Abstract

Systematic efforts to sequence the cancer genome have identified large numbers of relevant mutations and copy number alterations in human cancers; however, elucidating their functional consequences, and their interactions to drive or maintain oncogenic states, is still a significant challenge. Here we introduce REVEALER, a computational method that identifies combinations of mutually exclusive genomic alterations correlated with functional phenotypes, such as the activation or gene-dependency of oncogenic pathways or the sensitivity to a drug treatment. We use REVEALER to uncover complementary genomic alterations associated with the transcriptional activation of β-catenin and NRF2, MEK-inhibitor sensitivity, and KRAS dependency. REVEALER successfully identified both known and new associations demonstrating the power of combining functional profiles with extensive characterization of genomic alterations in cancer genomes.

38. Telomere and Telomerase Dynamics in Human Cells

Abstract

Accumulating evidence now implicates telomeres and telomerase as critical regulators genomic stability and replicative lifespan in mammalian cells. Disruption of telomere maintenance and/or telomerase expression contributes to the etiology of some degenerative diseases and may participate in the process of aging. Although telomere dysfunction and aberrant telomerase expression clearly play important roles in cancer development, the contribution of telomere biology to cancer is complex and involves both positive and negative influences on tumor development. Indeed, recent work from several laboratories suggests additional roles for telomeres and telomerase in both normal and malignant physiology. Understanding the complexity of telomere biology will provide further insights into chromosome biology in both normal and malignant cells.

41. Human breast cancer cells generated by oncogenic transformation of primary mammary epithelial cells.

Abstract

A number of genetic mutations have been identified in human breast cancers, yet the specific combinations of mutations required in concert to form breast carcinoma cells remain unknown. One approach to identifying the genetic and biochemical alterations required for this process involves the transformation of primary human mammary epithelial cells (HMECs) to carcinoma cells through the introduction of specific genes. Here we show that introduction of three genes encoding the SV40 large-T antigen, the telomerase catalytic subunit, and an H-Ras oncoprotein into primary HMECs results in cells that form tumors when transplanted subcutaneously or into the mammary glands of immunocompromised mice. The tumorigenicity of these transformed cells was dependent on the level of ras oncogene expression. Interestingly, transformation of HMECs but not two other human cell types was associated with amplifications of the c-myc oncogene, which occurred during the in vitro growth of the cells. Tumors derived from the transformed HMECs were poorly differentiated carcinomas that infiltrated through adjacent tissue. When these cells were injected subcutaneously, tumors formed in only half of the injections and with an average latency of 7.5 weeks. Mixing the epithelial tumor cells with Matrigel or primary human mammary fibroblasts substantially increased the efficiency of tumor formation and decreased the latency of tumor formation, demonstrating a significant influence of the stromal microenvironment on tumorigenicity. Thus, these observations establish an experimental system for elucidating both the genetic and cell biological requirements for the development of breast cancer.

Human mesothelial cells, endothelial cells, and type II kidney epithelial cells growing in culture devote approximately 3% of their total protein synthesis to the production of an Mr approximately 46-kD, pI 7.1, secreted glycoprotein (designated Sp46). Fibroblasts make about 1/10th as much Sp46 as these cell types, and their synthesis is dependent upon hydrocortisone. Keratinocytes, urothelial cells, conjunctival epithelial cells, and mammary epithelial cells do not make detectable amounts of Sp46. Mesothelial cells secrete Sp46 onto the substratum, and from there it is subsequently released into the medium. Immunofluorescence analysis using specific antisera discloses that Sp46 is deposited beneath cells as a fine coating on the substratum. In sparse cultures, Sp46 is detected in trails behind motile cells. In contrast, secreted fibronectin coalesces into fibers, most of which remain in contact with and on top of the cells; thus Sp46 does not preferentially bind to fibronectin. About 6 kD of the mass of human Sp46 is N-linked oligosaccharide, which is terminally sialated before secretion. Sp46 has a low glycine content, indicating that it is not a collagenlike protein. Its NH2-terminal sequence over the first 40 amino acids does not resemble any protein for which sequence information is available. Sp46 appears to be a novel extracellular glycoprotein, high-level constitutive expression of which is restricted to mesoderm-derived epithelial and endothelial cells. We therefore propose for it the name "mesosecrin."

43. The Effect of Receiver Pressure on the Observed Flow Pattern in the Hydrostatic Extrusion of Bimetal Rods

Abstract

An experimental study was conducted to determine the acceptable combinations of process variables for the production of a niobium-titanium superconducting alloy in a pure aluminum matrix by hydrostatic extrusion. Conditions for sound flow and defective flow are shown on plots of percent reduction in area versus die semicone angle. The superconductor used was a 56 percent Nb-44 percent Ti alloy surrounded by a matrix of 99.995 percent pure aluminum. The study was performed in three phases. Single-core billets were extruded into atmospheric pressure with reduction in area, die semicone angle, and the ratio of the volumes of the two phases being varied. Single-core billets were extruded into receiver pressure with reduction in area and die semicone angle being varied. Billets with three cores were extruded with reduction in area, die semicone angle, friction, and receiver pressure being varied. In all cases there was a light mechanical bond between the two materials produced by interference fitting of the cores into drilled holes. Experimental results showed that increasing the receiver pressure increases the range of acceptable process variables to produce sound flow. Although it was possible to produce sound flow, the results were not always consistent and repeatable. This is largely due to the great difference in strength between the soft aluminum and the hard superconductor and the difficulty in accurately controlling the bond strength. It is expected that the beneficial effect of receiver pressure will be even more pronounced with a tighter control on bonding.

44. Analysis of Combined Backward-Forward Extrusion

Abstract

The upper bound approach is used to analyze combined backward-forward extrusion. The deformation region is divided into five zones separated by planer and cylindrical surfaces of velocity discontinuities. The internal power of deformation and shear and friction losses are computed individually and summed. The pseudo-independent process parameter is the backward rate of flow with respect to which the total power of deformation is optimized. The optimal backward rate of flow is assumed to be the actual one. Thus, the backward rate of flow becomes a dependent parameter to be studied through this analysis. Conditions covering backward rates of flow from zero to maximum are demonstrated graphically. Examples are given for which combined flow results and for which either only forward flow or only backward flow occur.

Abstract

The upper bound approach is used to analyze the early stage of impact extrusion of a thin or thick-walled cup. It is shown that during the early stage of extrusion with a flat ram the ram pressure increases linearly with increasing penetration of the ram, increased area of contact between the tool and the workpiece, and poorer lubrication. With the same friction and process geometry the early stage of extrusion is made easier by using a nosed ram instead of a flat one. The optimum nose contour can be determined by a minimization technique.

Abstract

Li(sub x)Ni(1-x)O, Li(sub x)Mn(1-x)O, and Li(sub x)Co(1 -x)O. It is proposed in this investigation to measure the effect on the room temperature specific resistivity of additions of MgO to systems such as LixNi(1-x)O, LixMn(1-x)O, and LixCo(1-x)O. Because the divalent state of magnesium is so stable, it will not enter into the conductivity process directly, but it can be anticipated that it can confer several beneficial results on the conductivity in the above named oxide systems. It is realized that extremely large additions of MgO to any of the systems under consideration, as well as additions of MgO to systems in which the amount of lithium is very small, would serve only to increase the resistivity, but it can be anticipated that a region of compositions may be found in which the resistivity will be decreased. An additional reason for adding MgO, and perhaps ultimately the most important, is that existing theories do not explain the maximum in the activation energy observed at approximately 50% nickel in the system Li(sub 0.08)Ni(sub y)Co(1-0.08-y)O observed by Miller, Heikes and Johnston (J. Consideration is given to the thermodynamics of equilibria involving metal solutions, oxide solutions, and atmospheres of known 02 partial pressures. The standard free energy of formation of the oxides of the metals involved are regarded, in one case, as being markedly different (exemplified by the system Mn-Fe-O at 1400K) and, in the other case, as being nearly the same (exempli fied by the system Co-Ni-O at 1700K). Results are presented in the form of ternary phase diagrams.

Abstract

CD2 (T11, the T cell E receptor), a nonpolymorphic 47- to 55-kDa glycoprotein, is a T cell-specific surface protein that plays an important role in T lymphocyte adhesion, signal transduction, and differentiation. A natural ligand of CD2 is lymphocyte function associated Ag-3 (LFA-3 (CD58)), a widely expressed glycoprotein of 50 to 70 kDa. The physiologic interaction of CD2 with LFA-3 functions to increase intercellular adhesion and plays a role in T cell activation. This interaction, however, in the absence of other stimuli, has not previously been shown to induce intracellular signals such as Ca2+ mobilization or IL-2 production. To investigate whether cAMP may play a role in ligand-triggered CD2-mediated signal transduction, we have studied the ability of purified LFA-3 and anti-CD2 mAb to induce changes in intracellular cAMP content in murine Ag-specific T cell hybridomas that stably express wild-type and mutated human CD2 molecules. By using a RIA sensitive to the femtomolar range and specific for cAMP, we demonstrate that purified LFA-3, like anti-CD2 mAb, is capable of inducing marked, transient increases in the intracellular concentration of cAMP. Presentation of purified LFA-3, like anti-CD2 mAb, is capable of inducing marked, transient increases in the intracellular concentration of cAMP. Presentation of purified LFA-3 alone to CD2-expressing hybridoma cells, however, did not stimulate phosphatidylinositol turnover nor IL-2 production. The cytoplasmic domain of CD2 is necessary for these ligand-induced cAMP changes, demonstrating that LFA-3 binding to CD2 transduces a signal to the cell. Experiments using the phosphodiesterase inhibitor 3-isobutyl-1-methyl-xanthine showed that CD2-mediated regulation of cAMP levels occurs primarily by the stimulation of cAMP production rather than by the inhibition of cAMP degradation. These results demonstrate that the interaction of LFA-3 with CD2, in the absence of other stimuli, is capable of initiating intracellular biochemical changes and suggest that CD2/LFA-3 interactions may regulate T cell function at least in part through the generation of intracellular cAMP.
