Abnormal lipid metabolism, such as systemic increased free fatty acid, results in overproduction of pro-inflammatory enzymes and cytokines, which is crucial in the development of obesity-related osteoarthritis (OA). However, there are only a few drugs that target the lipotoxicity of OA. Recent researches have documented that the traditional Chinese medicine, Sparstolonin B (Ssn B), exerted anti-inflammatory effects in various diseases, but not yet in OA. On the basis of this evidence, our works purposed to evaluate the effect of Ssn B on free fatty acid (FFA) palmitate (PA)-stimulated human osteoarthritic chondrocytes and obesity-associated mouse OA model. We found that Ssn B suppressed PA-triggered inflammatory response and extracellular matrix catabolism in a concentration-dependent approach. In vivo, Ssn B treatment inhibited cartilage degeneration and subchondral bone calcification caused by joint mechanical imbalance and alleviated metabolic inflammation in obesity. Mechanistically, co-immunoprecipitine and molecular docking analysis showed that the formation of toll-like receptor 4 (TLR4)/myeloid differentiation protein-2 (MD-2) complex caused by PA was blocked by Ssn B. Subsequently, it leads to inactivation of PA-caused myeloid differentiation factor 88 (MyD88)-dependent nuclear factor-kappaB (NF-κB) cascade. Together, these findings demonstrated that Ssn B is a potential treatment agent for joint degenerative diseases in obese individuals.

Although expressive writing has proved to be beneficial on physical, mental, and social health of individuals, it has been restrained to lab-based experimental studies. In the real world, individuals may naturally engage with expressive writing when dealing with difficult times, especially when facing a tough health journey. Health blogging may serve as an easy-to-access method for self-therapy, if spontaneous expressive writing occurs. However, many posts may not be expressive enough to provide the therapeutic power. In this study, we build a Gaussian naive Bayes model to detect expressive writing in an online health community, CaringBridge. Because we lack full text data as training data, we use a method to learn model parameters from meta-analysis of the literature. The classifier reaches reasonable accuracy on the test set annotated by the authors. We also explore factors that may influence users' spontaneous expressive writing. We find gender, health condition, author type, and privacy settings can affect individuals' spontaneous expressive writing. Finally, we reflect on our methodology and results and provide design implications for online health communities.

Osteoarthritis (OA) is a frequently observed condition in aged people. OA cartilage is characterized by chondrocyte apoptosis, chondrocyte inflammation, and hyperactive catabolism of extracellular matrix. However, the specific molecular mechanisms remain unclear. Recent data has shown that Angptl4, a multifunctional cytokine, is involved in the regulation of inflammatory and apoptosis responses in different tissues. This study is aimed at defining the role of Angptl4 in the development of OA. We employed X-ray analysis, safranin O-fast green (S-O) staining, and hematoxylin staining to evaluate histomorphological characteristics in the knee joint of mice. Real-time quantitative polymerase chain reaction, Western blot assays, immunofluorescence staining, and enzyme-linked immunosorbent assays (ELISA) were performed to analyze the changes in gene and protein expression. Mechanically, our data demonstrated that Angptl4 knockdown improved the degradation of extracellular matrix and reduced TNF-α-mediated chondrocyte inflammation and apoptosis by suppressing sirtuin 1/NF-κB signaling pathway. In addition, animal studies showed that the suppression of Angptl4 expression might alleviate OA development. In conclusion, our findings revealed the underlying mechanisms of Angptl4 regulation in chondrocytes and its potential value in the treatment of OA.

Online Health Communities (OHCs) are known to provide substantial emotional and informational support to patients and family caregivers facing life-threatening diagnoses like cancer and other illnesses, injuries, or chronic conditions. Yet little work explores how OHCs facilitate other vital forms of social support, especially instrumental support. We partner with CaringBridge.org---a prominent OHC for journaling about health crises---to complete a two-phase study focused on instrumental support. Phase one involves a content analysis of 641 CaringBridge updates. Phase two is a survey of 991 CaringBridge users. Results show that patients and family caregivers diverge from their support networks in their preferences for specific instrumental support types. Furthermore, ``prayer support'' emerged as the most prominent support category across both phases. We discuss design implications to accommodate divergent preferences and to expand the instrumental support network. Online platforms provide new channels for people in need to seek help from friends and strangers. However, individuals often encounter psychological barriers that deter them from asking for help. For example, people might have different concerns about asking for help, including acknowledging incompetence, bothering others, and accruing social debt. These perceived social costs limit the potential benefits of help solicitations. In this study, we attempt to investigate whether anonymity (posting a question anonymously), ephemerality (allowing questions to be visible for only a short period), and system routing (having the system handle the question routing) could reduce social costs in a typical online help-seeking behavior-question asking. We built a platform to support these three features and conducted a controlled within-subjects experiment to test their effects on the social costs of posting questions. Results suggest that the presence of anonymity, ephemerality, and system routing reduce social costs. Further, we find that employing anonymity and system routing features did not lower the quality and quantity of answers to the questions in our system.

MicroRNAs (miRNAs) perform a variety of important cellular functions, including regulating the cell cycle, apoptosis and differentiation, amongst others. Recent research has demonstrated an essential function performed by miRNAs in regulating pyroptosis, which is a type of programmed cell death associated with inflammatory responses that plays a critical role in numerous diseases. Through direct or indirect action on proteins associated with the pyroptosis signaling pathway, miRNAs are involved in the pathological processes of cardiovascular, kidney and immune diseases, among others. The present review discusses the maturation process of miRNAs and the process of pyroptosis, with a specific focus on the transport of miRNAs to damaged cells via exosomes, shedding vesicles and protein stabilized complexes, as well as the role of different miRNAs in the regulation of pyroptosis through different gene and protein targets. The aim of the present review was to provide a novel insight into the regulatory role of miRNAs in pyroptosis and new treatment options for pyroptosis‑associated diseases.

Intervertebral disc degeneration (IVDD) is one of the major causes of low back pain, but effective therapies are still lacking because of its complicated pathology. It has been demonstrated that increased levels of interleukin-1β (IL-1β) may promote the development of IVDD. Cardamonin (CAR) is a chalcone extracted from Alpinia katsumadai and other plants. It exhibits an anti-inflammatory effect in multiple diseases. In the present study, we investigated the protective effects of CAR on rat nucleus pulposus (NP) cells under IL-1β stimulation in vitro and in a puncture-induced rat IVDD model in vivo. We explored the CAR treatment's inhibition of the expression of inflammatory factors such as cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), prostaglandin E2 (PGE2), nitric oxide (NO), tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6) in rat NP cells. Moreover, the up-regulation of matrix metalloproteinase-13 (MMP-13) and thrombospondin motifs 5 (ADAMTS-5) and the degradation of aggrecan and collagen II induced by IL-1β were reversed by CAR. Mechanistically, we demonstrated that CAR inhibited nuclear factor kappa B (NF-κB) signaling by activating the nuclear factor erythroid-derived 2-like 2 (Nrf2) in IL-1β-induced rat NP cells. Furthermore, the protective effect of CAR was shown in the IVDD model through persistent intragastric administration. Taken together, our results revealed that CAR could activate the Nrf2/HO-1 signaling axis and be a novel agent for IVDD therapy.

Various social media sites and online communities provide new channels for people in need to ask questions and seek help. However, individuals may still encounter psychological barriers that deter solicitation for assistance, which is more formally described as"social costs''. For example, it can be %concerns people have when seeking help can be the concerns of burdening others, the obligation of reciprocation, etc. To understand what could reduce social costs, we conducted a study in the context of Question-Answering (QA) and investigated the following three factors inspired by literature: anonymity (posting a question anonymously), recommendation (having the system handle the question routing), and ephemerality (allowing questions to be visible only for a short period). We built a QA platform to support these three features and conducted a randomized within-subject experiment to test their effects on social costs of posting questions. Results suggest the presence of anonymity, recommendation, and ephemerality reduces the social costs which provides design implications for future community building.

Affective computing, a field that bridges the gap between human affect and computational technology, has witnessed remarkable technical advancement. However, theoretical underpinnings of affective computing are rarely discussed and reviewed. This paper provides a thorough conceptual analysis of the literature to understand theoretical questions essential to affective computing and current answers. Inspired by emotion theories, we proposed the function-component-representation (FCR) framework to organize different conceptions of affect along three dimensions that each address an important question: function of affect (why compute affect), component of affect (how to compute affect), and representation of affect (what affect to compute). We coded each paper by its underlying conception of affect and found preferences towards affect detection, behavioral component, and categorical representation. We also observed coupling of certain conceptions. For example, papers using the behavioral component tend to adopt discrete representation, whereas papers using the physiological component tend to adopt dimensional representation. The FCR framework is not only the first attempt to organize different theoretical perspectives in a systematic and quantitative way, but also a blueprint to help conceptualize an AC project and pinpoint new possibilities. Future work may explore how the identified frequencies of FCR framework combinations may be applied in practice.

Online health communities rely on information about their users to provide services to members. We partner with the online health community CaringBridge.org to infer the health condition that users are discussing from their early writing on the site. We utilize the self-reported health condition data that is provided by users to train machine learning classifiers to predict the health condition of non-reporting users. An analysis of the classifier's errors reveals that users frequently discuss multiple health conditions. We present models with explainable features, enabling us to extract words for the enrichment of consumer health vocabularies and to support future designs connecting patients.

As the third most popular social network among millennials, Snapchat is well known for its picture and video messaging system that deletes content after it is viewed. However, the Stories feature of Snapchat offers a different perspective of ephemeral content sharing, with pictures and videos that are available for friends to watch an unlimited number of times for 24 hours. We conduct-ed an in-depth qualitative investigation by interviewing 18 participants and reviewing 14 days of their Stories posts. We identify five themes focused on how participants perceive and use the Stories feature, and apply a Goffmanesque metaphor to our analysis. We relate the Stories medium to other research on self-presentation and identity curation in social media.

This study aims at measuring last-night sleep quality from electroencephalography (EEG). We design a sleep experiment to collect waking EEG signals from eight subjects under three different sleep conditions: 8 hours sleep, 6 hours sleep, and 4 hours sleep. We utilize three machine learning approaches, k-Nearest Neighbor (kNN), support vector machine (SVM), and discriminative graph regularized extreme learning machine (GELM), to classify extracted EEG features of power spectral density (PSD). The accuracies of these three classifiers without feature selection are 36.68%, 48.28%, 62.16%, respectively. By using minimal-redundancy-maximal-relevance (MRMR) algorithm and the brain topography, the classification accuracy of GELM with 9 features is improved largely and increased to 83.57% in average. To investigate critical frequency bands for measuring sleep quality, we examine the features of each band and observe their energy changing. The experimental results indicate that Gamma band is more relevant to measuring sleep quality.

Projector-camera (pro-cam) systems create virtual environments for remote interaction with audio and video support. However, previous pro-cam systems require custom hardware and thus have not been used at-scale in the field. In this work, we present a pro-cam application to reinforce social relationships for an off-the-shelf pro-cam system. The ShareTable application users can use video chat in conjunction with sharing tabletop video, allowing them to connect while playing and interacting together.
