Pleosporales is a highly diverse (and the largest) order in Dothideomycetes, and it is widespread in decaying plants in various environments around the world. During a survey of fungal diversity in Sichuan Province, China, specimens of hyphomycetous and Thyridaria-like fungi were collected from dead branches of pine trees and cherry trees. These taxa were initially identified as members of Massarinaceae and Thyridariaceae through morphological examination. Phylogenetic analyses of the Thyridariaceae, combining ITS, SSU, LSU, RPB2, and TEF1 sequence data, indicated a distinct clade sister to Pseudothyridariella and Thyridariella, distinct from any genus in the family. Thus, a new genus, Vaginospora, is proposed to accommodate the type species Vaginospora sichuanensis, which is characterized by semi-immersed globose to oblong ascomata with an ostiolar neck, cylindrical to clavate asci with an ocular chamber, and hyaline to dark brown, fusiform, 3–5-transversely septate ascospores with an inconspicuous mucilaginous sheath. Based on the morphological comparisons and multi-locus phylogenetic analyses (ITS, SSU, LSU, RPB2, and TEF1) of the Massarinaceae, we have identified three collections belonging to the genus Helminthosporium, leading us to propose H. filamentosa sp. nov., H. pini sp. nov., and H. velutinum as a new host record. According to Phylogenetic analysis, H. pini formed an independent clade sister to H. austriacum and H. yunnanense, and H. filamentosa represents the closest sister clade to H. quercinum. Helminthosporium pini is distinct from H. austriacum by the shorter conidiophores and H. yunnanense by the longer and wider conidia. The H. filamentosa differs from H. quercinum in having longer conidiophores and smaller conidia. This study extends our understanding of diversity within Thyridariaceae and Helminthosporium. Phytopathogenic fungi in the genus Bipolaris are typically associated with poaceous hosts including major cereal crops. Morphological identification alone is often insufficient for species discrimination due to overlapping microscopic characters. Thus, molecular phylogenetic analyses have been utilised for accurate species identification and to establish evolutionary relationships within the genus. Although there are 138 species epithets of Bipolaris listed in the Index Fungorum, only 46 species have been identified and accurately placed within the genus based on ex-type derived DNA sequence data, with only four Bipolaris species reported from Sri Lanka. During this study, diseased and dead plant specimens of weedy grasses and cultivated rice were collected from several collection sites in Sri Lanka and associated fungal species were isolated. Morphological characters together with phylogenetic inference from the analyses of three loci, internal transcribed spacers 1 and 2 with 5.8S region (ITS), glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and translation elongation factor 1-α (TEF1), supported the introduction of two novel species namely Bipolaris adikaramae from Panicum maximum and B. petchii from Ischaemum sp. Morphological descriptions and illustrations are provided for the newly described taxa. In addition, B. shoemakeri from Echinochloa sp. and Oryza sativa and B. sivanesaniana from O. sativa were identified as novel records from Sri Lanka. This study suggests the need for extensive surveys of crops and associated weedy hosts in order to accurately assess the diversity of emerging species of phytopathogenic and opportunistic graminicolous fungi. Bipolaris sorokiniana (teleomorph, Cochliobolus sativus) is one of the wheat pathogens that can attack all wheat parts, including seeds, roots, shoots, and leaves. Black point, root rot, crown rot and spot blotch are the main diseases caused by B. sorokiniana in wheat. Seed infection by B. sorokiniana can result in black point disease, reducing seed quality and seed germination and is considered a main source of inoculum for diseases such as common root rot and spot blotch. Root rot and crown rot diseases, which result from soil-borne or seed-borne inoculum, can result in yield losses in wheat. Spot blotch disease affects wheat in different parts of the world and cause significant losses in grain yield. This review paper summarizes the latest findings on B. sorokiniana, with a specific emphasis on management using genetic, chemical, cultural, and biological control measures.

Double-stranded (ds)RNA fungal viruses are currently assigned to six different families. Those from the family Totiviridae are characterized by nonsegmented genomes and single-layer capsids, 300-450 Å in diameter. Helminthosporium victoriae virus 190S (HvV190S), prototype of recently recognized genus Victorivirus, infects the filamentous fungus Helminthosporium victoriae (telomorph: Cochliobolus victoriae), which is the causal agent of Victoria blight of oats. The HvV190S genome is 5179 bp long and encompasses two large, slightly overlapping open reading frames that encode the coat protein (CP, 772 aa) and the RNA-dependent RNA polymerase (RdRp, 835 aa). To our present knowledge, victoriviruses uniquely express their RdRps via a coupled termination-reinitiation mechanism that differs from the well-characterized Saccharomyces cerevisiae virus L-A (ScV-L-A, prototype of genus Totivirus), in which the RdRp is expressed as a CP/RdRp fusion protein due to ribosomal frameshifting. Here, we used transmission electron cryomicroscopy and three-dimensional image reconstruction to determine the structures of HvV190S virions and two types of virus-like particles (capsids lacking dsRNA and capsids lacking both dsRNA and RdRp) at estimated resolutions of 7.1, 7.5, and 7.6 Å, respectively. The HvV190S capsid is thin and smooth, and contains 120 copies of CP arranged in a "T = 2" icosahedral lattice characteristic of ScV-L-A and other dsRNA viruses. For aid in our interpretations, we developed and used an iterative segmentation procedure to define the boundaries of the two, chemically identical CP subunits in each asymmetric unit. Both subunits have a similar fold, but one that differs from ScV-L-A in many details except for a core α-helical region that is further predicted to be conserved among many other totiviruses. In particular, we predict the structures of other victoriviruses to be highly similar to HvV190S and the structures of most if not all totiviruses including, Leishmania RNA virus 1, to be similar as well.

The enigma of the transmissible disease of Helminthosporium victoriae has almost been resolved. Diseased isolates are doubly infected with two distinct viruses, the victorivirus Helminthosporium victoriae virus 190S and the chrysovirus HvV145S. Mixed infection, however, is not required for disease development. DNA transformation experiments and transfection assays using purified HvV190S virions strongly indicate that HvV190S alone is necessary for inducing disease symptoms. HvV145, like other chrysoviruses, appears to have no effect on colony morphology. This chapter will discuss the molecular biology of the two viruses and summarize recent results of characterization of host gene products upregulated by virus infection. Furthermore, the novel structural features of HvV190S capsid will be highlighted.

Environmentally damaging invasive plants can also serve as reservoir hosts for agricultural pathogens. Microstegium vimineum is an invasive C4 annual grass that is present throughout the midwestern and eastern United States. It can reach high densities in disturbed areas such as crop-forest interfaces, which creates the potential for pathogen spillover from M. vimineum to agricultural crops and native plants. A previous study that surveyed disease on M. vimineum found a large-spored Bipolaris species that was widespread on M. vimineum and also isolated from co-occurring native grasses. Here, we report that the large-spored fungus isolated from M. vimineum and the native grass Elymus virginicus is Drechslera gigantea, based on comparison with published descriptions of morphological traits, and establish that D. gigantea is a pathogen of M. vimineum and E. virginicus. We review the phylogenetic placement and taxonomic history of D. gigantea and propose that it be reassigned to the genus Bipolaris as Bipolaris gigantea. [ABSTRACT FROM AUTHOR]

The plants of rice varieties (Arco, Thaibonnet and Elio) were inoculated with three isolates of Helminthosporium rostrata (HR1 HR2 and HR3), isolated for the first time in Morocco from the rice seed of Taibonnet variety at the end of the growing season. The results obtained showed that all the isolates are able to induce the disease on rice plants and sporulate on the foliar lesions. HR1, HR2 and HR3 was respectively the most pathogenic on Elio (I.C = 113), Arco (I.C = 212.5), and Taibonnet (130.48). Results of 2007-2015 immunological studies on search of sources of barley's non-specific resistance to helminthosporium diseases (root rot, leaf spots: bacterial stripe, halo blight, etc) under conditions of Kirov region are presented in the article. Immunologic status of 380 spring barley samples from VIR's world collection and of 140 varieties and perspective lines bred in North-East Agricultural Research Institute are analyzed. Studies were conducted under conditions of natural and artificial epiphytoses of local infectious populations. As a whole, low frequency of obtaining of immunologically value samples is reported: in VIR's collection, it is equal to 9.5% average for all diseases and in collection of North-East Agricultural Research Institute - 11.4%. Much less samples had combination of resistance with productivity at standard level or higher - about 5.6%. At 3-7 year study of barley genofund under hard infectious backgrounds sources were revealed of resistance to one, two, or more diseases. Following varieties had complex resistance to three types of leaf spot: Medicum 336 (Russia), Mironovsky 86 and Surpriz (Ukraine), Margret (Germany), Korona Lashego (Poland), ^dac (Canada), and perspective lines of authors' breeding: 917-01, 138-09, and 247-09. Line 917-01 (Forward) created by a method of agricultural biotechnology passes State Tests. Variety Medicum 336 bred in Samara Agricultural Research Institute has resistance to all types of helminthosporioses. All samples were tested on yield structure elements and potential productivity. It is revealed that variety Medicum 336 has high level of 1000 grain mass and high amount of fruit-bearing shoots; varieties Mironovsky 86 and Natali have high level of 1000 grain mass; variety Surpriz - high level of 1000 grain mass and productivity; varieties Edem and ^dac - early ripening and productivity and so on. Perspective lines of spring bar- ley are created using of some sources which pass competitive tests now. Among them there are such lines as 43-05, 457-08, and 341 -08 that exceed standard varieties on productivity by 0.16- 0.63 t/ha or by 4-17%.

Exserohilum includes a number of plant pathogenic, saprobic and clinically relevant fungi. Some of these species are of great importance in human activities, but the genus has never been revised in a phylogenetic framework. In this study, we revise Exserohilum based on available ex-type cultures from worldwide collections, observation of the holotypes and/or protologues, and additional isolates from diverse substrates and geographical origins. Based on nine nuclear loci, i.e., ITS, LSU, act, tub2, cam, gapdh, his, tef1 and rpb2, as well as phenotypic data, the genus and species boundaries are assessed for Exserohilum. Three species, i.e., E. novae-zelandiae, E. paspali and E. sorghicola, are excluded from the genus and reallocated in Sporidesmiella and Curvularia, respectively, whereas E. heteropogonicola and E. inaequale are confirmed as members of Curvularia. Exserohilum rostratum is revealed as conspecific with species previously described in Exserohilum such as E. antillanum, E. gedarefense, E. leptochloae, E. longirostratum, E. macginnisii and E. prolatum. Additionally, E. curvatum is revealed as synonym of E. holmii, and E. fusiforme of E. oryzicola. A total of 11 Exserohilum phylogenetic species are described, illustrated and discussed, including one novel taxon, E. corniculatum. Summary: Wheat is one of the primary staple foods throughout the planet. Significant yield gains in wheat production over the past 40 years have resulted in a steady balance of supply versus demand. However, predicted global population growth rates and dietary changes mean that substantial yield gains over the next several decades will be needed to meet this escalating demand. A key component to meeting this challenge is better management of fungal incited diseases, which can be responsible for 15%–20% yield losses per annum. Prominent diseases of wheat that currently contribute to these losses include the rusts, blotches and head blight/scab. Other recently emerged or relatively unnoticed diseases, such as wheat blast and spot blotch, respectively, also threaten grain production. This review seeks to provide an overview of the impact, distribution and management strategies of these diseases. This study isolated and examined many isolate of Drechslera spp., Bipolaris spp. and Exserohilum from rice cultivar and treated the most changes that occurred in the taxonomy and nomenclature of the genus Helminthosporium according to the ICBN rules. This genus was considered as complex of species and deeply entrenched in the taxonomic literature. 100-668 species were recorded in the phytopathlolgical literature and database, but in this study only 49 species were fixed as correct names in Helminthosporium, and split it into four genera,also the common species of the genus Helmintho sporium were distributed the four genera: Drechslera (28 spp.), Bipolaris (10 spp.), Exserohilum (7spp.) and Marielliottia (3 spp.). Most of the species of Helminthosporium become synonym for species of the genera Drechslera, Bipolaris, Exserohilum and Marielliottia and the names of Helminthosporium stilled as basyname. Summary: Bipolaris sorokiniana is the causal agent of multiple diseases on wheat and barley and is the primary constraint to cereal production throughout South Asia. Despite its significance, the molecular basis of disease is poorly understood. To address this, the genomes of three Australian isolates of B. sorokiniana were sequenced and screened for known pathogenicity genes. Sequence analysis revealed that the isolate BRIP10943 harboured the ToxA gene, which has been associated previously with disease in the wheat pathogens Parastagonospora nodorum and Pyrenophora tritici‐repentis. Analysis of the regions flanking ToxA within B. sorokiniana revealed that it was embedded within a 12‐kb genomic element nearly identical to the corresponding regions in P. nodorum and P. tritici‐repentis. A screen of 35 Australian B. sorokiniana isolates confirmed that ToxA was present in 12 isolates. Sequencing of the ToxA genes within these isolates revealed two haplotypes, which differed by a single non‐synonymous nucleotide substitution. Pathogenicity assays showed that a B. sorokiniana isolate harbouring ToxA was more virulent on wheat lines that contained the sensitivity gene when compared with a non‐ToxA isolate. This work demonstrates that proteins that confer host‐specific virulence can be horizontally acquired across multiple species. This acquisition can dramatically increase the virulence of pathogenic strains on susceptible cultivars, which, in an agricultural setting, can have devastating economic and social impacts. The present aeromycological investigation was undertaken to study atmospheric fungal spores in Timişoara (western Romania). This study was carried out using a Hirst type volumetric sampler. The study revealed the existence of a rich airborne mycoflora. The atmospheric fungal spores were classified and evaluated into three groups (‘major’, ‘minor’ and ‘sporadic’) depending upon their catch percentage in the air.Cladosporium/Fusarium/Leptosphaeria-group,Alternaria, Helminthosporiumairborne fungal spores and airborne fungal fragments regularly recorded (frequency 100% of days).Cladosporiumaccounted for 81.09% of the outdoor fungal spores. The airborne fungal fragments have been identified as abundant in our geographic area. Spearman’s correlations were applied to meteorological parameters and airborne fungal spore concentrations. In addition, correlations were calculated between the fungal spore concentrations and the meteorological variables from the previous day. A total of eleven weather factors were selected for this investigation. Following Spearman’s correlations, I identified two patterns of behaviour: most of the airborne fungal spores prefer cloudiness, lower near-surface soil temperature, lower atmospheric pressure, higher relative humidity and precipitation (pattern A) while other spore concentrations favour increased sunshine, higher near-surface soil temperature and dry conditions (pattern B). The behaviour of some fungal spores during the warm season has proven unclear (pattern C). This study demonstrates the need for investigations throughout the year and the evaluation with complementary statistical methods, regarding the correct interpretation of airborne mycoflora relationships with meteorological parameters. Helminthosporium solani strains were isolated from potato tubers collected in Russia or taken from imported German and Dutch seed tubers. Sequences of the nuclear ribosomal genes and internal transcribed spacers (ITS) for all 24 tested strains were identical and had 100% similarity to the sequences from GenBank identified as Helminthosporium solani. The obtained molecular data confirmed the morphological identification based on the width and length of conidia, the shape of conidiophores and the colony morphology. Screening for resistance to the fungicides Score 250 SC (active ingredient difenoconazole 250 g/l), Quadris (azoxystrobin 250 g/l), Tecto 500 SC (thiabendazole 500g/l), Zeroxxe (colloidal silver particles (3 g/l) stabilized with amphoteric surfactant) was done. Agar blocks with pure cultures of the fungal strains were placed in the centre of Petri dishes containing malt agar amended with fungicide concentrations of 0.1, 1, 10, 100 and 1000 mg/l (accounted for the concentration of the active ingredient). Malt agar free of fungicide was used as the control. Growth inhibition of 50% (EC50) compared to the control was detected based on the doseresponse curves. Difenoconazole (EC50<0.12mg/l) and colloidal silver (EC50<76 mg/l) were the most effective fungicides. No strains resistant to the aforementioned fungicides were found. In most cases, azoxystrobin was effective against H. solani (EC50<7 mg/l), but there were several strains with high resistance to this fungicide (EC50>100 mg/l). Thiabendazole appears to be effective against the sensitive strains of H. solani (EC50< 7.3 mg/l); however, six studied strains from Russia and the Netherlands were found to be extremely resistant to it (EC50>1000 mg/l). The sequence of their ß-tubulin gene contained a SNP mutation in the 198 codon or 200 codon, translating to Gln (CAG) instead of Glu (GAG) or Tyr (TAC) instead of Phe (TTC), respectively. Bipolaris sorokiniana, a fungal endophyte, was isolated from the leaves ofCostus speciosus, a popular medicinal plant used to control diabetes. Fermentation of the fungus in potato dextrose broth and chromatographic purification of the ethyl acetate (EtOAc) extracts of the culture broth and mycelium yielded two rare sesquiterpenes helminthosporal acid (1) and helminthosporol (2), together with ergosterol. Compounds1and2showed a strong phytotoxic effect on lettuce seed germination and toxicity against brine shrimps. Compound1also showed antifungal activity. In the harvest year of 2004, a random sample of seed lots (n=165) from six rice-growing regions of Rio Grande do Sul (RS) State, Brazil, were examined under seed health blotter test. Fungi growth resembling Bipolaris or Drechslera spp. was isolated and identified at the species level. Observations on the anamorphic structures (conidiophores and conidia) allowed separation of isolates into three groups. Pseudothecia, asci and ascospores from sexual crosses between isolates from a same anamorphic group but originated from different regions showed the typical characteristics of three recognized species (anamorph/teleomorph): Bipolaris cynodontis (Cochliobolus cynodontis); B. curvispora (C. melinidis) and B. oryzae (C. miyabeanus). All species were widely distributed across the rice-growing regions. Overall, the most prevalent species were B. oryzae (44.6%) and B. curvispora (42,9%) followed by B. cynodontis (24,1%). This is the first report of B. cynodontis associated with rice seeds in Brazil and the preliminary identification of B. curvispora in rice by the observation of the teleomorph phase was confirmed. The regional occurrence of a complex of pathogenic Bipolaris species related to brown spot epidemics in other countries, may have epidemiological consequences in the region which deserve further investigation.Uma amostra aleatória (n=165) de lotes de sementes de seis regiões produtoras de arroz no Estado do Rio Grande do Sul (RS), Brasil, ano de colheita 2004, foi examinada em teste blotter de sanidade de sementes. Fungos semelhantes a espécies de Bipolaris e Drechslera foram isolados para identificação das espécies. A observação de estruturas da fase anamórfica (conidióforos e conídios) permitiu separar os isolados em três grupos. Cruzamentos sexuais induzidos em isolados do mesmo grupo, mas originados de diferentes regiões, resultaram em pseudotécio, ascos e ascósporos, permitindo a identificação de três espécies (anamorfo/teleomorfo): Bipolaris cynodontis (Cochliobolus cynodontis); B. curvispora (C. melinidis) e B. oryzae (C. miyabeanus). Todas as espécies estavam distribuídas nas seis regiões produtoras no Rio Grande do Sul. Em geral, a espécie mais prevalente nas amostras foi B. oryzae (44,6%) e B. curvispora (42,9%) seguido de B. cynodontis (24,1%). Este é o primeiro relato de B. cynodontis associada a sementes de arroz no Brasil, e confirma-se a ocorrência de B. curvispora pela observação da fase teleomórfica. A ocorrência de um complexo de espécies patogênicas de Bipolaris, relacionadas com a mancha parda em outros países, pode ter consequências epidemiológicas na região, o que merece futura atenção.

Four fungicides for seed treatments and one as foliar spray were tested in replicated field experiments in a strip plot design to determine the effect of fungicides on Helminthosporium leaf blight (caused by Cochliobolus sativus and Pyrenophora tritici-repentis ) severity and grain yield of wheat. Wheat seed cv. RR 21 was treated with fungicides, carbendazim (Areestin), triadimenol (Baytan), tebuconazole (Raxil), and carboxin + thiram (Vitavax 200B). Single foliar application of propiconazole (Tilt) was sprayed at flowering stage. Controls were included for both factors and treatments were replicated four times. Triadimenol and carboxin + thiram increased seed germination in both years. Triadimenol, tebuconazole, and carboxin + thiram reduced the number of infected seedlings and seedling root rot severity in both years. Number of tillers was higher in carboxin + thiram treated plots compared to other seed treatments. Compared to the control, carboxin + thiram increased grain yield by 9% and 8% in 2004 and 2005, respectively, and triadimenol by 6% in both years. The foliar spray of propiconazole significantly reduced Helminthosporium leaf blight severity and increased thousand-kernel weights. Propiconazole spray increased grain yield by 15% and 14% in 2004 and 2005, respectively. Therefore, seed treatment either with triadimenol or carboxin + thiram in combination with single post-flowering foliar spray of fungicides should minimize grain yield loss due to wheat foliar blight in South Asia. A transmissible disease of the plant pathogenic fungus Helminthosporium victoriae , the causal agent of Victoria blight of oats, was reported more than 50 years ago. Diseased, but not normal, isolates, of H. victoriae contain two distinct viruses designated according to their sedimentation values as victorivirus Helminthosporium victoriae virus 190S (HvV190S) and chrysovirus Helminthosporium victoriae 145S (HvV145S). Although a viral etiology of the disease was previously proposed, conclusive evidence was lacking. Here we present unequivocal evidence based on transfecting virus-free H. victoriae protoplasts with purified virus particles showing that HvV190S is essential for disease development. Furthermore, we show an expansion of the host range of HvV190S to include Cryphonectria parasitica and we also show similarity in a subset of phenotypic traits between HvV190S-infected RNA silencing deficient mutant (Δ dcl-2 ) of C. parasitica and a strain of H. victoriae . In virulence assays on detached American chestnut branches and Red Delicious apple fruits, HvV190S-infected C. parasitica strain Δ dcl-2 was markedly less virulent than wild type and virus-free Δ dcl-2 C. parasitica strains. Furthermore, the hypovirulent HvV190S-infected C. parasitica Δ dcl-2 strain exhibited strong antifungal activity in dual culture with the plant pathogenic fungus Sclerotinia sclerotiorum . No such inhibitory activity was observed in comparable dual cultures with wild type and virus-free Δ dcl-2 C. parasitica strains. Cyclohelminthols I–V ( 1 – 4 ) were isolated from Helminthosporium velutinum yone96. The positions of the carbons attached to chlorine atoms in 2 – 4 were determined by signal splitting due to isotopes in high resolution fully proton decoupled 13 C NMR spectra. Although these natural products gave poor electron circular dichroism (ECD) spectrum because of the presence of a single chromophore in the molecules, the introduction of extra chromophores enabled us to obtain a spectrum with potently split ECD couplings. Their chirality is discussed based on the exciton ECD coupling theory as well as simulated ECD spectra based on density functional theory molecular orbital calculations. The relative configurational relationship between H-6 and H-7 was determined by comparing their 3 J H-6/H-7 values with those of jesterones in literature, as well as with structurally related natural products. The relative configurations were confirmed by a remarkable magnetic shielding of the C1-methyl protons upon naphthoylation of 1 . Spliceosomal twin introns, “stwintrons”, have been defined as complex intervening sequences that carry a second intron (“internal intron”) interrupting one of the conserved sequence domains necessary for their correct splicing via consecutive excision events. Previously, we have described and experimentally verified stwintrons in species of Sordariomycetes, where an “internal intron” interrupted the donor sequence of an “external intron”. Here we describe and experimentally verify two novel stwintrons of the potato pathogen Helminthosporium solani . One instance involves alternative splicing of an internal intron interrupting the donor domain of an external intron and a second one interrupting the acceptor domain of an overlapping external intron, both events leading to identical mature mRNAs. In the second case, an internal intron interrupts the donor domain of the external intron, while an alternatively spliced intron leads to an mRNA carrying a premature chain termination codon. Morphological, cultural, and physiological characteristics of 19 Helminthosporium-like hyphomycetes isolated from the White Sea were studied. Taxonomic status of the isolates was verified using molecular genetics techniques. One of the isolates was identified as Alternatia sp., while the rest of the marine isolates belonged to the species Paradendriphiella salina (G.K. Sutherl.) Woudenb. & Crous. The specific features of the isolates studied were characterized as adaptive. Optimum salinity for their growth was 1-2% NaCl, which is lower than the value for the known open ocean isolates. This is probably due to relatively low salinity of White Sea (22-24‰) as compared with the ocean water (35‰). While the temperature optimum for growth was 22°C, growth and sporulation occurred at 6°C, which has not been reported for marine fungi isolated from warmer seawater. All isolates studied grew and sporulated efficiently on the medium supplied with the Fucus algae extract and in the sea water layer. Conidia of the isolates submerged in the sea water were propagated efficiently, unlike the soil-born fungi. epi -Cochlioquinone D ( 1 ) and its 12-α-hydroxyderivative ( 2 ) were isolated from Helminthosporium velutinum TS28 based on antifungal screening against Cochliobolus miyabeanus . Relative configurations were established by the NMR analyses except for C5 locating far from the other asymmetric centers. ECD discussions involving spectral comparison with those of other related molecules as well as those based on theoretical calculations disclosed not only the C5 configuration but also the chirality for C12–C22 polycyclic moiety. H. velutinum TS28 also afforded precochlioquinol D ( 3 ), just the side chain component, which led us to discuss their biosynthetic pathway. Although 1 and 2 inhibit C. miyabeanus effectively, the C-14 epimeric cochlioquinones scarcely inhibited in spite of very close structures. Present study further revealed that C. miyabeanus tightly recognizes both the polycyclic moiety and the side chain. In contrast, both cochlioquinones and epi -cochlioquinones exhibited potent cytotoxicity against human colon adenocarcinoma (COLO 201) cells regardless of the C14 configuration as well as type of side chains. The fungal maize pathogen Cochliobolus carbonum produces a phytotoxic and cytostatic cyclic peptide, HC-toxin, of structure cyclo(D-prolyl-L-alanyl-D-alanyl-L-Aeo), in which Aeo stands for 2-amino-9,10-epoxi-8-oxodecanoic acid. Here we report the isolation of a gene, TOXC, that is present only in HC-toxin-producing (Tox2+) fungal strains. TOXC is present in most Tox2+ strains in three functional copies, all of which are on the same chromosome as the gene encoding HC-toxin synthetase. When all copies of TOXC are mutated by targeted gene disruption, the fungus grows and sporulates normally in vitro but no longer makes HC-toxin and is not pathogenic, indicating that TOXC has a specific role in HC-toxin production and hence virulence. The TOXC mRNA is 6.5 kb and the predicted product has 2,080 amino acids and a molecular weight of 233,000. The primary amino acid sequence is highly similar (45 to 47% identity) to the β subunit of fatty acid synthase from several lower eukaryotes, and contains, in the same order as in other β subunits, domains predicted to encode acetyl transferase, enoyl reductase, dehydratase, and malonyl-palmityl transferase. The most plausible function of TOXC is to contribute to the synthesis of the decanoic acid backbone of Aeo.

Prototype victorivirus HvV190S employs stop/restart translation to express its RdRp from the downstream ORF in its bicistronic mRNA. The signals for this activity appear to include a predicted RNA pseudoknot directly upstream of the CP stop and RdRp start codons, which overlap in the motif AUGA. Here we used a dual-fluorescence system to further define which HvV190S sequences are important for stop/restart translation and found that the AUGA motif plus 38 nt directly upstream are both necessary and sufficient for this activity. This RNA cassette encompasses the predicted pseudoknot, and indeed substitutions that disrupted the pseudoknot disrupted the activity whereas complementary substitutions that restored the pseudoknot restored the activity. Replacement of this RNA cassette with those from other victoriviruses with a predicted pseudoknot in comparable position also supported stop/restart translation. Helminthosporium solani is a slow-growing fungal pathogen belonging to the family Massarinaceae. It causes blemishes on potato tubers, affecting processing and fresh market trade. Despite its worldwide distribution, little is known about the biology of H. solani. Here we report the generation of a draft genome sequence of H. solani with an estimated genome size of ~35 megabases, for use in phylogenetic and pathogenicity studies of this fungal pathogen. This sequence is also the first reference genome within the family Massarinaceae. We identified a large suite of genes in the H. solani genome that encode putative cell wall degrading enzymes. Based on comparison with other genomes, we speculate that H. solani is a hemibiotroph or necrotroph. The presence of a large number of genes in the glycoside hydrolase (GH) 10 and 43 families, which aid in the hydrolysis of glucoronoarabinoxylan, also suggests that H. solani may be able to survive on grass hosts and indicates the need to re-examine the life cycle and host range of this pathogen. The tubers of Bletilla formosana were fermented with eight plant pathogen fungi, respectively, and antioxidant activities and total phenolic content (TPC) of the crude extracts of fermented products and non-fermented products were investigated. The antioxidant activities were evaluated in three different test systems [DPPH, ABTS radical-scavenging activity, and ferric reducing-antioxidant power (FRAP)]. It was found that the extract of Helminthosporium maydis fermented B. formosana (FBF) possessed the highest TPC and exhibited a significant antioxidant activity compared with non-fermented product and other fermented products. Correlation analysis between antioxidant activities and TPC was also investigated. The good correlation between antioxidant activities and TPC revealed that the phenolic compounds might be the major contributors for the high antioxidant activities of the fermented B. formosana . Two phenolic compounds, curvularin and dehydrocurvularin, were isolated from H. maydis FBF, which had never been reported from plant of orchidaceae or H. maydis . Curvularin exhibited significant antioxidant activities, and was also present at a high concentration (0.373 mg/mg extract sample), implying an important role for the antioxidant activity of H. maydis FBF. The use of antagonist microorganisms against fungal plant pathogens is an attractive and ecologically alternative to the use of chemical pesticides. Streptomyces are beneficial soil bacteria and potential candidates for biocontrol agents. This study reports the isolation, characterization and antagonist activity of soil streptomycetes from the Los Petenes Biosphere Reserve, a Natural protected area in Campeche, Mexico. The results showed morphological, physiological and biochemical characterization of six actinomycetes and their inhibitory activity against Curvularia sp., Aspergillus niger, Helminthosporium sp. and Fusarium sp. One isolate, identified as Streptomyces sp. CACIS-1.16CA showed the potential to inhibit additional pathogens as Alternaria sp., Phytophthora capsici, Colletotrichum sp. and Rhizoctonia sp. with percentages ranging from 47 to 90 %. Sorghum grain mold fungi like Alternaria solani, Curvularia lunata, Fusarium moniliforme andHelminthosporium sp, were tested with hexane fraction (AMIE-HF), chloroform fraction (AMIE-CF), ethyl acetate fraction (AMIE-EAF), crude extract (AMIE) of Annona muricata, petroleum ether extract of Abutilon indicum (AIPE) and alkaloid enriched fraction of Evolvulus alsinoides (EAMAA I). AMIE HF at 1000, 2500 and 5000 ppm and AMIE EAF at 2500 and 5000 ppm concentration showed significant effect on F. moniliforme while AMIE CF and AMIE showed no effect. AIPE remarkably inhibited A. solani at all concentrations and C. lunata at 2500 and 5000 ppm concentration. Indigenous people especially in southern Nigeria use Corchorus olitorius L. (Jute) as a staple vegetable. Population dynamics, richness and frequency of occurrence of microflora isolates on healthy green leaves of wild okra were estimated within two weeks at weekly intervals using the dilution technique. This study was conducted in the University of Benin intend to show the diversity of microorganisms on the leaves of wild okra. The leaves were categorized based on their period of harvest into old, new and middle with a week interval between each harvest. After serial dilution in distilled water, isolation was done using nutrient agar for bacteria and potato dextrose agar for fungi. After incubation colony forming units per millimeter were counted, isolated, identified and characterized using standard microbiological techniques. The fungal diversity and frequency of occurrence were higher in the first sampling (61.50% and 62.07% respectively) than those of the second sampling (38.50% and 37.93%). Total viable microbial population in the second sampling after two weeks was higher (11.23 X 102cfu/ml) than in the first sampling after one week (10.00 X 102cfu/ml). The total cumulative bacterial count was higher (15.69X 102cfu/ml) than those of fungi (55.40 X 102 cfu/ml) during the studies. Bacterial genera isolated included; Staphylococcus, Bacillus, Micrococcus, Serratia and Proteus. Rhodotorula, Mucor, Trichoderma, Aspergillus, Penicillium, and Helminthosporium were the genera of fungi isolated. Silver scurf, caused by Helminthosporium solani, and black dot, caused by Colletotrichum coccodes, cause tuber blemishes on potato ( Solanum tuberosum L.) which affect processing and fresh market trade. Tubers from ten cultivars were collected at harvest from three organic farms in Wisconsin and categorized as symptomatic or asymptomatic based on visual symptoms of silver scurf and black dot and/or signs of H. solani and C. coccodes. Tuber incubation and PCR assays were performed on asymptomatic tubers to detect H. solani and C. coccodes. Tuber incubation and PCR assays were in slight to fair agreement (kappa coefficient <0.4) for detecting both pathogens. Most asymptomatic tubers tested were positive by one or both assays for H. solani (75 %) or C. coccodes (94 %). Minituber inoculation assays were also performed to screen potato lines for resistance to silver scurf. Of the 14 lines tested, a diploid interspecific hybrid, C287, had consistently low sporulation, suggesting it has partial resistance to silver scurf. Since the majority of tubers harvested are already infected with one or both pathogens further research should focus on organically acceptable management practices that may inhibit disease development in field and in storage. Abstract: Four prenylated flavonoids, isoglabratephrin, (+)-glabratephrin, tephroapollin-F and lanceolatin-A were isolated from Tephrosia apollinea L. growing in Egypt. The structures of compounds have been elucidated using physical and spectroscopic methods including (UV, IR, 1H NMR, 13C NMR, DEPT, 2D 1H–1H COSY, HSQC, HMBC and NOESY). The isolated flavonoids showed considerable antifungal activity against four phytopathogenic fungi, namely Alternaria alternata, Helminthosporium sp., Colletotrichum acutatum and Pestalotiopsis sp. in a dose-dependent manner using the agar well-diffusion bioassay. They differ significantly in their activity with tephroapollin-F was the most effective. In a test using a concentration of 4 mg/ml of tephroapollin-F, strong fungicidal activities (32.8–58.3%) were produced against the test fungi, where C. acutatum, Helminthosporium sp. and Pestalotiopsis sp. showed greater susceptibility, while A. alternata was the least susceptible. Using the same concentration, the two flavonoids isoglabratephrin and (+)-glabratephrin showed moderate activities with % inhibition of fungal growth were ranged between (16.1–37.8) against A. alternata, Helminthosporium sp. and Pestalotiopsis sp., while showed a strong antifungal activity against C. acutatum (% growth inhibition were 46.4 and 42.9, respectively). In all treatments, the flavonoid lanceolatin-A exhibited weak to moderate activities. Using lower concentrations of the test flavonoids (2 and 1 mg/ml), weak to moderate antifungal activities were observed against all of the test fungal strains. In all cases and regardless of the flavonoid tested, C. acutatum was the most susceptible, while A. alternata was the least. Fungi affecting leaves of Capsicum chinense Jacq., Heliconia sp., Veitichia merrillii Becc., Polianthes tuberosa L., Jatropha curcas L. and Thevethia ahouai A. DC. were identified by using traditional techniques. Eight strains identified as members of the genera Curvularia, Alternaria and Helminthosporium were isolated from leaves of damaged plants. The effectiveness of the systemic fungicides Azoxystrobin, Benomyl, Fosetil Al, Tiabendazole, Imazalil and Procloraz in controlling these pathogens were evaluated. Under the tested conditions, Imazalil caused 100% growth inhibition in all the fungal isolates, while the growth inhibition caused by Fosetil-Al and Procloraz ranged from 75 to 100%. Abstract: Silver scurf caused by Helminthosporium solani and dry rot caused by Fusarium spp. are tuber diseases of economic importance in potato-growing areas worldwide. Recently, the two pathogens have developed resistance to thiabendazole (TBZ), a post-harvest fungicide commonly used for their control. Therefore, alternative disease control strategies are needed. The present study assessed the efficacy of the biopesticides Bio-Save 10LP (Pseudomonas syringae-strain ESC-10; Ps10) and Bio-Save 11LP (P. syringae-strain ESC-11; Ps11) against silver scurf and dry rot. Approximately 30 isolates representing the genus Fusarium were obtained from symptomatic potato specimens with dry rot from New Brunswick (NB), Nova Scotia (NS), Prince Edward Island (PE) and Alberta (AB), Canada. Species isolated were Fusarium sambucinum, Fusarium tumidum, Fusarium coeruleum, Fusarium culmorum, and Fusarium avenaceum. H. solani isolated from AB, NB and PE was included in the study as the causal agent of silver scurf. The efficacy of P. syringae against F. sambucinum and H. solani was tested in vitro. Ps10 and Ps11 inhibited the growth of H. solani up to 68% (NB isolate) and 73% (PE isolate), respectively and the inhibition was more or less comparable with that of TBZ. F. sambucinum was not significantly inhibited by Ps10; however Ps11 significantly inhibited AB, PE and NB isolates by 43%, 28% and 54%, respectively. Conversely, TBZ inhibited AB, PE and NB isolates of Fusarium spp. in vitro by 86%, 88% and 100%, respectively. TBZ in combination with either Ps10 or Ps11 did not always reduce the growth of H. solani or Fusarium spp. in vitro compared to that of TBZ alone. Storage trials conducted in NB and PE assessed the efficacy of P. syringae against H. solani or Fusarium spp. in vivo and confirmed that the application of P. syringae or TBZ alone or in combination significantly reduced the incidence and/or severity of silver scurf and Fusarium dry rot. Ps11 alone or in combination with TBZ was significantly more effective than Ps10 in controlling silver scurf disease severity. The reduction in disease severity of dry rot and silver scurf in storage due to Ps10, Ps11, or TBZ or their combinations was consistently comparable. Results indicate that the use of P. syringae (strains ESC-11 or ESC-10) as a post-harvest treatment can contribute to the management of both silver scurf and Fusarium dry rot in potato storages. Abstract: Many naturally occurring plant volatile compounds are known for their anti-fungal properties. In this study, acetaldehyde and 2E-hexenal were chosen as prototype volatiles in order to investigate the use of volatile compounds for control of blemish pathogens in fresh-pack potato packaging. Pure cultures of the three main potato blemish pathogens, Pectobacterium atrosepticum (bacterial soft rot), Colletotrichum coccodes (black dot), and Helminthosporium solani (silver scurf), were used in the study. Pathogen cultures were exposed to the pure volatiles that were injected into the atmosphere of sealed jars for 4–8days at 23°C. Results showed that 2E-hexenal was the most effective of the two volatiles with 5μL/L providing complete inhibition of growth for all three pathogens in vitro. Cytological studies showed that a concentration of 2.5μL/L of 2E-hexenal was capable of inhibiting germination in both fungal pathogens. These results suggest that the primary mode of action of 2E-hexenal was inhibiting germination for fungi and suppressing bacterial growth. The quantities required to achieve pathogen inhibition are extremely low. This study suggests that these volatiles may be used to effectively manage potato postharvest blemish diseases in storage. A survey of the prevalence of skin blemish diseases in potatoes after the growing seasons of 2008 and 2009 was carried out on 247 potato lots representing different cultivars and production regions in Norway. The results showed the presence of silver scurf ( Helminthosporium solani) in all lots. Skin spot ( Polyscytalum pustulans) and black scurf ( Rhizoctonia solani) were found in 80% of the lots, and black dot ( Colletotrichum coccodes) and common scab caused by Streptomyces spp. were present in 50-70%. Also, powdery scab ( Spongospora subterranea) occurred in 65-80% of the lots, and root-lesion nematodes ( Pratylenchus spp.) were detected in 60% of the sub-samples that exhibited symptoms of common scab.
