Infertility has become one of the most common chronic diseases among reproductive- aged individuals, and male factors account for about 50 %. Zinc is a trace element that is essential for sperm quality and male reproductive system. Although several current studies with small sample sizes have investigated this relationship, the conclusions have been still controversial. Here, using a large population sample involved 25,915 participants from Changzhou Maternity and Child Health Care Hospital, we revealed an inverted “U”-shaped trend between seminal plasma zinc concentrations and sperm motility PR/PR + NP (PR, progressive motility; NP, non-progressive motility). The results showed that the highest values of sperm PR/PR + NP observed in the group with intermediate concentrations of zinc (Group 2, 0.25–2.11 mmol/L). The mean values were 43.17 ± 19.03 % and 56.64 ± 20.28 %, respectively. And the lowest values came out in the highest zinc levels group (Group 4, > 3.04 mmol/L). In vitro cell experiments also showed that zinc caused dose-dependent cytotoxicity for GC-2 cells at a threshold value. Furthermore, RNA-seq analysis demonstrated that high concentrations of zinc exerted toxic effects on GC-2 cells through immune injury. Taken together, our findings suggested that moderate amounts of zinc are crucial for human reproduction and excessive concentrations may have adverse effects on male fertility.

Drying is an important means to maintain the quality of peanut pods and reduce the risk of mildew. Microwave hot air combined drying is renowned for its efficacy, low-energy consumption, and superior processing quality, rendering it suitable for peanut pod drying. However, its efficacy hinges greatly on various parameters. To enhance drying efficiency and quality, the drying kinetics of peanut pods using microwave hot air combined drying equipment were investigated. Varying microwave power (1.2 kW, 2.4 kW, 3.6 kW), hot air temperature (30 °C, 35 °C, 40 °C, 45 °C), air velocity (0.4 m/s, 0.7 m/s, 1 m/s), and microwave radiation time (4 min, 6 min) were examined. Results indicated a decline in drying time, moisture ratio, and drying rate with increasing microwave power, hot air temperature, air velocity, and microwave radiation time. Peroxide and acid values complied with national standards. The Verma model exhibited optimal fitting. At 45 °C, peanut pod moisture diffusion coefficient was 3.022 × 10−9 m2 s−1, specific energy consumption at 30 °C was 5302 kJ/kg, and thermal efficiency was 42.6 %. The synergistic drying of peanut pods by microwave and hot air has a high moisture diffusion coefficient and thermal efficiency. The kinetic and energy analysis of the drying process can furnish some theoretical foundation for subsequent microwave hot air drying and equipment parameter adjustment.

IntroductionDespite the rapid growth of interdisciplinary resilience research in Chinese contexts, no study has systematically reviewed individual-level measurement scales for Chinese-speaking populations. We report a systematic review of scales developed for or translated/adapted to Chinese-speaking contexts, where we assessed how widely used scales fare in terms of their psychometric qualities.MethodsStudies included in this review must have been published in peer-reviewed English or Chinese journals between 2015-2020 and included self-reported resilience scales in Chinese-speaking populations. Searches were conducted in PsycINFO, CNKI (completed in May 2021), and PubMed (completed in January 2024). We developed coding schemes for extracting relevant data and adapted and applied an existing evaluation framework to assess the most frequently used resilience scales by seven methodological criteria.ResultsAnalyses of 963 qualified studies suggested that Chinese resilience scales were used in a diverse range of study contexts. Among 85 unique kinds of resilience measures, we highlighted and evaluated the three most frequently used translated scales and three locally developed scales (nine scales in total including variations such as short forms). In short, resilience studies in Chinese contexts relied heavily on the translated 25-item Connor-Davidson Resilience Scale, which scored moderately on the overall quality. The locally developed Resilience Scale for Chinese Adolescents and Essential Resilience Scale received the best ratings but could use further development.DiscussionWe discussed how future work may advance widely used scales, and specified seven methodological recommendations for future resilience scale development with existing and new scales in and beyond the Chinese study contexts. We further addressed issues and challenges in measuring resilience as a process and called on researchers to further develop/evaluate process measures for Chinese-speaking populations.

Peanut is an important oil crop and cash crop, with a wide range of applications in many fields such as the food industry, light industry, and chemical industry. Mechanized shelling is a necessary part of the post-production processing of peanuts, and it is also the key to determining the quality of peanut products. Reducing shelling damage is an effective way to improve the quality and comprehensive benefits of peanut products. Consequently, it is of great significance to strengthen the research on damage reduction in mechanized peanut-shelling. China is a large peanut producer, but the research on mechanized shelling started relatively late, and the existing technology is not compatible with the high-quality shelling requirements of farmers. This paper reviews the status of mechanized peanut-shelling technology, compares the technical characteristics and equipment development of the world’s important peanut producing countries, it summarizes and proposes the suggestions to reduce loss from the aspects of varieties, agronomy, technology, and technical equipment; further deepen innovative research; and strengthen the construction of peanut-shelling socialized service systems. It is expected to provide reference for effectively reducing damage and improving quality of China’s mechanized shelling, and promoting the sustainable development of peanut shelling industry.

In order to investigate the influence of fracture attitude variation on the permeability of fractures in deep rock mass under natural crustal stresses, based on the permeability law of a single fracture obeying the law of negative exponential change, the theoretical formula of coefficient of permeability of fractures with variable attitude subjected to 3D stresses is proposed. The Lagrange multiplier method is applied to analyze the change of variable attitude on fracture permeability, and influence factors and sensibility of fracture permeability with or without infillings are also analyzed. Statistic law of crustal stress in Chinese mainland is taken as an example, and the variable laws of the permeability of fractures in rock mass with depth and fracture attitude are analyzed. The results show that (1) the permeability of fractures in rock mass is obviously influenced by the attitude of fractures. For shallow rock mass, under the condition of the maximum principle stress distribution in the horizontal direction, the permeability of fractures gradually decreases with the increasing dip angle of fractures; but with the increasing depth, when the depth is more than 200 m and the strike of fracture is roughly consistent with the direction of large or medium principal stress, the permeability of fractures can increase with the increasing dip angle of fractures, especially the strike of fractures perpendicular to the direction of the minimum principle stress. For deep rock mass, the permeability of fracture is very weak, and the impact of fracture attitude change on its permeability is very slight. (2) For fractures with infillings, the change of elastic modulus ratio of rock block to infillings and Poisson’s ratio of infillings have little effect on fracture permeability. The conclusions can be used for reference for further investigating the variation of deep rock permeability characteristics.

There exist challenges in quantifying the length of individual telomeres at single nucleotide resolution. Here the authors report a method to capture telomeres and accurately analyse their length in human cell lines and patient peripheral blood leukocyte samples using single-molecule real-time sequencing.

Rapid drying of peanut is very important to maintain quality and reduce the risk of mildew, especially for peanut harvested in high moisture content in China. The modified mixed-flow dryer can dry freshly harvested peanut efficiently and evenly which is not available for traditional fixed-bed drying. However, the performances of energy and exergy of peanut drying using modified mixed-flow dryer are not clear, lacking of relevant data for equipment improvement and process formulation. Therefore the relevant investigation was carried out to evaluate the performance of fresh harvested peanut drying using industrial mixed-flow dryer with heat-pump full power heating and waste heat recovery The results show that the energy consumption is mainly consumed by heat-pump and centrifugal fan, accounting for 73.5% and 24.2% of all energy consumed respectively. The overall specific energy consumption and specific heat consumption are 1494.5 kJ/kg and 4706.1 kJ/kg respectively. The thermal efficiency and the energy efficiency are affected by the change of the temperature of airflow entering the drying chamber, both fluctuate over time, increase earlier then decrease later and rise again until the material temperature reach to a relatively steady temperature. And the energy efficiency of dryer (178.9%) is approximately triple of thermal efficiencies of drying chamber (56.1%) and dryer (59.4%) for the overall drying. The heat utilization ratio decreases gradually over drying time, with the overall value of 50.0% for drying chamber and 52.9% for dryer. The exergy efficiency of drying process earlier increases then decreases in the overall trend with the overall value of 51.3%. The exergy efficiencies of the drying chamber and whole dryer are both increased gradually with the overall value of 34.5% and 31.8% respectively. However, the variation tendencies of energy improvement potential rates are opposite to exergy efficiencies.

Golden cocoon silk fibers suffer a lot from limited yield and serious color losing, which greatly restrict their uses. The development of golden cocoon-like silk fibers can effectively address the above issues. Herein, a low-dosage chemicals, short process alternative approach to reactive dyeing of golden cocoon-like silk fibers with robust color fastness was proposed. Investigation involves the pretreatment of the above silk via one-bath method followed by reactive dyeing. The results indicate that the color of as-prepared golden cocoon-like silk fiber is comparable with the commercial golden cocoon one. The short-process dyeing and robust color fastness with reactive dyes of silk fiber can be realized via the introduction of alkali. Importantly, no obvious color degradation following cyclic washing was observed, with the value of color difference within 2.0. Additionally, the tensile behavior of reactive dyed golden cocoon-like silk fiber was not significantly decreased as compared with the raw fiber.

Graphene has a high electric heating conversion rate and can be used as a new heat source for far-infrared drying equipment to improve drying efficiency and reduce energy consumption. This study aims to investigate the drying kinetics of graphene far-infrared paddy drying equipment The infrared (IR) temperatures of 35, 40, 45, 50, 55, and 60 °C; distances between radiations of 25, 30, 35, and 40 mm; grain flow of 5.0, 6.0, 7.0, 8.0, and 9.0 kg/min; and dehumidifying air velocities of 4, 5.5, 7, and 8.5 m/s were studied for appropriate drying time and optimal paddy quality. Simultaneously, the drying kinetics, moisture diffusion coefficient, and activation energy were observed. Experimental results show that the coupling degree of Midilli was superior, specifically, the Deff of paddy was 4.83–16.371 × 10−9 m2/s, an activation energy of 47.44 kJ mol−1 and a heat efficiency of 71.11% were achieved at a drying temperature of 60 °C. The far infrared drying of graphene has high moisture diffusion coefficient and thermal efficiency. By analyzing the kinetics and energy of drying process, a basis for formulating drying strategies could be provided to realize the research and development of new drying machinery.

Hypertrophic scars (HSs) develop due to excessive collagen deposition and abnormal fibroblast proliferation during wound healing, significantly impacting patient quality of life. Three dosages of GA ointments were administered to rabbit ear HS models to investigate the potential efficacy and mechanism of gallic acid (GA) on HS. Daily application of ointment was performed on the matrix group, the GA ointment groups, and the silicone gel group for 28 days. (No drug treatment was performed on the skin and model groups as a blank group and vehicle group, and silicone gel ointment was topically administered to the silicone gel group as a positive control group.) Scar specimens were collected for histopathology analysis, RNA sequencing analysis, real-time quantitative polymerase chain reaction, and Western blot analysis at the first, second, and fourth weeks after the treatment. Low-dose and medium-dose GA effectively suppressed HS formation and markedly decreased fibroblast infiltration levels and scar thickness. Moreover, decreased expression of TRPC3 mRNA and TGF-β1, p-Smad2/3, and Smad2/3 protein was observed in the low- and medium-dose GA groups and the silicone gel group. This study provides evidence for the efficacy of GA in treating HS and sheds light on its potential underlying pharmacological mechanisms.

A box-type hot-air convection dryer integrated with parallel vertical fans, multiple air deflectors, a rack moving mechanism, and an opening heat pump system was designed and used. The airflow distributions in the inlet surface of the air-inlet chamber and drying chamber under empty-loading conditions were measured and the results showed excellent uniformities. The air temperature distributions at the inlet and outlet surfaces of the drying chamber, wet basis moisture content of the materials, energy consumption, product loss rate, and processing capacity were measured and analyzed as well under the loading of longans, lychees, and shiitake mushrooms. The results indicated that the maximum temperature difference at air-inlet surface of drying chamber was 4 °C. The dehydration rate was fast, and the moisture content of the wet basis decreased slowly in the earlier half stage of drying. However, the dehydration rate was slow, and the moisture content decreased at an accelerated rate in the latter half stage of drying. The higher the initial moisture of the material, the more obvious the above phenomenon. Further, the moisture uniformity of dried products reached 90%, with no products of mildewing and obviously insufficient drying found, which would support technique for the drying of fruits and vegetables.

A cell’s size affects the likelihood that it will die. But how is cell size controlled in this context and how does cell size impact commitment to the cell death fate? We present evidence that the caspase CED-3 interacts with the RhoGEF ECT-2 in Caenorhabditis elegans neuroblasts that generate “unwanted” cells. We propose that this interaction promotes polar actomyosin contractility, which leads to unequal neuroblast division and the generation of a daughter cell that is below the critical “lethal” size threshold. Furthermore, we find that hyperactivation of ECT-2 RhoGEF reduces the sizes of unwanted cells. Importantly, this suppresses the “cell death abnormal” phenotype caused by the partial loss of ced-3 caspase and therefore increases the likelihood that unwanted cells die. A putative null mutation of ced-3 caspase, however, is not suppressed, which indicates that cell size affects CED-3 caspase activation and/or activity. Therefore, we have uncovered novel sequential and reciprocal interactions between the apoptosis pathway and cell size that impact a cell’s commitment to the cell death fate. This study shows that in developing C. elegans neuroblasts, the caspase CED-3 interacts with the RhoGEF ECT-2, leading to changes in actomyosin and the unequal division of these cells. This reveals a non-canonical function of caspases, wherein they help establish ensure that the size of daughter cells fated for apoptosis is below a critical ’lethal’ threshold.

Valproic acid/sodium valproate (VPA) is a widely used anticonvulsant drug for maintenance treatment of bipolar disorders. In order to balance the efficacy and adverse events of VPA treatment, an individualized dose regimen is necessary. This study aimed to establish an individualized medication model of VPA for patients with bipolar disorder based on machine learning and deep learning techniques. The sequential forward selection (SFS) algorithm was applied for selecting a feature subset, and random forest was used for interpolating missing values. Then, we compared nine models using XGBoost, LightGBM, CatBoost, random forest, GBDT, SVM, logistic regression, ANN, and TabNet, and CatBoost was chosen to establish the individualized medication model with the best performance (accuracy = 0.85, AUC = 0.91, sensitivity = 0.85, and specificity = 0.83). Three important variables that correlated with VPA daily dose included VPA TDM value, antipsychotics, and indirect bilirubin. SHapley Additive exPlanations was applied to visually interpret their impacts on VPA daily dose. Last, the confusion matrix presented that predicting a daily dose of 0.5 g VPA had a precision of 55.56% and recall rate of 83.33%, and predicting a daily dose of 1 g VPA had a precision of 95.83% and a recall rate of 85.19%. In conclusion, the individualized medication model of VPA for patients with bipolar disorder based on CatBoost had a good prediction ability, which provides guidance for clinicians to propose the optimal medication regimen.

Hypertrophic scar (HS) is a typical pathological response during skin injury, which can lead to pain, itching, and contracture in patients and even affect their physical and mental health. The complexity of the wound healing process leads to the formation of HS affected by many factors. Several treatments are available for HS, whereas some have more adverse reactions and can even cause new injuries with exacerbated scarring. Traditional Chinese Medicine (TCM) has a rich source, and most botanical drugs have few side effects, providing new ideas and methods for treating HS. This paper reviews the formation process of HS, the therapeutic strategy for HS, the research progress of TCM with its relevant mechanisms in the treatment of HS, and the related new drug delivery system of TCM, aiming to provide ideas for further research of botanical compounds in the treatment of HS, to promote the discovery of more efficient botanical candidates for the clinical treatment of HS, to accelerate the development of the new drug delivery system and the final clinical application, and at the same time, to promote the research on the anti-HS mechanism of multiherbal preparations (Fufang), to continuously improve the quality control and safety and effectiveness of anti-HS botanical drugs in clinical application.

Unsteady one-dimensional partial differential equations for heat and mass transfer in peanut drying are used in mathematical simulations to compare the drying performance of a barrel-shaped bed in three ventilation modes: bottom up, inside out and outside in. Results obtained using the finite difference scheme show that the mathematical simulation reliably predicts the drying process of peanuts, and that the drying delay of bottom-up ventilation is the most significant at the beginning of drying. This is because the drying enhancement afforded by the high average velocity of air along the ventilation direction is insufficient to offset the drying delay caused by the large ventilation thickness compared with that afforded by the inside-out and outside-in ventilation. Furthermore, as the air volume flow increases, the time consumption and moisture content difference decrease, whereas the productivity, energy consumption, and uniformity increase for all three ventilation modes, consistent with the classical ventilation drying law. Under the same air volume flow, the time and energy consumptions for the bottom-up ventilation are the lowest, followed by those for the outside-in and inside-out ventilation. The productivity afforded by the bottom-up ventilation is the highest, followed by that by the outside-in and inside-out ventilation. The outside-in ventilation indicates the least moisture content difference and the best drying uniformity. Meanwhile, the inside-out ventilation indicates the most significant moisture content difference and the worst drying uniformity.

In order to assess the effectiveness of reconstructed energy dissipation facilities (EDFs) in open channels at hydropower stations, hydraulic prototype observation (HPO) tests are conducted to investigate the characteristics of discharge flow and the dynamic response of hydraulic structures during sluice opening periods. While hydraulic model tests (HMTs) are commonly utilized in laboratory settings to study these characteristics, experimental conditions cannot fully replicate the real-world operations of such structures. HPO tests are employed to examine flow patterns, free water surface fluctuations, and pulsating pressure changes in open channels under varying flood discharge conditions (FDCs). Flow patterns in open channels are recorded via video; free water surface fluctuations are measured using total-station and laser rangefinder instruments; and pulsating pressure is monitored with pressure sensors and data collection systems. Flow pattern observations concentrate on addressing adverse water flow phenomena, such as turbulence, surging, and backflow. The examination of free water surface fluctuations aims to verify whether the height of the guide wall along the open channel fulfills the necessary requirements and assess the effectiveness of energy dissipation of the EDF. To comprehend the variations in pulsating pressure within the continuous sill and the base slab, nine measurement points were established across three sections perpendicular to the continuous sill’s axis on three distinct elevation levels. Additionally, three measurement points were positioned on the reinforced base slab along the open channel’s axis. The findings indicate that the impact on the continuous sill caused by discharging water is more severe when the discharge rate of a single sluice gate reaches 500 m3/s than in other FDCs. To ensure the safe operation of open channels during flood discharge, the discharge rate for each sluice gate should be reduced to 250 m3/s. The dominant pulsation induced by discharge flow falls within the low-frequency range, resulting in minimal adverse effects on the stilling basin and guide wall. The flow pattern within the stilling basin remains stable under various FDCs, with no significant adverse hydraulic phenomena observed. Parameters, including free water surface fluctuations and pulsating pressure variations, lie within acceptable ranges. These observations suggest that the arrangement of the reconstructed energy dissipation facilities is generally effective following technical reconstruction.

Lapatinib is used for the treatment of metastatic HER2(+) breast cancer. We aim to establish a prediction model for lapatinib dose using machine learning and deep learning techniques based on a real-world study. There were 149 breast cancer patients enrolled from July 2016 to June 2017 at Fudan University Shanghai Cancer Center. The sequential forward selection algorithm based on random forest was applied for variable selection. Twelve machine learning and deep learning algorithms were compared in terms of their predictive abilities (logistic regression, SVM, random forest, Adaboost, XGBoost, GBDT, LightGBM, CatBoost, TabNet, ANN, Super TML, and Wide&Deep). As a result, TabNet was chosen to construct the prediction model with the best performance (accuracy = 0.82 and AUC = 0.83). Afterward, four variables that strongly correlated with lapatinib dose were ranked via importance score as follows: treatment protocols, weight, number of chemotherapy treatments, and number of metastases. Finally, the confusion matrix was used to validate the model for a dose regimen of 1,250 mg lapatinib (precision = 81% and recall = 95%), and for a dose regimen of 1,000 mg lapatinib (precision = 87% and recall = 64%). To conclude, we established a deep learning model to predict lapatinib dose based on important influencing variables selected from real-world evidence, to achieve an optimal individualized dose regimen with good predictive performance.

The ferroelectricity in hybrid perovskite CH3NH3PbI3 is under debate. Here, the authors predict a Pb-free hybrid perovskite N(CH3)4SnI3 with non-polar molecular cation N(CH3)4+ showing strong ferroelectricity, good stability, and high visible-light absorption, ideal for high-efficiency solar cells.

Methods to control protein degradation usually require prior genetic engineering of each specific protein of interest. Here the authors present a toolbox for targeted depletion of endogenous proteins in cells and organisms, which is based on light- and drug-controlled recombinant E3 ubiquitin ligases.

In order to solve problems such as poor applicability of headers, weak separation ability of threshing mechanisms and poor impurity-removal ability of cleaning devices in the existing seed harvest methods of Chinese milk vetch (Astragalus sinicus L.), a combined Chinese milk vetch seed harvester was designed in this paper. The parameters of the key components, such as the flexible anti pod-dropping seedling-lifting header, the longitudinal rod-teeth-type threshing device and the air-sieve-type layered impurity-controlled cleaning device, were designed and optimized. Aiming at reducing seed loss rate, breakage rate and impurity rate of Chinese milk vetch during the mechanical harvesting process, through multi-parameter optimization, the best combination of working parameters was obtained: machine forward speed was 3 km·h−1, rotation speed of the threshing drum was 550 r·min−1, rotation speed of the cleaning fan was 990 r·min−1 and the scale sieve’s opening was 35 mm. Field tests were performed under these parameters, and the results showed that the seed loss rate of Chinese milk vetch was 2.35%, the breakage rate was 0.22% and the impurity rate was 0.51%, which were better than the technical requirements of loss rate and breakage rate less than 5% and impurity rate less than 3% specified in relevant standards. The research results can solve the shortage problem of efficient seed harvest equipment in large-scale planting areas of Chinese milk vetch, and will further help to carry out seed harvest experiments on different varieties of Chinese milk vetch and other green manure varieties in paddy fields.

There is a pressing need for an automatic road extraction method due to the continuous development of transportation networks. Free from the influence of weather, satellite-mounted synthetic-aperture radar (SAR) opens the way for such a road detection application. This study introduces an automatic discrimination method based on a deep neural network (DNN) adjusted for roads from dual-polarimetric (VV and VH) Sentinel-1 SAR imagery. In this proposed method, the U-Net extended the convolutional neural network (CNN), is adjusted for road extraction from SAR images. To investigate the potential of using the U-Net based fully convolutional neural network (FCN) for road extraction, LeNet-5, is applied as a preliminary DNN model. Additionally, several training optimizations are introduced to improve accuracy. To assess the performance of the different polarization modes used in road extraction, both single-polarimetric and dual-polarimetric data were investigated. Moreover, four machine learning algorithms have been compared for accuracy and speed. As a result, the outcome evaluation of Precision, Recall, and F1 obtained by FCN is better than the original CNN, and the training time has been significantly reduced. The DNN model (CNN and FCN) is superior to machine learning methods in accuracy and elapsed computation time.

The quality of walnuts deteriorates owing to the poor quality of the shelling equipment. The improvement of shelling quality is urgently required for walnut processing. In this study, systematic research was carried out on the changes in walnut mechanical properties, mechanical model of walnut shelling, and the key parameters of the equipment. The key parameters were determined as the angles of the shelling conic roller (X1), speeds of the shelling conic roller (X2), clearance between the shelling conic roller and the static roller (X3), and the moisture content of walnuts (X4). The Box–Behnken design method was used for the experimental design, an analysis of variance was applied to determine the graded significance of each variable on the rate of high-quality kernel (RHQK) and rate of shell rushing (RSC), and the multi-objective optimization method was used to obtain the optimal parameters for RHQK and RSC. The ranking of factors affecting RHQK and RSC were: (X3) > (X4) > (X2) > (X1) for RHQK, and (X2) > (X3) > (X4) > (X1) for RSC. The ranks of significant interactive effects among the factors were as follows: (X1 X2) > (X2 X3) for RHQK and (X2 X3) > (X3 X4) > (X2 X4) for RSC. The multi-objective optimization results showed that the optimal combination was X1 = 15.83°, X2 = 17.93 rpm, X3 = 45 mm, and X4 = 9.5%, yielding RHQK = 84.54%, and RSC = 99.15%. The verification test of the optimal results further illustrates the accuracy of the optimization. The obtained results showed that the quality of walnut shelling can be improved by adjusting the moisture content of walnuts and optimizing key parameters of the equipment. This method also represents a potential solution for improving the shelling quality of other nuts.

The mechanism(s) through which mammalian kinase MELK promotes tumorigenesis is not understood. We find that the C. elegans orthologue of MELK, PIG-1, promotes apoptosis by partitioning an anti-apoptotic factor. The C. elegans NSM neuroblast divides to produce a larger cell that differentiates into a neuron and a smaller cell that dies. We find that in this context, PIG-1 MELK is required for partitioning of CES-1 Snail, a transcriptional repressor of the pro-apoptotic gene egl-1 BH3-only. pig-1 MELK is controlled by both a ces-1 Snail- and par-4 LKB1-dependent pathway, and may act through phosphorylation and cortical enrichment of nonmuscle myosin II prior to neuroblast division. We propose that pig-1 MELK-induced local contractility of the actomyosin network plays a conserved role in the acquisition of the apoptotic fate. Our work also uncovers an auto-regulatory loop through which ces-1 Snail controls its own activity through the formation of a gradient of CES-1 Snail protein.

In view of the poor effectiveness of existing potato cleaning methods in China and reflecting the findings of a research analysis of basic sizes and types of impurities on potato tubers, a gradient cleaning method for potato based on a multi-step dry-cleaning and wet cleaning operation was proposed. The method mainly consists of dry-cleaning and wet cleaning. The dry-cleaning stage, which combines vibration and brushing, could effectively remove impurities such as residual rhizomes, peeled potato skin, and large pieces of soil and crushed stone from the surface of potato tubers. The wet cleaning stage adopts the gradient cleaning method of pre-cleaning, rough cleaning and fine cleaning, which could further remove soil and crushed stone attached to the surface and hidden in the sprout eyes of potato tubers. The optimal parameter combination for the gradient cleaning method was determined as follows. The potato feeding amount was 3 t/h, the speed of the rubber chain rod mechanism was 25 r/min, the speed of the first and third brush roller was 40 r/min, the speed of the second and fourth brush roller was 56 r/min, the moving speed of the immersion mechanism conveying net chain was 0.04 m/s, the speed of the brush roller in the high pressure spray and brush roller combination mechanism was 40 r/min, the ultrasonic power was 1200 W, the ultrasonic frequency was 33 kHz, the bubble intensity was 300 L/min, and the moving speed of the conveying net chain in the ultrasonic and bubble combination mechanism was 0.05 m/s. Taking the impurity removal rate and damage rate of potato tuber as the test indexes, a potato cleaning performance test was carried out under the optimal parameters combination. The results showed that the average impurity removal rate and damage rate of potato tubers were 99.05% and 2.48%, respectively. Additionally, the operational performance fully met the requirements for potato cleaning. This study provides a new method for potato cleaning in China and can also provide a reference for cleaning other root and tuber crops.

Abstract Background Retinitis pigmentosa is a common genetic disease that causes retinal degeneration and blindness for which there is currently no curable treatment available. Vision preservation was observed in retinitis pigmentosa animal models after retinal stem cell transplantation. However, long-term safety studies and visual assessment have not been thoroughly tested in retinitis pigmentosa patients. Methods In our pre-clinical study, purified human fetal-derived retinal progenitor cells (RPCs) were transplanted into the diseased retina of Royal College of Surgeons (RCS) rats, a model of retinal degeneration. Based on these results, we conducted a phase I clinical trial to establish the safety and tolerability of transplantation of RPCs in eight patients with advanced retinitis pigmentosa. Patients were studied for 24 months. Results After RPC transplantation in RCS rats, we observed moderate recovery of vision and maintenance of the outer nuclear layer thickness. Most importantly, we did not find tumor formation or immune rejection. In the retinis pigmentosa patients given RPC injections, we also did not observe immunological rejection or tumorigenesis when immunosuppressive agents were not administered. A 203 m-high concrete gravity dam is a huge structure for which the dynamic responses, damage mechanism, and safety evaluation under an intense earthquake need to be investigated carefully and in detail. This article presents the results of the dynamic failure model test on the overflow section of the aforementioned dam. It presents the seismic characteristics, failure mode, and safety evaluation for the overflow section during a strong earthquake. Two models of the dam section have been tested on the shaking table to simulate earthquake effects in a laboratory. The first model considers the Chinese code seismic wave, while the second model considers the Wenchuan seismic wave, a natural wave. The peak ground acceleration (PGA) of the first crack during testing determines the seismic safety level of the dam, and the locations and forms of cracks at the end of testing indicate the vulnerable areas of the dam under two kinds of earthquake inputting. After the final analysis, the results of the experimental works for the overflow during an earthquake offer an important reference for engineering designers working on anti-seismic design.

A 203-m-high gravity dam being built in earthquake-prone areas needs to be investigated very carefully to determine its dynamic responses, damage mechanism, and safety evaluation. The dynamic characteristics, seismic responses, failure mode, and safety evaluation of the above structure are presented through dynamic fracture test for small-scale model on shaking table. Because the strength of the model material is very low, the traditional strain gauge is also not easy to be glued to the surface of model. It is difficult to measure the accurate strain data of small-scale model during testing. Therefore, Fiber Bragg Grating (FBG) strain sensor is presented to obtain the strain of small-scale model during testing, due to its high sensitivity. The dynamic strain and residual strain are obtained with the FBG sensors embedded in model. The FBG sensor is adhered to model material completely and shows advantages of ease for installation, high sensitivity, and reliability compared with traditional resistance strain gauge. The model during testing is submitted with earthquake wave from the Chinese Code. In the experiment, the peak ground acceleration (PGA) of the first crack in the model indicates the safety level of the gravity dam. The crack locations and forms determine the damageable part of gravity dam under intense earthquake. After the final analysis, the safety evaluation result of the gravity dam under strong earthquake is given in order to guide the implementation of the project.

Ultrasonic testing analysis is a crucial scientific component during the process and interpretation of the ultrasonic detection signal. Focusing on the ultrasonic testing characteristics, the time-invariant spectral analysis method cannot meet the processing requirements of the detection signal fully. Thus, S transform, the time-varying analysis method, was introduced into the ultrasonic testing data processing of the concrete structure. The acoustic wave phase velocity was derived based on the spectrum analysis, and the S transform time-frequency analysis method was established. Finally, based on the method of concrete experimental data set, studies show that the frequency energy spectrum with S transform can realize flexible and effective identification of defects in the concrete structure. Definitely, this analysis method can significantly improve the resolution and practicality of ultrasonic testing.

Objective: Infantile hemangiomas (IHs) are the most common vascular tumors of infancy. Oral propranolol has achieved great success in treating IHs since 2008. To minimize the systemic side events caused by oral administration of propranolol, topical timolol started to be applied in the treatment of IHs, especially for superficial lesions.Methods: We treated 724 children with superficial IHs using oral propranolol or topical timolol, and investigated the efficacy and safety of the two treatment patterns.Results: Both oral propranolol and topical timolol achieved a satisfactory therapeutic outcome, with an effective response rate of 97 and 96.4%, respectively. No significant differences in visual analog scale (VAS) improvement between the two groups were observed. Occurrence rate of systemic adverse events for patients treated with oral propranolol (3.9%) was significantly higher than that for patients treated with topical timolol (0%). Clinical response was not associated with gender, duration of treatment, lesion location, lesion size, gestational age, and progesterone use during pregnancy, but closely associated with age at treatment initiation, which indicated that younger age at treatment initiation predicted for a better regression rate.Conclusions: We recommend that topical timolol instead of oral propranolol could be the first-line therapy for superficial IHs because of its good efficacy and improved safety.

Concrete structures are often in different humidity conditions that have a significant impact on the elastic modulus of concrete, therefore, systematic research on the evolution of the law of concrete elastic modulus under different humidity conditions is needed. In this study, the variation laws of the water saturation of concrete specimens with strength grades C15, C20, and C30 were obtained, and then the influence laws of the water saturation on the concrete axial compressive strength were carried out, and the prediction model of elastic modulus of concrete with respect to water saturation was constructed. The results showed that the water saturation of concrete with strength grades C15, C20, and C30 increased with an extension of immersion time, and the water saturation showed an approximately linear rapid growth within three soaking hours, reaching 47.56%, 71.63%, and 47.29%, respectively. Note, the concrete reached saturation state when the soaking time was 240 h. The axial compressive strength with strength grades C15, C20, and C30 decreased with increased water saturation, and the axial compressive strength of saturated concrete decreased by 27.25%, 21.14%, and 20.76%, respectively, as compared with the dry state concrete. The elastic modulus of concrete with strength grades C15, C20, and C30 increased with increased water saturation, and the elastic modulus of saturated concrete was 1.18, 1.19, and 1.24 times higher than those of dry concrete, respectively.

Since copper azide (Cu(N3)2) has high electrostatic sensitivity and is difficult to be practically applied, silicon-based Cu(N3)2@carbon nanotubes (CNTs) composite energetic films with higher electrostatic safety were fabricated, which can be compatible with micro-electro mechanical systems (MEMS). First, a silicon-based porous alumina film was prepared by a modified two-step anodic oxidation method. Next, CNTs were grown in pores of the silicon-based porous alumina film by chemical vapor deposition. Then, copper nanoparticles were deposited in CNTs by electrochemical deposition and oxidized to Cu(N3)2 by gaseous hydrogen azide. The morphology and composition of the prepared silicon-based Cu(N3)2@CNTs energetic films were characterized by field emission scanning electron microscopy (FESEM), transmission electron microscopy (TEM) and X-ray diffraction (XRD), respectively. The electrostatic sensitivity of the composite energetic film was tested by the Bruceton method. The thermal decomposition kinetics of the composite energetic films were studied by differential scanning calorimetry (DSC). The results show that the exothermic peak of the silicon-based Cu(N3)2@CNTs composite energetic film is at the temperature of 210.95 °C, its electrostatic sensitivity is significantly less than that of Cu(N3)2 and its 50% ignition energy is about 4.0 mJ. The energetic film shows good electric explosion characteristics and is successfully ignited by laser.

An effective approach is introduced to predict the magnitude of reservoir-triggered earthquake (RTE), based on support vector machines (SVM) and fuzzy support vector machines (FSVM) methods. The main influence factors on RTE, including lithology, rock mass integrity, fault features, tectonic stress state, and seismic activity background in reservoir area, are categorized into 11 parameters and quantified by using analytical hierarchy process (AHP). Dataset on 100 reservoirs in China, including the 48 well-documented cases of RTE, are collected and used to train and validate the prediction models established with SVM and FSVM, respectively. Through numerical tests, it is found that both the SVM and FSVM models are effective in the prediction of the magnitude of RTE with high accuracy, provided that sufficient samples are collected. While the results of FSVM which is extended from SVM by introducing a fuzzy membership to reduce the influence of noises or outliers are found to be slightly less accurate than those of SVM in the current analysis of RTE cases. The reason might be attributed to the high discreteness of the sample data in the current study.

The therapeutic options for infantile hemangiomas (IHs) have been greatly altered since the introduction of oral propranolol for successful treatments of IHs. Recently, there is an increase in the application of topical timolol maleate for treating superficial IHs. In the present study, we developed a new formulation of timolol maleate 0.5% hydrogel and treated 321 patients with superficial IHs to evaluate its efficacy and safety in the treatment of superficial IHs. This new timolol hydrogel was applied three times daily with a mean duration of 7.1 months. Response to treatment was assessed according to cosmetic improvement by using visual analog scale (VAS). The average VAS improvement after treatment was 76.4, with 126 patients (39.3%) achieving excellent responses, 159 patients (49.5%) achieving good responses, 33 patients (10.3%) achieving fair responses, and three patients (0.9%) achieving poor responses. Age at treatment initiation (P = 0.0349) and lesion thickness (P = 0.0147) were significantly associated with therapeutic efficacy. No severe side effects were observed in all patients. In conclusion, this new topical timolol maleate 0.5% hydrogel appears to be a proper candidate for treating superficial IHs, and our study provides supportive evidence and experience of topical timolol maleate in treating superficial IHs.

In order to comprehensively evaluate the properties of rock-filled concrete (RFC) with the strength of C15, lab experimental test and in-situ test are applied to explore the mechanical, hydraulic, ultrasonic characteristics of RFC in Hantang reservoir dam. Four types of defects within RFC are shown from the appearance of borehole cores specimens: (1) large sized voids existing in interfacial transition zone (ITZ) between self-compacting concrete (SCC) and rock block (RB); (2) bad cohesion in ITZ; (3) joints within rock block; (4) voids within SCC. For hydraulic aspects, the average porosity of RFC is 14.10%; the permeability rate of RFC ranges from 2.41 Lu to 10.41 Lu, with the average of 11.32 Lu, occasionally more than 25.52 Lu due to interconnected defects. For ultrasonic aspects, the ultrasonic velocity of RFC conforms to lognormal distribution, with the average of 2993.3 m/s and standard deviation of 650.5 m/s. For mechanical aspects, the average cubic compressive strength of RFC is 22.55 Mpa, with the standard deviation of 4.09 Mp. Thus the data shows a relatively great dispersion due to uneven distribution of some defects in RFC, which deteriorate the quality of RFC. Through the experimental investigation, it is shown that the quality of massive RFC in the Hantang dam is obviously non-homogeneous, mainly influenced by construction technology.

BackgroundPhthalate exposure in pregnancy is typically estimated using maternal urinary phthalate metabolite levels. Our aim was to evaluate the association of urinary and placental tissue phthalates, and to explore the role of maternal and pregnancy characteristics that may bias estimates.MethodsFifty pregnancies were selected from the CANDLE Study, recruited from 2006 to 2011 in Tennessee. Linear models were used to estimate associations of urinary phthalates (2nd, 3rd trimesters) and placental tissue phthalates (birth). Potential confounders and modifiers were evaluated in categories: temporality (time between urine and placenta sample), fetal sex, demographics, social advantage, reproductive history, medication use, nutrition and adiposity. Molar and quantile normalized phthalates were calculated to facilitate comparison of placental and urinary levels.ResultsMetabolites detectable in >80% of both urine and placental samples were MEP, MnBP, MBzP, MECPP, MEOHP, MEHHP, and MEHP. MEP was most abundant in urine (geometric mean [GM] 7.00 ×102 nmol/l) and in placental tissue (GM 2.56 ×104 nmol/l). MEHP was the least abundant in urine (GM 5.32 ×101 nmol/l) and second most abundant in placental tissue (2.04 ×104 nmol/l). In aggregate, MEHP differed the most between urine and placenta (2.21 log units), and MEHHP differed the least (0.07 log units). MECPP was positively associated between urine and placenta (regression coefficient: 0.31 95% CI 0.09, 0.53). Other urine-placenta metabolite associations were modified by measures of social advantage, reproductive history, medication use, and adiposity.ConclusionPhthalates were ubiquitous in 50 full-term placental samples, as has already been shown in maternal urine. MEP and MEHP were the most abundant. Measurement and comparison of urinary and placental phthalates can advance knowledge on phthalate toxicity in pregnancy and provide insight into the validity and accuracy of relying on maternal urinary concentrations to estimate placental exposures.Impact statementThis is the first report of correlations/associations of urinary and placental tissue phthalates in human pregnancy. Epidemiologists have relied exclusively on maternal urinary phthalate metabolite concentrations to assess exposures in pregnant women and risk to their fetuses. Even though it has not yet been confirmed empirically, it is widely assumed that urinary concentrations are strongly and positively correlated with placental and fetal levels. Our data suggest that may not be the case, and these associations may vary by phthalate metabolite and associations may be modified by measures of social advantage, reproductive history, medication use, and adiposity.

Feature selection is an important topic in bioinformatics. Defining informative features from complex high dimensional biological data is critical in disease study, drug development, etc. Support vector machine-recursive feature elimination (SVM-RFE) is an efficient feature selection technique that has shown its power in many applications. It ranks the features according to the recursive feature deletion sequence based on SVM. In this study, we propose a method, SVM-RFE-OA, which combines the classification accuracy rate and the average overlapping ratio of the samples to determine the number of features to be selected from the feature rank of SVM-RFE. Meanwhile, to measure the feature weights more accurately, we propose a modified SVM-RFE-OA (M-SVM-RFE-OA) algorithm that temporally screens out the samples lying in a heavy overlapping area in each iteration. The experiments on the eight public biological datasets show that the discriminative ability of the feature subset could be measured more accurately by combining the classification accuracy rate with the average overlapping degree of the samples compared with using the classification accuracy rate alone, and shielding the samples in the overlapping area made the calculation of the feature weights more stable and accurate. The methods proposed in this study can also be used with other RFE techniques to define potential biomarkers from big biological data.
