Abstract Background This study describes the effectiveness of a novel active pharmaceutical ingredient, fluralaner (isoxazoline class), against important ectoparasites infesting cattle in Brazil. Methods A total of 13 studies involving a 5% fluralaner-based pour-on formulation (Exzolt 5%; further referred to as Exzolt) were conducted. Specifically, the effectiveness of this formulation was studied against Rhipicephalus microplus (6 studies), Cochliomyia hominivorax larvae (4 studies), Dermatobia hominis larvae (1 study) and Haematobia irritans flies (2 studies). Results The therapeutic efficacy of Exzolt was found to exceed 98% at 4 days post treatment (DPT), while persistent efficacy (> 90% efficacy) against repeated infestations of R. microplus was observed for up to 79 DPT. In field studies, ≥ 98% therapeutic efficacy was demonstrated at all study sites by 7 DPT, and a persistent efficacy (> 90% efficacy) was observed for 42, 49 or 56 DPT. Exzolt prevented C. hominivorax eggs from developing to the larval stage, thus mitigating the development of myiasis in cattle naturally and artificially infested with this screworm. The efficacy of Exzolt against D. hominis larvae was 98% at 3 DPT, while persistent efficacy (> 90% effectiveness) was found to last for up to 70 DPT. Against H. irritans, Exzolt showed therapeutic efficacy (≥ 90%) within the first day of treatment at both study sites, while persistent efficacy (≥ 90%) was observed for 7 DPT at one site and for 21 DPT at the other site. Conclusions Overall, the results from these studies confirm that Exzolt is therapeutically efficacious against the most important ectoparasites infesting cattle in Brazil. The novel active pharmaceutical ingredient, fluralaner, provides a new treatment option for farmers to control cattle ectoparasites, especially where there is resistance to other chemical classes. The ectoparasites Rhipicephalus microplus, Dermatobia hominis, and Haematobia hominis are a major obstacle to livestock productivity, a factor that is aggravated above all by the acquisition of drug resistance to many active ingredients. With the aim of minimizing this phenomenon, four clinical studies were conducted with a new formulation (benzoylphenylurea + organophosphate). In order to assess the anti-ixodid activity, trials were carried out on naturally and experimentally infested cattle. In the former, the carrapaticidal efficacy index was over 90% from the 7º to the 28º DPT (Days post-treatment). In the "Stall test" (artificial infestations), the maximum efficacy rate (100%) was achieved from the 20º to the 47º DPT. The mosquicidal activity of the new formulation against Haematobia irritans (natural infestation) remained above 90% from the 1º to the 14º DPT. It was more than 90% effective against Dermatobia hominis larvae from the 7º to the 21º DPT. Based on the results obtained in the four studies carried out, the unprecedented formulation can be recommended, in the dose and route prescribed, for the treatment and control of external parasites in cattle. The horn fly Haematobia irritans is a cosmopolitan hematophagous ectoparasite of great importance in livestock. In Mexico, H. irritans is distributed across the country, and is found during the whole year. The fluctuation of H. irritans population is related with climate conditions. Despite its wide distribution, the effects on animal health, and its negative impact on meat and milk production, little data exists on its infestation and epidemiology is limited. This paper is a review on the current situation of H. irritans in cattle in Mexico, its economic impact, control methods, perspectives, and research opportunities. [ABSTRACT FROM AUTHOR]

Haematobia irritans irritans (Linnaeus, 1758: Diptera: Muscidae), the horn fly, is an external parasite of penned and pastured livestock that causes a major economic impact on cattle production worldwide. Pesticides such as synthetic pyrethroids and organophosphates are routinely used to control horn flies; however, resistance to these chemicals has become a concern in several countries. To further elucidate the molecular mechanisms of resistance in horn fly populations, we sequenced the transcriptomes of ten populations of horn flies from the southern US possessing varying degrees of pesticide resistance levels to pyrethroids, organophosphates, and endosulfans. We employed an Illumina paired end HiSeq approach, followed by de novo assembly of the transcriptomes using CLC Genomics Workbench 8.0.1 De Novo Assembler using multiple kmers, and annotation using Blast2GO PRO version 5.2.5. The Gene Ontology biological process term Response to Insecticide was found in all the populations, but at an increased frequency in the populations with higher levels of insecticide resistance. The raw sequence reads are archived in the Sequence Read Archive (SRA) and assembled population transcriptomes in the Transcriptome Shotgun Assembly (TSA) at the National Center for Biotechnology Information (NCBI).

Horn flies, Haematobia irritans irritans (L.); house flies, MuscadomesticaL.; and stable flies, Stomoxys calcitrans (L.), are problematic to livestock production and, in the instance of M. domestica, to human health. p-Anisaldehyde was assessed for its bioactivity against the eggs of the three muscid species. The compound, which is found in many edible plants, was lethal to the eggs of H. irritans and S. calcitrans at a relatively low (=0.01%) concentration when eggs were exposed to the treated substrate and when exposed to fumigation (=0.0001%). Although M. domestica eggs were less vulnerable to the treated substrate than the other two muscids, egg hatch was delayed, and fumigation with a low p-anisaldehyde concentration (0.001%) induced substantial egg mortality. In many instances, larval development inside the eggs did not occur, while other larvae developed, swelled, and although they broke the egg casing, they failed to emerge. The possible utility of p-anisaldehyde against the eggs of the three muscid species as a possible integrated pest management tactic is discussed. [ABSTRACT FROM AUTHOR]

Seven mature Angus × Hereford crossbreed cows (544 ± 23.2 kg of body weight) fitted with duodenal and ruminal cannulas were used in a crossover design to evaluate the effects of horn fly (Haematobia irritans) (L.) infestation on horn fly-induced defensive behaviors, feed and water intake, as well as digestion characteristics of beef cows. Cows were randomly assigned to one of two treatment groups: 1) a horn fly infested (1,000 flies per cow) or 2) a noninfested control group. All cows were housed in individual screened pens. Fly counts and infestations were performed daily to maintain consistent populations throughout the study. Sudan hay (11.4 % crude protein, dry matter basis) was selected to mimic forage quality that range cattle frequently graze in the southwest region. Horn fly-induced host defensive behaviors were greater (P ≤ 0.0002) for horn fly infested cattle. Intake of water, dry matter, organic matter, crude protein, and neutral detergent fiber were similar (P ≥ 0.4765) among infested and noninfested cows. However, total volatile fatty acid concentration tended to increase (P = 0.0967) when horn flies were present. Dry matter fecal excretion and total tract digestion were not affected (P ≥ 0.2602) by horn fly infestation. These data indicate that horn fly infestations do not affect forage intake or digestion characteristics of beef cattle (Bos taurus) in confinement settings. [ABSTRACT FROM AUTHOR]

Bovine leukemia virus (BLV) is the agent responsible for enzootic bovine leukosis, the most common neoplastic disease in cattle. The horn fly, a major hematophagous pest of cattle, is able to transmit different diseases in cattle. However, its implication in BLV transmission under a natural environment is still discussed. The objectives of this work were to determine the presence of BLV in horn flies (by sequencing) and to evaluate the ability of horn flies to transmit BLV to cattle (through an experimental assay under a natural environment). To demonstrate the presence of BLV in the flies, 40 horn flies were collected from a BLV-positive cow with a sweep net and ten pools with four horn-fly mouthparts each were prepared. The presence of BLV was determined by nested PCR and sequencing. To demonstrate BLV transmission, other 40 flies were collected from the same BLV-positive cow with a sweep net. Eight homogenates containing five horn-fly mouthparts each were prepared and injected to eight cows of different breeds, and blood samples were collected every 21 days. Then, to evaluate the ability of horn flies to transmit BLV to grazing cattle under natural conditions, both infected and uninfected cattle from the experimental transmission assay were kept together in the same paddock with more than 200 horn flies per animal for 120 days. Blood samples were collected every 20 days and the number of flies determined. The sequencing results confirmed the presence of the provirus in horn flies. The results also confirmed that BLV transmission is a possible event, at least experimentally. However, the role of horn flies as vectors of BLV under a natural grazing system is still discussed.

Adult brachycera biting flies can significantly impact livestock through both direct effects (reduction of food intake, disturbance, painful bites, and blood loss) and indirect effects (pathogen transmission), leading to substantial economic losses and production damage. This study aimed to assess the presence of blood-sucking flies in six mixed-animal farm environments on the island of Mallorca (Balearic Islands, Spain) by employing multiple trapping methods. Additionally, distribution maps of brachycera biting fly species recorded in Spain were created, based on data extracted thorough review of scientific literature and citizen digital databases. Investigation of several pathogens, including equine infectious anemia virus (EIAV), Anaplasmataceae bacteria, and piroplasm protozoa, was carried out using different PCR targets (18S rRNA, 16S rRNA, groESL, and tat genes). Citizen science databases and literature review corroborated the consistent distribution trend for two Stomoxyinae species, underscoring the importance of citizen collaboration as a complement to traditional entomological surveillance. Our study confirmed the presence of two biting Stomoxyinae species: the prevalent stable fly Stomoxys calcitrans across all sampled farms, and the horn fly Haematobia irritans, which turned out to be less abundant. DNA barcoding techniques validated the identification of the two species. Neither EIAV nor bacterial/protozoan pathogens were detected using the selected PCR targets in either fly species. However, Wolbachia pipientis (clustered in the supergroup A together with the only sequence of W. pipientis from the USA) was identified through PCR targeting 16S rRNA, groESL and wsp genes in all pools of H. irritans (n = 13) collected from two of the examined farms. This study represents the first attempt to investigate pathogens in Stomoxyinae biting flies in Spain. The discovery of the endosymbiotic Wolbachia organism in H. irritans represents the first record in Spain and the second from Europe. Abstract Background The horn fly Haematobia irritans is a blood-sucking ectoparasite responsible for substantial economic loss of livestock. Like other hematophagous arthropods species, the successful blood-feeding of H. irritans is highly dependent on the modulation of the host’s hemostasis and immune system. Here, we evaluated the biological activity of hematobin (HTB), a protein recently identified in the H. irritans saliva, on macrophage biology. The goal was to understand the putative interactions between the components of H. irritans saliva and the early host immune responses. Results Thioglycolate-elicited peritoneal macrophages from BALB/c mice were stimulated by lipopolysaccharide (LPS) plus interferon-γ (IFN-γ) in the presence or absence of recombinant HTB. The presence of the salivary protein in the cultures inhibited nitric oxide production and decreased the inducible nitric oxide synthase (iNOS) expression induced by LPS plus IFN-γ. The tumor necrosis factor-α (TNF-α) and interleukin-12p40 (IL-12p40) levels were also reduced in the macrophages pre-incubated with HTB; these findings correlated to the decreased NF-κB expression. The biological activities described here were not associated with changes in annexin V binding to macrophages suggesting that HTB does not induce cell death. In addition, the activity of HTB seems to be specific to macrophages because no changes were observed in lymphocyte proliferation or cytokine production. Conclusions We describe here the first bioactive salivary protein of H. irritans. We characterized its ability to modulate macrophage inflammatory response, and the results can help explain how horn flies modulate the host immune system to feed on blood.

Haematobia irritans is a cosmopolitan obligate blood-feeding ectoparasite of cattle and is the major global pest of livestock production. Currently, H. irritans management is largely dependent on broad-spectrum pesticides, which has led to the development of insecticide resistance. Thus, alternative control methods are needed. Essential oils have been studied as an alternative due to their wide spectrum of biological activities against insects. Thus, the main aim of this study was to evaluate the insecticidal, repellent and antifeedant activity of the essential oils from Blepharocalyx cruckshanksii leaves and Pilgerodendron uviferum heartwood against horn flies in laboratory conditions. The composition of the essential oils was analyzed using gas chromatography coupled to mass spectrometry. Accordingly, α-pinene (36.50%) and limonene (20.50%) were the principal components of the B. cruckchanksii essential oil, and δ-cadinol (24.16%), cubenol (22.64%), 15-copaenol (15.46%) and δ-cadinene (10.81%) were the most abundant compounds in the P. uviferum essential oil. Mortality of flies and feeding behavior were evaluated by non-choice tests, and olfactory response was evaluated using a Y-tube olfactometer. Both essential oils were toxic to horn flies, with LC50 values for B. cruckchanksii essential oil of 3.58 µL L−1 air at 4 h, and for P. uviferum essential oil of 9.41 µL L−1 air and 1.02 µL L−1 air at 1 and 4 h, respectively. Moreover, the essential oils exhibited spatial repellency in the olfactometer using only 10 µg of each oil, and these significantly reduced the horn fly feeding at all doses evaluated. Although further laboratory and field studies related to the insectistatic and insecticide properties of these essential oils against H. irritans are necessary, B. cruckshanksii leaves and P. uviferum heartwood essential oils are promising candidates for horn fly management.

We evaluated the effects of four different 6-year duration control strategies on the resistance levels and frequency of the pyrethroid target site resistance alleles, superkdr (skdr) and kdr, at four field populations of Haematobia irritans irritans (Linnaeus, 1758) (Diptera: Muscidae) in Louisiana, USA. Consecutive use of pyrethroid ear tags for 6 years caused a significant increase in the resistance ratio to pyrethroids as well as the frequencies of both skdr and kdr resistance alleles. After 3 years of consecutive use of pyrethroid ear tags, followed by 1 year with no treatment, and followed by 2 years with organophosphate ear tags, the resistance ratio for pyrethroid was not significantly affected, the %R-skdr significantly dropped while the %R-kdr allele remained relatively high and stable. Similar results were observed when pyrethroid ear tags were used for three consecutive years, followed by 1 year with no treatment, and followed by 2 years with endosulfan ear tags; however, this treatment resulted in a slight increase in the resistance ratio for pyrethroids. In a mosaic, the resistance ratio for pyrethroids showed a 2.5-fold increase but the skdr-kdr genetic profiles did not change, as the %R alleles (skdr and kdr) remained low and stable through the 6 years. Lack of exposure to pyrethroid insecticides for 3 years significantly affected the skdr mutation but not the kdr mutation, preventing re-establishment of susceptibility to pyrethroids. SS-SR (skdr-kdr) individuals were responsible for the maintenance of the kdr mutation in two of the populations studied, and fitness cost seems to strongly affect the SR-RR genotype. A 4-yr study was conducted on native rangeland to assess the growth and reproductive performance of cows (Bos taurus) infested with naturally occurring seasonal populations of horn flies (Haematobia irritans). One hundred five Angus × Hereford cow-calf pairs were evaluated as a randomized complete block that was replicated across 4 yr. Cows were approximately 39 d postpartum at the beginning of each yearly trial and were randomly allocated to either an untreated control (UTC) or an insecticide-treated (TRT) herd. Horn fly populations were monitored throughout each yearly replication and blood serum progesterone levels were used to estimate postpartum interval lengths and days to pregnancy. Initial body weights of cows were collected in May with final body weights and calf weaning weights acquired in October of each year. Monthly horn fly control ranged from 85.55 to 99.57% throughout the 4 yr. Cows within UTC herds maintained on average 530.10 ± 94.74 more (P = 0.0015) flies per animal than TRT. However, no differences were detected between treatment groups for any of the reproductive parameters evaluated (P > 0.05). Despite a lack of difference in the reproductive parameters measured, TRT cows gained more (P = 0.0492) weight throughout the fly seasons when compared to UTC cattle. Furthermore, calves paired with insecticide-treated cows tended (P = 0.0680) to wean 16.28 ± 8.04 kg heavier than calves paired with cows exposed to naturally occurring horn fly populations. Pesticides are used worldwide to control arthropod parasites in cattle herds. The indiscriminate and/or inappropriate use of pesticides without veterinary guidance is a reality in several countries of South America. Improper pesticide use increases the chances of contamination of food and the environment with chemical pesticides and their metabolites. Reduction of these contamination events is an increasing challenge for those involved in livestock production. The horn fly, Haematobia irritans (Linnaeus) (Diptera: Muscidae), is one of the most economically important parasites affecting cattle herds around the world. As such, horn fly control efforts are often required to promote the best productive performance of herds. Pesticide susceptibility bioassays revealed that pyrethroid resistance was widespread and reached high levels in horn fly populations in the Brazilian state of Rondônia. The knockdown resistance (kdr) sodium channel gene mutation was detected in all horn fly populations studied (n = 48), and the super kdr sodium channel gene mutation was found in all homozygous resistant kdr individuals (n = 204). Organophosphate resistance was not identified in any of the fly populations evaluated. The horn fly is one of the most economically important parasites affecting cattle herds around the world.Pesticide control in horn fly populations promotes excellent initial results, but the excessive use of these compounds results in insecticide resistance issues.The selection pressure imposed by pesticides means that as pesticide effectiveness increases, so do the selection for and rapidity of resistance development in horn fly populations. Haematobia irritans is an obligate bloodsucking ectoparasite of cattle and is the global major pest of livestock production. Currently, H. irritans management is largely dependent upon broad-spectrum pesticides, which lately has led to the development of insecticide resistance. Thus, alternative control methods are necessary. Endophyte-infected grasses have been studied as an alternative due to their capability to biosynthesize alkaloids associated with anti-insect activities. Thus, the main aim of this study was to evaluate the antifeedant and repellent activity of lolines obtained from endophyte-infected tall fescue against H. irritans adults in laboratory conditions. The alkaloid extract (ALKE) was obtained by acid–base extraction. N-formyl loline (NFL) and N-acetyl loline (NAL) were isolated by preparative thin layer chromatography (pTLC) and column chromatography (CC), and the loline was prepared by acid hydrolysis of a NFL/NAL mixture. Loline identification was performed by gas chromatography coupled to mass spectrometry (GC/MS). Feeding behavior was evaluated by a non-choice test, and olfactory response was evaluated using a Y-tube olfactometer. Accordingly, all samples showed antifeedant activities. NFL was the most antifeedant compound at 0.5 µg/µL and 1.0 µg/µL, and it was statistically equal to NAL but different to loline; however, NAL was not statistically different to loline. NFL and NAL at 0.25 µg/µL were more active than loline. All samples except loline exhibited spatial repellency in the olfactometer. Thus, the little or non-adverse effects for cattle and beneficial activities of those lolines make them suitable candidates for horn fly management.

The control of parasite populations affecting cattle herds is an urgent need in tropical areas, where the presence of ectoparasites is a serious threat to animal health. The tick Rhipicephalus microplus as well as the fly Haematobia irritans cause considerable economic damages, so the control of ectoparasites in animals has been very important, demanding the use of chemical agents for an effective and safe control either for animals or for the environment. The objective of this study was to determine the effectiveness of Fipronil (Effipro Bovis) in the control of R. microplus and H. irritans in cattle under field conditions, applying field rotation as a strategy. The study was carried out in a herd of 14 animals: Holstein and mestizo Holstein F1 naturally infested. Animals were divided into the control and treated group; all of them evaluated before and after treatment. Three consecutive trials were conducted, applying the treatment with Effipro Bovis, each time infestation levels were reached above 10 ticks per animal. As a result of the first trial, there was a reduction in infestation intensity to 0.9 ticks per animal (97.6%) and, for H. irritans, 0.8 flies per animal (99.1%). A high control percentage was observed in the populations of ticks R. microplus in the first 24 hours, and flies H. irritans in the days following the treatment. The product Effipro Bovis kept controlled R. microplus populations for 35 days and H. irritans for 84 days, with an intermediate treatment of the product 42 days after the application. This study reports intense horn fly infestations of horses raised in an important wetland ecosystem, Sultan Marshes in Central Anatolia, Turkey. In total, seven horses raised together were found to be harbored over 500 flies per site of each animal. Totally, 376 fly specimens were collected from the horses by using the nets and were subjected to the laboratory for species identification. All flies were morphologically identified as the adults of Haematobia irritans . Partial fragments of mitochondrial cytochrome oxidase I ( mt-COI ) gene from totally 50 isolates were amplified for sequence and phylogenetic analyses. The mt-COI sequence analyses revealed no polymorphism among the isolates and explored a unique haplotype for H. irritans . A mean haplotype and nucleotide diversities of 0.8571 and 0.00695 were determined, respectively, within the COI data set of H. irritans , and newly characterized haplotype from Turkey exhibited a mean intraspecific genetic difference of 1.0 to the all published sequences of the isolates from several countries. The horn fly, Haematobia irritans (L.), is a blood-sucking livestock ectoparasite responsible for substantial livestock losses. In the present work, the potential use of recombinant hematobin (HTB), a horn fly salivary protein, as an antigen for cattle vaccination was investigated. In this trial, horn fly loads and H. irritans's blood intake were assessed in vaccinated (n=4) and control (n=4) crossbred dark-coated steers, which were naturally infected. The vaccinated group received a 1 ml subcutaneous injection of 100 mg of HTB protein emulsified in 500 ml of Incomplete Freund Adjuvant (AIF) on days 0 and 30. The control group received on the same days 1ml of distilled water emulsified in 500 ml of AIF. The vaccinated group had significantly more HTB-specific IgG antibodies after the HTB booster and had a lower fly load than the control group (206623 vs. 285623 flies per animal, respectively). The aim of this study was to determine the efficacy of the entomopathogenic fungus, Metarhizium anisopliae strain 134 (Ma134), for dairy cattle naturally infested with Haematobia irritans (L.) An aqueous formulation of 1 x 108 conidia/ml was applied by aspersion on a group of eight cows every seven days for a total of four applications, while a control group was sprayed with the same solution without conidial content. The degree of horn fly infestation was estimated daily in both groups by the direct count of adult flies found resting or feeding on the animals, and Abbott's formula was used to calculate the effectiveness of the formulation. The Ma134 formulation resulted in a 68.6 % reduction of the fly infestation during the study period, and a significant reduction was observed from the first week post-application (p < 0.05). The results show that the Ma134 formulation is effective in reducing infestations of horn fly, H. irritans, on dairy cattle under natural conditions. The horn fly, Haematobia irritans (L.) is an important cattle pest and traditionally has been managed using insecticides; however, many horn fly populations are insecticide-resistant in United States. Use of alternative control techniques has been limited because of the challenges of managing a fly pest on pastured cattle. After the discovery of a wild horn fly infected with Beauveria bassiana in Florida, the fungus was cultured and evaluated for efficacy against laboratory-reared horn flies. This fungal strain was selected for increased virulence by passage through laboratory-reared horn fly hosts to shorten interval from infection to fly death and subsequent conidia formation, properties important to future use of the fungus as a biological control agent against horn flies. After seven passages through horn fly hosts, fly mortality was not significantly accelerated as evaluated through LT50 values, but conidia were readily produced from these killed flies. Although further development is needed to improve fungal efficacy, this fungal strain holds promise as a biological control agent for inclusion in horn fly integrated pest management programs. Bovine trypanosomiasis is a hemoparasitic disease transmitted in Latin America mainly by biting flies of the family Tabanidae. The study aimed to evaluate infection by Trypanosoma vivax and Trypanosoma evansi in cattle specialized in milk production on a farm and their potential vectors. A direct parasitological and entomological study was performed using microscopy techniques and polymerase chain reaction (PCR) with two molecular markers to differentiate Trypanosoma species in blood samples of cattle and flies. Infection frequency with Trypanosoma vivax and Trypanosoma evansi in cattle was 3.6 and 0%, respectively. Characterization of vectors shows Haematobia irritans as the most frequent fly in the study area (97.1%), followed by Stomoxys calcitrans (2.8%). No horseflies were identified. T. vivax and T. evansi were found in proboscis and thorax-abdomen of biting flies Haematobia irritans and Stomoxys calcitrans, representing an epizootic behavior, atypical in South American countries. Strains of Salmonella enterica can be subdivided into clades that differ in their genetic composition, influencing mi-crobial ecology and bacterial transmission. Salmonella serovar Montevideo strains 1110 and 304, representatives of two different clades, were used to evaluate interactions with the various stages of horn fly development. Sterilized cattle dung was inoculated with Salmonella monocultures, and horn fly larvae were exposed to 10³, 105, and 107 colony-forming units (CFU)/g per strain. Salmonella supported horn fly development, and concentration-dependent differences in pupal survival suggested that Salmonella Montevideo 304 impacts adult emergence when larvae are reared in a high concentration. Viable bacteria of each strain were quantified from larvae, pupae, and newly emerged adults. Both strains were cultured from larvae at a mean ~105, regardless of concentration, and both strains survived pupation. Quantities of Salmonella 1110 were stationary through the midpupal stage, after which quantities declined in pupae reared in 105 and increased twofold in pupae reared in 107 CFU/g. Quantities of Salmonella 304 remained stationary throughout pupal development when reared in 105, yet increased 29-fold when reared in 107 CFU/g. At high densities, properties of Salmonella 304 may influence its interaction with horn fly larvae, enabling the bacteria to evade degradation during larval gut histolysis and to subsequently proliferate during the late stages of pupal development. This may account for the observed effect on adult emergence. Horn flies, Haematobia irritans irritans (L.), and stable flies, Stomoxys calcitrans (L.) (Diptera: Muscidae), are economically important blood-feeding ectoparasites of wild and domesticated animals, including cattle, Bos taurus L. Conventional insecticides are used for control of biting flies on cattle, but safety concerns and the buildup of insecticide resistance indicate the need for alternative control tactics. Many botanical extracts and oils are composed of more than one bioactive compound that can exert different modes of action, delaying or averting resistance. Plant genera that have shown repellency and toxicity against horn flies and stable flies include Allium, Azadirachta, Chrysanthemum, Cinnamomum, Cymbopogon, Derris, Eucalyptus, Festuca, Melaleuca, Melinus, Mentha, Nepeta, Nicotiana, Pelargonium, Pogostemon, Ricinus, Rosa, Syzygium, Vitex, and Zyloxanthum. Other botanically based methods for biting fly control have been investigated, such as the use of fatty acids, soybean trypsin inhibitors, and fungal endophytes on forage grasses. Many of the plantbased control methods have been shown to have strong effects against the two biting fly species, but work has only just begun on identifying and, in particular, developing botanically based tactics. O controle eficaz de Haematobia irritans (mosca-dos-chifres) e Rhipicephalus (Boophilus) microplus (carrapato-do-boi) é um fator crucial à maior rentabilidade da pecuária brasileira, porém ainda constitui-se um desafio. Um maior conhecimento das práticas adotadas no combate destes parasitos faz-se necessário para que se possam estruturar estratégias de controle mais próximas da realidade do produtor rural e mais fáceis de serem executadas. Este estudo caracterizou, através de entrevistas, as práticas adotadas no controle desses ectoparasitos em 23 propriedades da mesorregião do Triângulo Mineiro e Alto Paranaíba, Minas Gerais. A maioria dos entrevistados reconheceu a importância desses parasitos para a atividade pecuária, entretanto, diversos problemas relativos ao controle parasitário foram observados. O controle da mosca-dos-chifres e do carrapato-do-boi era uma prática rotineira em, respectivamente, 17,4% e 95,7% das propriedades, geralmente realizado em função de elevado grau de infestação dos animais. Mais de seis aplicações de ectoparasiticidas eram realizadas por ano em todas as propriedades que empregavam tratamentos para o controle da mosca e em 76,5% daquelas que combatiam o carrapato. Os produtos eram aplicados principalmente com bombas costais manuais (63,6%) e sem a contenção dos animais em 45,5% das propriedades. A diluição dos produtos segundo recomendações dos fabricantes era realizada por 45,4% dos entrevistados, porém, 63,6% aplicavam um volume de solução por animal menor do que o tecnicamente recomendado. Utilizava-se principalmente a associação de piretróides e organofosforados para o controle de ambos os parasitos. Apesar de utilizar rotineiramente ectoparasiticidas, o uso de equipamento de proteção individual (EPI) não era comum entre os entrevistados. A maioria dos entrevistados conhecia algumas características epidemiológicas dos parasitos, entretanto, o controle parasitário adotado na maior parte das propriedades tende a comprometer não apenas a eficácia dos tratamentos, mas a suscetibilidade dos parasitos e a sustentabilidade do controle.

Avaliou-se o grau de infestação natural por ectoparasitas em fêmeas bovinas de corte de três grupos genéticos. Foram realizadas contagens, de julho de 2003 a dezembro de 2004, de carrapatos (Rhipicephalus (Boophilus) microplus), moscas-dos-chifres (Haematobia irritans) e bernes (larvas de Dermatobia hominis) em fêmeas dos grupos genéticos Nelore (NE), Angus x Nelore (AN), Canchim x Nelore (CN) e Simental xNelore (SN) infestadas naturalmente. Foram feitas de 6 a 10 contagens em cada animal, totalizando 5.384 observações em fêmeas de sete estádios fisiológicos (bezerras, novilhas vazias, novilhas prenhes, vacas primíparas, com ou sem bezerro, e vacas pluríparas, com ou sem bezerro). Os dados, transformados por log10 (n + 1), foram analisados pelo método dos quadrados mínimos, com um modelo estatístico que incluiu os efeitos de grupo genético da fêmea, de animal dentro de grupo genético (erro a), de ano-época da contagem, de estádio fisiológico e da interação grupo genético x ano-época da contagem. A diferença entre os grupos genéticos dependeu do ano-época da contagem, contudo, em geral, as fêmeas nelores foram as menos infestadas pelo carrapato e as Angus x Nelore, as mais infestadas pela mosca-dos-chifres e pelo berne. Houve efeito de estádio fisiológico da fêmea em todas as três características estudadas, e as diferenças variaram com a característica. It was realized from 6 to 10 countings in each animal, totalizing 5,384 observations in females at seven physiological stages (calf; empty heifers, pregnant heifers, primiparous cows with or without calf; and pluriparous cows, with or without calf). Data, which were transformed by log10 (n + 1), were analyzed by the least square method with a statistical model that included effects of the female genetic group, of the animal within a genetic group (error a), year-season counting, physiological state, and year-season x genetic group interaction. Differences among genetic groups depended on year-season counting; however, in general, Nellore females were the least infected by ticks, and Angus x Nellore females were the most infected by horn flies and by beef-worms. There was a physiological stage effect of the female in all the three studied characteristics and the differences changed accordingly to the trait. Despite of the counting year-season x genetic group, there are differences among Nellore, Canchim x Nelore, Angus x Nellore and Simmental x Nelore genetic groups regarded to the degree of natural infestation by cattle tick, horn fly and beef-worm.

Horn flies, Haematobia irritans, a major cattle pest in the USA, cause substantial economic losses and current control methods rely heavily on insecticides. Three horn fly populations were evaluated for insecticide susceptibility to permethrin, β-cyfluthrin, and diazinon. Susceptibility was variable by population, with the greatest resistance exhibited by a 66-fold resistance ratio (RR) to permethrin and >14-fold RR to diazinon. Mechanisms of resistance were determined using molecular techniques and enzymatic assays. The knockdown resistance (kdr) genotype (L150F) associated with pyrethroid resistance, and a G262A mutation in acetylcholinesterase, previously associated with organophosphate resistance, were found in all field populations evaluated. Insensitivity of diazoxon at the acetylcholinesterase (AChE) target site was significantly different in horn flies from one of the field sites. For metabolic detoxifying enzymes, cytochrome P450 nor general esterases showed a significant difference between field strains and a laboratory susceptible strain. Pyrethroid resistance was likely due to the presence of the L150F mutation in the population. In vitro studies targeting the AChE enzyme did not support the notion that the G262A mutation was the sole cause of resistance to organophosphates, and, therefore, the exact resistance mechanism to diazinon was not able to be confirmed.

Este estudo teve como objetivo conhecer as espécies de muscídeos simbovinos e a área de eleição destas sobre o corpo do bovino de leite. Também estimou-se a flutuação populacional das espécies de moscas, relacionando-a com as médias mensais de temperatura e umidade relativa, bem como a precipitação pluviométrica em Capão do Leão, Rio Grande do Sul, Brasil. Para estimar a sazonalidade e a localização sobre bovinos, foram examinadas 20 vacas leiteiras a cada 30 dias, no período de abril/2007 a março/2008, conforme as divisões zootécnicas por meio da observação direta dos dípteros. Os muscídeos simbovinos verificados foram: Haematobia irritans, espécie mais abundante e encontrada em maior frequência na costela/no flanco, seguida por Stomoxys calcitrans, com maior ocorrência nos membros dianteiros e traseiros e Musca domestica, na cabeça, no ventre e na costela/no flanco dos bovinos. A sazonalidade foi influenciada principalmente pela temperatura, exercendo forte impacto negativo em situações extremas. Na temperatura média menor ou igual a 12,7°C e na temperatura acima de 22,5°C, não houve observação de dípteros.This study aimed to describe the symbovine species and which area of the dairy cattle body these flies selectes. In addition the fluctuation of these species population was estimated relative to the average monthly temperature, humidity and rainfall in Capão do Leão, Rio Grande do Sul, Brazil. To estimate seasonality and location on the cattle, 20 dairy cattle were examined through flies' direct observation every 30 days in the period from April/2007 to March/2008 according to the animal zootechnical divisions. The symbovine muscids checked were: Haematobia irritans, the most abundant species and found in greater frequency on the rib/flank, followed by Stomoxys calcitrans, with higher occurrence on the front and rear legs, and Musca domestica, on the head, belly and rib/flank. Seasonality was mainly influenced by temperature which had a strong and negative impact in extreme situations. There was no observation of flies with average temperature less than or equal to 12.7°C and above 22.5°C.

To investigate the kdr (knockdown resistance) resistance-associated gene mutation and determine its frequency in pyrethroid-resistant horn fly (Haematobia irritans) populations, a total of 1,804 horn flies of 37 different populations from all Brazilian regions (North, Northeast, Central-West, Southeast, and South) were molecular screened through polymerase chain reaction (PCR). The kdr gene was not detected in 87.08% of the flies. However, the gene was amplified in 12.92% of the flies, of which 11.70% were resistant heterozygous and 1.22% were resistant homozygous. Deviation from Hardy-Weinberg equilibrium (HWE) was found only in 1 ranch with an excess of heterozygous. When populations were grouped by region, three metapopulations showed significant deviations of HWE (Central-West population, South population and Southeast population). This indicates that populations are isolated one from another and kdr occurrence seems to be an independent effect probably reflecting the insecticide strategy used by each ranch. Although resistance to pyrethroids is disseminated throughout Brazil, only 48% of resistant populations had kdr flies, and the frequency of kdr individuals in each of these resistant populations was quite low. But this study shows that, with the apparent exception of the Northeast region, the kdr mechanism associated with pyrethroid resistance occurs all over Brazil.Com o objetivo de verificar a ocorrência e determinar a frequência da mutação kdr (knock down resistance) em populações de Haematobia irritans (mosca-dos-chifres) resistentes aos piretróides, foram analisados 1.804 indivíduos de 37 populações de todas as Regiões do Brasil. Com exceção da Região Nordeste, o kdr (knock down resistance gene) foi encontrado em populações de todas as regiões. A mutação não foi detectada em 87,08% dos indivíduos. Entretanto, o gene foi amplificado de 12,92% das moscas, das quais 11,70% se mostraram heterozigotas resistentes e 1,22% homozigotas resistentes. Em todas as populações verificou-se equilíbrio de acordo com a Lei de Hardy e Weinberg, exceto uma com excesso de heterozigotos. Entretanto, quando agrupamos diferentes populações numa metapopulação de acordo com a região geográfica, é possível observar um desvio nas populações Centro-Oeste, Sul e Sudeste, indicando isolamento populacional e que a ocorrência do kdr é provavelmente um efeito independente, talvez refletindo a estratégia de uso do inseticida de cada produtor. Apesar da resistência aos piretróides estar disseminada por todo o país, apenas 48% das populações resistentes apresentaram o kdr, e a frequência de indivíduos kdr nas populações resistentes se mostrou bastante baixa. À exceção da Região Nordeste, o mecanismo de resistência ligado ao kdr ocorre em todo o país.

Dezesseis vacas de raça Holandesa em lactação foram utilizadas para avaliar o efeito da administração de alho na alimentação sobre a alteração da carga de carrapato, mosca-do-chifre, mosca dos estábulos e mosca doméstica. Os tratamentos (T) foram constituídos por controle negativo (T1), alho a 100 g (T2) e 200 g vaca-1 (T3), por três dias, e o amitraz a 0,025% (T4). Para avaliação, foram contadas as fêmeas ingurgitadas de carrapato com comprimento superior a quatro mm e as moscas, antes da aplicação e no 1, 2, 3, 4, 5, 6, 7 e 14º dia após a aplicação de cada tratamento. Nos 1, 2 e 3º dias pós-tratamento foram coletadas amostras de leite para avaliação das propriedades organolépticas do leite cru e do queijo Minas-Frescal. Os valores médios de carrapatos, em comparação ao pré-tratamento, foram de 188,44; 106,28; 108,10 e 7,60%, respectivamente. O ectoparasiticida comercial foi o mais eficaz (p0,05) ao produto químico (T4). Nestes tratamentos, não foi detectado odor ou sabor de alho no leite e no queijo. Não houve diferença (p>0,05) entre os tratamentos constituídos no controle de moscas.Sixteen lactating Holstein cows were used to evaluate the effect of garlic supply on the load of tick, horn fly, stable fly and house fly. Treatments (T) consisted of a negative control (T1), garlic at 100 g (T2) and 200 g cow-1 (T3) for three days, and Amitraz 0.025% (T4). Engorged female ticks presenting length superior to 4.0 mm and flies were counted before application and on the 1st, 2nd, 3rd, 4th, 5th, 6th, 7th, and 14th day after each treatment application. On the 1st, 2nd, 3rd days after treatments, milk samples were collected to evaluate organoleptic properties in raw milk and in "Minas-Frescal" cheese. Tick mean values were 188.44, 106.28, 108.10, and 7.60%, respectively, relative to those of the pretreatment. Commercial ectoparasiticide was the most efficient (p0.05) to that of the chemical product (T4). In these treatments, no garlic flavor or taste was detected in milk and cheese. There was no difference (p>0.05) among treatments for fly control.

Haematobia irritans irritans (Linnaeus, 1758: Diptera: Muscidae), the horn fly, is an external parasite of penned and pastured livestock that causes a major economic impact on cattle production worldwide. Pesticides such as synthetic pyrethroids and organophosphates are routinely used to control horn flies; however, resistance to these chemicals has become a concern in several countries. To further elucidate the molecular mechanisms of resistance in horn fly populations, we sequenced the transcriptomes of ten populations of horn flies from the southern US possessing varying degrees of pesticide resistance levels to pyrethroids, organophosphates, and endosulfans. We employed an Illumina paired end HiSeq approach, followed by de novo assembly of the transcriptomes using CLC Genomics Workbench 8.0.1 De Novo Assembler using multiple kmers, and annotation using Blast2GO PRO version 5.2.5. The Gene Ontology biological process term Response to Insecticide was found in all the populations, but at an increased frequency in the populations with higher levels of insecticide resistance. Horn fly susceptibility to insecticides was evaluated in the state of Mato Grosso do Sul, Brazil, from October 2000 to September 2002. Insecticide bioassays (n=57) were conducted in 38 ranches from 14 municipalities throughout the state. Horn flies from wild populations were collected on cattle and exposed to filter papers impregnated with cypermethrin, permethrin, or diazinon and mortality was assessed after two hours. Resistance to cypermethrin was detected in all populations, with resistance ratios (RR) ranging from 27.6 to 91.3-fold. Blood-sucking diptera are important parasites in bovine production systems, especially regarding confinement conditions. Haematobia irritans, the horn fly, is one of the most troublesome species within bovine production systems, due to the intense stress imposed to the animals. An important aspect while studying the variability within a species is the study of the geographic structure of its populations and, attempting to find out the genetic flow of Brazilian populations of horn fly, the RAPD technique, which is suited for this purpose, has been used. The use of molecular markers generated from RAPD made it possible to identify the geographic origin of samples from different Brazilian geographic regions, as well as to estimate the genotypic flow among the different Brazilian populations of the horn fly.Dípteras hematófagos são importantes parasitas dentro de sistemas de produção de bovinos, especialmente em confinamento. A utilização dos marcadores moleculares gerados através da técnica de RAPD-PCR tornou possível a identificação da origem geográfica das amostras das diferentes regiões geográficas brasileiras, assim como, estimar o fluxo genotípico entre as diferentes populações brasileiras da mosca-dos-chifres.

The objective of the present study was to determine the population dynamics and the infestation extent of the fly H. irritans in naturally infested bovine. Thirty percent of the bovine mass of the units selected were sampled twice a month, and flies were counted by direct observation. The units chosen were representative from the North, Center and South of each province of the country. The climatic variables of average temperature and rainfall were correlated to the determined population dynamics. It was found that the fly H. irritans was present the whole year, with an intensity of 39.5 ±12.2 flies/animal. There a positive and direct relationship between the climatic variables of average temperature (r = 0.73), rainfalls (r = 0.63) and the presence of the fly on the animals. The months with the highest infestation intensity were during the period June-September, coinciding with the warmest and rainiest months in the country. The presence of the fly was revealed in 72.7% of the units sampled with 9.9% of high, 43.5% medium and 46.7% low infestations. [ABSTRACT FROM AUTHOR]

The horn fly, Haematobia irritans, is a serious pest of cattle in North America. The control of horn flies has primarily relied on insecticides. However, the heavy use of insecticides has led to the development of insecticide resistance in horn flies. Novel methods to control horn flies are greatly needed. Transgenic technology is an effective tool to genetically modify insects and may lead to novel methods of pest control based on genomic approaches. Here we report a piggyBac-mediated transformation of the horn fly via electroporation. Transformation with a DsRed fluorescent marker protein coding region was verified by PCR analysis of individual fly bodies and pupal cases and sequencing of PCR products. However, Southern blot analysis failed to indicate the DsRed gene was integrated into the horn fly genome. Thus, the electroporation protocol may have caused the DsRed gene to be integrated into bacterial symbionts of the horn fly. [ABSTRACT FROM AUTHOR]

In this study, we investigated the impact of endophyte ( Neotyphodium coenophialum) infection of cattle pasture upon the survival of the horn fly, Haematobia irritans, a major agricultural pest affecting livestock in many parts of the world. RESULTS In laboratory assays, where cattle dung collected from endophyte-infected (E+) tall fescue cultivar K-31 was used as the oviposition substrate, larval development was significantly reduced compared with development on cattle dung from steers that grazed uninfected (E−) tall fescue. Furthermore, studies with cattle dung supplemented with the alkaloid fraction extracted from the endophytic fungi revealed significant larval mortality, and HPLC analysis identified two alkaloids, peramine and lolitrem B. The development of larvae was shown to be significantly reduced in field-collected cattle dung. These results suggest that part of the toxicity of alkaloids contained in endophytes is transferred to faecal matter, causing an increase in mortality of H. irritans. Estudaram-se o comportamentos e os danos causados pela mosca-dos-chifres em vacas e bezerros Nelore antes da desmama. Foram utilizados dois grupos de 60 animais cada um, sendo um grupo com infestação natural de moscas e o outro sem moscas, por quatro anos consecutivos. Observou-se que o número de moscas das vacas não ultrapassou a média de 80 moscas/animal, em todos os anos experimentais. A maioria das vacas (83%) apresentou poucas moscas, enquanto a minoria (17%) teve maior quantidade. Os bezerros foram pouco infestados pelas moscas. Chuvas acima de 100 mm em curto espaço de tempo diminuíram a infestação de moscas. O ganho de peso das vacas tratadas com inseticida (sem infestação por moscas) bem como os seus bezerros foi maior do que as vacas controle, porém não foi estatisticamente significativo (P>0,05). A percentagem média de prenhez, nos quatro anos, das vacas tratadas foi 15% superior a das não tratadas e esta diferença foi causada provavelmente pela infestação de moscas nos touros.

Um teste de campo, realizado simultaneamente no Brasil (Eldorado do Sul, Rio Grande do Sul) e Argentina (Rafaela, Santa Fé), avaliou o comportamento da doramectina injetável sobre insetos adultos de Haematobia irritans. Em ambos os locais, utilizaram-se 40 animais (20 vacas e 20 novilhas), sendo que 20 foram tratados e 20 permaneceram como controle. Realizaram-se contagens de moscas nos dias 0, 1, 4, 7, 10, 14, 21, 28 e 35 pós-tratamento. Em comparação com o grupo controle, a eficácia observada no Brasil foi inferior a 50% no dia 1, e 84,4, 72,6, 81,4, 74,2, 79,2, 50,7 e 64,7% nos demais dias de observação enquanto na Argentina, foi de 97,3, 84,2, 95,9, 92,2, 93,6, 63,3, 73, 8 e 66,9%, respectivamente. Em ambos os países, ocorreu uma contribuição dessa avermectina para a diminuição da população de insetos adultos de H. irritans embora os resultados obtidos no Brasil tenham apresentado uma eficácia menor que os constatados na Argentina.
