Abstract Polycomb repressive complex 2 (PRC2) catalyzes the writing of the tri-methylated histone H3 at Lys27 (H3K27me3) epigenetic marker and suppresses the expression of genes, including tumor suppressors. The function of the complex can be partially antagonized by the SWI/SNF chromatin-remodeling complex. Previous studies have suggested that PRC2 is important for the proliferation of tumors with SWI/SNF loss-of-function mutations. In the present study, we have developed an EED-directed allosteric inhibitor of PRC2 termed BR0063, which exhibits anti-proliferative properties in a subset of solid tumor cell lines harboring mutations of the SWI/SNF subunits, SMARCA4 or ARID1A. Tumor cells sensitive to BR0063 exhibited several distinct phenotypes, including cell senescence, which was mediated by the up-regulation of CDKN2A/p16. Further experiments revealed that the expression of p16 was suppressed in the BR0063-resistant cells via DNA hypermethylation in the CpG island (CGI) promoter region, rather than via PRC2 occupancy. The expression of TET1, which is required for DNA demethylation, was found to be inversely correlated with p16 CGI methylation, and this may serve as a biomarker for the prediction of resistance to PRC2 inhibitors in SWI/SNF LOF tumors.

Abstract Serotonergic neurons in the central nervous system control behavior and mood, but knowledge of the roles of serotonergic circuits in the regulation of immune homeostasis is limited. Here, we employ mouse genetics to investigate the functions of enteric serotonergic neurons in the control of immune responses and find that these circuits regulate IgA induction and boost host defense against oral, but not systemic Salmonella Typhimurium infection. Enteric serotonergic neurons promote gut-homing, retention and activation of intestinal plasmacytoid dendritic cells (pDC). Mechanistically, this neuro-immune crosstalk is achieved through a serotonin-5-HT receptor 7 (HTR7) signaling axis that ultimately facilitates the pDC-mediated differentiation of IgA+ B cells from IgD+ precursors in the gut. Single-cell RNA-seq data further reveal novel patterns of bidirectional communication between specific subsets of enteric neurons and lamina propria DC. Our findings thus reveal a close interplay between enteric serotonergic neurons and gut immune homeostasis that enhances mucosal defense.

Abstract Introduction The evidence regarding the effectiveness of Lanzhou Lamb Rotavirus Vaccine (LLR) and RotaTeq (RV5) against gastroenteritis (RVGE) caused by emerging genotypes in Chinese children remains limited. Methods We conducted a test-negative case–control study using gastroenteritis surveillance data from four cities (2020–2023) in Guangdong Province, China. Children aged 2 months to 5 years hospitalized with acute gastroenteritis were enrolled. Cases were rotavirus-positive; controls were rotavirus-negative. Vaccine effectiveness (VE) was estimated using multivariable logistic regressions. Results Among 2650 children, 218 (8.2%) were rotavirus-positive, predominantly G8P[8]. Also, 1543 (58.23%) children were unvaccinated, while 632 (23.85%) and 475 (17.92%) received at least one dose of RV5 and LLR, respectively. Adjusted RV5 VE against any RVGE severity was 51.7% [95% confidence interval (CI) − 58.1–85.3%]) for one dose, 37.6% (95% CI − 58.5–75.4%) for two doses, and 64.1% (95% CI 38.0–79.2%) for three doses. For LLR, VE against any RVGE severity was 38.7% (95% CI 5.7–60.2%) for one dose, 74.6% (95% CI 35.3–90.0%) for two doses, and 58.8% (95% CI − 217.6–94.6%) for three doses. Against severe RVGE, RV5 VE was 67.2% (95% CI − 144.7–95.6%) for one dose, 74.0% (95% CI − 92.1–96.5%) for two doses, and 86.6% (95% CI 56.8–95.9%) for three doses. For LLR, VE against severe RVGE was 57.7% (95% CI 20.3–77.6%) for one dose, 73.4% (95% CI 11.9–92.0%) for two doses, and − 27.8% (95% CI − 949.7–84.4%) for three doses. Conclusions Both RV5 and LLR provided protection against RVGE, including the emerging G8P[8] genotype. Three doses of RV5 offered strong protection, while two doses of LLR also appeared to be an effective strategy against rotavirus infection.

Abstract Thallium, a highly toxic pollutant, shows greater toxicity to human than other common heavy metals such as mercury, lead, cadmium and its effective removal from wastewater gains great attention. The main restriction for the Tl+ removal is the interference of a high concentration of co-existing ions in wastewater. Therefore, the goal of the current work was to synthesis adsorbent with high selectivity for the Tl+ removal. Herein, the pore size sieving strategy was proposed and Prussian blue-impregnated biochar (BC@PB) particles was synthesized. More than 95% Tl+ can be removed even the concentrations of the coexistence ions (Na+, Cd2+, and Zn2+) 1,000 higher than the initial concentration of Tl+ (500 μg/L). BC@PB also showed large adsorption capacity (9365 μg/g) and more than 99% Tl+ (initial concentration, 500 μg/L) were removed in just 1 min. The BC@PB had excellent and stable Tl+ removal ability (> 99%) over a range of pH from 3 to 9, which covered the pH range of common thallium-containing wastewater. The density functional theory (DFT) calculation confirmed that not only hydrated volume but also the hydration free energy of ions, which governed the energy barrier for ions entering into narrow channels of BC@PB, played essential roles on the selectivity removal of Tl+. Overall, due to its high selectivity, high adsorption capacity and easy preparation process, the synthesized BC@PB particles based on the pore sizing sieving strategy, can be a promising candidate for the removal of thallium from wastewater.

Abstract Understanding the mechanical characteristics of rocks when subjected to direct tension is crucial for assessing the stability of rock formations. Within the scope of this research, a series of tests was conducted using Tage tuff to assess the deformation behavior and crack extension of rock under direct tension. The axial, lateral, and shear strain fields as well as crack propagation, localized deformation behavior, and failure mode of the rocks were analyzed using three-dimensional digital image correlation method. The results showed that the axial strain fields on the specimen surface were heterogeneous, with different locations and localization occurring in the pre-peak stage, which was similar to the evolution of shear strain, whereas the lateral strain only showed slight changes. The crack extension direction was inferred, indicating that both tensile and shear stress occurred in the tests. Furthermore, different stress–strain responses were observed for the inside and outside of the localized bands. Then, the surface patterns of specimen failure were scanned and analyzed to assess the failure mode and residual strength of the specimen under direct tensile stress. Finally, the results of direct tension, uniaxial compression, and Brazilian split tests for Tage tuff were compared, and the complete stress–strain curve of uniaxial tension (UT) was simulated using a nonlinear-variable-compliance constitutive equation. This study provides a deeper understanding into the damage behavior of rocks under direct tension.

Lung cancer, which is a leading cause of cancer-related deaths worldwide, frequently metastasizes to the bones, significantly diminishing patients’ quality of life and complicating treatment strategies. This study aims to develop an advanced 3D Mask R-CNN model, enhanced with the ConvNeXt-V2 backbone, for the automatic segmentation of bone tumors and identification of lung cancer metastasis to support personalized treatment planning. Data were collected from two hospitals: Center A (106 patients) and Center B (265 patients). The data from Center B were used for training, while Center A’s dataset served as an independent external validation set. High-resolution CT scans with 1 mm slice thickness and no inter-slice gaps were utilized, and the regions of interest (ROIs) were manually segmented and validated by two experienced radiologists. The 3D Mask R-CNN model achieved a Dice Similarity Coefficient (DSC) of 0.856, a sensitivity of 0.921, and a specificity of 0.961 on the training set. On the test set, it achieved a DSC of 0.849, a sensitivity of 0.911, and a specificity of 0.931. For the classification task, the model attained an AUC of 0.865, an accuracy of 0.866, a sensitivity of 0.875, and a specificity of 0.835 on the training set, while achieving an AUC of 0.842, an accuracy of 0.836, a sensitivity of 0.847, and a specificity of 0.819 on the test set. These results highlight the model’s potential in improving the accuracy of bone tumor segmentation and lung cancer metastasis detection, paving the way for enhanced diagnostic workflows and personalized treatment strategies in clinical oncology.

Objective: There is limited research on the effectiveness of orthopedic robot-assisted treatment for sarcopenia combined with femoral neck fractures (FNF) in clinical settings. This study aimed to compare the clinical outcomes of orthopedic robot-assisted screw placement with traditional manual screw placement for treating sarcopenia combined with FNF. Methods: The retrospective study included patients with FNF under the age of 65 who were admitted to the Department of Orthopedics at XX Hospital Affiliated to XX University between January 2019 and March 2022 and underwent internal fixation with cannulated screws. Grip strength of the dominant hand was measured, and the skeletal muscle index (SMI) was calculated based on the muscle tissue area at the level of the 12th thoracic vertebrae on chest CT divided by height squared. Sarcopenia was diagnosed when SMI was below 42.6 cm2/m2 for men or 30.6 cm2/m2 for women. Patients were then grouped based on whether they underwent orthopedic robot-assisted surgery, as the robot-assisted (RA) group or the freehand (FH) group. Follow-up evaluations over 24–36 months included various parameters such as operation time, blood loss, X-ray shots, number of needle drilling, cannulated nail parallelism, C-reactive protein (CRP) levels, time to weight-bearing, perioperative infection, VAS (visual analogue scale) scores at 1 and 6 months post-surgery, Harris hip score at 6 months post-surgery, and incidence of nonunions and femoral head necrosis within 24 months post-surgery. Results: A total of 127 participants were enrolled in this study, with 46 individuals in the RG group and 81 in the FG group. Across both groups, factors such as gender, age, SMI, injury sides, Garden classifications, intraoperative blood loss, preoperative CRP, and VAS scores at 6 months post-surgery did not show statistically significant differences (p > 0.05). The RA group exhibited shorter operation time, fewer intraoperative X-ray shots, less needle drilling, and lower postoperative CRP levels compared to the FH group. The VAS score of the RA group was lower one month after surgery, whereas the Harris score was higher six months post-surgery (p 0.05). Objective: Traumatic brain injury (TBI) is among the leading causes of death and disability globally. Identifying and assessing the risk of in-hospital mortality in traumatic brain injury patients at an early stage is challenging. This study aimed to develop a model for predicting in-hospital mortality in TBI patients using prehospital data from China. Methods: We retrospectively included traumatic brain injury patients who sustained injuries due to external forces and were treated by pre-hospital emergency medical services (EMS) at a tertiary hospital. Data from the pre-hospital emergency database were analyzed, including demographics, trauma mechanisms, comorbidities, vital signs, clinical symptoms, and trauma scores. Eligible patients were randomly divided into a training set (241 cases) and a validation set (104 cases) at a 7:3 ratio. Least absolute shrinkage and selection operator (LASSO) and multivariate logistic regression were employed to identify independent risk factors. Analyzed the discrimination, calibration, and net benefit of the nomogram across both groups. Results: 17.40 % (42/241) of TBI patients died in the hospital in the training set, while 18.30 % (19/104) in the validation set. Abstract Background Patients with cold tumors gain limited benefits from immune checkpoint blockade (ICB) therapy owing to low programmed cell death protein ligand 1 (PD‐L1) expression and minimal immune cell infiltration. Mild photothermal therapy (PTT) using black phosphorus nanosheets (BPNSs) is a promising approach to enhance the efficacy of ICB therapy. However, to ensure that BPNS‐based PTT‐enhanced ICB therapy is clinically adaptable, a noninvasive, bedside‐accessible imaging tool capable of monitoring the status of PD‐L1 is imperative. We demonstrated that positron emission tomography (PET) using [64Cu]HKP2202 precisely delineated PD‐L1 expression in tumors receiving PTT. Methods BPNSs were modified with polyethylene glycol to prepare BPNS@PEG, which were then characterized. MC38 cells and tumor allografts were treated with BPNS@PEG followed by 808 nm near‐infrared light exposure. PET using [64Cu]HKP2202 was performed to monitor PD‐L1 expression in vivo. We also evaluated whether the efficacy of ICB therapy improved after delivering BPNS@PEG‐based PTT. Results BPNS@PEG had a well‐defined lamellar structure with clear edges and an average size of 150 nm. PET and Western blotting assays indicated that PD‐L1 expression was upregulated after BPNS@PEG and NIR‐light treatment. Notably, the antitumor effect of anti PD‐L1 therapy was enhanced in mice treated with BPNS@PEG‐based PTT. Conclusions BPNS@PEG had the capacity to convert cold tumors into hot tumors to facilitate the efficacy of ICB therapy. Importantly, the complementary diagnostic PET radiotracer targeting PD‐L1 allowed real‐time monitoring of PD‐L1 expression in the tumor microenvironment to guide ICB administration, holding great potential to achieve efficient and precise tumor immunotherapy.

Natural basalt can be processed into rod-like basalt fibers (BFs) and flake-like basalt scales (BSs) through high-temperature melting, forming and other processes. BFs and BSs have great application prospects in the field of organic coatings. BFs possess a regular rod-like structure and excellent mechanical properties, which are mainly used to enhance the mechanical properties of organic coatings. BSs possess a relatively irregular flake-like structure, which can form “maze” effects and layered structures to enhance corrosion resistance of organic coatings. However, basalt materials have poor compatibility, adhesion, and dispersion in organic resins due to their chemical inertness, resulting in a weak basalt-resin interface. Basalt-resin interface is an important parameter that determines the service performance of basalt composite coatings. Therefore, it is necessary to improve basalt-resin interface through corresponding strategies, such as chemical etching, chemical grafting and plasma treatment. Firstly, the surface modification strategies for improving basalt-matrix interface were summarized in this paper. Secondly, we discussed the effects of BFs and BSs on corrosion resistance of organic coatings and corrosion protection mechanism of composite coatings. Finally, we reviewed the research progress of modified basalt materials reinforced organic coatings.

Background: Some systematic reviews (SRs) on triple therapy (consisting of long-acting β 2 -agonist, long-acting muscarinic antagonist, and inhaled corticosteroid, LABA/LAMA/ICS) for chronic obstructive pulmonary disease (COPD) have reported conflicting results. As the number of syntheses increases, the task of identifying and interpreting evidence becomes increasingly complex and demanding. Objectives: To provide a comprehensive overview of the efficacy and safety of triple therapy for COPD. Design: Overview of SRs. Methods: Two independent reviewers conducted comprehensive searches in PubMed, Embase, Web of Science, and the Cochrane Library to identify relevant SRs that compared triple therapy with any non-triple therapy for COPD, from the inception of these databases until 1 June 2023. The AMSTAR 2 and GRADE tools were utilized to assess the quality of the included studies and the evidence for each outcome. Results: Eighteen SRs encompassing 30 original studies and involving 47,340 participants were analyzed. The overall AMSTAR 2 rating revealed that 3 SRs were of low quality, 13 SRs were of critically low quality, and 2 SRs were of high quality. No high-certainty evidence revealed a significant advantage of triple therapy in improving lung function or reducing acute exacerbations. However, all evidence, including one high certainty, supported the benefits of improving quality of life. Regarding all-cause mortality, no significant difference was found when compared to LAMA or ICS/LABA; however, high-certainty evidence confirmed its effectiveness when compared with LABA/LAMA. Notably, high-certainty evidence indicated that triple therapy was associated with a significant increase in the risk of pneumonia compared to LABA/LAMA. Conclusion: Triple therapy demonstrated notable benefits in improving lung function, reducing exacerbations, improving quality of life, and reducing all-cause mortality. However, it is important to note that it may also significantly increase the risk of pneumonia. ObjectiveTo evaluate the efficacy and safety of intranasal insulin on postoperative cognitive dysfunction (POCD) in elderly patients after laparoscopic radical resection of colorectal cancer.MethodsOlder patients scheduled for laparoscopic radical resection of colorectal cancer at Beijing Luhe Hospital, Capital Medical University, between August 2023 and November 2023, were enrolled in this double-blind pilot study. Patients were randomized to the control and insulin groups at a 1:1 ratio. The primary outcome was the rate of POCD at postoperative 7 days.ResultsA total of 61 patients (30 in the insulin group) were analyzed. The insulin group had a significantly lower POCD rate compared with the control group at postoperative day 7 [4(13.3%) vs. 12 (38.7%), p = 0.024]. The serum levels of IL-6, TNF-α and S100β at T2-5 in the insulin group were significantly lower than those of the control group (IL-6: mean difference at T2, −4.14, p = 0.036; T3, −3.84, p = 0.039; T4, −3.37, p = 0.013; T5, −2.57, p = 0.042; TNF-α: mean difference at T2, −3.19, p = 0.002; T3, −2.35, p = 0.028; T4, −2.30, p = 0.019; T5, −1.96, p = 0.0181; S100β: mean difference at T2, −8.30, p = 0.019; T3, −23.95, p = 0.020; T4, −20.01, p = 0.023; T5, −17.67, p = 0.010). No insulin allergic reactions, nasal irritation, or hypoglycemic reactions were observed in either of the groups.ConclusionIntranasal insulin may decrease the risk of POCD and inhibit the elevated serum IL-6, TNF-α, and S100β levels in elderly patients after laparoscopic radical resection of colorectal cancer, which proves that intranasal insulin may be a promising therapeutic option for POCD.Clinical trial registrationIdentifier, ChiCTR2300074423.

Abstract As satellite imagery technology advances, remote sensing plays an increasingly prominent role in modern society. Nevertheless, the limitations of existing imaging sensors and complex atmospheric conditions constrain the quality of raw remote sensing data, posing challenges for interpretation and noise reduction. Super‐resolution technology focuses on enhancing low‐quality, low‐resolution remote sensing images. In this study, we introduce a method that utilizes a high‐order degradation model to generate low‐resolution remote sensing images. We employ a Generative Adversarial Network with a Convolutional Block Attention Module (CBAM‐GAN) to enhance these images, reducing noise interference and improving texture and feature display. Our approach outperforms other methods on the UCMerced‐LandUse, WHU‐RS19, and AID datasets. Specifically, it raises SSIM index scores to 0.9443, 0.8928, and 0.8633, respectively, exceeding baselines by 1.31%, 0.19%, and 1.30%. The MOS index also improves to 3.98, 3.96, and 3.83, respectively, representing a 2.31%, 8.20%, and 2.96% gain over the baseline. Our reconstruction produces superior results, demonstrating the effectiveness of our proposed method.

This paper investigates a fuzzy adaptive fast finite-time control method for nonlinear pure-feedback systems (PFSs) with time-varying actuator faults. Using the mean-value theorem (MVT) to handle the non-affine functions in the system and to effectively decouple the control input signals. By integrating the approximation ability of fuzzy logic systems (FLSs), the fast finite-time control theory, and the backstepping techniques, a fuzzy-based fast finite-time fault-tolerant control method is proposed. Meanwhile, the fault-tolerant controller designed in this paper can effectively compensates the actuator faults. The theoretical analysis verifies that the strategy ensures the convergence of the tracking error and all closed-loop signals are bounded in fast finite-time. Finally, the effectiveness and feasibility of the proposed control strategy are verified by two numerical simulations.

This paper investigates the fixed-time fuzzy adaptive tracking control method based on Barrier Lyapunov Functions (BLFs) for nonlinear pure-feedback systems with full-state constraints and actuator faults. The Mean-Value Theorem (MVT) is used to transform the pure-feedback systems (PFSs) with non-affine terms into a strict-feedback structure. The BLFs are constructed to ensure that all states of the system are specified within the constraints, and the approximation ability of Fuzzy Logic Systems (FLSs) is used to handle the unknown nonlinear functions. By using the backstepping technique, a fuzzy adaptive fixed-time controller is constructed to compensate for possible faults in the actuator. Theoretical analysis proves the convergence of the tracking error in fixed-time and the boundedness of all signals in the closed-loop system. Finally, simulation results verify the established theoretical conclusions and show the effectiveness of the proposed scheme.

Abstract Objective To evaluate and summarize systematic reviews of the effects and safety of awake prone positioning for COVID-19-related acute hypoxaemic respiratory failure. Methods A comprehensive search was conducted on PubMed, Embase, the Cochrane Library, Web of Science, CNKI, CSPD, CCD and CBM from their inception to March 28, 2023. Systematic reviews (SRs) of awake prone positioning (APP) for COVID-19-related acute hypoxaemic respiratory failure in adults were included. Two reviewers screened the eligible articles, and four reviewers in pairs extracted data and assessed the methodological quality/certainty of the evidence of all included SRs by AMSTAR 2 and GRADE tools. The overlap of primary studies was measured by calculating corrected covered areas. Data from the included reviews were synthesized with a narrative description. Results A total of 11 SRs were included. The methodological quality of SRs included 1 “High”, 4 “Moderate”, 2 “Low” and 4 “Critically low” by AMSTAR 2. With the GRADE system, no high-quality evidence was found, and only 14 outcomes provided moderate-quality evidence. Data synthesis of the included SR outcomes showed that APP reduced the risk of requiring intubation (11 SRs) and improving oxygenation (3 SRs), whereas reduced significant mortality was not found in RCT-based SRs. No significant difference was observed in the incidence of adverse events between groups (8 SRs). The corrected covered area index was 27%, which shows very high overlap among studies. Conclusion The available SRs suggest that APP has benefits in terms of reducing intubation rates and improving oxygenation for COVID-19-related acute hypoxemic respiratory failure, without an increased risk of adverse events. The conclusion should be treated with caution because of the generally low quality of methodology and evidence. Trial registration The protocol for this review was registered with PROSPERO: CRD42023400986. Registered 15 April 2023.

In this paper, a novel adaptive neural fault-tolerant control scheme is proposed for uncertain large nonlinear systems with sensor, actuator faults and dead zone. Due to the fault of the sensor, the actual state and the fault parameters are coupled, and a fault parameter separation method is designed for decoupling. The radial basis function neural network (RBFNN) is used to approximate the unknown interconnection functions in nonlinear systems, and combining the RBFNN and backstepping technology, an adaptive neural fault-tolerant controller is designed for nonlinear large-scale systems through ordinary Lyapunov function. The stability of the closed-loop system is verified by Lyapunov analysis, and obtained satisfactory tracking performance under the comprehensive influence of sensor, actuator faults and dead zone. Finally, the effectiveness of the proposed adaptive neural fault-tolerant control is illustrated by simulation of large-scale wind farm system.

Abstract Background To find a useful hypoxia non-invasive biomarker for evaluating early treatment response and prognosis to definitive chemoradiotherapy (dCRT) in patients with esophageal squamous cell carcinoma (ESCC), using blood oxygenation level-dependent (BOLD) magnetic resonance imaging (MRI). Methods The R2* values were obtained pre- and 2–3 weeks post-dCRT in 28 patients with ESCC using BOLD MRI. Independent samples t-test (normality) or Mann-Whitney U test (non-normality) was used to compare the differences of R2*-related parameters between the complete response (CR) and the non-CR groups. Diagnostic performance of parameters in predicting response was tested with receiver operating characteristic (ROC) curve analysis. The 3-year overall survival (OS) was evaluated using Kaplan Meier curve, log rank test, and Cox proportional hazards regression analysis. Results The post-R2*, ∆R2*, and ∆%R2* in the CR group were significantly higher than those in the non-CR group (P = 0.002, 0.003, and 0.006, respectively). The R2*-related parameters showed good prediction of tumor response, with AUC ranging from 0.813 to 0.829. The 3-year OS rate in patients with ∆R2* >-7.54 s− 1 or CR were significantly longer than those with ∆R2* ≤ -7.54 s− 1 (72.37% vs. 0.00%; Hazard ratio, HR = 0.196; 95% confidence interval, 95% CI = 0.047–0.807; P = 0.024) or non-CR (76.47% vs. 29.27%; HR = 0.238, 95% CI = 0.059–0.963; P = 0.044). Conclusions The preliminary results demonstrated that the R2* value might be a useful hypoxia non-invasive biomarker for assessing response and prognosis of ESCC treated with dCRT. BOLD MRI might be used as a potential tool for evaluating tumor oxygenation metabolism, which is routinely applied in clinical practice and beneficial to clinical decision-making. A large sample size was needed for further follow-up studies to confirm the findings.

Mudstone and shale formations developed in the first sub-member of the third member of the Shahejie Formation (Es31Member) in the Qikou Sag, Bohai Bay Basin due to continuous lacustrine sedimentation. Atomic force microscopy, nitrogen adsorption, high-pressure mercury intrusion porosimetry, and field emission scanning electron microscopy were used to quantify the pore structure of the three main lithofacies in the study area. The results show that the average porosity and permeability of the mudstone are 3.54% and 0.00087 md, respectively. The pore structure is complex, and only the laminated shale has a high correlation between porosity and permeability. The laminated shale contains intergranular and intercrystalline pores, whereas the bedded shale primarily has intergranular dissolution pores and intergranular pores. The pore diameters of the two shales are predominantly at the submicron level. The pores of the laminated shale are located in and between the felsic laminae, and the pore connectivity is high. This lithofacies type is believed to be the most promising location for continental shale oil exploration in desert regions. The bedded shale is dominated by isolated pores with a large pore radius and large fluctuation. In contrast, to the laminated shale, the bedded shale has a high content of brittle minerals and low bedding density and is more likely to produce high-angle hydraulic fractures. The rock facies should be the focus of future shale oil evaluations in desert regions.

The increasing environmental concerns of conventional antifouling coatings have led to the exploration of novel and sustainable solutions to address the biofouling caused by Limnoperna fortunei. As a rapidly expanding invasive species, the fouling process of Limnoperna fortunei is closely associated with microbial fouling, posing significant threats to the integrity of aquatic infrastructure and biodiversity. This review discusses recent progress in the development of non-toxic, eco-friendly antifouling coatings that are designed to effectively resist biofouling without using toxic chemicals. Recent research has focused on developing novel non-toxic coatings that integrate natural bioactive components with advanced material technologies. These formulations not only meet current environmental standards and exhibit minimal ecological impact, but also possess significant potential in preventing the attachment, growth, and reproduction of Limnoperna fortunei. This review aims to provide scientific guidance by proposing effective and sustainable solutions to address the ecological challenges presented by Limnoperna fortunei. The insights gained from current research not only reveal novel antifouling methods, but also identify key areas for further investigation aimed at enhancing performance and environmental compatibility.

The vertical distribution of the marine total suspended matter (TSM) concentration significantly influences marine material transport, sedimentation processes, and biogeochemical cycles. Traditional field observations are constrained by limited spatial and temporal coverage, necessitating the use of remote-sensing technology to comprehensively understand TSM variations over extensive areas and periods. This study proposes a remote-sensing approach to estimate the vertical distribution of TSM concentrations using MODIS satellite data, with the Bohai Sea and Yellow Sea (BSYS) as a case study. Extensive field measurements across various hydrological conditions and seasons enabled accurate reconstruction of in situ TSM vertical distributions from bio-optical parameters, including the attenuation coefficient, particle backscattering coefficient, particle size, and number concentration, achieving a determination coefficient of 0.90 and a mean absolute percentage error of 26.5%. In situ measurements revealed two distinct TSM vertical profile types (vertically uniform and increasing) and significant variation in TSM profiles in the BSYS. Using surface TSM concentrations, wind speed, and water depth, we developed and validated a remote-sensing approach to classify TSM vertical profile types, achieving an accuracy of 84.3%. Combining this classification with a layer-to-layer regression model, we successfully estimated TSM vertical profiles from MODIS observation. Long-term MODIS product analysis revealed significant spatiotemporal variations in TSM vertical distributions and column-integrated TSM concentrations, particularly in nearshore regions. These findings provide valuable insights for studying marine sedimentation and biological processes and offer a reference for the remote-sensing estimation of the TSM vertical distribution in other marine regions.

IntroductionLung cancer, with the highest global mortality rate among cancers, presents a grim prognosis, often diagnosed at an advanced stage in nearly 70% of cases. Recent research has unveiled a novel mechanism of cell death termed disulfidptosis, which is facilitated by glucose scarcity and the protein SLC7A11.MethodsUtilizing the least absolute shrinkage and selection operator (LASSO) regression analysis combined with Cox regression analysis, we constructed a prognostic model focusing on disulfidptosis-related genes. Nomograms, correlation analyses, and enrichment analyses were employed to assess the significance of this model. Among the genes incorporated into the model, CHRNA5 was selected for further investigation regarding its role in LUAD cells. Biological functions of CHRNA5 were assessed using EdU, transwell, and CCK-8 assays.ResultsThe efficacy of the model was validated through internal testing and an external validation set, with further evaluation of its robustness and clinical applicability using a nomogram. Subsequent correlation analyses revealed associations between the risk score and infiltration of various cancer types, as well as oncogene expression. Enrichment analysis also identified associations between the risk score and pivotal biological processes and KEGG pathways. Our findings underscore the significant impact of CHRNA5 on LUAD cell proliferation, migration, and disulfidptosis.ConclusionThis study successfully developed and validated a robust prognostic model centered on disulfidptosis-related genes, providing a foundation for predicting prognosis in LUAD patients.

Abstract The fates of terrestrial organic carbon (OCterr) during fluvial transport from land to ocean are still not well constrained. This study systematically examines the evolution and dynamics of OCterr along the river‐estuary‐coastal ocean continuum in three fluvial systems discharging to the Chinese marginal seas. The 14C‐depleted characteristics of bulk OC and molecular components of riverine suspended sediments and marine sediments suggest that the Chinese marginal seas are a significant sink of pre‐aged OCterr. Our study reveals significant apparent aging of OCterr within estuaries, likely due to degradation of (younger) labile components, and apparent rejuvenation of OCterr in shelf systems, likely reflecting inputs of younger OCterr from proximal sources along the sediment dispersal pathway. The aging and rejuvenation of OCterr along the river‐ocean continuum confounds the use of plant wax lipid 14C to constrain lateral transport times, and sheds light on more complex OCterr dynamics in marginal seas.

ObjectiveSepsis in pediatric patients can progress to severe sepsis, and identifying biomarkers of this progression may permit timely intervention to prevent it. This study aimed to investigate the ability of thrombin-antithrombin complex (TAT), α2-plasmininhibitor-plasmin complex (PIC) and tissue-type plasminogen activator-inhibitor complex (t-PAIC) to predict severe sepsis in pediatrics early.Methods148 eligible pediatric sepsis patients were enrolled in this study, and were then divided into those who progressed to severe sepsis (n = 50) or not (n = 98). Serum levels of TAT, PIC, and t-PAIC were analysed, and simplified pediatric critical illness score (PCIS) and DIC score were calculated on the day of pediatric sepsis diagnosis.ResultsCompared with sepsis patients, severe sepsis patients had higher levels of TAT, PIC and t-PAIC. Correlation analysis revealed that TAT, PIC and t-PAIC were significantly correlated with simplified PCIS and DIC score. ROC curve analysis suggested that TAT, PIC and t-PAIC could serve as biomarkers for predicting severe sepsis with the AUC up to 0.862, 0.759 and 0.851, respectively. Stratified analysis demonstrated that the patients with increased levels of TAT, PIC and t-PAIC had worse illness severity and clinical outcome. Univariate logistic regression analysis revealed that TAT, PIC and t-PAIC were all risk factors for severe sepsis, yet only TAT and t-PAIC were independent risk factors in multivariate model.ConclusionsTAT, PIC and t-PAIC could serve as biomarkers for predicting severe sepsis, and correlated with illness severity in pediatrics, what's more, serum levels of TAT and t-PAIC may be independent risk factors for pediatric severe sepsis.

Abstract The direct oxidation of methane to methanol (MTM) remains a significant challenge in heterogeneous catalysis due to the high dissociation energy of the C-H bond in methane and the high desorption energy of methanol. In this work, we demonstrate a breakthrough in selective MTM by achieving a high methanol space-time yield of 2678 mmol molCu−1 h−1 with 93% selectivity in a continuous methane-steam reaction at 400 °C. The superior performance is attributed to the confinement effect of 6-membered ring (6MR) voids in SSZ-13 zeolite, which host isolated Cu-OH single sites. Our results provide a deeper understanding of the role of Cu-zeolites in continuous methane-steam to methanol conversion and pave the way for further improvement.

Abstract The identification of effective drug targets and the development of bioactive molecules are areas of high need in cancer therapy. The phosphatidylinositol transfer protein alpha/beta isoform (PITPα/β) has been reported to play an essential role in integrating phosphoinositide trafficking and lipid metabolism in diverse cellular processes but remains unexplored as a potential target for cancer treatment. Herein, data analysis of clinical cancer samples revealed that PITPα/β expression is closely correlated with the poor prognosis. Target identification by chemical proteomic methods revealed that microcolin H, a naturally occurring marine lipopeptide, directly binds PITPα/β and displays antiproliferative activity on different types of tumour cell lines. Furthermore, we identified that microcolin H treatment increased the conversion of LC3I to LC3II, accompanied by a reduction of the level of p62 in cancer cells, leading to autophagic cell death. Moreover, microcolin H showed preeminent antitumour efficacy in nude mouse subcutaneous tumour models with low toxicity. Our discoveries revealed that by targeting PITPα/β, microcolin H induced autophagic cell death in tumours with efficient anti-proliferating activity, which sheds light on PITPα/β as a promising therapeutic target for cancer treatment.

Abstract Traffic congestion can lead to negative driving emotions, significantly increasing the likelihood of traffic accidents. Reducing negative driving emotions as a means to mitigate speeding, reckless overtaking, and aggressive driving behaviors is a viable approach. Among the potential methods, affective speech has been considered one of the most promising. However, research on humor-based affective speech interventions in the context of driving negative emotions is scarce, and the utilization of electroencephalogram (EEG) signals for emotion detection in humorous audio studies remains largely unexplored. Therefore, our study first designed a highly realistic experiment scenario to induce negative emotions experienced by drivers in congested traffic conditions. Subsequently, we collected drivers’ EEG signals and subjective questionnaire ratings during the driving process. By employing one-way analysis of variance (ANOVA) and t tests, we analyzed the data to validate the success of our experiment in inducing negative emotions in drivers during congested road conditions and to assess the effectiveness of humorous audio in regulating drivers’ negative emotions. The results indicated that humorous audio effectively alleviated drivers’ negative emotions in congested road conditions, with a 145.84% increase in arousal and a 93.55% increase in valence ratings compared to control conditions. However, it should be noted that humorous audio only restored drivers’ emotions to the level experienced during normal driving. Our findings offer novel insights into regulating drivers’ negative emotions during congested road conditions.

Abstract High levels of East Asian black carbon (BC) aerosols affect ecological and environmental sustainability and contribute to climate warming. Nevertheless, the BC sources in China, after implementing clean air actions from 2013‒2017, are currently elusive due to a lack of observational constraints. Here we combine dual-isotope-constrained observations and chemical-transport modelling to quantify BC’s sources and geographical origins in Shanghai. Modelled BC concentrations capture the overall source trend from continental China and the outflow to the Pacific. Fossil sources dominate (~70%) BC in relatively clean summer. However, a striking increase in biomass burning (15‒30% higher in a fraction of biomass burning compared to summer and 2013/2014 winter), primarily attributable to residential emissions, largely contributes to wintertime BC (~45%) pollution. It highlights the increasing importance of residential biomass burning in the recent winter haze associated with >65% emissions from China’s central-east corridor. Our results suggest clearing the haze problem in China’s megacities and mitigating climate impact requires substantial reductions in regional residential emissions, besides reducing urban traffic and industry emissions.

The geostationary earth orbit (GEO) represents a distinctive geosynchronous orbit situated in the Earth’s equatorial plane, providing an excellent platform for long-term monitoring of ionospheric total electron content (TEC) at a quasi-invariant ionospheric pierce point (IPP). With GEO satellites having limited dual-frequency coverage, the inclined geosynchronous orbit (IGSO) emerges as a valuable resource for ionospheric modeling across a broad range of latitudes. This article evaluates satellite differential code biases (DCB) of BDS high-orbit satellites (GEO and IGSO) and assesses regional ionospheric modeling utilizing data from international GNSS services through a refined polynomial method. Results from a 48-day observation period show a stability of approximately 2.0 ns in BDS satellite DCBs across various frequency signals, correlating with the available GNSS stations and satellites. A comparative analysis between GEO and IGSO satellites in BDS2 and BDS3 reveals no significant systematic bias in satellite DCB estimations. Furthermore, high-orbit BDS satellites exhibit considerable potential for promptly detecting high-resolution fluctuations in vertical TECs compared to conventional geomagnetic activity indicators like Kp or Dst. This research also offers valuable insights into ionospheric responses over mid-latitude regions during the March 2024 geomagnetic storm, utilizing TEC estimates derived from BDS GEO and IGSO satellites.

Objective There was no universally accepted classification system that describes LHBT lesions as a type of its' pathology in patients with shoulder pain. This study aimed to determine the correlation of anatomic variants of glenoid labrum attachment of long head of biceps tendon (LHBT) and to assess their association, if any, with its lesions in rotator cuff tear (RCT) patients. Methods All RCT patients from January 2016 to December 2019 were assessed arthroscopically to classify the LHBT labrum attachment according to its' anatomical location. A simplified classification was created to describe the LHBT as normal, tendinitis, subluxation or dislocation, partial tear and superior labral tear from anterior to posterior (SLAP) lesion beyond type II The RCT were classified as three types as partial, small to medium and large to massive. The correlation of variants of LHBT labral attachment with type of LHBT lesions in different RCT groups was evaluated. Results In total, 669 patients were included for evaluation. The attachment of the LHBT was entirely posterior in 23 shoulders (3.4%), posterior‐dominant in 81 shoulders (12.1%), and equal in 565 shoulders (84.4%). In equal distribution LHBT attachment group, age > 60 (odds ratio: 2.928, P < 0.001) and size of RCT (P < 0.001) were significant risk factors of LHBT lesions. In the analysis of all patients, comparing with the partial thickness rotator cuff tear (PTRCT), the odds ratio of small to medium RCT and large to massive RCT was 2.398 and 6.606 respectively. In addition, age > 60 (odds ratio: 2.854, P < 0.001) and size of RCT (P < 0.001) were significant risk factors of LHBT lesions. In posterior dominant group, size of RCT was a significant risk factor of LHBT lesions but not any others (P < 0.001). In entirely posterior group, no risk factor of LHBT lesions was found. It showed that the variation of LHBT attachment was not a significant risk factor of LHBT lesions in rotator cuff repaired patients (p = 0.075). Conclusions There are three types of LHBT labrum attachment in RCT patients on arthroscopic observation. 84.4% were equal distribution of LHBT attachment on glenoid labrum, followed by posterior‐dominant (12.1%) and entirely posterior type (3.4%) in present study. Although the variation of LHBT attachment was not a significant risk factor of LHBT lesion in rotator cuff repaired(RCR) patients, there were different risk factors among three LHBT labral attachment types. In RCR patients, age > 60 and RCT size were significant risk factors of LHBT lesions.

We investigated dual carbon isotopes within the vertical water column at sites 67-1 and 67-2 of the western equatorial Indian Ocean to determine the source and age of particulate organic carbon (POC) and thus evaluated the contributions of modern and fossil (aged) POC. The concentration of POC ranged from 7 to 47.3 μgC L−1, δ13CPOC values ranged from –31.8 to –24.4‰, and Δ14CPOC values ranged from –548 to –111‰. Higher values of δ13CPOC and Δ14CPOC near the surface indicated an influence of autochthonous POC, whereas decreasing trends toward the bottom suggested a contribution of aged OC sources to the total POC pool. The contribution of fossil POC was lower near the surface, accounting for only 12% and 6% of the total POC at sites 67-1 and 67-2, respectively; however, in the deeper layers below 1,000 m, the contribution of fossil POC increased to 52% and 44% of the total POC at the two sites. Mechanisms for the increased contributions of fossil OC within deeper POC include the inflow of aged OC from sediments resuspended near slopes, the adsorption of old dissolved organic carbon in deep water masses, and the impact of aged OC that may originate from hydrothermal sources. This study highlights the importance of aged OC in the carbon cycle of the equatorial Indian Ocean.

IntroductionThe design of high-pressure optical windows is one of the key tasks in deep-sea exploration, as it directly determines the reliability and maximum operating depth of underwater optical equipment.MethodIn this paper, mechanical modeling and finite element analysis methods are employed to study optical windows.Results and discussionResults show that changes in boundary conditions during loading significantly affect the stress distribution of the window. Based on this conclusion, an optimization design method is proposed, which involves the use of transition materials to reduce the impact of base deformation on the window. The study further analyzes the effects of the transition material’s Young’s modulus, Poisson’s ratio, thickness, and friction coefficient on the stress of the optical window. A calculation method for material selection criteria is proposed to select appropriate transition materials, and actual materials are used for verification. Finally, the reliability of the optimization design scheme based on transition materials is confirmed through high-pressure experiments.

Abstract Janus nanoparticles have aroused the interest of scholars because of their highly efficient emulsification of spilled oils in wastewater. In this work, interfacially active Janus hollow glass microparticles (J-HGMPs) of asymmetric wettability were designed and synthesized in order to achieve more efficient separation of emulsified oil droplets from oily wastewater. Surface characteristic techniques such as FTIR, SEM, zeta potential and contact angle measurements had been employed to assess the amphiphilic surface properties of J-HGMPs. The oil removal/recovery performance of J-HGMPs in different oil–water systems and their interfacial activities were studied. As a particulate emulsifier, J-HGMPs could remove/recover > 96% oil from oil–water mixed phase. The results showed that J-HGMPs had strong interfacial activities and anchored firmly at oil/water interfaces. This high adsorption energy was also evaluated and verified via the calculation of Gibbs free energy. Overall, this study provided a novel and low-cost oil recovery method via a convenient buoyancy force that could be effectively applied in the treatment of oil spills while achieving the goal of benign and green environmental protection.

Abstract Objectives The early detection and identification of stroke are essential to the prognosis of patients with suspected stroke symptoms out-of-hospital. We aimed to develop a risk prediction model based on the FAST score to identify the different types of strokes early for emergency medical services (EMS). Methods This retrospective observational study enrolled 394 stroke patients at a single center from January 2020 to December 2021. Demographic data, clinical characteristics, and stroke risk factors with patients were collected from the EMS record database. Univariate and multivariate logistic regression analysis was used to identify the independent risk predictors. The nomogram was developed based on the independent predictors, in which the discriminative value and calibration of the nomogram were verified by the receiver operator characteristic (ROC) curve and calibration plots. Results A total of 31.90% (88/276) of patients were diagnosed with hemorrhagic stroke in the training set, while 36.40% (43/118) in the validation set. The nomogram was developed based on the multivariate analysis, including age, systolic blood pressure, hypertension, vomiting, arm weakness, and slurred speech. Whether to use face priors in the face super-resolution (FSR) methods is a symmetry problem.Various face priors are used to describe the overall and local face features, making the generation of super-resolution face images expensive and laborious. FSR methods that do not require any prior information tend to focus too much on the local features of the face, ignoring the modeling of global information. To solve this problem, we propose a dual-path facial image super-resolution network (SwinDPSR) fused with Swin Transformer. The network does not require additional face priors, and it learns global face shape and local face components through two independent branches. In addition, the channel attention ECA module is used to aggregate the global and local face information in the above dual-path sub-networks, which can generate corresponding high-quality face images. The results of face super-resolution reconstruction experiments on public face datasets and a real-scene face dataset show that SwinDPSR is superior to previous advanced methods both in terms of visual effects and objective indicators. The reconstruction results are evaluated with four evaluation metrics: peak signal-to-noise ratio (PSNR), structural similarity (SSIM), learned perceptual image patch similarity (LPIPS), and mean perceptual score (MPS).

Variations in the coupling coefficient of loosely coupled transformers and dynamic loads have a significant impact on the overall performance of bidirectional inductive power transfer (BIPT) systems. However, a wide range of load and coupling coefficient variations are common in the actual charging process, which may cause the converter on both sides to operate in a hard switching state, resulting in switching noise, reduced efficiency, and potential safety concerns. In this paper, a triple-phase-shift control (TPSC) strategy is proposed to study the zero-voltage switching (ZVS) operating range and constant-voltage output (CVO) characteristics of the double-side-LCC (DS-LCC) topology. To ensure a CVO over the wide range of coupling coefficient variations, a dual-phase-shift control is introduced for AC voltage matching. Based on this, the third phase-shift angle control between the converters on both sides is introduced to ensure the ZVS realization. Meanwhile, the time-domain model is developed to analyze the rationality of the proposed third phase-shift angle and the ZVS operating range. Finally, the effectiveness of the proposed TPSC strategy is validated through a 1.5 kW experimental prototype with an air gap of 100–150 mm.

Annual outbreaks of floating Ulva prolifera blooms in the Yellow Sea have caused serious local environmental and economic problems. Rapid and effective monitoring of Ulva blooms from satellite observations with wide spatial-temporal coverage can greatly enhance disaster response efforts. Various satellite sensors and remote sensing methods have been employed for Ulva detection, yet automatic and rapid Ulva detection remains challenging mainly due to complex observation scenarios present in different satellite images, and even within a single satellite image. Here, a reliable and fully automatic method was proposed for the rapid extraction of Ulva features using the Tasseled-Cap Greenness (TCG) index from satellite top-of-atmosphere reflectance (RTOA) data. Based on the TCG characteristics of Ulva and Ulva-free targets, a local adaptive threshold (LAT) approach was utilized to automatically select a TCG threshold for moving pixel windows. When tested on HY1C/D-Coastal Zone Imager (CZI) images, the proposed method, termed the TCG-LAT method, achieved over 95% Ulva detection accuracy though cross-comparison with the TCG and VBFAH indexes with a visually determined threshold. It exhibited robust performance even against complex water backgrounds and under non-optimal observing conditions with sun glint and cloud cover. The TCG-LAT method was further applied to multiple HY1C/D-CZI images for automatic Ulva bloom monitoring in the Yellow Sea in 2023. Moreover, promising results were obtained by applying the TCG-LAT method to multiple optical satellite sensors, including GF-Wide Field View Camera (GF-WFV), HJ-Charge Coupled Device (HJ-CCD), Sentinel2B-Multispectral Imager (S2B-MSI), and the Geostationary Ocean Color Imager (GOCI-II). The TCG-LAT method is poised for integration into operational systems for disaster monitoring to enable the rapid monitoring of Ulva blooms in nearshore waters, facilitated by the availability of near-real-time satellite images.

Monoclonal antibodies require careful formulation due to their inherent stability limitations. Polysorbates are commonly used to stabilize mAbs, but they are prone to degradation, which results in unwanted impurities. KLEPTOSE® HPβCD (hydroxypropyl beta-cyclodextrin) has functioned as a stable stabilizer for protein formulations in our previous research. The current study investigates the collaborative impact of combining polysorbates and HPβCD as excipients in protein formulations. The introduction of HPβCD in formulations showed it considerably reduced aggregation in two model proteins, bevacizumab and ipilimumab, following exposure to various stress conditions. The diffusion interaction parameter revealed a reduction in protein–protein interactions by HPβCD. In bevacizumab formulations, the subvisible particle counts per 0.4 mL of samples in commercial formulations vs. formulations containing both HPβCD and polysorbates subjected to distinct stressors were as follows: agitation, 87,308 particles vs. 15,350 particles; light, 25,492 particles vs. 6765 particles; and heat, 1775 particles vs. 460 particles. Isothermal titration calorimetry (ITC) measurement indicated a weak interaction between PS 80 and HPβCD, with a KD value of 74.7 ± 7.5 µM and binding sites of 5 × 10–3. Surface tension measurements illustrated that HPβCD enhanced the surface activity of polysorbates. The study suggests that combining these excipients can improve mAb stability in formulations, offering an alternative for the biopharmaceutical industry.

The identification of multi-source signals with time-frequency aliasing is a complex problem in wideband signal reception. The traditional method of first separation and identification especially fails due to the significant separation error under underdetermined conditions when the degree of time-frequency aliasing is high. The single-mode recognition method does not need to be separated first. However, the single-mode features contain less signal information, making it challenging to identify time-frequency aliasing signals accurately. To solve the above problems, this article proposes a time-frequency aliasing signal recognition method based on multi-mode fusion (TRMM). This method uses the U-Net network to extract pixel-by-pixel features of the time-frequency and wave-frequency images and then performs weighted fusion. The multimodal feature scores are used as the classification basis to realize the recognition of the time-frequency aliasing signals. When the SNR is 0 dB, the recognition rate of the four-signal aliasing model can reach more than 97.3%.

Although laser technology brings convenience to production and daily life, it also causes high-energy damage. Therefore, there is an urgent need to develop optical limiting materials for laser protection. In this study, a novel nonlinear optical material, red/black phosphorus lateral heterojunction, is successfully prepared through solvothermal and ultrasonic treatment. Using the Z−scan method, the nonlinear optical properties of the red/black phosphorus heterojunction are determined at wavelengths of 532 and 1064 nm. These results indicate that the red/black phosphorus heterojunction exhibits reverse saturable absorption properties in 1.2.3-glycerol. Interestingly, the red/black phosphorus heterojunction shows an enhanced performance over red phosphorus by introducing the black phosphorus phase. Moreover, the red/black phosphorus heterojunction is doped into organically modified silicate gel glass with excellent broadband optical limiting performance. This study highlights the promising prospect of the red/black phosphorus heterojunction in the nonlinear optical and optical limiting fields.

Caprine arthritis encephalitis is an infectious disease caused by the caprine arthritis encephalitis virus that infects goats, sheep, and other small ruminants. An outbreak of CAEV could be extremely harmful to the goat farming industry and could cause severe economic losses. We designed specific primers and probes for the gag gene and established a TaqMan real-time quantitative polymerase chain reaction assay. This method’s correlation coefficient (R2) was >0.999, and the sensitivity of the assay to the plasmid-carried partial gag gene was approximately 10 copies/µL, 1000 times higher than that of conventional PCR. No specific fluorescence was detected for other sheep viruses. Using this method, we tested 776 asymptomatic sheep blood samples and 4 neurodegenerative sheep brain samples from six farms in eastern China, and the positivity rate was 0.77% (6/780). The gag gene was partially sequenced in the three positive samples and compared with the sequences from other representative strains in GenBank. The results revealed that all three strains belonged to the B1 subtype and were most closely related to the strains from Shanxi and Gansu, previously isolated in China, with their homology ranging from 97.7% to 98.9%. These results suggest that the designed RT-qPCR assay can be used to detect subclinical CAEV in sheep and that the virus is still present in eastern China.

The cytoskeleton is a fundamental component found in all eukaryotic organisms, serving as a critical factor in various essential cyto-biological mechanisms, particularly in the locomotion and morphological transformations of plant cells. The cytoskeleton is comprised of three main components: microtubules (MT), microfilaments (MF), and intermediate filaments (IF). The cytoskeleton plays a crucial role in the process of cell wall formation and remodeling throughout the growth and development of cells. It is a highly organized and regulated network composed of filamentous components. In the basic processes of intracellular transport, such as mitosis, cytokinesis, and cell polarity, the plant cytoskeleton plays a crucial role according to recent studies. The major flaws in the organization of the cytoskeletal framework are at the root of the aberrant organogenesis currently observed in plant mutants. The regulation of protein compartmentalization and abundance within cells is predominantly governed by the process of vesicle/membrane transport, which plays a crucial role in several signaling cascades.The regulation of membrane transport in eukaryotic cells is governed by a diverse array of proteins. Recent developments in genomics have provided new tools to study the evolutionary relationships between membrane proteins in different plant species. It is known that members of the GTPases, COP, SNAREs, Rabs, tethering factors, and PIN families play essential roles in vesicle transport between plant, animal, and microbial species. This Review presents the latest research on the plant cytoskeleton, focusing on recent developments related to the cytoskeleton and summarizing the role of various proteins in vesicle transport. In addition, the report predicts future research direction of plant cytoskeleton and vesicle trafficking, potential research priorities, and provides researchers with specific pointers to further investigate the significant link between cytoskeleton and vesicle trafficking.

Background: The readmission rate following hospitalization for chronic obstructive pulmonary disease (COPD) is surprisingly high, and frequent readmissions represent a higher risk of mortality and a heavy economic burden. However, information on all-cause readmissions in patients with COPD is limited. Objective: This study aimed to systematically summarize all-cause COPD readmission rates within 30 and 90 days after discharge and their underlying risk factors. Methods: Eight electronic databases were searched to identify relevant observational studies about COPD readmission from inception to 1 August 2022. Newcastle-Ottawa Scale was used for methodological quality assessment. We adopt a random effects model or a fixed effects model to estimate pooled all-cause COPD readmission rates and potential risk factors. Results: A total of 28 studies were included, of which 27 and 8 studies summarized 30- and 90-day all-cause readmissions, respectively. The pooled all-cause COPD readmission rates within 30 and 90 days were 18% and 31%, respectively. The World Health Organization region was initially considered to be the source of heterogeneity. We identified alcohol use, discharge destination, two or more hospitalizations in the previous year, and comorbidities such as heart failure, diabetes, chronic kidney disease, anemia, cancer, or tumor as potential risk factors for all-cause readmission, whereas female and obesity were protective factors. Conclusions: Patients with COPD had a high all-cause readmission rate, and we also identified some potential risk factors. Therefore, it is urgent to strengthen early follow-up and targeted interventions, and adjust or avoid risk factors after discharge, so as to reduce the major health economic burden caused by frequent readmissions. Trial registration: This systematic review and meta-analysis protocol was prospectively registered with PROSPERO (no. CRD42022369894).

Abstract Serine hydroxymethyltransferase 2 (SHMT2) plays an important role in converting serine to glycine and supplying carbon to one-carbon metabolism to sustain cancer cell proliferation. However, the expression, function, and underlying mechanisms of SHMT2 in clear cell renal cell carcinoma (ccRCC) remain largely unknown. In this study, we demonstrated that SHMT2 was upregulated in ccRCC tissues compared with controls and associated with patient survival. SHMT2 knockdown inhibited proliferation, migration, and invasion in ccRCC cells. Overexpression of SHMT2 promoted tumor progression. Mechanistically, SHMT2 depletion disrupted one-carbon metabolism, increased reactive oxygen species (ROS) levels, and decreased ATP levels via metabolic reprogramming, which destroyed cell homeostasis. The SHMT2 knockdown-induced stress activated autophagy. A mass of autophagosomes fused with lysosomes, resulting in lysosomal membrane permeabilization (LMP) and leakage of lysosomal contents into the cytoplasm, which eventually led to apoptosis. Our work reveals that SHMT2 functions as an oncogenic gene to promote ccRCC progression. SHMT2 depletion induces apoptosis by causing LMP through excessive activation of the autophagy-lysosome pathway via metabolic reprogramming.

In order to determine the mapping relationship between the off grid fault section and the fault current distribution, and find the shortest path between the off grid fault section and the fault current distribution, an off grid fault section location method based on Dijkstra algorithm was designed for the intelligent O＆amp;M equipment of the electro-optical communication network.The LightGBM algorithm in the data-driven method was selected to determine the mapping relationship between the off grid fault section and the fault current distribution through a large number of samples.Based on Dijkstra algorithm, the shortest path of mapping relationship was found, and the off network fault section was searched.The location platform for off grid fault section of intelligent operation and maintenance equipment of power optical communication network was designed to realize the location and alarm of off grid fault point, namely, signal loss of intelligent operation and maintenance equipment.The comparison experiment shows that the location result of this method was the closest to the actual location and the most accurate in the off network fault section location caused by the interruption of optical cable in the SDH ring network circuit area, the off network fault section location caused by the fault of optical cable in the multi way bus section, and the off network fault section location scenario caused by the fault of optical cable in the multi way bus section in the ring power area.It effectively improved the accuracy of fault location and has good applicability.

The core loss calculation is essential for the design and performance evaluation of high-frequency transformers. It is related to the dimensions, temperature rises performance, and operating efficiency of transformer designs. Some scholars have examined the correlation between the characteristics of the core and losses, such as the core air gap length and the core structure; some investigations have also analyzed the effects of harmonics, distortion flux, and other factors on core losses. However, in reality, the core operating environment of high-frequency transformers is very complex, and achieving the required accuracy of loss model calculations is difficult. This paper discussed the insufficiency of the classical Bertotti loss separation model in estimating core loss under non-sinusoidal excitation of high-frequency transformers. Based on the derivation and modification of the classical loss model, the magnetic performance test platform of the nanocrystalline core material was established. In order to obtain a higher-order correction calculation method for the core loss of high-frequency transformers under trapezoidal wave excitation, the experimentally measured loss curves of nanocrystalline materials were further corrected. Finally, the accuracy of the correction model was verified by changing the loss results by varying the trapezoidal wave rise time constant.

Abstract Background Comminuted fractures of the coronoid process significantly compromise the stability and function of the elbow joint. Reconstruction of the coronoid process with a prosthesis has been suggested as an alternative to restore the architecture. The purpose of this study was to investigate the strength and stability of various methods for the fixation of a coronoid prosthesis by finite element analysis. Materials and methods A coronoid prosthesis was designed based on the morphological information from computed tomography images acquired from 64 subjects in whom the top 40% of the coronoid process height was replaced. Four methods for the fixation of the prosthesis were suggested: (1) a double 2.0-mm fixation bolt, anterior to posterior; (2) a double 2.5-mm fixation bolt, anterior to posterior; (3) a single 4.0-mm fixation bolt, posterior to anterior; (4) a single 4.5-mm fixation bolt, posterior to anterior. The integrated prosthesis-bone constructs were analyzed via the finite element analysis of 10 simulated proximal ulna models with loading applied along the axis of the humerus and with three different elbow flexion angles (30°, 90°, and 130°). The maximum principal stress and the total deformation were quantified and compared. Results A coronoid prosthesis was developed. The maximum principal stress of the fixation bolts occurred around the neck of the fixation bolt. For a comparison of the strengths of the four fixation methods, the maximum principal stress was the lowest for fixation using a single 4.5-mm fixation bolt. The value of the maximum principal stress significantly decreased with increased elbow flexion angle for all fixation methods. The maximum deformation of the fixation bolts occurred at the head of the fixation bolt. For a comparison of the maximum deformations in the four fixation methods, the maximum deformation was the lowest for fixation using a single 4.5-mm fixation bolt. The value of the maximum deformation significantly decreased with increased elbow flexion angle for all fixation methods. Conclusions The present study suggested that fixation of a coronoid prosthesis with a single 4.5-mm fixation bolt from posterior to anterior is an excellent option in terms of the strength and stability. Objective To explore the clinical effects of wrist arthroscopy combined with oblique ulnar shortening osteotomy in the treatment of ulnar impaction syndrome. Methods This was a retrospective study of 60 patients with ulnar impaction syndrome who were admitted to our department from January 2016 to December 2019. According to different surgical methods, they were divided into an observation group and a control group, with 30 cases in each group. The control group was treated with oblique ulnar shortening osteotomy, and the observation group used wrist arthroscopy based on the control group. The two groups of patients were compared in terms of wrist function before and 12 months after surgery. We compared the Disabilities of the Arm, Shoulder, and Hand Score (DASH Score), Patient‐Rated Wrist Evaluation Score (PRWE Score), Visual Analog Score (VAS), and ulnar variation between the two groups at 12 months after surgery. The excellent and good rates by Mayo wrist score were compared between the two groups at the last follow‐up. Results All patients were followed up for 12–36 months, with an average of 14.5 months. Bone union was achieved at the oblique osteotomy of the ulna, with an average healing time of 13.6 weeks. The observation group was examined by wrist arthroscopy, and the triangular fibrocartilage complex (TFCC) Palmer classification was confirmed in three cases of type IIA, seven cases of type IIB, 14 cases of type IIC, and six cases of type IID. Compared with before surgery, the grip strength, flexion‐extension, ulnar radial deflection, and forearm rotation of the two groups of patients were significantly improved at 12 months after surgery. The DASH score, PRWE score, and VAS score of the observation group were better than those of the control group, and the difference in ulnar shortening length was not statistically significant between the two groups. The excellent and good rates of the observation group (93.3%) were better than those of the control group (87.5%) at the last follow‐up, and the difference was statistically significant. Conclusion Compared with oblique ulnar shortening osteotomy alone, combined wrist arthroscopy can better reduce the pain of patients with ulnar impaction syndrome, restore wrist function, and improve the excellent and good rates. Therefore, it is clinically worthy of promotion.

SYP71, a plant-specific Qc-SNARE with multiple subcellular localization, is essential for symbiotic nitrogen fixation in nodules in Lotus, and is implicated in plant resistance to pathogenesis in rice, wheat and soybean. Arabidopsis SYP71 is proposed to participate in multiple membrane fusion steps during secretion. To date, the molecular mechanism underlying SYP71 regulation on plant development remains elusive. In this study, we clarified that AtSYP71 is essential for plant development and stress response, using techniques of cell biology, molecular biology, biochemistry, genetics, and transcriptomics. AtSYP71-knockout mutant atsyp71-1 was lethal at early development stage due to the failure of root elongation and albinism of the leaves. AtSYP71-knockdown mutants, atsyp71-2 and atsyp71-3, had short roots, delayed early development, and altered stress response. The cell wall structure and components changed significantly in atsyp71-2 due to disrupted cell wall biosynthesis and dynamics. Reactive oxygen species homeostasis and pH homeostasis were also collapsed in atsyp71-2. All these defects were likely resulted from blocked secretion pathway in the mutants. Strikingly, change of pH value significantly affected ROS homeostasis in atsyp71-2, suggesting interconnection between ROS and pH homeostasis. Furthermore, we identified AtSYP71 partners and propose that AtSYP71 forms distinct SNARE complexes to mediate multiple membrane fusion steps in secretory pathway. Our findings suggest that AtSYP71 plays an essential role in plant development and stress response via regulating pH homeostasis through secretory pathway.

The quantitative characterization of the thermal conditions in the Tibetan Plateau has long been a focal point of global research. Downward shortwave radiation, as a crucial component, plays an important role in numerous land surface processes while also serving as a significant indicator of the plateau’s thermal state. In order to gain a more comprehensive understanding of the Earth’s radiation budget in the Tibetan Plateau region, this study undertook an evaluation of six radiation products (ISCCP-FH, CERES-SYN, GLASS DSR, Himawari-8, MCD18A1, and ERA5). Two sets of ground measurements (downward shortwave radiation values from 10 CMA sites and 6 sites provided by the National Tibetan Plateau Data Center) in 2015 and 2016 were used as validation data to verify the accuracy of the remote sensing products. The results show that in the Tibetan Plateau region, CERESC products show the highest accuracy among the six data products with a bias (relative bias) of −7.57 W/m2 (3.46%), RMSE (relative RMSE) of 32.77 W/m2 (14.99%), and coefficient of determination of 0.80. Among all products, only the ERA5 products overestimated the value of downward shortwave radiation in the Tibetan Plateau region with a bias (relative bias) of 15.62 W/m2 (7.14%). By employing a spatial resolution upscaling approach, we assessed the influence of varying spatial resolutions on the validation accuracy, with the results indicating minimal impact. Through an analysis of the impact of cloud factors and aerosol factors on the validation accuracy, it is deduced that ERA5, Himawari-8, and MCD18A1 products are significantly influenced by cloud factors, whereas the CERES-SYN product is notably affected by aerosol factors.
