Purpose The purpose of this study was to determine whether net metabolic cost of walking is affected by age per se. Methods We selected 10 healthy, active older adults (mean age 75 years) and 10 young adults (mean age 26 years), and determined their preferred overground walking speed. On the same day, in a morning and afternoon session, we had them walk at that speed overground and on a treadmill while we measured oxygen consumption rate. From the latter we subtracted the rate in sitting and calculated net metabolic cost. Results Anthropometrics were not different between the groups nor was preferred walking speed (1.27 m s−1 both groups). There was no difference in net metabolic cost of overground walking between older and young adults (e.g., in the morning 2.64 and 2.56 J kg−1 m−1, respectively, p > 0.05). In the morning session, net metabolic cost of walking was higher on the treadmill than overground in our older adults by 0.6 J kg−1 m−1 (p Conclusion First, there is no effect of age per se on metabolic cost of overground walking. Second, older adults tend to have higher metabolic cost of walking on a treadmill than walking overground at preferred speed, and adaptation may take a long time. The commonly reported age-related elevation of metabolic cost of walking may be due to confounding factors causing preferred walking speed to be lower in older adults, and/or due to older adults reacting differently to treadmill walking than young adults.

An ecosystem services (ES) approach to chemical risk assessment has many potential advantages, but there are also substantial challenges regarding its implementation. We report the findings of a multi-stakeholder workshop that evaluated the feasibility of adopting an ES approach to chemical risk assessment using currently available tools and data. Also evaluated is the added value such an approach would bring to environmental decision making. The aim was to build consensus across disparate stakeholders and to co-produce a common understanding of the regulatory benefits and feasibility of implementing an ES approach in European chemicals regulation. Workshop discussions were informed by proof of concept studies and resulted in the development of a novel tiered framework for assessing chemical risk to ES delivery. There was consensus on the substantial added value of adopting an ES-based approach for regulatory decision making. Ecosystem services provide a common currency and a ‘unifying approach’ across environmental compartments, stressors and regulatory frameworks. The ES approach informs prioritisation of risk and remedial action and aids risk communication and risk management. It facilitates a more holistic assessment, enables ES trade-offs to be compared across alternative interventions, and supports comparative risk assessments and a socio-economic analysis of management options and decisions. Key to realising this added value is a shift away from using a single threshold value to categorise risk, towards a consideration of the exposure-effect distribution for individual ES of interest. Also required is the development of an integrated systems-level approach across regulatory frameworks and agreement on specific protection goals and scenarios for framing environmental risk assessments. The need to further develop tools for extrapolating toxicity data to service providers and ES delivery, including logic chains and ecological production functions, was highlighted. Also agreed was the need for methods and metrics for ES valuation to be used in assessing trade-offs.

Aquatic ecosystems contribute to human well-being by delivering ecosystem services, but their protection has been given low priority in Africa. Lake Ziway in the Ethiopian Rift Valley basin provides services including irrigation, drinking water and fish food in the region. This paper reviews the biological resources and spatio- temporal variation of water quality of the lake focussing on nutrients, metals and pesticides. Lake Ziway is under increasing agricultural and urban pressure and is exhibiting deteriorating trends in several water quality and ecological parameters. Nutrients and trace metals, including PO43−, NO3−, NH4+, Ca2+, Cu and Ni of the lake have shown increasing temporal trends in concentration. Spatially, higher values of major parameters (e.g. NO3−, NH4+, K, Na and electrical conductivity) were observed at shoreline sites near floriculture farming. The water quality of the lake exceeded guideline values for drinking water (alkalinity and Fe) and for aquatic life (NH +, Fe, Cr, Cu and Se). The recently reported pesticides in the lake possibly cause ecological and human health effect. Accordingly, agriculture and urbanisation are affecting water quality of Lake Ziway, with likely negative effects on human health and the lake ecosystem function unless appropriate interventions are taken. Our results may be useful in assessing other African lakes subject to similar anthropogenic pressures in their catchments.

There is increasing research interest in the application of the ecosystem services (ES) concept in the environmental risk assessment of chemicals to support formulating and operationalising regulatory environmental protection goals and making environmental risk assessment more policy- and value-relevant. This requires connecting ecosystem structure and processes to ecosystem function and henceforth to provision of ecosystem goods and services and their economic valuation. Ecological production functions (EPFs) may help to quantify these connections in a transparent manner and to predict ES provision based on function-related descriptors for service providing species, communities, ecosystems or habitats. We review scientific literature for EPFs to evaluate availability across provisioning and regulation and maintenance services (CICES v5.1 classification). We found quantitative production functions for nearly all ES, often complemented with economic valuation of physical or monetary flows. We studied the service providing units in these EPFs to evaluate the potential for extrapolation of toxicity data for test species obtained from standardised testing to ES provision. A broad taxonomic representation of service providers was established, but quantitative models directly linking standard test species to ES provision were extremely scarce. A pragmatic way to deal with this data gap would be the use of proxies for related taxa and stepwise functional extrapolation to ES provision and valuation, which we conclude possible for most ES. We suggest that EPFs may be used in defining specific protection goals (SPGs), and illustrate, using pollination as an example, the availability of information for the ecological entity and attribute dimensions of SPGs. Twenty-five pollination EPFs were compiled from the literature for biological entities ranging from ‘colony’ to ‘habitat’, with 75% referring to ‘functional group’. With about equal representation of the attributes ‘function’, ‘abundance’ and ‘diversity’, SPGs for pollination therefore would seem best substantiated by EPFs at the level of functional group.

The objective of this case study was to explore the feasibility of using ecological models for applying an ecosystem services-based approach to environmental risk assessment using currently available data and methodologies. For this we used a 5 step approach: 1) selection of environmental scenario, 2) ecosystem service selection, 3) development of logic chains, 4) selection and application of ecological models and 5) detailed ecosystem service assessment. The study system is a European apple orchard managed according to integrated pest management principles. An organophosphate insecticide was used as the case study chemical. Four ecosystem services are included in this case study: soil quality regulation, pest control, pollination and recreation. Logic chains were developed for each ecosystem service and describe the link between toxicant effects on service providing units and ecosystem services delivery. For the soil quality regulation ecosystem service, springtails and earthworms were the service providing units, for the pest control ecosystem service it was ladybirds, for the pollination ecosystem service it was honeybees and for the recreation ecosystem service it was the meadow brown butterfly. All the ecological models addressed the spatio-temporal magnitude of the direct effects of the insecticide on the service providing units and ecological production functions were used to extrapolate these outcomes to the delivery of ecosystem services. For all ecosystem services a decision on the acceptability of the modelled and extrapolated effects on the service providing units could be made using the protection goals as set by the European Food Safety Authority (EFSA). Developing quantitative ecological production functions for extrapolation of ecosystem services delivery from population endpoints remains one of the major challenges. We feel that the use of ecological models can greatly add to this development, although the further development of existing ecological models, and of new models, is needed for this.

The feasibility and added value of an ecosystem services approach in retrospective environmental risk assessment were evaluated using a site-specific case study in a lowland UK river. The studied water body failed to achieve good ecological status temporarily in 2018, due in part to the exceedance of the environmental quality standard (annual average EQS) for zinc. Potential ecosystem service delivery was quantified for locally prioritised ecosystem services: regulation of chemical condition; maintaining nursery populations and habitats; recreational fishing; nature watching. Quantification was based on observed and expected taxa or functional groups within WFD biological quality elements, including macrophytes, benthic macroinvertebrates and fish, and on published functional trait data for constituent taxa. Benthic macroinvertebrate taxa were identified and enumerated before, during and after zinc EQS exceedance, enabling a generic retrospective risk assessment for this biological quality element, which was found to have good ecosystem service potential. An additional targeted risk assessment for zinc was based on laboratory-based species sensitivity distributions normalised using biotic-ligand modelling to account for site-specific, bioavailability-corrected zinc exposure. Risk to ecosystem services for diatoms (microalgae) was found to be high, while risks for benthic macroinvertebrates and fish were found to be low. The status of potential ecosystem service delivery (ESD) by fish was equivalent to high ecological status defined under the WFD, while ESD was higher for benthic macroinvertebrates than defined by WFD methods. The illustrated ecosystem services approach uses readily available data and adds significantly to the taxonomic approach currently used under the WFD by using functional traits to evaluate services that are prioritised as being important in water bodies. The main shortcomings of the illustrated approach were lack of: representation of bacteria and fungi; WFD predicted species lists for diatoms and macrophytes; site-specific functional trait data required for defining actual (rather than potential) ecosystem service delivery.

Background Previous research has shown that the mechanics of walking on a treadmill is similar to walking overground. However it might be true that the energetics of walking is not similar between overground and treadmill, especially for older adults. We hypothesized that a lack of or inadequate familiarization on a treadmill would increase the Metabolic Cost of Transport (MCoT) in older adults due to increased levels of anxiety and the novelty of the locomotor task. Methods 10 healthy elderly (5 males and 5 females, mean age of 75.3 SD(6.3) years) were recruited and they walked first at their overground Preferred Walking Speed (PWS) for 8 min to reach a steady state of oxygen consumption in the morning. After that the same speed was imposed on a dual-belt treadmill and they walked for 15 min to properly familiarize themselves with the treadmill. This was called the familiarization session (Session 1). In the afternoon they repeated both the overground and treadmill walking again in the same order and with the same protocol. This session was called the post-familiarization session (Session 2). Results The group average of the overground PWS was 1.28 SD(0.11) m/s (4.61 SD(0.40) km/hr). During the familiarization session, the group average of the Gross Cost of Transport (GCoT) was 3.47 SD(0.35) J/kg/m while walking overground and 4 SD(0.65) J/kg/m while walking on a treadmill. The Net Cost of Transport (NCoT) was 2.64 SD(0.37) J/kg/m while walking overground and 3.14 SD(0.64) J/kg/m while walking on a treadmill. During the post-familiarization session, the group average of the GCoT was 3.84 SD(0.35) J/kg/m while walking overground and 3.94 SD(0.67) J/kg/m while walking on a treadmill. The NCoT was 2.76 SD(0.39) J/kg/m while walking overground and 2.90 SD(0.68) J/kg/m while walking on a treadmill. Both the GCoT and NCoT were statistically significantly higher on a treadmill than overground during the familiarization session. This elevation was not present during the post-familiarization session. There were also no statistically significant differences in the Resting Metabolic Rate (RMR) between before walking overground and before walking on a treadmill in either the familiarization or the post-familiarization session. Discussion and Conclusion This shows that the energetics of walking can be different for even healthy elderly on a treadmill if they are not or inadequately familiarized to it. This underlines the importance of adequate familiarization to treadmill walking for elderly in trying to understand the MCoT in this population.

Recent genome-wide association studies have identified over 230 genetic risk loci for multiple sclerosis. Current experimental autoimmune encephalomyelitis (EAE) models requiring active induction of disease may not be optimally suited for the characterization of the function of these genes. We have thus used gene expression profiling to study whether spontaneous opticospinal EAE (OSE) or MOG-induced EAE mirrors the genetic contribution to the pathogenesis of multiple sclerosis more faithfully. To this end, we compared gene expression in OSE and MOG EAE models and analyzed the relationship of both models to human multiple sclerosis risk genes and T helper cell biology. We observed stronger gene expression changes and an involvement of more pathways of the adaptive immune system in OSE than MOG EAE. Furthermore, we demonstrated a more extensive enrichment of human MS risk genes among transcripts differentially expressed in OSE than was the case for MOG EAE. Transcripts differentially expressed only in diseased OSE mice but not in MOG EAE were significantly enriched for T helper cell-specific transcripts. These transcripts are part of immune-regulatory pathways. The activation of the adaptive immune system and the enrichment of both human multiple sclerosis risk genes and T helper cell-specific transcripts were also observed in OSE mice showing only mild disease signs. These expression changes may, therefore, be indicative of processes at disease onset. In summary, more human multiple sclerosis risk genes were differentially expressed in OSE than was observed for MOG EAE, especially in T(H)1 cells. When studying the functional role of multiple sclerosis risk genes and pathways during disease onset and their interactions with the environment, spontaneous OSE may thus show advantages over MOG-induced EAE. We performed a greenhouse experiment to test how the functional diversity of earthworms, the dominant group of soil macro-invertebrates in many terrestrial ecosystems, affects nitrogen cycling and plant growth. Three species were chosen to represent a range of functional traits: Lumbricus terrestris (large, mainly detritivorous, makes vertical permanent burrows open at the surface), Aporrectodea longa (medium-large, feeds on both detritus and soil, makes burrows more branched than L. terrestris), and Allolobophora chlorotica (small, geophagous, makes ephemeral burrows below the soil surface). Mesocosms with ryegrass (Lolium perenne) were inoculated with none to all three species (similar total biomass), using an experimental design (Simplex) suited to partition single species and diversity effects. Two contrasting N sources, urea or mammalian dung, were labelled with 15N so that the acquisition by plants and earthworms and recovery of applied 15N could be estimated. Over 3 months, plant production was higher with urea applications, but there were also species-specific earthworm effects: A. chlorotica and, to a lesser extent, A. longa increased shoot biomass, whereas L. terrestris increased root biomass. Earthworms did not affect soil N concentrations or leaching losses, whereas more N was leached under urea. A. chlorotica tended to increase dung-15N recovery in grass shoots, but in interaction with A. longa had the opposite effect, possibly through increased N immobilization in the microbial biomass. Earthworms assimilated negligible amounts of urea-15N but a substantial proportion (17% on average) of the dung-15N, with no clear-cut differences between species. Our findings show that earthworm species may have similar trophic response to N sources and yet different effects on plant N uptake and primary production, and that inter-specific earthworm interactions can result in non-additive diversity effects.

Turoctocog alfa (NovoEight) is a third-generation recombinant factor VIII (rFVIII) with a truncated B-domain that is manufactured in Chinese hamster ovary cells. No human or animal-derived materials are used in the process. The aim of this study is to describe the molecular design and purification process for turoctocog alfa. A five-step purification process is applied to turoctocog alfa: protein capture on mixed-mode resin; immunoaffinity chromatography using a unique, recombinantly produced anti-FVIII mAb; anion exchange chromatography; nanofiltration and size exclusion chromatography. This process enabled reduction of impurities such as host cell proteins (HCPs) and high molecular weight proteins (HMWPs) to a very low level. The immunoaffinity step is very important for the removal of FVIII-related degradation products. Manufacturing scale data shown in this article confirmed the robustness of the purification process and a reliable and consistent reduction of the impurities. The contribution of each step to the final product purity is described and shown for three manufacturing batches. Turoctocog alfa, a third-generation B-domain truncated rFVIII product is manufactured in Chinese hamster ovary cells without the use of animal or human-derived proteins. The five-step purification process results in a homogenous, highly purified rFVIII product.

Soil biodiversity is deteriorating in Europe due to an on-going intensification of agriculture, climate change and food production supporting measures of the European Common Agricultural Policy (CAP). Nevertheless, the CAP tries to take biodiversity into account via proposing a range of agri-environmental measures. These ES contribute to food security, climate change mitigation, water retention and plant biomass growth. Healthy soils also help to prevent erosion, desertification, and landslides and to stabilise crop yields. The provision of ES by soil biota is a result of their impact on soil processes in interaction with soil conditions as well as soil management practices of the farmers such as tillage or crop rotations. Some taxa amongst soil biota play key roles in regulating soil processes. With respect to biocontrol of soil-borne pests, the earthworm species Lumbricus terrestris is known to play an important role in suppressing toxigenic plant pathogens, such as Fusarium culmorum and its mycotoxin deoxynivalenol (DON). We highlight the importance of earthworms for pest control to conceptualise and show how farmers' management practices influence soil ecosystem services and outline how this can be examined in a socio-ecological context by providing a concrete example of an economical evaluation of ES provided by earthworms.

Earthworms are important for soil functioning in arable cropping systems and earthworm species differ in their response to soil tillage and crop residue management. Lumbricus terrestris (Linnaeus, 1758) are rare in intensively tilled arable fields. In two parallel field trials with either non-inversion (NIT) or conventional tillage (CT), we investigated the feasibility of inoculating L. terrestris under different crop residue management (amounts and placement). Simultaneously, we monitored the response of the existing earthworm communities to L. terrestris inoculation and to crop residue treatments in terms of earthworm density, species diversity and composition, ecological groups and functional diversity. L. terrestris densities were not affected by residue management. We were not able to infer effects of the inoculation on the existing earthworm communities since L. terrestris also colonized non-inoculated plots. In NIT and two years after trial establishment, the overall native earthworm density was 1.4 and 1.6 times higher, and the epigeic density 2.5 times higher, in treatments with highest residue application (S 100 ) compared to 25% (S 25 ) or no (S 0 ) crop residues, respectively. Residue management did not affect earthworm species composition, nor the functional trait diversity and composition, except for an increase of the community weighted means of bifide typhlosolis in S 0 compared to S 100 . In CT, however, crop residues did have a strong effect on species composition, ecological groups and functional traits. Without crop residues (S 0 ), epigeic density was respectively 20 and 30% lower than with crop residues placed on the soil surface (S 100 ) or incorporated (I 100 ). Community composition was clearly affected by crop residues. Trait diversity was 2.6 to 3 times larger when crop residues were provided, irrespective of placement. Crop residues in CT also resulted in heavier earthworms and in a shift in the community towards species with a thicker epidermis and cuticle, a feather typhlosolis shape, and a higher average cocoon production rate. We conclude that earthworm communities under conventional tillage respond more strongly to the amount of crop residue than to its placement. Under non-inversion tillage, crop residue amounts affected earthworm communities, but to a smaller degree than under conventional tillage.

The ecosystem services approach has gained broad interest in regulatory and policy circles for use in ecological risk assessment. Whilst identifying several challenges, scientific experts from European regulatory authorities,the chemical industry and academia considered the approach applicable to all chemical sectors and potentially contributing to greater ecological relevance for setting and assessing environmental protection goals compared to current European regulatory frameworks for chemicals. These challengeswere addressed in workshops to develop a common understanding across stakeholders on how the ecosystem services concept might be used in chemical risk assessment and what would need to be done to implement it. This paper describes the consensusoutcome of those discussions. Knowledge gaps and research needs were identified and prioritised, exploring the use of novel approaches from ecology, ecotoxicology and ecological modelling. Where applicable, distinction ismade between prospective and retrospective ecological risk assessment. For prospective risk assessment the development of environmental scenarios accounting for chemical exposure and ecological conditions was designatedas a top priority. For retrospective risk assessment the top priority research need was development of reference conditions for key ecosystem services and guidance for their derivation. Both prospective and retrospectiverisk assessment would benefit from guidance on the taxa and measurement endpoints relevant to specific ecosystem services and from improved understanding of the relationships between measurement endpointsfromstandard toxicity tests and the ecosystemservices of interest (i.e. assessment endpoints). The development of mechanistic models, which could serve as ecological production functions, was identified as a priority.A conceptual framework for future chemical risk assessment based on an ecosystem services approach is presented.presented.©

A recently developed membrane-mimetic model was applied to study membrane interaction and binding of the two anchoring C2-like discoidin domains of human coagulation factor VIIIa (FVIIIa), the C1 and C2 domains. Both individual domains, FVIII C1 and FVIII C2, were observed to bind the phospholipid membrane by partial or full insertion of their extruding loops (the spikes). However, the two domains adopted different molecular orientations in their membrane-bound states; FVIII C2 roughly was positioned normal to the membrane plane, while FVIII C1 displayed a multitude of tilted orientations. The results indicate that FVIII C1 may be important in modulating the orientation of the FVIIIa molecule to optimize the interaction with FIXa, which is anchored to the membrane via its γ-carboxyglutamic acid-rich (Gla) domain. Additionally, a structural change was observed in FVIII C1 in the coiled main chain leading the first spike. A tight interaction with one lipid per domain, similar to what has been suggested for the homologous FVa C2, is characterized. Finally, we rationalize known FVIII antibody epitopes and the scarcity of documented hemophilic missense mutations related to improper membrane binding of FVIIIa, based on the prevalent nonspecificity of ionic interactions in the simulated membrane-bound states of FVIII C1 and FVIII C2.

Background and aims - Intense rains are becoming more frequent. By causing waterlogging, they may increase soil erosion and soil surface compaction, hamper seedling establishment, and reduce plant growth. Since anecic earthworms make vertical burrows that improve water infiltration, we hypothesised that they can counteract such disturbance. Methods - In a field experiment, intact soil mesocosms with ryegrass (Lolium multiflorum), with or without introduced adult Lumbricus terrestris, underwent either a precipitation regime with two intense rain events (36 mm, at beginning and end of spring), or a control regime with the same cumulative rainfall but no intense events. Short-term response of soil moisture and lagged response of plant growth were measured, and soil macroporosity was quantified. Results - Intense rains reduced ryegrass shoot biomass (by 16–21 % on average) only in the absence of earthworms. Waterlogging duration aboveground was not affected, whereas soil moisture contents after intense rainfall tended to drop faster with earthworms present. Continuous vertical macropores were found only in the mesocosms to which earthworms had been added. The number of such macropores was 2.4 times higher under the intense precipitation regime, despite similar earthworm survival. Conclusions - We found that anecic earthworms can offset negative effects of intense rainfall on plant growth aboveground. Underlying mechanisms, such as macropore formation and enhanced nutrient cycling, are discussed. We also observed that altered precipitation patterns can modify earthworm burrowing behaviour, as earthworms had produced more burrows under the intense regime

Background: Tyrosine kinase inhibitors (TKIs) have improved the survival of patients with chronic myeloid leukaemia. Many patients have deep molecular responses, a prerequisite for TKI therapy discontinuation. We aimed to define precise conditions for stopping treatment. Methods: In this prospective, non-randomised trial, we enrolled patients with chronic myeloid leukaemia at 61 European centres in 11 countries. Eligible patients had chronic-phase chronic myeloid leukaemia, had received any TKI for at least 3 years (without treatment failure according to European LeukemiaNet [ELN] recommendations), and had a confirmed deep molecular response for at least 1 year. The primary endpoint was molecular relapse-free survival, defined by loss of major molecular response (MMR; >0·1% BCR-ABL1 on the International Scale) and assessed in all patients with at least one molecular result. Secondary endpoints were a prognostic analysis of factors affecting maintenance of MMR at 6 months in learning and validation samples and the cost impact of stopping TKI therapy. We considered loss of haematological response, progress to accelerated-phase chronic myeloid leukaemia, or blast crisis as serious adverse events. This study presents the results of the prespecified interim analysis, which was done after the 6-month molecular relapse-free survival status was known for 200 patients. The study is ongoing and is registered with ClinicalTrials.gov, number NCT01596114. Findings: Between May 30, 2012, and Dec 3, 2014, we assessed 868 patients with chronic myeloid leukaemia for eligibility, of whom 758 were enrolled. Median follow-up of the 755 patients evaluable for molecular response was 27 months (IQR 21–34). Molecular relapse-free survival for these patients was 61% (95% CI 57–64) at 6 months and 50% (46–54) at 24 months. Of these 755 patients, 371 (49%) lost MMR after TKI discontinuation, four (1%) died while in MMR for reasons unrelated to chronic myeloid leukaemia (myocardial infarction, lung cancer, renal cancer, and heart failure), and 13 (2%) restarted TKI therapy while in MMR. A further six (1%) patients died in chronic-phase chronic myeloid leukaemia after loss of MMR and re-initiation of TKI therapy for reasons unrelated to chronic myeloid leukaemia, and two (

The sustainable management of land for agricultural production has at its core a healthy soil, because this reduces the quantity of external inputs, reduces losses of nutrients to the environment, maximises the number of days when the soil can be worked, and has a pore structure that maximises both the retention of water in dry weather and drainage of water in wet weather. Soil health encompasses the physical, chemical, and biological features, but the use of biological indicators is the least well advanced. Sustainability also implies the balanced provision of ecosystem services, which can be more difficult to measure than single indicators. We describe how the key components of the soil food web contribute to a healthy soil and give an overview of the increasing number of scientific studies that have examined the use of biological indicators. A case study is made of the ecosystem service of water infiltration, which is quite an undertaking to measure directly, but which can be inferred from earthworm abundance and biodiversity which is relatively easy to measure. This highlights the difficulty of putting any monitoring scheme into practice and we finish by providing the considerations in starting a new soil health monitoring service in the UK and in maintaining biological monitoring in The Netherlands.

Urban soils are major sinks that provide the services of attenuating and detoxifying environmental pollutants. This significant ecosystem service of urban soil can be evaluated by the natural attenuation capacity (NAC). In this research, we develop a method to calculate the natural pollutant attenuation capacity of urban soils on the basis of 5 chemical and physical measurements. The ecosystem services (ES) approach is gaining broad interest in regulatory and policy arenas for use in landscape management and ecological risk assessment. It has the potential to bring greater ecological relevance to the setting of environmental protection goals and to the assessment of the ecological risk posed by chemicals. A workshop, organised under the auspices of the Society of Environmental Toxicology and Chemistry Europe, brought together scientific experts from European regulatory authorities, the chemical industry and academia to discuss and evaluate the challenges associated with implementing an ES approach to chemical ecological risk assessment (ERA). Clear advantages of using an ES approach in prospective and retrospective ERA were identified, including: making ERA spatially explicit and of relevance to management decisions (i.e. indicating what ES to protect and where); improving transparency in communicating risks and trade-offs; integrating across multiple stressors, scales, habitats and policies. A number of challenges were also identified including: the potential for increased complexity in assessments; greater data requirements; limitations in linking endpoints derived from current ecotoxicity tests to impacts on ES. In principle, the approach was applicable to all chemical sectors, but the scale of the challenge of applying an ES approach to general chemicals with widespread and dispersive uses leading to broad environmental exposure, was highlighted. There was agreement that ES-based risk assessment should be based on the magnitude of impact rather than on toxicity thresholds. The need for more bioassays/tests with functional endpoints was recognized, as was the role of modelling and the need for ecological production functions to link measurement endpoints to assessment endpoints. Finally, the value of developing environmental scenarios that can be combined with spatial information on exposure, ES delivery and service provider vulnerability was recognized.

Objectives There is convergent evidence for an important role of interleukin-16 (IL-16) in the pathogenesis of multiple sclerosis (MS). IL-16 serves as a chemoattractant for different immune cells that are involved in developing lesions. Here, we compared IL-16 levels of MS patients and controls and addressed the long-term effect of IFN-β, the most common immunomodulatory MS therapy, on IL-16 serum levels in MS patients over 2 years. Beyond this, we analysed the expression of IL-16 in two CD4+ T-cell subsets, Th1 and Th17 cells, which are important autoimmune mediators and affected by IFN-β treatment, derived from myelin-specific T-cell transgenic mice. Materials and methods IL-16 serum levels of 17 controls and of 16 MS patients before therapy and at months 1, 2, 3, 6, 9, 12 and 24 during IFN-β1a therapy were determined by ELISA. MRI was performed before therapy, at months 12 and 24. IL-16 expression of in vitro differentiated murine myelin oligodendrocyte glycoprotein (MOG)-specific Th1 and Th17 cells was quantified by real-time PCR. Results Before therapy, MS patients showed significantly elevated IL-16 levels compared with controls irrespective of disease activity determined by MRI. Therapy with IFN-β1a led to a significant linear decrease in IL-16 serum levels beginning after 2 months. MOG-specific Th17 cells expressed more IL-16 than Th1 cells. Conclusions Reduction in increased IL-16 levels may be of relevance for the therapeutic effect of IFN-β1a in MS. Easily accessible IL-16 serum levels hold a potential as biomarker of treatment efficacy in MS.

Soil organisms have an important role in aboveground community dynamics and ecosystem functioning in terrestrial ecosystems. However, most studies have considered soil biota as a black box or focussed on specific groups, whereas little is known about entire soil networks. Here we show that during the course of nature restoration on abandoned arable land a compositional shift in soil biota, preceded by tightening of the belowground networks, corresponds with enhanced efficiency of carbon uptake. In mid- and long-term abandoned field soil, carbon uptake by fungi increases without an increase in fungal biomass or shift in bacterial-to-fungal ratio. The implication of our findings is that during nature restoration the efficiency of nutrient cycling and carbon uptake can increase by a shift in fungal composition and/or fungal activity. Therefore, we propose that relationships between soil food web structure and carbon cycling in soils need to be reconsidered., Effects of habitat restoration on belowground organisms and ecosystem processes are poorly understood. Morriën and colleagues show that changes in the composition and network interactions of soil biota lead to improved carbon uptake efficiency when formerly cultivated land is restored.
