1. Marker-Assisted Breeding of Improved Maternal Haploid Inducers in Maize for the Tropical/Subtropical Regions

Abstract

For efficient production of doubled haploid (DH) lines in maize, maternal haploid inducer lines with high haploid induction rate (HIR) and good adaptation to the target environments is an important requirement. In this study, we present second-generation Tropically Adapted Inducer Lines (2GTAILs), developed using marker assisted selection (MAS) for qhir1, a QTL with a significant positive effect on HIR from the crosses between elite tropical maize inbreds and first generation Tropically Adapted Inducers Lines (TAILs). Evaluation of 2GTAILs for HIR and agronomic performance in the tropical and subtropical environments indicated superior performance of 2GTAILs over the TAILs for both HIR and agronomic performance, including plant vigor, delayed flowering, grain yield, and resistance to ear rots. One of the new inducers 2GTAIL006 showed an average HIR of 13.1% which is 48.9% higher than the average HIR of the TAILs. Several other 2GTAILs also showed higher HIR compared to the TAILs. While employing MAS for qhir1 QTL, we observed significant influence of the non-inducer parent on the positive effect of qhir1 QTL on HIR. The non-inducer parents that resulted in highest mean HIR in the early generation qhir1+ families also gave rise to highest numbers of candidate inducers, some of which showed transgressive segregation for HIR. The mean HIR of early generation qhir1+ families involving different non-inducer parents can potentially indicate recipient non-inducer parents that can result in progenies with high HIR. Our study also indicated that the HIR associated traits (endosperm abortion rate, embryo abortion rate, and proportion of haploid plants among the inducer plants) can be used to differentiate inducers vs. non-inducers but are not suitable for differentiating inducers with varying levels of haploid induction rates. We propose here an efficient methodology for developing haploid inducer lines combining MAS for qhir1 with HIR associated traits.

Abstract

More than 30 years i n formation about the fatty acid composit ion of wi ld plant seeds was collected f rom the appropriate pharmaceutical, botanical and chemical literature by the former Institute for Chemistry and Physics of Lipids of the Federal Center for Cereal, Potato and Lipid Research in Münster. Since it was very difficult to search this unique source of information, the collection was transferred into an electronically searchable database. This plan was supported by financial assistance of the German Ministry of Consumer Protection, Food and Agriculture. Since some month the database is available in the internet (http:\\www.bagkf.de/sofa) free of charge. Up to now more than 18.000 different tables with about 110.000 individual data are recallable from the database, by different types of search forms. An overview about the application of the different search forms as well as some examples for searches is given in this publication.

Abstract

Orange‐fleshed sweetpotato (OFSP) [Ipomoea batatas (L.) Lam] often lacks high dry matter (DM) in elite breeding material. The objectives were to examine the potential to combine yield with high root DM and high root β‐carotene (BC) content by variance‐covariance analysis and optimization of breeding scenarios for "OFSP dry and starchy," which was exemplarily shown for the humid tropics of Peru. The trial series across seasons suggested a good chance to combine yield and quality (high DM and high BC). The unique data were used to estimate variance components and correlations among four yield traits and four quality traits. For the optimization of selections in our breeding program, we used the selection response by Cochran's approach and developed a small R script. Responses to selection were predicted for storage root yield and a modified Elston index aggregating eight traits. The differences between two‐ and three‐stage selection scenarios were small. For storage root yield, the recommended optimized two‐stage selection increased the response relative to the standard (one‐stage selection 100%) to 141.6% for 2268 plots and to 189.1% for 540 plots; the corresponding optimized three‐stage selection increased the response to 144.3% for 2268 plots and to 190.5% for 540 plots. A two‐stage selection is recommended with two to three locations at the first stage and 6–12 locations at the second stage, according to low‐ or high‐test capacity, respectively. Optimization of breeding schemes is much more important for smaller breeding programs than those with larger test capacity, and this allows testing even larger numbers of genotypes in a small breeding program. Abstract

Additive Manufacturing (AM) of ceramics is a constantly emerging field of interest both in research and in industry. Binder jetting-based AM of ceramics in particular offers the opportunity to produce large ceramic parts with a high wall thickness at a high throughput. One limitation is that it requires flowable powders, which are generally coarse and thus exhibit only limited sintering activity. The resulting low sintered densities impede the commercial binder jetting-based production of dense oxide ceramics. We present an approach to efficiently increase the green density of binder jetted alumina parts by optimized slurry infiltration, which also leads to a significant increase in the sintered density. In a first step, alumina parts were fabricated via binder jetting, using a 20-µm-sized alumina powder, yielding relative green densities of about 47–49%. Initial sintering studies with powder compacts showed that sintering even above 1900 °C is not sufficient to achieve acceptable densification. Therefore, green samples were infiltrated with a highly filled ceramic slurry to fill the remaining pores (about 2–5 µm in size) with smaller particles and thus increase the packing density. Particle volume content (40–50 vol%), particle size (100–180 nm) and the infiltration procedure were adapted for tests on cuboid samples to achieve a high penetration of the green bodies and a high degree of pore filling. In this way, the relative green density could be increased starting from about 47% after binder jetting, to 73.4% after infiltration and drying. After sintering at 1675 °C densities above 90% could be achieved, yielding three-point bending strengths up to 145 MPa. As a conclusion, this approach can be regarded as a promising route for overcoming the drawbacks of the binder jetting process on the way to denser, mechanically more stable sintered alumina parts.

Abstract

Key message A breeding strategy combining genomic with one-stage phenotypic selection maximizes annual selection gain for net merit. Choice of the selection index strongly affects the selection gain expected in individual traits. Abstract Selection indices using genomic information have been proposed in crop-specific scenarios. Routine use of genomic selection (GS) for simultaneous improvement of multiple traits requires information about the impact of the available economic and logistic resources and genetic properties (variances, trait correlations, and prediction accuracies) of the breeding population on the expected selection gain. We extended the R package “selectiongain” from single trait to index selection to optimize and compare breeding strategies for simultaneous improvement of two traits. We focused on the expected annual selection gain (ΔGa) for traits differing in their genetic correlation, economic weights, variance components, and prediction accuracies of GS. For all scenarios considered, breeding strategy GSrapid (one-stage GS followed by one-stage phenotypic selection) achieved higher ΔGa than classical two-stage phenotypic selection, regardless of the index chosen to combine the two traits and the prediction accuracy of GS. The Smith–Hazel or base index delivered higher ΔGa for net merit and individual traits compared to selection by independent culling levels, whereas the restricted index led to lower ΔGa in net merit and divergent results for selection gain of individual traits. The differences among the indices depended strongly on the correlation of traits, their variance components, and economic weights, underpinning the importance of choosing the selection indices according to the goal of the breeding program. We demonstrate our theoretical derivations and extensions of the R package “selectiongain” with an example from hybrid wheat by designing indices to simultaneously improve grain yield and grain protein content or sedimentation volume.

14. Hydrograph Shape Impact on Sand Infiltration and Sediment Transport Dynamics in Gravel‐Bed Rivers

Abstract

Sand (i.e., < 2 mm) infiltration into a gravel bed is recognized to affect sediment dynamics of coarser fractions (i.e., entrainment and transport rate), near-bed velocities, bed morphology, and to pose significant engineering and ecological issues. Local hydraulics, sediment dynamics, and sand infiltration into the gravel matrix are related and mutually affect each other in gravel-bed rivers. However, relatively little research has been done to explore sediment dynamics and sand infiltration in a mobile gravel-bed under unsteady flow conditions. In the present study, we experimentally simulated three hydrographs of different durations of rising and falling limbs, and monitored sand infiltration over beds with a coarse and a fine grain size distribution. Results confirmed that the addition of sand enhances sediment dynamics. Moreover, the shape of the rising limb of the hydrograph was responsible for altering identified sand infiltration processes and bedload rate. Short time-to-peak floods (i.e., short time from the start to the peak of the hydrograph) experienced higher sediment transport rates during the falling limb than the rising limb of the hydrograph, favoring sand infiltration. In contrast, long time-to-peak floods (i.e., long time from the start to the peak of the hydrograph) experienced higher sediment transport rates during the rising limb than the falling limb of the hydrograph, resulting in reduced sand infiltration.

Abstract

Introduction Simulation based learning (SBL) has been used in diagnostic radiography for teaching and learning purposes. However, the method of offering the SBL opportunities has not been placed in the context of whether the experiences provide the student with a real life simulation. Moreover, in the light of the current staff shortages and healthcare issues in South Africa, placing students into the clinical practice without sound grounding in the application of theory into practice would be unfair to the student and patients. Thus, SBL could provide the opportunity for the student to learn in a safe teaching and learning environment. Methods A qualitative, meta- synthesis was undertaken using systematic literature searches. The existing literature in English was retrieved from databases (Medline, CINAHL and ScienceDirect). The keywords used were simulation based learning, simulation, radiography and diagnostic radiography. Results Themes and categories were developed from the literature. Theme 1: An authentic and realistic situation, which is relevant to the development of a professional in the context of the profession. Theme 2: Building confidence in a safe, reliable and nurturing environment. Theme 3: Active participation in a collaborative process. Conclusion The meta synthesis revealed three major themes that can be used as a framework to motivate for the use of SBL in a diagnostic radiography programme. Implications for practice The implementation of SBL that could prepare students for the workplace. SBL focused on specific competencies for clinical readiness.

Abstract

Introduction The focus on paediatric radiation dose reduction supports reevaluation of paediatric imaging protocols. This is particularly important in the neonates where chest radiographs are frequently requested to assess respiratory illness and line placement. This study aims to assess the impact of neonatal chest radiographic protocols on patient dose in four hospitals in different countries. Methods Exposure parameters, collimation, focus to skin distance (FSD) and radiation dose from 200 neonatal chest radiographs were registered prospectively. Inclusion criteria consisted of both premature and full-term neonates weighing between 1000 and 5000 g. Only data from the examinations meeting diagnostic criteria and approved for the clinical use were included. Radiation dose was assessed using dose area product (DAP). Results The lowest DAP value (4.58 mGy cm2) was recorded in the Norwegian hospital, employing a high kVp, low mAs protocol using a DR system. The Canadian hospital recorded the highest DAP (9.48), using lower kVp and higher mAs with a CR system, including the addition of a lateral projection. The difference in the mean DAP, weight, field of view (FOV) and kVp between the hospitals is statistically significant (p Conclusion Use of non-standardised imaging protocols in neonatal chest radiography results in differences in patient dose across hospitals included in the study. Using higher kVp, lower mAs and reducing the number of lateral projections to clinically relevant indications result in a lower DAP measured in the infant sample studied. Further studies to examine image quality based on exposure factors and added filtration are recommended. Implications for practice Reevaluation of paediatric imaging protocols presents an opportunity to reduce patient dose in a population with increased sensitivity to ionising radiation.

Abstract

Satellite-based broad-scale (i.e., global and continental) human settlement data are essential for diverse applications spanning climate hazard mitigation, sustainable development monitoring, spatial epidemiology and demographic modeling. Many human settlement products report exceptional detection accuracies above 85%, but there is a substantial blind spot in that product validation typically focuses on large urban areas and excludes rural, small-scale settlements that are home to 3.4 billion people around the world. In this study, we make use of a data-rich sample of 30 refugee settlements in Uganda to assess the small-scale settlement detection by four human settlement products, namely, Geo-Referenced Infrastructure and Demographic Data for Development settlement extent data (GRID3-SE), Global Human Settlements Built-Up Sentinel-2 (GHS-BUILT-S2), High Resolution Settlement Layer (HRSL) and World Settlement Footprint (WSF). We measured each product’s areal coverage within refugee settlement boundaries, assessed detection of 317,416 building footprints and examined spatial agreement among products. For settlements established before 2016, products had low median probability of detection and F1-score of 0.26 and 0.24, respectively, a high median false alarm rate of 0.59 and tended to only agree in regions with the highest building density. Individually, GRID3-SE offered more than five-fold the coverage of other products, GHS-BUILT-S2 underestimated the building footprint area by a median 50% and HRSL slightly underestimated the footprint area by a median 7%, while WSF entirely overlooked 8 of the 30 study refugee settlements. The variable rates of coverage and detection partly result from GRID3-SE and HRSL being based on much higher resolution imagery, compared to GHS-BUILT-S2 and WSF. Earlier established settlements were generally better detected than recently established settlements, showing that the timing of satellite image acquisition with respect to refugee settlement establishment also influenced detection results. Nonetheless, settlements established in the 1960s and 1980s were inconsistently detected by settlement products. These findings show that human settlement products have far to go in capturing small-scale refugee settlements and would benefit from incorporating refugee settlements in training and validating human settlement detection approaches.

Abstract

Selected doubled haploid lines averaged similar testcross performance as their original landraces, and the best of them approached the yields of elite inbreds, demonstrating their potential to broaden the narrow genetic diversity of the flint germplasm pool. Maize landraces represent a rich source of genetic diversity that remains largely idle because the high genetic load and performance gap to elite germplasm hamper their use in modern breeding programs. Production of doubled haploid (DH) lines can mitigate problems associated with the use of landraces in pre-breeding. Our objective was to assess in comparison with modern materials the testcross performance (TP) of the best 89 out of 389 DH lines developed from six landraces and evaluated in previous studies for line per se performance (LP). TP with a dent tester was evaluated for the six original landraces, ~ 15 DH lines from each landrace selected for LP, and six elite flint inbreds together with nine commercial hybrids for grain and silage traits. Mean TP of the DH lines rarely differed significantly from TP of their corresponding landrace, which averaged in comparison with the mean TP of the elite flint inbreds ~ 20% lower grain yield and ~ 10% lower dry matter and methane yield. Trait correlations of DH lines closely agreed with the literature; correlation of TP with LP was zero for grain yield, underpinning the need to evaluate TP in addition to LP. For all traits, we observed substantial variation for TP among the DH lines and the best showed similar TP yields as the elite inbreds. Our results demonstrate the high potential of landraces for broadening the narrow genetic base of the flint heterotic pool and the usefulness of the DH technology for exploiting idle genetic resources from gene banks.

Abstract

Epistasis seems to play a significant role in the manifestation of heterosis. However, the power of detecting epistatic interactions among quantitative trait loci (QTL) in segregating populations is low. We studied heterosis in Arabidopsis thaliana hybrid C24 x Col-0 by testing near-isogenic lines (NILs) and their triple testcross (TTC) progenies. Our objectives were to (i) provide the theoretical basis for estimating different types of genetic effects with this experimental design, (ii) determine the extent of heterosis for seven growth-related traits, (iii) map the underlying QTL, and (iv) determine their gene action. Two substitution libraries, each consisting of 28 NILs and covering ~61 and 39% of the Arabidopsis genome, were assayed by 110 single-nucleotide polymorphism (SNP) markers. With our novel generation means approach 38 QTL were detected, many of which confirmed heterotic QTL detected previously in the same cross with TTC progenies of recombinant inbred lines. Furthermore, many of the QTL were common for different traits and in common with the 58 QTL detected by a method that compares triplets consisting of a NIL, its recurrent parent, and their [F.sub.1] cross. While the latter approach revealed mostly (75%) overdominant QTL, the former approach allowed separation of dominance and epistasis by analyzing all materials simultaneously and yielded substantial positive additive x additive effects besides directional dominance. Positive epistatic effects reduced heterosis for growth-related traits in our materials.

Arabidopsis thaliana has emerged as a leading model species in plant genetics and functional genomics including research on the genetic causes of heterosis. We applied a triple testcross (TTC) design and a novel biometrical approach to identify and characterize quantitative trait loci (QTL) for heterosis of five biomass-related traits by (i) estimating the number, genomic positions, and genetic effects of heterotic QTL, (ii) characterizing their mode of gene action, and (iii) testing for presence of epistatic effects by a genomewide scan and marker x marker interactions. In total, 234 recombinant inbred lines (RILs) of Arabidopsis hybrid C24 x Col-0 were crossed to both parental lines and their [F.sub.1] and analyzed with 110 single-nucleotide polymorphism (SNP) markers. QTL analyses were conducted using linear transformations [Z.sub.1], [Z.sub.2], and [Z.sub.3] calculated from the adjusted entry means of TTC progenies. With [Z.sub.1], we detected 12 QTL displaying augmented additive effects. With [Z.sub.2], we mapped six QTL for augmented dominance effects. A one-dimensional genome scan with [Z.sub.3] revealed two genomic regions with significantly negative dominance x additive epistatic effects. Two-way analyses of variance between marker pairs revealed nine digenic epistatic interactions: six reflecting dominance x dominance effects with variable sign and three reflecting additive x additive effects with positive sign. We conclude that heterosis for biomass-related traits in Arabidopsis has a polygenic basis with overdominance and/or epistasis being presumably the main types of gene action.

Epistatic interactions among quantitative trait loci (QTL) contribute substantially to the variation in complex traits. The main objectives of this study were to (i) compare three- vs. four-step genome scans to identify three-way epistatic interactions among QTL belonging to a metabolic pathway, (ii) investigate by computer simulations the power and proportion of false positives (PFP) for detecting three-way interactions among QTL in recombinant inbred line (RIL) populations derived from a nested mating design, and (iii) compare these estimates to those obtained for detecting three-way interactions among QTL in RIL populations derived from diallel and different partial diallel mating designs. The single-nucleotide polymorphism haplotype data of B73 and 25 diverse maize inbreds were used to simulate the production of various RIL populations. Compared to the three-step genome scan, the power to detect three-way interactions was higher with the four-step genome scan. Higher power to detect three-way interactions was observed for RILs derived from optimally allocated distance-based designs than from nested designs or diallel designs. The power and PFP to detect three-way interactions using a nested design with 5000 RILs were for both the 4-QTL and the 12-QTL scenario of a magnitude that seems promising for their identification.

Primary causes of heterosis are still unknown. Our goal was to investigate the extent and underlying genetic causes of heterosis for five biomass-related traits in Arabidopsis thaliana. We (i) investigated the relative contribution of dominance and epistatic effects to heterosis in the hybrid C24 x Co1-0 by generation means analysis and estimates of variance components based on a triple testcross (TTC) design with recombinant inbred lines (RILs), (ii) estimated the average degree of dominance, and (iii) examined the importance of reciprocal and maternal effects in this cross. In total, 234 RILs were crossed to parental lines and their [F.sub.1]'s. Midparent heterosis (MPH) was high for rosette diameter at 22 days after sowing (DAS) and 29 DAS, growth rate (GR), and biomass yield (BY). Using the [F.sub.2]-metric, directional dominance prevailed for the majority of traits studied but reciprocal and maternal effects were not significant. Additive and dominance variances were significant for all traits. Additive x additive and dominance x dominance variances were significant for all traits but GR. We conclude that dominance as well as digenic and possibly higher-order epistatic effects play an important role in heterosis for biomass-related traits. Our results encourage the use of Arabidopsis hybrid C24 X Co1-0 for identification and description of quantitative trait loci (QTL) for heterosis for biomass-related traits and further genomic studies.

Abstract

Fused deposition modeling (FDM) is limited by the availability of application specific functional materials. Here we illustrate printing of non-conventional polymer nanocomposites (CNT- and graphene-based polybutylene terephthalate (PBT)) on a commercially available desktop 3D printer leading toward printing of electrically conductive structures. The printability, electrical conductivity and mechanical stability of the polymer nanocomposites before and after 3D printing was evaluated. The results show that 3D printed PBT/CNT objects have better conductive and mechanical properties and a better performance than 3D printed PBT/graphene structures. In addition to that, printing more than one material (multi-materials) and challenges in using abrasive conductive fillers (i.e., CNT and graphene) are also discussed. Overall this study demonstrates that a commercially available desktop 3D printer can be used to fabricate low-cost functional objects.

Favorable epistatic gene complexes may be important for hybrid performance of maize (Zea mays L.). This study was conducted to assess the importance of epistasis in per se and testcross performance for grain yield and grain moisture in four crosses among four elite European flint maize lines by generation means analyses as well as genome-wide tests for significant digenic epistatic effects between marker loci. For each cross, six generations (P1, P2, [F.sub.1], [F.sub.2], BC1, BC2) and testcrosses of these generations plus the [F.sub.2]-Syn1, [F.sub.2]-Syn2, and Fz-Syn3 generations in combination with an unrelated dent tester were evaluated in four environments. Testcross generation means of [bar.P], [bar.BC], [F.sub.1], [F.sub.2], [F.sub.2]-Syn1, [F.sub.2]-Syn2, and [F.sub.2]-Syn3 did not significantly differ from each other for grain yield and grain moisture, indicating that epistasis between unlinked and moderately linked loci was negligible in its net effect. Depending on the cross, QTL mapping for per se and testcross performance with the dent tester was conducted with 71 to 344 lines ([F.sub.3] to [F.sub.6]) grown in four environments. In genome-wide two-way ANOVAs, significant epistatic interactions were found with only a few marker pairs that did not improve the fit of the model after including main-effect QTLs previously detected by composite interval mapping. Poor correspondence of the results from per se and testcross analyses reflects dominance and epistatic interactions between parental and tester alleles. Our results suggest that epistasis is of minor importance for both traits with regard to the optimum type of population ([F.sub.2] vs. BC) in recycling breeding of elite maize inbreds. The magnitude of the genotypic correlation between line per se performance (LP) and testcross performance (TP) is crucial for optimum testing schemes in hybrid breeding as well as simultaneous improvement of commercial hybrids and their inbred parents. The objectives of this study were to (i) obtain estimates of the correlation, and (ii) determine quantitative trait loci (QTL) correspondence between LP and TP within four populations ([F.sub.3] to [F.sub.6], lines) derived from intrapool crosses of European flint maize (Zea mays L.). The number of lines evaluated for both LP and TP ranged from 65 to 280 across the four populations. The LP and TP with a dent inbred tester were evaluated for grain yield, grain moisture, kernel weight, protein concentration, and plant height in four to five environments. Composite interval mapping (CIM) using a joint restriction fragment length polymorphism (RFLP) linkage map of all populations was conducted separately for LP and TP in each population, with sample sizes ranging from 71 to 344. Genotypic correlations between LP and TP, [r.sub.g] (LP, TP), were low to intermediate for grain yield (0.28-0.56) across populations and intermediate to high for the other traits (0.52-0.87). The magnitude of [r.sub.g] (LP, TP) across populations for grain yield was neither associated with the ratio between the genotypic variances for LP and TP nor with the evidence for dominance in LP or epistasis in LP or TP. Genotypic correlations between observed TP and its prediction based on QTL positions and effects for LP were smaller than corresponding values of [r.sub.g] (LP, TP) for all traits. Except for grain yield, more than half of the QTL were in common to LP and TP in the largest population A x [B.sup.I]. From simulation studies it is known that the allocation of experimental resources has a crucial effect on power of QTL detection as well as on accuracy and precision of QTL estimates. In this study we used a rely large experimental data set composed of 976 [F.sub.5], maize testcross progenies evaluated in 19 environments and cross-validation to assess the effect of sample size (N), number of test environments (E), and significance threshold on the number of detected QTL, the proportion of the genotypic variance (E), and by them, and the corresponding bias of estimates for grain yield, grain moisture, and plant height. In addition, we used computer simulations to compare the usefulness of two cross-validation schemes for obtaining unbiased estimates of QTL effects. The maximum, validated genotypic variance explained by QTL in this study was 52.3% for grain moisture despite the large number of detected QTL, thus confirming the infinitesimal model of quantitative genetics. In both simulated and experimental data, the effect of sample size on power of QTL detection as well as on accuracy, and precision of QTL estimates was large. The number of detected QTL and the proportion of genotypic variance explained by QTL generally increased more with increasing N than with increasing E. The average bias of QTL estimates and its range were reduced by increasing N and E. Cross-validation performed well with respect to yielding asymptotically unbiased estimates of the genotypic variance explained by QTL. On the basis of our findings, recommendations for planning of QTL mapping experiments and allocation of experimental resources are given

46. Congruency of quantitative trait loci detected for agronomic traits in testcrosses of five populations of European maize

Congruency of estimated positions and effects of QTL in different samples of the same cross or different crosses is an indicator of the reliability of these estimates and their usefulness in marker-assisted selection (MAS). We investigated the influence of the sample and genetic background on QTL congruency among five populations of European maize (Zea mays L.). Three samples derived from the same cross comprised 344 (A x BI) and 109 (A x [B.sup.II]) [F.sub.2:3] as well as 71 [F.sub.4:5] (A x [B.sup.III]) lines. Two other crosses comprised 109 (A x C) and 84 (C x D) [F.sub.3:4] lines. All lines were topcrossed to the same inbred tester and evaluated in four or five environments. A combined linkage map of RFLP marker data from all five populations was used in composite interval mapping (CIM). The total number of QTL identified for five agronomically important traits was 42 in A x [B.sup.I], 18 in A x [B.sup.II], 20 in A x [B.sup.III], 28 in A x C, and 23 in C x D. Averaged across traits, the proportion p of the genetic variance explained by these QTL varied between 50.4% in the largest population A x Bt and 30.7% in a population of considerably smaller size (A x [B.sup.II]). Cross validation (CV) yielded substantially lower estimates of p. Between 10 and 24% of the 42 QTL from A x [B.sup.I] were also detected within a 20-cM interval in the other four populations. Incongruent QTL among A x B samples were due to the low power of QTL detection and the large bias in QTL estimates. The genetic correlations between predicted (based on QTL positions from one population) and observed phenotypic values in another population were highest among A x B samples with a maximum of 0.68 for plant height. Congruency of QTL was found for kernel weight, protein concentration, and plant height and was mainly attributable to one or few QTL of moderate to large size. Abstract

Background: Acute ischemic stroke (AIS) patients risk grave disability when treatment is delayed. The benchmark for door to needle (DTN) time for thrombolysis in AIS is less than 60 minutes; The American Stroke Association's (ASA) Target Stroke, Phase II (TSP II) sets a higher standard of less than 45 minutes. Emergency department caregivers at a 377 bed community hospital with primary stroke center designation were educated about the TSP II toolbox; however DTN times in 2016 were less than 60 minutes only 29% of the time with a mean time of 78 minutes, a medium time of 77 minutes and a range of 178 minutes. Purpose: Using a lean methodology approach we organized team debriefings to evaluate cases of time delay with the aim to develop strategies to improve outcome. Methods: As a process improvement project, an interprofessinal team was assembled to debrief and evaluate all patients receiving thrombolytic therapy. A comprehensive checklist was developed to include all of the critical components of care along with target benchmark timelines. Each data point was scrutinized to evaluate for possible time delays. The caregiver team was encouraged to objectively identify time delays and look for themes. Through root cause analysis, the most common theme identified was the performance of unnecessary tasks that could wait until after initiation of therapy. The other theme identified was delay in critical interventions such as timely lowering of blood pressure to prevent hemorrhage in patients undergoing thrombolysis. We engaged in coaching conversations among members of the team in an effort to promote better critical thinking and better patient outcomes. Results: From January, 2017 to August, 2018 there were 69 thrombolytic therapy cases The DTN rate of less than 60 minutes was 78%. Mean, median and mode times were 53,51 and 57 minutes respectively. The range was from 28-93 minutes. DTN in 45 minutes or less occurred at a rate of 30%. Conclusion: Improved outcome for exemplary professional practices can be afforded through a systematic approach to debrief about time sensitive treatments. Bringing team members together for a guided discussion designed to look for themes that lead to performance deficits will develop better patient outcomes.
