLA class I allotypes using heterotetramer combinatorial coding. This strategy allowed in-depth ex vivo profiling of the vaccine-induced CD8+ T cell responses using phenotypic and activation markers. mRNA vaccination of TNFi-treated and untreated IBD patients induced robust spike-specific CD8+ T cell responses with a predominant central memory and activated phenotype, comparable to those in healthy controls. Prominent non-spike-specific CD8+ T cell responses were observed in SARS-CoV-2 experienced donors prior to vaccination. Non-spike-specific CD8+ T cells persisted and spike-specific CD8+ T cells notably expanded after vaccination in these patient cohorts. 24. Pain and fatigue in adult patients with multiple osteochondromas: The Netherlands.

Background: Multiple Osteochondromas (MO) is a rare genetic disorder characterised by the presence of numerous benign bone tumours, known as osteochondromas. Within the spectrum of debilitating symptoms associated with MO, pain is recognized as a major problem. Interestingly, our clinical observations suggest that fatigue is also a significant concern but has merely been touched upon in MO literature. This study aims to (1) assess the level of pain and fatigue in adult patients with MO; (2) compare fatigue in MO to healthy subjects and patients with Rheumatoid Arthritis (RA); (3) identify associated variables for pain and fatigue in patients with MO. Methods: In this cross-sectional study, 353 adult MO patients completed a survey with validated questionnaires on pain, fatigue and psychosocial factors. Pain and fatigue were assessed with the Numeric Rating Scale (NRS), and fatigue was also measured with the Checklist Individual Strength (CIS). Fatigue (CIS) was compared with reference scores of healthy subjects and patients with RA, using a one-sample t-test. Multiple linear regression models for pain and fatigue were developed using a-priori selected independent variables based on a theoretical framework (ICF-model). Results: Pain was reported by 87.8% (NRS = 3.19±2.6) and fatigue by 90.4% (NRS = 4.1±2.6) of patients with MO. Fatigue scores for MO (CIS = 84.1±15.3) were significantly higher (p<0.001) compared to reference scores of healthy subjects and patients with RA. The multivariable analysis for pain provided a final regression model with six variables (R
2 = 0.445, p<0.001) of which fear avoidance beliefs and fatigue had the strongest association. For the fatigue models NRS (R2 = 0.455, p<0.001) and CIS (R2 = 0.233, p<0.001), the strongest associations were found with anxiety and depression respectively. Conclusions: Pain and fatigue are highly prevalent in patients with MO. Fatigue is significantly higher compared to healthy subjects and patients with RA. We investigate the properties (e.g. star formation rate, dust attentuation, stellar mass and metallicity) of a sample of infrared luminous galaxies at z \sim 1 via near-IR spectroscopy with Subaru-FMOS. Our sample consists of Herschel SPIRE and Spitzer MIPS selected sources in the COSMOS field with photometric redshifts in the range 0.7 < z-phot < 1.8, which have been targeted in 2 pointings (0.5 sq. deg.) with FMOS. We find a modest success rate for emission line detections, with candidate H{\alpha} emission lines detected for 57 of 168 SPIRE sources (34 per cent). By stacking the near-IR spectra we directly measure the mean Balmer decrement for the H{\alpha} and H{\beta} lines, finding a value of
= 0.51\pm0.27 for = 10^12 Lsol sources at = 1.36. By comparing star formation rates estimated from the IR and from the dust uncorrected H{\alpha} line we find a strong relationship between dust attenuation and star formation rate. This relation is broadly consistent with that previously seen in star-forming galaxies at z ~ 0.1. Finally, we investigate the metallicity via the N2 ratio, finding that z ~ 1 IR-selected sources are indistinguishable from the local mass-metallicity relation. We study the Higgs sector of the minimal supersymmetric standard model extended with vector-like quarks, at the one-loop level. The radiative corrections to the tree-level masses of the scalar Higgs bosons are calculated by including the contributions from the loops of top quark, vector-like quarks, and their scalar superpartners, for a reasonable parameter region. We find that the mass of the lightest scalar Higgs boson at the one-loop level should be larger than 85 GeV, if we take into account the negative experimental result for the Higgs search at LEP2. As the radiative corrections are calculated in some detail, we also find that the mass of the lightest scalar Higgs boson at the one-loop level is bounded from above at 280 GeV, This upper bound is increased from a previous result. It is found that there are wide regions in the parameter space that allow the strongly first-order electroweak phase transition, where the Higgs boson mass is larger than the experimental lower bound of 115 GeV, and the masses of the scalar color octet is around 200 GeV. In the minimal supersymmetric standard model (MSSM), a strongly first-order electroweak phase transition (EWPT) is only possible in a confined parameter region where one of the scalar top quarks is lighter than the top quark and the other one is as heavy as the SUSY breaking scale. If the MSSM is enlarged to accommodate vector-like quarks and their superpartners, we find that the strongly first-order EWPT is possible without requiring light scalar top quark at the one-loop level, in the limit where the lightest scalar Higgs boson of the MSSM behaves like the Higgs boson of the standard model and the other Higgs bosons are all as heavy as the SUSY breaking scale. We study the Higgs self couplings of a supersymmetric $E_6$ model that has two Higgs doublets and two Higgs singlets. The lightest scalar Higgs boson in the model may be heavier than 112 GeV, at the one-loop level, where the negative results for the Higgs search at the LEP2 experiments are taken into account. The contributions from the top and scalar top quark loops are included in the radiative corrections to the one-loop mass of the lightest scalar Higgs boson, in the effective potential approximation. The effect of the Higgs self couplings may be observed in the production of the lightest scalar Higgs bosons in $e^+e^-$ collisions at the International Linear Collider (ILC) via double Higgs-strahlung process. For the center of mass energy of 500 GeV with the integrated luminosity of 500 fb$^{-1}$ and the efficiency of 20 %, we expect that at least 5 events of the lightest scalar Higgs boson may be produced at the ILC via double Higgs-strahlung process., Comment: 21 pages, 5 figures

The possibility of explicit CP violation is studied in a supersymmetric model proposed by Dine, Seiberg, and Thomas, with two effective dimension-five operators. The explicit CP violation may be triggered by complex phases in the coefficients for the dimension-five operators in the Higgs potential, and by a complex phase in the scalar top quark masses. Although the scenario of explicit CP violation is found to be inconsistent with the experimental data at LEP2 at the tree level, it may be possible at the one-loop level. The Dine-Seiberg-Thomas model (DSTM) is the simplest version of the new physics beyond the minimal supersymmetric standard model (MSSM), in the sense that its Higgs sector has just two dimension-five operators, which are obtained from the power series of the energy scale for the new physics in the effective action analysis. We study the possibility of spontaneous CP violation in the Higgs sector of the DSTM, which consists of two Higgs doublets. We find that the CP violation may be triggered spontaneously by a complex phase, obtained as the relative phase between the vacuum expectation values of the two Higgs doublets. At the tree level, for a reasonably established parameter region, the masses of the three neutral Higgs bosons and their corresponding coupling coefficients to a pair of $Z$ bosons in the DSTM are calculated such that the results are inconsistent with the experimental constraint by the LEP data. Thus, the LEP2 data exclude the possibility of spontaneous CP violation in the DSTM at the tree level. On the other hand, we find that, for a wide area in the parameter region, the CP symmetry may be broken spontaneously in the Higgs sector of the DSTM at the one-loop level, where top quark and scalar top quark loops are taken into account. The upper bound on the radiatively corrected mass of the lightest neutral Higgs boson of the DSTM is about 87 GeV, in the spontaneous CP violation scenario. This article explores how a chemically reactive solute will disperse across mobile to immobile phase when injected into the fluid flowing within a long circular tube. To model this process, we utilized mathematical modeling, including advection-diffusion equations for flow of fluid within the tube and first-order chemical reaction equations to account for reversible and irreversible reactions on the tubes' wall. We proposed a numerical method based on an explicit finite difference scheme to solve the governing equations for the dispersion of a chemically reactive solute. We used an upwind method with a conservative representation in the diffusion component to discretize the advection-diffusion equation. To ensure the stability of our proposed numerical scheme, we computed the time step constraint condition so that the maximum principle for the discrete governing equation holds. We also verified the performance of our proposed scheme through computational results that were compared with previous studies. One of our key findings was that the depletion coefficient D
0 achieved a quasi-steady state for larger absorption rates. We also observed that the advection coefficient D1 initially increased with an increasing absorption rate, but eventually declined due to phase exchange kinetics. The dispersion coefficient D2 also decreased with a rising absorption rate due to a low-velocity gradient in the middle region. Our study showed that rapid distributions are possible under certain conditions, such as a high Damköhler number (Da ≥ 10) and a high absorption rate (Γ>5). Computational results show that the proposed scheme can be useful in developing an efficient pulmonary drug delivery system for periodic inhalation of drugs to determine the optimal frequency of injection. Background: Humoral responses after SARS-CoV-2 vaccination are greatly impaired in multiple sclerosis (MS) patients on fingolimod. Effects of repeated vaccination and infections on long-term responses are unclear. Methods: Prospective study in 60 MS patients on fingolimod measuring humoral responses after up to four vaccinations and 8 months after fourth vaccination. Results: Anti-WH1 antibody titers increased with each additional vaccination. At long-term follow-up titers increased further and most patients developed new humoral responses against the BA.1 omicron variant. Conclusion: Repeated SARS-CoV-2 vaccinations boost humoral immunity and, probably together with SARS-CoV-2 infections, induce humoral responses on the long-term in almost all patients. We find that, at the one-loop level, the spontaneous CP violation is possible in a supersymmetric standard model that has an extra chiral Higgs triplet with hypercharge Y=0. At the tree level, this triplet-extended supersymmetric standard model (TESSM) cannot have any reasonable parameter spaces for the spontaneous CP violation, because the experimental constraints on the coupling coefficient of the neutral Higgs boson to a pair of $Z$ bosons exclude them. By contrast, at the one-loop level, we find that there are experimentally allowed parameter regions, where the spontaneous CP violation may take place. Assuming that the squark masses are not degenerate, the radiative corrections due to the stop and sbottom quarks give rise to CP phases, which trigger the CP violation explicitly in the Higgs sector of the MNMSSM. The masses, the branching ratios for dominant decay channels, and the total decay widths of the five neutral Higgs bosons in the MNMSSM are calculated in the presence of the explicit CP violation. We study the scalar Higgs sector of the next-to-minimal supersymmetric standard model with an extra U(1), which has two Higgs doublets and a Higgs singlet, in the light leptophobic $Z'$ scenario where the extra neutral gauge boson $Z'$ does not couple to charged leptons. In this model, we find that the sum of the squared coupling coefficients of the three neutral scalar Higgs bosons to $ZZ$, normalized by the corresponding SM coupling coefficient is noticeably smaller than unity, due to the effect of the extra U(1), for a reasonable parameter space of the model, whereas it is unity in the next-to-minimal supersymmetric standard model.
