Abstract Pyroptosis, an inflammatory regulated cell death (RCD) mechanism, is characterized by cellular swelling, membrane rupture, and subsequent discharge of cellular contents, exerting robust proinflammatory effects. Recent studies have significantly advanced our understanding of pyroptosis, revealing that it can be triggered through inflammasome- and caspase-independent pathways, and interacts intricately with other RCD pathways (e.g., pyroptosis, necroptosis, ferroptosis, and cuproptosis). The pathogenesis of pulmonary fibrosis (PF), including idiopathic pulmonary fibrosis (IPF) and other interstitial lung diseases, involves a multifaceted interplay of factors such as pathogen infections, environmental pollutants, genetic variations, and immune dysfunction. This chronic and progressive interstitial lung disease is characterized by persistent inflammation, extracellular matrix (ECM) accumulation, and fibrotic alveolar wall thickening, which potentially contribute to deteriorated lung function. Despite recent advances in understanding pyroptosis, the mechanisms by which it regulates PF are not entirely elucidated, and effective strategies to improve clinical outcomes remain unclear. This review strives to deliver a comprehensive overview of the biological functions and molecular mechanisms of pyroptosis, exploring its roles in the pathogenesis of PF. Furthermore, it examines potential biomarkers and therapeutic agents for anti-fibrotic treatments.

ABSTRACT: The objective of this study was to examine the aroma profiles of 12 Gouda cheeses sold in China and to determine which aromas were preferred by young Chinese consumers (n = 110). The consumers selected 11 descriptors of the aromas of the Gouda cheeses in a check-all-that-apply questionnaire. These 11 descriptors were used by a panel of experts for sensory analysis to perform a quantitative descriptive analysis of the cheeses. A principal component analysis of the data from the quantitative descriptive analysis revealed that the characteristic aromas of young Gouda cheeses, medium-aged Gouda cheeses, and aged Gouda cheeses were “milky” and “whey”; “creamy” and “sour”; and “rancid” and “nutty,” respectively. The results of a penalty analysis combined with the check-all-that-apply results and the preference scores showed that the 3 groups of young Chinese consumers (those who often ate cheese, occasionally ate cheese, and never ate cheese) preferred the Gouda cheeses with “milky” or “creamy” aromas and did not enjoy those with “sour” or “rancid” aromas. Occasional cheese eaters comprised the majority of the young Chinese consumers, and they were more tolerant of the Gouda cheeses with “whey” and “sulfury” aromas than those who often or never ate cheese. In addition, we identified a positive correlation between the consumers' preferences for the aromas of the Gouda cheeses and their willingness to pay for them. Overall, the results of this study should help promote the development of Gouda cheeses and associated products that meet the preferences of young Chinese consumers.

Abstract Human metapneumovirus (HMPV) is a newly identified pathogen causing acute respiratory tract infections in young infants worldwide. Since the initial document of HMPV infection in China in 2003, Chinese scientists have made lots of efforts to prevent and control this disease, including developing diagnosis methods, vaccines and antiviral agents against HMPV, as well as conducting epidemiological investigations. However, effective vaccines or special antiviral agents against HMPV are currently not approved, thus developing early diagnosis methods and knowing its epidemiological characteristics will be beneficial for HMPV control. Here, we summarized current research focused on the epidemiological characteristics of HMPV in China and its available detection methods, which will be beneficial to increase the public awareness and disease control in the future.

As two major pests of cowpea in South China, bean flower thrips [Megalurothrips usitatus (Bagnall)] and flower thrips [Frankliniella intonsa (Trybom)] always occur on the same plant. In this study, the two-sex life table parameters of these two species were investigated on three bean pods: cowpea (Vigna unguiculata L. var. Chunqiu Wujiadou), snap bean (Phaseolus vulgaris L. var. Yonglong 3), and green bean (Phaseolus vulgaris L. var. Linghangzhe) using standard cut bean protocols. Longevity of female adult M. usitatus showed no significant difference on cowpea and snap bean, but was shortest on green bean, with the highest fecundity on snap bean, followed by cowpea and green bean. Longevity and fecundity of female adult F. intonsa were both highest on cowpea, followed by snap bean and green bean. On snap bean, the population parameters (R0, GRR, T) of M. usitatus were higher than F. intonsa, whereas r and λ were not different, suggesting that M. usitatus were more competitive on snap bean. On cowpea, the population parameters (R0, GRR, r, λ) of F. intonsa were higher than M. usitatus, suggesting a competitive advantage of F. intonsa on cowpea. To better mimic field conditions, we investigated the survival and fecundity of adult M. usitatus under two distinct scenarios: when they could feed directly on cut surfaces of cowpeas (standard lab protocol), and when they could not. Adult survival of M. usitatus is not significantly influenced by whether or not they fed directly on cut surfaces of cowpeas. However, a notable decrease in fecundity was observed in M. usitatus that fed on surface-covered cut pods. This suggested that direct feeding on cut surfaces of cowpea did have an impact on thrips’ fecundity, offering fresh perspectives for future research into the growth and development of thrips in a laboratory. Our study suggests that experiments involving cut surfaces may be misleading, and further investigations are ongoing to address these concerns.

This study aimed to investigate the influence of varying weight ratios of Shuanla and Erjingtiao peppers (10:0, 8:2, 6:4, 5:5, 4:6, 2:8, and 0:10, corresponding to samples PA, PB, PC, PD, PE, PF, and PG, respectively) on the sensory attributes, chromatism, acid values (AVs), peroxide values (POVs), capsaicinoids, and volatile organic compounds (VOCs) of seven chili oil samples. GC–IMS was employed to detect the VOCs of the chili oil samples, which were subsequently analyzed using multivariate statistical methods. The results revealed significant differences in pungency among the samples, with the PA sample exhibiting the strongest pungency. The PG sample demonstrated the highest values for a*, b*, and C*, while the PA sample displayed the highest L* and h*. The AVs of seven samples ranged from 0.490 ± 0.005 to 1.727 ± 0.015 mg/g. The POVs of the chili oil samples, ranging from 0.094 ± 0.000 to 0.127 ± 0.002 g/100 g, were significantly lower than those of extra virgin olive oil, 0.183 ± 0.001 g/100 g. The contents of capsaicinoids ranged from 15.26 ± 0.07 g/kg in the PA sample to 0.38 ± 0.00 g/kg in the PG sample (p < 0.05). Additionally, 56 volatile flavor substances were identified, and 10 key flavor compounds (ROAV ≥ 1) were screened among them. Multivariate data analysis via OPLS-DA indicated that 20 VOCs (VIP > 1) could serve as flavor markers in a clustering heat map to differentiate among the seven chili oil varieties. The findings of this study provide a valuable reference for the promotion of Shuanla and Erjingtiao peppers in chili oil production and the development of specific flavor profiles in chili oil to cater to diverse consumer preferences.

ABSTRACT: Milk fan is an acid-curd cheese with strong national characteristics (a traditional dairy product of the Bai nationality with a shape like a piece of paper) and a long history in Yunnan province, China. In our previous study, we characterized the microbial community diversity of milk fan, but the succession of microorganisms associated with flavor formation in milk fan is still unknown. Therefore, we examined the predominant microorganisms and their correlations with the formation of flavor in the fermentation of sour juice and drying of milk fan by gas chromatography mass spectrometry, high-throughput 16S rDNA sequencing, intergenic spacer sequencing and metatranscriptome analysis. We found that the relative abundances of Lactobacillus and Issatchenkia initially decreased and then increased with time during the fermentation of sour juice. However, the relative abundances of Acetobacter, Leuconostoc, Lactococcus, Geotrichum, and Dipodascus initially increased and then decreased. During the drying step, the relative abundances of Lactobacillus and Issatchenkia continuously increased and became the dominant microorganisms in the milk fan. The metatranscriptomes generated from the milk fan showed that “carbohydrate metabolism,” “translation,” and “signal transduction” were the main metabolic functions of the microbial communities. Rhodotorula and Yarrowia contained more differentially expressed genes than other genera, which indicated they may be associated with the production of the characteristic flavor. Furthermore, a Pearson correlation analysis showed that Lactococcus, Rhodotorula, Candida, Cutaneotrichosporon, and Yarrowia were significantly positively correlated with more aroma-active compounds, mainly ethyl acetate, 2-heptanone, isovaleraldehyde, butyric acid, nonanal, and hexanal. In conclusion, these findings contribute to a better understanding of the flavor production mechanism during the production of milk fan.

The traditionally produced pea paste (PP) suffers from suboptimal flavor and inferior quality. Based on the study of single-strain fermentation, we further selected S. cerevisiae, Z. rouxii, and L. paracasei for PP production by dual-strain fermentation (SL, ZL). By combining intelligent sensory technology, gas chromatography–mass spectrometry (GC-MS), and ultra-high-performance liquid chromatography (UPLC) technology, the aroma and taste characteristics of SL- and ZL-fermented PP were compared. The electronic nose and tongue revealed the differences in the aroma and taste characteristics between the two fermentation methods for fermenting PP. In total, 74 volatile compounds (VOCs) in PP were identified through GC-MS analysis. In contrast, the number of VOCs and the concentrations of alcohols and acids compounds in SL were higher than in ZL. Among the 15 VOCs that were common to both and had significant differences, the concentrations of ethanol, 1-pentanol, and ethyl acetate were higher in SL. For taste characteristics, SL demonstrated significantly higher levels of sweet and bitter amino acids, as well as tartaric acid, compared with ZL. These results elucidate the flavor differences of dual-strain fermented PP, providing a theoretical basis for selecting suitable strains for fermenting PP.

Abstract Background Inflammatory bowel diseases (IBDs) pose significant challenges in terms of treatment non‐response, necessitating the development of novel therapeutic approaches. Although biological medicines that target TNF‐α (tumour necrosis factor‐α) have shown clinical success in some IBD patients, a substantial proportion still fails to respond. Methods We designed bispecific nanobodies (BsNbs) with the ability to simultaneously target human macrophage‐expressed membrane TNF‐α (hmTNF‐α) and IL‐23. Additionally, we fused the constant region of human IgG1 Fc (hIgG1 Fc) to BsNb to create BsNb‐Fc. Our study encompassed in vitro and in vivo characterization of BsNb and BsNb‐Fc. Results BsNb‐Fc exhibited an improved serum half‐life, targeting capability and effector function than BsNb. It's demonstrated that BsNb‐Fc exhibited superior anti‐inflammatory effects compared to the anti‐TNF‐α mAb (infliximab, IFX) combined with anti‐IL‐12/IL‐23p40 mAb (ustekinumab, UST) by Transwell co‐culture assays. Notably, in murine models of acute colitis brought on by 2,4,6‐trinitrobenzene sulfonic acid（TNBS) and dextran sulphate sodium (DSS), BsNb‐Fc effectively alleviated colitis severity. Additionally, BsNb‐Fc outperformed the IFX&UST combination in TNBS‐induced colitis, significantly reducing colon inflammation in mice with colitis produced by TNBS and DSS. Conclusion These findings highlight an enhanced efficacy and improved biostability of BsNb‐Fc, suggesting its potential as a promising therapeutic option for IBD patients with insufficient response to TNF‐α inhibition. Key points A bispecific nanobody (BsNb) was created to target TNF‐α and IL‐23p19, exhibiting high affinity and remarkable stability. BsNb‐Fc inhibited the release of cytokines in CD4+T cells during co‐culture experiments. BsNb‐Fc effectively alleviated colitis severity in mouse model with acute colitis induced by DSS or TNBS, outperforming the IFX&UST combination.

ObjectiveThis study aimed to investigate the epidemiological characteristics of common pathogens contributing to childhood lower respiratory tract infections (LRTIs) in Xiangtan City, Hunan Province before and during the coronavirus disease 2019 (COVID-19) pandemic.MethodsA total of 11,891 enrolled patients, aged 1 month to 14 years, diagnosed with LRTIs and admitted to Xiangtan Central Hospital from January 2018 to December 2021 were retrospectively reviewed in this study. Specifically, the epidemiological characteristics of these pathogens before and during the COVID-19 pandemic were analyzed.ResultsThere was a significant decrease in the number of children hospitalized with LRTIs during the COVID-19 pandemic (2020–2021) compared to data from 2018 to 2019 (before the COVID-19 pandemic). Of these cases, 60.01% (7,136/11,891) were male and 39.99% (4,755/11,891) were female. 78.9% (9,381/11,891) cases occurred in children under 4 years of age. The average pathogen detection rate among 11,891 hospitalized LRTIs children was 62.19% (7,395/11,891), with the average pathogen detection rate of 60.33% (4,635/7,682) and 65.57% (2,670/4,209) before and during COVID-19 pandemic, respectively. The detection rates of adenovirus (ADV), bordetella pertussis (BP) and moraxella catarrhalis (M. catarrhalis) decreased dramatically, while the detection rates of influenza viruses (IFV), parainfluenza viruses (PIV), respiratory syncytial virus (RSV), haemophilus influenzae (H. influenzae), streptococcus pneumoniae (S. pneumoniae), and staphylococcus aureus (S. aureus) increased significantly during the COVID-19 pandemic. Overall, RSV, mycoplasma pneumoniae (MP), H. influenzae, and IFV were the major pathogens causing LRTIs in hospitalized children before and during the COVID-19 pandemic.ConclusionPublic health interventions for COVID-19 prevention are beneficial to reduce the incidence of LRTIs in children by limiting the prevalence of ADV, MP, BP, and M. catarrhalis, but which have limited restrictive effects on other common LRTIs-associated pathogens. Collectively, the data in this study comprehensively investigated the effects of COVID-19 pandemic on the epidemiological characteristics of respiratory pathogens, which will be beneficial for improving early preventive measures.

Abstract Background The black cutworm, Agrotis ipsilon, is a serious global underground pest. Its distinct phenotypic traits, especially its polyphagy and ability to migrate long distances, contribute to its widening distribution and increasing difficulty of control. However, knowledge about these traits is still limited. Results We generated a high-quality chromosome-level assembly of A. ipsilon using PacBio and Hi-C technology with a contig N50 length of ~ 6.7 Mb. Comparative genomic and transcriptomic analyses showed that detoxification-associated gene families were highly expanded and induced after insects fed on specific host plants. Knockout of genes that encoded two induced ABC transporters using CRISPR/Cas9 significantly reduced larval growth rate, consistent with their contribution to host adaptation. A comparative transcriptomic analysis between tethered-flight moths and migrating moths showed expression changes in the circadian rhythm gene AiCry2 involved in sensing photoperiod variations and may receipt magnetic fields accompanied by MagR and in genes that regulate the juvenile hormone pathway and energy metabolism, all involved in migration processes. Conclusions This study provides valuable genomic resources for elucidating the mechanisms involved in moth migration and developing innovative control strategies.

Random mutagenesis is the natural opportunity for proteins to evolve and biotechnologically it has been exploited to create diversity and identify variants with improved characteristics in the mutant pools. Rational mutagenesis based on biophysical assumptions and supported by computational power has been proposed as a faster and more predictable strategy to reach the same aim. In this work we confirm that substantial improvements in terms of both affinity and stability of nanobodies can be obtained by using combinations of algorithms, even for binders with already high affinity and elevated thermal stability. Furthermore, in silico approaches allowed the development of an optimized bispecific construct able to bind simultaneously the two clinically relevant antigens TNF-α and IL-23 and, by means of its enhanced avidity, to inhibit effectively the apoptosis of TNF-α-sensitive L929 cells. The results revealed that salt bridges, hydrogen bonds, aromatic-aromatic and cation-pi interactions had a critical role in increasing affinity. We provided a platform for the construction of high-affinity bispecific constructs based on nanobodies that can have relevant applications for the control of all those biological mechanisms in which more than a single antigen must be targeted to increase the treatment effectiveness and avoid resistance mechanisms.

This study aimed to investigate the volatile flavor compounds and tastes of six kinds of sauced pork from the southwest and eastern coastal areas of China using gas chromatography–ion mobility spectroscopy (GC-IMS) combined with an electronic nose (E-nose) and electronic tongue (E-tongue). The results showed that the combined use of the E-nose and E-tongue could effectively identify different kinds of sauced pork. A total of 52 volatile flavor compounds were identified, with aldehydes being the main flavor compounds in sauced pork. The relative odor activity value (ROAV) showed that seven key volatile compounds, including 2-methylbutanal, 2-ethyl-3, 5-dimethylpyrazine, 3-octanone, ethyl 3-methylbutanoate, dimethyl disulfide, 2,3-butanedione, and heptane, contributed the most to the flavor of sauced pork (ROAV ≥1). Multivariate data analysis showed that 13 volatile compounds with the variable importance in projection (VIP) values > 1 could be used as flavor markers to distinguish six kinds of sauced pork. Pearson correlation analysis revealed a significant link between the E-nose sensor and alcohols, aldehydes, terpenes, esters, and hetero-cycle compounds. The results of the current study provide insights into the volatile flavor compounds and tastes of sauced pork. Additionally, intelligent sensory technologies can be a promising tool for discriminating different types of sauced pork.

Cotton aphid Aphis gossypii Glover damages plants such as cotton directly by feeding on leaves and indirectly by transmitting viruses and excreting honeydew, which interferes with photosynthesis. The control of A. gossypii is still dominated by the frequent use of insecticides, which leads to a gradual increase in pesticide resistance in A. gossypii. Research is therefore needed on non-pesticide controls. In this study, seven plant essential oils (EOs) of Ocimum sanctum L., Ocimum basilicum L., Ocimum gratissimum L., Mentha piperita L., Mentha arvensis L., Tagetes erecta L., and Lavandula angustifolia Mill. were examined as potential controls for A. gossypii. We used life tables and electrical penetration graphs (EPG) to explore the effects of these EOs on the growth, development, and feeding behavior of A. gossypii, followed by a study of effects of the EOs on honeydew secretion by A. gossypii as a measure of their antifeedant activity. We found that the EOs of O. sanctum, M. piperita, M. arvensis and T. erecta significantly extended the pre-adult developmental period. Also, adult longevity, number of oviposition days, and total fecundity of A. gossypii treated with the EOs of M. arvensis or T. erecta were all significantly reduced. Aphids treated with the EOs of O. sanctum, M. piperita, or L. angustifolia showed significant reductions in their net reproductive rate (R0), intrinsic rate of increase (rm), and finite rate of increase (λ), and significant increases in mean generation time (T). In terms of their effects on the feeding behavior of A. gossypii, all seven EOs significantly reduced the total duration of phloem feeding (E2 waves), the number of phloem-feeding bouts, and the proportion of time spent in secretion of saliva into phloem sieve elements (E1 waves) and phloem feeding (E2). The total duration and number of E1 waves (saliva secretion) were significantly reduced by the EOs of O. sanctum, O. gratissimum, and M. arvensis. For C waves (probing in non-vascular tissues), the total duration spent in this behavior was significantly increased by the EOs of O. gratissimum, M. piperita, and L. angustifolia, but the number of such probing events was increased only by L. angustifolia EO. The EOs of O. basilicum, M. arvensis, and T. erecta significantly increased the total duration of ingestion of xylem sap (G waves), while the total time of mechanical difficulty in stylet penetration (F waves) was increased by M. arvensis. The total duration and number of the non-probing events (Np waves) were significantly increased by EOs of O. sanctum and O. basilicum. After treatment with all seven of these EOs, the area covered by honeydew was significantly reduced compared with the control. Studies have analyzed that EOs of O. sanctum, M. piperita, and T. erecta were most effective, followed by the EOs of M. arvensis and L. angustifolia, and finally the EOs of O. basilicum and O. gratissimum. In the present study, the EOs of O. sanctum, M. piperita, and T. erecta were found to have potential for the development as antifeedants of A. gossypii, and these data provide a basis for future research on non-pesticide chemical control of A. gossypii.

The adulteration of soymilk (SM) into raw bovine milk (RM) to gain profit without declaration could cause a health risk. In this study, electronic nose (E-nose) and headspace-gas chromatography ion-mobility spectrometry (HS-GC-IMS) were applied to establish a rapid and effective method to identify adulteration in RM with SM. The obtained data from HS-GC-IMS and E-nose can distinguish the adulterated samples with SM by principal component analysis. Furthermore, a quantitative model of partial least squares was established. The detection limits of E-nose and HS-GC-IMS quantitative models were 1.53% and 1.43%, the root mean square errors of prediction were 0.7390 and 0.5621, the determination coefficients of prediction were 0.9940 and 0.9958, and the relative percentage difference were 10.02 and 13.27, respectively, indicating quantitative regression and good prediction performances of SM adulteration levels in RM were achieved. This research can provide scientific information on the rapid, non-destructive and effective adulteration detection for RM.

ABSTRACT: Diacetyl and acetoin are key aroma components of fermented milk but are produced in low concentrations by starter cultures. In this study, we expressed NADH oxidase, acetolactate synthase, and inactivated acetolactate decarboxylase in Lacticaseibacillus casei TCS to generate recombinant L. casei strains, and investigated the effects of the genes encoding these enzymes on diacetyl and acetoin production during milk fermentation. In the single-gene recombinant strains tested, diacetyl concentrations were highest in milk fermented by L. casei TCSI-nox (nox gene overexpressed, 3.68 mg/kg), whereas acetoin concentrations were highest in milk fermented by L. casei TCS-ΔalsD (alsD gene deleted, 32.94 mg/kg). Moreover, diacetyl and acetoin concentrations were higher in the inducible strains than in the corresponding constitutive strains (e.g., TCSI-nox vs. TCSC-nox, and TCSI-ΔalsD-nox vs. TCSC-ΔalsD-nox). This phenomenon was also reflected in the protein expression levels and enzyme activities. In the double-gene recombinant strains tested, the highest concentrations of diacetyl and acetoin were produced by L. casei TCSI-ΔalsD-nox (nox overexpressed and alsD deleted, 4.66 mg/kg, 69.62 mg/kg, respectively). The triple-gene recombinant L. casei TCS-ΔalsD-nox-alsS produced the highest concentrations of diacetyl and acetoin, which were 2.38 and 11.19 times, respectively, the concentrations produced by the original strain. These results show that the nox, alsS, and alsD genes make key contributions to the biosynthesis of diacetyl and acetoin by L. casei. The modification of multiple genes had a synergistic effect, leading to greatly increased synthesis of diacetyl and acetoin by L. casei during its fermentation of milk.

ABSTRACT: 3-Methylbutanal is a key volatile compound that imparts a nutty flavor to Cheddar cheese. Lactococcus lactis has been successfully applied as a starter to increase the level of 3-methylbutanal produced during the ripening of cheese. However, the mechanism of action and genetic diversity of this bacterium for 3-methylbutanal biosynthesis remains unclear. In this study, we investigated the association between the L. lactis genotype and phenotype in the biosynthesis of 3-methylbutanal via both direct and indirect pathways. Fourteen strains of L. lactis were screened for the capacity to produce 3-methylbutanal, and strain 408 (>140 μM) produced the highest among all tested strains, which exhibited both α-keto acid decarboxylase and α-ketoacid dehydrogenase activities. Furthermore, the results of a sodium meta-arsenite inhibition experiment showed that the 3-methylbutanal–producing capacities of each strain declined to various degrees. The kdcA gene, which encodes the direct pathway component α-ketoacid decarboxylase, was detected in 4 of the 14 strains, of which only strain 408 contained the full-length gene. We then characterized the genes associated with the indirect pathway by detecting the expression levels of the pdh gene cluster, ack, and pta, which were expressed at relatively higher levels in a high-yield strain than in a low-yield strain. As a result, these L. lactis strains were divided into 3 categories according to gene diversity, gene expression, and 3-methylbutanal production. The results of this study refine our knowledge of the genetic determinants of 3-methylbutanal biosynthesis in L. lactis and explain the effect of both synthesis pathways on 3-methylbutanal production.

A systematic flavoromics-based analysis of samples of 12 commercially available Gouda cheeses was performed to determine their key volatile components, the contribution of these components to the aromas of the cheeses, and which aromas were preferred by a panel of Chinese consumers. The sensory analysis results show that the Chinese consumers preferred young and medium cheeses, and that sensory attributes such as ‘milk’ and ‘cream’ were the most popular. Seventy-seven aroma compounds were identified by gas chromatography–mass spectrometry, and 28 of these were determined to be aroma-active compounds by gas chromatography–olfactometry analysis and calculation of their odour activity values. Partial least-squares analysis revealed that compounds such as diacetyl and acetoin correlated with aromas preferred by the Chinese consumers, while isobutyric acid, hexanoic acid and valeric acid correlated with aromas disliked by the Chinese consumers. Finally, the flavour contribution of each aroma-active compound was validated through aroma reorganisation and omission experiments.

Stinky tofu is a traditional Chinese food with wide consumption in China. Nevertheless, the dynamic changes in the flavour of stinky tofu during storage have yet to be investigated. In this study, the flavour changes of stinky tofu over six different storage periods were comprehensively analysed through sensory, electronic nose and gas chromatography-mass spectrometry (GC-MS) analyses. The results of the sensory and electronic nose analyses confirmed the changes in the flavour of stinky tofu across different storage periods. In the GC-MS analysis, 60 volatile compounds were detected during storage, and the odour activity values indicated that 29 of these 60 compounds significantly contributed to the aroma profile. During storage, the alcohol concentration of the stinky tofu gradually decreased while the acid and ester concentrations increased. According to a partial least squares analysis, 2-phenylethyl acetate, 2-phenylethyl propanoate, p-cresol, and phenylethyl alcohol, which were detected after 10 days of storage, promoting the release of an overripe apple-like odour from the stinky tofu. Findings regarding the flavour changes and characteristics of stinky tofu during different storage periods can provide a potential reference for recognising the quality of these products.

Recombinant antibodies such as nanobodies are progressively demonstrating to be a valid alternative to conventional monoclonal antibodies also for clinical applications. Furthermore, they do not solely represent a substitute for monoclonal antibodies but their unique features allow expanding the applications of biotherapeutics and changes the pattern of disease treatment. Nanobodies possess the double advantage of being small and simple to engineer. This combination has promoted extremely diversified approaches to design nanobody-based constructs suitable for particular applications. Both the format geometry possibilities and the functionalization strategies have been widely explored to provide macromolecules with better efficacy with respect to single nanobodies or their combination. Nanobody multimers and nanobody-derived reagents were developed to image and contrast several cancer diseases and have shown their effectiveness in animal models. Their capacity to block more independent signaling pathways simultaneously is considered a critical advantage to avoid tumor resistance, whereas the mass of these multimeric compounds still remains significantly smaller than that of an IgG, enabling deeper penetration in solid tumors. When applied to CAR-T cell therapy, nanobodies can effectively improve the specificity by targeting multiple epitopes and consequently reduce the side effects. This represents a great potential in treating malignant lymphomas, acute myeloid leukemia, acute lymphoblastic leukemia, multiple myeloma and solid tumors. Apart from cancer treatment, multispecific drugs and imaging reagents built with nanobody blocks have demonstrated their value also for detecting and tackling neurodegenerative, autoimmune, metabolic, and infectious diseases and as antidotes for toxins. In particular, multi-paratopic nanobody-based constructs have been developed recently as drugs for passive immunization against SARS-CoV-2 with the goal of impairing variant survival due to resistance to antibodies targeting single epitopes. Given the enormous research activity in the field, it can be expected that more and more multimeric nanobody molecules will undergo late clinical trials in the next future.Systematic Review Registration

The oriental armyworm Mythimna separata (Walker) is a worldwide migratory pest that threatens food security in China. Previous studies have clarified the general migration regularity of M. separata in East Asia, but knowledge of migration routes over northeast China, especially in autumn as well as the impact by climate change in recent years, is still limited. From 2017 to 2020, we monitored the migration of M. separata in northeast China by searchlight traps and simulated moth trajectories by the HYSPLIT model. The M. separata moths had three main migration periods in northeast China, and there were obvious seasonal differences at different latitudes. The spring and summer migrants (late May to early-mid June and late July to early August) came from the southern Shandong Province and migrated to the north via Hebei/Shandong-Liaoning-Jilin-Heilongjiang. Moreover, more than half of the autumn trapped insects from late August to mid-late September migrated in the provinces where the sites are located, southward via Heilongjiang-Jilin-Liaoning. These results clarified the migration route of M. separata in northeast China and their relationship with the insect source in north China, thereby providing a theoretical basis for regional monitoring, early warning, and management of the pest.

Plutella xylostella (L.) is one of the most serious pests of cruciferous vegetables. Our previous work demonstrated that the essential oil of Artemisia lavandulaefolia DC. exhibits promising insecticidal activities against P. xylostella. This study further characterizes the key components that are responsible for the insecticidal effect. In total, 47 compounds (96.52% of the total compounds) were identified from the total oil using GC-MS, and the major compounds were eucalyptol (21.57%), D(+)-camphor (17.33%), (-)-4-terpineol (9.96%) and caryophyllene oxide (10.96%). Among them, (-)-4-terpineol showed significantly larvicidal and fumigant activities against P. xylostella. The LD50 of (-)-4-terpineol was 43.15 mg/mL at 12 h and 31.22 mg/mL at 24 h for 3rd instar larvae, and the LC50 for adults was 8.34 mg/mL at 12 h and 7.35 mg/mL at 24 h. In addition, the adults treated with (-)-4-terpineol showed varying degrees of inhibitory activity toward glutathione S-transferase, catalase, acetylcholinesterase and Na+/K+-ATPase at different post-treatment intervals and concentrations. The results indicate that (-)-4-terpineol has promising insecticidal activities against P. xylostella, and it has good inhibitory effects on the four enzymes of P. xylostella adults.

In this study, an artificial starter culture was prepared using the core microbial species of JIUYAO to produce Chinese rice wine (CRW). The fermentation activity and flavor profiles of CRW samples fermented with traditional JIUYAO, a commercial starter culture, and our artificial starter culture were compared. The optimal protectant combination for lyophilization of the artificial starter was established as 15.09% skim milk, 4.45% polyethylene glycol, 1.96% sodium glutamate, and 11.81% maltodextrin. A comparative analysis revealed that the ethanol content of the three CRW samples was similar. The total acid content of the CRW sample fermented with the artificial starter (7.10 g/L) was close to that of the sample fermented with JIUYAO (7.35 g/L), but higher than that of the sample fermented with the commercial starter (5.40 g/L). An electronic nose analysis revealed that the olfactory fingerprints of the CRW samples fermented with JIUYAO and the artificial starter resembled each other. For both above mentioned samples, the flavor profiles determined by gas chromatography–mass spectrometry indicated some differences in the variety and content of the aroma compounds, but the key odorants (odor activity values ≥1), such as isoamyl acetate, ethyl acetate, phenyl alcohol, and isoamyl alcohol, were similar.

ATP-binding cassette transporter B1 (ABCB1, or P-glycoprotein) is known to be an important participant in multidrug resistance in mammals, and it also has been proved as a transporter for some insecticides in several lepidopteran insects, yet the precise function of this transporter in Spodoptera frugiperda is unknown. Here, we generated a SfABCB1 knockout strain of the S. frugiperda using the CRISPR/Cas9 system to explore its potential roles in determining susceptibility to chemical insecticides or Bt toxins. Bioassay results showed that the susceptibility of SfABCB1 knockout strain to beta-cypermethrin, chlorantraniliprole and emamectin benzoate were significantly increased compared with the wild-type strain DH19, whereas there were no changes to Bt toxins for Cry1Ab, Cry1Fa and Vip3Aa. Our results revealed that SfABCB1 plays important roles in the susceptibility of S. frugiperda to beta-cypermethrin, chlorantraniliprole and emamectin benzoate, and imply that overexpression of ABCB1 may contribute to beta-cypermethrin, chlorantraniliprole and emamectin benzoate resistance in S. frugiperda.

Post-harvest diseases of mango reduce fruit quality and cause severe yield losses with completely unmarketable fruits. The most common diseases of mangos are anthracnose (Colletotrichum gloeosporioides). In this study, the antibacterial activities of essential oils from Artemisia scoparia, Artemisia lavandulaefolia, and Artemisia annua against C. gloeosporioides were tested. The results showed that the essential oil of A. scoparia was more effective by the agar diffusion method; the EC50 value was 9.32 µL/mL. The inhibition rate was 100%, at a concentration of 10 μL/mL, through the spore germination method. The morphological changes of the mycelium were observed by scanning electron microscopy (SEM), the mycelia treated with essential oils showed shrinking, deformity, fracture, and dryness through SEM. A. scoparia essential oil was inoculated in vivo and subjected to paroxysm testing under natural conditions. A. scoparia had significantly inhibitory activity, and the inhibition rate was 66.23% in vivo inoculation tests after 10 days. The inhibition rate was 92.06% in the paroxysm test under natural conditions after 15 days. Finally, A. acoparia essential oil was analyzed by gas chromatography-mass spectrometry. The main compounds were 2-ethenyl-Naphthalene (23.5%), 2,4-pentadiynyl-Benzene (11.8%), 1,2-dimethoxy-4-(2-propenyl)-Benzene (10.0%), β-Pinene (8.0%), and 1-methyl-4-(1-methylethyl)-1,4-Cyclohexadiene (6.3%). The results have revealed the potential use of A. scoparia essential oil against post-harvest fungal pathogens C. gloeosporioides.

Many plants show significant biological activity against pests due to their unique chemical constituents. It is important to identify effective constituents for their development and utilization as botanical pesticides. Our previous study showed that Artemisia lavandulaefolia essential oil had biological activity against Plutella xylostella. Here, we isolated and identified the constituents of essential oil from A. lavandulaefolia by silica gel column chromatography. The main constituents identified were eucalyptol and caryophyllene oxide, and they were confirmed by gas chromatography–mass spectrometry (GC–MS). Eucalyptol and caryophyllene oxide showed strong contact toxicity against P. xylostella larvae after 24 h of application (Median lethal dose, LD50 = 76.97 μL/mL and 20.71 mg/mL. Furthermore, the two active constituents against P. xylostella adults showed significant fumigant activity (Mmedian lethal concentration, LC50 = 3.25 μL/L and 1.06 mg/L, respectively. Finally, we measured the detoxification enzymes and acetylcholinesterase of the larvae treated with active constituents. The eucalyptol-treated larvae displayed enhanced carboxylesterase (CarE) and glutathione-S-transferase (GST) activities in an in vivo experiment, but it was lower for acetylcholinesterase (AchE) activity. The activities of the CarE and GST significantly decreased when exposed to caryophyllene oxide. In general, the two active constituents, eucalyptol and caryophyllene oxide, showed high insecticidal activity, which demonstrates their potential to be used as natural insecticides.

The case-based reasoning (CBR) method can effectively predict the future health condition of the system based on past and present operating data records, so it can be applied to the prognostic and health management (PHM) framework, which is a type of data-driven problem-solving. The establishment of a CBR model for practical application of the Ground Special Vehicle (GSV) PHM framework is in great demand. Since many CBR algorithms are too complicated in weight optimization methods, and are difficult to establish effective knowledge and reasoning models for engineering practice, an application development using a CBR model that includes case representation, case retrieval, case reuse, and simulated annealing algorithm is introduced in this paper. The purpose is to solve the problem of normal/abnormal determination and the degree of health performance prediction. Based on the proposed CBR model, optimization methods for attribute weights are described. State classification accuracy rate and root mean square error are adopted to setup objective functions. According to the reasoning steps, attribute weights are trained and put into case retrieval; after that, different rules of case reuse are established for these two kinds of problems. To validate the model performance of the application, a cross-validation test is carried on a historical data set. Comparative analysis of even weight allocation CBR (EW-CBR) method, correlation coefficient weight allocation CBR (CW-CBR) method, and SA weight allocation CBR (SA-CBR) method is carried out. Cross-validation results show that the proposed method can reach better results compared with the EW-CBR model and CW-CBR model. The developed PHM framework is applied to practical usage for over three years, and the proposed CBR model is an effective approach toward the best PHM framework solutions in practical applications.

Resource-continuity over spatial and temporal scales plays a central role in the population abundance of polyphagous pests in an agricultural landscape. Shifts in the agricultural land use in a region may alter the configuration of key resource habitats, resulting in drastic changes in pest abundance. Apolygus lucorum (Meyer-Dür) (Hemiptera: Miridae) is a pest of cotton in northern China that has become more serious in recent years following changes in the region’s cropping systems. However, no evidence from the landscape perspective has yet been gathered to account for the increasing population of A. lucorum in China. In this study, we investigated the effects of landscape composition on the population abundance of A. lucorum in cotton fields in July and August of 2016, respectively. We found that increased acreage planted to cotton actually had a negative effect on the abundance of A. lucorum, while planting of other crops (e.g., vegetables, soybean, and peanut) was positively associated with the mirid’s population abundance in cotton fields. Maize production only displayed a positive effect on population abundance in August. Our results suggested that the decreasing of cotton area may weaken the trap-kill effect on A. lucorum, and the extension of other crops and maize potentially enhance the continuity of resources needed by A. lucorum. Combined effects of these two aspects may promote an increased population density of A. lucorum in the agriculture district. In the future, when possible, management strategies in key regional crops should be coordinated to reduce resource continuity at the landscape or area-wide scale to lower A. lucorum populations across multiple crops.

Objective This meta-analysis was conducted to examine the possible association between serum zinc concentration and cervical cancer risk. Methods PubMed, WanFang, China National Knowledge Infrastructure, and SinoMed databases were searched for relevant articles published between January 1980 and September 2017. Results were combined using a random-effects model, and pooled standardized mean differences (SMD) and 95% confidence intervals (CI) were calculated to compare serum zinc levels in patients with cervical cancer versus controls. Publication bias was evaluated using Begg’s funnel plot and Egger’s regression asymmetry test. Results Twelve articles regarding serum zinc levels and cervical cancer were included in this meta-analysis. Combined results showed that serum zinc levels in cervical cancer cases were significantly lower than in controls without cervical cancer (summary SMD –1.379, 95% CI –1.527, –1.231), with high heterogeneity ( I 2 = 98.8%). Analysis of data stratified by geographic location showed a significant association between serum zinc levels and cervical cancer risk in Asian populations (summary SMD –1.391, 95% CI –1.543, –1.239). Conclusions Higher serum zinc levels may be a protective factor for cervical cancer in Asian women.
