Abstract Aims Surveys and research on the applications of the hepatic venous pressure gradient (HVPG) are important for understanding the current status and future development of this technology in China. This article aimed to investigate the status of hepatic venous pressure gradient measurement in China in 2022. Methods We investigated the overall status of HVPG technology in China—including hospital distribution, hospital level, annual number of cases, catheters used, average cost, indications, and current challenges by using online questionnaire. By counting the number and percentages of cases of these results, we hope to clarify the current status of HVPG measurements in China. Results According to the survey, 85 hospitals in China used HVPG technology in 2022 distributed across 29 provinces. A total of 4989 HVPG measurements were performed in all of the surveyed hospitals in 2022, of which 2813 cases (56.4%) were measured alone. The average cost of HVPG measurement was 5646.8 ± 2327.9 CNY. Of the clinical teams who performed the measurements (sometimes multiple per hospital), 94.3% (82/87) used the balloon method, and the majority of the teams (72.4%, 63/87) used embolectomy catheters. Conclusions This survey clarified the clinical application status of HVPG in China and confirmed that some medical institutions in China have established a foundation for this technology. It is still necessary to continue promoting and popularizing this technology in the future.

Globally, water scarcity threatens food security, especially in arid and semi-arid food-producing regions, such as the Hetao Irrigation District (HID), where the water consumption during the non-growth season (spring irrigation, autumn irrigation) accounts for more than 30 % of the total irrigation volume. Meanwhile, mapping the characteristics of spatial and temporal evolution of the area of cropland irrigated in the spring and autumn and the pattern of water consumption is crucial for strengthening the management of agricultural water use. Evaporation and infiltration of irrigation water are difficult to capture in time due to insufficient timeliness or accuracy of single remote sensing data. This study integrates seven remote sensing datasets (Landsat5, Landsat7, Landsat8, HJ-1A/B, GF-1, Sentinel-2 and MODIS) for the first time to build a high spatial-temporal resolution dataset, and combines the water index NDWI and MNDWI to extract the maximum water range formed after irrigation by manually adjusting the threshold method. In addition to the global land use/cover dataset (GLC FCS30–1985_2020), the cropland layer was superimposed to accurately identify the cropland spring irrigation and autumn irrigation range from 2000 to 2021 in the HID. In the past 20 years, spring-irrigated (SI) areas increased by 1012.60 km2 (+49.96 %), and the autumn-irrigated (AI) areas decreased by 512.63 km2 (-11.02 %). Combined with the quantity of water diversion and displacement, the characteristics of irrigation water consumption in the non-growth season were further analysed. The results showed that the water consumption of spring irrigation increased by 0.67×108 m3 (+22.26 %), and that of autumn irrigation increased by 4.34×108 m3 (+38.82 %). The decrease of autumn- irrigated area and the increase of water consumption were mainly related to the stability of autumn irrigation water, imperfect irrigation and drainage conditions and insufficient field water management. In 2020­2021, the area of non-growth “autumn irrigation - spring irrigation” repeated irrigation is about 1271.52 km2, which can be used as a key control area. This study provides a theoretical basis and direction for rational water use in non-growth seasons of irrigation districts.

ObjectivesThis study aimed to evaluate whether the “weekend effect” would affect the time metrics and the prognosis of acute ischemic stroke (AIS) patients who underwent endovascular treatment (EVT) due to basilar artery occlusion (BAO).MethodsClinical data of AIS patients who underwent EVT due to BAO between December 2019 and July 2023 were retrospectively analyzed. At the time when the patients were admitted, the study population was divided into the weekdays daytime group and weekends nighttime group. In the subgroup analysis, the study cohort was divided into four groups: the weekdays daytime group, weekdays nighttime group, weekend daytime group, and weekend nighttime group. A good outcome was defined as a modified Rankin Scale score of ≤3 at 90 days after EVT. Time metrics [e.g. onset-to-door time (ODT) and door-to-puncture time (DPT)] and clinical outcomes were compared using appropriate statistical methods.ResultsA total of 111 patients (88 male patients, mean age, 67.7 ± 11.7 years) were included. Of these, 37 patients were treated during weekdays daytime, while 74 patients were treated during nights or weekends. There were no statistically significant differences in ODT (P = 0.136), DPT (P = 0.931), and also clinical outcomes (P = 0.826) between the two groups. Similarly, we found no significant differences in the time metrics and clinical outcomes among the four sub-groups (all P > 0.05).ConclusionThis study did not reveal any influence of the “weekend effect” on the time metrics and clinical outcomes in AIS patients who underwent EVT due to BAO at a comprehensive stroke center.

Abstract Background Spinal subarachnoid hematoma (SSH) is a known but rare entity that can cause cauda equina compression. The occurrence of SSH associated with aneurysmal subarachnoid hemorrhage has rarely been described in the literature. Case presentation A 56-year-old woman presented with subarachnoid hemorrhage secondary to a ruptured middle cerebral artery aneurysm and was managed with coiling embolization without stent assistance. There was no history of either lumbar puncture or the use of anticoagulants. The patient developed severe lumbago radiating to bilateral legs nine days after the procedure. Subsequent magnetic resonance imaging demonstrated a SSH extending from L5 to S2 and wrapping around the cauda equina. The patient was treated with intravenous methylprednisolone (250 mg/day) for four consecutive days, followed by a taper of oral prednisolone (20 mg/day) until complete recovery. Magnetic resonance imaging at one month follow-up revealed complete resolution of the SSH. Conclusions Here, we report a case of acute cauda equina syndrome caused by a SSH after aneurysmal subarachnoid hemorrhage, which will facilitate timely intervention of patients with this disorder.

Fully biodegradable mulch film enhances temperature and moisture retention during the early stages of maize growth while naturally degrading in the later stages, providing an environmentally friendly alternative to conventional plastic mulch films. However, there is no consensus on its impact on maize growth and yield. The present study utilized field test data from spring maize covered with fully biodegradable mulch film in the Xiliaohe Plain, aiming to improve the Decision Support System for Agrotechnology Transfer (DSSAT) model while focusing on soil temperature, irrigation, rainfall, and evapotranspiration. The parameters of the DSSAT model were calibrated and validated using field test data from 2016 to 2018. The improved DSSAT model accurately simulated the maize growth process under various induction periods of fully biodegradable mulch film. The simulation accuracy of this model was as follows: MRE < 10%, nRMSE < 12%, and R2 ≥ 0.80. Moreover, the yield of spring corn covered with fully biodegradable mulch film was predicted using meteorological data from 2019 to 2023. This study suggests that regions such as the Xiliaohe Plain, which share climatic conditions, should opt for fully biodegradable mulch film with an induction period of approximately 80 days to ensure high yields across different hydrological years.

The impact of fertilization and irrigation on heavy metal accumulation in saline–alkali soil and its underlying mechanisms are critical issues given the constraints that soil salinization places on agricultural development and crop quality. This study addressed these issues by investigating the effects of adjusting organic fertilizer types, proportions, and irrigation volumes on the physicochemical properties of lightly to moderately saline–alkali soils and analyzing the interaction mechanisms between microorganisms and heavy metals. The results indicate that the rational application of organic fertilizers combined with supplemental irrigation can mitigate soil salinity accumulation and water deficits, and reduce the soil pH, thereby enhancing soil oxidation, promoting nitrogen transformation and increasing nitrate–nitrogen levels. As the proportion of organic fertilizers increased, heavy metal residues, enrichment, and risk indices in the crop grains also increased. Compared to no irrigation, supplemental irrigation of 22 mm during the grain-filling stage increased soil surface Cd content, Zn content, and the potential ecological risk index (HRI) by 10.2%, 3.1%, and 8%, respectively, while simultaneously reducing the heavy metal content in grains by 12–13.5% and decreasing heavy metal enrichment. Principal component analysis revealed the primary factors influencing Cu and Zn residues and Cd accumulation in the crop grains. Soil salinity was significantly negatively correlated with soil pH, organic matter, total nitrogen, and ammonium nitrogen, whereas soil organic matter, total nitrogen, ammonium nitrogen, soil pH, oxidation–reduction potential, soluble nitrogen, and microbial biomass nitrogen were positively correlated. The accumulation and residues of Zn and Cu in the soil were more closely correlated with the soil properties compared to those of Cd. Specifically, Zn accumulation on the soil surface was primarily related to aliphatic organic functional groups, followed by soil salinity. Residual Zn in the crop grains was primarily associated with soil oxidation–reduction properties, followed by soil moisture content. The accumulation of Cu on the soil surface was mainly correlated with the microbial biomass carbon (MBC), whereas the residual Cu in the crop grains was primarily linked to the soil moisture content. These findings provide theoretical insights for improving saline–alkali soils and managing heavy metal contamination, with implications for sustainable agriculture and environmental protection.

To mitigate the issues of severe farmland soil salinization, the environmental degradation stemming from the overuse of chemical fertilizers, and suboptimal soil composition, a study was conducted to investigate the influence of different types and ratios of organic fertilizers on the physical and chemical attributes of saline–alkali soil. This study aimed to investigate the relationship between different types and proportions of organic fertilizers, soil moisture, organic fertilizer application rates, organic carbon molecular structure, and the soil environment in saline–alkali soils. Reducing the application of chemical fertilizers and substituting them with organic fertilizers can improve the soil quality of saline–alkali lands. The results indicated that replacing a part of the urea with organic fertilizer in saline–alkali farmland reduced the soil salinity by 11.1 to 22.8% in the 0–60 cm soil layer, decreased the soil pH by 0.11 to 1.52%, and increased the soil redox potential (Eh) values by 2.5 to 4.3% in the 0–20 cm layer of the mild and moderate saline–alkali soils. It also decreased the accumulation of the soil organic matter (OM) during the growing season. Compared to commercial organic fertilizers, natural organic fertilizers increased the accumulation of the soil soluble carbon (DOC) and nitrogen (DON), resulting in less soil salinity accumulation. When commercial organic fertilizer was applied in a 1:1 ratio with inorganic fertilizer, the salt accumulation was minimized. Compared to conventional fertilization, organic fertilizer reduced the accumulation of the NH4+-N (ammonium nitrogen) and NO3−-N (nitrate nitrogen) in the soil by 3.1 to 22.6%. In comparison to conventional chemical fertilizers, the application of organic fertilizer in the mild and moderate saline–alkali soils increased the accumulation of the DOC, DON, microbial biomass carbon (MBC), microbial biomass nitrogen (MBN), and microbial quotient during the grain-filling stage. Specifically, it increased the DOC, DON, and DOC/DON by 12.7 to 26.7%, 12 to 59.3%, and 15.2 to 35.5%, respectively. The application of commercial organic fertilizer in the mild saline–alkali soils increased the MBC, MBN, MBC/SOC, and MBN/TN by 37.1, 65.6, 36.7, and 4.7%, respectively. Through analyzing the relative proportions of soil surface organic carbon functional groups during the grain filling period, we observed that, after the application of organic fertilizer, the OM in the mildly salinized soils primarily originated from terrestrial plant litter, whereas, in moderately salinized soils, the OM was mainly derived from microbial sources.

In order to explore the effect of fishponds on soil water, salt transport and salinization in cropland wasteland, a study on soil water balance and salt distribution pattern in a cropland–wasteland–fishpond system was carried out in 2022–2023 in a typical study area selected from the Yichang Irrigation Area of the Hetao Irrigation District. A water balance model was established for the cropland–wasteland–fishpond system to analyze the effects of irrigation on soil salinity at the boundaries of the cropland, wasteland, and fishpond. The results showed that the lateral recharge from the cropland to the wasteland during spring irrigation in 2022 was 24 mm, the lateral recharge generated by fishponds to wasteland was 18 mm, and the lateral recharge from fishponds to fishpond boundaries was 34 mm. In the fertility period of 2023, the lateral recharge from cropland to wasteland was 15 mm, the lateral recharge from fishponds to wasteland was 9 mm, and the lateral recharge from fishponds to fishpond boundaries was 21 mm. Due to the low salinity content of fishpond water, it diluted the groundwater of the wasteland, and the soil salinity at the boundary between the wasteland and the fishpond was monitored. The data show that the soil salinity at the boundary of the fishpond was smaller than that of the wasteland, which indicates that the migration of fishpond water to the wasteland will not lead to an increase in the soil salinity of the wasteland, but rather to a decrease in the soil salinity of the wasteland. Fishpond regulation has a significant impact on soil and groundwater, and when the topographic conditions of the Hetao irrigation area allow, the model of cropland–wasteland–fishpond can be appropriately adopted to solve land degradation and increase the economic income of farmers; the results of the study provide a contribution for the improvement of the management of land use and soil salinization in the Hetao irrigation area.

Background This study aimed to establish and validate a nomogram model for predicting 90‐day mortality in patients with acute basilar artery occlusion receiving endovascular thrombectomy. Methods and Results A total of 242 patients with basilar artery occlusion undergoing endovascular thrombectomy were enrolled in our study, in which 172 patients from 3 stroke centers were assigned to the training cohort, and 70 patients from another center were assigned to the validation cohort. Univariate and multivariate logistic regression analyses were adopted to screen prognostic predictors, and those with significance were subjected to establish a nomogram model in the training cohort. The discriminative accuracy, calibration, and clinical usefulness of the nomogram model was verified in the internal and external cohorts. Six variables, including age, baseline National Institutes of Health Stroke Scale score, Posterior Circulation–Alberta Stroke Program Early CT (Computed Tomography) score, Basilar Artery on Computed Tomography Angiography score, recanalization failure, and symptomatic intracranial hemorrhage, were identified as independent predictors of 90‐day mortality of patients with basilar artery occlusion and were subjected to develop a nomogram model. The nomogram model exhibited good discrimination, calibration, and clinical usefulness in both the internal and the external cohorts. Additionally, patients were divided into low‐, moderate‐, and high‐risk groups based on the risk‐stratified nomogram model. Conclusions Our study proposed a novel nomogram model that could effectively predict 90‐day mortality of patients with basilar artery occlusion after endovascular thrombectomy and stratify patients with high, moderate, or low risk, which has a potential to facilitate prognostic judgment and clinical management of stroke.

Abstract Glioma is the most common malignant tumor of the brain and enhancing the efficacy of chemotherapy in glioma is critical for improving patients’ prognosis. In this study, a glioma-targeting drug delivery system is constructed using biodegradable periodic mesoporous organosilica nanoparticles (PMO) that are modified with lactoferrin (Lf) ligands. The obtained PMO is doped with thioether groups and can be degraded in the high concentration of glutathione in tumor cells. The surface area and pore volume of PMO are 772 cm2/g and 0.98 cm3/g, respectively and the loading capacity of doxorubicin (Dox) is as high as 20%. The results of the confocal laser scanning microscope show that the uptake of PMO-Lf@Dox by C6 cells is higher than PMO@Dox. The quantitative analysis of the flow cytometer further demonstrates that more PMO-Lf@Dox enter C6 cells, indicating that the modification of lactoferrin can significantly increase the uptake of C6 cells. Finally, the therapeutic efficacy results show that Lf-modified PMO enhances the inhibitory effect of Dox on C6 cells when incubated for 24 h and 72 h. In summary, this lactoferrin receptor-mediated PMO drug carrier with biodegradability in glutathione in tumor cells can be used to enhance drug delivery into glioma without long-term accumulation in vivo. PurposeTo evaluate the prognostic value of the cerebral blood volume (CBV) index for 90-day functional outcomes in patients with acute ischemic stroke (AIS) treated within a late therapeutic window.MethodsWe retrospectively reviewed patients who underwent pre-treatment computed tomography perfusion (CTP) and endovascular thrombectomy (EVT) for large-vessel occlusion (LVO) of the anterior circulation within the late therapeutic window between January 2021 and February 2023. Clinical data, the Alberta Stroke Program Early Computed Tomography Score (ASPECTS) based on unenhanced computed tomography (CT), and perfusion parameters, including ischemic core, hypoperfusion volume, mismatch volume between the core and penumbra, and CBV index, were assessed and compared between patients who achieved favorable outcomes (defined as a modified Rankin Scale score of 0–2).ResultsOf the 118 patients, 56 (47.5%) had favorable outcomes. In the univariate analysis, age, National Institutes of Health Stroke Scale (NIHSS) score at admission, ASPECTS score, CBV index, and ischemic core volume were significantly associated with functional outcomes (P < 0.05). In multivariate analyses, age (odds ratio [OR], 1.060; 95% confidence interval [CI] 1.013–1.110, P = 0.012), NIHSS score at admission (OR, 1.126; 95% CI 1.031–1.229, P = 0.009), and CBV index (OR, 0.001; 95% CI 0.000–0.240, P = 0.014) were independent predictors of a 90-day favorable outcome.ConclusionA high CBV index was independently associated with favorable outcomes in patients who underwent mechanical thrombectomy within the late therapeutic window. In addition, a higher CBV index reflects improved blood flow and favorable digital subtraction angiography collateral status.

Purpose. Preoperative oral carbohydrate (CHO) is a rapid postoperative rehabilitation protocol that improves perioperative outcomes and is widely used in adult surgical patients. However, pregnant women are excluded because of the possibility of aspiration due to delayed gastric emptying. This meta-analysis was conducted to evaluate the efficacy of preoperative oral CHO in elective cesarean section. Methods. PubMed, Embase, Web of Science, and the Cochrane Library were searched from inception to July 2023. Randomized controlled trials were included. The risk of bias was assessed using the Cochrane tool. Risk ratios and 95% confidence intervals were calculated. Meta-analysis was performed using random-effects models to estimate risk ratios and mean differences (MDs) with 95% confidence intervals (CIs). The outcomes included thirst and hunger scores, incidence of vomiting and nausea, time to flatus, and homeostatic model assessment of insulin resistance (HOMA-IR). Results. A total of nine studies with 1211 patients were included in the analysis. The levels of thirst and hunger were evaluated using a 10-point visual analog scale, with 0 representing the best and 10 representing the worst. The severity of hunger (weighted mean difference (WMD: −2.34, 95% CI: −3.13 to −1.54), time to flatus (WMD: −3.51 hours, 95% CI: −6.85 to −0.17), and HOMA-IR (WMD: −1.04, 95% CI: −1.31 to −0.77) were significantly lower in the CHO group compared to the control group. However, there were no significant differences in the severity of thirst or the incidence of vomiting and nausea between the CHO and control groups. Conclusion. Preoperative oral CHO during cesarean section alleviates thirst and hunger, shortens the time of postoperative flatus, and reduces HOMA-IR. However, the available evidence is insufficient to reach a clear consensus on the benefits or harms of preoperative oral CHO during cesarean section. Therefore, it is premature to make a definitive recommendation for or against its routine use.

Brackish water has been increasingly used worldwide for irrigation to relieve crop water stress. However, crop yield reductions inevitably occur under uncontrolled irrigation with saline waters, especially in areas with shallow and saline groundwater. It is essential to quantify the movement of soil water and salts in a soil-plant system and to optimize irrigation patterns with brackish water to alleviate the harm of salt stress on crop growth. Therefore, a two-year cornfield experiment was carried out during 2020–2021 to evaluate differences in soil water and salt movement under different irrigation regimes: a) brackish water irrigation with high (HB), medium (MB), and low (LB) irrigation depths, and b) freshwater irrigation with medium irrigation depths (MF). Moreover, the HYDRUS (2D/3D) model was used to evaluate soil water contents (SWC), electrical conductivities of the saturation paste extract (ECe), and fluxes, distributions, and mass balances of soil water and salts for the HB, MB, LB, and MF regimes. Meanwhile, oxygen isotope (18O) was used to calibrate and verify the simulation accuracy of the HYDRUS (2D/3D) model by determining the proportion of water and salt sources in a soil-plant system.The results showed that HYDRUS (2D/3D) could precisely capture SWC and ECe under different irrigation treatments, with the root mean square error (RMSE) of 0.004–0.017 cm3 cm−3 and 0.023–0.372 dS m−1 for SWC and ECe, respectively. The highest differences in SWCs and ECe in plots under brackish water and freshwater irrigations occurred in the 0–20 cm soil layer. The stress areas in the soil profile due to water and salt stresses under MB decreased and increased by 1.8 cm2 and 2010.0 cm2, respectively, compared with MF, accounting for 0.04% and 40.2% of the root zone. However, average root water uptake (RWU) in both years under MB decreased by 1.9% compared to MF. Additionally, RWU showed a nonlinear relationship with irrigation depth when brackish water was used for irrigation. The highest RWU occurred under MB, where the average RWU in both years increased by 7.7% and 12.4% compared with HB and LB, respectively. Therefore, the MB treatment could be recommended to increase crop yield and alleviate the freshwater shortage in agricultural production. Moreover, the findings of this study improve our understanding of the mechanisms of soil water and salt movement and groundwater contributions in a system under brackish water irrigation with different irrigation depths.

Abstract Objective This study aimed to develop and validate a nomogram to predict the risk of pancreatitis after percutaneous transhepatic biliary stent insertion (PTBS) in patients with malignant biliary obstruction (MBO). Materials and methods We enrolled 314 patients who underwent PTBS for MBO from March 2016 to July 2021 in this retrospective study. We used univariate analysis to identify potential risk factors, while a multivariate logistic regression model was employed to establish a nomogram for predicting the risk of pancreatitis. The discrimination and calibration of the nomogram were evaluated by estimating the area under the receiver operator characteristic curve (AUC) and by bootstrap resampling and visual inspection of the calibration curve. The clinical utility of the nomogram was assessed using decision curve analysis (DCA). Results After the procedure, 41 (13.1%) patients developed pancreatitis. Based on multivariate logistic regression analysis, young age (OR = 2.57, 95% CI 1.16 to 5.69), stent insertion across the papilla (OR = 6.47, 95% CI 2.66 to 15.70), and visualization of the pancreatic duct (OR = 15.40, 95% CI 6.07 to 39.03) were associated with an elevated risk of pancreatitis. Importantly, the performance of the nomogram was satisfactory, with an identical AUC (0.807, 95% CI 0.730 to 0.883) and high-level agreement between predicted and observed probabilities as suggested in calibration curves. The DCA curve subsequently confirmed the clinical utility. Conclusion A predictive nomogram for pancreatitis after PTBS in patients with MBO was successfully established in the present study.

Regulating the plastic film mulching duration was a good strategy to decrease plastic residues and to avoid the stress from low soil nitrate concentration (SNC). However, few studies focused on soil nitrogen dynamic under different mulching durations, especially for the impacts of soil nitrogen loss on crop yield and comprehensive benefits have not been revealed. In this study, HYDRUS (2D/3D) model was used to evaluate the differences of SNC and NO3-N leaching (NL) under no mulching (NM), mulching for the seedling stage (PMS), mulching for the elongation stage (PME), mulching for the tasseling stage (PMT), and mulching for the entire corn growth season (PMW). Additionally, a two-year (2019–2020) field experiment was carried out to measure SNC, NL, N2O, and NH3 emissions and to calibrate and validate the model accuracy. The effect of soil nitrogen loss on the comprehensive benefits under different mulching durations was further analyzed using a comprehensive multi-objective model. The results showed that HYDRUS (2D/3D) could well capture SNC dynamic under different mulching treatments, with average the normalized root mean square error of 16.5% in the validation period. There was no apparent difference in SNC, N2O emission, NH3 emission, and NL under different mulching durations in the seedling stage. However, the corresponding difference markedly increased with corn growth and development, especially in 2019. The average SNC (0–20 cm soil depth) under NM was higher by 9.2%, 25.0%, 21.2%, and 19.4% than PMS, PME, PMT, and PMW, respectively. Nitrogen emission (N2O and NH3) showed an increase trend with the shortening of mulching duration, i.e., NM>PMS>PME >PMT >PMW, while an opposite trend was found for NL. In general, PME was found to be the optimal mulching duration with the highest comprehensive evaluation score of 1782.7. Therefore, PME can be recommended to apply in this region to promote the developments of sustainable agriculture.

Given the low water and fertiliser use efficiency and the extensive distribution of sand interlayered soil in the Hetao irrigation district (HID), this study aimed to investigate the effects of different irrigation and fertilisation regimes on root parameters and yield in spring maize grown in sand interlayered soil. A two-year field plot experiment was conducted using the spring maize “Ximeng 3358” under three irrigation and nitrogen levels. Root length (RL), surface area (RS), diameter (RD), volume (RV), and length density (RLD), grain yield, and water use efficiency (WUE) were examined. Root growth was inhibited at the sand layer, with approximately 72.46–87.37% of the roots concentrated in the 0–40 cm soil layer. Notably, the proportion of roots in the bottom layer was 24.61–87.37% higher than that in the sub-bottom layer. Moreover, RL, RS, RD, and RV peaked in the medium irrigation and nitrogen fertilisation (I2F2) treatment. Furthermore, correlation analysis showed that the root parameters were significantly positively correlated with yield and WUE, with RS being most correlated to yield and WUE. Roots at a narrow row spacing of 20 cm (NR20) and at a depth of 10–20 cm were strongly correlated with yield and WUE. Conclusively, the I2F2 treatment can be used as the optimal combination of water and nitrogen for sand interlayered soil farmlands.

In order to investigate the influence of freshwater fish ponds on water and salt transport in cultivated wasteland in salinized areas, a typical study area was selected in the middle and lower reaches of the Hetao Irrigation District in China in the Yichang Irrigation Domain, and the temporal and spatial changes in the salinity of soil and salinity of groundwater and fish pond water in the cultivated–wasteland–fish pond system were characterized through the monitoring of the environmental information of soil and groundwater at the boundaries of the cultivated land, wasteland, and fish ponds. Salinity changes and groundwater migration in different periods were determined, and the response of soil salinity to the depth of groundwater burial was analyzed, as well as the effect of fish ponds on soil salinization. The results showed that the amount of groundwater migrating from cropland to wasteland during the simulation period in 2022 was 2700 m3, the amount of groundwater migrating from wasteland to fish ponds was 630 m3, and the amount of groundwater migrating from fish ponds to wasteland during the fall watering period was 440 m3. From an overall perspective, the average soil salinity of wasteland was 1.56 times higher than that of the boundary of fish ponds. Not only do fish ponds play a positive role in the ecosystem, but they also have a desalinization effect that reduces soil salinity significantly. Groundwater depth and soil salinity have an exponential relationship; when the depth of groundwater is greater than 1.75 m, the soil salinity varies little with the depth of groundwater and the soil salinity is less than 0.66 ds/m, which can be determined as the critical depth, and the average depth of groundwater in cultivated land in the study area is 1.5 m. Therefore, it is necessary to reasonably control the water level of fish ponds, so as to make the groundwater depth of cultivated land control between 1.75 m and 2.0 m, and to prevent soil salinization.

Efficient deep-water offshore wind power installation platforms with a pressurized self-elevating mat are a new type of equipment used for installing offshore wind turbines. However, the unstable internal pressure of the pressurized self-elevating mat can cause serious harm to the platform. This paper studies the pneumatic control system of the self-elevating mat to improve the precision of its pressure control. According to the pneumatic control system structure of the self-elevating mat, the pneumatic model of the self-elevating mat is established, and a conventional PID controller and fuzzy PID controller are designed and established. It can be seen via Simulink simulation that the fuzzy PID controller has a smaller adjustment time and overshoot, but its anti-interference ability is relatively weak. The membership degree and fuzzy rules of the fuzzy PID controller are optimized using a neural network algorithm, and a fuzzy neural network PID controller based on BP neural network optimization is proposed. The simulation results show that the overshoot of the optimized controller is reduced by 9.71% and the stability time is reduced by 68.9% compared with the fuzzy PID. Finally, the experiment verifies that the fuzzy neural network PID controller has a faster response speed and smaller overshoot, which improves the pressure control accuracy and robustness of the self-elevating mat and provides a scientific basis for the engineering applications of the self-elevating mat.

Soil salinization affects more than 25% of land globally. Subsurface pipe drainage is known for its effectiveness in improving saline–alkali land. The red clay layer (RCL) hinders soil improvement in the Hetao Irrigation District of Inner Mongolia, China. The soil water and salt migration rules at different buried depths and RCL were studied based on the field subsurface pipe drainage test and simulation using the DRAINMOD-S model (Version 6.1). The following implications can be drawn from the results: (1) Although the RCL affected the accuracy of the model, the calibrated statistical results met the application requirements, and the DRAINMOD-S model can be used to analyze subsurface pipe drainage under different distribution conditions of the RCL. (2) The RCL can reduce the drainage efficiency of the subsurface pipe, specifically when the distribution is shallow. (3) The soil desalting rate increased with an increase in the buried depth of the subsurface pipe. The desalination effect of shallow soil was better than that of deep soil. The RCL reduced the drainage and salt removal efficiency of the subsurface pipe. Burying the subsurface pipe as far above the RCL as possible should be considered. Thus, it is feasible to apply the DRAINMOD-S model to relevant studies.

Abstract Blockade of programmed cell death 1 ligand (PD-L1) has been used to treat triple-negative breast cancer (TNBC), and various strategies are under investigation to improve the treatment response rate. Inhibition of glutamine metabolism can reduce the massive consumption of glutamine by tumor cells and meet the demand for glutamine by lymphocytes in tumors, thereby improving the anti-tumor effect on the PD-L1 blockade therapy. Here, molybdenum disulfide (MoS2) was employed to simultaneously deliver anti-PDL1 antibody (aPDL1) and V9302 to boost the anti-tumor immune response in TNBC cells. The characterization results show that MoS2 has a dispersed lamellar structure with a size of about 181 nm and a size of 232 nm after poly (l-lysine) (PLL) modification, with high stability and biocompatibility. The loading capacity of aPDL1 and V9302 are 3.84% and 24.76%, respectively. V9302 loaded MoS2 (MoS2-V9302) can effectively kill 4T1 cells and significantly reduce glutamine uptake of tumor cells. It slightly increases CD8+ cells in the tumor and promotes CD8+ cells from the tumor edge into the tumor core. In vivo studies demonstrate that the combination of aPDL1 and V9302 (MoS2-aPDL1-V9302) can strongly inhibit the growth of TNBC 4T1 tumors. Interestingly, after the treatment of MoS2-aPDL1-V9302, glutamine levels in tumor interstitial fluid increased. Subsequently, subtypes of cytotoxic T cells (CD8+) in the tumors were analyzed according to two markers of T cell activation, CD69, and CD25, and the results reveal a marked increase in the proportion of activated T cells. The levels of cytokines in the corresponding tumor interstitial fluid are also significantly increased. Additionally, during the treatment, the body weights of the mice remain stable, the main indicators of liver and kidney function in the blood do not increase significantly, and there are no obvious lesions in the main organs, indicating low systemic toxicity. In conclusion, our study provides new insights into glutamine metabolism in the tumor microenvironment affects immune checkpoint blockade therapy in TNBC, and highlights the potential clinical implications of combining glutamine metabolism inhibition with immune checkpoint blockade in the treatment of TNBC. Abstract Background To assess the clinical outcomes after endovascular thrombectomy (EVT) in elderly large vessel occlusion (LVO)-related acute ischemic stroke (AIS) patients with atrial fibrillation (AF). Methods Between January 2019 and December 2020, consecutive AF patients who received EVT due to anterior-circulation stroke were enrolled. The primary outcome was modified Rankin scale (mRS) score at 90 days. Secondary outcomes included all-cause mortality, the recanalization status after EVT (assessed using modified thrombolysis in cerebral infarction scale, mTICI) and any intracranial hemorrhage (ICH). A multivariate logistic regression model was performed to identify predictors of the functional outcome. Results A total of 148 eligible patients were finally enrolled. Among them, 42 were ≥ 80 years old. Compared to their younger counterparts, patients aged ≥80 years had lower likelihood of good functional outcome (mRS score 0–2) at 90 days (26.2% vs. 48.1%, P = 0.015), less satisfied recanalization (mTICI, 2b-3) (78.6% vs. 94.3%, P = 0.004) and higher all-cause mortality rate (35.7% vs. 14.2%, P = 0.003). A multivariable logistic regression analysis showed that age ≥ 80 years at baseline were the significant predictors for a poor functional outcome (OR: 3.72, 95% CI: 1.17–11.89, p = 0.027). Intravenous thrombolysis (IVT) prior to EVT and longer time intervals from onset of symptoms to EVT tended to be associated with poor functional outcome in patients ≥80 years old. Conclusions Age ≥ 80 years was a significant predictor of unfavorable outcomes after EVT for AIS patients with AF. An increased risk of adverse events must be balanced against the benefit from EVT in elderly patients with AF.

Abnormal levels of intracellular hydrogen sulphide (H2S) have been associated with different pathological conditions, including cancer. Here the authors report the design of a H2S-responsive and -depleting nanoplatform that, combined with NIR-II photodynamic properties, can be used for H2S imaging and cancer therapy.

Controlling drainage during the growth stage is one of the means to provide suitable water and fertilizer conditions for crops, alleviate environmental pollution, and increase crop yield. Therefore, in this study, we studied three drainage treatments: free drainage (FD) and growth-stage subsurface controlled drainage (CD) at depths of 40 cm (CWT1) and 70 cm (CWT2). We used the HYDRUS-2D model to simulate the dynamic changes of NO3-N in the 0–100 cm soil layer as well as NO3-N uptake by crops, leaching after irrigation and fertilization, and loss through subsurface pipes in 2020 (model calibration period) and 2021 (model validation period). The degree of agreement between the simulated and measured values was high, indicating a high simulation accuracy. CD increased the soil NO3-N content and crop NO3-N uptake, and decreased NO3-N leaching and loss. We observed significant differences in the soil NO3-N content after irrigation at the budding stage of oilseed sunflower between CD and FD, with the largest difference seen for the 0–40 cm soil layer. CD increased crop yield, and the average oilseed sunflower yield of the CWT1 and CWT2 treatments increased by 4.52% and 3.04% relative to the FD treatment (p < 0.05). CD also enhanced nitrogen use efficiency. In moderately salinized soil, CD at 40 cm (CWT1) reduced the nutrient difference in vertical and horizontal directions while retaining water and fertilizer. CWT1 stabilized the groundwater depth, reduced the hydraulic gradient of groundwater runoff, and decreased the drainage flow rate. The NO3-N leaching and loss dropped, which promoted crop nitrogen uptake and utilization, improved nitrogen use efficiency, reduced nitrogen loss, and had a positive effect on protecting the soil and water environment. The results demonstrate that CD is a suitable drainage method for the experimental area.

The subsurface pipe drainage project is essential in farmland drainage operations and is globally recognized as an effective saline–alkali land improvement measure owing to its efficient drainage capacity and low land occupation rate. This study aimed to establish enhanced methods for improving saline–alkali land by combining ditching with subsurface pipe drainage. The ditching was conducted at a depth of 60 cm based on the existing subsurface pipe arrangement. The calibrated DRAINMOD-S model was employed to simulate the test area with different ditching depths and subsurface pipe arrangement parameters. Furthermore, the law of soil water and salt transport in the subsurface pipe drainage system at different ditching depths was investigated. After ditching, the total unit drainage volume of leaching increased by an average of 14.65% over two years and the water storage of different soil layers in the different plots decreased by 1.37–1.48 mm on average. Ditching demonstrated a superior salt-leaching effect in areas with subsurface pipe layouts. The soil desalination rate of different soil layers increased by 6.40–13.40% on average, with a more significant impact on the surface soil desalination rate. The effect of the increased desalination rate was more apparent as the ditching depth increased. However, as the buried depth of the subsurface pipe increased, the relationship between the ditching depth and soil desalination rate became insignificant. Ditching improved the salt-leaching effect of subsurface pipe drainage projects, which can effectively reduce the cost of subsurface pipe burial, consequently promoting subsurface pipe use.

The Hetao Irrigation District is a typical salinized irrigation district in China, and soil salinization restricts agricultural development. To explore the spatial and temporal variability of soil water and salt and the coupling relationship in the Hetao Irrigation District, a field experiment was carried out at the scale of the Yichang Irrigation District branch canal in the downstream of the Hetao Irrigation District. Fifty-three soil sampling points were established to analyze the spatial and temporal variability of soil water content and total salt content and the coupling relationship using geostatistics and the coupling degree model. The results showed that soil water content in the study area belonged to medium variability and weak variability, and soil total salt content belonged to strong variability and medium variability. The theoretical models of soil water content and total salt content semi-variance function in the study area following the Gaussian model, with the block-base ratio less than 25%, with strong spatial autocorrelation, and the spatial correlation gradually increased with the increase of soil depth. The total salt content of the soil in the study area was interpolated with higher accuracy using radial basis functions as compared to ordinary kriging interpolation. In terms of temporal changes in salinity, the average salt accumulation rate of the 0–100 cm soil layer in the study area was 20.17% when salinity increased from May to June; the average desalination rate was 16.37% when salinity decreased from June to August. The main factors affecting soil salinity in cultivated land during the growing period were irrigation, precipitation, and planting crops, and the main factors affecting soil salinity in wasteland were precipitation and topography. The average coupling degree of soil water and salt in wasteland in the study area was lower than that of cultivated land, ranging from 65.15% to 86.59% of that of cultivated land. The level of coordination is marginal coordination for cultivated land and marginal disorder for wasteland. The study provides a theoretical basis for the prevention and control of soil salinization in arid areas.

Background and purposePseudo-occlusion of the cervical internal carotid artery (cICA) refers to an absence of intraluminal contrast on computed tomography angiography (CTA), while the artery is patent on digital subtraction angiography during endovascular thrombectomy. We aimed to compare the outcomes between the cICA pseudo-occlusion and true occlusion after thrombectomy.MethodsWe retrospectively analyzed patients with apparent cICA occlusion on CTA who underwent thrombectomy between January 2016 and August 2021, and divided them into the pseudo-occlusion and true occlusion groups based on angiographic exploration. Recanalization failure was defined as a modified Thrombolysis in Cerebral Infarction score of 0–2a. Poor outcome was defined as a 90-day modified Rankin Scale score of 3–6. Propensity score matching was performed to compare the outcomes. Sensitivity analysis using multivariate-adjusted regression in the original cohort was conducted to test the robustness of the findings.ResultsOf the 146 patients included, 79 patients (54.1%) had cICA pseudo-occlusion and 67 patients (45.9%) had true occlusion. Following 1:1 propensity score-matched analysis, patients with pseudo-occlusion had an increased likelihood of recanalization failure (OR, 3.18; 95% CI, 1.06–9.59; P = 0.040) and poor outcome (OR, 2.80; 95% CI, 1.07–7.30; P = 0.035) compared with patients with true occlusion. Sensitivity analysis showed that cICA pseudo-occlusion remained independently associated with recanalization failure (OR, 2.55; 95% CI, 1.07–6.09; P = 0.036) and poor outcome (OR, 2.48; 95% CI, 1.08–5.67; P = 0.032).ConclusionsPatients with cICA pseudo-occlusion on CTA treated with thrombectomy had an increased risk of reperfusion failure and poor outcome compared with true occlusion patients.

Abstract Gas‐mediated sonodynamic therapy (SDT) has the potential to become an effective strategy to improve the therapeutic outcome and survival rate of cancer patients. Herein, titanium sulfide nanosheets (TiSX NSs) are prepared as cascade bioreactors for sequential gas–sonodynamic cancer therapy. TiSX NSs themselves as hydrogen sulfide (H2S) donors can burst release H2S gas. Following H2S generation, TiSX NSs are gradually degraded to become S‐defective and partly oxidized into TiOX on their surface, which endows TiSX NSs with high sonodynamic properties under ultrasound (US) irradiation. In vitro and in vivo experiments show the excellent therapeutic effects of TiSX NSs. In detail, large amounts of H2S gas and reactive oxygen species (ROS) can simultaneously inhibit mitochondrial respiration and ATP synthesis, leading to cancer cell apoptosis. Of note, H2S gas also plays important roles in modulating and activating the immune system to effectively inhibit pulmonary metastasis. Finally, the metabolizable TiSX NSs are excreted out of the body without inducing any significant long‐term toxicity. Collectively, this work establishes a cascade bioreactor of TiSX NSs with satisfactory H2S release ability and excellent ROS generation properties under US irradiation for programmed gas–sonodynamic cancer therapy.

ObjectivesThe aim of the study was to assess the effect of the stroke health management model on the prognosis and recurrence of mild to moderate ischemic stroke, guided by the stroke health manager based on the patients' needs. In addition, up-to-date evidence of healthcare resource allocation, planning, and optimization is provided.MethodsThe current research was a retrospective, observational, single-center, history-controlled study with patients divided into two groups, namely, the intervention group and the control group, following the guidance of the stroke health manager. The control group patients received standard medical care during hospitalization, which consisted of advice on healthy lifestyle choices carried out by the bed nurse, but no structured education, WeChat group, or clinical consultation was included. The intervention group patients, in addition to the standard medical care, received health management and health education from the stroke health manager, and after hospital discharge, the patients were followed up over the telephone by the health manager to see if there was any recurrence or readmission.ResultsFrom 1 January 2018 to 31 December 2020, 382 patients with acute ischemic stroke were enrolled in this study. Through the univariate regression analysis, we found that SHM intervention was associated with a significantly lower risk of recurrence (HR = 0.459). We constructed a nomogram based on the significant variables from the regression analysis and also analyzed the association between the control group and the SHM intervention group among all subgroups using the Cox proportional hazards model to assess the effect of the stroke health management model. Most patients in this study had a total risk point between 170 and 270. The C-index value was 0.76, and the time-dependent AUC for predicting recurrence was >0.7.ConclusionThe stroke health manager-guided management model based on patients' needs can better control the risk factors of stroke and significantly reduce the recurrence rate of mild to moderate ischemic stroke within 1 year.

The treatment of complex cerebrovascular diseases (CCVDs) at the skull base, such as complex intracranial aneurysms, carotid-cavernous sinus fistulas, and intracranial artery traumatic injuries, is a difficult clinical problem despite advances in endovascular and surgical therapies. Covered stents or stent graft insertion is a new concept for endovascular treatment that focuses on arterial wall defect reconstruction, differing from endovascular lesion embolization or flow diverter therapies. In recent years, covered stents specifically designed for cerebrovascular treatment have been applied in the clinical setting, allowing thousands of patients with CCVDs to undergo intraluminal reconstruction treatment and achieving positive results, even in the era of flow diverters. Since there is no unified reference standard for the application of covered stents for treating CCVDs, it is necessary to further standardize and guide the clinical application of this technique. Thus, we organized authoritative experts in the field of neurointervention in China to write an expert consensus, which aims to summarize the results of covered stent insertion in the treatment of CCVDs and propose suitable standards for its application in the clinical setting. Based on the contents of this consensus, clinicians can use individualized intraluminal reconstruction treatment techniques for patients with CCVDs.

Background and purposeFutile recanalization (FR), defined as functional dependence despite successful reperfusion, is common in patients who experience an acute stroke after thrombectomy. We aimed to determine the predictors of FR in patients who underwent thrombectomy in late time windows (6 h or more after symptom onset).MethodsThis retrospective review included patients who underwent thrombectomy for acute anterior circulation large vessel occlusion from October 2019 to June 2021. Successful reperfusion was defined as a modified Thrombolysis in Cerebral Infarction (mTICI) score of 2b/3. Functional dependence at 90 days was defined as a modified Rankin scale score of 3–6. Multivariate analysis and a receiver operating characteristic (ROC) curve were used to identify the predictors of FR in patients treated in delayed time windows.ResultsOf the 99 patients included, FR was observed in 51 (51.5%). In the multivariate analysis, older age (OR, 1.12; 95% CI, 1.04–1.22; P = 0.005), female sex (OR, 3.79; 95% CI, 1.08–13.40; P = 0.038), a higher National Institutes of Health Stroke Score (NIHSS) score upon admission (OR, 1.11; 95% CI, 1.02–1.22; P = 0.023), and an increased number of passes per procedure (OR, 2.07; 95% CI, 1.11–3.86; P = 0.023) were independently associated with FR after thrombectomy. The ROC curve indicated that the model that combined age, female sex, baseline NIHSS score, and the number of passes per procedure (area under the curve, 0.84; 95% CI, 0.75–0.90, P < 0.001) was able to predict FR accurately.ConclusionsOlder age, female sex, higher NIHSS score upon admission, and an increased number of passes per procedure were independent predictors of FR in patients who experienced acute ischemic strokes after thrombectomy in late time windows.

Transdermal biosensing focuses on the qualitative and quantitative collection and detection of biomarkers on and in the skin layers. In comparison to the blood‐based diagnosis, transdermal sensing technologies provide pain‐free, convenient, patient‐friendly, and infection‐avoiding methods to collect biosignals from transdermal samples such as interstitial fluid (ISF) and sweat. This article summarizes the latest trends in transdermal biosensing including the technologies and their application blueprints. Specifically, the current transdermal biosensing technologies including microneedles, microfluidics, and reverse iontophoresis are introduced first. Subsequently, their roles in daily health status monitoring, disease diagnosis and therapy monitoring are discussed in detail. Finally, an overview of the future blueprints and application prospects of transdermal biosensing is provided.

Abstract Background Anti-angiogenic therapy has been widely applied to the clinical treatment of malignant tumors. However, the efficacy of such treatments has been called into question, especially in triple-negative breast cancer (TNBC). Bevacizumab, the first anti-angiogenic agent approved by FDA, actually increases invasive and metastatic properties of TNBC cells, resulting from the activation of Wnt/β-catenin signaling in response to hypoxia. As a critical receptor of Wnt/β-catenin signaling, Frizzled-7 (Fzd7) is aberrantly expressed in TNBC, indicating Fzd7 a potential target for developing drugs to be combined with anti-angiogenic agents. Methods Hybridoma technique and antibody humanization technique were utilized to generate a Fzd7-targeting antibody (SHH002-hu1). Biolayer interferometry (BLI) assay and near infrared (NIR) imaging were conducted to detect the affinity and targeting ability of SHH002-hu1. Next, whether SHH002-hu1 could suppress the invasion and migration of TNBC cells induced by Bevacizumab were validated, and the underlying molecular mechanisms were elucidated by luciferase reporter and western blot assays. The nude-mice transplanted TNBC models were established to assess the anti-TNBC activities of SHH002-hu1 when combined with Bevacizumab. Then, the effects on putative TNBC stem-like cells and Wnt/β-catenin signaling were evaluated by immunofluorescence (IF). Further, the tumor-initiating and self-renew capacity of TNBC cells were studied by secondary nude mouse xenograft model and sphere formation assay. In addition, the effects of SHH002-hu1 on the adaptation of TNBC cells to hypoxia were evaluated by the detection of vasculogenic mimicry (VM) and hypoxia-inducible factor-1α (HIF-1α) transcriptional activity. Results The novel humanized antibody targeting Fzd7 (SHH002-hu1) exhibited extremely high affinity with Fzd7, and specifically targeted to Fzd7+ cells and tumor tissues. SHH002-hu1 repressed invasion, migration and epithelial-mesenchymal cell transformation (EMT) of TNBC cells induced by Bevacizumab through abating Wnt/β-catenin signaling. SHH002-hu1 significantly enhanced the capacity of Bevacizumab to inhibit the growth of TNBC via reducing the subpopulation of putative TNBC stem-like cells, further attenuating Bevacizumab-enhanced tumor-initiating and self-renew capacity of TNBC cells. Moreover, SHH002-hu1 effectively restrained the adaptation of TNBC cells to hypoxia via disrupting Wnt/β-catenin signaling. Conclusion SHH002-hu1 significantly enhances the anti-TNBC capacity of Bevacizumab, and shows the potential of preventing TNBC recurrence, suggesting SHH002-hu1 a good candidate for the synergistic therapy together with Bevacizumab.

Biodegradable mulches have the same temperature- and moisture-preservation effects as ordinary plastic mulches before degradation. After degradation, rainwater enters the soil through the damaged parts, improving precipitation utilization. Under drip irrigation with mulching, this study explores precipitation utilization of biodegradable mulches under different precipitation intensities and the effects of different biodegradable mulches on the yield and water use efficiency (WUE) of spring maize in the West Liaohe Plain, China. In this paper, in situ field observation experiments were conducted for three consecutive years from 2016 to 2018. Three types of white degradable mulch films were set up, with induction periods of 60 d (WM60), 80 d (WM80), and 100 d (WM100). Three types of black degradable mulch films were also used, with induction periods of 60 d (BM60), 80 d (BM80), and 100 d (BM100). Precipitation utilization, yield, and WUE under biodegradable mulches were studied, with ordinary plastic mulches (PM) and bare land (CK) set as controls. The results showed that as precipitation increased, the effective infiltration of precipitation decreased first and then increased. When precipitation reached 89.21 mm, plastic film mulching no longer affected precipitation utilization. Under the same precipitation intensity, the precipitation effective infiltration ratio increased as the damage to the biodegradable film increased. Still, the intensity of this increase gradually decreased as the damage increased. The highest yield and WUE were observed for the degradable mulch film with an induction period of 60 days in years with normal rainfall and for the degradable mulch film with an induction period of 100 days in dry years. In the West Liaohe Plain, maize planted under film receives drip irrigation. We recommend that growers select a degradable mulch film with a degradation rate of 36.64% and an induction period of approximately 60 days in years with normal rainfall, and a degradable mulch film with an induction period of 100 days in dry years.

The impact of different combinations of water–nitrogen coupling on maize yield and the environment needs investigation. Low, medium, and high levels of irrigation and N application gradients were studied through field experiments to elucidate the suitable water–nitrogen coupling zone for spring maize in the West Liaohe Plain during three hydrological year patterns under drip irrigation with plastic film. The effects of different water–nitrogen couplings on maize yield, water- and nitrogen-use efficiencies (WUE and NUE), and post-harvest soil alkali-hydrolyzable N residues were studied under integrated drip irrigation by varying the application rates of water and fertilizer. A multi-objective optimization of water–nitrogen coupling zones was performed by integrating maize yield, harvest index, WUE, and soil environmental effects. Results show that with an increase in irrigation and N application rate, the residual amount of alkali-hydrolyzable N increased slowly within a certain range. Upon exceeding a certain amount, residual N increased rapidly, and more N entered the soil environment. The NUE of moderate water–nitrogen coupling treatment was high, with lower environmental risk of residual alkali-hydrolyzable N. Moderate irrigation yielded the highest harvest index in the normal hydrological year. Irrigation rate had a higher impact on yield compared to nitrogen application, because of drip irrigation under plastic film. An appropriate irrigation amount results in a higher WUE and the application of N application must be adjusted according to the rainfall in a particular year. This study highlights the need for structuring water–nitrogen coupling zones specifically for different hydrological years.

While antiretroviral drugs have transformed the lives of HIV-infected individuals, chronic treatment is required to prevent rebound from viral reservoir cells. People living with HIV also are at higher risk for cardiovascular and neurocognitive complications, as well as cancer. Finding a cure for HIV-1 infection is therefore an essential goal of current AIDS research. This review is focused on the discovery of pharmacological inhibitors of the HIV-1 Nef accessory protein. Nef is well known to enhance HIV-1 infectivity and replication, and to promote immune escape of HIV-infected cells by preventing cell surface MHC-I display of HIV-1 antigens. Recent progress shows that Nef inhibitors not only suppress HIV-1 replication, but also restore sufficient MHC-I to the surface of infected cells to trigger a cytotoxic T lymphocyte response. Combining Nef inhibitors with latency reversal agents and therapeutic vaccines may provide a path to clearance of viral reservoirs.

Merkel cell polyomavirus (MCV or MCPyV) is an alphapolyomavirus causing human Merkel cell carcinoma and encodes four tumor (T) antigen proteins: large T (LT), small tumor (sT), 57 kT, and middle T (MT)/alternate LT open reading frame proteins. We show that MCV MT is generated as multiple isoforms through internal methionine translational initiation that insert into membrane lipid rafts. The membrane-localized MCV MT oligomerizes and promiscuously binds to lipid raft-associated Src family kinases (SFKs). MCV MT-SFK interaction is mediated by a Src homology (SH) 3 recognition motif as determined by surface plasmon resonance, coimmunoprecipitation, and bimolecular fluorescence complementation assays. SFK recruitment by MT leads to tyrosine phosphorylation at a SH2 recognition motif (pMTY114), allowing interaction with phospholipase C gamma 1 (PLCγ1). The secondary recruitment of PLCγ1 to the SFK-MT membrane complex promotes PLCγ1 tyrosine phosphorylation on Y783 and activates the NF-κB inflammatory signaling pathway. Mutations at either the MCV MT SH2 or SH3 recognition sites abrogate PLCγ1-dependent activation of NF-κB signaling and increase viral replication after MCV genome transfection into 293 cells. These findings reveal a conserved viral targeting of the SFK-PLCγ1 pathway by both MCV and murine polyomavirus (MuPyV) MT proteins.
