Anhang 82, Der CD30-Rezeptor ist ein Mitglied der Tumor Nekrose Faktor Rezeptor Familie. Die Stimulation des CD30-Rezeptors vermittelt vielfältige Zellantworten, unter anderem die Auslösung von Apoptose. So ist es erstaunlich, dass der Rezeptor auf stark proliferierenden Tumoren, wie dem Hodgkin Lymphom (HL) und dem Anaplastischen Grosszelligen Lymphom (ALCL) überexprimiert wird. Während die Simulation des CD30-Rezeptors in den Tumorzellen des ALCL Apoptose auslöst, zeigen sich die Tumorzellen des HL resistent gegenüber CD30-vermittelter Apoptose. Da eine Mutation des CD30-Rezeptors als Ursache ausgeschlossen werden kann, liegt die Vermutung nahe, dass die nachgeschaltete Signaltransduktion diese unterschiedliche Reaktion der Zellen verursacht. In Gewebeschnitten CD30+ Tumoren wurde deshalb mittels in situ Hybridisierung vergleichend die Expression der TNFR-Signaltransduktionskomponenten in Tumorzellen des HL, ALCL und umliegenden gesunden Zellen untersucht. Es zeigte sich eine Überexpression der Transkripte von TRAF1, A20 und cIAP-2 differentiell in den Tumorzellen des HL im Vergleich zu denen des ALCL. Zudem konnte die Induktion der Expression dieser Proteine durch CD30-Rezeptorstimulation und die anti-apoptotische Wirkung von TRAF1 belegt werden. Bis heute ist der Weg, auf dem der CD30-Rezeptor in der Lage ist Apoptose auszulösen, nicht bekannt. Die bisher identifizierten Signaltransduktionskomponenten des Rezeptors weisen keinerlei pro-apoptotische Eigenschaften auf. Daher existiert die Vermutung, dass bisher unbekannte Proteine mit CD30 wechselwirken und dies Apoptose vermitteln kann. Mittels Hefe-2-Hybrid-Technik wurde hier versucht, neuartige Interaktionspartner des Rezeptors zu identifizieren. Die Untersuchungen wurden an cDNAs verschiedener HL- und ALCL-Zelllinien mit den Ködern für CD30, TRAF1 und TRAF5 durchgeführt. Hiermit konnte die bisher unbekannte Interaktion der Proteine CD30 und TRAF5 mit hUBC-9 gezeigt werden. Eine mögliche durch hUBC-9 vermittelte Sumotylierung der Proteine konnte jedoch weder nachgewiesen, noch ausgeschlossen werden. Um weitergehend zu untersuchen, ob dysfunktional mutierte Proteine der Apoptosevermittlung die Apoptoseresistenz der HL- Tumorzellen verursachen, wurden die apoptotischen Schlüsselproteine Caspase-3, FLICE and Apaf-1 auf mRNA-Ebene mittels RT-PCR und Sequenzanalyse untersucht. Die Mutationsanalysen der cDNAs von Apaf-1, FLICE und Caspase-3 in CD30+ Zelllinien, nicht-lymphatischen Tumorzelllinien und nicht-malignen Referenzproben ergab keine Abweichungen für die Transkripte von FLICE. In allen generierten cDNA-Proben von Caspase-3 konnte eine generelle Abweichung von der Zielsequenz, die zu einem Austausch von zwei Aminosäuren der Proteinsequenz führt, festgestellt werden. Die Untersuchung von Apaf-1-Transkripten führte zu der Charakterisierung dreier neuartiger Formen der Apaf-1-mRNA, die vermutlich Spleißvarianten darstellen., CD30 is a prominent member of the tumour necrosis factor receptor (TNFR) family. One of the major functions of these receptors is believed to be the induction of apoptosis. Therefore it is surprising that this receptor is overexpressed by highly proliferating tumours such as Hodgkin lymphoma (HL) and anaplastic large cell lymphoma (ALCL). Currently it is well known that the stimulation CD30 leads to apoptosis of ALCL tumour cells but not of Hodgkin- Reed-Sternberg (HRS) cells of HL. These very different results of CD30 stimulation might be related to the diverse expression of CD30-associated signal transduction components. Therefore the expression of these signal transduction components were examined in tumour cells of ALCL, HL and normal tissue by in situ hybridisation. The study revealed a differential transcript overexpression of the anti apoptotic proteins TRAF1, A20 and cIAP-2 in tumour cells of HL in contrast to the low expression levels in ALCL tumour cells. Additionally it was demonstrated that stimulation of CD30 receptor leads to the induction of TRAF1, A20 and cIAP-2 in ALCL and B-HL cell lines. Furthermore the anti apoptotic function of TRAF1 was shown. Up to now it is still unknown how CD30 is capable of inducing apoptosis. All identified components of CD30-associated signal transduction seem to miss any apoptotical functionality. Hence the reasonable assumption arises that other related signal transduction components exist that induce apoptotic pathways. To search for putative novel interactions, a two-hybrid screening was performed in cDNA of HL-, ALCL-cell lines with CD30 and its signal transduction components TRAF1 and TRAF5 as baits. A novel protein-protein interaction of CD30 and TRAF5 with the human ubiquitin conjugating enzyme 9 (hUBC-9) had been discovered. Unfortunately it could not be revealed whether this interaction leads to hUBC-9 mediated SUMO-1 modification of these proteins or not. To examine whether dysfunctional mutations of pro apoptotic proteins are the cause for apoptotic resistance the key players of apoptotic execution caspase-3, FLICE and Apaf-1 were investigated on mRNA level utilizing RT-PCR and sequence analysis methods. Therefore the cDNA of several CD30 positive tumour cell lines, cell lines of other tumour entity and normal tissue had been prepared and investigated. The mutational analysis of FLICE revealed no aberrations to the original identified sequence. All of the investigated cDNA probes for caspase-3 showed an aberration of two amino acids in contrast to the formerly publicised protein sequence. The study of the Apaf-1 transcripts did not disclose the expected tumour related alterations but led instead to the identification of three novel forms of Apaf-1 which probably represent splicing variants.
