'Regent' is the fungal resistant grapevine cultivar with the highest acreage in Germany and an important resistance donor in international breeding programs. It carries the resistance loci Rpv3.1 as well as Ren3 and Ren9 against downy and powdery mildew, respectively. As the parents of 'Chambourcin', the resistant paternal ancestor of 'Regent', did not coincide with the breeder's information, the germplasm repository of JKI Geilweilerhof was screened to find the missing ancestors. SSR marker analysis revealed that 'Joannes Seyve 11369' and 'Plantet' are the true parents of 'Chambourcin' and not 'Seyve Villard 12-417' and 'Chancellor'. Furthermore, the origin of the resistance loci Ren3 and Ren9 could be traced back to the genotypes 'Seibel 4614' and 'Munson'. Since the breeder Hermann Jaeger mentioned 'Munson' as a direct descendant of Vitis aestivalis Michx. var. linsecomii (Buckley) L. H. Plant roots are essential for water and nutrient uptake and contribute to the plants' response to environmental stress factors. As the hidden half of a plant, investigation of root systems is highly challenging, most of available methods are destructive and very labour-intensive. In this proof-of-concept study, a non-invasive X-ray micro computed tomography (X-ray µCT) method was applied to investigate the phenotypic variation of the complex three-dimensional (3D) architecture of grapevine roots as a function of genotype and soil. Woody cuttings of 'Calardis Musqué', 'Villard Blanc' and V3125 ('Schiava Grossa' x 'Riesling') were cultivated in polypropylene columns filled with two different soil types, clay loam and sandy loam, for 6 weeks. Afterwards, the columns were scanned once using the technique of X-ray µCT. The received raw data were analysed for the reconstruction of 3D root system models (3D model), which display a non-destructive visualization of whole, intact root systems with a spatial resolution of 42 µm. The 3D models of all investigated plants (in total 18) were applied to quantify root system characteristics precisely by measuring adventitious root length, lateral root length, total root length, root system surface area, root system volume and root growth angles from the woody cutting relative to a horizontal axis. The results showed that: (i) early root formation and root growth differed between genotypes, especially between 'Calardis Musqué' and 'Villard Blanc'; and (ii) the soil type does influence adventitious root formation of V3125, but had minor effects on 'Calardis Musqué' and 'Villard Blanc'. In conclusion, this innovative, high-resolution method of X-ray µCT is suitable for high resolution phenotyping of root formation, architecture, and rooting characteristics of grapevine woody cuttings in a non-destructive manner, e.g. to investigate root response to drought stress and would provide new insights into phylloxera root infection., VITIS - Journal of Grapevine Research, Vol. 60 No. 1 (2021): Vitis

A biparental population segregating for downy mildew resistance was studied to identify resistance linked molecular markers. The progeny of 202 individuals from a cross of V3125 (susceptible breeding line) with 'Börner' (resistant rootstock) was phenotyped in the field in four seasons and by evaluating artificially infected leaf discs. QTL mapping revealed a major resistance locus on chromosome 5 that explained up to 17.4 % of the phenotypic variance. This new resistance locus was named Rpv14. It was transmitted from the male grandparent V. cinerea Arnold to 'Börner' and is associated with the marker GF05-13., VITIS - Journal of Grapevine Research, Vol. 55 No. 2 (2016): Vitis

In a double-blind, randomised, cross-over trial clofibrate and a combination of polyenyl phosphatidyl choline (PPC) plus clofibrate were tested in 67 patients with hyperlipoproteinemia. Each treatment lasted for 4 weeks and was separated by a 4 week placebo period. The daily doses were clofibrate 1.2 g and PPC 1.8 g + clofibrate 1.2 g. respectively. The results revealed that polypenyl phosphatidyl choline prevented the elevation of LDL-cholesterol induced by clofibrate treatment, and that the lipid-lowering potency of the combination did not differ significantly from that of clofibrate. Since elevation of LDL-cholesterol is considered to increase the risk of coronary heart disease, the combination appears to offer a therapeutic advantage. Despite the significance of this clinical observation, a final decision may only be obtained from a prospective, long term investigation in patients with coronary heart diseases and hyperlipoproteinemia.

The metabolic activity of the red cell glycolytic pathway hexose monophosphate shunt (HMP) with dependent glutathione system was studied in patients with hyperthyroidism (n = 10), hyperlipoproteinemia (n = 16), hypoglycemia (n = 25) and hyperglycemia (n = 23). In uncontrolled diabetics and patients with hyperthyroidism the mean value of glucose phosphate isomerase (GPI), glucose-6-phosphate dehydrogenase (G-6-PD), glutathione reductase (GR) was increased, whereas these enzyme activities were reduced in patients with hypoglycemia. Apart from a few values of hexokinase (HK) which were lower than normal the results in hyperlipoproteinemia patients remained essentially unchanged, including the intermediates such as 2,3-diphosphoglycerate (2,3-DPG), adenosine triphosphate (ATP) and reduced glutathione (GSH). While increased rates of 2,3-DPG and ATP in hypoglycemia patients were obtained, these substrates were markedly reduced in diabetics.

Serum proinsulin and insulin levels were measured on 55 normal or overweight women before and after oral glucose administration. The proinsulin proportion of basal total insulin was 70% in women of normal weight. With increasing overweight the relation shifted in favour of insulin. After stimulation with glucose, proinsulin levels were significantly raised, analogous to total insulin, but les marked than the latter. The increased total insulin excretion in obesity was, therefore, largely due to insulin and less to proinsulin. The greater the overweight the later maximal insulin levels were reached after oral glucose administration: proinsulin peaks occurred later than insulin peaks. Measurement of areas from single values and corresponding times for proinsulin and insulin, after stimulation, indicated their significant correlation with the degree of overweight. In women of more than 70% overweight (Broca index), reactive proinsulin and insulin excretion decreased again despite an increase in body weight. They had a definitely reduced carbohydrate tolerance. After reduction in body weight previously increased proinsulin levels fell again. The significance of higher proinsulin levels in fasting subjects, which increased after stimulation and with overweight but were in percentage terms less than those of reactive insulin, remains unexplained.

Healthy fasted volunteers were subjected to an acute oral ethanol load over 12 h after a diet of 3 days with high linolenic acid content. Free fatty acids, triglycerides, glycerol, phospholipids, cholesterol and insulin, as well as the fatty acid pattern of triglycerides in the plasma, were determined during the test. The test was repeated with nicotinic acid added. The lipid values obtained and the comparisons of fatty acid composition both indicate that the predominant role of peripheral lipolysis in the genesis of acute ethanol-induced hypertriglyceridemia, in spite of the possibility of enhanced synthesis of palmitic acid in the liver.

Innerhalb eines Jahres konnte bei 14 640 erstmals untersuchten Patienten 36mal eine bis dahin nicht bekannte Sarkoidose diagnostiziert werden. Der Haufigkeit nach fand sich ein deutliches Uberwiegen der akuten Verlaufsform (23 Patienten) gegenuber primar-chronischen Fallen (13 Patienten). Auf die Klinik der Erkrankung wird anhand des vorliegenden Patientengutes eingegangen. Die Diagnose sollte stets histologisch gesichert werden; dies war in 52% durch Leberblindpunktion, in 100% durch Mediastinoskopie und in 50% durch Bronchoskopie moglich. Die Problematik der einzelnen bioptischen Methoden wird diskutiert.

After healthy persons, aged between 20 and 35 years, had consumed either ethanol or ethanol and fructose, triglycerides, free glycerol, FFA, phospholipids and total cholesterol were determined. After a basic dosage of 0.5 g ethanol/kg body weight, each person received a maeintenance dosage of 0.1 g ethanol/kg body weight and hour. Control experiments were carried out on persons receiving only fructose and on fasting persons who consumed no ethanol. After 8 hrs, triglyerides rose in the ethanol group by 107 % , in the ethanol-fructose group by 63 %. FFA exhibited in both ethanol and ethanol-fructose groups an initial decrease, with a secondary increase in the ethanol group. The initial decrease was greater in the ethanol-fructose group. A significant rise in free glycerol by 419 % was observed 30 min after the intake of combined ethanol/fructose. Free glycerol rose under ethanol alone by 144 %. The ohospholipids exhibited a slight rise in the ethanol group; no significant changes occured in the cholesterol. The blood ethanol level was lower under ethanol-fructose than under ethanol alone. The addition of fructose diminishes the ethanol-induced hypertriglyceridemia. Our investigations give further proof that, under short-term ethanol load, the fatty acids necessary for the increased triglyceride synthesis in the liver, originate predominantly from a peripheral lipolysis, and that changes in liver metabolism depending on the oxidation of ethanol are not of less importance for the development of the acute ethanol-induced hyperlipidemia.
