Yeni ve yeniden çıkan hastalıklar, insan ve hayvan sağlığını tehdit ettiğinden tüm dünyada araştırılması öncelikli patojenler arasında bulunmaktadır. Bu patojenlerin ülkelere nasıl geldiği ve bulaşma siklusunun nasıl olduğu yapılan araştırmalar ile aydınlatılmaya çalışılmaktadır. Bu patojenlerin taşınmasında göçmen kuşlar önemli bir rol oynamaktadır. Bu tez çalışması kapsamında da göçmen kuşların kullandığı göç rotaları incelenerek Samsun ve Hatay illerindeki kuşlardan Flavivirus, Nairovirus ve Phleboviruslerin araştırılması amacıyla numuneler toplandı. Toplamda 117 kuştan elde edilen 464 numuneye belirtilen virus aileleri için RT-PCR ve Zika virus için Real Time RT-PCR yöntemleri kullanılarak analizler yapıldı. Hatay'dan toplanan bir kartalda Batı Nil Virusu tespit edildi. Yapılan genetik analizlere göre de Genetik Hat 1 Alttip 1a olarak sınıflandırıldı. Hatay'dan toplanan üç kuşta (flamingo, pelikan ve leylek) ve Samsun'dan toplanan 14 kuşta (4 ördek, 5 keklik, 4 bıldırcın ve 1 güvercin) ise Toscana virus (TOSV) tespit edildi. Samsun bölgesinden toplanan kuşların %25'i TOSV yönünden pozitif bulunduğundan bu hastalığa dikkat edilmesi gerekmektedir. Yapılan analizlere göre de Hatay'dan bir kuş Genotip A ve geri kalan 16 kuş ise Genotip B olarak sınıflandırılmıştır. Pozitif tespit edilen dokular hücre kültürlerine inokule edildikten sonra güvercin, ördek ve kekliğe ait organlardan TOSV izolasyonu yapılmıştır. Kuşlarda tespit edilen BNV ve TOSV ile ülkemizde bu hastalıkların sirkülasyonunda kuşların rolleri olabileceği görüldü. Ayrıca bu çalışma ile dünyada ilk defa kuşlarda TOSV tespiti yapılmıştır. Bu bulgu ile kuşların TOSV sirkülasyonuna katıldığı ve virusun ülkeler arasında vahşi/göçmen kuşlar aracılığıyla taşınabileceğine dair kanıt elde edilmiştirKeywords: Arbovirus, Migrating Birds, Toscanavirus, West Nile Virus, Wildlife. Emerging and re-emerging diseases that threaten human and animal health have priority for researches among the other pathogens all over the World. How these pathogens arrive in countries and how the transmission cycle occurs are explained by researches. Migrating birds have important role in transmissions of these pathogens. The migrating routes used by migratory birds were examined and samples were collected to investigate Flavi- Nairo ve Phleboviruses from the birds in Samsun and Hatay provinces, within the scope of this thesis.464 samples collected from 117 birds were analyzed by RT-PCR and qRT-PCR for these viruses. An eagle sample which collected from Hatay was found positive for West Nile Virus (WNV) and it was classified as Genetic Line 1 Subtype 1a according to genetic analysis. Toscana virus (TOSV) was detected in three birds (flamingo, pelicans and storks) collected from Hatay and 14 birds (4 ducks, 5 partridges, 4 quails and 1 pigeon) collected from Samsun. It should be noted that 25% of birds collected from Samsun region are found positive for TOSV. One bird from Hatay was classified as Genotype A while the other 16 birds were classified as Genotype B. Moreover, TOSV isolated from pigeon, duck and partridge's organs after the positive tissues were inoculated into cell cultures. Detection of WNV and TOSV in birds showed that birds have a role in the circulation of these infections in our country. In addition, this is the first detection of TOSV in birds. Bu tez çalışmasında mezbahada kesime getirilen sığırlarda Bovine Herpes Virus -1 (BoHV-1) ve Bovine Parainfluenza virus 3 ( BPIV-3) dağılımının araştırılması amacıyla sığırlardan kan serumu, tracheal swap, nazal swap ve lezyonlu akciğer dokusu örnekleri toplandı. Serolojik açıdan homolog antikorlar Virus Nötralizasyon Testi (VNT) ve moleküler açıdan kontrol BoHV-1 için PCR ve BPIV-3 için RT-PCR yöntemi kullanılarak yapıldı.Toplam 146 sığır örneklerinin 73 tanesi Ekim- Kasım aylarında ve 73 tanesi Şubat- Mart aylarında toplandı.BoHV-1 antikorları yönünden toplamda 42 tanesi (% 28.7) serum pozitif , BPIV-1 yönünden 52 adet (%35.6) serum pozitif olarak bulundu. Toplam örnekler içinde17 tanesi (% 11.6) hem BoHV-1 hem de BPIV-3 yönünden pozitif olarak değerlendirildi. Toplam örnekler içerisinde BoHV-1 yönünden pozitif kabul edilen serumların % 40.4'ü BPIV-3 yönünden de pozitif olarak bulundu. Yine toplam örnekler içerisinde BPIV-3 yönünden pozitif olanların % 32.6'ü BoHV-1 yönünden de pozitif bulundu.Ekim- Kasım aylarında alınan 73 kan serumu örneğinin 13 tanesi (% 17.8) BoHV-1 antikorları yönünden, 32 tanesi (% 43.8) BPIV-3 antikorları yönünden pozitif bulundu. Serum örneklerinin 5 tanesi (% 6.8) hem BoHV-1, hem de BPIV-3 yönünden pozitif bulundu. BoHV-1 yönünden pozitif bulunan örneklerden % 38.4'ü BPIV-3 yönünden de pozitif olarak belirlendi. BPIV-3 yönünden pozitif bulunan örneklerin % 15.6'sı BoHV-1 yönünden de pozitif olarak belirlendi.Şubat- Mart aylarında toplanan 73 kan serumu örneğinin 29 tanesi (% 39.7) BoHV-1 antikorları yönünden, 20 tanesi (% 27.3) BPIV-3 antikorları yönünden pozitif bulundu. Serum örneklerinin 12 tanesi (% 16.4) hem BoHV-1 hem de BPIV-3 yönünden pozitif bulundu. BoHV-1 yönünden pozitif bulunan örneklerden % 41.3'ü aynı zamanda BPIV-3 yönünden de pozitif olarak belirlendi. BPIV-3 yönünden pozitif bulunan % 60'ı aynı zamanda BoHV-1 yönünden de pozitif bulundu.Moleküler açıdan yapılan kontrolde BoHV-1 için pozitiflik tespit edilemedi. BPIV-3 için 1 adet nazal swap örneği pozitif bulundu.Anahtar kelimeler: BoHV-1, BPIV-3, Bovine herpesvirus -1, Bovine Parainfluenza -3, PCR In this study, Bovine Herpes Virus -1 (BoHV-1) and Bovine Parainfluenza virus 3 ( BPIV-3) investigated in cattle and collected serum samples, tracheal swaps, nasal swaps and lungs samples that have lesions from the slaughterhouse.Virus Neutralization Test (VNT) used for serological studies and PCR used for BoHV-1 and RT-PCR used for BPIV-3 for molecular studies. Totally 146 cattle samples tested and 73 samples collected in September- October and 73 samples collected in February- March.A total of 42 samples (28.7%) were detected seropositivite for BoHV-1 and 52 samples (35.6%) were detected seropositive for BPIV-3. Total of 17 samples (11.6%) were detected seropositive for BoHV-1 and BPIV-3. The samples which are seropositive for BoHV-1 were detected 40.4% seropositive for BPIV-3. The samples which are seropositive for BPIV-3 were detected 32.6% seropositive for BoHV-1.Thirteen samples (17.8%) were detected seropositive for BoHV-1 and 32 samples (43.8%) were detected seropositive for BPIV-3 in Semptember- October term. In this term, 5 serum samples (6.8%) were detected positive for both BoHV-1 and BPIV-3. The samples which are seropositive for BoHV-1 were detected 38.4% seropositive for BPIV-3. The samples which are seropositive for BPIV-3 were detected 15.6% seropositive for BoHV-1.Twentynine samples (39.7%) were detected seropositive for BoHV-1 and 20 samples (27.3%)Were detected seropositive for BPIV-3 in February-March term. In this term, 12 samples (16.4%) were detected positive for both BoHV-1 and BPIV-3. The samples which are seropositive for BoHV-1 were detected 41.3% seropositive for BPIV-3. The samples which are seropositive for BPIV-3 were detected 60% seropositive for BoHV-1.In molecular studies, none of the samples were detected positive for BoHV-1 and 1 sample of nasal swap were detected positive for BPIV-3.Key Words: BoHV-1, BPIV-3, Bovine herpesvirus-1, Bovine Parainfluenza ? 3, PCR 67
