Abstract

The emergence of the global pandemic caused by the novel SARS-CoV-2 virus has motivated scientists to find a definitive treatment or a vaccine against it in the shortest possible time. Current efforts towards this goal remain fruitless without a full understanding of the behavior of the virus and its adaptor proteins. This review provides an overview of the biological properties, functional mechanisms, and molecular components of SARS-CoV-2, along with investigational therapeutic and preventive approaches for this virus. Since the proteolytic cleavage of the S protein is critical for virus penetration into cells, a set of drugs, such as chloroquine, hydroxychloroquine, camostat mesylate have been tested in clinical trials to suppress this event. In addition to angiotensin-converting enzyme 2, the role of CD147 in the viral entrance has also been proposed. Mepolizumab has shown to be effective in blocking the virus's cellular entrance. Antiviral drugs, such as remdesivir, ritonavir, oseltamivir, darunavir, lopinavir, zanamivir, peramivir, and oseltamivir, have also been tested as treatments for COVID-19. Regarding preventive vaccines, the whole virus, vectors, nucleic acids, and structural subunits have been suggested for vaccine development. Mesenchymal stem cells and natural killer cells could also be used against SARS-CoV-2. All the above-mentioned strategies, as well as the role of nanomedicine for the diagnosis and treatment of SARS-CoV-2 infection, have been discussed in this review. (C) 2020 Elsevier B.V. Abstract

Objective: The genus Ferula L. includes perennial flowering plants belonging to the Apiaceae family. This genus is a rich source of biologically active phytochemicals such as sulfur-containing derivatives, coumarins, sesquiterpenes, sesquiterpene lactones, sesquiterpene coumarins, glucuronic acid, galactose, arabinose, rhamnose, and daucane esters. Over the last decade, considerable attention has been paid to biological activities of these compounds; it is assumed that the most prominent biological features of the genus Ferula are their cytotoxic effects. This article discusses cytotoxic activity of the genus Ferula and their important compounds. Materials and Methods: In this mini-review article, papers published from 1990 to April 2016 were included and the following information was discussed; cytotoxic activity of the genus Ferula and their important compounds, the type of cell line used in vitro, concentrations of the extracts/active compound that were used, and the underlying mechanisms of action through which Ferula-related chemicals induced cytotoxicity. In addition, we explained different mechanisms of action through which the active constituents isolated from Ferula, could decrease cellular growth. Conclusion: It is highly recommended that potent and effective compounds that were isolated from Ferula plants and found to be appropriate as adjuvant therapy for certain diseases, should be identified. Also, the versatile biological activities of sesquiterpene coumarins suggest them as promising agents with a broad range of biological applications to be used in the future. [ABSTRACT FROM AUTHOR]

16. Chemical Composition and Biological Profile of Johreniopsis stricticaulisAerial Parts Essential Oil

Abstract

AbstractThe chemical composition of Johreniopsis stricticaulisessential oil was obtained by hydro-distillation method from its aerial parts, and analyzed by gas chromatography-mass spectrometry (GC/MS). Totally the current essential oil was mainly constituted of sesquiterpene hydrocarbons. Germacrene-B (23.0 %) was the most abundant compound, and the other major compounds were α-cubebene (10.02 %), γ-elemene (5.37 %) and limonene (4.72 %). Also antioxidant, antibacterial and antifungal activities of essential oil was tested. Antioxidant assay showed that the essential oil of J. stricticaulishas moderate activity, which was weaker than positive control butylated hydroxytoluene (BHT) (IC5018.0 and 0. 13 mg/ml for essential oil and BHT, respectively). Moreover the essential oil of J. stricticaulisexhibited significant antibacterial and antifungal activities against Staphylococcus aureusas well as Candida albicans,respectively; however, it had no antibacterial effects on Escherichia coli. These results proved that essential oil of J. stricticaulisincludes sesquiterpenes compounds with moderate antioxidant and antibacterial properties. This is the first report on chemical composition and biological activities of J. stricticaulisessential oil.

17. An in-situforming implant formulation of naltrexone with minimum initial burst release using mixture of PLGA copolymers and ethyl heptanoate as an additive: In-vitro, ex-vivo, and in-vivorelease evaluation

Abstract

An in-situ forming implant formulation of naltrexone (NTX) was achieved based on a minimum initial burst release of NTX in the in-vitrorelease medium using a Box-Behnken design. Variables such as percent of copolymer by weight, copolymer composition (PLGA 756s: PLGA 504H), and percent ethyl heptanoate by weight as an additive in formulation were investigated. The in-vitro, ex-vivo, and in-vivorelease of the optimized formulation was investigated. The rabbit-blood concentrations of the optimized formulation and Vivitrol®were compared to ensure their equivalency. The initial burst release of the optimized formulation in the in-vitrorelease over the first 24 h, 6.18 ± 0.91%, was significantly (p < 0.05) lower than that of the formulation containing 100% of PLGA 504H (17.45 ± 1.07%) and 100% of PLGA 756s (11.82 ± 1.03%). The Cmaxof NTX (21.06 ± 2.9 ng/mL) from the optimized formulation was close to that of Vivitrol®(21.11 ± 2.89 ng/mL). Also, the absolute bioavailability (F) and the range of serum concentration of NTX (C) of the ISFI formulation (F = 18.29, C = 6.18–22.84) were similar to Vivitrol®(F = 16.83, C = 6.83–23.09). These results indicate that the optimized formulation can reach an effective therapeutic concentration for treating opioid and alcohol dependence.
