Parkinson’s disease (PD) is the second most prevalent neurodegenerative disease worldwide and represents a challenge for clinicians. The present study aims to investigate the effects of cerebrolysin and/or lithium on the behavioral, neurochemical and histopathological alterations induced by reserpine as a model of PD. The rats were divided into control and reserpine-induced PD model groups. The model animals were further divided into four subgroups: rat PD model, rat PD model treated with cerebrolysin, rat PD model treated with lithium and rat PD model treated with a combination of cerebrolysin and lithium. Treatment with cerebrolysin and/or lithium ameliorated most of the alterations in oxidative stress parameters, acetylcholinesterase and monoamines in the striatum and midbrain of reserpine-induced PD model. It also ameliorated the changes in nuclear factor-kappa and improved the histopathological picture induced by reserpine. It could be suggested that cerebrolysin and/or lithium showed promising therapeutic potential against the variations induced in the reserpine model of PD. However, the ameliorating effects of lithium on the neurochemical, histopathological and behavioral alterations induced by reserpine were more prominent than those of cerebrolysin alone or combined with lithium. It can be concluded that the antioxidant and anti-inflammatory effects of both drugs played a significant role in their therapeutic potency.

Annona squamosa is a medicinal plant that has been used in folk medicine since antiquity. The goal of this study is to see how effective Annona squamosa leaf extract (A.S.L.E) or its niosomal-entrapped preparation is at protecting skin from UVA irradiation. The prepared niosomal-entrapped A.S.L.E has been characterized via spectrophotometry and transmission electron microscopy imaging. Furthermore, the entrapment efficiency and in vitro release of A.S.L.E were determined. In this study, ex vivo and freshly prepared samples from the dorsal region of the rats’ skin were used as biological samples, which were divided into five groups: control UVA-unexposed, unprotected UVA-exposed, A.S.L.E-protected UVA-exposed, and niosomal-entrapped A.S.L.E UVA-exposed. UVA irradiation was performed by exposing the skin samples to a UVA-producing lamp for 4 h. Samples from various groups were then examined using FTIR spectroscopy, histopathology, and protein electrophoresis methods. The results showed that A.S.L.E has a skin protective effect against UVA irradiation. The niosomal-entrapped A.S.L.E was more effective than the native plant leaf extract in protecting skin from the damaging effects of UVA. Therefore, the nanotechnologically formulated preparation, niosomal-entrapped A.S.L.E, can be used as an effective photoprotector (sunscreen) against the adverse effects of UVA radiation. Olive oils are more expensive compared with other vegetable oils. Therefore, adulterating such expensive oil is prevalent. The traditional methods for olive oil adulteration detection are complex and require pre-analysis sample preparation. Therefore, simple and precise alternative techniques are required. In the present study, the Laser-induced fluorescence (LIF) technique was implemented for detecting alteration and adulteration of olive oil mixed with sunflower or corn oil based on the post-heating emission characteristics. Diode-pumped solid-state laser (DPSS, λ = 405 nm) was employed for excitation and the fluorescence emission was detected via an optical fiber connected to a compact spectrometer. The obtained results revealed alterations in the recorded chlorophyll peak intensity due to olive oil heating and adulteration. The correlation of the experimental measurements was evaluated via partial least-squares regression (PLSR) with an R-squared value of 0.95. Moreover, the system performance was evaluated using receiver operating characteristics (ROC) with a maximum sensitivity of 93%.

The present study aimed at investigating the antidepressant and antioxidant actions of near-infrared (NIR) laser at a wavelength of 830 nm and power of 100 mW which applied transcranially on an animal model of depression induced by repeated doses of reserpine (0.2 mg/kg). Thirty male Wistar adult rats were divided into three groups: rat model of depression; rat model of depression irradiated with laser for 14 days after induction of depression; and the control group that was given the drug vehicle and sham-exposed to the laser. Forced swimming test (FST) was used to verify the induction of animal model of depression and to screen the effect of antidepressant effect of low-level laser at the end of the experiment. Monoamine level, oxidative stress markers, and activities of acetylcholinesterase (AchE) and monoamine oxidase (MAO) were determined in the cortex and hippocampus of the rat brain. Reserpine resulted in depletion of monoamines and elevation in the oxidative stress markers and change in the enzymatic activities measured in both brain areas. Laser irradiation has an inhibitory action on the monoamine oxidase (MAO) in the cortex and hippocampus leading to elevation of the monoamine levels and attenuation of the oxidative stress in the studied areas. FST has emphasized the antidepressant effect of the utilized laser irradiation parameters on the behavioral level. The present findings provide evidence for the antidepressant and antioxidant actions of NIR low-power laser in the rat model of depression. Accordingly, low-laser irradiation may be presented as a potential candidate modality for depression treatment.

Introduction: Caffeine and nicotine are the most widely consumed psychostimulants worldwide. Although the effects of each drug alone on the central nervous system have been studied extensively, the literature on the neurochemical and electrophysiological effects of their combined treatments is scarce. The present study investigated the cortical electrophysiological and neurochemical alterations induced by acute administration of caffeine and nicotine in rats. Methods: The rats received caffeine and nicotine at a 1-hour interval between the two treatments. Results: Caffeine and nicotine administration resulted in a significant decrease in the concentrations of cortical amino acid neurotransmitters, namely glutamate, aspartate, glycine, and taurine, while γ-aminobutyric acid (GABA) significantly increased. Increased cortical lipid peroxidation and reduced glutathione and nitric oxide levels and acetylcholinesterase and Na⁺/K⁺-ATPase activities were also observed. The Electroencephalogram (EEG) showed an increase in delta frequency power band, whereas theta, beta-1, and beta-2 decreased after caffeine and nicotine treatment. Conclusion: These findings suggest that caffeine and nicotine adversely exacerbate their stimulant effects manifested by the EEG changes mediated by increasing cholinergic transmission and disturbing the balance between the excitatory and inhibitory amino acids leading to oxidative stress.

Seizures, the main symptom of epilepsy, are provoked due to a neurological disorder that underlies the disease. The accurate detection of seizures is a crucial step in any procedure of treatment. In the present study, electrocorticogram (ECoG) signals were recorded from awake and freely moving animals implanted with cortical electrodes before and after pentylenetetrazol, the chemo-convulsant injection. ECoG signals were segmented into 4-s epochs and labeled. Twenty-four linear and non-linear features were extracted from the time and frequency domains of the ECoG signals. The extracted features either individually or in combinations were fed to an automatic support vector machine (SVM) classification system. SVM classifier was trained with 5 min of ictal and non-ictal labeled ECoG signals to build the hyperplane that separates two sets of training signals. Sensitivity, specificity, and accuracy were determined for the testing dataset using the different feature combinations. It has been found that some linear features either individually or in combinations outperform non-linear features in terms of the accuracy for seizure detection. The maximum accuracy achieved by the system was 95.3% and has been obtained only after linear and non-linear features were combined. ECoG signals were classified without pre-processing or removal of artifacts to reduce the required computational time to be suitable for online implementation purposes. This may prove the detection system’s robustness and supports its use in online seizure detection protocols.

Cognitive impairment is one of the serious side effects that cancer-treated patients suffer from after treatment by doxorubicin (DOX). Investigating the mechanisms underlying this impairment is crucial for its treatment or prevention. The current study investigates the cortical and hippocampal neurochemical changes induced by an acute dose of DOX (20 mg/kg, i.p.) and evaluates the neuroprotective effect of nanocurcumin (NC) (50 mg/kg, p.o.) against these changes. Animals were randomly divided into four groups, control, rats treated with either NC or DOX, and the fourth group treated with NC prior to DOX. Cortical dopamine level has significantly increased (71.88 %) after DOX injection. This was associated with a significant rise in the levels of lipid peroxidation (183.99 %, 201.4 %) and nitric oxide (36.54 %, 55 %) and a significant reduction in reduced glutathione (13 %, 21.44 %) in the cortex and hippocampus, respectively. In addition, DOX inhibited the cortical and hippocampal activities of acetylcholinesterase (94.82 %, 62.75 %) and monoamine oxidase (64.40 %, 68.84 %), respectively. Protection with NC mitigates the changes induced in the oxidative stress parameters by DOX. However, the effect on the activities of AchE and MAO was insignificant. This was reflected in the level of dopamine that showed non-significant changes in comparison to control and DOX-treated rats. The present findings indicate that oxidative stress, inhibition in AchE, MAO, and the subsequent elevation in dopamine could have a crucial role in mediating the chemo-brain adverse effects induced by DOX. In addition, protection with NC mitigated some of these adverse effects thus rendering DOX more tolerable.

Inflammatory pathway and disruption in glutamate homeostasis join at the level of the glia, resulting in various neurological disorders. In vitro studies have provided evidence that membrane proteins connexions (Cxs) are involved in glutamate release, meanwhile, excitatory amino-acid transporters (EAATs) are crucial for glutamate reuptake (clearance). Moreover, pannexin-1 (Panx-1) activation is more detrimental to neurons. Their expression patterns during inflammation and the impacts of IκB kinase (IKK) inhibition, morphine, and galantamine on the inflammatory-associated glutamate imbalance remain elusive. To investigate this, rats were injected with saline or lipopolysaccharide. Thereafter, vehicles, morphine, galantamine, and BAY-117082 were administered in different groups of animals. Subsequently, electroencephalography, enzyme-linked immunosorbent assay, western blot, and histopathological examinations were carried out and various indicators of inflammation and glutamate level were determined. Parallel analysis of Cxs, Panx-1, and EAAts in the brain was performed. Our findings strengthen the concept that unregulated expressions of Cxs, Panx-1, and EAATs contribute to glutamate accumulation and neuronal cell loss. Nuclear factor-kB (NF-κB) pathway can significantly contribute to glutamate homeostasis via modulating Cxs, Panx-1, and EAATs expressions. BAY-117082, via inhibition of IkK, promoted the anti-inflammatory effects of morphine as well as galantamine. We concluded that NF-κB is an important component of reshaping the expressions of Cxs, panx-1, and EAATs and the development of glutamate-induced neuronal degeneration.

Studying light penetration in biological tissues became a very important concern in various medical applications. It is an essential factor required to resolve the optical dose in many diagnostic and therapeutic procedures. The absorption and scattering properties of the inspected tissue control how deep the light will travel inside the tissue. However, these optical properties are highly dependent on the wavelength of the light source. In this work, the light transmission through different regions of the rat’s head was investigated and the minimum laser power required to reach different parts of the head is also determined using 808-nm semiconductor laser diode. The power variation in different regions of the head is estimated using Monte Carlo simulation. Absorption and scattering coefficients of the head layers were calculated using integrating sphere measurements and Kubelka-Munk model. The absorption coefficient of the skin was 0.19 ± 0.071 mm−1, 0.024 ± 0.11 mm−1 for skull, and 0.35 ± 0.13 mm−1 for the brain, while the scattering coefficients were 7.35 ± 1.09, 2.71 ± 0.37, and 13.04 ± 0.36 mm−1 for skin, skull, and brain, respectively. The obtained results provide a relationship between laser incident power and the depth in the rat’s head showing a higher optical transmission at the frontal part of the head than the middle or back regions due to the variations in the skull thickness. Therefore, the study revealed that the transmitted power of 808 nm laser at different incident locations on the head is nonlinear and variable due to different skull’s thickness.

Metal nanoparticles, in general, and silver nanoparticles (AgNPs), in particular, have been the focus of numerous studies over the last two decades. Recently, the green synthesis of metal nanoparticles has been favored over chemical synthesis due to its low toxicity and easy preparation. The present study aims to investigate the dose-dependent effects of green synthesized AgNPs on rats’ brains. Thirty-four Wistar male rats were divided into four groups. The first, second, and third groups were administered for 14 days with three different doses (0.5, 5, 10 mg/Kg) of AgNPs, respectively. The fourth group, which served as a control group, was given normal saline for the same period. The effects of the green synthesized AgNPs on the cortical and hippocampal levels of the oxidative stress markers (MDA, NO, and GSH) as well as the activity of acetylcholinesterase (AchE) and the monoamine neurotransmitters (DA, NE, and 5H-T) were investigated. AgNPs showed minimal oxidative stress in the cortex and hippocampus for the administered doses. However, AgNPs showed an inhibitory effect on AchE activity in a dose-dependent manner and a decrease in the 5H-T and NE levels. The green synthesized AgNPs showed an ultrastructural change in the cellular membranes of the neurons. The green synthesis of AgNPs has reduced their cytotoxic oxidative effects due to their capping with biologically compatible and boosting molecules such as flavonoids. However, other neurotoxic effects were observed in a dose-dependent manner.

Noninvasive brain stimulation/modulation is a rapidly emerging technique that has been implemented in different clinical applications. The commonly noninvasive techniques used in neurological manipulations include photobiomodulation (PBM), transcranial electrical stimulation (TES), transcranial magnetic stimulation (TMS), and ultrasound stimulation (USS). These techniques have the ability to excite, inhibit, or modulate neuronal activity in targeted brain areas to obtain the required therapeutic effects. However, each technique owns its unique mechanism of action that relies on specific parameters suitable for treating certain neurological disorders. Neurological disorders such as epilepsy, Parkinson's disease (PD), Alzheimer's disease (AD), and depression have been treated by one or more of these noninvasive techniques. The therapeutic outcomes of these techniques for neurological diseases are promising, yet with limitations. In the present review, the mechanisms of action of these different brain stimulation/modulation modalities were explored and a synopsis of their applications in the treatment of certain neurological disorders was provided. Moreover, methodological issues, limitations, and open questions were presented. Furthermore, some future directions were suggested.

The antidepressant effect of curcumin-coated iron oxide nanoparticles (Cur-IONPs) was investigated in the current study using depression rat model induced by reserpine. IONPs were synthesized by curcumin as a reducing agent producing Cur-IONPs. Rats were divided into control, depression rat model, and depressed rats treated with Cur-IONPs. After treatment rat behavior was evaluated using forced swimming test (FST). Serotonin (5-HT), norepinephrine (NE), dopamine (DA), monoamine oxidase (MAO), acetylcholinesterase (AchE), Na+, K+, ATPase, lipid peroxidation (MDA), reduced glutathione (GSH), glutathione-s-transferase (GST) and nitric oxide (NO) were measured in the cortex and hippocampus. In depressed rats, FST showed increased immobilization time and reduced swimming time. This was associated with a significant decrease in 5-HT, NE, DA and GSH and a significant increase in MDA and NO levels and GST, MAO, AchE and Na+, K+, ATPase activities in the cortex and hippocampus. Treatment with Cur-NONPs for two weeks increased the swimming time reduced the immobility time, and elevated 5-HT, NE and DA levels. Cur-IONPs attenuated the oxidative stress induced by reserpine and restored the MAO, AchE and Na+, K+, ATPase. The present green method used curcumin in the IONPs synthesis and has several merits; obtaining nanoform of iron oxide, increasing the bioavailability of curcumin and reducing the oxidative stress induced by iron. The present antidepressant effect of Cur-IONPs could be attributed to the ability of Cur-IONPs to restore monoamine neurotransmitter levels by increasing their synthesis and reducing their metabolism. In addition, the antioxidant activity of curcumin prevented oxidative stress in the depressed rats.

Alzheimer's disease (AD) is a progressive neurodegenerative disease that afflicts millions of people all over the world. Intracerebroventricular (ICV) injection of a sub-diabetogenic dose of streptozotocin (STZ) was established as an experimental animal model of AD. The present study was conducted to evaluate the efficacy of curcumin nanoparticles (CNs) against the behavioral, neurochemical and histopathological alterations induced by ICV-STZ. The animals were divided into: control animals, the animal model of AD that received a single bilateral ICV microinjection of STZ, and the animals protected by a daily oral administration of CNs for 6 days before the ICV-STZ injection. The animals of all groups were subjected to surgical operation on the 7th day of administration. Then the administration of distilled water or CNs was continued for 8 days. The ICV-STZ microinjection produced cognitive impairment as evident from the behavioral Morris water maze (MWM) test and induced oxidative stress in the cortex and hippocampus as indicated by the significant increases in lipid peroxidation and nitric oxide (NO) levels and the significant decrease in reduced glutathione (GSH) levels. It also produced a significant increase in acetylcholinesterase (AChE) and tumor necrosis-alpha (TNF-ɑ) and a significant decrease in Na+,K + -ATPase. In addition, a significant increase in amino acid neurotransmitters occurred in the hippocampus, whereas a significant decrease was obtained in the cortex of STZ-induced AD rats. CNs ameliorated the behavioral, immunohistochemical and most of the neurochemical alterations induced by STZ in the hippocampus and cortex. It may be concluded that CNs might be considered as a promising therapeutic agent for the treatment of AD.

Aim The current study aims to investigate the impact of paradoxical (REM) sleep deprivation and/or epileptic seizures on rat's cortical brain tissues. Main methods Animals were divided into four groups; control, epileptic, REM sleep deprived and epileptic subjected to REM sleep deprivation. Electrocorticogram (ECoG) signals were recorded and quantitatively analyzed for each group. Concentrations of amino acid neurotransmitters; proinflammatory cytokines; and oxidative stress parameters; and acetylcholinesterase activity were determined in the cortex of the animals in different groups. Key findings Results showed significant variations in the spectral distribution of ECoG waves in the epilepsy model, 24- and 48-hours of REM sleep deprivation and their combined effects indicating a state of cortical hyperexcitability. Significant increases in NO and taurine and significant decrement in glutamine, GABA and glycine were determined. In REM sleep deprived rats significant elevation in glutamate, aspartate, glycine and taurine and a significant lowering in GABA were obtained. This was accompanied by significant reduction in AchE and IL-β. In the cortical tissue of epileptic rats deprived from REM sleep significant increases in lipid peroxidation, TNF-α, IL-1β, IL-6 and aspartate and a significant reduction in AchE were observed. Significance The present data indicate that REM sleep deprivation induces an increase in lipid peroxidation and storming in proinflammatory cytokines in the cortex of rat model of epilepsy during SRS. These changes are associated with a decreased seizure threshold as inferred from the increase in alpha and Beta waves and a decrease in Delta waves of ECoG.

The use of technology in agriculture has grown in recent years with the era of data analytics affecting every industry. The main challenge in using technology in agriculture is identification of effectiveness of big data analytics algorithms and their application methods. Pest management is one of the most important problems facing farmers. The cotton leafworm,Spodoptera littoralis(Boisd.) (CLW) is one of the major polyphagous key pests attacking plants includes 73 species recorded at Egypt. In the present study, several machine learning algorithms have been implemented to predict plant infestation with CLW. The moth of CLW data was weekly collected for two years in a commercial hydroponic greenhouse. Furthermore, among other features temperature and relative humidity were recorded over the total period of the study. It was proven that the XGBoost algorithm is the most effective algorithm applied in this study. Prediction accuracy of 84 % has been achieved using this algorithm. The impact of environmental features on the prediction accuracy was compared with each other to ensure a complete dataset for future results. In conclusion, the present study provided a framework for applying machine learning in the prediction of plant infestation with the CLW in the greenhouses. Based on this framework, further studies with continuous measurements are warranted to achieve greater accuracy.

The aim of this work was to develop a digital dynamic cardiac phantom able to mimic gated myocardial perfusion single photon emission computed tomography (SPECT) images. A software code package was written to construct a cardiac digital phantom based on mathematical ellipsoidal model utilizing powerful numerical and mathematic libraries of python programing language. An ellipsoidal mathematical model was adopted to create the left ventricle geometrical volume including myocardial boundaries, left ventricular cavity, with incorporation of myocardial wall thickening and motion. Realistic myocardial count density from true patient studies was used to simulate statistical intensity variation during myocardial contraction. A combination of different levels of defect extent and severity were precisely modeled taking into consideration defect size variation during cardiac contraction. Wall thickening was also modeled taking into account the effect of partial volume. It has been successful to build a python-based software code that is able to model gated myocardial perfusion SPECT images with variable left ventricular volumes and ejection fraction. The recent flexibility of python programming enabled us to manipulate the shape and control the functional parameters in addition to creating variable sized-defects, extents and severities in different locations. Furthermore, the phantom code also provides different levels of image filtration mimicking those filters used in image reconstruction and their influence on image quality. Defect extent and severity were found to impact functional parameter estimation in consistence to clinical examinations. A python-based gated myocardial perfusion SPECT phantom has been successfully developed. The phantom proved to be reliable to assess cardiac software analysis tools in terms of perfusion and functional parameters. The software code is under further development and refinement so that more functionalities and features can be added.

The present study designed to investigate the protective effect of curcumin nanoparticles (CUR-NPs) on the cardiotoxicity induced by doxorubicin. Rats were divided into four groups; control, rats treated daily with CUR-NPs (50 mg/kg) for 14 days, rats treated with an acute dose of doxorubicin (20 mg/kg) and rats treated daily with CUR-NPs for 14 days injected with doxorubicin on the 10th day. After electrocardiogram (ECG) recording from rats at different groups, rat decapitation was carried out and the heart of each rat was excised out to measure the oxidative stress parameters; lipid peroxidation (MDA), nitric oxide (NO) and reduced glutathione (GSH) and the activities of Na,K,ATPase and acetylcholinesterase (AchE). In addition, the levels of dopamine (DA), norepinephrine (NE) and serotonin (5-HT) were determined in the cardiac tissues. Lactate dehydrogenase (LDH) activity was measured in the serum. The ECG recordings indicated that daily pretreatment with CUR- NPs has prevented the tachycardia (i.e. increase in heart rate) and ameliorated the changes in ST wave and QRS complex induced by doxorubicin. In addition, CUR-NPs prevented doxorubicin induced significant increase in MDA, NO, DA, AchE and LDH and doxorubicin induced significant decrease in GSH, NE, 5-HT and Na,K,ATPase. According to the present findings, it could be concluded that CUR-NPs have a protective effect against cardiotoxicity induced by doxorubicin. This may shed more light on the importance of CUR-NPs pretreatment before the application of doxorubicin therapy.

The present study aims to evaluate the efficacy of selenium (Se) alone or combined with carbamazepine (CBZ) against the adverse effects induced by the chemoconvulsant pentylenetetrazole (PTZ) in the cortex of adult male rats. Electrocorticogram (ECoG) and oxidative stress markers were implemented to evaluate the differences between treated and untreated animals. Animals were divided into five groups: control group that received i.p. saline injection, PTZ-treated group that received a single i.p. injection of PTZ (60 mg/kg) for induction of seizures followed by a daily i.p. injection of saline, Se-treated group that received an i.p. injection of sodium selenite (0.3 mg/kg/day) after PTZ administration, CBZ-treated group that received orally CBZ (80 mg/kg/day) after PTZ administration, and combination (Se plus CBZ)-treated group that received an oral administration of CBZ (80 mg/kg/day) followed by an i.p. injection of sodium selenite (0.3 mg/kg/day) after PTZ administration. Quantitative analyses of the ECoG indices and the neurochemical parameters revealed that Se and CBZ have mitigated the adverse effects induced by PTZ. The main results were decrease in the number of epileptic spikes, restoring the normal distribution of slow and fast ECoG frequencies and attenuation of most of the oxidative stress markers. However, there was an increase in lipid perioxidation marker in combined treatment of CBZ and Se. The electrophysiological and neurochemical data proved the potential of these techniques in evaluating the treatment’s efficiency and suggest that supplementation of Se with antiepileptic drugs (AEDs) may be beneficial in ameliorating most of the alterations induced in the brain as a result of seizure insults and could be recommended as an adjunct therapy with AEDs.

In this paper, different window sizes for EEG signal segmentation are investigated in order to optimize the performance of seizure detection systems. To differentiate between epileptic and non-epileptic epochs, the time axis of the EEG signal is divided into non-overlapping windows. The window period should be long enough for the lapse to be informative but not too long for it to stay stationary. Hence, the KPSS test is used to determine signal stationarity for different window sizes, then the optimal window is chosen such that it corresponds to the smallest number of non-stationary segments in the signal of interest. The seizure detection system is then applied to the piece-wise stationary segments. Compared to the exhaustive examination, it is found that the KPSS test optimal window results in the highest sensitivity.

The present study investigated the efficacy of cationic liposome-encapsulated carotenoids (lutein or beta-carotene) as a treatment in an animal model of fibromyalgia (FM). Preparation and characterization of the nano-sized cationic liposomal carotenoids have been carried out. FM has been induced in the experimental animals via successive subcutaneous reserpine injection (1 mg/kg). Animals were divided into four groups; control, reserpinized (Res), reserpinized and cationic liposomal lutein-treated (Res + CL-Lut), and reserpinized and liposomal beta-carotene-treated (Res + CL-Bc). Levels of norepinephrine (NE), dopamine (DA), and serotonin (5-HT), and oxidative stress markers (MDA, H2O2, NO, and GSH) were determined in the brain's cortical tissue of the different groups of animals. Furthermore, the spectral analysis of the electrocorticogram (ECoG) was carried out. Animal behavior was tested for different animal groups. Results showed a significant reduction in monoamines, an elevation of oxidative stress markers, a shift in the ECoG frequency band power, and a change in pain threshold of the reserpinized animals. A return to a non-significant difference from the control values of all the measured parameters has been obtained after two weeks of cationic liposomal carotenoid preparations treatment. The present findings shed more light on the validity of the reserpine model of FM and provide evidence for the antidepressant, antioxidant, and anti-nociceptive potential of the cationic liposomal carotenoids. The present results proofed that the natural product preparations on a nano-sized scale could be a good alternative to the pharmacological interventions for FM treatment.

The iron oxide nanoparticles (IONPs) have enormous applications in biomedical fields. The change in properties of IONPs such as morphology, size, surface charge, agglomeration, and magnetization have marked impact on their effectiveness. In this work, evaluation of the physico-chemical properties of different green synthesized IONPs compared with their chemical synthesized one has been carried out. IONPs were fabricated chemically (Chem-IONPs) or through a green route using curcumin (Cur-IONPs), aloe Vera (AV-IONPs), and green tea (GT-IONPs) extracts. All prepared IONPs were characterized using transmission electron microscopy (TEM), zeta potential measurements, UV–Vis spectroscopy, X-ray diffraction (XRD), vibrating sample magnetometer (VSM), and differential scanning calorimetry (DSC) techniques. Additionally, the DPPH scavenging activity of the prepared nanoparticles was tested. The present study showed in a comparative way the effect of different routes of IONPs synthesis on their physicochemical properties. These different properties may have great usage in different biomedical applications.

The aim of the present work was to investigate the neurochemical changes induced in the cerebellum of rat model of Parkinson's disease (PD). Rats were divided into two groups; control and rat model of PD induced by the intrastriatal injection of rotenone. As compared to control, a significant increase in the excitatory amino acid neurotransmitters; glutamate and aspartate together with a significant decrease in the inhibitory amino acids, GABA, glycine and taurine were observed in the cerebellum of rat model of PD. This was associated with a significant increase in lipid peroxidation, nitric oxide and tumor necrosis factor-α and a significant decrease in reduced glutathione. A significant decrease in acetylcholinesterase and a significant increase in Na+,K+-ATPase were recorded in the cerebellum of rat model of PD. In addition the cerebellar sections from rat model of PD showed marked necrosis of Purkinje cells, irregular damaged cells, cytoplasmic shrinkage, necrosis and perineuronal vacuolation. The present results indicate that the disturbance in the balance between the excitatory and inhibitory amino acids may have a role in the pathogenesis of PD. According to the present neurochemical and histopathological changes, the cerebellum should be taken into consideration during the treatment of PD.

Aims Parkinson's disease (PD) is the second most prevalent neurodegenerative disease affecting the population. The present study investigates the potential therapeutic effect of cerebrolysin (CBL), as a neurotrophic factor mimic, on the behavioral and biochemical alterations induced in 6-hydroxydopamine (6-OHDA) – lesioned rats as a model of PD. Main methods The animals were divided into 3 experimental groups; control group, Parkinsonian model group through bilateral microinjection of 6-OHDA into substantia nigra (SN) and CBL-treated group which received a daily intraperitoneal administration of CBL (2.5 ml/kg) initiated 24 h after induction of Parkinsonism for 21 days. Key findings Treatment of Parkinsonian animals with CBL succeeded in restoring the midbrain and striatum dopamine levels. In addition, it normalized the increased MDA and NO levels recorded in the Parkinsonian animals and replenished the decreased level of midbrain GSH. In addition to the recorded recovery of the biochemical parameters, there was a parallel improvement in the animal's behavioral aspects. Significance The findings of the present study provide evidence for the promising therapeutic effect of CBL in the present 6-OHDA rat model of PD through counteracting oxidative stress, replenishing dopamine content and enhancing behavioral outcomes.

The present study aims to evaluate the antidepressant efficacy of curcumin nanoparticles on rat model of depression induced by reserpine. Rats were divided into control, the rat model of depression induced by daily i.p. injection of reserpine (0.2 mg/kg for 21 days), and the rat model of depression treated daily with the formulated CNPs (20 mg/kg for 7 and 15 days). The behavioral evaluation was assessed for all groups of animals by the forced swim test (FST). Monoamine neurotransmitters were measured in the cortex and hippocampus via fluorescence spectroscopy. The electrophysiological evaluation was carried out by recording and analyzing the electrocorticogram (ECoG) in a group of animals that served as self-control. The chronic administration of reserpine resulted in a significant decrease in the duration of the active phase in the FST; a significant decrease in the cortical and hippocampal levels of serotonin, norepinephrine, and dopamine; a significant increase in spectral power of alpha and delta waves; and a significant decrease in spectral power of theta, beta-1, and beta-2 waves with respect to control values. Administration of CNPs for 7 days has improved the performance of animals in FST indicated by the increase in the duration of the active phase. Additionally, the levels of serotonin and dopamine have been restored; however, the level of norepinephrine has been not completely recovered in both cortex and hippocampus. A significant increase in alpha and beta-2 waves, an increase in theta and beta-1, and a decrease in delta waves have been recorded after 7 days of treatment. Extending treatment with CNPs for 15 days has succeeded in restoring the active phase in FST and monoamine level in the cortex and hippocampus to control like values. In addition, ECoG waves have returned to near control like values. It could be concluded that the formulated nanocurcumin has an effective and potent antidepressant activity that was evidenced by electrical, chemical, and behavioral tests.

Status epilepticus (SE) has functional and structural consequences resulting in brain damage. The present study aims to investigate the role of taurine and green tea extract in the neuroprotection against oxidative stress and changes in acetylcholinesterase (AChE) and Na + ,K + -ATPase activities during SE induced by pilocarpine in the hippocampus of adult male rats. Animals received an oral administration of either taurine (100 mg/kg) or green tea extract containing 100 mg/kg epigallocatechin gallate for 3 days before the induction of SE with pilocarpine (380 mg/kg, i.p.) and were sacrificed 1 h after pilocarpine injection. Data indicated that a state of oxidative stress has evolved during SE as evident from the significant increase in lipid peroxidation level and significant decrease in reduced glutathione (GSH) level. Significant decreases in AChE and Na + ,K + -ATPase activities were also recorded. Pretreatment of rats with taurine exaggerated the increase in lipid peroxidation and failed to prevent the decrease in Na + ,K + -ATPase activity resulting from pilocarpine. However, taurine pretreatment prevented the reduced activity of hippocampal AChE induced by pilocarpine during SE. Pretreatment of rats with green tea extract prevented the increase in lipid peroxidation occurring during SE. However, it failed to inhibit the decrease in Na + ,K + -ATPase activity. In conclusion, taurine pretreatment failed to reduce the oxidative stress induced during SE. In contrast, pretreatment of rats with green tea extract ameliorated the oxidative stress induced by pilocarpine and this may assist in reducing the insults of hyperexcitability and excitotoxicity that occur during SE and thereby reduce neuronal damage.

Febrile seizures (FS) are convulsions associated with high body temperature. It has a high incidence in children from the age of 6months to 5years and may have adverse consequences in adulthood. The experimental model of FS could be induced in animals via hyperthermia. The present study was designed to investigate persistent electroencephalographic (EEG), neurochemical and behavioral alterations in adult animals that had experienced complex FS at their immature age. EEG signals were obtained from the cortex of both FS and control normothermic groups of animals. A spectrophotometric assay was carried out to determine oxidative stress parameters (malondialdehyde, nitric oxide, reduced glutathione) and acetylcholinesterase activity in the cortex and hippocampus of FS and control animals. Behavioral assessment of seizure threshold and severity were investigated via a sub-convulsive dose of nicotine in adult animals. Alterations in the oxidant/antioxidant system and AChE activity were obtained in the cortex and hippocampus of FS animals in comparison to control animals. EEG spectral analysis displayed significant changes in all EEG frequency bands. A decrease in seizure latency and an increase in seizure severity were also observed. The present study provides evidence for long-lasting abnormalities in the cortex and hippocampus of adult animals subjected to complex FS at their developmental age, which may be correlated to the underlying mechanism of epileptogenesis and its related co-morbidities.

Transcranial low-level infrared laser is a modality of therapy based on the principle of photons delivered in a non-invasive manner through the skull for the treatment of some neurological conditions such as psychological disorders, traumatic brain injuries, and neurodegenerative diseases among others. In the present study, effects of low-level infrared laser irradiation with different radiation powers (80, 200, and 400 mW, continuous wave) were investigated on normal animals subjected to forced swimming test (FST). Results indicated that there are changes in FST parameters in animals irradiated with laser; the lowest dose provoked a significant increase in animal activity (swimming and climbing) and a significant decrease in animal’s immobility, while the highest laser dose resulted in a complete inverse action by significantly increasing animal immobility and significantly decreasing animal activity with respect to control animals. The lowest dose (80 mW) of transcranial laser irradiation has then utilized on animals injected with a chronic dose of reserpine (0.2 mg/kg i.p. for 14 days) served as an animal model of depression. Laser irradiation has successfully ameliorated depression induced by reserpine as indicated by FST parameters and electrocorticography (ECoG) spectral analysis in irradiated animals. The findings of the present study emphasized the beneficial effects of low-level infrared laser irradiation on normal and healthy animals. Additionally, it indicated the potential antidepressant activity of the low dose of infrared laser irradiation.

Electrical activity recording from the brains of awake animals is a corner stone in the study of the neurophysiological basis of behavior. To meet this need, a microelectrode driver suitable for the animal of interest has to be developed. In the present study a miniature microdrive was developed specifically for the leopard toad, Bufo regularis, however, it can be used for other small animals. The microdrive was designed to meet the following requirements: small size, light weight, simple and easy way of attaching and removing, advancing and withdrawing of microelectrode in the animal brain without rotation, can be reused and made from inexpensive materials. To assess the performance of the developed microdrive, we recorded auditory evoked potentials from different auditory centers in the toad's brain. The potentials were obtained from mesencephalic, diencephalic and telencephalic auditory sensitive areas in response to simple and complex acoustic stimuli. The synthetic acoustical tones introduced to the toad were carrying the dominant frequencies of their mating calls.

Brain cooling has pronounced effects on seizures and epileptic activity. The aim of the present study is to evaluate the anticonvulsant effect of brain cooling on the oxidative stress and changes in Na(+), K(+)-ATPase and acetylcholinesterase (AchE) activities during status epilepticus induced by pilocarpine in the hippocampus of adult male rat in comparison with α-lipoic acid. Rats were divided into four groups: control, rats treated with pilocarpine for induction of status epilepticus, rats treated for 3 consecutive days with α-lipoic acid before pilocarpine and rats subjected to whole body cooling for 30 min before pilocarpine. The present findings indicated that pilocarine-induced status epilepticus was accompanied by a state of oxidative stress as clear from the significant increase in lipid peroxidation (MDA) and superoxide dismutase (SOD) and significant decrease in reduced glutathione and nitric oxide (NO) levels and the activities of catalase, AchE and Na(+), K(+)-ATPase. Pretreatment with α-lipoic acid ameliorated the state of oxidative stress and restored AchE to nearly control activity. However, Na(+), K(+)-ATPase activity showed a significant decrease. Rats exposed to cooling for 30 min before the induction of status epilepticus revealed significant increases in MDA and NO levels and SOD activity. AchE returned to control value while the significant decrease in Na(+), K(+)-ATPase persisted. The present data suggest that cooling may have an anticonvulsant effect which may be mediated by the elevated NO level. However, brain cooling may have drastic unwanted insults such as oxidative stress and the decrease in Na(+), K(+)-ATPase activity.

The present study aims to investigate the effects of paradoxical sleep deprivation (PSD) on the waking EEG and amino acid neurotransmitters in the hippocampus and cortex of rats. Animals were deprived of paradoxical sleep for 72h by using the multiple platform method. The EEG power spectral analysis was carried out to assess the brain's electrophysiological changes due to sleep deprivation. The concentrations of amino acid neurotransmitters were assessed in the hippocampus and cortex using HPLC. Control data showed slight differences from normal animals in the delta, theta and alpha waves while an increase in the beta wave was obtained. After 24h of PSD, delta relative power increased and the rest of EEG wave's power decreased with respect to control. After 48h and 72h the spectral power analysis showed non-significant changes to control. The amino acid neurotransmitter analysis revealed a significant increase in cortical glutamate, glycine and taurine levels while in the hippocampus, glutamate, aspartate, glutamine and glycine levels increased significantly. Both the waking EEG and neurotransmitter analyses suggest that PSD induced neurochemical and electrophysiological changes that may affect brain proper functionality.
