Abstract Exposure to mixtures of toxic metals is known to cause adverse health effects through epigenetic alterations. Here we aimed to examine the unexplored area of aberrant DNA methylation in the H19/IGF2 domain following combined toxic metal exposure. An in vitro epigenotoxicity assay using the human normal liver epithelial cell line THLE-3 was conducted. When THLE-3 cells were exposed to specific concentrations of either organic arsenic or MeHgCl, an increase in the H19 lncRNA levels and a marked reduction in the IGF2 mRNA levels were observed. In contrast, combined exposures coupled with CdCl2 resulted in the transcriptional repression of H19 and transcriptional activation of IGF2. It should be noted that the correlation between the dysregulated expression of H19/IGF2 and the hypermethylated CpG sites within the H19 differentially methylated region (DMR) was statistically significant. Furthermore, we performed transcriptomic analysis of the hepatocytes exposed to toxic metal combinations indicating enrichment of pro-inflammatory and anti-proliferative pathways compared to the unexposed cells. Our results suggest that hazardous metal mixtures may trigger epigenetic aberrations at the H19/IGF2 locus. We propose that altered CpG methylation in the H19 DMR could be a candidate biomarker for hepatic epigenotoxicity, in part, due to environmental exposure.

Background: Currently, the efficient production of chimeric mice and their survival are still challenging. Recent researches have indicated that preimplantation embryo culture media and manipulation lead to abnormal methylation of histone in the H19/Igf2 promotor region and consequently alter their gene expression pattern. This investigation was designed to evaluate the relationship between the methylation state of histone H3 and H19/Igf2 expression in mice chimeric blastocysts. Methods: Mouse 129/Sv embryonic stem cells (mESCs) expressing the green fluorescent protein (mESCs-GFP) were injected into the perivitelline space of 2.5 days post-coitis (dpc) embryos (C57BL/6) using a micromanipulator. H3K4 and H3K9 methylation, and H19 and Igf2 expression was measured by immunocytochemistry and q-PCR, respectively, in blastocysts. Results: Histone H3 trimethylation in H3K4 and H3K9 in chimeric blastocysts was significantly less and greater, respectively (p< 0.05), than in controls. H19 expression was significantly less (p< 0.05), while Igf2 expression was less, but not significantly so, in chimeric than in control blastocysts. Conclusions: Our results showed, that the alteration ofH3K4me3 and H3K9me3 methylation, change H19/Igf2 expression in chimeric blastocysts. [ABSTRACT FROM AUTHOR]

To study the alleviating effects of flavonoids from Cuscuta chinensis (CCFs) on ovary injury in female offspring of pregnant mice exposed to BPA, five groups (n = 20) of pregnant mice were intragastrically administrated with BPA (5 mg/kg/day) and CCFs (20 mg/kg/day, 30 mg/kg/day, 40 mg/kg/day) at pregnancy days 1–18. The ovaries and serum of F1 female mice were collected at postnatal day (PND) 21 and PND 56 for the detection of related indicators. The ovarian and testicular histomorphologies were observed with hematoxylin-eosin staining (H&E). The levels of follicle-stimulating hormone (FSH) and luteinizing hormone (LH) and the contents of estradiol (E2), progesterone (P4), and testosterone (T) in serum were detected by radioimmunoassay. The contents of ovarian and testicular estrogen receptor alpha (ERα) and estrogen receptor beta (ERβ) were detected by enzyme-linked immunosorbent assay (ELISA). The expression of caspase-7, caspase-9, bcl-2, and bax in ovaries and testes of offspring mice were detected by Western blot, and apoptosis in ovaries and testes was detected by TUNEL. The mRNA relative transcription levels of ERα, progesterone receptor (PgR), DNA methyltransferase1 (Dnmt1), DNA methyltransferase3A (Dnmt3A), and DNA methyltransferase3B (Dnmt3B) were detected by real-time quantitative PCR (RT-qPCR). The ovary of female offspring with PND 56 was treated with bisulfite sequence PCR (BSP). Our results showed that, compared with the BPA group, 40 mg/kg CCFs significantly reduced the ovarian index of F1 females and the ovarian cytoapoptosis (P < 0.01). CCFs also can alleviate the injure of the levels of serum hormone, hormone receptors, and DNMTs induced by BPA in F1 females at PND 21 and PND 56. Forty milligrams per kilogram of CCFs significantly inhibited the hypermethylation of the H19/Igf2 imprinted gene induced by BPA (P < 0.01). It indicated that CCFs adjusted H19/Igf2 methylation by increasing the expression of DNMTs, thereby increasing the levels of reproductive hormones and receptors along with reducing the cytoapoptosis. Abstract The sexually dimorphic expression of H19/IGF2 is evolutionarily conserved. To investigate whether the expression of H19/IGF2 in the female porcine eye is sex-dependent, gene expression and methylation status were evaluated using quantitative real-time PCR (qPCR) and bisulfite sequencing PCR (BSP). We hypothesized that H19/IGF2 might exhibit a different DNA methylation status in the female eye. In order to evaluate our hypothesis, parthenogenetic (PA) cells were used for analysis by qPCR and BSP. Our results showed that H19 and IGF2 were over-expressed in the female eye compared with the male eye (3-fold and 2-fold, respectively). We observed a normal monoallelic methylation pattern for H19 differentially methylated regions (DMRs). Compared with H19 DMRs, IGF2 DMRs showed a different methylation pattern in the eye. Taken together, these results suggest that elevated expression of H19/IGF2 is caused by a specific chromatin structure that is regulated by the DNA methylation status of IGF2 DMRs in the female eye.

Objective: To investigate the consequences of removing embryos from the incubator daily, and exposing them to ambient air at 37.0 °C for short periods of time, on embryo development, gene expression and the health of the conceptus. Methods: An in vitro cross-sectional study was undertaken using CBA F1 mouse zygotes exposed to ambient air at 37.0 °C for 3, 5, and 8 min daily during their entire in vitro development. Blastocysts were analyzed for total cell numbers, apoptosis, and H19 and Igf2 expression and transferred to pseudopregnant recipient females to examine pregnancy, fetal and placental weight, and placental H19 and Igf2 expression. Results: Short exposure of pre-implantation embryos to ambient air at 37.0 °C resulted in a statistically significant reduction in blastocyst development and total cell numbers, increased apoptotic cell index, and impaired H19 and Igf2 expression in the blastocyst. Implantation rates were significantly reduced and fetuses and placentae displayed a significant reduced weight on day 18 of pregnancy. Placentae displayed significantly reduced levels of H19 and Igf2 expression. Conclusions: Exposing mouse pre-implantation embryos to ambient air at 37.0 °C, even for brief periods for routine micromanipulations is detrimental to normal embryonic development. Our results highlight the importance of how small alterations in the culture environment can have major consequences for the health of the embryo.

The imprinted genes of primate embryonic stem cells (ESCs) often show altered DNA methylation. It is unknown whether these alterations emerge while deriving the ESCs. Here we studied the methylation patterns of two differentially methylated regions (DMRs), SNRPN and H19/IGF2 DMRs, during the derivation of monkey ESCs. We show that the SNRPN DMR is characteristically methylated at maternal alleles, whereas the H19/IGF2 DMR is globally highly methylated, with unusual methylation on the maternal alleles. These methylation patterns remain stable from the early stages of ESC derivation to late passages of monkey ESCs and following differentiation. Importantly, the methylation status of H19/IGF2 DMR and the expression levels of IGF2, H19, and DNMT3B mRNAs in early embryo-derived cells were correlated with their capacity to generate genuine ESC lines. Thus, we propose that these markers could be useful to predict the outcomes of establishing an ESC line in primates.

The H19-IGF2 imprinted gene region could be implicated in the risk of developing impaired renal function (IRF). Our aim was to determine the association of several common H19-IGF2 variants and IRF in a cohort of elderly healthy individuals. The study involved 675 individuals >65 years of age, 184 with type 2 diabetes mellitus (T2DM), and 105 with IRF (estimated glomerular filtration rate [eGFR] <60). They were genotyped for two common H19 single nucleotide polymorphisms (SNPs) (rs2839698 and rs10732516), one H19-IGF2 intergenic indel (rs201858505), and one indel in the 3′UTR of the IGF2. For the H19 SNPs, we also determined the allele present in the methylated chromosome through genotyping the DNA digested with a methylation-sensitive endonuclease. None of the four H19-IGF2 variants was associated with IRF in our cohort. We found a significantly higher frequency of the 3′UTR IGF2 deletion (D) in the eGFR <60 group ( p = 0.01; odds ratio = 1.16, 95% confidence interval = 1.10-2.51). This association was independent of age and T2DM, two strong predictors of IRF. In conclusion, a common indel variant in the 3′UTR of the IGF2 gene was associated with the risk of IRF. This association could be explained by the role of IGF2 in podocyte survival, through regulation of IGF2 expression by differential binding of miRNAs to the indel sequences. The H19/insulin-like growth factor 2 ( Igf2) gene plays vital roles during development, but their expression and methylation status have not been elucidated in male cloned animals. In this study, we investigated the methylation status of imprinting control region (ICR) and differentially methylated region (DMR) of the H19/Igf2 locus in the sperm and testes of naturally reproduced Saanen dairy goats (NG, control), transgenic cloned dairy goats (CG), and the first generation of mating the CGs with the female NGs (FG). Bisulfite sequencing polymerase chain reaction revealed that the H19 ICR was highly methylated in the sperm of all testing groups (97.07% ± 0.47%, 93.73% ± 0.85%, and 96.67% ± 0.54%). Notably, the methylation level of H19 ICR was decreased in the testes of CG goats compared with that of NG goats (73.83% ± 3.28% vs. 97.27% ± 0.71%, p < 0.05), while the Igf2 DMR2 was highly methylated in the sperm of these three groups (89.57% ± 1.74%, 87.23% ± 1.11%, and 83.2% ± 1.27%). However, the Igf2 DMR2 was abnormally hypomethylated in the testes of CG goats (27.33% ± 1.005%, p < 0.001), and moderately methylated in FG goats (53.1% ± 1.47%, p < 0.01) compared with that in NG goats (82.67% ± 0.70%). Furthermore, we found significantly decreased expression of H19 ( p < 0.01) and increased expression of Igf2 ( p < 0.01) in CG goats. Thus, we infer that somatic cell nuclear transfer has clear effect on genomic imprinting in the testes, but not sperm of cloned goats. [ABSTRACT FROM AUTHOR]

A subset of autosomal genes undergo genomic imprinting which results in expression from only the paternal or maternal chromosome. While this phenomenon is restricted to mammals and angiosperms, the underlying silencing mechanisms appear to be evolutionarily conserved. A biallelically unmethylated DNaseI hypersensitive region (A6-A4) between the imprinted Igf2 and H19 genes is conserved in humans and mice and functions as a tissue-specific maintenance element for the imprinted growth factor IGF2. In order to analyse A6-A4 for potentially conserved transcriptional maintenance properties, we have generated transgenic Drosophila harbouring the element in a reporter construct. These flies depicted silencing of the reporter genes lacZ and mini-white. The silenced state of the mini-white gene showed variegation and sensitivity to temperature changes. In addition, two members of the conserved Polycomb group, Enhancer of zeste and Posterior sex combs, were needed for repression. Polycomb group proteins are essential for gene silencing during development. Our results indicate that Polycomb group proteins may also be involved in the regulation of mammalian imprinted genes.
