This work investigates the effect of Ru on the precipitation of the topologically close-packed (TCP) phase from the γ-γʹ microstructural degradation analysis. The addition of Ru is found to delay the precipitation time of the TCP phase from 100 h to 1000 h after thermal exposure at 950 °C. The effect of Ru on the γ-γʹ microstructural degradation is also investigated. Adding Ru slightly decreases the coarsening rate of the γʹ size and promotes the reduction of the volume fraction of γʹ phase. The reduction of the volume fraction of γʹ phase causes the ''inverse partitioning'' effect and increases the solution limit of the TCP forming element in the matrix. These help inhibit the precipitation of the TCP phases in Ni-based single crystal superalloys during thermal exposure.

This work investigates the effect of microstructural degradation on the overtemperature rupture performance in Ru-containing Ni-based single crystal superalloys. Unlike the initial samples, the rupture lives in the degraded sample are largely lost, and adding Ru in the degraded sample increases the rupture life at 1120 °C/100 MPa and 1180 °C/100 MPa. Based on the difference in the microstructure and microstructural evolution in the initial and degraded samples. Their influence on the rupture performance can be derived. The enlarged width of the γ matrix is the major reason for the reduction of the rupture life and increase in the strain for the degraded sample compared to the initial samples. The existence of the dislocation network and the inhibition of the precipitation of the TCP phase are the major reasons for the lower rupture life reduction in the 5Ru degraded sample than in the 3Ru degraded sample. The mobility of the dislocation increases with the temperature, weakening the benefit effect caused by Ru.

Abstract Background Progressive peritoneal fibrosis is a worldwide public health concern impacting patients undergoing peritoneal dialysis (PD), yet there is no effective treatment. Our previous study revealed that a novel compound, micheliolide (MCL) inhibited peritoneal fibrosis in mice. However, its mechanism remains unclear. Brahma-related gene 1 (BRG1) is a key contributor to organ fibrosis, but its potential function in PD-related peritoneal fibrosis and the relationship between MCL and BRG1 remain unknown. Methods The effects of MCL on BRG1-induced fibrotic responses and TGF-β1-Smads pathway were examined in a mouse PD model and in vitro peritoneal mesothelial cells. To investigate the targeting mechanism of MCL on BRG1, coimmunoprecipitation, MCL-biotin pulldown, molecular docking and cellular thermal shift assay were performed. Results BRG1 was markedly elevated in a mouse PD model and in peritoneal mesothelial cells cultured in TGF-β1 or PD fluid condition. BRG1 overexpression in vitro augmented fibrotic responses and promoted TGF-β1-increased-phosphorylation of Smad2 and Smad3. Meanwhile, knockdown of BRG1 diminished TGF-β1-induced fibrotic responses and blocked TGF-β1-Smad2/3 pathway. MCL ameliorated BRG1 overexpression-induced peritoneal fibrosis and impeded TGF-β1-Smad2/3 signaling pathway both in a mouse PD model and in vitro. Mechanically, MCL impeded BRG1 from recognizing and attaching to histone H3 lysine 14 acetylation by binding to the asparagine (N1540) of BRG1, in thus restraining fibrotic responses and TGF-β1-Smad2/3 signaling pathway. After the mutation of N1540 to alanine (N1540A), MCL was unable to bind to BRG1 and thus, unsuccessful in suppressing BRG1-induced fibrotic responses and TGF-β1-Smad2/3 signaling pathway. Conclusion Our research indicates that BRG1 may be a crucial mediator in peritoneal fibrosis and MCL targeting N1540 residue of BRG1 may be a novel therapeutic strategy to combat PD-related peritoneal fibrosis.

Ru is crucial in improving creep properties in Nickel-based single-crystal superalloys. The effect of Ru content on the creep behavior of nickel-based single-crystal superalloy was studied. Ru exhibits unprecedented mechanical properties, increasing the creep life at 1120 °C but decreasing it at 1180 °C. This is due to the competition in the stress concentration formed between the topologically close-packed phases and the creep cavity. They are both related to the dissociation of the γ′ phase. The effect of oxidation caused by the addition of Ru is lower than these two factors. This work helps to recognize the comprehensive effect of Ru and can provide guidance for alloy design in the future.

Hydrogen-fueled and hydrogen-hybridized aircraft engines are a new trend in the aviation industry for environmental reasons. Single crystalline Ni-based superalloys are the most commonly used engine materials and their hydrogen embrittlement properties need urgent investigation. In this study, the hydrogen embrittlement behavior and underlying fracture mechanism of a second-generation Ni-based single crystal superalloy with electrochemical hydrogen pre-charge were investigated. The superalloy showed tremendous susceptibility to hydrogen embrittlement with reduced strength and ductility. A large number of micropores and cracks on the fracture surface are found in hydrogen-charged specimens, leading to embrittlement and ultimate cracking. More dislocations, stacking faults and DSBs are observed in specimens with hydrogen uptake. Hydrogen-induced micropores first form at the γ/γ′ interface and then propagate into the γ′ phase, leading to cracking, which was analyzed using in situ environmental studies with a transmission electron microscope. Hydrogen reduces the cohesive strength between the γ- and γ′-phase and accelerates crack propagation along the voids. Hydrogen embrittlement fracture in Ni-based single crystal superalloys is due to synergistic hydrogen-enhanced local plasticity, strain-induced vacancies and decohesion in the hydrogen-induced cracking process.

In this work, the effect of directional solidification methods on solid solution windows in two Ni-based single-crystal superalloys is investigated. The sample prepared using the liquid metal cooling (LMC) method was found to contain smaller dendrite spacing, lower dendrite segregation and eutectic, and a smaller size of γ′ phase than that of the high-speed rapid solidification (HRS) method. The alloy-2 contained higher Cr, Co and lowered Re exhibited similar dendrite spacing and volume fraction of eutectic, and the lower dendrite segregation and smaller size of γ′ phase than that of the other alloy-1. The solution treatment window is broadened by adopting the LMC method or changing the composition to alloy-2. This is mainly realized by decreasing the lower limit temperature, and the upper limit temperature remains unchanged. According to the eutectic only contains the melt temperature, a similar eutectic is responsible for the unchanged upper limit temperature. At this state, the lower bound of the temperature is the dissolution of the γʹ phase in the dendrite core. The smaller size of γ′ phase and the elements' partition ratio are responsible for the change in the solid solution window.

The aim of this study was to determine the status of weed Solanum nigrum L. as a transitional host for Meloidogyne enterolobii and its effect on the population base of the nematodes in the next season. The nematode species infecting S. nigrum L. in a fallow field was identified by morphological identification and molecular diagnosis, and parasitic characteristics of the nematodes in S. nigrum L., including development of the nematode in S. nigrum L., the histopathological response of S. nigrum L. to M. enterolobii, and the host suitability of S. nigrum L., were studied. The M. enterolobii soil population density was evaluated before and after S. nigrum L. planting. Species identification revealed that it was M. enterolobii infection. Developmental observation indicated that juveniles of M. enterolobii developed fast in S. nigrum L., establishing feeding sites by 5 days after inoculation (DAI) and forming obvious egg masses on the root at 25 DAI. Histopathological observation showed the typical susceptible response of S. nigrum L., including giant cells with thick cell walls, uniformly dense cytoplasm, and less vacuolation, mainly inside the vascular cylinder. Host suitability assays suggested that S. nigrum L. is a good host for M. enterolobii with an average reproduction factor (RF) of 48.04 ± 14.71. Population densities assays revealed that S. nigrum L. increased the population density of M. enterolobii for two consecutive years from 0.48 ± 0.25 and 0.53 ± 0.31 J2/cm3 to 1.33 ± 0.16 and 1.56 ± 0.43 J2/cm3 of soil. These results indicated that M. enterolobii could reproduce well by infecting S. nigrum L. during the fallow season, and it increased the population base of M. enterolobii to the next season during vegetable production, which suggested a novel direction for the control of root-knot nematodes by controlling weeds as transitional hosts of M. enterolobii in the fallow season.

It is challenging to study how topologically close-packed phases (TCPs) transform between one phase to another. Here the authors use atomic-resolved tools to look at the transformation between μ and P phases, revealing an intrinsic link between seemingly unrelated TCP configurations.

Oxygen-affected cracking commonly presents on thin Ni-based single crystal superalloy components serving in high temperature and oxidizing environments. This study uses a newly developed in-situ thermal-stress environmental transmission electron microscope to investigate the oxidation and fracture behaviors of Ni-based single crystal superalloy at 650°C under stress. The in-situ oxidation was found to change the tensile fracture mode from the close-packed {111} planes of plastic fracture to $ \{001\} $ planes adjacent to γ/γ′ interfaces of brittle fracture. The microanalysis also revealed that the γ′ cuboids, γ phase, and γ/γ interface exhibit different oxidation behavior, thus underscoring the thickness debit effect.

The cucurbit vegetable chieh-qua (Benincasa hispida var. chieh-qua How) is an important crop in South China and southeast Asian countries. Viral diseases cause substantial loss of chieh-qua yield. To identify the viruses that affect chieh-qua in China, ribosomal RNA-depleted total RNA sequencing was performed using chieh-qua leaf samples with typical viral symptoms. The virome of chieh-qua comprises four known viruses (melon yellow spot virus (MYSV), cucurbit chlorotic yellows virus (CCYV), papaya ringspot virus (PRSV) and watermelon silver mottle virus (WSMoV) and two novel viruses: cucurbit chlorotic virus (CuCV) in the genus Crinivirus and chieh-qua endornavirus (CqEV) in the genus Alphaendornavirus. The complete genomes of the two novel viruses in chieh-qua and three other isolates of CuCV in pumpkin, watermelon and cucumber were determined and the recombination signals of pumpkin and watermelon isolates of CuCV were detected. A reverse transcriptase PCR indicated that the dominant viruses of chieh-qua in Hainan are MYSV (66.67%) and CCYV (55.56%), followed by CuCV (27.41%), WSMoV (7.41%), cucumber mosaic virus (8.15%), zucchini yellow mosaic virus (6.67%), PRSV (6.67%) and CqEV (35.56%). Our findings support diagnostic and prevalence studies of viruses infecting chieh-qua in China, enabling sustainable control strategies for cucurbit viruses worldwide.

The Ni-based superalloy is used as the turbine blade, which is subject to the coupling effect of temperature and super-gravity during service. As the Ni-based superalloys are difficult to become homogenous after using the solid solution heat treatment, a study on morphology and composition distribution of Ni-base superalloys with segregation during microstructural degradation is necessary. This study investigates the microstructure of the ex-service turbine blade and cast samples subjected to the high-temperature centrifugal test. The difference in the size and shape factor of the γ′ phase decreased with the stress caused by the super-gravity condition, indicating a higher magnitude of homogenization degree. The higher stress will also promote the merge of the sub-grain boundaries, leading to a lower density and higher orientational deviation of the sub-grain boundaries.

Abstract Background Albumin-globulin ratio (AGR), a variable based on serum albumin and non-albumin proteins, has been demonstrated as a predictor of mortality in patients with malignant neoplasm. The aim of this study was to evaluate the prognostic value of AGR on peritoneal dialysis (PD) patients. Methods We retrospectively analyzed 602 incident PD patients from January 1st, 2008, to December 31st, 2017, at our center and followed them until December 31st, 2018. Kaplan-Meier curves and multivariate Cox regression models were applied to analyze the association between AGR and all-cause of mortality and cardiovascular mortality. Results The median follow-up time was 32.17 (interquartile range = 32.80) months. During follow-up, 131 (21.8%) patients died, including 57 patients (43.5%) who died due to cardiovascular diseases. Meloidogyne enterolobii, a highly pathogenic root-knot nematode species, causes serious damage to agricultural production worldwide. Collagen is an important part of the nematode epidermis, which is crucial for nematode shape maintenance, motility, and reproduction. In this study, we report that a novel collagen gene, Me-col-1, from the highly pathogenic root-knot nematode species Meloidogyne enterolobi was required for the egg formation of this pathogen. Me-col-1 encodes a protein with the size of 35 kDa, which is closely related to collagen found in other nematodes. Real-time PCR assays showed that the expression of Me-col-1 was highest in eggs and lowest in pre-parasitic second-stage juveniles (preJ2). Interestingly, knockdown of Me-col-1 did not compromise the survival rate of preJ2 but significantly reduced the egg production and consequentially caused 35.79% lower multiplication rate (Pf/Pi) compared with control. Our study provides valuable information for better understanding the function of collagen genes in the nematode life cycle, which can be used in the development of effective approaches for nematode control.

Spodoptera frugiperda is a highly destructive and polyphagous pest that causes severe damage to various crops, especially maize. The wide use of chemical insecticides to control S. frugiperda results in resistance against commonly used chemicals and resistant mutations will expand in populations accompanied by a spread to vulnerable areas. Consequently, more effective and friendly strategies must be explored to minimize losses caused by S. frugiperda. Entomopathogenic nematodes (EPN) are good candidates for the biological control of different species of insect pests, including S. frugiperda. In the current study, the infective capabilities of the EPN species HbSD, belonging to Hetrerorhabditis bacteriophora, were evaluated against S. frugiperda under laboratory, greenhouse and field conditions. In laboratory assays, HbSD was highly virulent against 3rd/5th instar larvae, which was related to HbSD concentration and exposure durations. In greenhouse assays, spraying aqueous HbSD also showed good performance in killing larvae on maize leaves. However, the virulence of HbSD decreased in field trials where many adverse factors affecting survival and efficacy were encountered by HbSD. Overall, our study provides an alternative EPN for the biological control of S. frugiperda with the potential to be developed as a sustainable option for efficient pest management.

Early detection of pathogens before the planting season is valuable to forecast disease occurrence. Therefore, rapid and reliable diagnostic approaches are urgently needed, especially for one of the most aggressive root knot nematodes, Meloidogyne enterolobii. In this study, we developed a novel primer–TaqMan probe set aimed at M. enterolobii. The primer–probe set was successfully applied in the identification and quantification of M. enterolobii via qPCR technology. It was also suitable for improved PCR technology, known as ddPCR analyses, and this work presents the first application of this technology for plant parasitic nematodes. Compared with qPCR, ddPCR exhibited better performance with regard to analytical sensitivity, which can provide a more accurate detection of M. enterolobii concealed in field soil. In addition, we generated standard curves to calculate the number of eggs in soil using the qPCR and ddPCR platforms. Hopefully, the results herein will be helpful for forecasting disease severity of M. enterolobii infection and adopting effective management strategies.

The genus Meloidogyne is one of the most economically damaging plant parasitic nematodes on a worldwide basis. To date, only the dragon fruits in Brazil were reported infected by M. javanica or M. incognita in previous surveys. From 2019–2021, we conducted our investigation in 62 orchards from 10 counties of Hainan, which are the main producing areas of dragon fruits in China. The collected roots were visually scored by gall index to evaluate the distribution and severity of diseases caused by Meloidogyne. Then, the isolated nematodes were further confirmed by a combination of morphological and molecular analysis. This survey revealed that the root-knot nematodes were widely distributed in red dragon fruit planting areas in Hainan and caused severe symptoms including stunting, wilting, and yellowing of the stems. The nematode populations were identified as M. enterolobii and M. javanica, among which, M. enterolobii accounted for a larger proportion, 93.9%. To our knowledge, this is the first report of M. enterolobii causing diseases on red dragon fruit. The results obtained from this survey will provide an insight into the presence of root-knot nematodes in dragon fruit plants. Also, these results will raise attention for developing effective management strategies to prevent further spread of root-knot nematodes on dragon fruit in Hainan.

It is known that Ni-based superalloys experience microstructural rafting during high temperature straining. This paper presents a comparative study of the different rafting models by means of finite element modeling and experimentation investigation. It was found that uniaxial elastic loading does not alter the misfit or misfit isotropy of the structure, but causes repartitioning of the misfit into elastic strains in the γ and γ' phases. This causes an elastic strain energy anisotropy and hydrostatic stress anisotropy among the vertical and horizontal γ phase channels. Both anisotropies predict a raft structure that is contrary to the experimental observation. Through a carefully designed pre-treatment at 750 °C and under a uniaxial stress of 750 MPa, an anisotropic dislocation structure was created on the {001} γ'/γ interfaces without altering the cuboidal morphology of the γ' phase. This allows the evaluation of the influence of dislocations on the rafting behavior of the alloy. It was found that rafting occurred in the pre-treated samples containing anisotropic dislocation structures during thermal exposure without applied stress, instead of isotropic corsening, and that the raft structures conform to the expectations base don the anistropic dislocation strucutres. This demonstrates that an anisotropic dislocation structure on the {001} γ'/γ interfaces is a direct cause of rafting. This is attributted to the fact that dislocations on γ'/γ interfaces help to relax the local misfit strains, causing some to expand at the expense of others. At the same time, the formation of the anisotropic dislocation structure is a direct result of the anisotropy of elastic strains induced by the applied bia stess in during the pre-treatment. These explain the complex interplay of the elastic and plastic models reported the literature.

Background. Neutrophil to lymphocyte ratio (NLR) is a new inflammatory marker; the relationship between NLR and adverse cardiovascular (CV) prognosis has been gradually emphasized in the general population. However, their association in peritoneal dialysis (PD) patients remains unclear. Methods. From January 1, 2010, to May 31, 2017, a total of 1652 patients were recruited. NLR was categorized in triplicates: NLR≤2.74, 2.743.96. Kaplan-Meier cumulative incidence curve and multivariable COX regression analysis were used to determine the relationship between NLR and the incidence of adverse CV outcome, while a competitive risk model was applied to assess the effects of other outcomes on adverse CV prognosis. Besides, forest plot was investigated to analyze the adverse CV prognosis in different subgroups. Results. During follow-up, 213 new-onset CV events and 153 CV disease (CVD) deaths were recorded. Multivariable COX regression models showed that the highest tertile of NLR level was associated with increased risk of CV events (HR=1.39, 95%CI=1.01‐1.93, P=0.046) and CVD mortality (HR=1.81, 95%CI=1.22‐2.69, P=0.003), while compared to the lowest tertile. Chronic kidney disease is a common disease closely related to renal tubular inflammation and oxidative stress, and no effective treatment is available. Activation of the nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasome is an important factor in renal inflammation, but the mechanism remains unclear. Micheliolide (MCL), which is derived from parthenolide, is a new compound with antioxidative and anti-inflammatory effects and has multiple roles in tumors and inflammatory diseases. In this study, we investigated the effect of MCL on lipopolysaccharide- (LPS-) induced inflammation in renal tubular cells and the related mechanism. We found that MCL significantly suppressed the LPS-induced NF-κB signaling and inflammatory expression of cytokines, such as tumor necrosis factor-α and monocyte chemoattractant protein-1 in a rat renal proximal tubular cell line (NRK-52E). MCL also prevented LPS- and adenosine triphosphate-induced NLRP3 inflammasome activation in vitro, as evidenced by the inhibition of NLRP3 expression, caspase-1 cleavage, and interleukin-1β and interleukin-18 maturation and secretion. Additionally, MCL inhibited the reduction of mitochondrial membrane potential and decreases the release of reactive oxygen species (ROS). Moreover, MCL can prevent NLRP3 inflammasome activation induced by rotenone, a well-known mitochondrial ROS (mROS) agonist, indicating that the mechanism of MCL’s anti-inflammatory effect may be closely related to the mROS. In conclusion, our study indicates that MCL can inhibit LPS-induced renal inflammation through suppressing the mROS/NF-κB/NLRP3 axis in tubular epithelial cells.

Prior studies have shown that the levels of some platelet (PLT) indices were associated with mortality in patients undergoing hemodialysis. We aimed to investigate whether the changes in PLT indices associated with mortality in patients on peritoneal dialysis (PD). A single-center, retrospective observational cohort study was conducted in incident PD patients from 1 January 2006 to 31 December 2012, and followed up until 31 December 2014. Cox proportional hazard models were used to examine the relationships between the levels of PLT indices including PLT, plateletcrit (PCT), mean platelet volume (MPV), platelet distribution width (PDW), platelet large cell ratio (PLCR), and mortality. Of 1324 patients, 276 (20.8%) died during follow-up (median, 37; IQR, 3–107.4 months), among which 134 were due to cardiovascular diseases (CVD). The highest tertile of PLT levels at baseline was associated with increased risk for cardiovascular mortality after adjustment for demographic, clinical characteristics, and laboratory variables (hazard ratio [HR]:1.93; 95% confidence interval [CI]: 1.16–3.20). The similar treads were also observed in the middle and the highest tertile of the PCT level (HR: 1.68, 95%CI: 1.00–2.81 and HR: 1.89, 95%CI: 1.14–3.14, respectively). In addition, the highest tertile of PCT was associated with increased all-cause mortality (HR: 1.41, 95%CI: 1.01–1.96). However, none of the associations in MPV, PDW, and PLCR analyses reached statistical significance (HR: 0.71, 95%CI: 0.43–1.16; HR: 0.72, 95%CI: 0.45–1.18 and HR: 0.74, 95%CI: 0.46–1.19, respectively). These results suggest that higher PLT and PCT may be associated with higher risk for cardiovascular mortality in incident PD patients. Additional studies are needed to investigate whether correction of these two PLT indices reduces the risk.

This study investigated a novel hypothesis on the influences of residual or pre-dislocation structures on creep life of Ni-based single crystal superalloys. Different types of anisotropic dislocation structures were created by pre-straining the samples at a relatively low temperature under different constant bias load conditions. After such pre-straining treatment, the samples were subjected to creep testing at higher temperatures under uniaxial tension. It was found that the pre-straining treatment was effective in creating different anisotropic dislocation structures without affecting the γ′ phase cuboid morphology and that the samples containing different anisotropic dislocation structures have all significantly reduced creep lives. The difference in the anisotropy of the dislocation structures has little influence on the reduction of the creep life or the rafting morphology of the γ′ phase cuboids. By using a Re-free alloy, the influence of the TCP phase precipitation on creep life is also clarified. These observations suggest that interruption to high temperature straining is the main contributor to the reduced creep lives. This finding raises a serious concern on the validity, or reliability, of creep life assessment based on continuous laboratory creep tests for real life applications, where the services are almost always interrupted into intermittent sessions. In this work, micrometer copper-zinc alloy particles-reinforced particleboard wood flour/poly (lactic acid) (mCu-Zn/PWF/PLA) wood plastic composites with high gloss and antibacterial properties for 3D printing were prepared by a melt blending process. The structure and properties of the composites with different contents of mCu-Zn were analyzed by means of mechanical testing, dynamic mechanical analysis, thermogravimetric analysis, differential scanning calorimetry, X-ray diffraction, scanning electron microscopy, and antibacterial testing. The results showed that the mechanical properties, thermal stability, and antibacterial performance of the composites were significantly improved, as mCu-Zn was added into the wood plastic composites. When adding 2 wt.% mCu-Zn, the flexural strength of mCu-Zn/PWF/PLA composites (with 5 wt.% of particleboard wood flour) (PWF) increased by 47.1% compared with pure poly (lactic acid) (PLA), and 18.9% compared with PWF/PLA wood plastic composites. The surface gloss was increased by 1142.6% compared with PWF/PLA wood plastic composites. Furthermore, the inhibition rates of mCu-Zn/PWF/PLA composites against Escherichia coli reached 90.43%. Therefore, this novel high gloss and antibacterial wood plastic composites for fused deposition modeling (FDM) 3D printing have potential applications in personalized and classic furniture, art, toys, etc.

Twinning structures and their interfacial segregation play a key role in strengthening of magnesium alloys. Micro-steps are frequently existed in the incoherent twin boundaries, while the effect of them on interface and interfacial segregation is still not clear. In this work, we performed an atomic-scale microstructure analysis using high-angle annular dark field scanning transmission electron microscopy (HAADF-STEM) to explore the effect of micro-steps on twin and its interfacial segregation in Mg-Ag alloy. Diffraction pattern of the incoherent {10 1 ¯ 1} twin shows that the misorientation has a slight tilt of 5° from its theoretical angle of 125° due to the accumulated effects of the micro-steps and their misfit dislocations in twin boundaries. Most of the micro-steps in {10 1 ¯ 1} twin boundary are in the height of 2 d ( 10 1 ¯ 1 ) and 4 d ( 10 1 ¯ 1 ) , respectively, and both of them have two types according to whether there are dislocations on the micro-steps. The twin boundary is interrupted by many micro-steps, which leads to a step-line distributed interfacial segregation. Moreover, the Ag tends to segregate to dislocation cores, which results in the interruption of interfacial segregation at the micro-steps with dislocations.

Pectate lyases are known to play a key role in pectin degradation by catalyzing the random cleavage of internal polymer linkages (endo-pectinases). In this paper, four novel cDNAs, designated Hg-pel-3, Hg-pel-4, Hg-pel-6 and Hg-pel-7, that encode pectate lyases were cloned and characterized from the soybean cyst nematode, Heterodera glycines. The predicted protein sequences of HG-PEL-3, HG-PEL-4 and HG-PEL-6 differed significantly in both their amino acid sequences and their genomic structures from other pectate lyases of H. glycines (HG-PEL-1, HG-PEL-2 and HG-PEL-7). A phylogenetic study revealed that the pectate lyase proteins of H. glycines are clustered into distinct clades and have distinct numbers and positioning of introns, which suggests that the pectate lyase genes of H. glycines may have evolved from at least two ancestral genes. A Southern blot analysis revealed that multiple Hg-pel-6-like genes were present in the H. glycines genome. In situ hybridization showed that four novel pectate lyases (Hg-pel-3, Hg-pel-4, Hg-pel-6 and Hg-pel-7) were actively transcribed in the subventral esophageal gland cells. A semi-quantitative RT-PCR assay supported the finding that the expression of these genes was strong in the egg, pre-parasitic second-stage juvenile (J2) and early parasitic J2 stages and that it declined in further developmental stages of the nematode. This expression pattern suggests that these proteins play a role in the migratory phase of the nematode life cycle. Knocking down Hg-pel-6 using in vitro RNA interference resulted in a 46.9% reduction of the number of nematodes that invaded the plants and a 61.5% suppression of the development of H. glycines females within roots compared to the GFP-dsRNA control. Plant host-derived RNAi induced the silencing of the Hg-pel-6gene, which significantly reduced the nematode infection levels at 7 Days post inoculation (dpi). Similarly, this procedure reduced the number of female adults at 40 dpi, which suggests the important roles of this gene in the early stages of parasitism. Our combined data suggest that two types of pectate lyases are present in the H. glycines genome and may have different roles during infection.

The soybean cyst nematode, Heterodera glycines, is an important pest of soybeans. Although resistance is available against this nematode, selection for virulent races can occur, allowing the nematode to overcome the resistance of cultivars. There are abundant field populations, however, little is known about their genetic diversity. In order to elucidate the differences between races, we investigated the transcriptional diversity within race 3 and race 4 inbred lines during their compatible interactions with the soybean host Zhonghuang 13. Six different race-enriched cDNA libraries were constructed with limited nematode samples collected from the three sedentary stages, parasitic J2, J3 and J4 female, respectively. Among 689 putative race-enriched genes isolated from the six libraries with functional annotations, 92 were validated by quantitative RT-PCR (qRT-PCR), including eight putative effector encoding genes. Further race-enriched genes were validated within race 3 and race 4 during development in soybean roots. Gene Ontology (GO) analysis of all the race-enriched genes at J3 and J4 female stages showed that most of them functioned in metabolic processes. Relative transcript level analysis of 13 selected race-enriched genes at four developmental stages showed that the differences in their expression abundance took place at either one or more developmental stages. This is the first investigation into the transcript diversity of H. glycines races throughout their sedentary stages, increasing the understanding of the genetic diversity of H. glycines.
