To investigate the intestinal fermentation characteristics of different endopolysaccharides (MPS) sub-fractions MPS-1, MPS-2, MPS-3, and MPS-4 from Monascus purpureus mycelium, an in vitro pig colon model was used to determine the changes in molecular weight of the polysaccharides, the contents of short-chain fatty acids (SCFAs), and gut microbiota at different fermentation times. The results showed that after intestinal fermentation, the acetic acid, propionic acid, and butyric acid production in each MPS treatment group was higher. At 48 h of fermentation, the total SCFAs content produced by the MPS-2 and MPS-3 treatment groups increased significantly (PMPS-2>MPS-1>MPS-4 in descending order. The 16S rDNA results revealed that after MPS treatment, Firmicutes and Bacteroidetes were the main dominant bacteria at the phylum level, and the ratio of Firmicutes/Bacteroidetes was reduced. At the genus level, with the extension of fermentation time, the abundance of beneficial bacteria such as Bacteroides, Prevotella, and Parabacteroides increased, while the abundance of harmful genera, including Streptococcus and Clostridium, decreased significantly (P

The market value of sea urchin gonads is determined by the specific characteristics associated with gonad size and texture. Formulated feeds can effectively promote the gonad growth of sea urchins but cannot assure essential gonad texture traits. The objective of this study was to investigate the impact of vitamin C (VC) on the gonad growth, texture, collagen content, and the expression of genes involved in the collagen synthesis of sea urchins (Mesocentrotus nudus). Graded amounts of VC (0, 3000 and 6000 mg/kg) were supplemented to make three formulated feeds. Fresh kelp (Saccharina japonica) was used as the control diet. Each diet was randomly distributed to three tanks of M. nudus. The results indicated that the gonadosomatic index (GSI) and texture traits of M. nudus fed C3000 were significantly greater than those fed C0 and C6000. Collagen type I (Col I) in the gonads of M. nudus fed C3000 showed significantly greater areas than those fed C0 and C6000. Consistently, the expression levels of collagen alpha-1 (colp1α) of M. nudus fed C3000 were significantly higher than those fed C0 and C6000. As for the transforming growth factor beta (tgf-β)/Smads pathway, the expression levels of collagen synthesis genes (tgf-β receptor 1 and 2, smad nuclear-interacting protein 1 (snip1) and prolyl 4-hydroxylase subunit beta (p4hβ)) in the C3000 group were significantly greater than those in the C0, C6000 and kelp groups. On the contrary, the expression levels of collagen degradation genes (lysyl oxidase-like 2 (loxl2) and matrix metalloproteinase 14 (mmp14)) in the C3000 group were significantly lower than those in the C0, C6000 and kelp groups. In conclusion, VC at an addition level of 3000 mg/kg significantly increased the gonad texture and collagen contents of M. nudus, which could be accomplished by increasing collagen synthesis and inhibiting collagen degradation through the tgf-β/Smads pathway. These results could contribute to better understanding the beneficial effects of VC addition on the gonad texture quality of M. nudus.

To solve the problems of measurement errors led by mismatches of dense feature matching in machine vision structural deflection measurement, this paper proposes a dense feature extraction, matching, and dual-step mismatch-removal-based full-field structural dynamic deflection measurement method. First, the of dense feature detection and matching theory is introduced to extract the SIFT feature points on a structural surface in an image sequence and matched by FLANN to trace the structure movement, and the mechanisms and causes of mismatches are analyzed. Then, a dual-step mismatch removal method combining RANSAC and Structural Displacement Continuity Restriction (SDCR) is introduced to achieve full-field dynamic beam deflection measurement. The proposed method is validated through indoor cantilever beam experiments, and results show that the method can effectively eliminate a large number of SIFT feature mismatches (accounting for approximately 55% of the total matched feature points). The full-field dynamic displacement field of the beam can be measured with the correctly matched dense feature points by converting dense feature point displacements into continuous and uniform spatiotemporal deflections of the structure. A comparison with the GOM Correlate Professional DIC measurement system was conducted, and the maximum measurement error of the cantilever beam dynamic displacement of the proposed method is between 0.024 and 0.053 mm, the root mean squared error of displacement is approximately 0.01 mm, and the correlation coefficient between two deflection–time curves reaches 0.9964. The proposed algorithm is proven to be effective in full-field displacement measurement and has great potential in future structural health monitoring of bridges.

Silicon-based solid-state batteries (Si-SSBs) are now a leading trend in energy storage technology, offering greater energy density and enhanced safety than traditional lithium-ion batteries. This review addresses the complex challenges and recent progress in Si-SSBs, with a focus on Si anodes and battery manufacturing methods. It critically analyzes the lithiation process in Si anodes, emphasizing the significant volumetric expansion and evolving solid-electrolyte interphase (SEI), both of which are crucial for the electrochemical performance and durability of batteries. Innovations in anode design, such as morphological optimization and compositional alloying, have been explored to reduce mechanical stress and improve the electrochemical properties. Moreover, we elaborate in detail the essential steps in the layer-by-layer construction and encapsulation of Si-SSBs, which are vital for their stability and function. Finally, this review offers a thorough examination of present scientific and technological developments, providing insights and prospective pathways for researchers and industry professionals dedicated to advancing SSB technology.

In sustainable aquaculture systems, copper sulfate (CuSO4) is widely applied as a disinfectant to control parasitic infections and algal growth. However, aquatic organisms may suffer from exposure to excessive concentrations of Cu. Elevated Cu concentrations could activate damage to the respiratory functions of aquatic animals. Thus, this study explored the effects exerted by ferulic acid (FA) on respiratory metabolism, oxidation-related lesions, and the apoptosis parameters of the gills and red blood cells in copper sulfate pentahydrate (CuSO4·5H2O)-treated carp (Cyprinus carpio var. Jian). When the 30-day feeding experiment was completed, the carp were exposed to 12.5 μM of Cu for 4 days. The results indicated that the Cu decreased the oxygen consumption and ammonia excretion rates in the carp, reduced the metabolic- and antioxidant-related enzyme activities and glutathione levels in the carp, and enhanced the caspase activities and reactive oxygen species and malondialdehyde levels in the gills of the carp. Moreover, in addition to the changes in the above parameters, the Cu decreased the cell numbers and hemoglobin concentrations and increased the phosphatidylserine exposure and cytochrome c levels in the red blood cells of the carp. These results demonstrate that Cu is capable of decreasing respiratory metabolism and increasing oxidation-related lesions and apoptosis inside the gills and red blood cells of the fish. However, dietary FA quenched the Cu-induced apoptosis and oxidative lesions by reversing the same biomarker indicators, thereby suppressing the Cu-induced decrease in respiratory metabolism. Thus, FA can be used as a suppressor of Cu stress in fish.

To solve the problem of low accuracy in automatic concrete crack image segmentation and the non-standardization of concrete crack image datasets, an exposure-based concrete crack image capture scene characterization method was proposed, and the optimal exposure interval for crack segmentation was presented by multiple scene image capture experiments. First, current public crack datasets were collected and analyzed, and it was shown that improper spatial resolution, mislabeling, overexposure, and defocus are frequent non-standardization problems in crack dataset production. Through the analysis of the photoelectric principle in concrete crack imaging, an equivalent exposure was set as a core indicator for scene characterization. Twenty-one indoor scenes were designed by varying the illumination intensity and exposure time, and the experimental results showed that an equivalent exposure can be a core control index for scene characterization. The grayscale distribution law of concrete crack images was analyzed with four specimens’ images captured indoors in 50 exposure scenes, and the segmentation accuracy of an image from each scene was calculated through comparison with corresponding manually labeled binary files. The experiment’s results revealed that 5~50 lx·s was the optimal equivalent exposure interval for concrete crack image segmentation, in which better segmentation accuracy was achieved with an F1 score of up to 96.3%.

Li-ion cells with a LiMnxFe1−xPO4 (LMFP) and LiNi1−x−y−zCoxMnyAlzO2 (NCMA) blending cathode show their benefits of lower cost and higher safety compared to barely NCMA cathode-based cells. However, the rate capability of LMFP material is relatively poor compared to NCMA or even LiFePO4, which is because of the low electronic conductivity of LMFP material and the 1D diffusion channel in its structure. This work discusses the effect on electrochemical performance when blends of various ratios of LMFP are used in an NCMA cathode, with data verified by a 5 Ah pouch cell. This work further investigated the interaction between NCMA and LMFP during charge/discharge. Combining results from experiment and simulation, it evidences that blending more LMFP does not always lead to worse discharge rate but reduces charge rate. Moreover, it is found that, in a constant current discharge/charge process, although the system is under continuous discharge/charge, LMFP works intermittently. This leads to different diffusion polarization states of LMFP in the discharge/charge process and further results in a difference in discharge/charge rate capability. Therefore, to improve rate capability, especially charging rate, using smaller-sized or doped LMFP to improve its diffusion coefficient is an optimized strategy.

To improve the accuracy of concrete crack measurement with a machine vision method in structural health monitoring and in technical status evaluation, a subpixel crack measurement method based on the partial area effect was proposed. (1) First, a pixelwise crack image segmentation method was established through a multi-step process of multi-threshold fusion and morphology operation, and a novel pixel degree crack width calculation method was developed with the extraction of the middle points, the center line and its normal, and the intersection of the center line normal and crack edges. (2) Then, a subpixel algorithm based on the partial area effect was introduced to locate vertical, horizontal, and oblique cracks in subpixel crack edges, and the subpixel crack width could be calculated along the crack center line pixelwise. (3) Finally, the proposed method was verified by indoor concrete beam crack measurement tests with a digital microscope, and the results show that the maximum relative errors of the subpixel width of the horizontal, vertical, and oblique straight cracks measured by the proposed method were 3.06%, 8.97%, and 5.16%, respectively. The absolute error of the crack length was less than 0.30 mm, and the measurement accuracy could reach 0.01 pixels. The subpixel crack measurement method provides a novel possible solution for structural health monitoring.

ABSTRACT The microbial network of the soil-root continuum plays a key role in plant growth. To date, limited information is available about the microbial assemblages in the rhizosphere and endosphere of endangered plants. We suspect that unknown microorganisms in roots and soil play an important role in the survival strategies of endangered plants. To address this research gap, we investigated the diversity and composition of the microbial communities of the soil-root continuum of the endangered shrub Helianthemum songaricum and observed that the microbial communities and structures of the rhizosphere and endosphere samples were distinguishable. The dominant rhizosphere bacteria were Actinobacteria (36.98%) and Acidobacteria (18.15%), whereas most endophytes were Alphaproteobacteria (23.17%) as well as Actinobacteria (29.94%). The relative abundance of rhizosphere bacteria was higher than that in endosphere samples. Fungal rhizosphere and endophyte samples had approximately equal abundances of the Sordariomycetes (23%), while the Pezizomycetes were more abundant in the soil (31.95%) than in the roots (5.70%). The phylogenetic relationships of the abundances of microbes in root and soil samples also showed that the most abundant bacterial and fungal reads tended to be dominant in either the soil or root samples but not both. Additionally, Pearson correlation heatmap analysis showed that the diversity and composition of soil bacteria and fungi were closely related to pH, total nitrogen, total phosphorus, and organic matter, of which pH and organic matter were the main drivers. These results clarify the different patterns of microbial communities of the soil-root continuum, in support of the better conservation and utilization of endangered desert plants in Inner Mongolia. IMPORTANCE Microbial assemblages play significant roles in plant survival, health, and ecological services. The symbiosis between soil microorganisms and these plants and their interactions with soil factors are important features of the adaptation of desert plants to an arid and barren environment. Therefore, the profound study of the microbial diversity of rare desert plants can provide important data to support the protection and utilization of rare desert plants. Accordingly, in this study, high-throughput sequencing technology was applied to study the microbial diversity in plant roots and rhizosphere soils. We expect that research on the relationship between soil and root microbial diversity and the environment will improve the survival of endangered plants in this environment. In summary, this study is the first to study the microbial diversity and community structure of Helianthemum songaricum Schrenk and compare the diversity and composition of the root and soil microbiomes.

Introduction Post-COVID-19 depression (PCD) is a possible sequela of COVID-19. Some doctors have used acupuncture to treat PCD, but no systematic review or meta-analysis has yet evaluated its efficacy and safety for the treatment of PCD. The aim of this systematic review is to assess the efficacy and safety of acupuncture therapy for PCD.Methods and analysis Two reviewers will independently search the Cochrane Central Register of Controlled Trials (CENTRAL), Medline (PubMed), Excerpt Medica Database (EMBASE), China National Knowledge Infrastructure (CNKI), Chinese Biomedical Literature Database (CBM), Chinese Scientific Journal Database (VIP) and Wan-Fang Database from inception to 24 January 2023. Study selection, data extraction and assessment of study quality will be independently performed by two reviewers. If a meta-analysis is appropriate, Review Manager V.5.3 will be used for data synthesis; otherwise, a descriptive analysis will be conducted. Data will be synthesised using a fixed-effects or random-effects model, according to the results of a heterogeneity test. The results will be presented as risk ratios with 95% CIs for dichotomous data, and weighted mean differences or standardised mean differences with 95% CIs for continuous data.Ethics and dissemination The entire process used for this systematic review does not use private information, so ethical approval is not required. The results of this meta-analysis will be disseminated through publication in a peer-reviewed journal and/or conference presentations.PROSPERO registration number CRD42022379312.

ABSTRACT Arbuscular mycorrhizal fungi (AMF) have been reported to have a wide distribution in terrestrial ecosystems and to play a vital role in ecosystem functioning and symbiosis with Stipa grasses. However, exactly how AMF communities in the rhizosphere change and are distributed along different Stipa population with substituted distribution and their relationships remain unclear. Here, the changes and distribution of the rhizosphere AMF communities and their associations between hosts and the dynamic differences in the glomalin-related soil protein (GRSP) in the rhizosphere soil of seven Stipa species with spatial substitution distribution characteristics in arid and semiarid grasslands were investigated. Along with the substituted distribution of the Stipa populations, the community structures, taxa, species numbers, and alpha diversity index values of AMF in the rhizosphere changed. Some AMF taxa appeared only in certain Stipa species, but there was no obvious AMF taxon turnover. When the Stipa baicalensis population was replaced by the Stipa gobica population, the GRSP tended to decline, whereas the carbon contribution of the GRSP tended to increase. Stipa grandis and Stipa krylovii had a great degree of network modularity of the rhizosphere AMF community and exhibited a simple and unstable network structure, while the networks of Stipa breviflora were complex, compact, and highly stable. Furthermore, with the succession of zonal populations, the plant species, vegetation coverage, and climate gradient facilitated the differentiation of AMF community structures and quantities in the rhizospheres of different Stipa species. These findings present novel insights into ecosystem functioning and dynamics correlated with changing environments. IMPORTANCE This study fills a gap in our understanding of the soil arbuscular mycorrhizal fungal community distribution, community composition changes, and diversity of Stipa species along different Stipa population substitution distributions and of their adaptive relationships; furthermore, the differences in the glomalin-related soil protein (GRSP) contents in the rhizospheres of different Stipa species and GRSP’s contribution to the grassland organic carbon pool were investigated. These findings provide a theoretical basis for the protection and utilization of regional biodiversity resources and sustainable ecosystem development.

The objective of this study was to investigate the effects of Scoparia dulcis extract (SDE) on stress induced by high stocking density and Cu and trichlorfon exposure in crucian carp (Carassius auratus). The results showed that these stressors exerted detrimental effects in fish, such as inhibition of growth performance, reduced feed intake, and interruption of fish locomotion. Under high stocking density, dietary SDE supplementation increased the content of reduced glutathione (GSH) and the activities of amylase, catalase (CAT), and glutathione reductase (GR) and decreased the content of malonaldehyde (MDA) in the intestine of crucian carp. A similar trend was presented in the hepatopancreas under Cu exposure. Dietary SDE supplementation enhanced the activities of CAT, superoxide dismutase (SOD), glutathione peroxidase (GPx), lactate dehydrogenase, glutamate-oxaloacetate transaminase, and glutamate-pyruvate transaminase in the muscle of crucian carp under trichlorfon exposure. The optimum dietary SDE supplementation levels were 4.07, 4.33, and 3.95 g kg−1 diet based on the recovery rate of weight gain (RWG), feed intake (FI), and inhibitory rate of rollover (IR) for crucian carp under high stocking density and Cu and trichlorfon exposure, respectively. Overall, dietary supplementation with SDE may be a useful nutritional strategy for relieving these stresses in aquatic animals.

The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING)-mediated senescence-associated secretory phenotype (SASP) pathway has recently been identified in the suppression and promotion of cancers. However, its practical role in carcinogenesis remains to be comprehensively elucidated. Here, we describe an investigation analysing SASP activity and its correlations with DNA damage response (DDR), genomic mutations, and cell proliferation in gastric carcinogenesis among 30 cases with available endoscopic submucosal dissection (ESD) specimens of early neoplastic lesions (including low-grade dysplasia [LGD], high-grade dysplasia [HGD], and intramucosal carcinoma). The positive cells of senescence-associated β-galactosidase staining and cGAS, STING, interferon-regulatory factor 3 (IRF3), and signal transducer and activator of transcription 6 (STAT6) expression levels using immunostaining were elevated in HGD and in cancers. Similarly, increased expression of the Fanconi anemia group D2 (FANCD2) protein, tumour suppressor p53 binding protein 1 (TP53BP1), and replication protein A (RPA2) (i.e., primary DDR factors) was detected in HGD and in cancers; these increased expression levels were closely correlated with high expression of Ki67 and minichromosome maintenance complex component 7 (MCM7) proteins. Moreover, genomic mutations in TP53 gene were detected in 56.67% of the evaluated cases (17/30) using next-generation sequencing, and positive staining was verified in HGD and in cancers. Statistical analysis revealed that cell proliferation closely correlated with the expression of DDR factors, of which TP53BP1 was positively associated with SASP factors and IRF3 was positively correlated with cell proliferation. In addition, an analysis evaluating clinical features demonstrated that STAT6-positive cases showed a longer progression-free survival time than STAT6-negative cases. Our evaluation, conducted using a limited number of specimens, suggests SASP may be prevalent in early gastric neoplastic lesions and could be activated by accelerated cell proliferation-induced DDR. The clinical significance of SASP still needs to be determined.

Brain injury poses a heavy disease burden in the world, resulting in chronic deficits. Therapies for brain injuries have been focused on pharmacologic, small molecule, endocrine and cell-based therapies. Endogenous neural stem cells (eNSCs) are a group of stem cells which can be activated in vivo by damage, neurotrophic factors, physical factor stimulation, and physical exercise. The activated eNSCs can proliferate, migrate and differentiate into neuron, oligodendrocyte and astrocyte, and play an important role in brain injury repair and neural plasticity. The roles of eNSCs in the repair of brain injury include but are not limited to ameliorating cognitive function, improving learning and memory function, and promoting functional gait behaviors. The activation and mobilization of eNSCs is important to the repair of injured brain. In this review we describe the current knowledge of the common character of brain injury, the roles and mechanism of eNSCs in brain injury. And then we discuss the current mobilization strategy of eNSCs following brain injury. We hope that a comprehensive awareness of the roles and mobilization strategy of eNSCs in the repair of cerebral ischemia may help to find some new therapeutic targets and strategy for treatment of stroke.

Arbuscular mycorrhizal fungi (AMF) are considered to be an essential indicator of ecosystem biodiversity and can increase a plant’s ability to withstand arid conditions. Despite the obvious significance of AMF in the root and rhizosphere system, little is known about how the AMF variety varies between the soil and roots of endangered plants and how this varies depending on habitats in dry and semiarid regions. This study aimed to address this research gap by investigating the characteristics and diversity of AMF colonization in Zygophyllaceae. Using Illumina MiSeq high-throughput sequencing, indigenous AMF in the roots and rhizosphere soil of three endangered plants (Tetraena mongolica, Sarcozygium xanthoxylon, and Nitraria tangutorum Bobr) were investigated. The three threatened plants had different AMF populations in their root and rhizosphere soils, according to a hierarchical clustering analysis. AMF communities in rhizosphere soil were more sensitive to LDA than root AMF communities based on linear discriminant analysis effect size (LEfSe). Glomus, Septoglomus, and Rhizophagus were seen to function as dominant fungi as the soil and root AMF populations carried out their various tasks in the soil and roots as a cohesive collective. Distance-based redundancy analysis (db-RDA) showed that pH, total phosphorus, and accessible potassium were closely associated with AMF communities. The pH of the soil appears to be an important factor in determining AMF community stability. These findings can serve as a guide for the use of AM fungus in the rehabilitation of agricultural land in arid regions. In summary, our work contributed new knowledge for the scientific preservation of these endangered plant species and for the further investigation of the symbiotic link between AMF and endangered plant species.

Split Hopkinson pressure bar (SHPB) tests are usually used to determine the dynamic mechanical strength of basalt-fiber-reinforced concrete (BFRC), but this test method is time-consuming and expensive. This paper makes predictions about the dynamic mechanical strength of BFRC by employing machine learning (ML) algorithms and feature sets drawn from experimental data from prior works. However, there is still the problem of improving the accuracy of the dynamic mechanical strength prediction by the BFRC, which remains a challenge. Using stacking ensemble learning and genetic algorithms (GA) to optimize parameters, this study proposes a prediction method that combines these two techniques for obtaining accurate predictions. This method is composed of three parts: (1) the training uses multiple base learners, and the algorithms employed by the learners include extreme gradient boosting (XGBoost), gradient boosting (GB), random forest (RF), and support vector regression (SVR); (2) multi-base learners are combined using a stacking strategy to obtain the final prediction; and (3) using GA, the parameters are optimized in the prediction model. An experiment was conducted to compare the proposed approach with popular techniques for machine learning. In the study, the stacking ensemble algorithm integrated the base learner prediction results to improve the model’s performance and the GA further improved prediction accuracy. As a result of the application of the method, the dynamic mechanical strength of BFRC can be predicted with high accuracy. A SHAP analysis was also conducted using the stacking model to determine how important the contributing properties are and the sensitivity of the stacking model. Based on the results of this study, it was found that in the SHPB test, the strain rate had the most significant influence on the DIF, followed by the specimen diameter and the compressive strength.

Summary Root‐associated microbiomes play significant roles in plant productivity, health and ecological services. However, our current understanding of the microbial assemblages in the rhizosphere and endosphere of herbage is still limited. To gain insights into these microbial assemblages, Illumina MiSeq high‐throughput sequencing was performed to investigate the characteristics of microbial communities of an herbage, Leymus chinensis. Hierarchical clustering analysis and principal coordinate analysis (PCoA) results showed that microbial communities of the rhizosphere and endosphere samples were clearly distinguished. Rhizosphere soil communities showed a greater sensitivity than root endosphere communities using linear discriminant analysis (LDA) effect size (LEfSe). Rhizosphere and endosphere communities performed their respective functions in the soil as a cohesive collective, and Rhizobiales were observed to function as generalists. Redundancy analysis (RDA) and variance partitioning analysis (VPA) results revealed that the contribution of the interaction between soil physicochemical parameters and soil enzymes was greater than their individual contributions. In summary, this study is the first to elucidate the microbial diversity and community structure of L. chinensis and compare the diversity and composition between rhizospheric and endosphere microbiomes.

The Xe ^44+ 2.7203 Å line, which has been proposed as one of the diagnostic lines for the x-ray imaging crystal spectrometer on ITER, is observed on the EAST tokamak together with its several satellite lines. The observations are made under high electron temperature ( T _e ) conditions (core T _e > 5 keV). Most of the observed xenon lines are identified by comparing the experiment results with the atomic simulation results. The first ion temperature measurements made by the xenon spectra on EAST are also reported in this article. These xenon spectra observations contribute to the justification for using xenon as the diagnostic impurity in x-ray crystal spectrometers in future reactor-scale high-temperature plasmas.

The cGAS (GMP-AMP synthase)-mediated senescence-associated secretory phenotype (SASP) and DNA-induced autophagy (DNA autophagy) have been extensively investigated in recent years. However, cGAS-mediated autophagy has not been elucidated in cancer cells. The described investigation revealed that active DNA autophagy but not SASP activity could be detected in the BT-549 breast cancer cell line with high micronucleus (MN) formation. DNA autophagy was identified as selective autophagy of free genomic DNA in the cytoplasm but not nucleophagy. The process of DNA autophagy in the cytosol could be initiate by cGAS and usually cooperates with SQSTM1-mediated autophagy of ubiquitinated histones. Cytoplasmic DNA, together with nuclear proteins such as histones, could be derived from DNA replication-induced nuclear damage and MN collapse. The inhibition of autophagy through chemical inhibitors as well as the genomic silencing of cGAS or SQSTM1 could suppress the growth and survival of cancer cells, and induced DNA damage could increase the sensitivity to these inhibitors. Furthermore, expanded observations of several other kinds of human cancer cells indicated that high relative DNA autophagy or enhancement of DNA damage could also increase or sensitize these cells to inhibition of DNA autophagy.

The formation of micronuclei (MN) is prevalent in human cancer cells and its role in activating the senescence-associated secretory phenotype (SASP) machinery has been identified recently. However, the role of MN in regulation of SASP signaling still needs to define in practical cancers. Here, we reported that in colorectal cancer cells the expression of NAT10 (N-acetyltransferase 10) could mediate MN formation through DNA replication and NAT10-positive MN could activate SASP by binding to cGAS. The chemical inhibition of NAT10 by Remodelin or genomic depletion could markedly reduce MN formation, SASP activation, and senescence in colorectal cancer cells. Cell stress such as oxidative or hypoxia could upregulate NAT10 and its associated MN formation senescence and expression of SASP factors. Statistical analysis of clinical specimens revealed correlations between NAT10 expression, MN formation, SASP signaling, and the clinicopathological features of colorectal cancer. Our data suggest that NAT10 increasing MN formation and SASP pathway activation, promoting colorectal cancer progression.

Ewing sarcoma (ES) is the second most common malignant bone and soft tissue tumor in children and adolescents. Despite advances in comprehensive treatment, patients with ES metastases still suffer poor outcomes, thus, emphasizing the need for detailed genetic profiles of ES patients to identify suitable molecular biomarkers for improved prognosis and development of effective and targeted therapies. In this study, the next generation sequencing Ion AmpliSeq™ Cancer Hotspot Panel v2 was used to identify cancer-related gene mutations in the tissue samples from 20 ES patients. This platform targeted 207 amplicons of 2800 loci in 50 cancer-related genes. Among the 20 tissue specimens, 62 nonsynonymous hotspot mutations were identified in 26 cancer-related genes, revealing the molecular heterogeneity of ES. Among these, five novel mutations in cancer-related genes (KDR, STK11, MLH1, KRAS, and PTPN11) were detected in ES, and these mutations were confirmed with traditional Sanger sequencing. ES patients with KDR, STK11, and MLH1 mutations had higher Ki-67 proliferation indices than the ES patients lacking such mutations. Notably, more than half of the ES patients harbored one or two possible 'druggable' mutations that have been previously linked to a clinical cancer treatment option. Our results provided the foundation to not only elucidate possible mechanisms involved in ES pathogenesis but also indicated the utility of Ion Torrent sequencing as a sensitive and cost-effective tool to screen key oncogenes and tumor suppressors in order to develop personalized therapy for ES patients.
