1. Increase of Proliferative Activity of Leukaemic Blast Cells from Human Peripheral Blood in Liquid Culture

Abstract

The proliferative behaviour of leukaemic blast cells from the peripheral blood of 12 patients suffering from acute leukaemia was investigated in short term liquid culture. In 3 cases of AML an increase of the blast cell number was observed exceeding the initial value whereas in 9 other cases the cell number decreased more or less rapidly. In 9 of these patients the proliferation kinetics of the cultured leukaemic blast cells were studied with 3H-thymidine labelling. In all these cases the labelling index increased during the first days of culture, in 3 cases to values of 48 %, 45 % and 38 % on day 3. Only in 5 cases, however, did an absolute increase of the blast cells incorporating 3H-thymidine occur. Here the doubling times of the proliferating leukaemic blast cells were estimated to be 12, 13, 14, 28 and 55 h. Since a doubling time of 12 to 14 h seems to be too short to be explained only by an exponential growth of the initially proliferating cells it is postulated that leukaemic blast cells in a G0- or long G1-phase were present in the peripheral blood of these patients and that these entered the cell cycle during liquid culture.

Abstract

The liquid argon calorimeter of the H 1 detector presently taking data at the HERA ep collider at DESY, Hamburg, is described. The main physics requirements and the most salient design features relevant to this calorimeter are given. The aim to have smooth and hermetic calorimetric coverage over the polar angular range 4° ≤ θ ≤ 154° is achieved by a single liquid argon cryostat containing calorimeter stacks structured in wheels and octants for easy handling. The absorber materials used are lead in the electromagnetic part and stainless steel in the hadronic part. The read-out system is pipelined to reduce the dead time induced by the high trigger rate expected at the HERA collider where consecutive bunches are separated in time by 96 ns. The main elements of the calorimeter, such as the cryostat, with its associated cryogenics, the stack modules, the read-out, calibration and trigger electronics as well as the data acquisition system are described. Performance results from data taken in calibration runs with full size H 1 calorimeter stacks at a CERN test beam, as well as results from data collected with the complete H 1 detector using cosmic rays during the initial phase of ep operations are presented. The observed energy resolutions and linearities are well in agreement with the requirements.

Abstract

The high luminosity of PEP‐II in combination with the vertexing possibilities of the BABAR‐detector [1] offer unique opportunities on light meson spectroscopy. The basic interest in this domain is the search for exotic states. Many of the candidates for conventional mesons are under heavy discussion because of analysis ambiguities, mixing with conventional mesons and some are only seen in selected reactions. Many ambiguities can be resolved using a clean initial state which restricts the final state to specific quantum numbers, such as weak decays of Ds±‐mesons into three pseudoscalars, allowing only a few resonances to occur. The selections and the results of the Dalitz plot analyses of the decays Ds± → Ks0Ks0π± and Ds± → π+π−π± are presented. These decays are of particular interest for the discussion of the existence and the spin of isoscalar resonances above 1.5 GeV/c2. In this mass region the glueball ground state is conjectured to lie and might mix strongly with other isoscalar JPC = 0++‐states.

Abstract

Technical aspects of the three major components of the H1 detector at the electron-proton storage ring HERA are described. This paper covers the detector status up to the end of 1994 when a major upgrading of some of its elements was undertaken. A description of the other elements of the detector and some performance figures from luminosity runs at HERA during 1993 and 1994 are given in a paper previously published in this journal.

Abstract

General aspects of the H1 detector at the electron-proton storage ring HERA as well as technical descriptions of the magnet, luminosity system, trigger, slow-control, data acquisition and off-line data handling are given. The three major components of the detector, the tracking, calorimeter and muon detectors, will be described in a forthcoming article. The present paper describes the detector that was used from 1992 to the end of 1994. After this a major upgrade of some components was undertaken. Some performance figures from luminosity runs at HERA during 1993 and 1994 are given.

Abstract

From 17 patients with different forms of acute leukaemia, mononuclear blood cells were cultured in diffusion chambers (DC) implanted intraperitoneally into pre-irradiated mice. In 14 patients, growth of blast cells could be observed during the culture period of up to 21 days. To question whether this growth of blast cells was due only to proliferation of the initially proliferating fraction or whether a re-entry of resting leukaemic cells into proliferation was involved, various 3H-thymidine (3H-TdR) labelling studies were carried out. The absolute increase of blast cells in EC showed no correlation with the fraction of leukaemic blast cells in DNA-synthesis in the implanted cell suspension as measured by 3H-TdR labelling in vitro. Furthermore, in 2 patients where the kinetic behaviour of initially labelled leukaemic blast cells was followed during DC culture, the increase in total blast cells could only be attributed to a small extent to proliferation of those cells initially in the cell cycle. Lastly, "in vivo" labelling during the culture period showed that in one case 25% and in another case 60% of the blast cells in DC were proliferating. The conclusion is that, owing to the stimulation of the diffusion chamber milieu and possibly also due to removal of an in vivo inhibition, in most cases of acute leukaemia resting leukaemic blast cells can apparently re-enter the active cell cycle. This has relevance for an understanding of the self-maintenance of the leukaemic cell population and may also be a reason for relapse of leukaemia after the usual cytostatic drug treatment which affects mainly the proliferating leukaemic cells.

Peripheral blood cells of 21 patients with different forms of acute leukemia were cultured in diffusion chambers (5 x 10(5) cells/chamber) implanted intraperitoneally in 650 R preirradiated host mice over a period of up to 21 days. In patients with acute myeloid leukemia (AML), acute erythroleukemia (AEL), or acute myelomonocytic leukemia (AMMoL), the total number of cells which developed during this culture period exceeded the implanted value and also the values for normal peripheral blood cells from ten controls. In acute undifferentiated leukemia (AUL), two out of six patients showed considerable growth whereas the others, and also two patients with acute lymphoid leukemia (ALL), had poor growth. Differential counts revealed that the rise in total cells was due mainly to proliferation of blast cells and formation of granulopoietic cells. The latter exceeded the numbers from normal peripheral blood cells in 9 out of 13 patients with AML, AEL, or AMMoL and in 2 out of 6 patients with ALL. The production of granulopoiesis was not restricted to proliferating cells, but included mature cells which were of abnormal morphology in some cases. From the amount of granulopoiesis and the time of its development it was assumed that they were at least partly derived from leukemic blast cells. Chromosome analyses to decide whether the granulopoietic cells were of leukemic or normal cell origin are in progress.

Growth of mononuclear cells from human peripheral blood from 10 normal individuals was tested in diffusion chamber culture over a period up to 17 or 21 days. After an initial decrease during the first few days an increase of the total cell number was observed with maximal values on day 13. In all individuals growth of undifferentiated blast cells, lymphocytes, plasma cells, immature and mature granulopoietic cells, macrophages, and megakaryocytes occurred. In all individuals the different cell types had similar growth patterns in diffusion chamber culture. The considerable numerical variations which were seen in the granulopoietic cells were probably due to different stem cell concentrations in the peripheral blood of the investigated individuals. The results indicate that the diffusion chamber technique is a valuable method for the detection of haemopoietic stem cells and the culture of lymphocytic cells in man.

Abstract

The diffusion chamber (DC) system of in vivo culture is useful for testing human leukemic peripheral blood cells for several reasons. With this technique, stem cells apparently preceding the granulopoietic progenitor cells (CFU-c) can be detected. Arguments for this come from diffusion chamber studies on normal human peripheral blood cells where an absolute increase in CFU-c was found (Hoelzer et al., 1976a) and also from DC culture of human bone marrow where CFU-d (cells that form granulocytic colonies in DC) as precursors of CFU-c could be distinguished on the basis of different velocity sedimentation profiles and different 3H-thymidine suicidal fractions (Jacobsen et al., 1978). Differentiation of cells in the DC system occurs not only into the granulopoietic/macrophage series but also into megakaryocytes and, even if rarely, into erythropoiesis, as well as into lymphatic cell lines. Furthermore, the cultures can be maintained over long periods; in our present experiments leukemic cells have been cultured for up to 2 months and Hodgkin cells for even longer (Boecker et al., 1975). Thus the DC system seems to be suitable for sustaining the growth of leukemic cells of an earlier “stemness” than normal CFU-c and seems also to promote differentiation into haemopoietic cell lines other than granulopoiesis.

18. Synthesis of Lewis Acid-Base Adducts Between Potassium Dihydrogenpnictogenides and Group 13 Lewis Acids.

Abstract

Lewis acid-base adducts between potassium dihydrogenpnictogenides [K(18c6)PnH 2 ] (Pn=P, As) and Trialkylgallanes (GaMe 3 , GaEt 3 , GatBu 3 ) as well as BH 3 were synthesized. In the course of these investigations, we were able to fully characterize a row of compounds of the type [K(18c6)(thf) 0-2 ][PnH 2 (ER 3 ) 2 ] (E=B: R=H; E=Ga: R=Me, Et, tBu). Based on the successful syntheses of these species, we also synthesized several compounds of the composition [K(18c6)(thf) 0-2 ][Pn(SiMe 3 ) 2 (ER 3 ) n ] (n=1, 2) starting from [KPn(SiMe 3 ) 2 ] (Pn=P and As) and ER 3 (E=B, Al, In: R=Me; E=Ga: R=Me, Et, tBu). All isolated compounds, were examined by NMR spectroscopy, infra-red spectroscopy and elemental analysis. A selection of the compounds presented were also examined by means of X-ray structure analysis on the single crystal. The obtained adducts of negatively charged [PnH 2 ] - and [Pn(SiMe 3 ) 2 ] - fragments with Lewis acids are a class of compounds that has scarcely been investigated to date., (© 2024 The Author(s). From 17 patients with different forms of acute leukaemia, mononuclear blood cells were cultured in diffusion chambers (DC) implanted intraperitoneally into pre-irradiated mice. In 14 patients, growth of blast cells could be observed during the culture period of up to 21 days. To question whether this growth of blast cells was due only to proliferation of the initially proliferating fraction or whether a re-entry of resting leukaemic cells into proliferation was involved, various 3H-thymidine (3H-TdR) labelling studies were carried out.
