Abstract

BackgroundAlthough recent sequencing studies have revealed that 10% of childhood gliomas are caused by rare germline mutations, the role of common variants is undetermined and no genome-wide significant risk loci for pediatric central nervous system tumors have been identified to date.MethodsMeta-analysis of 3 population-based genome-wide association studies comprising 4069 children with glioma and 8778 controls of multiple genetic ancestries. Replication was performed in a separate case-control cohort. Quantitative trait loci analyses and a transcriptome-wide association study were conducted to assess possible links with brain tissue expression across 18 628 genes.ResultsCommon variants in CDKN2B-AS1 at 9p21.3 were significantly associated with astrocytoma, the most common subtype of glioma in children (rs573687, P-value of 6.974e-10, OR 1.273, 95% CI 1.179-1.374). The association was driven by low-grade astrocytoma (P-value of 3.815e-9) and exhibited unidirectional effects across all 6 genetic ancestries. For glioma overall, the association approached genome-wide significance (rs3731239, P-value of 5.411e-8), while no significant association was observed for high-grade tumors. Predicted decreased brain tissue expression of CDKN2B was significantly associated with astrocytoma (P-value of 8.090e-8).ConclusionsIn this population-based genome-wide association study meta-analysis, we identify and replicate 9p21.3 (CDKN2B-AS1) as a risk locus for childhood astrocytoma, thereby establishing the first genome-wide significant evidence of common variant predisposition in pediatric neuro-oncology. We furthermore provide a functional basis for the association by showing a possible link to decreased brain tissue CDKN2B expression and substantiate that genetic susceptibility differs between low- and high-grade astrocytoma.

Abstract

Integrated models are a popular tool for analyzing species of conservation concern. Species of conservation concern are often monitored by multiple entities that generate several datasets. Individually, these datasets may be insufficient for guiding management due to low spatio-temporal resolution, biased sampling, or large observational uncertainty. Integrated models provide an approach for assimilating multiple datasets in a coherent framework that can compensate for these deficiencies. While conventional integrated models have been used to assimilate count data with surveys of survival, fecundity, and harvest, they can also assimilate ecological surveys that have differing spatio-temporal regions and observational uncertainties. Motivated by independent aerial and ground surveys of lesser prairie-chicken, we developed an integrated modeling approach that assimilates density estimates derived from surveys with distinct sources of observational error into a joint framework that provides shared inference on spatio-temporal trends. We model these data using a Bayesian Markov melding approach and apply several data augmentation strategies for efficient sampling. In a simulation study, we show that our integrated model improved predictive performance relative to models that analyzed the surveys independently. We use the integrated model to facilitate prediction of lesser prairie-chicken density at unsampled regions and perform a sensitivity analysis to quantify the inferential cost associated with reduced survey effort., Comment: 22 pages; 5 figures, 1 table, submitted to Biometrics

Abstract

Objective To evaluate the association between maternal BMI and congenital heart defects (CHDs) in the offspring when including live births, stillbirths, aborted and terminated pregnancies and to investigate if maternal interpregnancy weight changes between the first and second pregnancy influences the risk of foetal CHDs. Methods A nationwide cohort study of all singleton pregnancies in Denmark from 2008 to 2018. Data were retrieved from the Danish Foetal Medicine Database, which included both pre- and postnatal diagnoses of CHDs. Children or foetuses with chromosomal aberrations were excluded. Odds ratios were calculated with logistic regression models for CHDs overall, severe CHDs and five of the most prevalent subtypes of CHDs. Results Of the 547 105 pregnancies included in the cohort, 5 442 had CHDs (1.0%). Risk of CHDs became gradually higher with higher maternal BMI; for BMI 25-29.9 kg/m2, adjusted odds ratio (aOR) 1.17 (95% CI 1.10-1.26), for BMI 30-34.9 kg/m2, aOR 1.21 (95% CI 1.09-1.33), for BMI 35-39.9 kg/m2, aOR 1.29 (95% CI 1.11-1.50) and for BMI ≥ 40 kg/m2, aOR 1.85 (95% CI 1.54-2.21). Data was adjusted for maternal age, smoking status and year of estimated due date. The same pattern was seen for the subgroup of severe CHDs. Among the atrioventricular septal defects (n = 231), an association with maternal BMI ≥ 30 kg/m2 was seen, OR 1.67 (95% CI 1.13-2.44). 109 654 women were identified with their first and second pregnancies in the cohort. Interpregnancy BMI change was associated with the risk of CHDs in the second pregnancy (BMI 2 to < 4 kg/m2: aOR 1.29, 95% CI 1.09-1.53; BMI ≥ 4 kg/m2: aOR 1.36, 95% CI 1.08-1.68). Abstract

Finite temperature lattice QCD is probed by varying the temporal boundary conditions of the fermions. We develop the emerging physical behavior in a study of the quenched case and subsequently present first results for a fully dynamical calculation comparing ensembles below and above the phase transition. We show that for low temperature spectral quantities of the Dirac operator are insensitive to boundary conditions, while in the deconfined phase a non-trivial response to a variation of the boundary conditions sets in.

Abstract

We present results for excited meson spectra from $N_f=2$ clover-Wilson configurations provided by the CP-PACS Collaboration. In our study we investigate both low and high spin mesons. For spin-0 and spin-1 mesons, we are especially interested in the excited states. To access these states we construct several different interpolators from quark sources of different spatial smearings and calculate a matrix of correlators. For this matrix we then solve a generalized eigenvalue problem. 45. 'Dead birds flying': can north American rehabilitated raptors released into the wild mitigate anthropogenic mortality?

Abstract

As the human footprint expands to meet societal energy needs, as do the impacts to wildlife. Raptors in particular are highly susceptible to anthropogenic caused mortality. Industry sectors are encouraged to offset these causes of mortality. Several options to mitigate these losses have been proposed, including raptor rehabilitation. However, its role as a conservation tool is untested. Currently no peer‐reviewed demographic analyses exist using post‐release data from rehabilitated raptors to evaluate its effectiveness at continental scales. Our objectives were to estimate annual survival of rehabilitated and wild raptors, and then use those estimates in demographic models to assess potential effects at individual and population levels. We hypothesized that rehabilitated raptors would survive similarly to their wild counterparts after an acclimation period, and that longer‐lived species (K‐selected) would benefit most from these releases. We used US Geological Survey Bird Banding Lab band‐recovery data (1974–2018) from 20 raptor species for modeling survival of rehabilitated individuals (n = 125 740) in comparison to wild birds (n = 1 913 352). Results from 17 species with adequate recovery data indicated that five species rehabilitated ≠ wild survival, two species had uncertain estimates, and 10 species rehabilitated ≈ wild survival by years two and three post‐release. We acquired admission (n = 69 707) and release (n = 25 740) data from 24 rehabilitation centers across the US (2012–2021). We integrated survival, fecundity and numbers of releases into demographic models. These models quantified the extent to which rehabilitated raptors may contribute to broader conservation efforts, especially in the context of individual take. All but two species, had measurable numbers of individuals added to the population regardless of the number of releases. The general pattern was for K‐selected species to yield larger benefits from rehabilitated supplementation to the population. We simulate two dynamical, mass degenerate light quarks on 16^3x32 lattices with a spatial extent of 2.4 fm using the Chirally Improved Dirac operator. The simulation method, the implementation of the action and signals of equilibration are discussed in detail. Abstract

We study the excitations of hadrons containing a single heavy quark. We present meson and baryon mass splittings and ratios of meson decay constants resulting from quenched and dynamical two-flavor configurations. Light quarks are simulated using the Chirally Improved (CI) lattice Dirac operator. The heavy quark is approximated by a static propagator, appropriate for the $b$ quark on our lattices ($1/a \sim 1-2$ GeV).
