The entomopathogenic fungus Conoideocrella luteorostrata has recently been implicated in natural epizootics among exotic elongate hemlock scale (EHS) insects in Christmas tree farms in the eastern United States. Since 1913, C. luteorostrata has been reported from various plant feeding Hemiptera in the southeastern United States, but comprehensive morphological and phylogenetic studies of U.S. populations are lacking. The recovery of multiple strains of C. luteorostrata from mycosed EHS in North Carolina provided an opportunity to conduct pathogenicity assays and morphological and phylogenetic studies to investigate genus- and species-level boundaries among the Clavicipitaceae. Pathogenicity assays confirmed that C. luteorostrata causes mortality of EHS crawlers, an essential first step in developing this fungus as a biocontrol. Morphological studies revealed that conidia aligned with previous measurements of the Paecilomyces-like asexual state of C. luteorostrata, with conidiophore morphology consistent with historical observations. Additionally, a Hirsutella-like synanamorph was observed in select C. luteorostrata strains. In both a four-locus, 54-taxon Clavicipitaceae-wide phylogenetic analysis including D1-D2 domains of the nuclear 28S rRNA region (28S), elongation factor 1 alpha (EF1-α), DNA-directed RNA polymerase II subunit 1 (RPB1), and DNA-directed RNA polymerase II subunit 2 (RPB2) and a two-locus, 38-taxon (28S and EF1-α) phylogenetic analysis, all three Conoideocrella species were resolved as strongly supported monophyletic lineages across all loci and both methods (maximum likelihood and Bayesian inference) of phylogenetic inference except for 28S for C. tenuis. Despite the strong support for individual Conoideocrella species, none of the analyses supported the monophyly of Conoideocrella with the inclusion of Dussiella. Due to the paucity of RPB1 and RPB2 sequence data, EF1-α provided superior delimitation of intraspecies groupings for Conoideocrella and should be used in future studies. Further development of C. luteorostrata as a biocontrol against EHS will require additional surveys across diverse Hemiptera and expanded pathogenicity testing to clarify host range and efficacy of this fungus.

BackgroundTrichoderma reesei is an organism extensively used in the bioethanol industry, owing to its capability to produce enzymes capable of breaking down holocellulose into simple sugars. The uptake of carbohydrates generated from cellulose breakdown is crucial to induce the signaling cascade that triggers cellulase production. However, the sugar transporters involved in this process in T. reesei remain poorly identified and characterized.ResultsTo address this gap, this study used temporal membrane proteomics analysis to identify five known and nine putative sugar transporters that may be involved in cellulose degradation by T. reesei. Docking analysis pointed out potential ligands for the putative sugar transporter Tr44175. Further functional validation of this transporter was carried out in Saccharomyces cerevisiae. The results showed that Tr44175 transports a variety of sugar molecules, including cellobiose, cellotriose, cellotetraose, and sophorose.ConclusionThis study has unveiled a transporter Tr44175 capable of transporting cellobiose, cellotriose, cellotetraose, and sophorose. Our study represents the first inventory of T. reesei sugar transportome once exposed to cellulose, offering promising potential targets for strain engineering in the context of bioethanol production.

The use of pathogens and other natural enemies to suppress pest populations is a key component of integrated pest management (IPM) strategies. As biocontrol agents may interact synergistically, additively, or antagonistically, it is crucial to assess their interactions for effective pest management. In this study, we investigated the effect of three entomopathogenic fungus (EPF) species—Beauveria bassiana (Bals.‐Criv.) Vuill. CEP 091, Akanthomyces muscarius (Petch) Spatafora, Kepler & B. Shrestha CEP 182, and Cordyceps fumosorosea (Wize) Kepler, B. Shrestha & Spatafora CEP 315 (all Hypocreales)—on the survival, fecundity, and searching behavior of Orius insidiosus (Say) (Hemiptera: Anthocoridae). All three EPF species were found to be pathogenic to O. insidiosus, resulting in a significant decrease in survival compared with the control group. Fecundity remained unaffected by the fungal treatments. No effect of EPF on searching behavior of the predator was found. In relation to the searching behavior, individuals of 2 days post‐infections remained in the food patch longer than individuals of 7 days post‐infections, regardless of treatment. These findings provide valuable insights into the compatibility of EPF and O. insidiosus as combined biocontrol agents for managing whiteflies, aphids, and thrips. Understanding the interactions between EPF and predatory insects is crucial for developing IPM strategies that maximize pest control efficacy. One of our studies on lignicolous aquatic fungi in the Philippines yielded two new records of freshwater Sordariomycetes in the genera Sporoschisma and Trichoderma. The newly collected specimens, UPVMI 24–0003 and UPVMI 24–0004, exhibit morphological similarities to the holotypes of Trichoderma pinicola (SFC 20130926-S233) and Sporoschisma chiangraiense (MFLU 18–1722), respectively. Combined ITS-TEF1-α phylogenetic analysis showed that UPVMI 24–0003 clustered with the ex-type strain of T. pinicola (KACC 48486), and with T. simplex (HMAS 248842) as a sister taxon. The isolate UPVMI 24–0004 grouped with three strains of S. chiangraiense (MFLUCC 18-0703, MFLUCC 18-0336, and GMB0090), with 97% ML and 1.00 BYPP bootstrap supports in the ITS phylogeny. We provide detailed descriptions and illustrations for both T. pinicola and S. chiangraiense, along with a synopsis of morphological characters for freshwater Sporoschisma species. Natural products biosynthesized from biocontrol fungi in the rhizosphere can have both beneficial and deleterious effects on plants. Herein, we performed a comprehensive analysis of natural product biosynthetic gene clusters (BGCs) from the widely used biocontrol fungus Trichoderma afroharzianum T22 (ThT22). This fungus encodes at least 64 BGCs, yet only seven compounds and four BGCs were previously characterized or mined. We correlated 21 BGCs of ThT22 with known primary and secondary metabolites through homologous BGC comparison and characterized one unknown BGC involved in the biosynthesis of eujavanicol A using heterologous expression. In addition, we performed untargeted transcriptomics and metabolic analysis to demonstrate the activation of silent ThT22 BGCs via the "one strain many compound" (OSMAC) approach. Collectively, our analysis showcases the biosynthetic capacity of ThT22 and paves the way for fully exploring the roles of natural products of ThT22.

The Old-World bollworm, Helicoverpa armigera (Lepidoptera: Noctuidae), is a significant threat to crops worldwide and has become resistant to traditional synthetic insecticides. The present study investigated the pathogenicity of the entomopathogenic fungi (EPF) Metarhizium robertsii (Hypocreales: Clavicipitaceae) strain WG-04 or Beauveria bassiana (Hypocreales: Cordycipitaceae) strain WG-10, and an entomopathogenic nematode (EPN) species Heterorhabditis bacteriophora against the second and fourth instar larvae of H. armigera. Both fungal species and H. bacteriophora were evaluated, singly or in combination. After 24 and 48 h post-application of the fungal spores (106 spores/mL), H. bacteriophora was introduced at a rate of 50 infective juveniles/mL, and mortality was observed at 3-, 5-, and 7-days post-treatment. Adult emergence, egg hatching, and percentage pupation were recorded. The integration of both types of biocontrol agents exhibited additive and synergistic interactions in larval stages, and enhanced mortality was recorded when EPF was used in combination with the nematodes. In the individual application of all three biocontrol agents alone, the order of efficacy was H. bacteriophora > B. bassiana > M. robertsii; however, in joint treatments, the increase in mortality and decrease in percentage egg hatching, pupation, and adult emergence was determined to be directly linked to the exposure period of the H. bacteriophora. The results of this study suggest that combining H. bacteriophora with EPF could provide a solid foundation for an economically viable method for managing H. armigera larvae in chickpea fields. In the course of our ongoing research targeting the identification of potential biocontrol agents from entomopathogenic fungi (EPF), we explored a solid-state rice fungal extract of Blackwellomyces roseostromatus BCC56290 derived from infected lepidopteran larvae. Chemical and biological prospections afforded four unprecedentedly reported natural products differentiated into a dimeric naphthopyran bioxanthracene ES-242 derivative (1) and three cyclodepsipeptides (2–4) in addition to two known cyclodepsipeptides, cardinalisamides B (5) and C (6). Chemical structures of the isolated compounds were elucidated through comprehensive 1D/2D NMR and HR-ESI-MS data together with comparisons to the reported literature. The absolute configuration of the isolated cyclodepsipeptides was determined using Marfey's method. Nematophagous fungi (NF) form part of the soil microbiota and are natural enemies of nematodes, helping to regulate nematode populations. A verticillate NF isolated from soil from Tepalcingo, Mexico, was morphologically and molecularly characterised. This fungus was cultured in two different liquid media—Czapek-Dox broth (CzDoxB) and sweet potato dextrose broth (SPDB)—for 21 days. The ovicidal (OA) and larvicidal (LA) activities of fungal liquid culture filtrates (LCFs) were assessed in 96-well microtitre plates at different concentrations against Haemonchus contortus after 48 h. The morphological and molecular identification revealed the presence of Lecanicillium psalliotae. Additionally, the groups of compounds associated with nematocidal activity were determined from a qualitative chemical profile (QCP) using different reagents. The highest OA of the LCFs was obtained at 25 mg/mL from SPDB and CzDoxB and amounted to 97.2 and 99.06%, respectively. Meanwhile, the highest LA recorded with these LCFs at 100 mg/mL was 54.27% and 96.8%, respectively. The QCP revealed the presence of alkaloids and tannins in both LCFs that have previously been associated with nematocidal activity. Lecanicillium psalliotae exerted an important effect on H. contortus and could be of significance in future studies focused on the control and prevention of haemonchosis in small ruminants. Diaphorina citri Kuwayama (Asian citrus psyllid, ACP) is a devastating pest due to transmission of Liberibacter spp. to citrus plants. We identified presumed entomopathogenic fungi (EF) naturally associated with field-collected adult ACP, both killed (mycosed) and alive, at 14 field sites in the Gulf Coastal Plain of NE Mexico. On dead ACP, five species of EF (Hypocreales) were observed: Hirsutella citriformis Speare (Ophiocordycipitaceae), Cordyceps javanica (Frieder. & Bally) Kepler, Shrestha & Spatafora, Beauveria cf. bassiana (Bals.) Vuill., and Akanthomyces cf. lecanii (Cordycipitaceae); also, Niveomyces J.P.M. Araújo & C. de Bekker (Cordycipitaceae), a hyperparasite of Ophiocordycipitaceae, in this case H. citriformis. The most prevalent fungi on field ACP were H. citriformis and C. javanica, on dead and on live, incubated insects, respectively. In laboratory bioassays, C. javanica and H. citriformis conidia from cultures killed >60% of ACP adults. H. citriformis, C. javanica, and A. lecanii killed >60% nymphs. Niveomyces was observed only growing on H. citriformis fructifications on field-killed ACP; it was absent from live incubated insects, indicating Niveomyces is a mycoparasite on H. citriformis. Niveomyces cultures (ARSEF 10230) on agar grew as a Pleurodesmospora synanamorph; its LSU rRNA region D1–D2 sequences (Genbank OM780101) were 99.81% identical to Niveomyces sequences. Conidia from Niveomyces cultures were entomopathogenic on ACP nymphs and adults. These are the first reports of Niveomyces associated with H. citriformis; of the in vitro dimorphic growth (synanamorphs) Niveomyces/Pleurodesmospora; and of experimental verification of entomopathogenicity of Niveomyces. This study aims to report three new species of Conoideocrella and Moelleriella from Yunnan Province, Southwestern China. Species of Conoideocrella and Moelleriella parasitize scale insects (Coccidae and Lecaniidae, Hemiptera) and whiteflies (Aleyrodidae, Hemiptera). Based on the phylogenetic analyses of the three-gene nrLSU, tef-1α, and rpb1, it showed one new record species (Conoideocrella tenuis) and one new species (Conoideocrella fenshuilingensis sp. nov.) in the genus Conoideocrella, and two new species, i.e., Moelleriella longzhuensis sp. nov. and Moelleriella jinuoana sp. nov. in the genus Moelleriella. The three new species were each clustered into separate clades that distinguished themselves from one another. All of them were distinguishable from their allied species based on their morphology. Morphological descriptions, illustrations, and comparisons of the allied taxa of the four species are provided in the present paper. The Western flower thrips (WFT), Frankliniella occidentaliss (Pergande) (Thysanoptera: Thripidae), is the main pest of many vegetables and ornamental plants in greenhouses in Antalya province (southwestern part of Turkey). The objectives of this study were: (i) to isolate indigenous Entomopathogenic fungi (EPFs) from naturally infected individuals of WFT in the coastal area of Antalya, where greenhouse cultivation is intensively done, (ii) to identify EPF isolates using DNA sequencing, and (iii) to assess the effectiveness of the isolates against various developmental stages [larvae (1st and 2nd instars), pupa and adult (only females)] of WFT under laboratory conditions. In pathogenicity assays, all isolates were tested at three different conidial concentrations (1 × 105, 1 × 106 and 1 × 107 conidia/ml), using the spray method. Two Beauveria bassiana (Balsamo) Vuillemin (Deuteromycota: Hyphomycetes) isolates (M48 and M49) and Isaria fumosorosea Wise formerly [formerly known as Paecilomyces fumosoroseus (Wise) (Ascomycota: Hypocreales)] isolate (M50) were the most pathogenic, causing 100% mortality of WFT larvae within 7–10 days at the highest concentration (1 × 107 conidia/ml). These three isolates also caused a mortality of more than 85% in the pupae of WFT. The Lecanicillium psalliotae (Treschew) Zare & W. Gams (Ascomycota: Hypocreales) isolate (DOA3) was the most virulent against adult females of WFT, causing 100% morality at the highest concentration 10 days after treatment. The current study's findings suggested that certain EPF extracted from WFT cadavers may provide an important foundation for the creation of bioproducts, as well as a possible alternative technique for regulating WFT. Amphichorda has been previously accepted as a member of the Cordycipitaceae and currently it is considered a member of the Bionectriaceae. The substrates of Amphichorda were complex and varied, being mainly animal faeces. This study reports two new species of Amphichorda from Yunnan Province in south-western China. Based on the five-gene (nrSSU, nrLSU, tef‐1α , rpb1 and rpb2) sequence and ITS data phylogenetic analysis, two new species, namely A. excrementa and A. kunmingensis, are proposed and a detailed description of the new species is provided. Amphichorda excrementa and A. kunmingensis were isolated from animal faeces in the park. One of the most commonly used fungi in pest control, which has a wide range of hosts, is Beauveria bassiana. This entomopathogen has adapted to different environments and can be found in several places, so in the present work, 60 soil samples were collected in 2020 in Acatlán de Pérez Figueroa, Oaxaca, by means of Galleria mellonella, used as bait for its isolation. Five isolates were obtained from this procedure, which were evaluated in larvae of Galleria mellonellaat 24, 48, 72, 96, 120, 144, and 168 h in order to know their efficacy in mortality and mycosis. In the methodology, the DNA of the fungi was extracted and the ITS region of the 5.8S rRNA gene was amplified by PCR, the product was sequenced and the sequences were compared with others existing at the National Center for Biotechnological Information. The result was one isolate of Beauveria pseudobassiana and four of Beauveria bassiana, of which isolate 11 showed 100% mortality and mycosis at 96 h, unlike the commercial isolate and the control, which needed an additional 72 h to kill all larvae, in addition to showing 20% and 60% of mycosis, respectively. The time of 96 h was significantly different (p# 0.05) because during this time, most of the larvae died and mycosis occurred. Trichoderma afroharzianum (Hypocreales) is known as an important mycoparasite and biocontrol fungus and feeds on fungal material by parasitizing other fungi. Recent studies indicate that this species is also an ear rot pathogen in Europe. Here, the complete mitochondrial genome of three T. afroharzianum strains was sequenced using next-generation sequencing and comparatively characterized by the reported Trichoderma mitogenomes. T. afroharzianum mitogenomes were varying between 29 511 bp and 29 517 bp in length, with an average A + T content of 72.32%. These mitogenomes contain 14 core protein coding genes (PCGs), 22 tRNAs, two rRNAs, one gene encoding the ribosomal protein S3, and three or four genes including conserved domains for the homing endonucleases (HEGs; GIY-YIG type and LAGLIDADG type). All PCGs are initiated by ATG codons, except for atp8, and all are terminated with TAA. A significant correlation was observed between nucleotide composition and codon preference. Four introns belonging to the group I intron class were predicted, accounting for about 14.54% of the size of the mitogenomes. Phylogenetic analyses confirmed the positions of T. afroharzianum strains within the genus of Trichoderma and supported a sister group relationship between T. afroharzianum and T. simmonsii. The recovered trees also supported the monophyly of all included families and of the genus of Acremonium. Species of the family Polycephalomycetaceae grow on insects or entomopathogenic fungi and are distributed from tropical to subtropical regions. This study proposed four new species of hyperparasitic fungi from China based on six molecular markers (ITS, SSU, LSU, TEF-1α, RPB1 and RPB2) phylogenetic analyses and morphological characteristics. The four new species, i.e. Pleurocordyceps litangensis, Polycephalomyces jinghongensis, Po. multiperitheciatae and Po. myrmecophilus, were described and illustrated. Pl. litangensis, exhibiting a hyperparasitic lifestyle on Ophiocordyceps sinensis, differed from Pleurocordyceps other species in producing subulate β-phialides and ovoid or elliptic α-conidia. Po. jinghongensis was distinct from Polycephalomyces other species, being parasitic on Ophiocordyceps sp., as producing oval or long oval-shaped α-conidia and columns of β-conidia. Po. multiperitheciatae differed from Polycephalomyces other species as having synnemata with fertile head, linear β-conidia and parasitic on Ophiocordyceps multiperitheciata. Po. myrmecophilus was distinct from Polycephalomyces other species, being parasitic on the fungus Ophiocordyceps acroasca, as producing round or ovoid α-conidia and elliptical β-conidia without synnemata from the colonies. These four species were clearly distinguished from other species in the family Polycephalomycetaceae by phylogenetic and morphological characteristics. The complete mitochondrial genome of Clonostachys farinosa was assembled using the strain CSC22A0184 that was isolated from the lichen Parmotrema clavuliferum. The total length of the mitogenome was 51,551 bp and contained 49 genes: 15 protein-coding genes, two ribosomal RNA genes, 26 transfer RNA genes, and six open reading frames. The GC content of the mitogenome was 28.5% and had a base composition of 36.4% A, 12.6% C, 15.9% G, and 35.1% T. Phylogenetic analysis based on 14 protein-coding genes indicates that C. farinosa is clustered in the family Bionectriaceae. This is the first study of the mitogenome of C. farinosa, which is helpful for understanding the evolution of species within the genus Clonostachys. [ABSTRACT FROM AUTHOR]

Tenebrio molitor L., also known as the mealworm, is a polyphagous insect pest that infests various stored grains worldwide. Both the adult and larval stages can cause significant damage to stored grains. The present study focused on isolating entomopathogenic fungi from an infected larval cadaver under environmental conditions. Fungal pathogenicity was tested on T. molitor larvae and pupae for 12 days. Entomopathogenic fungi were identified using biotechnological methods based on their morphology and the sequence of their nuclear ribosomal internal transcribed spacer (ITS). The results of the insecticidal activity indicate that the virulence of fungi varies between the larval and pupal stages. In comparison to the larval stage, the pupal stage is highly susceptible to Metarhizium rileyi, exhibiting 100% mortality rates after 12 days (lethal concentration 50 [LC50] = 7.8 × 106 and lethal concentration 90 (LC90) = 2.1 × 1013 conidia/mL), whereas larvae showed 92% mortality rates at 12 days posttreatment (LC50 = 1.0 × 106 and LC90 = 3.0 × 109 conidia/mL). The enzymatic analyses revealed a significant increase in the levels of the insect enzymes superoxide dismutase (4.76–10.5 mg−1) and glutathione S‐transferase (0.46–6.53 mg−1) 3 days after exposure to M. rileyi conidia (1.5 × 105 conidia/mL) compared to the control group. Abstract Background The stink bugs, Nezara viridula L. and Piezodorus guildinii Westwood (Hemiptera: Pentatomidae) are the most important and widespread species of polyphagous stink bugs in the tropical and subtropical regions of Latin America, which affect the quality and yield of the common bean (Phaseolus vulgaris L.). The use of synthetic chemical insecticides is the major control strategy to manage stink bugs in common beans and alternatives are needed. In this study, mortality and median Lethal Time (LT50) of two Cuban isolates of the entomopathogenic fungus Beauveria bassiana (18 S-O-R and 96 P-O-E), as well as one commercial Cuban isolate (Bb-18), at a concentration of 1 × 108 conidia/ml were evaluated. These evaluations were conducted against both stink bug species using Petri dish bioassays and a semi-field experiment in common beans. Results In Petri dish bioassays, the isolates 18 S-O-R and 96 P-O-E caused 100% mortality of both N. viridula and P. guildinii. This was significantly higher than for isolate Bb-18, which caused 86.3% N. viridula and 81.3% P. guildinii mortality. In the semi-field experiment, when pooling both stink bug species, total mortality after 14 days was 91.3% for 18 S-O-R, 80.0% for 96 P-O-E and 73.8% for Bb-18 isolates. LT50 value for isolate 18 S-O-R tested under laboratory conditions was 6.04 ± 0.18 days for N. viridula and 5.32 ± 0.14 days for P. guildinii at the same concentration of 1 × 108 conidia/ml. LT50 value for isolate 18 S-O-R in semi field was 6.79 ± 0.37 days for N. viridula and 7.71 ± 0.32 days for P. guildinii at 1 × 108 conidia/ml. Conclusion The study highlights the potential of B. bassiana 18 S-O-R as a promising candidate for control of stink bugs in common bean under tropical conditions as an alternative to conventional chemical insecticides in integrated pest management (IPM) programs. Moving forward, further research should focus on validating the efficacy under diverse field conditions and integrating application methods into practical IPM approaches. Future use of B. bassiana will enhance sustainability and reduce environmental impacts associated with pesticide use.

Trichoderma spp. are ubiquitous rhizosphere fungi capable of producing several classes of secondary metabolites that can modify the dynamics of the plant-associated microbiome. However, the bacterial-fungal mechanisms that mediate these interactions have not been fully characterized. Here, a random barcode transposon-site sequencing (RB-TnSeq) approach was employed to identify bacterial genes important for fitness in the presence of Trichoderma atroviride exudates. We selected three rhizosphere bacteria with RB-TnSeq mutant libraries that can promote plant growth: the nitrogen fixers Klebsiella michiganensis M5aI and Herbaspirillum seropedicae SmR1, and Pseudomonas simiae WCS417. As a non-rhizosphere species, Pseudomonas putida KT2440 was also included. From the RB-TnSeq data, nitrogen-fixing bacteria competed mainly for iron and required the siderophore transport system TonB/ExbB for optimal fitness in the presence of T. atroviride exudates. In contrast, P. simiae and P. putida were highly dependent on mechanisms associated with membrane lipid modification that are required for resistance to cationic antimicrobial peptides (CAMPs). A mutant in the Hog1-MAP kinase (Δtmk3) gene of T. atroviride showed altered expression patterns of many nonribosomal peptide synthetase (NRPS) biosynthetic gene clusters with potential antibiotic activity. In contrast to exudates from wild-type T. atroviride, bacterial mutants containing lesions in genes associated with resistance to antibiotics did not show fitness defects when RB-TnSeq libraries were exposed to exudates from the Δtmk3 mutant. Unexpectedly, exudates from wild-type T. atroviride and the Δtmk3 mutant rescued purine auxotrophic mutants of H. seropedicae, K. michiganensis and P. simiae. Metabolomic analysis on exudates from wild-type T. atroviride and the Δtmk3 mutant showed that both strains excrete purines and complex metabolites; functional Tmk3 is required to produce some of these metabolites. This study highlights the complex interplay between Trichoderma-metabolites and soil bacteria, revealing both beneficial and antagonistic effects, and underscoring the intricate and multifaceted nature of this relationship.

Background: The stink bugs, Nezara viridula L. and Piezodorus guildinii Westwood (Hemiptera: Pentatomidae) are the most important and widespread species of polyphagous stink bugs in the tropical and subtropical regions of Latin America, which affect the quality and yield of the common bean (Phaseolus vulgaris L.). The use of synthetic chemical insecticides is the major control strategy to manage stink bugs in common beans and alternatives are needed. In this study, mortality and median Lethal Time (LT50) of two Cuban isolates of the entomopathogenic fungus Beauveria bassiana (18 S-O-R and 96 P-O-E), as well as one commercial Cuban isolate (Bb-18), at a concentration of 1 × 108 conidia/ml were evaluated. These evaluations were conducted against both stink bug species using Petri dish bioassays and a semi-field experiment in common beans. Results: In Petri dish bioassays, the isolates 18 S-O-R and 96 P-O-E caused 100% mortality of both N. viridula and P. guildinii. This was significantly higher than for isolate Bb-18, which caused 86.3% N. viridula and 81.3% P. guildinii mortality. In the semi-field experiment, when pooling both stink bug species, total mortality after 14 days was 91.3% for 18 S-O-R, 80.0% for 96 P-O-E and 73.8% for Bb-18 isolates. LT50 value for isolate 18 S-O-R tested under laboratory conditions was 6.04 ± 0.18 days for N. viridula and 5.32 ± 0.14 days for P. guildinii at the same concentration of 1 × 108 conidia/ml. LT50 value for isolate 18 S-O-R in semi field was 6.79 ± 0.37 days for N. viridula and 7.71 ± 0.32 days for P. guildinii at 1 × 108 conidia/ml. Conclusion: The study highlights the potential of B. bassiana 18 S-O-R as a promising candidate for control of stink bugs in common bean under tropical conditions as an alternative to conventional chemical insecticides in integrated pest management (IPM) programs. Moving forward, further research should focus on validating the efficacy under diverse field conditions and integrating application methods into practical IPM approaches. Trichoderma species (Hypocreaceae) is one of the world's most common widely used plant growth-promoting fungus and, ecologically and economically important genus. A survey was conducted for species of the genus Trichoderma from 165 soil samples in Aydin province, Türkiye in 2018 and 2019. We obtained 88 isolates of the genus Trichoderma with macroscopic and molecular features were identified from soil samples. Single conidia colonies of Trichoderma isolates were cultured different media for morphological characteristics. Tr49 isolate was recognized Trichoderma guizhouense and this isolate isolation of soil samples from red beet (Beta vulgaris subsp. vulgaris var. conditiva) field. The isolate was identified grounded on molecular and phylogenetic analysis with three gene (ITS; internal transcribed spacer, tef1-α; translation elongation factor 1 alpha, rpb2; RNA polymerase II subunit) primers. We provide the first North American report of the lichenicolous fungus Pronectria fragmospora. Based on multigene phylogenetic analyses we found this species to be nested within the genus Trichonectria and the new combination Trichonectria fragmospora is proposed. We provide a description and photos of the North American material on Evernia prunastri and a phylogenetic tree showing its placement within the genus. The knowledge of entomopathogenic fungus in social wasps is limited, however, association appears to be more common than past thinking. In this study, we recorded the occurrence of eight social wasp species infected: Agelaia fulvofasciata, Agelaia testacea, Apoica cf. pallida, Polistes rufiventris, Polybia dimidiata, Polybia liliacea, Polybia striata, Synoeca virginea, Polistes (Aphanilopterus), and one Mischocyttarus (Mischocyttarus) morphotypes by Ophiocordyceps sphecocephala for the first, and added some new geographic range extension records for the already reported association of Ophiocordyceps humbertii for some locations in the Neotropics with a brief behavioral diagnosis of the species. Trichoderma spp. are globally distributed and are considered significant fungal resources. This study presents the discovery of 11 new species of Trichoderma: T. caeruleum, T. gongcheniae, T. graminicola, T. graminis, T. hongkuii, T. parapeberdyi, T. neoguizhouense, T. neohongkuii, T. parahamatum, T. parahongkuii, and T. shaanxiensis. All of these new species were isolated from soils, except for T. caeruleum, T. graminicola, T. graminis, and T. neohongkuii, which were found as endophytes in Poaceae plants. The phylogenetic position of these novel species was determined by analysing the concatenated sequences of the second largest nuclear RNA polymerase subunit encoding gene (rpb2) and the translation elongation factor 1-alpha encoding gene (tef1). The results of the phylogenetic analysis revealed that each new species formed a distinct lineage: T. gongcheniae, T. graminicola, T. graminis, T. neoguizhouense, T. parapeberdyi, and T. shaanxiensis belong to the Harzianum Clade, T. hongkuii, T. parahongkuii, and T. neohongkuii are new members of the Koningii Clade, T. parahamatum belongs to the Hamatum Clade, and T. caeruleum does not fall within any of the named clades. The study also provided a detailed description of the morphology and cultural characteristics of the newly discovered Trichoderma species. Two new termite-pathogenic species, Ophiocordyceps globiperitheciata and O. longistipes, are described from Yunnan Province, China. Six-locus (ITS , nrSSU , nrLSU , tef-1α, rpb1 and rpb2) phylogenetic analyses in combination with morphological observations were employed to characterize these two species. Phylogenetically, O. globiperitheciata is most closely related to Hirsutella cryptosclerotium and O. communis, whereas O. longistipes shares a sister relationship with O. fusiformis. However, O. globiperitheciata differs from H. cryptosclerotium by parasitizing Blattodea and producing clavate, unbifurcated stromata. Ophiocordyceps globiperitheciata is distinguished from O. communis by multiple stromata, shorter asci and ascospores. Ophiocordyceps longistipes differs from O. fusiformis in producing larger stromata, perithecia, asci and ascospores, as well as smaller citriform or oval conidia. Microbial communities are an important component of mangrove ecosystems. In order to reveal the diversity of endophytic fungi in the mangrove ferns Acrostichum speciosum and A. aureum in China, the internal transcribed spacer (ITS) regions of endophytic fungi in four plant tissues (leaves, petioles, roots, and rhizomes) from three locations (Zhanjiang, Haikou, and Wenchang) were sequenced. The richness, species composition, and community similarity were analyzed. The main results are as follows: the dominant fungi in A. speciosum and A. aureum belonged to the phyla Ascomycota and Basidiomycota, accounting for more than 75% of the total identified fungi; in terms of species composition at the operational taxonomic unit (OTU) level, the endophytic fungi in A. aureum were more diverse than those in A. speciosum, and the endophytic fungi in rhizomes were more diverse than in other tissues. In Zhanjiang, both A. speciosum and A. aureum showed the richest diversity of endophytic fungi, both at the OTU classification level and in terms of species composition. Conversely, the richness of endophytic fungi in the samples of A. speciosum from Wenchang and Haikou is extremely low. The regional differences in dominant fungi increase with the degrading of taxonomic levels, and there were also significant differences in the number of unique fungi among different origins, with Zhanjiang samples having a larger number of unique fungi than the other locations. There were significant differences in the dominant fungi among different tissues, with Xylariales being the dominant fungi in rhizomes of A. speciosum and Hypocreales being the dominant fungi in the petioles, roots, and rhizomes of A. aureum. Overall, the community similarity of endophytic fungi among locations is moderately dissimilar (26–50%), while the similarity between tissues is moderately similar (51–75%). The low diversity of endophytic fungi could be one of the main reasons for the endangerment of A. speciosum. Mushrooms are important organisms because of their human nutritional and medicinal value. With the expansion of the cultivation of edible mushrooms, fungal diseases have become a major problem in limiting their production. Numerous fungi can cause mushroom deformation or rots. In this publication we report on fungal diseases found during Morchella cultivation in China, with emphasis on morphology and phylogeny to characterise species. The key findings include 1) establishment of a new family Albomorchellophilaceae in Hypocreales, and a novel monotypic genus Albomorchellophila with the type species A. morchellae. Divergence time estimates indicate that Albomorchellophilaceae diverged from its sister family Calcarisporiaceae at ca. 105 (92–120) MYA; 2) the phylogeny and morphology of the family Pseudodiploosporeaceae (Hypocreales) is revised. The family contains a single genus Pseudodiploospora. Intraspecific genetic analyses of Pseudodiploospora longispora reveals significant base differences within strains, especially in the regions of protein-coding genes RPB 2 and TEF; 3) four fungicolous taxa, i.e., Cylindrodendrum alicantinum, Hypomyces aurantius, Hypomyces rosellus, and Trichothecium roseum, are reported as putative pathogens on cultivated morels for the first time. In addition, the previously reported pathogens of morels, Clonostachys rosea, Clonostachys solani, Hypomyces odoratus, and Pseudodiploospora longispora are also detailed in their symptoms and morphology; 4) the phylogeny and morphology of "Zelopaecilomyces" previously placed within Pseudodiploosporeaceae are re-assessed. "Zelopaecilomyces" is proved to be introduced through a chimerism of gene fragments sourced from two distinct organisms. Consequently, it is recommended that "Zelopaecilomyces" should not be recognised due to the mixed up molecular data in phylogeny and a lack of support from morphological evidence. Furthermore, this study discusses the voucher specimen Paecilomyces penicillatus (CBS 448.69), which may contain two mixed taxa, i.e., Pseudodiploospora longispora and a member of Penicillium. Publications on pathogenic fungi of cultivated mushrooms is sporadically, which leads to a lack of understanding of causal agents. As a follow up to the diseases of morel cultivation, we also review the fungal diseases of cultivated mushrooms reported over the last four decades. More than 130 pathogens affect the growth and development of the main cultivated mushrooms. The taxonomic diversity of these pathogens is high, distributed in 58 genera, 40 families, 20 orders, 12 classes and six phyla. The host infected are from Ascomycota to Basidiomycota, mainly being reported from Agaricus bisporus, Cordyceps militaris, Morchella spp., and Pleurotus spp. This study not only enriches our current knowledge on the diversity of pathogens of cultivated mushrooms, especially morels, but also recognizes the importance of some taxa as potential pathogens. Taxonomic investigation and accurate identification are initial and key steps to understanding pathogen-mushroom interactions, and will result in better disease management strategies in the mushroom industry. Ophiocordyceps sinensis is a famous traditional Chinese medicine adapted to the alpine environment of the Qinghai-Tibet Plateau and adjacent regions. Clarification of the species diversity of Ophiocordyceps sinensis and its relatives could expand the traditional medicinal resources and provide insights into the speciation and adaptation. The study is prompted by the discovery of a new species, O. megala, described here from a biodiversity hotspot in the Hengduan Mountains, China. Combined morphological, ecological, and phylogenetic evidence supports its distinctiveness from O. sinensis, O. xuefengensis, and O. macroacicularis. Additionally, based on the phylogenetic construction of Ophiocordyceps, a special clade was focused phylogenetically on the more closely related O. sinensis complex, which was defined as the O. sinensis- species complex lineage. A total of 10 species were currently confirmed in this lineage. We made a comprehensive comparison of the sexual/asexual morphological structures among this species complex, distinguishing their common and distinctive features. Furthermore, using the method of species distribution modelling, we studied the species ocurrences in relation to climatic, edaphic, and altitudinal variables for the eight species in the O. sinensis-species complex, and determined that their potential distribution could extend from the southeastern Qinghai-Tibet Plateau to the Xuefeng Mountains without isolating barrier. Thus, the biodiversity corridor hypothesis was proposed around the O. sinensis-species complex. Our study highlights the phylogeny, species diversity, and suitable distribution of the O. sinensis-species complex lineage, which should have a positive implication for the resource discovery and adaptive evolution of this unique and valuable group. Ophiocordyceps is the largest genus in Ophiocordycipitaceae and has a broad distribution with high diversity in subtropical and tropical regions. In this study, two new species, pathogenic on lepidopteran larvae are introduced, based on morphological observation and molecular phylogeny. Ophiocordyceps fenggangensis sp. nov. is characterised by having fibrous, stalked stroma with a sterile tip, immersed perithecia, cylindrical asci and filiform ascospores disarticulating into secondary spores. Ophiocordyceps liangii sp. nov. has the characteristics of fibrous, brown, stipitate, filiform stroma, superficial perithecia, cylindrical asci and cylindrical-filiform, non-disarticulating ascospores. A new combination Ophiocordyceps musicaudata (syn. Cordyceps musicaudata) is established employing molecular analysis and morphological characteristics. Ophiocordyceps musicaudata is characterised by wiry, stipitate, solitary, paired to multiple stromata, yellowish, branched fertile part, brown stipe, immersed perithecia, cylindrical asci and cylindrical-filiform, non-disarticulating ascospores. Summary: Five lists of names for protection assembled by working groups set up under Art. F.2.1 are approved. The lists contain one family name along with 39 names of genera and 10 names of species, to be treated as protected against listed and unlisted synonymous or homonymous names. Lists were compiled by working groups (WGs) approved by the International Commission on the Taxonomy of Fungi and the Nomenclature Committee for Fungi: on Homobasidiomycetes, Heterobasidiomycetes, Orbiliomycetes, Sordariomycetes (excluding Diaporthales, Hypocreales and Magnaporthales) and Miscellaneous fungi. The list from the Pyricularia/Magnaporthe WG did not require any formal action. Acceptance of two proposals to conserve names under Art. 14.1 associated with two of the names (Typhula, Typhulaceae) originally included in the list for protection from the Homobasidiomycetes WG led to the removal of those two names from consideration for protection. The names Drechslerella and Dactylellina, initially included in the list from the Orbiliomycetes WG, were withdrawn from that list. In relation to two proposals under Art 14.1 that are connected to the list from the Dothideomycetes WG, the following family name is not recommended for conservation: Pleomassariaceae against Prosthemiaceae; and the following generic name is not recommended for conservation: Pleomassaria against Prosthemium. As a result of consideration of the set of proposals that contained the two proposals connected to Typhulaceae, the following family name is recommended for conservation: Phyllotopsidaceae against Sclerotiaceae; the following two names of genera are recommended for conservation: Macrotyphula against Sclerotium, and Typhula with a conserved type; and the following species name is recommended for rejection under Art. 56: Sclerotium fulvum. A modified proposal resulted in a recommendation to conserve the name Peziza medicaginis against Sporonema phacidioides. Fusarium disease of sugarbeet is a threat to sugarbeet production and can cause significant losses. During routine evaluation, a unique Fusarium species was solely isolated from unknown dark galls in sugarbeets from Wilkin County, MN, U.S.A. Analysis of the RPB2 gene placed this isolate in the Fusarium solani FSSC 5 clade (100.0% similarity) but uniquely separated from the potato F. solani. Sequencing of this Fusarium species by Minion Nanopore Systems resulted in a draft genome assembly of 19 contigs with a genome size of 59.3 Mb and GC content of 50.66%. A total of 18,900 protein-coding genes, 37 secondary metabolite clusters, 1,559 secreted proteins, 668 carbohydrate-active enzymes, and 404 effectors were predicted in the genome. Pathogenicity tests showed mild vascular discoloration in inoculated sugarbeets unaccompanied by external symptoms, suggesting weak pathogenic ability of this species, indicative of an opportunistic pathogen. Background: The genus Beauveria (Bals.) Vuill. includes many species, some of which are limited to specific regions while others are distributed worldwide. The diversity of Beauveria species is poorly investigated in Syria and most studies lack proper diagnosis of species. Entomopathogenic isolates of this genus were obtained using the Galleria Bait Method. This study aimed to identify these isolates based on morphological characterizations combined with molecular data, using nuclear ribosomal internal transcribed spacer (ITS) and elongation factor 1-alpha (EF1-α) sequences. The diversity of this genus in Syria has also been evaluated using a phylogenetic analysis of available ITS sequences of Syrian isolates in the GenBank. Results: Two entomopathogenic isolates, B195 and B243, were detected in the soil of agro-ecosystems in the Syrian coastal region. Morphological and molecular information revealed that these two isolates belong to Beauveria bassiana (Bals.) Vuill. (Hypocreales: Cordycipitaceae) with 514 bp and 284 bp for the sequences of each isolate for ITS and EF1-α, respectively. Pathogenicity test showed 100% mortality of Galleria mellonella L. larvae 2–3 days post-fungal exposure for both isolates. The phylogenetic tree showed that all Syrian sequences of Beauveria clustered within the species B. bassiana, with a considerable intraspecific diversity, except for two isolates previously identified as B. bassiana, which are closely related to Beuveria pseudobassiana S.A. Rehner and Humber. Conclusions: This study presents a morpho-molecular characterization of two Syrian soil-sourced B. bassiana isolates highly pathogenic to G. mellonella larvae and clarifies their phylogenetic placement. Depending on our findings, further exploration studies of the genus Beauveria in Syria are still needed to better our understanding of the diversity and distribution of this entomopathogen in Syria. The genus Samsoniella was erected based on orange cylindrical to clavate stromata, superficial perithecia and conidiophores with Isaria-like phialides and to segregate them from the Akanthomyces group. In this study, based on morphological features and multigene (SSU, LSU, TEF, RPB1 and RPB2) phylogenetic analysis six Samsoniella species parasitizing spiders were collected in China. Three of them belong to known species S. alpina, S. erucae and S. hepiali. Three new species S. anhuiensis sp. nov., S. aranea sp. nov. and S. fusiformispora sp. nov. are illustrated and described. They are clearly distinct from other species in Samsoniella occurring in independent subclades. Furthermore, among the four insect-pathogenic fungi specimens collected from similar sites, three of them were identified as the new species described below. Our study significantly broadens the host range of Samsoniella from Insecta to Arachnida, marking a noteworthy expansion in understanding the ecological associations of these fungi. Additionally, the identification of both mononematous and synnematous conidiophores in our study not only expands the knowledge of Samsoniella species but also provides a basis for future research by comparing the ecological significance between these conidiophore types. In conclusion, our study enhances the understanding of Samsoniella diversity, presenting a refined phylogenetic framework and shedding light on the ecological roles of these fungi in spider parasitism. Akanthomyces, a group of fungi with rich morphological and ecological diversity in Cordycipitaceae (Ascomycota, Hypocreales), has a wide distribution amongst diverse habitats. By surveying arthropod-pathogenic fungi in China and Southeast Asia over the last six years, nine Akanthomyces spp. were found and identified. Five of these were shown to represent four known species and an undetermined species of Akanthomyces. Four of these were new species and they were named A. kunmingensis and A. subaraneicola from China, A. laosensis from Laos and A. pseudonoctuidarum from Thailand. The new species were described and illustrated according to the morphological characteristics and molecular data. Akanthomyces araneogenus, which was isolated from spiders from different regions in China, Thailand and Vietnam, was described as a newly-recorded species from Thailand and Vietnam. The phylogenetic positions of the nine species were evaluated, based on phylogenetic inferences according to five loci, namely, ITS, nrLSU, TEF, RPB1 and RPB2. In this study, we reviewed the research progress achieved for Akanthomyces regarding its taxonomy, species diversity, geographic distribution and major host/substrate associations. Chinese fir (Cunninghamia lanceolata) is a special fast-growing commercial tree species in China with high economic value. In recent years, leaf blight disease on C. lanceolata has been observed frequently. The diversity of Fusarium species associated with leaf blight on C. lanceolata in China (Fujian, Guangxi, Guizhou, and Hunan provinces) was evaluated using morphological study and molecular multi-locus analyses based on RNA polymerase second largest subunit (RPB2), translation elongation factor 1-alpha (TEF-1a), and RNA polymerase largest subunit (RPB1) genes/region as well as the pairwise homoplasy index tests. A total of five Fusarium species belonging to four Fusarium species complexes were recognized in this study. Two known species including Fusarium concentricum and F. fujikuroi belonged to the F. fujikuroi species complex, and three new Fusarium species were described, i.e., F. fujianense belonged to the F. lateritium species complex, F. guizhouense belonged to the F. sambucinum species complex, and F. hunanense belonged to the F. solani species complex. To prove Koch's postulates, pathogenicity tests on C. lanceolata revealed a wide variation in pathogenicity and aggressiveness among the species, of which F. hunanense HN33-8-2 caused the most severe symptoms and F. fujianense LC14 led to the least severe symptoms. To our knowledge, this study also represented the first report of F. concentricum, F. fujianense, F. fujikuroi, F. guizhouense, and F. hunanense causing leaf blight on C. lanceolata in China. Entomopathogens tend to have a slow speed of kill when used for targeting agricultural insect pests. Relating temperature as a driver of this speed is important to predict pest mortality, and extending this to a degree‐day infection model has rarely been studied. Many species of wireworms (Coleoptera: Elateridae), the larvae of click beetles, are subterranean and generalist agricultural pests that can be difficult to control with pesticides. Targeting adult beetles, however, may be an effective method to reduce larval recruitment. Metarhizium brunneum Petch (Hypocreales), an entomopathogenic fungus, kills click beetles but the mortality rate and speed of kill are expected to vary according to temperature. Using a thermal gradient plate to simulate daily oscillating temperatures in Agassiz, British Columbia, Canada, for April, May, and June, the effectiveness of M. brunneum strains LRC112 and F52 in causing mortality to Agriotes obscurus (L.) and Agriotes lineatus (L.) click beetles was studied in the laboratory. Mortality was fastest in beetles exposed to June temperatures and slowest in those exposed to April temperatures, with differences among beetle species × M. brunneum strain combinations. Higher temperatures resulted in more rapid mycelial outgrowth and conidiation in beetle cadavers, with only A. obscurus infected with M. brunneum LRC112 attaining near 100% conidiation. The number of degree days required to kill 50% of the beetles (LDD50) was least for A. obscurus infected with M. brunneum LRC112 (176) followed by A. obscurus × M. brunneum F52 (212), A. lineatus × M. brunneum LRC112 (215), and A. lineatus × M. brunneum F52 (292). Hypothetical calculations showed that M. brunneum exposure earlier in the season resulted in a longer time to kill 50% of the beetles (LT50) but the earliest LT50 calendar date. Later M. brunneum exposure dates resulted in lower LT50's, but later LT50 dates. This conceptual work demonstrates that daily temperature oscillations, seasonality, and degree days must be considered to predict the efficacy and speed of kill of different fungal entomopathogen strains when targeting different click beetle species. This is the first record of the fungus Flavocillium subprimulinum in Mexico. The isolate was taxonomically characterised and cultured in potato dextrose broth (PDB), Czapek–Dox broth (CzDoxB), and sweet potato dextrose broth (SPDB) to obtain its filtrates (FLCF). The nematocidal activity (NA) of three FLCF concentrations was assessed against Haemonchus contortus L3. Protease activity (PA) was assessed with SDS-PAGE, followed by a zymogram. The NA of the FLCF reached 94.43% in PDB and 95.82% in CzDoxB, respectively, at 100 mg/mL. Lower mortality (64%) was found in SPDB at 100 mg/mL. SDS-PAGE showed bands (in PBS) of ~25, ~40, and ~55 kDa. The zymogram showed protein bands (PBs) with PA in the media, including PBs of ~14, ~40, and ~55 kDa. This study establishes the basis for exploring the potential use of this fungus against H. contortus, which is considered the most pathogenic parasite affecting lambs. [ABSTRACT FROM AUTHOR]

Beauveria bassiana (B. bassiana) is a significant entomopathogenic fungus (EPF) in agriculture as a sprayable biocontrol agent. It has the potential to be established as an endophyte (ENP) in various crops, resulting in beneficial effects for the host plants, including resistance to pest insects and increased growth and yield. However, it is not known whether a B. bassiana strain has such a favorable impact on the plant, since it is a common soil microorganism. Therefore, techniques that allow strain monitoring will be advantageous. To date, methods for detecting or monitoring a specific EPF strain after external application are scarce. In the present study, an in planta nested PCR technique was standardized to differentiate between three B. bassiana strains (GHA, PTG4, and BB37) established as endophytes in bean plants under laboratory conditions by detecting the insertion profile of four group I introns located in the 28S gene of B. bassiana ribosomal DNA. This technique recognized a distinct pattern of bands of different sizes for each strain, with a sensitivity of 1 pg per 10 ng of plant DNA. With the aim of evaluate the thermal tolerance of twelve suspoemulsion formulations containing conidia of I. javanica (at low and high moisture content) as an active ingredient and the biological effectiveness in the greenhouse, we developed twelve oil-based formulations of I. javanica with canola, castor bean, corn, soybean, peanut and mineral oil and surfactant mixtures: six formulations with conidia at low humidity (5-7%) and six with conidia at higher humidity (40-50%). Formulations were developed using the HLB system; Span 80 (HLB 4.3) and Tween 80 (HLB 15) like surfactants were mixed with oils. Thermotolerance was evaluated after exposure to 45 °C for 2, 8 and 24 h. Additionally, to evaluate the functionality of the formulations, greenhouse bioassays were performed using tomato plants infested with Bemisia tabaci. To determine the viability and shelf life, 10 mL of each formulation was stored for 5 and 9 months, and the storage temperature oscillated between 12 and 34 °C (at typical room temperature), with 12 / 12 h light / dark cycles, after 9 months of storage. The viability of unformulated conidia and those contained in the formulations was >82% after exposure to 45 °C for 2 h. The B (mineral oil), C (corn, soybean and peanut oils), D (corn oil), E (canola and soybean oils), and F (castor oil) formulations (low humidity), unformulated conidia and PAE-SIN caused mortality ranging from 43-56%. The conidia in formulations on oil exhibited 53-65% viability at 24 h and were statistically similar to each other but significantly different from the conidia (unformulated) with the viability 5%; at 72 h, the viability of conidia in all formulations was 78-88%, and the values were statistically similar to each other but significantly different from that of the conidia (unformulated) with the viability 6%. The formulations evaluated in this work presented lethality in B. tabaci nymphs 1-4, it is also suggested that these formulations confer thermal protection acquired by the formulation since unformulated conidia are drastically affected. Four new species of the genus Niveomyces are described from Thailand. They were found as mycoparasites on: Ophiocordyceps infecting flies (Diptera) for Niveomyces albus; ants (Hymenoptera) for N. formicidarum; and leafhoppers (Hemiptera) for N. hirsutellae and N. multisynnematus. A new genus, Pseudoniveomyces with two species: Pseudoniveo. blattae (type species), parasitic on Ophiocordyceps infecting cockroaches, and Pseudoniveo. arachnovorum, found on a spider egg sac, are also described. These fungi share a common feature which is a sporothrix-like asexual morph. Based on our molecular data, Sporothrix insectorum is shown to be affiliated to the genus Niveomyces, and thus a new combination N. insectorum comb. nov. is proposed. Niveomyces coronatus, N. formicidarum and N. insectorum formed the N. coronatus species complex found on ant-pathogenic Ophiocordyceps from different continents. Pseudoniveomyces species are distinguished from Niveomyces spp. based on the presence of fusoid macroconidia in culture and a red pigment diffused in the medium, resembling to Gibellula and Hevansia. The molecular phylogenetic analyses also confirmed its generic status. The host/substrates associated with the genera within Cordycipitaceae were mapped onto the phylogeny to demonstrate that mycoparasitism also evolved independently multiple times in this family. Entomogenous fungi are one of the most diverse fungal groups among ascomycetes. During investigations of macro-fungi biodiversity in Kaihua and Putuo in Zhejiang Province, China, in 2021, several entomopathogenic fungal specimens were collected from subtropical forests. After a careful examination of morphology and multi-gene phylogenetic analyses applying nrSSU, nrLSU, tef1, rpb1 and rpb2, two novel species were discovered, described herein as Blackwellomyces kaihuaensis and Metarhizium putuoense, and further compared with other similar species. Claviceps (Clavicipitaceae, Hypocreales) was erected in 1853, although ergotism had been well-known for a much longer time. By 2000, about 70 taxa had been described in Claviceps, of which eight species and six varieties were based on Japanese type or authentic specimens. Most of these Japanese Claviceps taxa are based on lost specimens or have invalid names, which means many species practically exist only in the scientific literature. The ambiguous identities of these species have hindered taxonomic resolution of the genus Claviceps. Consequently, we sought and collected more than 300 fresh specimens in search of the lost Japanese ergots. Multilocus phylogenetic analyses based on DNA sequences from LSU, TEF-1a, TUB2, Mcm7, and RPB2 revealed the phylogenetic relationships between the Japanese specimens and known Claviceps spp., as well as the presence of biogeographic patterns. Based on the phylogenetic analysis, host range and morphology, we re-evaluated Japanese Claviceps and recognised at least 21 species in Japan. Here we characterised 14 previously described taxa and designated neo-, lecto- and epi-types for C. bothriochloae, C. imperatae, C. litoralis, C. microspora, C. panicoidearum and C. yanagawaensis. Two varieties were elevated to species rank with designated neotypes, i.e. C. agropyri and C. kawatanii. Molecular phylogenetic and chemical analyses, and morphological characterization of collections of North American Paraisaria specimens support the description of two new species and two new combinations for known species. P. cascadensis sp. nov. is a pathogen of Cyphoderris (Orthoptera) from the Pacific Northwest USA and P. pseudoheteropoda sp. nov. is a pathogen of cicadae (Hemiptera) from the Southeast USA. New combinations are made for Ophiocordyceps insignis and O. monticola based on morphological, ecological, and chemical study. A new cyclopeptide family proved indispensable in providing chemotaxonomic markers for resolving species in degraded herbarium specimens for which DNA sequencing is intractable. This approach enabled the critical linkage of a 142-year-old type specimen to a phylogenetic clade. The eff ectiveness of two biological control agents, Chrysoperla carnea (Neuroptera: Chrysopidae) and the entomopathogenic fungus Beauveria bassiana (Ascomycota: Hypocreales) against nymphs and adults of Neophilaenus campestris (Hemi ptera: Aphrophoridae) was determined under laboratory conditions. First, diff erent nymphal stages of N. campestris were presented to diff erent larval stages of C. carnea. Second, the eff ect of the foam produced by N. campestris nymphs on the larvae of C. carnea predation was evaluated. Finally, four concentrations of a wild strain of B. bassiana, BbGEp1, were sprayed on plants in order to determine their lethality for adults of N. campestris. Second and 3rd-instar larvae of green lacewing larvae are capable of capturing and killing 3rd and 5th-instar nymphs of N. campestris. The percentage of 3rd-instar lacewing larvae that killed nymphs was signifi cantly higher than that were killed by second-instar larvae. Second-instar larvae killed signifi cantly more 3rd-instar nymphs than 5th-instar nymphs. Third instar lacewing larvae killed an average (± SEM) of 1.50 ± 0.31 5th-instar nymphs and 2nd-instar larvae killed very few nymphs. Spittlebug foam reduced, but did not prevent predation. The lethality of the entomopathogenic B. bassiana BbGEp1 used against adults of N. campestris was characterized by an LC50 value of 1.61 x 106 conidia/mL and LT50 of 3.63 days at 1 x 107 conidia/mL. The present study provides new and valuable data on the activity of two promising biological control agents of vectors of the bacterium Xylella fastidiosa. Further research is needed to confi rm the results presented here and on the cost eff ectiveness of using these control agents as alternatives to synthetic insecticides for preventing the further spread of X. fastidiosa in Europe. In the course of our ongoing research targeting the identification of potential biocontrol agents from entomopathogenic fungi (EPF), we explored a solid-state rice fungal extract of Blackwellomyces roseostromatus BCC56290 derived from infected lepidopteran larvae. Chemical and biological prospections afforded four unprecedentedly reported natural products differentiated into a dimeric naphthopyran bioxanthracene ES-242 derivative (1) and three cyclodepsipeptides (2–4) in addition to two known cyclodepsipeptides, cardinalisamides B (5) and C (6). Chemical structures of the isolated compounds were elucidated through comprehensive 1D/2D NMR and HR-ESI-MS data together with comparisons to the reported literature. The absolute configuration of the isolated cyclodepsipeptides was determined using Marfey’s method. All isolated compounds were assessed for their antimicrobial, cytotoxic, and nematicidal activities with some compounds revealing significant activities.

AbstractThe aim of this study was to characterize a new fungal species, Acremonium conglutinatum, isolated from air samples collected in Wando, South Korea. Phylogenetic analysis based on the internal transcribed spacer and large subunit regions revealed its unique position within the genus Acremonium. The isolated strain displayed distinct morphological characteristics, including ellipsoid or bent-ellipsoid conidia formed in clusters on the phialides. These features differentiate the new species from closely related species within the genus. This study describes the morphological and molecular characteristics of A. conglutinatum and emphasizes its phylogenetic relationships with other Acremonium spp. The identification of this novel species contributes to our understanding of the diversity and ecological role of Acremonium.
