The healing of living tendons is the result of the combined effects of endogenous and exogenous mechanisms. Biodegradable materials are widely used in the prevention of tendon adhesions caused by exercise-induced tendon injury. Decimeter waves and sodium hyaluronate are common surgical biomaterials to prevent tendon adhesion. This study used the male white Laiheng chicken as the animal model and decimeter waves and sodium hyaluronate as the research object to analyze their effects on controlling and preventing tendon adhesion and promoting tendon healing during sports injury surgery. The study provides evidence indicating that biodegradable materials, decimeter waves, and sodium hyaluronate can encourage the healing of the of the tendon after sports injuries.

The surface-enhanced Raman scattering (SERS) technique has displayed a broad application prospect in disease diagnosis owing to its high sensitivity, fast responsiveness, and high specificity. In this study, we developed a SERS-based immunoassay for the detection of cryptococcal capsular polysaccharide (glucuronoxylomannan, GXM), which is an important biomarker of cryptococcosis. The coupled localized surface plasmon resonance effect between magnetic SERS substrates and SERS tags gave rise to an enhanced Raman signal upon the formation of sandwich complexes, which contributes to the sensitive and specific detection of GXM. As a result, the developed method provided a lower limit of detection (1.25 ng/mL) than conventional methods, such as latex agglutination, lateral flow assay, and enzyme-linked immunosorbent assay. Additionally, a recovery experiment was performed to detect GXM in human serum, which also validated the potential advantages of a SERS-based immunoassay in the clinical diagnosis of cryptococcosis.

Re-endothelialization is a critical problem to inhibit postoperative restenosis, and gene delivery exhibits great potential in rapid endothelialization. Unfortunately, the therapeutic effect is enormously limited by inefficient specificity, poor biocompatibility and in vivo stability owing largely to the complicated in vivo environment. Herein, we developed a series of platelet membrane (PM) cloaked gene complexes based on natural bovine serum albumin (BSA) and polyethyleneimine (PEI). The gene complexes aimed to accelerate re-endothelialization for anti-restenosis via pcDNA3.1-VEGF165 (VEGF) plasmid delivery. Based on BSA and PM coating, these gene complexes exhibited good biocompatibility, stability with serum and robust homing to endothelium-injured site inherited from platelets. Besides, they enhanced the expression of VEGF protein by their high internalization and nucleus accumulation efficiency, and also substantially promoted migration and proliferation of vascular endothelial cells. The biological properties were further optimized via altering PEI and PM content. Finally, rapid recovery of endothelium in a carotid artery injured mouse model (79% re-endothelialization compared with model group) was achieved through two weeks' treatment by the PM cloaked gene complexes. High level of expressed VEGF in vivo was also realized by the gene complexes. Moreover, neointimal hyperplasia (IH) was significantly inhibited by the gene complexes according to in vivo study. The results verified the great potential of the PM cloaked gene complexes in re-endothelialization for anti-restenosis. STATEMENT OF SIGNIFICANCE: Rapid re-endothelialization is a major challenge to inhibit postoperative restenosis. Herein, a series of biodegradable and biocompatible platelet membrane (PM) cloaked gene complexes were designed to accelerate re-endothelialization for anti-restenosis via pcDNA3.1-VEGF165 (VEGF) plasmid delivery. The PM cloaked gene complexes provided high VEGF expression in vascular endothelial cells (VECs), rapid migration and proliferation of VECs and robust homing to endothelium-injured site. In a carotid artery injured mouse model, PM cloaked gene complexes significantly promoted VEGF expression in vivo, accelerated re-endothelialization and inhibited neointimal hyperplasia due to their good biocompatibility and superior specificity. Overall, the optimized PM cloaked gene complexes overcomes multiple obstacles in gene delivery for re-endothelialization and can be a promising candidate for gene delivery and therapy of postoperative restenosis.

Soil salinity constrains agricultural development in arid regions. Flax is an economically important crop in many countries, and screening or breeding salinity-resistant flax cultivars is necessary. Based on the previous screening of flaxseed cultivars C71 (salt-sensitive) and C116 (salt-tolerant) as test materials, flax seedlings stressed with different concentrations of NaCl (0, 100, 150, 200, and 250 mmol/L) for 21 days were used to investigate the effects of salt stress on the growth characteristics, osmotic regulators, and antioxidant capacity of these flax seedlings and to reveal the adaptive responses of flax seedlings to salt stress. The results showed that plant height and root length of flax were inhibited, with C116 showing lower growth than C71. The concentrations of osmotic adjustment substances such as soluble sugars, soluble proteins, and proline were higher in the resistant material, C116, than in the sensitive material, C71, under different concentrations of salt stress. Consistently, C116 showed a better rapid scavenging ability for reactive oxygen species (ROS) and maintained higher activities of antioxidant enzymes to balance salt injury stress by inhibiting growth under salt stress. A transcriptome analysis of flax revealed that genes related to defense and senescence were significantly upregulated, and genes related to the growth and development processes were significantly downregulated under salt stress. Our results indicated that one of the important adaptations to tolerance to high salt stress is complex physiological remediation by rapidly promoting transcriptional regulation in flax.

In this study, duplex surface treatments were used to prepare a ZrO2/Cr bilayer coating on zirconium alloy cladding for enhancing the wear and corrosion behaviors. The surface and cross-section morphology of coated Zr-4 alloy was characterized; the results show that the Cr- and ZrO2/Cr-coated samples had similar morphology, and more obvious surface undulates could be observed on the ZrO2/Cr coating than the pure Cr coating owing to the rough surface of the plasma electrolytic oxidation coating. Wear and electrochemical behavior in 1200 mg/L H3BO3 and 2.2 mg/L LiOH solutions of original and coated Zr-4 alloy were investigated. The electrochemical corrosion test indicated the coated Zr-4 alloy exhibited better corrosion resistance behavior than the original Zr-4 alloy. The potentiodynamic polarization curves and corrosion morphology suggest the pitting corrosion occurred on the surface of the original and coated Zr-4 alloy. The ZrO2/Cr-coated Zr-4 alloy had better corrosion resistance due to the dual protection of the PEO layer and Cr coating. The wear behavior of the original and coated Zr-4 alloy was also investigated under a constant load of 5 N. The results reveal that the coated Zr-4 alloy had better wear resistance, and the PEO layer was found to significantly enhance the wear resistance of the Zr-4 alloy.

To compare the oncological outcomes and major complications of laparoscopic radical hysterectomy (LRH) and abdominal radical hysterectomy (ARH) for stage IB1 cervical cancer (FIGO 2009) with a tumour size less than 2 cm.We retrospectively compared the oncological outcomes and major complications of 1207 stage IB1 cervical cancer patients with a tumour size less than 2 cm who received LRH (n = 546) or ARH (n = 661) in 37 hospitals.(1) There was no significant difference in 3-year overall survival (OS; 97.3% vs. 98.5%, P = 0.288) or 3-year disease-free survival (DFS; 95.1% vs. 95.4%, P = 0.792) between LRH (n = 546) and ARH (n = 661).(2) The rate of any 1 complication refers to the incidence of one or more complications in a patient, which was higher with LRH than ARH (OR = 4.047, 95% CI = 2.035-8.048, P 0.001). Additionally, intraoperative complications occurred with LRH (OR = 12.313, 95% confidence intervals [CI] = 1.571-96.493, P = 0.017), and postoperative complications (OR = 3.652, 95% CI = 1.763-7.562, P 0.001) were higher with LRH than ARH. The ureteral injury rate was higher with LRH than with ARH (1.50% vs. 0.20%, OR = 9.814, 95% CI = 1.224-78.712, P = 0.032). The ureterovaginal fistula rate was higher with LRH than ARH. The rates of obturator nerve injury, bladder injury, vesicovaginal fistula, rectovaginal fistula, venous thromboembolism, bowel obstruction, chylous leakage, pelvic haematoma, and haemorrhage were similar between the groups.The oncological outcomes of LRH and ARH for stage IB1 cervical cancer patients with a tumour size less than 2 cm do not differ significantly. However, incidences of any 1 complication, intraoperative complications, and postoperative complications were higher with LRH than ARH, with complications manifesting mainly as ureteral injury and uterovaginal fistula.

Respiratory syncytial virus (RSV) is a major cause of acute respiratory tract infections in children and is a public health threat globally. To investigate the spatiotemporal dynamics of RSV evolution, we performed systematic phylogenetic analysis using all available sequences from the GenBank database, together with sequences from Shanghai, China. Both RSV-A and RSV-B appear to have originated in North America, with an inferred origin time of 1954.0 (1938.7-1967.6) and 1969.7 (1962.6-1975.5), respectively. BA-like strains of RSV-B, with a 60-nt insertion, and the ON1 strain of RSV-A, with a 72-nt insertion, emerged in 1997.6 (1996.2-1998.6) and 2010.1 (2009.1-2010.3), respectively. Since their origin, both genotypes have gradually replaced the former circulating genotypes to become the dominant strain. The population dynamic of RSV-A showed a seasonal epidemic pattern with obvious expansion in the periods of 2006-2007, 2010-2011, 2011-2012, and 2013-2014. Thirty fixed amino acid substitutions were identified during the divergence of NA4 from GA1 genotypes of RSV-A, and 13 were found during the divergence of SAB4 from GB1 of RSV-B. Importantly, ongoing evolution has occurred since the emergence of ON1, including four amino acid substitutions (I208L, E232G, T253K, and P314L). RSV-A genotypes GA5, NA4, NA1, and ON1 and RSV-B genotypes CB1, SAB4, BA-C, BA10, BA7, and BA9 were co-circulating in China from 2005 to 2015. In particular, RSV-A genotype ON1 was first detected in China in 2011, and it completely replaced GA2 to become the predominant strain after 2016. These data provide important insights into the evolution and epidemiology of RSV.

Although nanostructures and oxide dispersion can reduce radiation-induced damage in materials and enhance radiation tolerance, previous studies prove that MoS2 nanocomposite films subjected to several dpa heavy ion irradiation show significant degradation of tribological properties. Even in YSZ-doped MoS2 nanocomposite films, irradiation leads to obvious disordering and damage such as vacancy accumulation to form lamellar voids in the amorphous matrix, which accelerates the failure of lubrication. However, after thermal annealing in vacuum, YSZ-doped MoS2 nanocomposite films exhibit high irradiation tolerance, and their wear duration remains unchanged and the wear rate was nearly three orders of magnitude lower than that of the as-deposited films after 7 dpa irradiation. This successful combination of anti-irradiation and self-adaptive lubrication mainly results from the manipulation of the nanosize and the change of composition by annealing. Compared with the smaller nanograins in as-deposited MoS2/YSZ nanocomposite films, the thermally annealed MoS2 nanocrystals (7-15 nm) with fewer intrinsic defects exhibited remarkable stabilization upon irradiation. Abundant amorphous nanocrystal phases in ion-irradiated thermally annealed films, where each has advantages of their own, greatly inhibit accumulation of voids and crack growth in irradiation; meanwhile, they can be easily self-assembled under induction of friction and achieve self-adaptive lubrication.

Caregiving services are currently the weakest part of China’s social security system for the elderly. It is well needed to investigate the fac-tors affecting the unmet care needs of the elderly with disabilities. Based on the Behavioral Model of Health Services Use (BMHSU), this paper explores the needs and influencing factors of care services for the disabled elderly in urban and rural areas using data from the 2018 Project of Social Policy Support System for Disadvantaged Families in China. The demand for care services of the disabled elderly in central and western areas is significantly higher than that in eastern, along with that in rural areas significantly higher than that in urban areas. The demands for care services of the disabled elderly in urban and rural areas are significantly affected by tendency factors, resource factors, and need factors. Urban and rural attributes, worried pension and LCI are the common influencing factors for the care service demand of the disabled elderly from economically disadvantaged families and ordinary families. The demands for care services of the disabled elderly were associated with tendencies, resources, and needs, increased chronic disease prevention and mental health services benefit caregivers.

Alveolar macrophages (AMs) are specialized tissue-resident macrophages that orchestrate the immune response in allergic inflammation and asthma. However, what signals direct AMs to cross talk with other immune cells remains unclear. Here, we report that autocrine motility factor receptor (AMFR), an endoplasmic reticulum–resident E3 ubiquitin ligase, is upregulated in AMs of asthma and is critical for this condition. AMFR deficiency significantly decreased allergy-induced T helper 2 (Th2) and eosinophilic inflammation, with less granulocyte-macrophage colony-stimulating factor (GM-CSF) production in AMs. Mechanistically, following thymic stromal lymphopoietin (TSLP) stimulation, AMFR associated directly with cytokine-inducible SH2-containing protein (CIS), induced the ubiquitination of Lys48-linked polyubiquitination of CIS, and consequently blocked the inhibitory effect of CIS on signal transducer and activator of transcription 5 (STAT5) phosphorylation and the downstream pathway activation in AMs. In conclusion, our results demonstrate that AMFR serves a crucial role in promoting inflammation in asthma through regulating AM function, and may emerge as a new potential drug target for asthma therapy.

Soil salinization seriously affects the growth and distribution of flax. However, there is little information about the salt tolerance of flax. In this study, the salt tolerance of 200 diverse flax accessions during the germination stage was evaluated, and then the Genome-wide Association Study (GWAS) was carried out based on the relative germination rate (RGR), relative shoot length (RSL) and relative root length (RRL), whereby quantitative trait loci (QTLs) related to salt tolerance were identified. The results showed that oil flax had a better salt tolerance than fiber flax. A total of 902 single nucleotide polymorphisms (SNPs) were identified on 15 chromosomes. These SNPs were integrated into 64 QTLs, explaining 14.48 to 29.38% (R2) of the phenotypic variation. In addition, 268 candidate genes were screened by combining previous transcriptome data and homologous gene annotation. Among them, Lus10033213 is a single-point SNP repeat mapping gene, which encodes a Glutathione S-transferase (GST). This study is the first to use GWAS to excavate genes related to salt tolerance during the germination stage of flax. The results of this study provide important information for studying the genetic mechanism of salt tolerance of flax, and also provide the possibility to improve the salt tolerance of flax.

Nursing informatics competency has crucial functions in the development of a hospital and patient safety. On the basis of the theory of self-concept, there has been an inadequate study of the influence of transformational leadership on nursing informatics competency mediated by innovation self-efficacy. A descriptive correlational research design was conducted for 586 Chinese nurses from five public hospitals. The measurements were obtained from a questionnaire on transformational leadership, nursing informatics competency, innovation self-efficacy, and a sociodemographic datasheet. We identified a positive correlation between transformational leadership and nursing informatics competency. Innovation self-efficacy had a mediating effect. Managers need to provide measures to shape their transformational leadership style and promote innovation self-efficacy of nurses, which enhances nursing informatics competency of nurses.

BackgroundAnti-CD3-based bispecific T cell engagers (BiTE) showed limited clinical efficacy in solid tumors. This is partly due to the difficulty and scarcity of T cells infiltrating into tumor microenvironment (TME). Macrophages are major component of immune cell infiltrate in the TME and can constitute up to 50% of a solid tumor mass. Signal regulatory protein-α (SIRPα) is a major myeloid cell inhibitory receptor that engages the ”don’t eat me” signal from CD47 expressed on tumors. Similar to BITE where T cells are activated by the CD3 antibody, we constructed a novel bispecific macrophage engager (BiME) where macrophage is activated by a SIRPα inhibitory antibody that is directed to a particular tumor via the tumor associated antigen (TAA) antibody, resulting in phagocytosis of the tumor. Previously, we have developed a human SIRPα monoclonal antibody called ES004-B4, that blocks CD47 binding. ES004 greatly augments antibody dependent cellular phagocytosis killing of cancer cell lines. We utilize ES004 to make a BiME called ES028 where the SIRPα antibody is linked to a Claudin18.2 antibody, targeting claudin18.2-expressing cancers like gastric cancer.MethodsThrough Elpiscience BiME platform, we have generated a panel of anti-Claudin18.2/SIRPα bispecific antibodies, including different anti-Claudin18.2 arm and anti-SIRPα arm positions, ratios and IgG isotypes. The binding and blocking ability of these bispecific antibodies were evaluated by ELISA and FACS. In vitro function activity was determined by phagocytosis assay using human monocyte derived macrophage and mouse bone marrow derived macrophage. In vivo anti-tumor efficacy was investigated in a syngeneic tumor model with hSIRPα knock-in mice.ResultsWe demonstrate that an anti-Claudin18.2/SIRPα bispecific antibody ES028 exhibited super anti-cancer effects, with improved phagocytosis of cancer in vitro and extended survival of claudin18.2-expressing tumor burden mice. In the syngeneic model, ES028 showed almost 100% tumor growth inhibition in the SIRPα knock-in models without causing cytokine storm. ES028 could not induce phagocytosis of Claudin18.2 negative cells, proving its specificity and selectivity.ConclusionsWe have developed a bispecific macrophage engager (BiME) platform that is capable of activating phagocytosis to kill cancer cells. A number of preclinical programs are ongoing to design specific BiME for particular tumor indication. Our SIRPα-based macrophage engager ES028 is the first to have reached the pre-clinical stage with a demonstrated favorable safety profile and promising therapeutic efficacy. Taken together, these results indicate that the bi-specific macrophage engager platform is feasible and could be a powerful weapon in the battle towards the elimination of cancers.

This study examined gender differences in mental health of bereaved parents related to the gender of deceased only child in China, an only-child society with traditional culture of son preference, using data drawn from the China Family Planning Survey on Vulnerable Households in 2017. The findings indicated that parents with deceased only child suffered from more negative mental health symptoms than nonbereaved parents. For only-child-death families, there were no statistically significant gender differences in mental health of parents, and the gender of the deceased only child was basically unrelated to maternal/paternal mental health. Due to the implementation of one-child policy in China, both sons and daughters are highly prized and equally relied on by aging parents owing to the irreplaceability of the only child, which might moderate the effects of traditional culture of son preference on bereaved parental mental health.

A rapid growing number of cases on victimization among left-behind children were reported and child victimization has become an important social issue in China. The frequent victimization events, together with its negative impact, lead the Chinese government to actively embark on innovation in protection service against victimization for left-behind children, especially at school and community levels. Drawing on representative data from Survey on Children and Old Adults Left-behind in Rural China, this study aims to test whether school-based and community-based protection service innovations have significant association with left-behind children's victimization incidence in China. According to the regression results, this study found that there was significantly negative association between protection service and victimization incidence among left-behind children in China. To be specific, school-based protection service was significantly associated with left-behind children's lower victimization incidence, and furthermore, combined protection service (both school-based and community-based protection services), which has significantly negative association with victimization incidence as well, achieved a more pronounced effect than school-based protection service only. Protection service innovations launched by Chinese government were proved to be effective in protecting left-behind children from victimization. The government shall lay more emphasis on innovation in protection service, as well as speed up the implementation of protection service policy nationwide.

Background Human Rhinoviruses (HRVs) are a common respiratory virus causing acute exacerbations of asthma, chronic obstructive pulmonary disease (COPD) and other related diseases. The sequence of 5’ untranslated region (UTR) and VP4/VP2 coding region of the genome are usually analyzed to classify HRV as HRV-A, HRV-B and HRV-C. Given the interest for new viral strains, we identified the phylogenetic characteristics and molecular epidemiological analysis of human rhinovirus in Shanghai from 2012 to 2020. Methods 6711 throat swabs and sputum samples from patients with acute respiratory infection (ARI) were collected, then the real-time reverse-transcriptase polymerase chain reaction (RT-PCR) method was used to detect HRV. And the fragment of 5 ' untranslated region (5'UTR) and the VP4/VP2 region were amplified for sequencing and gene typing. Results In total, 480/6711(7.2%) HRV-positive samples were detected from 2012–2020, in which 160 samples were further genotyped to HRV-A (109/160, 68.13%), HRV-B (32/160, 20.00%), HRV-C (19/160, 11.88%). We found 13 samples with unclassified results using 5'UTR. Then, these unclassified sample were genotyped to HRV-A and HRV-C by sequencing the VP4/VP2 region. The high incidence of HRV infection occurs in the fall and it is more prevalent in children than in adults. Conclusion Our study suggests that it is very significant to continuously detect and analyze the genotype distribution and epidemic trend of HRV, which can provide data support for disease prevention and treatment. And the 5' UTR and VP4/VP2 should be combined for HRV genotyping accurately.

Human rhinoviruses (HRVs) cause acute upper and lower respiratory tract infections and aggravation of asthma and chronic obstructive pulmonary disease. The 5' untranslated region (5' UTR) and the VP4/VP2 region are widely used for genotyping of HRVs. Members of the species Rhinovirus A and Rhinovirus C have been reported to be more frequently associated with severe disease than members of the species Rhinovirus B. We report the clinical and molecular epidemiological characteristics of HRVs circulating from 2012 to 2020 in Shanghai. A total of 5832 nasopharyngeal swabs from patients with acute respiratory infections were collected. A real-time reverse transcription polymerase chain reaction assay was used for virus detection. The 5' untranslated region and VP4/VP2 region were amplified and sequenced for genotyping and phylogenetic analysis. The overall rate of rhinovirus detection was 2.74% (160/5832), with members of species A, B, and C accounting for 68.13% (109/160), 20.00% (32/160), and 11.88% (19/160) of the total, respectively. Objective To investigate the long-term oncological outcomes of laparoscopic radical hysterectomy (LRH) and abdominal radical hysterectomy (ARH) for the treatment of stage IB2/IIA2 cervical cancer without preoperative adjuvant therapy. Methods We conducted a multicenter, retrospective, case-matching study. The differences in overall survival (OS) and disease-free survival (DFS) between the LRH and ARH were compared under the conditions of real-world study and case-control matching (1:1 matching). Results There was no significant difference in the outcomes of LRH (n = 580) and ARH (n = 1653) in 5-year OS and DFS (OS: 80.6% vs. 86.1%, p = 0.421; DFS: 78.6% vs. 80.7%, p = 0.376). After 1:1 matching, there was no difference in 5-year OS and DFS between LRH (n = 554) and ARH (n = 554) (OS: 80.4% vs. 84.5%, p = 0.993; DFS: 79.0% vs. 78.8%, p = 0.695). Before and after matching, the surgical approach was not an independent risk factor for 5-year OS and DFS, and postoperative adjuvant therapy affected patient prognosis. Further subgroup analysis suggested that there was no difference in LRH (n = 313) and ARH (n = 1092) in 5-year OS or DFS in patients who underwent standard postoperative adjuvant therapy (OS: 83.0% vs. 87.7%, p = 0.992; DFS: 79.0% vs. 82.5%, p = 0.323). After 1:1 pairing, the 5-year OS and DFS in LRH (n = 295) and ARH (n = 295) showed no difference. Before and after matching, the surgical approach was not an independent risk factor affecting the 5-year OS and DFS. Conclusions There was no difference in the oncological outcomes between laparoscopic and abdominal surgery in patients with stage IB2/IIA2 cervical cancer without preoperative adjuvant therapy. The network intervention has caused some unavoidable problems to the networked control system, such as network induction delay, packet loss, communication protocol selection, drive mode, network scheduling, etc., which will have an impact on the performance of the control system. Based on the Truetime simulation toolbox, this paper constructs a networked control system simulation model by using PD controller, with DC servo motor as the controlled object, in order to investigate the effect of time delay on the performance of network control system and the the effects of several other factors on the networked control system performance under certain time delay condition.

Genomic imprinting is an epigenetic phenomenon that causes biased expression of maternally and paternally inherited alleles. In flowering plants, genomic imprinting predominantly occurs in the triploid endosperm and plays a vital role in seed development. In this study, we identified 248 candidate imprinted genes including 114 maternally expressed imprinted genes (MEGs) and 134 paternally expressed imprinted genes (PEGs) in flax (Linum usitatissimum L.) endosperm using deep RNA sequencing. These imprinted genes were neither clustered in specific chromosomal regions nor well conserved among flax and other plant species. MEGs tended to be expressed specifically in the endosperm, whereas the expression of PEGs was not tissue-specific. Imprinted single nucleotide polymorphisms differentiated 200 flax cultivars into the oil flax, oil-fiber dual purpose flax and fiber flax subgroups, suggesting that genomic imprinting contributed to intraspecific variation in flax. The nucleotide diversity of imprinted genes in the oil flax subgroup was significantly higher than that in the fiber flax subgroup, indicating that some imprinted genes underwent positive selection during flax domestication from oil flax to fiber flax. Moreover, imprinted genes that underwent positive selection were related to flax functions. Thirteen imprinted genes related to flax seed size and weight were identified using a candidate gene-based association study. Therefore, our study provides information for further exploration of the function and genomic variation of imprinted genes in the flax population.

Genomic imprinting is an epigenetic phenomenon caused by the biased expression of maternally and paternally inherited alleles. In flowering plants, genomic imprinting predominantly occurs in triploid endosperm and plays a vital role in seed development. In this study, we identified 241 candidate imprinted genes including 143 maternally expressed imprinted genes (MEGs) and 98 paternally expressed imprinted genes (PEGs) in flax (Linum usitatissimumL.) endosperm using deep RNA sequencing. The conservation of imprinting in plants is very limited and imprinting clustering is not a general feature. MEGs tends to be endosperm expression specific, while PEGs are non-tissue specific. The aim of this study is to explore whether the single nucleotide polymorphism rs2275294 in the ZNF512B gene is related to the length of survival of patients with amyotrophic lateral sclerosis (ALS). This prospective study examined 212 patients with ALS, who were genotyped at the rs2275294 locus in ZNF512B using the ligase method. Genotype was compared with clinical data and survival. Kaplan–Meier survival analysis and Cox hazard regression were used to identify risk factors of shorter survival. Our results were meta-analyzed together with previous work in order to examine the potential association between the rs2275294-C allele and survival. Of the 212 patients, 166 carried the CC + CT genotype at the rs2275294 locus, while 46 carried the TT genotype. Patients with the C allele showed significantly shorter survival than those without it (34.13 ± 1.9 vs. 45.32 ± 5.7 months, p = 0.036). Cox analysis identified the C allele and time from symptom onset to diagnosis as risk factors for shorter survival. Meta-analysis of 447 patients in China and Japan confirmed the rs2275294-C allele to be an independent risk factor of shorter survival in ALS patients. The C allele at the rs2275294 locus in ZNF512B is a risk factor for shorter survival in patients with ALS.

To compare oncological outcomes of laparoscopic radical hysterectomy (LRH) and abdominal radical hysterectomy (ARH) for low-risk cervical cancer.We retrospectively compared the 3-year overall survival (OS) and 3-year disease-free survival (DFS) of 1269 low-risk cervical cancer patients with FIGO 2009 stage IA2, IB1 and IIA1 with a tumour size 2 cm, no lymphovascular space invasion (LVSI), superficial stromal invasion and no lymph node involvement on imaging, and who received LRH (n = 672) and ARH (n = 597) between 2009 and 2018 at 47 hospitals.In the total study population, LRH and ARH showed similar 3-year OS (98.6% vs. 98.9%, P = 0.850) and DFS rates (95.7% vs. 96.4%, P = 0.285). LRH was not associated with worse 3-year OS (HR 0.897, 95% CI 0.287-2.808, P = 0.852) or DFS (HR 0.692, 95% CI 0.379-1.263, P = 0.230) as determined by multivariable analysis. After propensity score matching in 1269 patients, LRH (n = 551) and ARH (n = 551) still showed similar 3-year OS (98.4% vs. 98.8%, P = 0.704) and DFS rates (95.5% vs. 96.3%, P = 0.249). LRH was still not associated with worse 3-year OS (HR 0.816, 95% CI 0.262-2.541, P = 0.725) or DFS (HR 0.694, 95% CI 0.371-1.296, P = 0.251).Among patients with low-risk cervical cancers 2 cm, no LVSI, superficial stromal invasion, and no lymph node involvement on imaging, no significant differences were observed in 3-year OS or DFS rates between LRH and ARH.

Objectives Monoacylglycerol lipase participates in organ protection by regulating the hydrolysis of the endocannabinoid 2-arachidonoylglycerol. This study investigated whether blocking monoacylglycerol lipase protects against postresuscitation myocardial injury and improves survival in a rat model of cardiac arrest and cardiopulmonary resuscitation. Design Prospective randomized laboratory study. Setting University research laboratory. Subjects Male Sprague-Dawley rat (n = 96). Interventions Rats underwent 8-minute asphyxia-based cardiac arrest and resuscitation. Surviving rats were randomly divided into cardiopulmonary resuscitation + URB602 group, cardiopulmonary resuscitation group, and sham group. One minute after successful resuscitation, rats in the cardiopulmonary resuscitation + URB602 group received a single dose of URB602 (5 mg/kg), a small-molecule monoacylglycerol lipase inhibitor, whereas rats in the cardiopulmonary resuscitation group received an equivalent volume of vehicle solution. The sham rats underwent all of the procedures performed on rats in the cardiopulmonary resuscitation and cardiopulmonary resuscitation + URB602 groups minus cardiac arrest and asphyxia. Measurements and main results Survival was recorded 168 hours after the return of spontaneous circulation (n = 22 in each group). Compared with vehicle treatment (31.8%), URB602 treatment markedly improved survival (63.6%) 168 hours after cardiopulmonary resuscitation. Next, we used additional surviving rats to evaluate myocardial and mitochondrial injury 6 hours after return of spontaneous circulation, and we found that URB602 significantly reduced myocardial injury and prevented myocardial mitochondrial damage. In addition, URB602 attenuated the dysregulation of endocannabinoid and eicosanoid metabolism 6 hours after return of spontaneous circulation and prevented the acceleration of mitochondrial permeability transition 15 minutes after return of spontaneous circulation. Conclusions Monoacylglycerol lipase blockade may reduce myocardial and mitochondrial injury and significantly improve the resuscitation effect after cardiac arrest and cardiopulmonary resuscitation.

Strong specificity for cancer cells is still the main challenge to deliver drugs for the therapy of cancer. Herein, we developed a convenient strategy to prepare a series of 5-boronopicolinic acid (BA) modified tumor-targeting drug delivery systems (T-DDSs) with strong tumor targeting function. An anti-tumor drug of camptothecin (CPT) was encapsulated into poly(lactide-co-glycolide)-g-polyethylenimine (PLGA-PEI) to form drug-loaded nanoparticles (NP/CPT). Then, the surface of NP/CPT was coated by BA with different polymer and BA molar ratios of 1:1, 1:5, 1:10 and 1:20 via electrostatic interaction to obtain T-DDSs with enhanced biocompatibility and specificity for tumor cells. The introduced BA can endow drug-loaded NPs with high targeting ability to tumor cells because of the overexpression of sialic acids (SA) in tumor cells, which possessed strong interaction with BA. Those T-DDSs exhibited good biocompatibility according to the results of MTT assay, hemolysis test and cellular uptake. Moreover, they were capable of decreasing the viability of breast cancer cell line 4T1 and MCF-7 cells with no obvious cytotoxicity for endothelial cells. Especially, T-DDS with 1:20 molar ratio displayed much higher cellular uptake than other groups, and also exhibited highly efficient in vivo anti-tumor effect. The significantly high targeting function and biocompatibility of T-DDSs improved their drug delivery efficiency and achieved good anti-tumor effect. The BA decorated T-DDSs provides a simple and robust strategy for the design and preparation of DDSs with good biocompatibility and strong tumor-specificity to promote drug delivery efficiency.

Postoperative restenosis remains the key problem of coronary arterial disease and seriously threatens human health. Currently, no effective strategies can thoroughly prevent restenosis owing to its complicated progression. We hypothesize the multi-effect drug of endothelium-protective epigenetic inhibitor (JQ1) may act well in anti-restenosis via exerting multiple functions and acting on different cells. Furthermore, homing to endothelium injured site and pH-triggered drug release can enhance targeted therapeutic efficacy by JQ1 preferential accumulation at injured site and in vascular smooth muscle cells (VSMCs). To verify our hypothesis and develop targeted drug delivery system (DDS) with controlled release ability for anti-restenosis, a pH-responsive nanoparticle was prepared to encapsulate JQ1 and further camouflaged by platelet membrane (PM). The resulting biomimetic and responsive DDS showed optimized drug release behavior and rapid homing to injured site. Moreover, the internalization of DDS in VSMCs was much higher than other groups without PM, which indicated that it inherited natural specificity for VSMCs from PM and obtained dual-targeting function to enhance therapeutic efficacy. The DDS presented significant anti-inflammatory effect via inhibiting NF-κB signaling pathway and secretion of pro-inflammatory cytokines from macrophages. Besides, VSMCs proliferation was suppressed by the DDS with little effect on vascular endothelial cell proliferation and in vivo re-endothelialization at suitable JQ1 concentration. More importantly, the DDS substantially inhibited intima hyperplasia without obvious pathological variation on main organs using a mouse model of carotid artery injury. In summary, we created a versatile DDS for the promotion of JQ1 targeting delivery and responsive release at injured site to mitigate restenosis. The DDS demonstrated outstanding performance via a synergistic and comprehensive effect of multiple factors for anti-restenosis, providing a promising method for clinical applications.
