Abstract

A study was conducted to examine the binding of Ca+ and Mg2+ with SynCam which was a synthetic hybrid of CaM that could activate all the CaM enzymes. The thermodynamic binding parameters of Ca2+-CaM and Mg2+-CaM complexes were characterized through the use of flow dialysis and microcalorimetry under varying conditions. Results revealed that Ca2+-induced change in CaM dynamics contributed to the latter's capabilities while Mg2+ functioned as allosteric effector.

Abstract

A novel chromosome mapping technique was developed based on a low coverage shotgun approach and directed sequencing steps. The method was utilized for nucleotide sequencing of the 115 kb chromosomal fragment of the Bacillus subtilis genome. Low coverage shotgun approach for the mapping of the 115 kb chromosomal fragment indicated the presence of 21 open reading frames that belong to seven transcription units. The G+C content of the segment is greater than 47% and contains genes that regulate the synthesis of exopolysaccharide.

Abstract

Electrospray ionization mass spectrometry (ESI-MS) studies on the binding of Ca and Mg ions to four calmodulins, (CaM) SynCaM 1, SynCaM 8, and SynCaM 12A and SynCaM 18A, indicate that CaMs can bind four to six Ca ions and two Mg ions on auxiliary sites. This binding takes place between pH 6 and 8. There are three kinds of sites, the first one being of high affinity and the other two of low affinity. The auxilarry sites probably have a regulatory role since both Mg2+ ions and Ca2+ ions do not bind to the same site.

Abstract

The lacA gene encoding beta-galactosidase in Bacillus subtilis and a regulator gene, lacR were analyzed to determine its characterization. The experiment utilized a transposon mutaganesis method. Results revealed that putative operon containing lacA is a negative regulator. On the other hand, lacR effectively encodes a product related to a family of regulators and includes the lactose repressor of Escherichia coli.

Abstract

Chalcone 4 (compound 1) is a small molecule that neutralizes the CXC chemokine CXCL12 and prevents it from acting on the CXCR4 and CXCR7 receptors. To overcome its poor solubility in aqueous buffers, we designed highly soluble analogues of compound 1, phosphate, l-seryl, and sulfate, all inactive by themselves on CXCL12 but when cleaved in vivo into 1, highly active locally at a low dose in a mouse airway hypereosinophilia model.

Abstract

Regulation of cellular responses to external stimuli such as hormones, neurotransmitters, or cytokines is achieved through the control of all steps of the complex cascade starting with synthesis, going through maturation steps, release, distribution, degradation and/or uptake of the signalling molecule interacting with the target protein. One possible way of regulation, referred to as scavenging or neutralization of the ligand, has been increasingly studied, especially for small protein ligands. It shows innovative potential in chemical biology approaches as well as in disease treatment. Neutralization of protein ligands, as for example cytokines or chemokines can lead to the validation of signalling pathways under physiological or pathophysiological conditions, and in certain cases, to the development of therapeutic molecules now used in autoimmune diseases, chronic inflammation and cancer treatment. This review explores the field of ligand neutralization and tries to determine to what extent small chemical molecules could substitute for neutralizing antibodies in therapeutic approaches.

Abstract

Platelets have an essential role in primary haemostasis to arrest bleeding at a site of vessel wall disruption. Insufficient platelet activation or a reduced platelet number give rise to bleeding syndromes. On the other hand, high platelet activation contributes to thrombosis, which often starts with platelet deposition on a damaged atherosclerotic plaque. The formation of vaso-occlusive platelet thrombi or aggregates, usually with repeated embolization, is a major cause of arterial thrombosis, as occurring in the heart and brains, and resulting in myocardial infarction and so-called cerebrovascular accidents (stroke). Although there is little doubt that coronary and cerebral artery diseases are multifactorial disorders, where also abnormal coagulation, fibrinolysis, vessel wall function and blood flow dynamics can play a role, intervention studies show that in particular antiplatelet drugs provide a risk reduction of one out of every four cases.keywordsplatelet activationhuman plateletplatelet adhesioncalcium responsecollagen receptorthese keywords were added by machine and not by the authors. Abstract

Ditopic dynamic combinatorial. libraries were generated and screened toward inhibition of the bifunctional enzyme HPr kinase/phosphatase from Bacillus subtilis. The libraries were composed of all possible combinations resulting from the dynamic interconversion of 16 hydrazides and five monoaldehyde or dialdehyde building blocks, resulting in libraries containing up to 440 different constituents. Of all possible acyl hydrazones formed, active compounds containing two terminal cationic heterocyclic recognition groups separated by a spacer of appropriate structure could be rapidly identified using a dynamic deconvolution procedure. Thus, parallel testing of sublibraries where one specific component was excluded basically revealed all the essential components. A potent ditopic inhibitor, based on 2-amino-benzimidazole, was identified from the process., QC 20100525
