Abstract Recent advances in high-throughput measurement technologies have enabled the analysis of molecular perturbations associated with disease phenotypes at the multi-omic level. Such perturbations can range in scale from fluctuations of individual molecules to entire biological pathways. Data-driven clustering algorithms have long been used to group interactions into interpretable functional modules; however, these modules are typically constrained to a fixed size or statistical cutoff. Furthermore, modules are often analyzed independently of their broader biological context. Consequently, such clustering approaches limit the ability to explore functional module associations with disease phenotypes across multiple scales. Here, we introduce AutoFocus, a data-driven method that hierarchically organizes biomolecules and tests for phenotype enrichment at every level within the hierarchy. As a result, the method allows disease-associated modules to emerge at any scale. We evaluated this approach using two datasets: First, we explored associations of biomolecules from the multi-omic QMDiab dataset (n = 388) with the well-characterized type 2 diabetes phenotype. Secondly, we utilized the ROS/MAP Alzheimer’s disease dataset (n = 500), consisting of high-throughput measurements of brain tissue to explore modules associated with multiple Alzheimer’s Disease-related phenotypes. Our method identifies modules that are multi-omic, span multiple pathways, and vary in size. We provide an interactive tool to explore this hierarchy at different levels and probe enriched modules, empowering users to examine the full hierarchy, delve into biomolecular drivers of disease phenotype within a module, and incorporate functional annotations.

Abstract In-depth multiomic phenotyping provides molecular insights into complex physiological processes and their pathologies. Here, we report on integrating 18 diverse deep molecular phenotyping (omics-) technologies applied to urine, blood, and saliva samples from 391 participants of the multiethnic diabetes Qatar Metabolomics Study of Diabetes (QMDiab). Using 6,304 quantitative molecular traits with 1,221,345 genetic variants, methylation at 470,837 DNA CpG sites, and gene expression of 57,000 transcripts, we determine (1) within-platform partial correlations, (2) between-platform mutual best correlations, and (3) genome-, epigenome-, transcriptome-, and phenome-wide associations. Combined into a molecular network of > 34,000 statistically significant trait-trait links in biofluids, our study portrays “The Molecular Human”. We describe the variances explained by each omics in the phenotypes (age, sex, BMI, and diabetes state), platform complementarity, and the inherent correlation structures of multiomics data. Further, we construct multi-molecular network of diabetes subtypes. Finally, we generated an open-access web interface to “The Molecular Human” ( http://comics.metabolomix.com ), providing interactive data exploration and hypotheses generation possibilities.

Abstract Background Immunoglobulin (Ig) glycosylation modulates the immune response and plays a critical role in ageing and diseases. Studies have mainly focused on IgG glycosylation, and little is known about the genetics and epidemiology of IgA glycosylation. Methods We generated, using a novel liquid chromatography-mass spectrometry method, the first large-scale IgA glycomics dataset in serum from 2423 twins, encompassing 71 N- and O-glycan species. Results We showed that, despite the lack of a direct genetic template, glycosylation is highly heritable, and that glycopeptide structures are sex-specific, and undergo substantial changes with ageing. We observe extensive correlations between the IgA and IgG glycomes, and, exploiting the twin design, show that they are predominantly influenced by shared genetic factors. A genome-wide association study identified eight loci associated with both the IgA and IgG glycomes (ST6GAL1, ELL2, B4GALT1, ABCF2, TMEM121, SLC38A10, SMARCB1, and MGAT3) and two novel loci specifically modulating IgA O-glycosylation (C1GALT1 and ST3GAL1). Validation of our findings in an independent cohort of 320 individuals from Qatar showed that the underlying genetic architecture is conserved across ancestries. Conclusions Our study delineates the genetic landscape of IgA glycosylation and provides novel potential functional links with the aetiology of complex immune diseases, including genetic factors involved in IgA nephropathy risk.

Plutonic xenoliths from volcanic arcs provide unique insights into transcrustal magmatic systems in subduction zone settings. At Santorini volcano in the Central Aegean Volcanic Arc (Greece), plutonic xenoliths occur throughout a sequence of lavas and pyroclastic rocks erupted within the last ~360 ka. They are mineralogically variable, ranging from troctolites to olivine gabbros, gabbros, gabbronorites, and diorites. Today’s data-driven world requires earth and environmental scientists to have skills at the intersection of domain and data science. These skills are imperative to harness information contained in a growing volume of complex data to solve the world’s most pressing environmental challenges. Despite the importance of these skills, Earth and Environmental Data Science (EDS) training is not equally accessible, contributing to a lack of diversity in the field. This creates a critical need for EDS training opportunities designed specifically for underrepresented groups. In response, we developed the Earth Data Science Corps (EDSC) which couples a paid internship for undergraduate students with faculty training to build capacity to teach and learn EDS using Python at smaller Minority Serving Institutions. EDSC faculty participants are further empowered to teach these skills at their home institutions which scales the program beyond the training lead by our team. Using a Rasch modeling approach, we found that participating in the EDSC program had a significant impact on undergraduate learners’ comfort and confidence with technical and nontechnical data science skills, as well as their science identity and sense of belonging in science, two critical aspects of recruiting and retaining members of underrepresented groups in STEM. Abstract When someone violates a social norm, others may think that some sanction would be appropriate. We examine how the experience of emotions like anger and disgust relate to the judged appropriateness of sanctions, in a pre-registered analysis of data from a large-scale study in 56 societies. Across the world, we find that individuals who experience anger and disgust over a norm violation are more likely to endorse confrontation, ostracism and, to a smaller extent, gossip. Moreover, we find that the experience of anger is consistently the strongest predictor of judgments of confrontation, compared to other emotions. Although the link between state-based emotions and judgments may seem universal, its strength varies across countries. Aligned with theoretical predictions, this link is stronger in societies, and among individuals, that place higher value on individual autonomy. Thus, autonomy values may increase the role that emotions play in guiding judgments of social sanctions.

Abstract Proteogenomics studies generate hypotheses on protein function and provide genetic evidence for drug target prioritization. Most previous work has been conducted using affinity-based proteomics approaches. These technologies face challenges, such as uncertainty regarding target identity, non-specific binding, and handling of variants that affect epitope affinity binding. Mass spectrometry-based proteomics can overcome some of these challenges. Here we report a pQTL study using the Proteograph™ Product Suite workflow (Seer, Inc.) where we quantify over 18,000 unique peptides from nearly 3000 proteins in more than 320 blood samples from a multi-ethnic cohort in a bottom-up, peptide-centric, mass spectrometry-based proteomics approach. We identify 184 protein-altering variants in 137 genes that are significantly associated with their corresponding variant peptides, confirming target specificity of co-associated affinity binders, identifying putatively causal cis-encoded proteins and providing experimental evidence for their presence in blood, including proteins that may be inaccessible to affinity-based proteomics.

National commemorations are historical events, too. While re-imagining the past to correspond with contemporary sentiments, they are themselves open to reinterpretation by future observers. The 50th anniversary of the 1916 Easter Rising was intended to be a liminal event, paying tribute to the veterans of the revolutionary generation while recalibrating the national narrative to be more conciliatory and less militaristic. Yet, the anniversary later became neglected as one of the sparks of the conflict in Northern Ireland and subsequently served as the grand negative example of what needs to be avoided during the preparations for the Easter Rising centenary in 2016. The organisers of this centenary eventually introduced a highly diverse programme that held inclusivity at its centre. Nonetheless, with Brexit or Covid-19 in mind, one might wonder what the afterlife of the centennial mosaic of narratives will look like. Will the popular success of the event last, or will it be overshadowed or even neglected in the future?

Abstract Background Bardet-Biedl syndrome (BBS) is an autosomal recessive, genetically heterogeneous, pleiotropic disorder caused by variants in genes involved in the function of the primary cilium. We have harnessed genomics to identify BBS and ophthalmic technologies to describe novel features of BBS. Case presentation A patient with an unclear diagnosis of syndromic type 2 diabetes mellitus, another affected sibling and unaffected siblings and parents were sequenced using DNA extracted from saliva samples. Corneal confocal microscopy (CCM) and retinal spectral domain optical coherence tomography (SD-OCT) were used to identify novel ophthalmic features in these patients. The two affected individuals had a homozygous variant in C8orf37 (p.Trp185*). SD-OCT and CCM demonstrated a marked and patchy reduction in the retinal nerve fiber layer thickness and loss of corneal nerve fibers, respectively. Conclusion This report highlights the use of ophthalmic imaging to identify novel retinal and corneal abnormalities that extend the phenotype of BBS in a patient with syndromic type 2 diabetes.

The auto-balancing mechanism is used in the spin-dry regime of the laundry machine. The high rotating speed and unbalanced mass inside the drum create centrifugal force, which is the cause of vibration. The auto-balancing mechanism consists of a set of balls in the circular guiding track. During the spin-dry process, the balls settle in the opposite position to an unbalanced mass. The centrifugal force of the balls compensates the one of the unbalanced masses. The paper deals with the non-linear numerical simulation of the imbalance settlement and the following parametric study. The solution to the problem is demonstrated on an industrial laundry machine with a maximum capacity of 7 kg of dry laundry and a maximum rotating speed of 930 rpm. The proposed numerical model allows us to investigate the behavior of the auto-balancing mechanism and predict the vibration amplitudes of the system.

Coho salmon (Oncorhynchus kisutch) are highly sensitive to 6PPD-Quinone (6PPD-Q). Details of the hydrological and biogeochemical processes controlling spatial and temporal dynamics of 6PPD-Q fate and transport from points of deposition to receiving waters (e.g., streams, estuaries) are poorly understood. To understand the fate and transport of 6PPD and mechanisms leading to salmon mortality Visualizing Ecosystem Land Management Assessments (VELMA), an ecohydrological model developed by US Environmental Protection Agency (EPA), was enhanced to better understand and inform stormwater management planning by municipal, state, and federal partners seeking to reduce stormwater contaminant loads in urban streams draining to the Puget Sound National Estuary. This work focuses on the 5.5 km2 Longfellow Creek upper watershed (Seattle, Washington, United States), which has long exhibited high rates of acute urban runoff mortality syndrome in coho salmon. We present VELMA model results to elucidate these processes for the Longfellow Creek watershed across multiple scales–from 5-m grid cells to the entire watershed. Our results highlight hydrological and biogeochemical controls on 6PPD-Q flow paths, and hotspots within the watershed and its stormwater infrastructure, that ultimately impact contaminant transport to Longfellow Creek and Puget Sound. Simulated daily average 6PPD-Q and available observed 6PPD-Q peak in-stream grab sample concentrations (ng/L) corresponds within plus or minus 10 ng/L. Most importantly, VELMA’s high-resolution spatial and temporal analysis of 6PPD-Q hotspots provides a tool for prioritizing the locations, amounts, and types of green infrastructure that can most effectively reduce 6PPD-Q stream concentrations to levels protective of coho salmon and other aquatic species.

IntroductionThe results indicate that post-traumatic growth does indeed occur after overcoming the severe form of COVID-19. It suggests that this posttraumatic growth most often occurred through a reassessment of priorities and an appreciation of life itself and loved ones. COVID-19 disease has been one of the most discussed and researched topics for several years, as it dramatically affects everyone’s daily life.MethodsThe qualitative study presented here focuses on health psychology, especially post-traumatic growth after overcoming a severe form of the COVID-19 disease. We worked with adult people who had either been treated in the intensive care unit or had been hospitalized with severe pneumonia.ResultsThematic analysis was used to determine categories and subcategories. The study presented here contributes to knowledge about the COVID-19 experience by mapping a Slovakian sample of adult participants.DiscussionThe results obtained by Thematic analysis help us better understand how people experience the disease, especially those who have overcome a severe form of the disease and thus had a borderline experience when their lives were directly threatened, as well as their overall health.

Abstract Background Although most retirees are satisfied, some do not feel well in retirement. The resource-based dynamic perspective explains retirement dissatisfaction as the lack of resources. This study focused on psychological resources, specifically on the role of rational/irrational beliefs and retirement concepts in retirement satisfaction. While irrational beliefs have many consequences, we know little about their role in retirement experiences, nor do we know about the benefits/harm of retirement concepts for retirement satisfaction. We assumed that not succumbing to irrational beliefs and conceptualizing retirement actively and positively add to psychological resources helping to adjust to retirement and retirement satisfaction. Our objective was to examine whether irrational beliefs and retirement concepts contributed to satisfaction or dissatisfaction in recent retirees. Methods 200 recent retirees (average retirement time 2.8 years) completed questionnaires containing the Irrational Belief Scale, the Satisfaction with Retirement Scale, and the Retirement Lifestyles Questionnaire, which determines the inclination toward four retirement concepts: Transition to Old Age, New Start, Continuation, and Imposed Disruption. The Pearson correlation coefficients were used to estimate the relationship between irrational beliefs, retirement concepts, and retirement satisfaction. We used a parallel mediation model with multiple mediators in the mediation analysis where irrational beliefs were the independent variable, retirement satisfaction was the dependent variable, and the four retirement concepts were mediators. Results We confirmed higher retirement satisfaction in recent retirees who conceptualize retirement as a New Start and Continuation and higher retirement dissatisfaction in those who see retirement as an Imposed Disruption or Transition to Old Age. The general irrational beliefs had a weaker direct impact on retirement satisfaction than the more specific retirement concepts. Inclination to general irrational beliefs appeared only weakly reflected in retirement dissatisfaction. However, a negative view of retirement as an imposed disruption might increase this inclination by intensifying retirement dissatisfaction. Conclusions Our results show a negative retirement concept as an imposed disruptive event that amplifies the impact of general irrational beliefs and leads to retirement dissatisfaction in recent retirees. It suggests that using rational-emotive behavior therapy and interventions to change the negative perception of retirement could be effective in increasing retirement satisfaction.

Chemokines orchestrate many aspects of tumorigenic processes such as angiogenesis, apoptosis and metastatic spread, and related receptors are expressed on tumor cells as well as on inflammatory cells (e.g., tumor-infiltrating T cells, TILs) in the tumor microenvironment. Expressional changes of chemokines and their receptors in solid cancers are common and well known, especially in affecting colorectal cancer patient outcomes. Therefore, the aim of this current systematic review and meta-analysis was to classify chemokines as a prognostic biomarker in colorectal cancer patients. A systematic literature search was conducted in PubMed, CENTRAL and Web of Science. Information on the chemokine expression of 25 chemokines in colorectal cancer tissue and survival data of the patients were investigated. The hazard ratio of overall survival and disease-free survival with chemokine expression was examined. The risk of bias was analyzed using Quality in Prognosis Studies. Random effects meta-analysis was performed to determine the impact on overall respectively disease survival. For this purpose, the pooled hazard ratios (HR) and their 95% confidence intervals (CI) were used for calculation. Twenty-five chemokines were included, and the search revealed 5556 publications. A total of thirty-one publications were included in this systematic review and meta-analysis. Overexpression of chemokine receptor CXCR4 was associated with both a significantly reduced overall survival (HR = 2.70, 95%-CI: 1.57 to 4.66, p = 0.0003) as well as disease-free survival (HR = 2.68, 95%-CI: 1.41 to 5.08, p = 0.0026). All other chemokines showed either heterogeneous results or few studies were available. The overall risk of bias for CXCR4 was rated low. At the current level of evidence, this study demonstrates that CXCR4 overexpression in patients with colorectal cancer is associated with a significantly diminished overall as well as disease-free survival. Summed up, this systematic review and meta-analysis reveals CXCR4 as a promising prognostic biomarker. Nevertheless, more evidence is needed to evaluate CXCR4 and its antagonists serving as new therapeutic targets.

Abstract Feces from 142 animals were collected on 15 farms in the region of Brittany, France. Each sample was directly collected from the rectum of the animal and identified with the ear tag number. Animals were sampled three times, at 5, 15 and 22 weeks of age. After DNA extraction from stool samples, nested PCR was performed to amplify partial 18S-rDNA and 60 kDa glycoprotein genes of Cryptosporidium. The parasite was detected on all farms. One hundred out of 142 calves (70.4%) were found to be parasitized by Cryptosporidium. Amplified fragments were sequenced for Cryptosporidium species identification and revealed the presence of C. parvum (43.8%), C. ryanae (28.5%), and C. bovis (27%). One animal was infected with Cryptosporidium ubiquitum. The prevalence of these species was related to the age of the animal. C. parvum caused 86.7% of Cryptosporidium infections in 5-week-old calves but only 1.7% in 15-week-old animals. The analysis of the results showed that animals could be infected successively by C. parvum, C. ryanae, and C. bovis for the study period. C. parvum gp60 genotyping identifies 6 IIa subtypes of which 74.5% were represented by IIaA15G2R1. This work confirms previous studies in other countries showing that zoonotic C. parvum is the dominant species seen in young calves.

Abstract The current worldwide context promoting agroecology and green agriculture require the discovery of new ecofriendly and sustainable plant protection tools. Plant resistance inducers, called also elicitors, are one of the most promising alternatives fitting with such requirements. We produced here a set of 30 molecules from pyroglutamic acid, bio-sourced from sugar beet byproducts, and examined for their biological activity on the major agro-economically pathosystem wheat-Zymoseptoria tritici. Foliar application of the molecules provided significant protection rates (up to 63% disease severity reduction) for 16 among them. Structure–activity relationship analysis highlighted the importance of all chemical groups of the pharmacophore in the bioactivity of the molecules. Further investigations using in vitro and in planta antifungal bioassays as well as plant molecular biomarkers revealed that the activity of the molecules did not rely on direct biocide activity towards the pathogen, but rather on the activation of plant defense mechanisms dependent on lipoxygenase, phenylalanine ammonia-lyase, peroxidase, and pathogenesis-related protein pathways. This study reports a new family of bio-sourced resistance inducers and provides new insights into the valorization of agro-resources to develop the sustainable agriculture of tomorrow.

Many approaches to identify therapeutically relevant neoantigens couple tumor sequencing with bioinformatic algorithms and inferred rules of tumor epitope immunogenicity. However, there are no reference data to compare these approaches, and the parameters governing tumor epitope immunogenicity remain unclear. Here, we assembled a global consortium wherein each participant predicted immunogenic epitopes from shared tumor sequencing data. 608 epitopes were subsequently assessed for T cell binding in patient-matched samples. By integrating peptide features associated with presentation and recognition, we developed a model of tumor epitope immunogenicity that filtered out 98% of non-immunogenic peptides with a precision above 0.70. Pipelines prioritizing model features had superior performance, and pipeline alterations leveraging them improved prediction performance. These findings were validated in an independent cohort of 310 epitopes prioritized from tumor sequencing data and assessed for T cell binding. This data resource enables identification of parameters underlying effective anti-tumor immunity and is available to the research community.

The contribution presents the verification of the methodology of accelerated creep tests from the point of view of obtaining more information about the stress–strain behaviour of the investigated materials using the Digital Image Correlation method. Creep tests are performed on SAC305 and SACX0807 lead-free solders and are supplemented by numerical modelling using the finite element method, considering the viscoplastic model based on the theory of Perzyna, Chaboche, and Norton. The stress–strain behaviour of both solders appears to be very similar at applied strain rates of 0.0002–0.0026%/s and applied creep stresses of 15–28 MPa. Initially, the viscoplastic model is calibrated using an analytical approach. Then, the finite element model updating approach is used to optimise the material parameters based on the simultaneous simulations of creep and tensile tests. As a result, the total objective function value is reduced almost five times due to optimisation. The proposed type of accelerated test with an hourglass specimen proves to be suitable for calibrating the considered class of viscoplastic models. The main benefit is that a single specimen is required to obtain creep curves on various stress levels.

The present study examines the Big Five traits and socio-demographic factors as predictors of both traditional left-right and liberal-conservative positions of Slovak voters (N = 704). As shown in previous research, Open-Mindedness and Conscientiousness are significant but weak predictors of both political axes, while Agreeableness plays a surprising role in predicting left-right orientation. To overcome the limitations of traditional political axes, three latent variables of the Slovak political space were discovered by exploratory factor analysis (EFA) of political preferences: sympathy towards old/new government, socially conservative parties, and a non-populist coalition. The former is significantly predicted by Open-Mindedness, while the latter is predicted by Conscientiousness. Overall, the Big Five traits predict a small but significant variance in political variables.

Abstract People cooperate every day in ways that range from largescale contributions that mitigate climate change to simple actions such as leaving another individual with choice – known as social mindfulness. It is not yet clear whether and how these complex and more simple forms of cooperation relate. Prior work has found that countries with individuals who made more socially mindful choices were linked to a higher country environmental performance – a proxy for complex cooperation. Here we replicated this initial finding in 41 samples around the world, demonstrating the robustness of the association between social mindfulness and environmental performance, and substantially built on it to show this relationship extended to a wide range of complex cooperative indices, tied closely to many current societal issues. We found that greater social mindfulness expressed by an individual was related to living in countries with more social capital, more community participation and reduced prejudice towards immigrants. Our findings speak to the symbiotic relationship between simple and more complex forms of cooperation in societies.

The phenomenon of the privacy paradox states that people are concerned about their data, but do not behave accordingly by avoiding disclosure of personal data, for instance, when using social media. Since findings about the influence of privacy concerns on privacy-related behavior are controversial, the present study investigates whether social norms or individual privacy concerns provide a better explanation of disclosure behavior. Therefore, a social media app dummy was developed and the social norm manipulated by altering the amount and the sensitivity of information provided in a fictitious app user profile. Regarding social norm factors, the results of the online experiment (N = 42) revealed that only the sensitivity of the data in the dummy profile demonstrated a medium effect on the amount of personal data given by the participants. Considering individual factors, the less concerned individuals were about their privacy, the higher were the amount and the degree of sensitivity of the personal information participants disclosed. In addition, participants differentiated between sensitive or non-sensitive information when disclosing their data. They disclosed sensitive information significantly less often, regardless of the dummy profile they saw. Altogether, the study results do not support the privacy paradox and demonstrate implications for media education.

Four T2D subtypes were previously identified: severe insulin deficient, severe insulin resistant, mild obesity-related, and mild age-related diabetes. Here, the authors show that these subtypes can be translated to an Arabic population and identify distinct subtype-specific metabolic and proteomic signatures.

Abstract Rare‐earth metal‐mediated group‐transfer polymerization (REM‐GTP) offers distinctive features over common polymerization techniques, such as living character, a broad scope of functional monomers, high activity, excellent control of the polymeric parameters as well as inherent chain‐end functionalization. Through the latter, polymers with reactive end‐groups become feasible, opening the pathway for further post‐polymerization functionalization. In this study, a straightforward graft‐to immobilization of the Michael‐type polymer poly(diethyl vinylphosphonate) (PDEVP) on multi‐walled carbon nanotubes (MWCNT) is reported. Hence, a customized azide initiator is synthesized and studied in the CH bond activation with various lanthanide‐based catalysts and the subsequent polymerization of diethyl vinylphosphonate (DEVP). The successful attachment of the azide end‐group is demonstrated via electrospray ionization mass spectrometry (ESI‐MS) and the synthesized polymers are subjected to immobilization on multi‐walled carbon nanotubes in a graft‐to approach. The prepared MWCNT:PDEVP composites are analyzed via thermogravimetric analysis (TGA), elemental analysis (EA), Raman spectroscopy, X‐Ray photoelectron spectroscopy (XPS), and transmission electron microscopy (TEM) and the versatility of this approach is shown via the stabilization of MWCNT dispersions in water.

The research aim was to find out whether providing information on psychological preparation would produce changes in pre-retirees retirement planning intentions and advance retirement concepts. In the experiment, we divided the pre-retirees (N = 567, mean age of 54.91, SD = 3.55 years, working full-time) into four groups (with and without pre-test, with and without the information provided). There were no differences between the pre-test and post-test conditions. However, the results showed that participants acknowledged the need to prepare for retirement even before the intervention. The participants also rated themselves as psychologically prepared for retirement. The results also indicated which pre-retirees could benefit from psychological preparation: those who believe they would have a disadvantageous and unfavorable retirement transition. The next group is the ones who tend to conceptualize their retirement as an imposed disruption as they also perceive their psychological resources for a successful transition as insufficient.

Exercise-induced hemolysis occurs as the result of intense physical exercise and is caused by metabolic and mechanical factors including repeated muscle contractions leading to capillary vessels compression, vasoconstriction of internal organs and foot strike among others. We hypothesized that exercise-induced hemolysis occurred in endurance racehorses and its severity was associated with the intensity of exercise. To provide further insight into the hemolysis of endurance horses, the aim of the study was to deployed a strategy for small molecules (metabolites) profiling, beyond standard molecular methods. The study included 47 Arabian endurance horses competing for either 80, 100, or 120 km distances. Blood plasma was collected before and after the competition and analyzed macroscopically, by ELISA and non-targeted metabolomics with liquid chromatography–mass spectrometry. A significant increase in all hemolysis parameters was observed after the race, and an association was found between the measured parameters, average speed, and distance completed. Levels of hemolysis markers were highest in horses eliminated for metabolic reasons in comparison to finishers and horses eliminated for lameness (gait abnormality), which may suggest a connection between the intensity of exercise, metabolic challenges, and hemolysis. Utilization of omics methods alongside conventional methods revealed a broader insight into the exercise-induced hemolysis process by displaying, apart from commonly measured hemoglobin and haptoglobin, levels of hemoglobin degradation metabolites. Obtained results emphasized the importance of respecting horse limitations in regard to speed and distance which, if underestimated, may lead to severe damages.

Scaled rolling contact fatigue tests, used to practically simulate the wear of the wheel and rail material under laboratory conditions, are typically classified into two categories. Tests in the first category use twin-disc stands, while the second group of test rigs use two discs of different diameters considering the rail disc as the larger one. The latter setup is closer to the real situation, but problems can occur with high contact pressures and tractions. The focus of this paper is on two main contributions. Firstly, a case study based on finite element analysis is presented, allowing the optimization of the specimen geometry for high contact pressures. Accumulated plastic deformation caused by cycling is responsible for abrupt lateral deformation, which requires the use of an appropriate cyclic plasticity model in the finite element analysis. In the second part of the study, two laser profilers are used to measure the dimensions of the specimen in real time during the rolling contact fatigue test. The proposed technique allows the changes in the specimen dimensions to be characterized during the test itself, and therefore does not require the test to be interrupted. By using real-time values of the specimen’s dimensional contours, it is possible to calculate an instantaneous value of the slip ratio or the contact path width.
