Iron (Fe) reduction and oxidation are important biogeochemical processes coupled to decomposition, nutrient cycling, and mineral weathering, but factors controlling their rates and spatial distribution with depth are poorly understood in terrestrial soils. In aquatic ecosystems, Fe reduction often occurs below a zone of oxic sediments. We tested an alternative conceptual model for Fe redox cycling in terrestrial soils using a deep humid tropical forest soil profile. We hypothesized that Fe reduction in anaerobic microsites scales with depth variation in labile C and Fe availability, as opposed to bulk oxygen (O2). We measured bulk O2 at multiple depths from 0.1 to 5 m quasi-continuously over 18 months and sampled soils from surface to bedrock (~7 m). Median O2 mixing ratios declined from 19.8 ± 1.2 % at 0.25 m to 16.1 ± 1.0 % at 1 m, but did not consistently decrease below 1 m, challenging a recent model of regolith development. Reduced Fe (Fe(II)) extractable in 0.5 M hydrochloric acid was greatest in 0–0.1 m soil and declined precipitously with depth, and did not correspond with visible gleying in B horizons. We observed similar depth trends in potential Fe reduction under anaerobic conditions. Depth trends in Fe(II) also closely mirrored short-term soil respiration and bulk soil C. Labile C stimulated Fe reduction at 0–0.1 m depth, whereas addition of short-range-ordered Fe oxides had no effect. Cultivable Fe-reducing bacterial abundance was four orders of magnitude greater in surface soil (0–0.1 m) than below 1 m. Although cultivable Fe oxidizing bacteria were typically also more abundant in surface soil, addition of labile C and nitrate stimulated Fe oxidizers in deep soil by two orders of magnitude under anaerobic conditions. This implies that infiltration of nitrate (and possibly C) from shallow soil water could potentially promote biotic Fe oxidation, a critical step in bedrock weathering, 7 m below. Together, these data suggest that C, Fe, and nutrient availability increase microbial Fe reduction and oxidation in surface (vs deeper) soil microsites despite high bulk O2, in contrast to the depth segregation of electron accepting processes often observed in aquatic ecosystems. Furthermore, the greatest capacity for Fe redox cycling can occur in A horizons that do not display gleying or mottling.

The importance of lignin as a recalcitrant constituent of soil organic matter (SOM) remains contested. Associations with iron (Fe) oxides have been proposed to specifically protect lignin from decomposition, but impacts of Fe-lignin interactions on mineralization rates remain unclear. Oxygen (O2) fluctuations characteristic of humid tropical soils drive reductive Fe dissolution and precipitation, facilitating multiple types of Fe-lignin interactions that could variably decompose or protect lignin. We tested impacts of Fe addition on 13C methoxyl-labeled lignin mineralization in soils that were exposed to static or fluctuating O2. Iron addition suppressed lignin mineralization to 21% of controls, regardless of O2 availability. However, Fe addition had no effect on soil CO2 production, implying that Fe oxides specifically protected lignin methoxyls but not bulk SOM. Iron oxide-lignin interactions represent a specific mechanism for lignin stabilization, linking SOM biochemical composition to turnover via geochemistry.

Humid tropical forests support large stocks of surface soil carbon (C) that exhibit high spatial variability over scales of meters to landscapes (km). Reactive minerals and organo-metal complexes are known to contribute to C accumulation in these ecosystems, although potential interactions with environmental factors such as oxygen (O2) availability have received much less attention. Reducing conditions can potentially contribute to C accumulation, yet anaerobic metabolic processes such as iron (Fe) reduction can also drive substantial C losses. We tested whether these factors could explain variation in soil C (0–10 and 10–20 cm depths) over multiple spatial scales in the Luquillo Experimental Forest, Puerto Rico, using reduced iron (Fe(II)) concentrations as an index of reducing conditions across sites differing in vegetation, topographic position, and/or climate. Fine root biomass and Fe(II) were the best overall correlates of site (n = 6) mean C concentrations and stocks from 0 to 20 cm depth (r = 0.99 and 0.98, respectively). Litterfall decreased as reducing conditions, total and dead fine root biomass, and soil C increased among sites, suggesting that decomposition rates rather than C inputs regulated soil C content at the landscape scale. Strong relationships between Fe(II) and dead fine root biomass suggest that reducing conditions suppressed particulate organic matter decomposition. The optimal mixed-effects regression model for individual soil samples (n = 149) showed that aluminum (Al) and Fe in citrate/ascorbate and oxalate extractions, Fe(II), fine root biomass, and interactions between Fe(II) and Al explained most of the variation in C concentrations (pseudo R2 = 0.82). The optimal model of C stocks was similar but did not include fine root biomass (pseudo R2 = 0.62). In these models, soil C concentrations and stocks increased with citrate/ascorbate-extractable Al and oxalate-extractable Fe. However, soil C decreased with citrate/ascorbate-extractable Fe, an index of Fe susceptible to anaerobic microbial reduction. At the site scale (n = 6), ratios of citrate/ascorbate to oxalate-extractable Fe consistently decreased across a landscape O2 gradient as C increased. We suggest that the impact of reducing conditions on organic matter decomposition and the presence of organo-metal complexes and C sorption by short-range order Fe and Al contribute to C accumulation, whereas the availability of an Fe pool to sustain anaerobic respiration in soil microsites partially attenuates soil C accumulation in these ecosystems.

It has been proposed that the large soil carbon (C) stocks of humid tropical forests result predominantly from C stabilization by reactive minerals, whereas oxygen (O2) limitation of decomposition has received much less attention. We examined the importance of these factors in explaining patterns of C stocks and turnover in the Luquillo Experimental Forest, Puerto Rico, using radiocarbon (14C) measurements of contemporary and archived samples. Samples from ridge, slope, and valley positions spanned three soil orders (Ultisol, Oxisol, Inceptisol) representative of humid tropical forests, and differed in texture, reactive metal content, O2 availability, and root biomass. Mineral-associated C comprised the large majority (87 ± 2%, n = 30) of total soil C. Turnover of most mineral-associated C (66 ± 2%) was rapid (11 to 26 years; mean and SE: 18 ± 3 years) in 25 of 30 soil samples across surface horizons (0-10 and 10-20 cm depths) and all topographic positions, independent of variation in reactive metal concentrations and clay content. Passive C with centennial-millennial turnover was typically much less abundant (34 ± 3%), even at 10-20 cm depths. Carbon turnover times and concentrations significantly increased with concentrations of reduced iron (Fe(II)) across all samples, suggesting that O2 availability may have limited the decomposition of mineral-associated C over decadal scales. Steady-state inputs of mineral-associated C were statistically similar among the three topographic positions, and could represent 10-25% of annual litter production. Observed trends in mineral-associated Δ14C over time could not be fit using the single-pool model used in many other studies, which generated contradictory relationships between turnover and Δ14C as compared with a more realistic two-pool model. The large C fluxes in surface and near-surface soils documented here are supported by findings from paired 14C studies in other types of ecosystems, and suggest that most mineral-associated C cycles relatively rapidly (decadal scales) across ecosystems that span a broad range of state factors.

Prostate cancer is one of the most common forms of cancer among American men, and worry about the disease has psychological, behavioral, and biological consequences. To better understand prostate cancer--specific worry, the authors tested a model of the interrelationships among family history of prostate cancer, perceived risk of and worry about prostate cancer, and perceived risk of and worry about other diseases. Men who attended prostate cancer-screening appointments at a general urology practice (n = 209) were given a brief anonymous self-report measure. Structural equation modeling (LISREL) results indicated: (1) perceived risk of prostate cancer mediated the relationship between family history of prostate cancer and prostate cancer worry; (2) perceived risk of other diseases increased perceived risk of prostate cancer; and (3) prostate cancer worry and increased other disease worry. Numerous predictive equations have evolved for the estimation of resting energy expenditure (REE) since it is not generally practical or feasible to use indirect calorimetry in practice settings. Recently, the Mifflin-St Jeor Equation (MSJE), derived from a population of 498 healthy subjects and published in 1990, was recommended as the standard for calculating REE by the Application of Indirect Calorimetry in ADA Evidence-Based Guides for Practice Expert Consensus Panel of the American Dietetic Association. Although no equation can replace the accuracy of a personalized indirect calorimetry measurement, this equation reportedly explained a substantial amount of the variation of the measured REE (R2 = 0.71) and provided the most accurate estimate (78% within +/-10% limit) of actual REE in the largest percentage of nonobese and obese in a defined test population. Importantly, the MSJE was derived on a diverse sample of healthy adults (N = 498), including females (n = 247) and males (n = 251), subjects aged 19 to 78 (45 +/- 14) years, and nonobese (n = 264) and obese (n = 234) individuals. This article presents 2 charts by gender from which an estimated REE can be easily derived from known height (inches) and weight (pounds) with an individual adjustment of -5 kcal/year of age. The charts are limited to the source sample from which the data were derived since the estimated REE for individuals falling within these parameters will be more accurate. This study evaluated food preferences and eating behaviors of individuals with Dravet syndrome. Patients diagnosed with Dravet syndrome were recruited, as well as a control group composed of siblings of patients with epilepsy (any form). The Food Preference Questionnaire and the Child Eating Behavior Questionnaire were completed by caregivers along with two open-ended questions regarding eating challenges. Seventy-eight participants (45 with Dravet syndrome and 33 controls) were included. Compared to controls, mean scores for food preference were lower for fruits (p = .000099), meats and fish (p = .00094), and snacks (p = .000027) in Dravet syndrome. People with Dravet syndrome also had less emotional overeating (p = .0085) and food enjoyment (p = .0012), but more slowness in eating (p = .00021) and food fussiness (p = .0064). In a subgroup analysis of only pediatric (age <18 years) patients, similar results were observed for both food preferences and eating habits. In qualitative data, caregivers most commonly reported difficulties with fixation on specific foods. This study demonstrates specific food preferences and challenging eating behaviors in individuals with Dravet syndrome. Poorly drained depressions within tile-drained croplands can have disproportionate environmental and agronomic impacts, but mechanisms controlling nutrient leaching remain poorly understood. We monitored nitrate and soluble reactive phosphorus (SRP) leaching using zero-tension soil lysimeters across a depression to upland gradient over 2 years in a corn-soybean (Zea mays L.-Glycine max [L.] Merr.) field in Iowa. We also measured stable isotopes (δ 15 N and δ 18 O) of nitrate to examine its sources and transformations. SRP concentrations peaked during winter and early spring after phosphorus (P) fertilization (mean = 3 mg P L -1 ), with highest values in the depression, and SRP was relatively stable thereafter (mean = 0.3 mg P L -1 ) irrespective of periods of high soil moisture that led to widespread iron (Fe) reduction across the field. During a near-average precipitation year, nitrate stable isotopes indicated direct leaching of fertilizer nitrate within days of application, followed by nitrification of fertilizer ammonium and several weeks of denitrification in depressional soils. Nevertheless, nitrate concentrations remained high (mean = 28 mg N L -1 ) in the depression despite strong isotopic evidence for denitrification (>48% N removal). During a wet year, nitrate concentrations were lower in the depression than upland and nitrate isotopes were highly variable, consistent with nearly complete nitrate removal by denitrification in the depression and significant denitrification in upland soils. We conclude that poorly drained depressional soils can potentially decrease nitrate leaching via denitrification under sustained wet conditions, but they inconsistently denitrify and are vulnerable to high nitrate and SRP losses when soils are not saturated, especially following fertilization., (© 2024 The Author(s). We examined how individual differences in masculine honor beliefs (MHBs) related to third-person perceptions of revenge pornography (i.e., a man distributing nude photos of his ex-girlfriend) in response to a break-up. Consistent with our hypotheses, higher levels of MHBs were associated with higher ratings of the woman as negative, higher levels of victim-blaming the woman, and higher ratings of revenge pornography as justified. Furthermore, participants perceived the woman as more at fault, and the man's behavior as more justified, when she cheated on him prior to his revenge (compared to when she did not cheat). Overall, our results suggest that both higher levels of MHBs and instances of infidelity inspire beliefs that disempower women victims. Sequence-based genetic testing identifies causative variants in ~ 50% of individuals with developmental and epileptic encephalopathies (DEEs). Aberrant changes in DNA methylation are implicated in various neurodevelopmental disorders but remain unstudied in DEEs. We interrogate the diagnostic utility of genome-wide DNA methylation array analysis on peripheral blood samples from 582 individuals with genetically unsolved DEEs. We identify rare differentially methylated regions (DMRs) and explanatory episignatures to uncover causative and candidate genetic etiologies in 12 individuals. Using long-read sequencing, we identify DNA variants underlying rare DMRs, including one balanced translocation, three CG-rich repeat expansions, and four copy number variants. We also identify pathogenic variants associated with episignatures. Finally, we refine the CHD2 episignature using an 850 K methylation array and bisulfite sequencing to investigate potential insights into CHD2 pathophysiology. Quantifying nitrate leaching in agricultural fields is often complicated by inability to capture all water draining through a specific area. We designed and tested undisturbed soil monoliths (termed "soil block mesocosms") to achieve complete collection of drainage. Each mesocosm measures 1.5 m × 1.5 m × 1.2 m and is enclosed by steel on the sides and bottom with a single outlet to collect drainage. We compared measurements from replicate mesocosms planted to corn (Zea mays L.) with a nearby field experiment with tile-drained plots ("drainage plots"), and with drainage from nearby watersheds from 2020 through 2022 under drought conditions. Annual mesocosm drainage volumes were 6.5-24.6 cm greater than from the drainage plots, likely because the mesocosms were isolated from the subsoil and could not store groundwater below the drain depth, whereas the drainage plots accumulated infiltration as groundwater. Thus, we obtained consistent nitrate leaching measurements from the mesocosms even when some drainage plots yielded no water. Despite drainage volume differences, mean flow-weighted nitrate concentrations were similar between mesocosms and drainage plots in 2 of 3 years. Mesocosm annual drainage volume was 8.7 cm lower to 16.7 cm higher than watershed drainage, likely due to lagged influences of groundwater. Corn yields were lower in mesocosms than drainage plots in 2020, but with irrigation, yields were similar in subsequent years. Mean 2020 surface soil moisture and temperature were similar between the mesocosms and nearby fields. Purpose: Evidence is growing that high salt intake is an independent risk factor for obesity, but the mechanisms are unknown. Our novel working hypothesis is that high salt intake drives cortisol production, which in turn, drives obesity. The current study aimed to demonstrate an acute cortisol response following a single high salt meal., Methods: Eight participants (age 30.5 ± 9.8 years [mean ± SD], 50% female), consumed high salt (3.82 g; 1529 mg sodium) and low salt (0.02 g; 9 mg sodium) meals in a randomized cross-over design., Results: Urinary and salivary cortisol and plasma adrenocorticotropic hormone (ACTH) demonstrated order effects. When high salt was given second, there was a peak above baseline for urinary cortisol (26.3%), salivary cortisol (9.4%) and plasma ACTH (4.1%) followed by a significant decline in each hormone (treatment*time, F[9, 18] = 2.641, p = 0.038, partial η 2  = 0.569; treatment*time, F[12, 24] = 2.668, p = 0.020, partial η 2  = 0.572; treatment*time, F[12, 24] = 2.580, p = 0.023, partial η 2  = 0.563, respectively), but not when high salt was given first (p > 0.05 for all)., Conclusion: These intriguing findings provide partial support for our hypothesis and support a need for further research to elucidate the role of high salt intake in cortisol production and, in turn, in the aetiology of obesity., Trial Registration Number: ACTRN12623000490673; date of registration 12/05/2023; retrospectively registered., (© 2024. Chemicals in the systemic circulation can undergo hepatic xenobiotic metabolism, generate metabolites, and exhibit altered toxicity compared with their parent compounds. This article describes a 2-chamber liver-organ coculture model in a higher-throughput 96-well format for the determination of toxicity on target tissues in the presence of physiologically relevant human liver metabolism. This 2-chamber system is a hydrogel formed within each well consisting of a central well (target tissue) and an outer ring-shaped trough (human liver tissue). The target tissue chamber can be configured to accommodate a three-dimensional (3D) spheroid-shaped microtissue, or a 2-dimensional (2D) cell monolayer. Culture medium and compounds freely diffuse between the 2 chambers. Human-differentiated HepaRG liver cells are used to form the 3D human liver microtissues, which displayed robust protein expression of liver biomarkers (albumin, asialoglycoprotein receptor, Phase I cytochrome P450 [CYP3A4] enzyme, multidrug resistance-associated protein 2 transporter, and glycogen), and exhibited Phase I/II enzyme activities over the course of 17 days. Histological and ultrastructural analyses confirmed that the HepaRG microtissues presented a differentiated hepatocyte phenotype, including abundant mitochondria, endoplasmic reticulum, and bile canaliculi. Liver microtissue zonation characteristics could be easily modulated by maturation in different media supplements. Furthermore, our proof-of-concept study demonstrated the efficacy of this coculture model in evaluating testosterone-mediated androgen receptor responses in the presence of human liver metabolism. Background: Elicitation of patients' preferences is an integral part of shared decision-making, the recommended approach for prostate cancer decision-making. Existing decision aids for this population often do not specifically focus on patients' preferences. Healium is a brief interactive web-based decision aid that aims to elicit patients' treatment preferences and is designed for a low health literate population., Objective: This study used a randomized controlled trial to evaluate whether Healium, designed to target preference elicitation, is as efficacious as Healing Choices, a comprehensive education and decision tool, in improving outcomes for decision-making and emotional quality of life., Methods: Patients diagnosed with localized prostate cancer who had not yet made a treatment decision were randomly assigned to the brief Healium intervention or Healing Choices, a decision aid previously developed by our group that serves as a virtual information center on prostate cancer diagnosis and treatment. Assessments were completed at baseline, 6 weeks, and 3 months post baseline, and included decisional outcomes (decisional conflict, satisfaction with decision, and preparation for decision-making), and emotional quality of life (anxiety/tension and depression), along with demographics, comorbidities, and health literacy., Results: A total of 327 individuals consented to participate in the study (171 were randomized to the Healium intervention arm and 156 were randomized to Healing Choices). The majority of the sample was non-Hispanic (272/282, 96%), White (239/314, 76%), married (251/320, 78.4%), and was on average 62.4 (SD 6.9) years old. Within both arms, there was a significant decrease in decisional conflict from baseline to 6 weeks postbaseline (Healium, P≤.001; Healing Choices, P≤.001), and a significant increase in satisfaction with one's decision from 6 weeks to 3 months (Healium, P=.04; Healing Choices, P=.01). Human-driven environmental changes shape ecological communities from local to global scales. Within cities, landscape-scale patterns and processes and species characteristics generally drive local-scale wildlife diversity. However, cities differ in their structure, species pools, geographies and histories, calling into question the extent to which these drivers of wildlife diversity are predictive at continental scales. In partnership with the Urban Wildlife Information Network, we used occurrence data from 725 sites located across 20 North American cities and a multi-city, multi-species occupancy modelling approach to evaluate the effects of ecoregional characteristics and mammal species traits on the urbanization-diversity relationship. Among 37 native terrestrial mammal species, regional environmental characteristics and species traits influenced within-city effects of urbanization on species occupancy and community composition. Species occupancy and diversity were most negatively related to urbanization in the warmer, less vegetated cities. Additionally, larger-bodied species were most negatively impacted by urbanization across North America. Our results suggest that shifting climate conditions could worsen the effects of urbanization on native wildlife communities, such that conservation strategies should seek to mitigate the combined effects of a warming and urbanizing world., (© 2023. Confidence in model estimates of soil CO 2 flux depends on assumptions regarding fundamental mechanisms that control the decomposition of litter and soil organic carbon (SOC). Multiple hypotheses have been proposed to explain the role of lignin, an abundant and complex biopolymer that may limit decomposition. We tested competing mechanisms using data-model fusion with modified versions of the CN-SIM model and a 571-day laboratory incubation dataset where decomposition of litter, lignin, and SOC was measured across 80 soil samples from the National Ecological Observatory Network. We found that lignin decomposition consistently decreased over time in 65 samples, whereas in the other 15 samples, lignin decomposition subsequently increased. These "lagged-peak" samples can be predicted by low soil pH, high extractable Mn, and fungal community composition as measured by ITS PC2 (the second principal component of an ordination of fungal ITS amplicon sequences). The highest-performing model incorporated soil biogeochemical factors and daily dynamics of substrate availability (labile bulk litter:lignin) that jointly represented two hypotheses (C substrate limitation and co-metabolism) previously thought to influence lignin decomposition. In contrast, models representing either hypothesis alone were biased and underestimated cumulative decomposition. Denitrification bioreactors are an effective edge-of-field conservation practice for nitrate (NO 3 ) reduction from subsurface drainage. However, these systems may produce other pollutants and greenhouse gases during NO 3 removal. Here a dual-chamber woodchip bioreactor system experiencing extreme low-flow conditions was monitored for its spatiotemporal NO 3 and total organic carbon dynamics in the drainage water. Near complete removal of NO 3 was observed in both bioreactor chambers in the first two years of monitoring (2019-2020) and in the third year of monitoring in chamber A, with significant (p < 0.01) reduction of the NO 3 -N each year in both chambers with 8.6-11.4 mg NO 3 -N L -1 removed on average. Based on the NO 3 removal observed, spatial monitoring of sulfate (SO 4 ), dissolved methane (CH 4 ), and dissolved nitrous oxide (N 2 O) gases was added in the third year of monitoring (2021). In 2021, chambers A and B had median hydraulic residence times (HRTs) of 64 h and 39 h, respectively, due to varying elevations of the chambers, with drought conditions making the differences more pronounced. In 2021, significant production of dissolved CH 4 was observed at rates of 0.54 g CH 4 -C m -3 d -1 and 0.07 g CH 4 -C m -3 d -1 in chambers A and B, respectively. In chamber A, significant removal (p < 0.01) of SO 4 (0.23 g SO 4 m -3 d -1 ) and dissolved N 2 O (0.21 mg N 2 O-N m -2 d -1 ) were observed, whereas chamber B produced N 2 O (0.36 mg N 2 O-N m -2 d -1 ). Considering the carbon dioxide equivalents (CO 2 e) on an annual basis, chamber A had loads (~12,000 kg CO 2 e ha -1 y -1 ) greater than comparable poorly drained agricultural soils; however, the landscape-scale impact was small (<1 % change in CO 2 e) when expressed over the drainage area treated by the bioreactor. Pathogenic variants in HCN1 are an established cause of developmental and epileptic encephalopathy (DEE). To date, the stratification of patients with HCN1 -DEE based on the biophysical consequence on channel function of a given variant has not been possible. Here, we analysed data from eleven patients carrying seven different de novo HCN1 pathogenic variants located in the transmembrane domains of the protein. All patients were diagnosed with severe disease including epilepsy and intellectual disability. The functional properties of the seven HCN1 pathogenic variants were assessed using two-electrode voltage-clamp recordings in Xenopus oocytes. All seven variants showed a significantly larger instantaneous current consistent with cation leak. The impact of each variant on other biophysical properties was variable, including changes in the half activation voltage and activation and deactivation kinetics. These data suggest that cation leak is an important pathogenic mechanism in HCN1 -DEE. Furthermore, published mouse model and clinical case reports suggest that seizures are exacerbated by sodium channel blockers in patients with HCN1 variants that cause cation leak. Much of the US Corn Belt has been drained with subsurface tile to improve crop production, yet poorly drained depressions often still flood intermittently, suppressing crop growth. Impacts of depressions on field-scale nutrient leaching are unclear. Poor drainage might promote denitrification and physicochemical retention of phosphorus (P), but ample availability of water and nutrients might exacerbate nutrient leaching from cropped depressions. We monitored nitrate, ammonium, and reactive P leaching across multiple depression-to-upland transects in north-central Iowa, using resin lysimeters buried and retrieved on an annual basis. Crops included conventional corn/soybean (Zea mays/Glycine max) rotations measured at fields with and without a winter rye (Secale cereale) cover crop, as well as juvenile miscanthus (Miscanthus × giganteus), a perennial grass. Leaching of nitrogen (N) and P was greater in depressions than in uplands for most transects and years. The median difference in nutrient leaching between paired depressions and uplands was 56 kg N ha -1 year -1 for nitrate (p = 0.0008), 0.6 kg N ha -1 year -1 for ammonium (p = 0.03), and 2.4 kg P ha -1 year -1 for reactive P (p = 0.006). Transects managed with a cover crop or miscanthus tended to have a smaller median difference in nitrate (but not ammonium or P) leaching between depressions and uplands. Cropped depressions may be disproportionate sources of N and P to downstream waters despite their generally poor drainage characteristics, and targeted management with cover crops or perennials might partially mitigate these impacts for N, but not necessarily for P., (© 2023 The Authors. Lignin is an abundant and complex plant polymer that may limit litter decomposition, yet lignin is sometimes a minor constituent of soil organic carbon (SOC). Accounting for diversity in soil characteristics might reconcile this apparent contradiction. Tracking decomposition of a lignin/litter mixture and SOC across different North American mineral soils using lab and field incubations, here we show that cumulative lignin decomposition varies 18-fold among soils and is strongly correlated with bulk litter decomposition, but not SOC decomposition. Climate legacy predicts decomposition in the lab, and impacts of nitrogen availability are minor compared with geochemical and microbial properties. Lignin decomposition increases with some metals and fungal taxa, whereas SOC decomposition decreases with metals and is weakly related with fungi. Decoupling of lignin and SOC decomposition and their contrasting biogeochemical drivers indicate that lignin is not necessarily a bottleneck for SOC decomposition and can explain variable contributions of lignin to SOC among ecosystems., (© 2023. A major limitation of transient optical spectroscopy is that relatively high laser fluences are required to enable broadband, multichannel detection with acceptable signal-to-noise levels. Under typical experimental conditions, many condensed phase and nanoscale materials exhibit fluence-dependent dynamics, including higher order effects such as carrier-carrier annihilation. With the proliferation of commercial laser systems, offering both high repetition rates and high pulse energies, have come new opportunities for high sensitivity pump-probe measurements at low pump fluences. However, experimental considerations needed to fully leverage the statistical advantage of these laser systems have not been fully described. Here, we demonstrate a high repetition rate, broadband transient spectrometer capable of multichannel shot-to-shot detection at 90 kHz. Importantly, we find that several high-speed cameras exhibit a time-domain fixed pattern noise resulting from interleaved analog-to-digital converters, which is particularly detrimental to the conventional "ON/OFF" modulation scheme used in pump-probe spectroscopy. Using a modified modulation and data processing scheme, we achieve a noise level of 10-5 in 4 s for differential transmission, an order of magnitude lower than for commercial 1 kHz transient spectrometers for the same acquisition time. We leverage the high sensitivity of this system to measure the differential transmission of monolayer graphene at low pump fluence. X-ray photoemission and X-ray absorption spectroscopy are important techniques to characterize chemical bonding at surfaces and are often used to identify the strength and nature of adsorbate-substrate interactions. In this study, we judge the ability of X-ray spectroscopic techniques to identify different regimes of chemical bonding at metal-organic interfaces. To achieve this, we sample different interaction strength regimes in a comprehensive and systematic way by comparing two topological isomers, azulene and naphthalene, adsorbed on three metal substrates with varying reactivity, namely the (111) facets of Ag, Cu, and Pt. Using density functional theory, we simulate core-level binding energies and X-ray absorption spectra of the molecular carbon species. The simulated spectra reveal three distinct characteristics based on the molecule-specific spectral features which we attribute to types of surface chemical bonding with varying strength. We find that weak physisorption only leads to minor changes compared to the gas-phase spectra, weak chemisorption leads to charge transfer and significant spectral changes, and strong chemisorption leads to a loss of the molecule-specific features in the spectra. Atmospheric nitrous oxide (N 2 O) is a potent greenhouse gas thought to be mainly derived from microbial metabolism as part of the denitrification pathway. Here we report that in unexplored peat soils of Central and South America, N 2 O production can be driven by abiotic reactions (≤98%) highly competitive to their enzymatic counterparts. Extracted soil iron positively correlated with in situ abiotic N 2 O production determined by isotopic tracers. Moreover, we found that microbial N 2 O reduction accompanied abiotic production, essentially closing a coupled abiotic-biotic N 2 O cycle. Anaerobic N 2 O consumption occurred ubiquitously (pH 6.4-3.7), with proportions of diverse clade II N 2 O reducers increasing with consumption rates. Our findings show that denitrification in tropical peat soils is not a purely biological process but rather a 'mosaic' of abiotic and biotic reduction reactions. We predict that hydrological and temperature fluctuations differentially affect abiotic and biotic drivers and further contribute to the high N 2 O flux variation in the region., (© 2022. Anti-seizure medications that block sodium channels are generally considered contraindicated in Dravet syndrome. There is, however, considerable debate about the sodium-channel blocker phenytoin, which is often used for status epilepticus, a frequent feature of Dravet syndrome. We describe four patients with Dravet syndrome in whom long-term phenytoin therapy reduced seizure frequency and duration. In two patients, phenytoin produced prolonged periods without status epilepticus for the first time. Attempting to wean phenytoin in all patients after 1 to 20 years of use resulted in seizure exacerbation. Reintroducing phenytoin improved seizure control, suggesting phenytoin is beneficial in some patients with Dravet syndrome., (© 2022 The Authors. Brain voltage-gated sodium channel NaV1.1 (SCN1A) loss-of-function variants cause the severe epilepsy Dravet syndrome, as well as milder phenotypes associated with genetic epilepsy with febrile seizures plus. Gain of function SCN1A variants are associated with familial hemiplegic migraine type 3. Novel SCN1A-related phenotypes have been described including early infantile developmental and epileptic encephalopathy with movement disorder, and more recently neonatal presentations with arthrogryposis. Here we describe the clinical, genetic and functional evaluation of affected individuals. Thirty-five patients were ascertained via an international collaborative network using a structured clinical questionnaire and from the literature. We performed whole-cell voltage-clamp electrophysiological recordings comparing sodium channels containing wild-type versus variant NaV1.1 subunits. Findings were related to Dravet syndrome and familial hemiplegic migraine type 3 variants. We identified three distinct clinical presentations differing by age at onset and presence of arthrogryposis and/or movement disorder. The most severely affected infants (n = 13) presented with congenital arthrogryposis, neonatal onset epilepsy in the first 3 days of life, tonic seizures and apnoeas, accompanied by a significant movement disorder and profound intellectual disability. Twenty-one patients presented later, between 2 weeks and 3 months of age, with a severe early infantile developmental and epileptic encephalopathy and a movement disorder. One patient presented after 3 months with developmental and epileptic encephalopathy only. Associated SCN1A variants cluster in regions of channel inactivation associated with gain of function, different to Dravet syndrome variants (odds ratio = 17.8; confidence interval = 5.4-69.3; P = 1.3 × 10-7). Functional studies of both epilepsy and familial hemiplegic migraine type 3 variants reveal alterations of gating properties in keeping with neuronal hyperexcitability. While epilepsy variants result in a moderate increase in action current amplitude consistent with mild gain of function, familial hemiplegic migraine type 3 variants induce a larger effect on gating properties, in particular the increase of persistent current, resulting in a large increase of action current amplitude, consistent with stronger gain of function. Clinically, 13 out of 16 (81%) gain of function variants were associated with a reduction in seizures in response to sodium channel blocker treatment (carbamazepine, oxcarbazepine, phenytoin, lamotrigine or lacosamide) without evidence of symptom exacerbation. Our study expands the spectrum of gain of function SCN1A-related epilepsy phenotypes, defines key clinical features, provides novel insights into the underlying disease mechanisms between SCN1A-related epilepsy and familial hemiplegic migraine type 3, and identifies sodium channel blockers as potentially efficacious therapies. Background: Propofol is the cornerstone of deep sedation during pediatric esophagogastroduodenoscopy (EGD), though adjuvant dexmedetomidine may provide propofol-sparing benefits., Objective: The objective of the study was to evaluate whether adjuvant dexmedetomidine decreases the total propofol dose in pediatric patients undergoing EGD., Methods: This single-center, retrospective, cohort study evaluated the total propofol dose in pediatric patients undergoing EGD with and without the use of adjuvant dexmedetomidine. Secondary outcomes included the change in hemodynamics across the perioperative continuum and post-procedure recovery time. A multivariable general linear regression was performed to identify associated variables for recovery time post-procedure., Results: A total of 159 patients were included in the study; 88 patients received dexmedetomidine and propofol (DEX-PRO), and 71 patients received propofol only (PRO). The median [interquartile range (IQR)] propofol dose in the DEX-PRO group was 0.26 [IQR, 0.17-0.36] mg kg -1  min -1 which was not significantly different than the PRO group at 0.27 [IQR, 0.21-0.34] mg kg -1  min -1 , p = 0.730. Evaluation of secondary end points showed the DEX-PRO group had more cases of post-anesthesia care unit (PACU) hypotension (61% vs. 34%, p = 0.001) and a longer recovery time (32.9 ± 14.1 vs. 25.6 ± 10.8 min, p < 0.001) versus the PRO group, respectively. Multivariable linear regression demonstrated that age and the use of dexmedetomidine were associated with prolonged recovery., Conclusion: Adjuvant dexmedetomidine did not reduce propofol requirements compared with propofol alone in pediatric patients undergoing EGD. Phthalic acid esters (phthalates) are a class of industrial chemicals that cause developmental and reproductive toxicity, but there are significant gaps in knowledge of phthalate toxicity mechanisms. There is evidence that phthalates disrupt retinoic acid signaling in the fetal testis, potentially disrupting control of spatial and temporal patterns of testis development. Our goal was to determine how a phthalate would interact with retinoic acid signaling during fetal mouse testis development. We hypothesized that mono-(2-ethylhexyl) phthalate (MEHP) would exacerbate the adverse effect of all-trans retinoic acid (ATRA) on seminiferous cord development in the mouse fetal testis. To test this hypothesis, gestational day (GD) 14 C57BL/6 mouse testes were isolated and cultured on media containing MEHP, ATRA, or a combination of both compounds. Cultured testes were collected for global transcriptome analysis after one day in culture and for histology and immunofluorescent analysis of Sertoli cell differentiation after three days in culture. ATRA disrupted seminiferous cord morphogenesis and induced aberrant FOXL2 expression. MEHP alone had no significant effect on cord development, but combined exposure to MEHP and ATRA increased the number of FOXL2-positive cells, reduced seminiferous cord number, and increased testosterone levels, beyond the effect of ATRA alone. In RNA-seq analysis, ATRA treatment and MEHP treatment resulted in differential expression of genes 510 and 134 genes, respectively, including 70 common differentially expressed genes (DEGs) between the two treatments, including genes with known roles in fetal testis development. MEHP DEGs included RAR target genes, genes involved in angiogenesis, and developmental patterning genes, including members of the homeobox superfamily.
