Alfalfa (Medicago sativa. L.) is a globally significant autotetraploid legume forage crop. However, despite its importance, establishing efficient gene editing systems for cultivated alfalfa remains a formidable challenge. In this study, we pioneered the development of a highly effective ultrasonic-assisted leaf disc transformation system for Gongnong 1 alfalfa, a variety widely cultivated in Northeast China. Subsequently, we created a single transcript CRISPR/Cas9 (CRISPR_2.0) toolkit, incorporating multiplex gRNAs, designed for gene editing in Gongnong 1. Both Cas9 and gRNA scaffolds were under the control of the Arabidopsis ubiquitin-10 promoter, a widely employed polymerase II constitutive promoter known for strong transgene expression in dicots. To assess the toolkit’s efficiency, we targeted PALM1, a gene associated with a recognizable multifoliate phenotype. Utilizing the CRISPR_2.0 toolkit, we directed PALM1 editing at two sites in the wild-type Gongnong 1. Results indicated a 35.1% occurrence of editing events all in target 2 alleles, while no mutations were detected at target 1 in the transgenic-positive lines. To explore more efficient sgRNAs, we developed a rapid, reliable screening system based on Agrobacterium rhizogenes-mediated hairy root transformation, incorporating the visible reporter MtLAP1. This screening system demonstrated that most purple visible hairy roots underwent gene editing. Notably, sgRNA3, with an 83.0% editing efficiency, was selected using the visible hairy root system. As anticipated, tetra-allelic homozygous palm1 mutations exhibited a clear multifoliate phenotype. These palm1 lines demonstrated an average crude protein yield increase of 21.5% compared to trifoliolate alfalfa. Our findings highlight the modified CRISPR_2.0 system as a highly efficient and robust gene editing tool for autotetraploid alfalfa.

Background: Interdigital tinea pedis is the most common type of foot infection, which is often treated by topical or systemic antifungals. Due to the increase in antifungal resistance, antifungal socks are becoming potential alternatives for the daily management of tinea pedis. Methods: In this study, antifungal fibres were adopted to produce interdigital hygiene socks to split the third and fourth toe seams of the feet. In vitro antifungal activity was first examined to verify the effectiveness of the socks. Preventive efficacy against tinea pedis was then evaluated among healthy participants, followed by therapeutic effect detection in patients diagnosed with tinea pedis by analysing the improvement in total symptom scores (TTS). Results: The interdigital-type hygiene socks exhibited apparent antifungal activities in vitro. BackgroundJob burnout has become an important factor affecting the mental and physical health and work efficiency of college counselors, and indirectly affects the quality and development of talent cultivation for college students. ObjectiveTo explore the relationship between job stress, job crafting, and job burnout among college counselors, and to test the mediating role of job crafting between job stress and job burnout, in order to take targeted measures to alleviate job stress and job burnout of college counselors, reduce associated health risks, and improve the effectiveness of higher education. MethodsAn anonymous questionnaire survey was conducted among 400 counselors from social network communication groups by convenience sampling. The Counselor Work Stress Scale, Job Crafting Scale, and Maslach Burnout Inventory-General Survey were used. Harman's single-factor method was used to evaluate common method bias in the survey data. One-way ANOVA was applied to test the difference in job stress, job crafting, and job burnout among college counselors by demographic characteristics, and chi-square test was used to analyze the difference in reporting job burnout. Partial correlation analysis was used to evaluate the correlation between selected variables. Structural equation modeling was used to analyze the relationship of job stress, job crafting, and job burnout among college counselors, and Bootstrap analysis was used to test if there was a mediating effect of job crafting on the relationship between job stress and job burnout. ResultsOf the 390 questionnaires recovered, there were 338 valid questionnaires (86.67%). Among the included subjects, the mean scores of job stress, job crafting, and job burnout were (2.70±0.62), (3.77±0.62), and (2.09±1.09), respectively. The positive rate of job burnout was 76.9% (260/338), with a positive rate of 72.8% in exhaustion dimension and 59.8% in cynicism dimension. Background. Coronary artery disease (CAD) is a type of cardiovascular disease which is one of the leading causes of death around the world. The presence of coronary stenosis is considered a pivotal indicator in the diagnosis of various CADs. The main purpose of this paper was to investigate the feasibility of an anchor-free deep learning (DL) method, fully convolutional one-stage object detection (FCOS), in coronary artery stenosis automatic detection. Methods. First, 2786 invasive coronary angiography (ICA) images from 130 patients were randomly divided into training, validation, and testing datasets using the 10-fold cross-validation approach. Then, FCOS was compared with other three widely used anchor-based DL models: single shot multibox detector (SSD), faster region-based convolutional network (Faster R-CNN), and you only look once (YOLOv3), in terms of precision, recall, F1 score, average precision (AP), and average recall (AR). Finally, the performances of different models in the detection of stenosis were compared in either single or multiple lesion scenarios using statistical tests. Results. FCOS achieved significantly superior precision (96.14% ± 0.53%), recall (94.36% ± 0.79%), F1 score (95.22% ± 0.56%), AP0.50 (93.36% ± 0.93%), AR0.50:0.95 (64.73% ± 1.46%), APsmall (55.04 ± 0.96%), APmedium (59.97 ± 1.13%), and APlarge (68.09 ± 5.18%) compared to Faster R-CNN and YOLOv3. Moreover, FCOS demonstrated significantly higher AR0.50:0.95 and APsmall compared to SSD. Regardless of the presence of single or multiple coronary stenoses in ICA images, FCOS also outperformed Faster R-CNN and YOLOv3. Furthermore, it showed significantly higher AR0.50:0.95 compared to SSD when in the multiple stenosis scenario. Conclusions. It is feasible to use the anchor-free DL model FCOS in detecting coronary stenosis based on ICA images.

The escalating degradation of urban eco-environments has underscored the significance of ecological security in sustainable urban development. Green infrastructure bridges green spaces in cities and increases ecosystem connectivity, thereby optimizing urban ecological security patterns. This study uses Nanjing as a case study and adopts a research paradigm that involves identifying ecological sources, constructing resistance surfaces, and subsequently extracting corridors within the ecological security pattern. This method amalgamates the evaluation of green infrastructure supply and demand, leading to the identification of both ecological corridors and nodes. The findings reveal that while the supply of green infrastructure in Nanjing is low in the city center and high in the suburbs, demand is high in the central area and low in the periphery, indicating a spatial mismatch between supply and demand. Ecological corridors and nodes are categorized into the core, important, and general levels based on their centrality and areas of supply–demand optimization. The connectivity, supply capacity, and supply–demand relationship of green infrastructure in Nanjing have been enhanced to varying degrees through the ecological security pattern optimization. The results of this study can serve as a decision-making reference for optimizing green infrastructure network patterns and enhancing urban ecological security.

Current point cloud registration methods predominantly focus on extracting geometric information from point clouds. In certain scenarios, i.e., when the target objects to be registered contain a large number of repetitive planar structures, the point-only based methods struggle to extract distinctive features from the similar structures, which greatly limits the accuracy of registration. Moreover, the deep learning-based approaches achieve commendable results on public datasets, but they face challenges in generalizing to unseen few-shot datasets with significant domain differences from the training data, and that is especially common in industrial applications where samples are generally scarce. Moreover, existing registration methods can achieve high accuracy on complete point clouds. However, for partial point cloud registration, many methods are incapable of accurately identifying correspondences, making it challenging to estimate precise rigid transformations. This paper introduces a domain-adaptive multimodal feature fusion method for partial point cloud registration in an unsupervised manner, named DAMF-Net, that significantly addresses registration challenges in scenes dominated by repetitive planar structures, and it can generalize well-trained networks on public datasets to unseen few-shot datasets. Specifically, we first introduce a point-guided two-stage multimodal feature fusion module that utilizes the geometric information contained in point clouds to guide the texture information in images for preliminary and supplementary feature fusion. Secondly, we incorporate a gradient-inverse domain-aware module to construct a domain classifier in a generative adversarial manner, weakening the feature extractor’s ability to distinguish between source and target domain samples, thereby achieving generalization across different domains. Experiments on a public dataset and our industrial components dataset demonstrate that our method improves the registration accuracy in specific scenarios with numerous repetitive planar structures and achieves high accuracy on unseen few-shot datasets, compared with the results of state-of-the-art traditional and deep learning-based point cloud registration methods.

In the face of geopolitical challenges and climate change, economic progress, safe production, and environmental protection have emerged as important directions for chemical industry development. However, the rational optimization of the chemical industry layout under the backdrop of ecological environmental protection necessitates further exploration. This study explores the evolution and future development direction of the chemical industry layout within the coastal region of Jiangsu Province, China, using the CA–Markov model. The findings reveal a trend of spatial agglomeration growth among coastal chemical enterprises, with Moran’s Index increasing from 0.109 in 2007 to 0.206 in 2017. The petrochemical industry, in particular, demonstrated the most significant agglomeration effect, with approximately 52.10% being concentrated in 14 coastal industrial parks in 2017. Under the constraints of the ecological environment and policy guidance, the land area allocated for the chemical industry experienced a reduction of over 10%, further strengthening the emphasis on spatial agglomeration. Chemical industries along Jiangsu’s coast have become agglomerated and concentrated near industrial parks and ports. Their spatial distribution and connectivity were mainly influenced by factors such as convenient transportation, the ecological environment, local policies, the distance from residential areas, and industrial agglomeration. Under different scenarios—including natural growth, ecological environment constraints, and policy guidance—chemical industries show diverse spatial patterns. Ecological environmental constraints and policy guidance can provide various intervention methods for the government to promote the optimization direction and focus of the chemical industry layout while minimizing its impact on the ecological environment.

In the late Qing Dynasty, religious periodicals by Western missionaries were made legal in China, and subsequently became an important manner of their missionary cause. Among them, Child’s Paper 小孩月报 (1875–1881) by John Marshall Willoughby Farnham, a Protestant missionary from the United States, endeavoured to convert child readers by carrying children’s stories of moral and emotional education. By concentrating on the educational elements of Child’s Paper, this article inspects how conversion was achieved via the intertextual interpretation of Christian doctrines within these educational elements. Specifically, how the image of little Christians and urchins, respectively, represents salvation and redemption in Christian morals. This article holds that the missionaries’ stress on the authority of Christian discourse in the education of Chinese children makes evident an increasing emphasis on the reformative effects of Christianity on Chinese children. Moreover, the conversion-education efforts by missionaries also construed helping Chinese children gain a cross-cultural perspective on Western religion, and arguably inspired later Chinese intellectuals’ to create newspapers for the purpose of the pre-primary education of Chinese children.

Autoimmune hepatitis (AIH) results from an autoimmune-mediated chronic inflammatory response against liver cells. Defective self-tolerance and dysfunctional dendritic cells (DCs) play a regulatory role in AIH. Itaconate has recently attracted attention in the field of immunometabolism because of its crucial role as an anti-inflammatory metabolite that negatively regulates the inflammatory response. However, the underlying mechanism of itaconate mediation of DCs in AIH remains unclear. In this study, we found that itaconate acts as an anti-inflammatory factor in the liver. Endogenous itaconate levels were significantly increased in mice with S100-induced AIH model and correlated with upregulation of the immune-responsive gene 1 expression. However, the anti-inflammatory response from endogenously itaconate may not represent the effects exogenously-produced itaconate. We investigated the anti-inflammatory response from exogenous itaconate in S100-induced AIH, and our results showed that itaconate treatment attenuated liver histopathological damage, hepatocyte apoptosis, aminotransferase elevation, and IL-6 production in the S100-induced AIH model. In addition, Itaconate decreased glycolysis to suppress the maturation of DCs in the liver and spleen of AIH models, thereby directly regulating differentiation of Th17 and Tregs in vivo. Due to their complex genotypes, low in vitro regeneration rates, and difficulty in obtaining transgenic plants, studies concerning basic biological research and molecular breeding in Tartary buckwheat (TB) are greatly limited. In this study, the hypocotyls of 60 genotypes of TB (TBC1~60) were used as explants. Of these, TBC14 was selected due to a high callus induction rate of 97.78% under dark and a proliferation coefficient (PC) of 28.2 when cultured on MS medium supplemented with 2.0 mg/L of 2,4-D and 1.5 mg/L of 6-BA. Subsequently, the samples of the calli obtained from TBC14 were collected at 0, 10, 20, and 30 d, and their transcriptomes were sequenced where identified. GO enrichment led to the detection of the most significant active gene set, which was the DNA binding transcription factor activity. The DEGs related to the pathways concerning metabolism, the biosynthesis of secondary metabolites, and hormone signal transduction were the most enriched in the KEGG database. The sets of MYB, AP2/ERF, and bHLH TFs exhibited the highest number of DEGs. Using this enrichment analysis, 421 genes encoding TFs, 47 auxin- and cytokinin-related genes, and 6 signal transduction-associated genes were screened that may play significant roles in callus formation (CF) in TB. Furthermore, FtPinG0008123200.01 (bZIP), a key gene promoting CF, was screened in terms of the weighted gene co-expression network associated with the various stages of CF. Our study not only provides valuable information about the molecular mechanism of CF but also reveals new genes involved in this process.

Pheromones play a vital role in regulating fish behavior, including reproduction, aggregation, hazard recognition and food location. To gain a better understanding of chemical communication in fish produced by density changes, this study analyzed the metabolites released by turbot (Scophthalmus maximus) under different stocking densities. The experiment was conducted at low (LD: 3.01 kg/m2), medium (MD: 6.62 kg/m2) and high (HD: 10.84 kg/m2) densities for 15 days. High-throughput non-targeted metabolomics (LC-MS/MS) was used to identify variations in metabolites released into the aquatic environment by turbot at different densities. Results showed that 29 and 47 metabolites were significantly upregulated in the MD and HD groups, respectively, compared with the LD group. Among them, hexadecanedioic acid, xanthine, phenethylamine, proline and styrene were significantly upregulated in the MD vs. LD, HD vs. MD and HD vs. LD. The VIP diagram of OPLS-DA alignment showed that phenethylamine was the most important metabolite shared by MD vs. LD, HD vs. MD and HD vs. LD. Key gene changes in the GH/IGF-1 signaling pathway, HPI axis of turbot were studied using qRT-PCR for density treatment. The results demonstrated that the expression of GH, GHR and IGF-1 was significantly lower, while the expression of CRH and ACTH was higher in the HD group. Additionally, plasma levels of cortisol, glucose, triglycerides and T3 were also highest in the HD group compared with the LD and MD groups. Phenylethylamine concentration was positively correlated with the HPI axis and negatively correlated with the GH/IGF-1 signaling pathway. To investigate the impact of phenethylamine accumulation on turbot, an acute treatment experiment with phenethylamine was set up. Its concentration in the aquatic environment was set at 0 (CON), 10−7 (LP) and 10−5 (HP) mol/L via exogenous addition, and turbot were exposed to these environments for 2 days. There was a high degree of concordance between the GH/IGF-1 signaling pathway (GH, GHR, IGF-1), HPI axis (CRH, ACTH) and plasma physiological changes (cortisol, glucose, triglycerides, T3) in the phenethylamine-treated group and the density-treated group. Therefore, accumulation of phenethylamine with increasing stocking density may be a potential cause of density stress. Phenylethylamine has a dose-dependent and trace effect as a pheromone.

The noise and low clarity in magnetic resonance imaging (MRI) images impede the doctor's diagnosis. An MRI image enhancement method is proposed in the non-subsampled contourlet transform (NSCT) domain. The coefficients of the NSCT are classified as noise component, weak edges component and strong edges component by feature clustering. We modified the transform coefficients to enhance the MRI images. The coefficients corresponding to noise are set to zero, the coefficients corresponding to strong edges are essentially unchanged, and the coefficients corresponding to weak edges are enhanced by a simplified nonlinear gain function. It is shown that the proposed MRI image enhancement method has advantages in visual quality and objective evaluation indexes compared to the state-of-the-art methods.

Abstract Background Mitogen-activated protein kinases (MAPKs) plays essential roles in the development, hormone regulation and abiotic stress response of plants. Nevertheless, a comprehensive study on MAPK family members has thus far not been performed in Tartary buckwheat. Results Here, we identified 16 FtMAPKs in the Fagopyrum tataricum genome. Phylogenetic analysis showed that the FtMAPK family members could be classified into Groups A, B, C and D, in which A, B and C members contain a Thr-Glu-Tyr (TEY) signature motif and Group D members contain a Thr-Asp-Tyr (TDY) signature motif. Promoter cis-acting elements showed that most Pro FtMAPks contain light response elements, hormone response elements and abiotic stress response elements, and several Pro FtMAPks have MYB-binding sites, which may be involved in the regulation of flavonoid biosynthesis-related enzyme gene expression. Synteny analysis indicated that FtMAPKs have a variety of biological functions. Protein interaction prediction suggested that MAPKs can interact with proteins involved in development and stress resistance. Correlation analysis further confirmed that most of the FtMAPK genes and transcription factors involved in the stress response have the same expression pattern. The transient transformation of FtMAPK1 significantly increased the antioxidant enzymes activity in Tartary buckwheat leaves. In addition, we also found that FtMAPK1 can respond to salt stress by up-regulating the transcription abundance of downstream genes. Conclusions A total of 16 MAPKs were identified in Tartary buckwheat, and the members of the MAPK family containing the TDY motif were found to have expanded. The same subfamily members have relatively conserved gene structures and similar protein motifs. Tissue-specific expression indicated that the expression of all FtMAPK genes varied widely in the roots, stems, leaves and flowers. Most FtMAPKs can regulate the expression of other transcription factors and participate in the abiotic stress response. Our findings comprehensively revealed the FtMAPK gene family and laid a theoretical foundation for the functional characterization of FtMAPKs.

Seismic data processing plays a key role in the field of geophysics. The collected seismic data are inevitably contaminated by various types of noise, which makes the effective signals difficult to be accurately discriminated. A fundamental issue is how to improve the signal-to-noise ratio of seismic data. Due to the complex characteristics of noise and signals, it is a challenge for the denoising model to suppress noise and recover weak signals. To suppress random noise in seismic data, we propose a multi-scale deformable convolution neural network denoising model based on U-Net, named MSDC-Unet. The MSDC-Unet mainly contains modules of deformable convolution and dilated convolution. The deformable convolution can change the shape of the convolution kernel to adjust the shape of seismic signals to fit different features, while the dilated convolution with different dilation rates is used to extract feature information at different scales. Furthermore, we combine Charbonnier loss and structure similarity index measure (SSIM) to better characterize geological structures of seismic data. Several examples of synthetic and field seismic data demonstrate that the proposed method is effective in the comprehensive results in terms of quantitative metrics and visual effect of denoising, compared with two traditional denoising methods and two deep convolutional neural network denoising models.

Land salinization is a global environmental problem, and how to manage saline soils and promote healthy ecosystems has become a major challenge. China-Singapore Tianjin Eco-City is located in coastal land reclamation areas, so salinization is severe in this region. In this study, geostatistical methods, the ordinary kriging method, and principal component analysis were used. Vertical sampling was performed over three layers (0–20 cm, 20–40 cm, and 40–60 cm) at 184 locations within the study area to produce a total of 542 soil samples. It was found that areas with soluble salt contents greater than 3000 mg/kg account for over 90% of the study area, and high soluble salt content in surface layer soils is the dominant factor in soil salinization. Na+, Cl−, and SO42- are the primary control factors that determine the coefficient of variation of the soils’ soluble salt content. Total salinity and Na+, Cl−, SO42-, K+, and Mg2+ reflect on the salinization of the soils, while effective phosphorus, available potassium, and soil organic carbon reflect on the state of soil nutrition. Based on our results, we proposed site-specific and scientific soil remediation and greening measures.

ObjectiveThere are a number of symptoms associated with Wilson's disease (WD), including motor function damage. The neuropathological mechanisms underlying motor impairments in WD are, however, little understood. In this study, we explored changes in the motor execution network topology in WD.MethodsWe conducted resting-state functional magnetic resonance imaging (fMRI) on 38 right-handed individuals, including 23 WD patients and 15 healthy controls of the same age. Based on graph theory, a motor execution network was constructed and analyzed. In this study, global, nodal, and edge topological properties of motor execution networks were compared.ResultsThe global topological organization of the motor execution network in the two groups did not differ significantly across groups. In the cerebellum, WD patients had a higher nodal degree. At the edge level, a cerebello-thalamo-striato-cortical circuit with altered functional connectivity strength in WD patients was observed. Specifically, the strength of the functional connections between the cerebellum and thalamus increased, whereas the cortical-thalamic, cortical-striatum and cortical-cerebellar connections exhibited a decrease in the strength of the functional connection.ConclusionThere is a disruption of the topology of the motor execution network in WD patients, which may be the potential basis for WD motor dysfunction and may provide important insights into neurobiological research related to WD motor dysfunction.

Tartary buckwheat (TB) is a pseudocereal rich in flavonoids, mainly including flavonols and anthocyanins. The flavonoid 3′-hydroxylase (F3′H) is a key enzyme in flavonoid biosynthesis and is encoded by two copies in TB genome. However, its biological function and effects on flavonol and anthocyanin synthesis in TB have not been well validated yet. In this study, we cloned the full-length FtF3′H1 gene highly expressed in all tissues (compared with FtF3′H2) according to TB flowering transcriptome data. The corresponding FtF3′H1 protein contains 534 amino acids with the molecular properties of the typical plant F3′H and belongs to the CYP75B family. During the flowering stage, the FtF3′H1 expression was highest in flowers, and its expression pattern showed a significant and positive correlation with the total flavonoids (R2 > 0.95). Bone morphogenetic proteins (BMPs), which belong to the transforming growth factor beta (TGF-β) family, are critical for the control of developmental processes such as dorsal-ventral axis formation, somite and tooth formation, skeletal development, and limb formation. Despite Oplegnathus having typical healing beak-like teeth and tooth development showing a trend from discrete to healing, the potential role of BMPs in the development of the beak-like teeth is incompletely understood. In the present study, 19 and 16 BMP genes were found in O. fasciatus and O. punctatus, respectively, and divided into the BMP2/4/16, BMP5/6/7/8, BMP9/10, BMP12/13/14, BMP3/15 and BMP11 subfamilies. Similar TGFb and TGF_β gene domains and conserved protein motifs were found in the same subfamily; furthermore, two common tandem repeat genes (BMP9 and BMP3a-1) were identified in both Oplegnathus fasciatus and Oplegnathus punctatus. Selection pressure analysis revealed 13 amino acid sites in the transmembrane region of BMP3, BMP7, and BMP9 proteins of O. fasciatus and O. punctatus, which may be related to the diversity and functional differentiation of genes within the BMP family. The qPCR-based developmental/temporal expression patterns of BMPs showed a trend of high expression at 30 days past hatching (dph), which exactly corresponds to the ossification period of the bones and beak-like teeth in Oplegnathus. Tissue-specific expression was found for the BMP4 gene, which was upregulated in the epithelial and mesenchymal tissues of the beak-like teeth, suggesting that it also plays a regulatory role in the development of the beak-like teeth in O. punctatus. Our investigation not only provides a scientific basis for comprehensively understanding the BMP gene family but also helps screen the key genes responsible for beak-like tooth healing in O. punctatus and sheds light on the developmental regulatory mechanism.

Previous studies have suggested that microwave (MW) radiation with certain parameters can induce spatial memory deficits. However, the effect of MW on the topological organization of the brain network is still unknown. This work aimed to investigate the topological organization of the brain network in rats with spatial memory impairments induced by acute microwave (MW) radiation. The Morris water maze (MWM) test and resting-state functional magnetic resonance imaging were performed to estimate the spatial memory ability and brain network topological organization of the rats after MW exposure. Compared with the sham group, the rats exposed to 30 mW/cm2 1.5 GHz MW radiation exhibited a significantly decreased normalized clustering coefficient (γ) (p = 0.002) 1 d after the exposure and a prolonged average escape latency (AEL) (p = 0.014) 3 d after the exposure. Moreover, after 10 mW/cm2 1.5 GHz MW radiation, a significantly decreased γ (p = 0.003) was also observed in the rats, without any changes in AEL. In contrast, no adverse effects on AEL or topological parameters were observed after 9.375 GHz MW radiation. In conclusion, the rats with spatial memory deficits induced by MW radiation exhibited disruptions in the topological organization of the brain network. Moreover, these topological organization disruptions emerged earlier than behavioral symptom onset and could even be found in the rats without a decline in the performance of the spatial memory task. Therefore, it is possible to use the topological parameters of the brain network as early and sensitive indicators of the spatial memory impairments induced by acute MW radiation.

Inflammatory cytokines including TNF-α and IL-1β impair intestinal barrier function in aging by disrupting intestinal tight junction integrity. Icariin (ICA) has a variety of pharmacological effects. Indeed, ICA produces anti-inflammatory, anti-oxidative stress, and inhibitory effects on microRNA (miRNA) expression. This study was to explore whether ICA could alleviate inflammation-associated intestinal barrier function impairment in aging and its underlying mechanism. Of particular interest, network pharmacology prediction indicated the potential therapeutic impacts of ICA for the treatment of colitis. Then, rats were used to study whether ICA has a protective effect on the reduction of tight junctions caused by inflammatory cytokines. Next, Caco-2 cell monolayers were used to explore the mechanism by which ICA alleviates the down-regulation of tight junctions. Network pharmacology prediction revealed that ICA alleviated colitis via suppressing oxidative stress. After ICA intervention, expressions of inflammatory cytokines were reduced, but tight junctions, antioxidant enzymes in aging rats were up-regulated. ICA reversed the TNF-α-induced decrease in abundance of Occludin protein in Caco-2 cell monolayers. Meanwhile, ICA alleviated the increase in permeability and expression of miR-122a. However, the protective effect of ICA was markedly attenuated after transfection with miR-122a mimics. In conclusion, ICA reduced the expressions of Occludin, Claudin1, and Claudin5 in colon, which were related to the reduction of TNF-α and IL-1β and alleviation of colonic in vivore. And ICA attenuated TNF-α-induced Occludin disruption and epithelial barrier impairment by decreasing miR-122a expression in Caco-2 cell monolayers.

With the spread of COVID-19 worldwide, online education is rapidly catching on, even in some underdeveloped countries and regions. Based on Bandura's ternary learning theory and literature review, this paper takes online learning of college students as the research object and conducts an empirical survey on 6,000 college students in East China. Based on literature review and factor analysis and structural equation model, this paper discusses the relationship among learning cognition, learning behavior, and learning environment in online learning of college students. The learning behavior includes interactive communication, self-discipline mechanism, classroom learning, and study after class. The learning environment includes teaching ability, knowledge system, platform support, process control, and result evaluation; learning cognition includes learning motivation, information perception, and adaptability. It is found that the learning environment has a significant positive impact on learning behavior, and learning cognition has a significant positive impact on learning behavior. It is uncertain whether the learning environment significantly impacts learning cognition. At the learning environment level, the teaching ability (0.59) has the most significant impact on the learning environment, followed by result evaluation (0.42), platform support (0.40), process control (0.33), and knowledge system (0.13). In terms of the influence on learning behavior, classroom learning has the most significant impact (0.79), followed by self-discipline mechanism (0.65), study after class (0.54), and interactive communication (0.44). In terms of learning cognition, information perception (0.62) has the most significant influence, followed by learning motivation (0.50) and adaptability (0.41). This paper suggests strengthening the construction of platforms and digital resources to create a more competitive learning environment. Improve process management and personalized online services, constantly stimulate students' enthusiasm for independent online learning, and cultivate students' online independent learning ability to promote the sustainable and healthy development of online education.

Purpose This study aimed to assess the protective effects of silibinin on blue light-emitting diode (LED)-induced retinal ganglion cells (RGCs) damage.Methods Silibinin was applied in RGCs damage in vitro model to test its protective effects. Cell viability was assessed with the MTT method and cell apoptosis was evaluated by TUNEL and Annexin V/propidium iodide staining. The expressions of apoptosis related proteins and influenced signalling pathways were measured using western blotting and immunohistochemistry. Inflammatory factors induced by RGC damage were detected using ELISA method.Results It was found that silibinin in 50 and 100 μM treatment showed a significant protective effect in RGCs under blue light damage. Apoptosis assay showed that silibinin treatment could significantly improve the apoptotic status of RGCs. When the potentially affected signal pathway was considered, blue light would down-regulate the expression of MEK1/ERK/CREB. The levels of inflammatory factors (TNF-α, IL-1β, IL-6 and IL-10) were significantly regulated by silibinin treatment.Conclusions Silibinin pretreatment would demonstrate protective effect against blue light induced acute RGCs damage. Silibinin treatment has a direct suppression of apoptosis and inflammation through the activation of MEK/ERK/CREB pathway in vitro.

Tartary buckwheat (Fagopyrum tataricum Gaertn.) is a coarse cereal with strongly abiotic resistance. The MYB family plays a regulatory role in plant growth, development, and responses to biotic and abiotic stresses. However, the characteristics and regulatory mechanisms of MYB transcription factors in Tartary buckwheat remain unclarified. Here, this study cloned the FtMYB22 gene from Tartary buckwheat, and investigated its involvement in responding to individual water deficit and salt stress in Arabidopsis. Sequence analysis highlighted that the N-termini of FtMYB22 contained two highly conserved SANT domains and one conserved domain from the SG20 subfamily. Nucleus-localized FtMYB22 did not have individual transcriptional activation activity. Water deficiency and salt stress induced the high expression of the GUS gene, which was driven by the promoter of FtMYB22. Yeast stress experiments showed that the overexpression of FtMYB22 significantly reduced the growth activity of transgenic yeast under water deficit or salt stress. Consistently, the overexpression of FtMYB22 reduced the salt and water deficit stress resistance of the transgenic plants. In addition, physiological parameters showed that transgenic plants had lower proline and antioxidant enzyme activity under stress conditions. Compared to the wild-type (WT), transgenic plants accumulated more malondialdehyde (MDA), H2O2, and O2−; they also showed higher ion permeability and water loss rates of detached leaves under stress treatments. Notably, FtMYB22 was involved in plant stress resistance through an ABA-dependent pathway. Under stress conditions, the expression of RD29A, RD29B, PP2CA, KIN1, COR15A, and other genes in response to plant stress in transgenic lines was significantly lower than that in the WT (p < 0.05). Furthermore, yeast two-hybrid assay showed that there was a significant interaction between FtMYB22 and the ABA receptor protein RCAR1/2, which functioned in the ABA signal pathway. Altogether, FtMYB22, as a negative regulator, inhibited a variety of physiological and biochemical reactions, affected gene expression and stomatal closure in transgenic plants through the ABA-dependent pathway, and reduced the tolerance of transgenic Arabidopsis to water deficiency and salt stress. Based on these fundamental verifications, further studies would shed light on the hormone signal response mechanism of FtMYB22.

Abiotic stresses such as drought and salinity are major environmental factors limiting plant productivity. Autophagy-related genes are extensively involved in plant growth, development, and adverse stress responses, which have not yet been characterized in Tartary buckwheat (Fagopyrum tataricum, TB). In this study, we verified that drought stress could induce autophagy in TB roots. Next, 49 FtATGs in the whole genome of TB were identified. All FtATGs were randomly distributed in 8 known chromosomes, while 11 FtATGs were predictably segmental repeats. As the core component of autophagy, there were 8 FtATG8s with similar gene structures in TB, while FtATG8s showed high expression at the transcription level under drought and salt stresses. The cis-acting element analysis identified that all FtATG8 promoters contain light-responsive and MYB-binding elements. FtATG8s showed a cell-wide protein interaction network and strongly correlated with distinct stress-associated transcription factors. Furthermore, overexpression of FtATG8a and FtATG8f enhanced the antioxidant enzyme activities of TB under adverse stresses. Remarkably, FtATG8a and FtATG8f may be vital candidates functioning in stress resistance in TB. This study prominently aids in understanding the biological role of FtATG genes in TB.

The experience of environmental governance in the Yangtze River Delta has formed the practical paths of cross-administrative cooperation and eco-civilization adaptation to economic development. As a result of a scientific analysis of policy texts on collaborative environmental governance in this region, this paper explores differences and core concerns, uncovering the development vein and mapping out the internal logic in order to provide a reference example for multi-regional governance. The policy has shifted from decentralization to authority, from universality to precision, from sustainable development to a community of common ecological destiny, from authoritarianism to co-governance, and from institutional norms to propaganda and guidance. Since the beginning of the new century, the internal logic of environmental governance policy in the Yangtze River Delta has been in line with the trend of coordinated development. In the future, efforts should be made to deepen the trinity mechanism of decision making, implementation, and supervision. When making decisions, we should further emphasize the unified standard of centralized environmental management and adhere to precise pollution control. Implementation will gradually establish the three-dimensional coordination mechanism of region, function, and role; supervision will involve the platform “internet + environment”, and the assessment will involve “pressure mechanism + environment”.

Electromagnetic pulse (EMP) radiation was reported to be harmful to hippocampal neurons. However, the mechanism underlying EMP-induced neuronal damage remains unclear. In this paper, for the first time, we attempted to investigate the involvement of ferroptosis in EMP-induced neuronal damage and its underlying mechanism. In vivo studies were conducted with a rat model to examine the association of ferroptosis and EMP-induced hippocampal neuronal damage. Moreover, in vitro studies were conducted with HT22 neurons to investigate the underlying mechanism of EMP-induced neuronal ferroptosis. In vivo results showed that EMP could induce learning and memory impairment of rats, ferroptotic morphological damages to mitochondria, accumulation of malonaldehyde (MDA) and iron, overexpression of prostaglandin-endoperoxide synthase 2 (PTGS2) mRNA, and downregulation of GPX4 protein in rat hippocampus. In vitro results showed that EMP could induce neuronal death, MDA accumulation, iron overload, PTGS2 overexpression, and GPX4 downregulation in HT22 neurons. These adverse effects could be reversed by either lipid peroxides scavenger ferrostatin-1 or overexpression of GPX4. These results suggest that EMP radiation can induce ferroptosis in hippocampal neurons via a vicious cycle of lipid peroxides accumulation and GSH/GPX4 axis downregulation. Lipid peroxides and the GSH/GPX4 axis provide potential effective intervention targets to EMP-induced hippocampal neuronal damage.

The spotted knifejaw (Oplegnathus punctatus) is a marine economic fish with high ecological value, food value, and fishing value, and its growth has obvious sex dimorphism. The rapid identification of its sex is beneficial to the development of sex determination and breeding. In this study, the method of comparative genomics and PCR amplification was used to further establish a rapid detection method for the recombinant RhoGEF10 gene in O. punctatus, which can quickly, accurately, and efficiently identify the sex of the O. punctatus to be tested. The homologous comparison results of male and female individuals showed that the DNA fragment length of the RhoGEF10 gene on the X1 chromosome was 326 bp, and the DNA fragment length on the Y chromosome was 879 bp. Therefore, it can be concluded that there is an insert fragment of 553 bp on the Y chromosome. PCR amplification results showed that the two DNA fragments of 879 bp and 326 bp were amplified in the Y chromosome and X1 chromosome of the male O. punctatus (X1X2Y), respectively, and the 879 bp fragment was a unique marker fragment of the recombinant RhoGEF10 gene; The female O. punctatus (X1X1X2X2) only a single DNA fragment of 326 bp was amplified. At the same time, the inserted fragment of the male individual resulted in partial inactivation of the RhoGEF10 protein, which in turn resulted in a slowing of peripheral nerve conduction velocity and thinning of the myelin sheath in male O. punctatus. The method shortens the time for accurate identification of the O. punctatus RhoGEF10 gene recombination and improves the detection efficiency. It is of great significance and application value in the research of nerve conduction and myelin development, male and female sex identification, the preparation of high male seedlings, and family selection based on the RhoGEF10 gene in the O. punctatus.

Soil salinization is a global problem that limits crop yields and threatens agricultural development. Auxin-induced expansins contribute to plant salt tolerance through cell wall loosening. However, how auxins and expansins contribute to the adaptation of the halophyte quinoa (Chenopodium quinoa) to salt stress has not yet been reported. Here, auxin was found to contribute to the salt tolerance of quinoa by promoting the accumulation of photosynthetic pigments under salt stress, maintaining enzymatic and nonenzymatic antioxidant systems and scavenging excess reactive oxygen species (ROS). The Chenopodium quinoa expansin (Cqexpansin) family and the auxin pathway gene family (Chenopodium quinoa auxin response factor (CqARF), Chenopodium quinoa auxin/indoleacetic acid (CqAux/IAA), Chenopodium quinoa Gretchen Hagen 3 (CqGH3) and Chenopodium quinoa small auxin upregulated RNA (CqSAUR)) were identified from the quinoa genome. Combined expression profiling identified Chenopodium quinoa α-expansin 50 (CqEXPA50) as being involved in auxin-mediated salt tolerance. CqEXPA50 enhanced salt tolerance in quinoa seedlings was revealed by transient overexpression and physiological and biochemical analyses. Furthermore, the auxin pathway and salt stress-related genes regulated by CqEXPA50 were identified. The interaction of CqEXPA50 with these proteins was demonstrated by bimolecular fluorescence complementation (BIFC). The proteins that interact with CqEXPA50 were also found to improve salt tolerance. In conclusion, this study identified some genes potentially involved in the salt tolerance regulatory network of quinoa, providing new insights into salt tolerance.

Abstract Background Castleman’s disease (CD) is an uncommon type of benign proliferation of the lymphoid tissue, characterized by local or systemic lymphadenopathy that most frequently appears in the mediastinum; involvement of the kidney is uncommon, and proliferation originating from the kidney is extremely rare. Herein, we report a rare case of hyaline vascular Castleman’s disease (HV-CD) in a 56-year-old male patient and discuss its morphological characteristics and differential diagnoses including mantle cell lymphoma (MCL), follicular lymphoma (FL), and nodal marginal zone lymphoma (NMZL). Case presentation A right upper-middle renal mass was detected after physical examination in a 56-year-old man without any clinical symptoms and a previous partial resection of the right kidney. Microscopically, the lymphoid follicles were increased in number and had expanded mantle zones and atrophic germinal centers. Vascular proliferation and hyalinization in the interfollicular zones were observed. Immunohistochemical staining showed CD20-positive cells in the mantle zones; CD21 and CD35 were expressed in the dendritic cells, CD3 was positive in a small number of T cells, and CD38 and CD138 were positive in the plasma cells. Additionally, Ki-67 expression was positive in the follicle centers. In contrast, staining for Bcl-2 in the germinal centers and cyclin D1 were negative. The immunohistochemical analysis combined with the morphological results supported the diagnosis of HV-CD. The patient recovered well after surgery. Conclusions Primarily renal HV-CD without lymph node hyperplasia or clinical symptoms is extraordinarily rare and different from the multicentric-type CD (MCD) with kidney involvement. Therefore, it is extremely important to improve the awareness of this diagnosis. Attention should be paid to the difference between HV-CD and common lymph node reactive hyperplasia, MCL, FL, NMZL, and so on. To avoid misdiagnosis as a renal malignant tumor requiring radical resection, distinguishing these diseases is crucial.

Abstract Background Tartary buckwheat (Fagopyrum tataricum) is an edible cereal crop whose sprouts have been marketed and commercialized for their higher levels of anti-oxidants, including rutin and anthocyanin. UDP-glucose flavonoid glycosyltransferases (UFGTs) play an important role in the biosynthesis of flavonoids in plants. So far, few studies are available on UFGT genes that may play a role in tartary buckwheat flavonoids biosynthesis. Here, we report on the identification and functional characterization of seven UFGTs from tartary buckwheat that are potentially involved in flavonoid biosynthesis (and have varying effects on plant growth and development when overexpressed in Arabidopsis thaliana.) Results Phylogenetic analysis indicated that the potential function of the seven FtUFGT proteins, FtUFGT6, FtUFGT7, FtUFGT8, FtUFGT9, FtUFGT15, FtUFGT40, and FtUFGT41, could be divided into three Arabidopsis thaliana functional subgroups that are involved in flavonoid biosynthesis of and anthocyanin accumulation. A significant positive correlation between FtUFGT8 and FtUFGT15 expression and anthocyanin accumulation capacity was observed in the tartary buckwheat seedlings after cold stress. Overexpression in Arabidopsis thaliana showed that FtUFGT8, FtUFGT15, and FtUFGT41 significantly increased the anthocyanin content in transgenic plants. Unexpectedly, overexpression of FtUFGT6, while not leading to enhanced anthocyanin accumulation, significantly enhanced the growth yield of transgenic plants. When wild-type plants have only cotyledons, most of the transgenic plants of FtUFGT6 had grown true leaves. Moreover, the growth speed of the oxFtUFGT6 transgenic plant root was also significantly faster than that of the wild type. At later growth, FtUFGT6 transgenic plants showed larger leaves, earlier twitching times and more tillers than wild type, whereas FtUFGT15 showed opposite results. Conclusions Seven FtUFGTs were isolated from tartary buckwheat. FtUFGT8, FtUFGT15, and FtUFGT41 can significantly increase the accumulation of total anthocyanins in transgenic plants. Furthermore, overexpression of FtUFGT6 increased the overall yield of Arabidopsis transgenic plants at all growth stages. However, FtUFGT15 shows the opposite trend at later growth stage and delays the growth speed of plants. These results suggested that the biological function of FtUFGT genes in tartary buckwheat is diverse.

Abstract Background Because flavonoids and trichomes play crucial roles in plant defence, their formation requires fine transcriptional control by multiple transcription factor families. However, little is known regarding the mechanism of the R2R3-MYB transcription factors that regulate both flavonoid metabolism and trichome development. Results Here, we identified a unique SG4-like-MYB TF from Tartary buckwheat, FtMYB8, which harbours the C2 repression motif and an additional TLLLFR repression motif. The expression profiles of FtMYB8 combined with the transcriptional activity of P FtMYB8 promoter showed that FtMYB8 mRNA mainly accumulated in roots during the true leaf stage and flowering stage and in bud trichomes and flowers, and the expression of this gene was markedly induced by MeJA, ABA and UV-B treatments but repressed by dark treatment. Overexpression of FtMYB8 in Arabidopsis reduces the accumulation of anthocyanin/proanthocyanidin by specifically inhibiting TT12 expression, which may depend on the interaction between FtMYB8 and TT8. Interestingly, this interaction may also negatively regulate the marginal trichome initiation in Arabidopsis leaves. Conclusions Taken together, our results suggest that FtMYB8 may fine-tune the accumulation of anthocyanin/proanthocyanidin in the roots and flowers of Tartary buckwheat by balancing the inductive effects of transcriptional activators, and probably regulate trichome distribution in the buds of Tartary buckwheat.

Effect of the diamond nanoparticle on the friction property of the sliding friction pair is investigated using the molecular dynamics simulation. A molecular dynamics simulation model without the diamond nanoparticle is also set up. The simulation is conducted at an external load of 60 nN. The results confirm that adding diamond nanoparticle can improve the friction property. With increasing the diameter, the angular velocity, and the indentation depth decrease inversely. Also, when the diameter varies from 32.13 Å to 49.98 Å, the friction force, and the contact area are both inversely proportional to 3.6 power of the diameter. The relationship between friction force and contact area shows a linear increase law, which indicates that friction force is affected by the contact area.

Hydrostatic journal bearings possess superior motion accuracy owing to their error averaging effect and often work under different speeds and external loads in precision applications. This paper researches the error averaging effect of hydrostatic journal bearings considering the influences of shaft rotating speeds and external loads. A new mathematical model is established based on a time division method where the rotational movement of the shaft is divided into many time intervals. The averaging coefficients of hydrostatic journal bearings are analyzed in detail. The results show that the averaging coefficients are affected by the combined effects of rotational speeds, and external loads and wave number of roundness errors. Finally, some useful guidelines are given for the accuracy design of hydrostatic journal bearings.

Coming along with high water reuse in sustainable and intensive recirculating aquaculture systems (RASs), the waste products of fish in rearing water is continuously accumulated. Nitrate, the final product of biological nitrification processes, which may cause aquatic toxicity to fish in different degrees when exposed for a long time. Therefore, the present study was conducted to evaluate the impact of chronic nitrate exposure on intestinal morphology, immune status, barrier function, and microbiota of juvenile turbot. For that, groups of juvenile turbot were exposed to 0 (control check, CK), 50 (low nitrate, L), 200 (medium nitrate, M), and 400 (high nitrate, H) mg L−1 nitrate-N in small-sized recirculating aquaculture systems. After the 60-day experiment period, we found that exposure to a high concentration of nitrate-N caused obvious pathological damages to the intestine; for instance, atrophy of intestinal microvilli and necrosis in the lamina propria. Germ cells play a key role in gonad development. As precursors, primordial germ cells (PGCs) are particularly important for germline formation. However, the origination and migration patterns of PGCs are poorly studied in marine fish, especially for viviparous economic species. The vasa gene has been widely used as a germ cell marker to identify a germline because vasa RNA is a component of germ plasm. In this study, we described the expression pattern of black rockfish (Sebastes schlegelii) vasa (Ssvas) in gonadal formation and development by in situ hybridization. The results showed that Ssvas failed in localization at the cleavage furrows until the late gastrula stage, when PGCs appeared and migrated to the genital ridge and formed elongated gonadal primordia at 10 days after birth. This study firstly revealed the PGCs origination and migration characteristics in viviparous marine fish. Furthermore, we microinjected chimeric mRNA containing EGFP and the 3′untranslated region (3′UTR) of Ssvas into zebrafish (Danio rerio) and marine medaka (Oryzias melastigma) fertilized eggs for tracing PGCs. We found that, although Sebastes schlegelii lacked early localization, similar to red seabream (Pagrus major) and marine medaka, only the 3′UTR of Ssvas vasa 3′UTR of black rockfish was able to label both zebrafish and marine medaka PGCs. In comparison with other three Euteleostei species, besides some basal motifs, black rockfish had three specific motifs of M10, M12, and M19 just presented in zebrafish, which might play an important role in labeling zebrafish PGCs. These results will promote germ cell manipulation technology development and facilitate artificial reproduction regulation in aquaculture.
