Abstract

The goal of this research was to create a rectangular microstrip patch antenna, examine its return loss, and see how it stacked up against a circular microstrip patch antenna. In order to evaluate the antenna layout, twenty random samples were gathered. The suggested layout divides patch antennas into two groups, with group 1 consisting of rectangular microstrip patch antennas (10 samples) and group 2 consisting of circular patch antennas (20 samples). The 80/0.05 power split was used to develop the G power calculator. The antenna has a return loss value of -28.1 dB, which is better than the circular patch antenna, and an oscillation frequency of 6.5 GHz. The significance level was calculated to be 0.04. Antenna performance was researched and developed with Ansoft HFSS version 15.0. The rectangular patch antenna achieves a return loss of -28.5 within the scope of the investigation, which is much lower than the return loss of the circular microstrip patch antenna, which is 13.3. Abstract

Many circuit design applications rely on an intermediate sequence to carry a decision to the next circuit stage. The decision may be carried by a weighted pattern of N bits, with the weights being selected in a way that optimizes the circuit implementation or some aspect of performance. For example, when the weights are consecutive powers of 2 beginning with 1 = 20, we have the standard binary representation. As another example, when all the weights are 1, we have the unary representation that encodes a value k by k asserted bits and N–k unasserted bits (a weight-k bit vector of length N). In this paper, we present the design of a circuit that screens a unary representation to verify that the represented value falls between preset lower and upper limits l and u, passing through any string that represents a value in the interval [l, u] and outputting the all-0 s bit pattern otherwise. Our mixed analog–digital circuit implementation, based on switched-capacitor arrays, provides a decision output within a clock cycle of 4 ns for 16-bit unary representation, when realized with 0.15 μm TSMC technology. The latter results were obtained with normal, per-bit capacitance of 200 fF and single-clock-cycle operation. As an added benefit, our filtering circuit can form the basis for designing a cost-effective Hamming decoder circuit.

Abstract

Target occupancy is often insufficient to elicit biological activity, particularly for RNA, compounded by the longstanding challenges surrounding the molecular recognition of RNA structures by small molecules. Here we studied molecular recognition patterns between a natural-product-inspired small-molecule collection and three-dimensionally folded RNA structures. Mapping these interaction landscapes across the human transcriptome defined structure–activity relationships. Although RNA-binding compounds that bind to functional sites were expected to elicit a biological response, most identified interactions were predicted to be biologically inert as they bind elsewhere. We reasoned that, for such cases, an alternative strategy to modulate RNA biology is to cleave the target through a ribonuclease-targeting chimera, where an RNA-binding molecule is appended to a heterocycle that binds to and locally activates RNase L1. Overlay of the substrate specificity for RNase L with the binding landscape of small molecules revealed many favourable candidate binders that might be bioactive when converted into degraders. We provide a proof of concept, designing selective degraders for the precursor to the disease-associated microRNA-155 (pre-miR-155), JUN mRNA and MYC mRNA. Thus, small-molecule RNA-targeted degradation can be leveraged to convert strong, yet inactive, binding interactions into potent and specific modulators of RNA function.

Abstract

α-Synuclein is an important drug target for the treatment of Parkinson’s disease (PD), but it is an intrinsically disordered protein lacking typical small-molecule binding pockets. In contrast, the encoding SNCA mRNA has regions of ordered structure in its 5′ untranslated region (UTR). Here, we present an integrated approach to identify small molecules that bind this structured region and inhibit α-synuclein translation. A drug-like, RNA-focused compound collection was studied for binding to the 5′ UTR of SNCA mRNA, affording Synucleozid-2.0, a drug-like small molecule that decreases α-synuclein levels by inhibiting ribosomes from assembling onto SNCA mRNA. This RNA-binding small molecule was converted into a ribonuclease-targeting chimera (RiboTAC) to degrade cellular SNCA mRNA. RNA-seq and proteomics studies demonstrated that the RiboTAC (Syn-RiboTAC) selectively degraded SNCA mRNA to reduce its protein levels, affording a fivefold enhancement of cytoprotective effects as compared to Synucleozid-2.0. As observed in many diseases, transcriptome-wide changes in RNA expression are observed in PD. Syn-RiboTAC also rescued the expression of ~50% of genes that were abnormally expressed in dopaminergic neurons differentiated from PD patient–derived iPSCs. These studies demonstrate that the druggability of the proteome can be expanded greatly by targeting the encoding mRNAs with both small molecule binders and RiboTAC degraders. [ABSTRACT FROM AUTHOR]

Abstract

We present a new comparator design featuring wide-range and high-speed operation using only conventional digital CMOS cells. Our comparator exploits a novel scalable parallel prefix structure that leverages the comparison outcome of the most significant bit, proceeding bitwise toward the least significant bit only when the compared bits are equal. This method reduces dynamic power dissipation by eliminating unnecessary transitions in a parallel prefix structure that generates the N-bit comparison result after [log 4N]+[log16N]4 CMOS gate delays. Our comparator is composed of locally interconnected CMOS gates with a maximum fan-in and fan-out of five and four, respectively, independent of the comparator bitwidth. The main advantages of our design are high speed and power efficiency, maintained over a wide range. Additionally, our design uses a regular reconfigurable VLSI topology, which allows analytical derivation of the input-output delay as a function of bitwidth. HSPICE simulation for a 64-b comparator shows a worst case input-output delay of 0.86 ns and a maximum power dissipation of 7.7 mW using 0.15-μ TSMC technology at 1 GHz. © 2012 IEEE.

Abstract

The discovery of biofunctional natural products (NPs) has relied on the phenotypic screening of extracts and subsequent laborious work to dereplicate active NPs and define cellular targets. Herein, NPs present as crude extracts, partially purified fractions, and pure compounds were screened directly against molecular target libraries of RNA structural motifs in a library-versus-library fashion. We identified 21 hits with affinity for RNA, including one pure NP, nocathiacin I (NOC-I). The resultant data set of NOC-I-RNA fold interactions was mapped to the human transcriptome to define potential bioactive interactions. Interestingly, one of NOC-I's most preferred RNA folds is present in the nuclease processing site in the oncogenic, noncoding microRNA-18a, which NOC-I binds with low micromolar affinity. This affinity for the RNA translates into the selective inhibition of its nuclease processing in vitro and in prostate cancer cells, in which NOC-I also triggers apoptosis. In principle, adaptation of this combination of experimental and predictive approaches to dereplicate NPs from the other hits (extracts and partially purified fractions) could fundamentally transform the current paradigm and accelerate the discovery of NPs that bind RNA and their simultaneous correlation to biological targets.

Abstract

Yoghurt is a fermented dairy product developed by fermentation of milk with Streptococcus thermophilus and Lactobacillus delbrueckii subsp. bulgaricus. Assimilation of fruit pulp can boost the nutritional and health benefits of the yoghurt to many folds. The prime objective of preparing fruit yoghurt with the addition of persimmon pulp was to enhance the nutritional value and to improve overall acceptance. Yoghurt was prepared with the incorporation of persimmon in the concentration of 5%, 10% & 15% to assess the best combination. Evaluation of physico-chemical and organoleptic properties was performed during storage. The product was stored for 28 days at 4o C and quality parameters were assessed at an interval of 7 days. Acidity, total solids, and moisture content of fruit yoghurt increased while pH, lactose, protein, fat and textural profile values decreased significantly (p < 0.01) during storage. The phenolic content of the controlled sample was 175 mg GAE/g, which increased to 180, 214 and 250 mg GAE/g with 5%, 10% & 15% persimmon combinations respectively. Syneresis value for controlled sample was 2.49% which decreased to 2.45%, 1.94% & 1.10% in 5, 10 & 15% persimmon blends. Additionally, the sensorial properties of yoghurt blended with 10% fruit pulp gained higher acceptability during storage at 4o C.

Abstract

Background: Although guidelines exist for advanced and variant bladder cancer management, evidence is limited/conflicting in some areas and the optimal approach remains controversial.Objective: To bring together a large multidisciplinary group of experts to develop consensus statements on controversial topics in bladder cancer management.Design: A steering committee compiled proposed statements regarding advanced and variant bladder cancer management which were assessed by 113 experts in a Delphi survey. Statements not reaching consensus were reviewed; those prioritised were revised by a panel of 45 experts prior to voting during a consensus conference.Setting: Online Delphi survey and consensus conference.Participants: The European Association of Urology (EAU), the European Society for Medical Oncology (ESMO), experts in bladder cancer management.Outcome measurements and statistical analysis: Statements were ranked by experts according to their level of agreement: 1-3 (disagree), 4-6 (equivocal), and 7-9 (agree).
