This study aims to discuss the application value of KMC algorithm optimized by heuristic method in basketball big data analysis and visual management. Because the data in basketball big data is too complicated and incomplete, the extraction of information is not direct and effective enough. Based on the metaheuristic K-Means clustering (KMC) algorithm, the weights and genetic algorithm are introduced to optimize it, and the University of California at Irvine (UCI) data set is applied to analyze the big data clustering performance of the optimized KMC algorithm. The 2018-2019 season National Basketball Association (NBA) shooting guards are selected as the research objects, and the optimized KMC algorithm is used to process the data and analyze the NBA scoring functional factors. It is found that the number of clusters increased from 2 to 16. After optimization, the Between-Within Proportion (BWP) value of the KMC algorithm only drops by 0.35, and the improved BWP (IBWP) value only drops by 0.288, which shows the smallest drop among all the algorithms. When the number of nodes is 4, the running time of the optimized KMC algorithm for processing the COVTYPE data set is 1922 s after optimization, and the running time for processing the IRIS data set is the shortest (113 s). When the number of parallel nodes is 10, the speedup ratio of the optimized KMC algorithm for processing COVTYPE data set is 4.16, and the maximal expansion rate is 0.81. The clustering accuracy of traditional KMC algorithm is 89.33%. After optimization, the clustering accuracy of KMC algorithm is 98.67%. The leader factor, offensive contribution factor, shooting stability factor, and passing ability factor in the core grouping are all at the maximum, which are 0.59, 0.51, 0.47, and 0.43, respectively. The optimized KMC algorithm has been shown to reduce the number of iterations, reduce convergence time, and improve clustering accuracy. Immunoglobulin-A vasculitis (IgAV) is an immune-related systemic vasculitis with an unclear etiology. Genetic predisposition is now considered to be closely associated with the development of the disease, and it is essential to reveal the relationship between them. To explore the role of heredity in the disease, we performed a genome-wide association study (GWAS) of 496 IgAV cases and 7165 controls using an Illumina Infinium Global Screening Array chip.In the first stage of analysis, a significant correlation between the major histocompatibility complex (MHC) and IgAV was observed. Subsequently, human leukocyte antigen (HLA) analysis was conducted using a new large-scale Han-MHC reference panel. Fine mapping of IgAV risk in the MHC region indicated that two amino acid positions, 120 and 11, of HLA-DRB1 and three potential HLA alleles (HLA-DRB1∗04, HLA-DRB1∗16, and HLA-DRB1∗16:02) were significantly associated.Further stepwise conditional analysis demonstrated that 3 amino acid positions (120, 26, 96) of HLA-DRB1 and 6 HLA-DRB1 alleles (HLA-DRB1*04, HLA-DRB1*16, HLA-DRB1*01, HLA-DRB1*12:02, HLA-DRB1*10, and HLA-DRB1*15:02) were independent signals. Among them, the most significant signal was HLA-DRB1 amino acid Ser120 (OR = 1.59, p = 3.19 × 10Our study confirms that the pathogenesis of IgAV has a genetic component and that HLA-DRB1 is strongly associated with susceptibility to IgAV.

In order to explore the comprehensive performance of a new type of brake-circular disc brake, a multi-physics coupling analysis was established using ANSYS Workbench software to study the heat generation mechanism and air-cooled heat dissipation effect of the brake under different working conditions. Under the condition of a single braking condition, the maximum temperature of the peripheral disc brake is 98.1℃; under the condition of continuous braking, the maximum temperature of the brake is 170.22℃, and the highest temperature occurs on the outer surface. A thermo-fluid-solid coupling analysis of the peripheral disc brake is established. Compared with the heat generation of a single braking condition, the maximum temperature of the brake is reduced by 14.77℃, which shows that the effect of forced air cooling is not obvious. Use the test bench to compare the finite element analysis results. It can be seen from the bench experiment that as the cyclic braking progresses, the maximum temperature of each braking cycle basically increases in a gradient manner, and the experimental and simulation results are basically consistent. The maximum error between the experiment and the finite element simulation is 8.29℃ , Verified the reliability of finite element analysis.

BackgroundElevated red blood cell distribution width (RDW) and decreased platelet count (PLT) can be clinically relevant to the prognosis in cancer patients. However, their prognostic values in patients with diffuse large B-cell lymphoma (DLBCL) need to be further explored.MethodsHealthy donors (n = 130) and patients with DLBCL (n = 349) were included and evaluated retrospectively in this study. The prognostic influence of clinical and pathological factors including RDW and PLT on overall survival (OS) and progression-free survival (PFS) were studied by Kaplan-Meier curves. To evaluate the independent prognostic relevance of RDW and PLT, univariate and multivariate Cox proportional hazards regression models were applied. The adjusted IPI model was established based on the results of multivariate analysis, and verified by Harrell’s C statistical analysis.ResultsKaplan-Meier curves indicated that an elevated RDW value and thrombocytopenia are poor factors for OS (P P = 0.006) and PFS (P = 0.003,P P = 0.003) and decreased PLT count (HR =1.749, 95%CI = 1.010–3.028,P = 0.046) were both independent prognostic factors. The c-index of IPI and NCCN-IPI were increased when RDW level and PLT were supplemented in our cohort.ConclusionsOur study shows that elevated RDW level and decreased PLT are independent poor prognostic factors in newly diagnosed DLBCL patients. Adding RDW and PLT to the IPI score may improve its predictive ability, and the adjusted IPI may be more powerful in predicting the survival of DLBCL patients in the rituximab era.

Based on an acoustic emission (AE) test system, pull-out tests were conducted to study the bond behavior between corroded steel bars and concrete with different corrosion degrees. The characteristics of the AE signal for the pull-out specimens were analyzed. The results showed that the AE location was consistent with the actual crack development. The characteristics of the AE signal reflected the bond behavior, the failure process and the characteristics of the specimens under different degrees of corrosion. Two critical states of crack development and the process of splitting failure slip were obtained. As a result, the stress distribution tends to be uniform along the steel bar with increasing degree of corrosion, and the peak value gradually shifted toward the free end.

People with schizophrenia have a shortened life expectancy, with cardiovascular disease (CVD) being the primary contributor to this excessive mortality. A total of 466 inpatients with schizophrenia and 507 healthy community controls in the Chinese mainland were recruited in this study. Sociodemographic information, medical history, and smoking history were recorded. In addition, total cholesterol (TC), fasting blood glucose (FBG), triglycerides (TG), and high-destiny lipoprotein cholesterol (HDL-C) were analyzed. The 10-year CVD risk was significantly higher in patients with schizophrenia compared with healthy controls. Male schizophrenia patients had significantly higher Framingham risk scores (FRS) than the females. Patients with schizophrenia carried significantly greater risk factors of CVD; body-mass index (BMI), TG and smoking prevalence were significantly higher than in the health community controls, while FBG and HDL-C were on the contrary. Smoking was significantly associated with FRS among schizophrenia inpatients. Collectively, these results suggest that Han Chinese mainland patients with schizophrenia harbor a high 10-year CVD risk when compared with healthy controls, especially in males. CVD in schizophrenia patients requires greater attention by clinicians and researchers.

Background: The gas human exhaled contains many volatile organic compounds (VOCs), which is related to the health status of body. Analysis of VOCs has been proposed as a noninvasive diagnostic tool for certain cancers. Detailed research on the VOCs in gas exhaled by cell can characterize cell type specific metabolites and may be helpful to detect the cancer markers in clinical practice. Methods: Solid-phase microextraction (SPME) gas chromatography–mass spectrometry was used to detect VOCs in the headspace of tissue culture flask in non-Hodgkin’s lymphoma (NHL) cell line JEKO and acute mononuclear leukemia cell line SHI-1, to elaborate the characteristic gaseous biomarkers of hematological malignancies. While macrophage cells and lymphocytic cells were acted as control. The blank group was only the RPMI 1640 medium containing 10% fetal calf serum that without cells. Results: Comparing with control group, the concentration of dimethyl sulfide, 2,4-dimethylheptane, methylbenzene, o-xylene, dodecane, and 1,3-di-tert-butylbenzene in JEKO cells was relatively higher, while the concentration of ethanol, hexanal, and benzaldehyde was lower. In SHI-1 cells, the levels of 2,4-dimethylheptane, benzene, 4-methyldecane, chloroform, 3,7-dimethyl dodecane, and hexadecane were significantly elevated, but the levels of hexanol and cyclohexanol were distinctly reduced. Conclusions: This pilot study revealed that the malignant hematological cells could change the components of VOCs in the cell culture flask in a cell type-specific pattern. The traits of VOCs in our setting offered new strategy for hematological malignancies tracing, and would act as potential biomarkers in diagnosis of malignant hematological diseases.

In a low-resource setting, an ideal disease diagnostic modality is an eye visualization assay that can be performed in minutes by untrained personnel. This paper reports the development of a self-catalytic colorimetric assay to detect the existence of cervical cancer cells by naked eye. The assay consists of a target specific initiator/catalyst, an acid self-catalytic system, and a pH indicator solution. By taking the advantage that trace amount of nucleus staining molecule, hemalum, can function as the initiator, a stained cervical cancer cell with an enlarged nucleus can initiate the self-catalytic reaction in a defined reaction condition to produce a significant amount of acid, which leads to a color change of a pH indicator solution, thereby indicating the presence of the cancer cell. Amplification of at least a trillion fold was achieved within 1.5 min. Due to the minimum engagement of instruments, simplicity and low cost, the assay is a potential tool in clinical to function as a “yes/no” check to facilitate quick decisions for cervical cancer screening. By alternating the method whereby the initiator is associated with the target, the assay can be expanded to screen and detect protein or nucleic acid based biomarkers related to other epidemic diseases in the resource-limited settings.

An enzymeless glucose biosensor based on polypyrrole nanofibers-supporting Au nanoparticles (Au/PPyNFs) was investigated in this study. The Au/PPyNFs heterogeneous composite materials were synthesized in-situ via hydrogen bonding interactions for the assembly of polyethyleneimine (PEI) on the surface of polypyrrole nanofibers (PPyNFs). By changing the molar ratio of PPy to HAuCl4, Au/PPyNFs with different Au loadings were obtained. The morphology and composition of Au/PPyNFs were characterized using SEM, TEM, FTIR, XRD and XPS, respectively. The hybrids exhibited a high electrocatalytic activity toward glucose oxidation, which is prerequisite for the catalysts to be applied in amperometric glucose sensors. By using the nonenzymatic glucose sensor based on Au/PPyNFs, 0.2-13 mM glucose can be detected with a sensitivity of 1.003 mu A cm(-2) mM(-1) and a good linearity (R-2=0.9993) between current density and glucose concentration. The proposed glucose sensor provides a promising strategy to construct fast, sensitive, and anti-interfering amperometric sensors for early diagnosis and prevention of diabetes. Citral, 3,7-dimethyl-2,6-octadienal, is a key component of the essential oils extracted from several lemon-scented herbal plants. Besides its antifungal activity, the anticancer effect of citral was studied in recent years. In this study, we investigated the effect of citral on the acute promyelocytic leukemia cell line NB4. Citral treatment had an antiproliferative effect in NB4 cells via the induction of apoptosis assessed by morphology, proliferation assay, DNA electrophoresis, Annexin V-FITC/PI staining and caspase-3 activation. And citral induced apoptosis of NB4 cells in a dose- and time-dependent manner. In addition, citral treatment induced decreased mitochondrial membrane potential, indicating that citral induced apoptosis via the mitochondrial pathway. Bax up-regulation and Bcl-2 down-regulation on mRNA level and NF-κB down-regulation on protein level was found in this study, suggesting that Bcl-2, Bax and NF-κB may be involved in the mechanism of the apoptotic effect of citral on NB4 cells. These data suggest that citral has a potential therapeutic effect on leukemia.

Many reports have demonstrated that human bone marrow mesenchymal stem cells (BMMSCs) are resistant to several chemotherapeutic agents or ionic radiation when compared with sensitive tumor cell lines; however, the underlying molecular mechanism is rarely known. In our previous studies, we found that p53 family member p73 was not expressed in BMMSCs with or without the treatment of chemotherapeutic drugs, and the exogenous induction of p73 protein could reduce the resistance of BMMSCs to the drugs. In order to elucidate which factor leads to the inhibition of p73 expression, we used a methylation-specific polymerase chain reaction to investigate the epigenetic methylation status of the p73 gene promoter CpG region. Our data showed that the p73 gene promoter was hypermethylated in BMMSCs but not in tumor cell lines, which were sensitive toward chemotherapeutic agents. Using the demethylation agent 5-aza-2'-deoxycytidine significantly reactivated p73 expression both at the transcriptional and at the protein level. In addition, the treatment of 5-aza-2'-deoxycytidine rendered BMMSCs more sensitive to chemotherapeutic agents through the process of enhanced apoptosis cell death. Taken together, our results suggest that the silencing of the p73 gene mediated by promoter hypermethylation may play a crucial role in leading to the high resistance of BMMSCs to chemotherapeutic drugs and thus we conclude that the p73 gene may be an important element regulating human BMMSCs in response to DNA damage.

Gold nanoparticles supported on multi-walled carbon nanotubes (Au/MWCNTs) were prepared as electrocatalysts for an electrochemical sensor of acetylene. The catalysts were characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD) and X-ray photoelectron spectroscopy (XPS). The electrochemical experiments showed that the catalysts exhibited a distinctly higher activity for the acetylene electro-oxidation than Au-NPs and Au/C catalysts. In typical cyclic voltammograms, the onset potential of Au/MWCNTs for the acetylene oxidation is 0.73 V. which is 0.19 V more negative than that of Au-NPs. The acetylene sensor test showed good linear response towards acetylene concentration in a wide range from 10 ppm to 50 ppm with a steady response current for 300 s, good sensitivity of 80 nA/ppm and fast 90% response time (t(90)) of 25 s to 50 ppm acetylene. In this paper, sugar’s production, consumption and trade status in China are reviewed systematically, and the background and reasons for the substantial increase in sugar import are analyzed. We found that the main reasons for China sugar industry’s weakness and low international competitiveness include inefficient production technology, low level of domestic support and protection, and high production cost. And now it is difficult to resist the dumping of foreign sugar with lower price, and China sugar industrial’s safety issue therefore has been highlighted. Based on the analysis and forecast of the gap between domestic sugar production and demand in 2020, we put forward policy options for further developing china’s sugar industry.

Human adipose derived mesenchymal stem cells (ADMSCs) are multipotential stem cells, originated from the vascular stromal compartment of fat tissues which can be used as an alternative cell source for many different cell therapies. However, their response to chemotherapeutic agants remains unknown. Here we assessed the acute direct effects of individual chemotherapeutic drug on ADMSCs. Using an in vitro culture system, the response of ADMSCs to the three chemotherapeutic agents cisplatin, comptothecin and vincristine was determined in comparison with that of testicular germ cell tumour (TGCT) cell line. The recovery of cell numbers following exposure to chemotherapeutic agents were also evaluated. Our results showed that human ADMSCs were resistant to chemo-therapeutic agents which are commonly used in clinic, the full recovery was seen respectively in ADMSCs after the drug treatment. Moreover, ADMSCs maintained their stem cell characteristics in vitro after the exposure to all chemotherapeutic agents.
