The thermal resistances (R-values) of airspaces depends on the emittance of all surfaces around an airspace, dimensions, heat-flow direction, and the temperatures of the bounding surfaces. Assessing the energy performance of building envelope components and fenestration systems requires accurate results for the R-values of any enclosed spaces. The evaluation of reflective insulation R-values has evolved to include use of computational fluid dynamics and surface-to-surface radiation to quantify convective and radiative contributions to the heat transfer across airspaces of all types. This paper compares an advanced and validated model for calculating enclosed airspace R-values with the widely-used ISO 6946 and airspace R-values in the ASHRAE Handbook-Fundamentals. The impact of construction and installation defects on the thermal performance of reflective insulation has not been previously evaluated. In this research, an advanced model was used to evaluate a construction defect and dimensional aspect ratios that one-dimensional methods do not address. Imperfect installation and defects that result in air movement into or through a reflective insulation assembly reduces the thermal resistance of the assembly. Additionally, the amount of thermal resistance reduction depends on the amount and temperature of invasive air or the size of internal defects that allows natural convection inside the reflective airspace. In this study, these performance issues are evaluated quantitatively using computer simulation techniques. The differences in results obtained using methods that are currently being used to evaluate the R-value and the advantages of the advanced method for evaluating the reflective insulation performance for different applications are discussed. For the case considered in this study, the results showed that the failure to achieve parallel surfaces results in less than a 5% decrease in thermal resistance. Also, the results showed that internal air gaps between airspaces result in negligible loss in R-value unless air gaps that allow circulation between airspaces are created.

The unique nerve terminal targeting of botulinum neurotoxin type A (BoNT/A) is due to its capacity to bind two receptors on the neuronal plasma membrane: polysialoganglioside (PSG) and synaptic vesicle glycoprotein 2 (SV2). Whether and how PSGs and SV2 may coordinate other proteins for BoNT/A recruitment and internalization remains unknown. Here, we demonstrate that the targeted endocytosis of BoNT/A into synaptic vesicles (SVs) requires a tripartite surface nanocluster. Live-cell super-resolution imaging and electron microscopy of catalytically inactivated BoNT/A wildtype and receptor-binding-deficient mutants in cultured hippocampal neurons demonstrated that BoNT/A must bind coincidentally to a PSG and SV2 to target synaptic vesicles. We reveal that BoNT/A simultaneously interacts with a preassembled PSG-synaptotagmin-1 (Syt1) complex and SV2 on the neuronal plasma membrane, facilitating Syt1-SV2 nanoclustering that controls endocytic sorting of the toxin into synaptic vesicles. Syt1 CRISPRi knockdown suppressed BoNT/A- and BoNT/E-induced neurointoxication as quantified by SNAP-25 cleavage, suggesting that this tripartite nanocluster may be a unifying entry point for selected botulinum neurotoxins that hijack this for synaptic vesicle targeting.

The phospholipid and free fatty acid (FFA) composition of neuronal membranes plays a crucial role in learning and memory, but the mechanisms through which neuronal activity affects the brain’s lipid landscape remain largely unexplored. Saturated FFAs, particularly myristic acid (C14:0), strongly increase during neuronal stimulation and memory acquisition, suggesting the involvement of phospholipase A1 (PLA1) activity in synaptic plasticity. Here, we show that genetic ablation of the DDHD2 isoform of PLA1 in mice reduced memory performance in reward-based learning and spatial memory models prior to the development of neuromuscular deficits, and markedly reduced saturated FFAs across the brain. DDHD2 was shown to bind to the key synaptic protein STXBP1. Using STXBP1/2 knockout neurosecretory cells and a haploinsufficientSTXBP1+/-mouse model of STXBP1 encephalopathy that is also associated with intellectual disability and motor dysfunction, we show that STXBP1 controls the targeting of DDHD2 to the plasma membrane and the generation of saturated FFAs in the brain. Our findings suggest key roles for DDHD2 and STXBP1 in the lipid metabolism underlying synaptic plasticity, learning and memory.

2019 coronavirus disease (COVID-19) is a disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). In addition to respiratory illness, COVID-19 patients exhibit neurological symptoms lasting from weeks to months (long COVID). It is unclear whether these neurological manifestations are due to an infection of brain cells. We found that a small fraction of human induced pluripotent stem cell (iPSC)-derived neurons, but not astrocytes, were naturally susceptible to SARS-CoV-2. Based on the inhibitory effect of blocking antibodies, the infection seemed to depend on the receptor angiotensin-converting enzyme 2 (ACE2), despite very low levels of its expression in neurons. The presence of double-stranded RNA in the cytoplasm (the hallmark of viral replication), abundant synthesis of viral late genes localized throughout infected cells, and an increase in the level of viral RNA in the culture medium (viral release) within the first 48 h of infection suggested that the infection was productive. Productive entry of SARS-CoV-2 requires the fusion of the viral and cellular membranes, which results in the delivery of the viral genome into the cytoplasm of the target cell. The fusion is triggered by proteolytic cleavage of the viral surface spike protein, which can occur at the plasma membrane or from endosomes or lysosomes. We found that SARS-CoV-2 infection of human neurons was insensitive to nafamostat and camostat, which inhibit cellular serine proteases, including transmembrane serine protease 2 (TMPRSS2). Inhibition of cathepsin L also did not significantly block infection. In contrast, the neuronal infection was blocked by apilimod, an inhibitor of phosphatidyl-inositol 5 kinase (PIK5K), which regulates early to late endosome maturation.IMPORTANCE COVID-19 is a disease caused by the coronavirus SARS-CoV-2. Millions of patients display neurological symptoms, including headache, impairment of memory, seizures, and encephalopathy, as well as anatomical abnormalities, such as changes in brain morphology. SARS-CoV-2 infection of the human brain has been documented, but it is unclear whether the observed neurological symptoms are linked to direct brain infection. The mechanism of virus entry into neurons has also not been characterized. Here, we investigated SARS-CoV-2 infection by using a human iPSC-derived neural cell model and found that a small fraction of cortical-like neurons was naturally susceptible to infection. The productive infection was ACE2 dependent and TMPRSS2 independent. We also found that the virus used the late endosomal and lysosomal pathway for cell entry and that the infection could be blocked by apilimod, an inhibitor of cellular PIK5K.

Acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the causative agent of the COVID-19 pandemic. Despite vaccinations, the development and use of neutralizing antibodies against the viral surface spike proteins, and small molecule inhibitors targeting the viral replication machinery, COVID-19 remains a global public health crisis. Emerging mutations in the viral genome have the potential to reduce prophylactic and therapeutic efficacy of virus-directed treatments. Targeting host cell factors required for infection could, therefore, be a potential strategy to overcome this problem since mutations in the viral genome are unlikely to bypass the requirement for the targeted host factor or function. The enzymatic activity of N-myristoyltransferases (NMTs) are essential to mediate stable membrane binding and function of a diverse class of cellular proteins, many of which regulate intracellular membrane trafficking. Here we report that nanomolar concentrations of the NMT inhibitor IMP-1088 inhibited SARS-CoV-2 spreading in human cells by compromising the infectivity of released viral particles, which was reduced by up to 90%. IMP-1088 also inhibited human Respiratory syncytial virus, the main cause of viral death in infants world-wide, but not the mosquito-delivered alphavirus Semliki Forest virus and the vesiculovirus Vesicular stomatitis virus. The antiviral effect of IMP-1088 against SARS-CoV-2 displayed remarkably slow reversibility, was well tolerated by cells, and is, therefore, a promising candidate for COVID-19 prophylaxis and therapy.

This paper presents the synthesis and isolation of a new binuclear complex of yttrium with anthranilic acid (HA). The complex [Y2(HA)6(H2O)4] Cl6.2C2H5OH (C1) was obtained as single crystals that its X-ray analysis revealed its triclinic P-1 space group in addition to anti-prismatic geometry around each of the yttrium ions. In the complex, the anthranilic acid ligands are bidentate, zwitter ionic and neutral, and the yttrium ions’ charge is only compensated by six chloride ions. The cytotoxicity of this complex against human breast cancer MDA-MB-231 cells, prostate cancer PC-3 cells, and bladder cancer T-24 cells was evaluated. This yttrium complex displayed more cytotoxic activity against the bladder cancer cells with an IC50 value of 307.7 μg/ml (223 μM). On the other hand, the activities of complex C1 against the MDA-MB-231 and PC-3 cells were less significant respectively with IC50 values of 1097 μg/ml (796 μM) and 921 μg/ml (669 μM).

Graphical abstract Sleep/wake cycle-dependent differences in molecular parameters controlling mitochondrial ability to regulate cellular levels of calcium and associated ROS production., Introduction Incidents of myocardial infarction and sudden cardiac arrest vary with time of the day, but the mechanism for this effect is not clear. We hypothesized that diurnal changes in the ability of cardiac mitochondria to control calcium homeostasis dictate vulnerability to cardiovascular events. Objectives Here we investigate mitochondrial calcium dynamics, respiratory function, and reactive oxygen species (ROS) production in mouse heart during different phases of wake versus sleep periods. Methods We assessed time-of-the-day dependence of calcium retention capacity of isolated heart mitochondria from young male C57BL6 mice. Rhythmicity of mitochondrial-dependent oxygen consumption, ROS production and transmembrane potential in homogenates were explored using the Oroboros O2k Station equipped with a fluorescence detection module. Changes in expression of essential clock and calcium dynamics genes/proteins were also determined at sleep versus wake time points. Results Our results demonstrate that cardiac mitochondria exhibit higher calcium retention capacity and higher rates of calcium uptake during sleep period. This was associated with higher expression of clock gene Bmal1, lower expression of per2, greater expression of MICU1 gene (mitochondrial calcium uptake 1), and lower expression of the mitochondrial transition pore regulator gene cyclophilin D. Protein levels of mitochondrial calcium uniporter (MCU), MICU2, and sodium/calcium exchanger (NCLX) were also higher at sleep onset relative to wake period. While complex I and II-dependent oxygen utilization and transmembrane potential of cardiac mitochondria were lower during sleep, ROS production was increased presumably due to mitochondrial calcium sequestration. Conclusions Taken together, our results indicate that retaining mitochondrial calcium in the heart during sleep dissipates membrane potential, slows respiratory activities, and increases ROS levels, which may contribute to increased vulnerability to cardiac stress during sleep-wake transition. This pronounced daily oscillations in mitochondrial functions pertaining to stress vulnerability may at least in part explain diurnal prevalence of cardiac pathologies.

Assessing the energy performance of building components with enclosed airspaces requires accurate determination of the thermal resistance ( R-value) of the airspaces. The R-value of enclosed airspace depends on its size and orientation, direction of heat transfer through the airspace, and temperatures and emissivities of all surfaces that define the airspace. In previous studies, practical correlations were developed to determine the R-values for vertical enclosed airspaces, horizontal enclosed airspaces with upward heat flow and downward heat flow, and 30° and 45° sloped-enclosed airspaces with downward heat flow. However, to the authors’ best knowledge, there is no such practical correlations available to determine the R-values for wide ranges of dimensions and operating conditions for 30° and 45° sloped-enclosed airspaces with upward heat flow. This paper focused on the thermal performance of 45° sloped-enclosed airspaces with upward heat flow, and the predicted R-values were compared with the R-values provided in ASHRAE Handbook of Fundamentals at different conditions. The dependence of the R-value on the aspect ratio of the enclosed airspaces was also investigated. As well, considerations were given to quantify the potential increase in the R-value of enclosed airspace when a thin sheet having different values of emissivity on both sides was placed in the middle of the airspace. The results showed that depending on the value of the effective emittance and the thickness of the airspace, the R-value could be tripled by incorporating thin a sheet in the middle of the enclosed airspace. Finally, practical correlation were developed to determine the effective R-values of 45° sloped-enclosed airspaces with upward heat flow for wide ranges of aspect ratio, temperature difference across the airspace, mean temperature, and effective emittance. The results showed that the calculated R-values using this correlation were in good agreement with the predicted R-values.

The measurements for the short-wave solar reflectivity of a Reflective Coating Material (RCM) with various cleaning operations that were obtained in a previous study were used in this study to conduct numerical simulations in order to assess the moisture and energy performance of cool and black roofs when they were subjected to the weather conditions of Saudi Eastern Province and Kuwait City. The results of the numerical simulations showed that black roofs always work with less moisture than cool roofs. Because the highest relative humidity in the different components of the black and cool roofs was well below 80%, there was no risk of condensation and mold growth in these roofs. For both weather conditions of Saudi Eastern Province and Kuwait City, the results showed that installing cool roofs have resulted in increasing the heating energy loads in relation to black roofs. Conversely, the results showed that the decrease in the cooling energy loads due to installing cool roofs were typically much greater than the increase in the heating energy loads. As such, cool roofs have resulted in net energy savings in relation to black roofs. Replacing black roof by cool roof in Saudi climate, the results showed that the annual energy savings in the total energy load was 25% and 34% as a result of installing cool roof with RCM at no cleaning and weekly homemade cleaning, respectively. Additionally, replacing black roof by cool roof in Kuwaiti climate, the results showed that the annual energy savings in the total energy load was 23% and 31% a result of installing cool roof with RCM at no cleaning and weekly homemade cleaning, respectively.

A novel series of 5-amino-6-substituted-3-methyl-1-phenyl-1H-pyrazolo[3,4-b]selenolo[3,2-e]pyrazines (3a–e) was synthesized by the reaction of the chloro pyrazolo[3,4-b]pyrazine carbonitrile 1 with selenium element in the presence of sodium borohydride and ethanol, followed by the reaction with α-halo alkylating agents to produce the selanyl-alkylated derivatives 2a–e. The latter compounds underwent Thorpe-Ziegler cyclization upon heating with ethanolic sodium ethoxide solution to afford the target selenolopyrazolopyrazine compounds. The 5-amino-3-methyl-1-phenyl-1H-pyrazolo[3,4-b]selenolo[3,2-e]pyrazine-6-carboxamide (3b) was used as a versatile precursor for synthesis of new heterocyclic fused to the pyrazoloselenolopyrazine moiety namely: pyrimidine and imidazopyrimidine. Assignment of the chemical structures for the newly synthesized compounds was confirmed on the bases of elemental and spectral techniques including FT-IR, 1H NMR, 13C NMR, and mass spectra. Furthermore, certain compounds were screened for their antimicrobial activity which revealed remarkable activities against various pathogenic strains of bacteria and fungi. Alternatively, some of these compounds exhibited promising anticancer action against some colon and breast cancer cells.

Heat transmission across an enclosed space is determined by the type of filling gas, the temperatures and long-wave emissivities of all surfaces that define the space, and the heat flow direction. The ASHRAE Handbook of Fundamentals provides the thermal resistances (R-values) of enclosed airspaces with only five effective emittance values (E) for vertical, horizontal, and 45° airspaces. ASHRAE R-values do not include the case of 30° sloped airspaces. In addition, ASHRAE R-values ignore the impact of the airspace aspect ratio (A) on R-values. However, many previous studies, as well as this study, have shown that A can have a significant effect on the R-value. Previously, correlations were developed for determining the R-values for vertical (90°) airspaces subjected to horizontal heat flow, horizontal (0°) airspaces subjected to up and down heat flow, 45° airspaces subjected to up and down heat flow, and 30° airspaces subjected to downward heat flow. To the authors’ knowledge, no such correlation existed for determining the R-value of 30° airspaces subjected to upward heat flow, which is developed in this paper. The potential increase in R-value by placing a thin layer of varied emittance on both sides in the middle of the airspace was also considered. Architects and building designers can use the developed correlation to compute the R-values of airspaces of various values for A and E and various operating conditions. This correlation along with the previous correlations can be included in the current energy models (e.g., EnergyPlus, ESP-r, DOE, and Design Builder).

Objectives: Cisplatin (CISP) is an effective chemotherapy used in the treatment of various types of cancer, but it causes nephrotoxicity and other adverse effects. Thymoquinone (THY) is an effective anti-inflammatory and antioxidant compound, which might protects against many chemotherapies associated toxicities. However, THY applications are hindered by its poor solubility and low bioavailability. This study evaluated the efficacy of a novel nanoparticle (NP) encapsulting THY to overcome its poor solubility, enhance its bioavilability, efficacy for the protection against CISP-induced nephrotoxicity in an Ehrlich solid carcinoma (ESC) mice model. Methods: Four treatment groups were included: 1) control, 2) tumour, 3) CISP, and 4) CISP + NP THY. Results: The results showed that NP THY was effective in preventing CISP-induced kidney toxicity in ESC mice and improved its function and pathology. NP THY effectively ameliorated CISP-induced oxidative stress conditions in the kidney tissue via increasing the levels of antioxidants both non-enzymatic (GSH) and enzymatic (SOD and CAT). NP THY, also, significantly reduced CISP-induced kidney inflammation by reducing TNF-α, IL-1β, and NF-kB levels. NP THY didn’t hinder the antitumor activity of CISP as shown by tumour weight and histological examination data. Conclusions: In conclusion, NP THY could be an adjuvant therapy to CISP cancer treatment to prevent associated nephrotoxicity and other adverse effects without compromising CISP antitumor efficacy.

The building envelope provides thermal comfort, an excellent visual view, and sunlight for the occupants. It consists of two parts: (i) an opaque (non-transparent) part (e.g., walls and roofs) and (ii) a transparent part (e.g., windows, curtain walls, and skylight devices). Recently, the use of fully-glazed facades, especially in large cities, has increased due to their aesthetical and structural advantages. This has led this study to review the performance of the currently passive smart glazing technologies. Phase Change Materials (PCMs) as latent energy storage material is the focus of this review, as well as other individual and combined techniques, including shading systems, solar cells (photovoltaic), and chromogenic (thermotropic and thermochromic) materials. PCM-integrated glazing systems have been extensively studied and rapidly developed over the past several decades from the standpoint of unique system designs, such as passive, active, and passive/active mixed designs, intelligent management, and sophisticated controls. In the academic literature, numerous studies on PCM-integrated building envelopes have been conducted, but a comprehensive review of PCM-integrated GUs combined with other passive and active techniques using dialectical analysis and comparing the climatic conditions of each study using Köppen-Geiger climate classification climate classification has been performed only rarely. Consequently, the primary objective of this study is to reduce this discrepancy for all types of glazing, excluding glazed roofs. This review article also contains literature tables as well as highlights, limitations, and further research suggestions at the end of each subsection.

Recent years have reported a rise in the occurrence of gastric ulceration especially among young children and adults. This study investigated the mechanism by which two types of Saudi honey: Alnahal Aljawal honey (Wadi) or Bin Ghaithan honey (Talh) exerted their antiulcer potential in indomethacin-induced gastric ulceration. Four cohorts of rats were used: Group 1; Healthy controls, Group 2; Ulcerative animals, Group 3; Ulcerative + Wadi honey treatment, Group 4; Ulcerative + Talh honey treatment. We profiled the levels of different indicators of oxidative stress including the activities of gastric mucosal glutathione superoxide dismutase (SOD), catalase (CAT), peroxidase (GPx), reduced glutathione (GSH), and lipid peroxidation (measured as malondialdehyde; MDA). CRP content, IL-10, and plasma tumor necrosis factor-α were also evaluated. The stomach was visually examined for macroscopic lesions and using light microscope for histopathological changes in the glandular mucosa. Wadi or Talh honey significantly reduced the ulcer indices, and essentially protected the glandular mucosa from lesions. Wadi or Talh honey also significantly reduced the gastric mucosal concentrations of GPx, SOD and GSH. In addition, the administration of Wadi or Talh honey decreased gastric mucosal plasma TNF-α and MDA, CRP content, and IL-10 levels. In conclusion, Wadi or Talh honey possibly exerted their antiulcer potential via restoring the homeostasis and stabilizing the enzymatic (SOD and GPx) and non-enzymatic (GSH) antioxidants as well as reducing the levels of inflammatory cytokines (TNF-α, CRP content, IL-10 and, NF-κB activity), and inhibiting the lipid peroxidation in the gastric mucosa. Consequently, Wadi or Talh honey may be of beneficial therapy for patients diagnosed with gastric ulceration. Clinical studies need to be conducted to further support these findings.

In this paper, an integrated model that coupled CONTAM and WUFI was developed to assess the indoor air quality (IAQ), moisture, and thermal comfort performance. The coupling method of CONTAM and WUFI is described based on the exchange of airflow rate control variables as infiltration, natural and mechanical ventilation parameters between heat and moisture flow balance equations in WUFI and contaminant flow balances equations in CONTAM. To evaluate the predictions of the integrated model compared to single models of CONTAM and WUFI, four scenarios were used. These scenarios are airtight-fan off, airtight-fan on, leaky-fan off, and leaky-fan on, and were defined for a three-story house subjected to three different climate conditions of Montreal, Vancouver, and Miami. The measures of the simulated indoor CO2, PM2.5, and VOCs obtained by CONTAM; the simulated indoor relative humidity (RH), predicted percentage of dissatisfied (PPD), and predicted mean vote (PMV) obtained by WUFI; and those obtained by the integrated model are compared separately for all scenarios in Montreal, Vancouver, and Miami. Finally, the optimal scenarios are selected. The simulated results of the optimal scenarios with the integrated model method (−28.88% to 46.39%) are different from those obtained with the single models. This is due to the inability of the single models to correct the airflow variables.

Many parts of the building envelopes contain enclosed airspaces. Also, the insulating glass units in fenestration systems, such as curtain walls, windows, and skylight devices, contain enclosed spaces that are normally filled with air or heavy gas such as argon, xenon, or krypton. The thermal resistance (R-value) of an enclosed space depends mainly on the type of the filling gas, emissivity of all surfaces that bound the space, the size and orientation of the space, the direction of heat flow through the space, and the respective temperatures of all surfaces that define the space. Assessing the energy performance of building envelopes and fenestration systems, subjected to different climatic conditions, requires accurate determination of the R-values of the enclosed spaces. In this study, a comprehensive review is conducted on the thermal performance of enclosed airspaces for different building applications. This review includes the computational and experimental methods for determining the effective R-value of enclosed reflective airspaces. Also, the different parameters that affect the thermal performance of enclosed airspaces are discussed. These parameters include the following: (a) dimensions, (b) inclination angles, (c) directions of heat flow, (d) emissivity of all surfaces that bound the space, and (e) operating conditions. Moreover, numerical simulations are conducted using a previously developed and validated model to investigate the effect of the inclination angle, direction of heat transfer, and the coating emissivity on the R-values of enclosed spaces when they are filled with different types of gases.

Thermoelectric devices are currently being used in the applications of cooling and generating electricity. This study mainly focuses on using these devices for both applications toward cooling down computer chips. An important aspect in designing the cooling system is to minimize the non-uniformity of the temperature distribution in the computer chip so as to reduce the thermal stresses in it. Another aspect in designing the cooling system is to minimize its power requirements. To investigate these two aspects, the temperatures of the cold chip areas can be allowed to increase, but not to exceed a certain temperature threshold, by installing Thermoelectric Generators (TEGs) on these areas that can harvest electrical power from the chip wasted heat. Thereafter, the chip hotspot areas can be cooled down by installing Thermoelectric Coolers (TECs) on these areas that can be powered by the harvested electrical power from the TEGs in order to maintain the temperatures of these hotspots to be less than or equal a certain temperature threshold. This cooling technique is called “sustainable self-cooling framework” for cooling chip hotspots. However, the question is: can the harvested electrical power by the TEGs be enough to power the TECs for cooling chip hotspots? In this study, a 3D model is developed to optimize the performance of both TEGs and TECs. Thereafter, this model is validated against experimental data of TEC and TEG. The results showed that the model predictions were in good agreements with the experimental data to within ±4%. Also, considerations are given in this study to optimize the performance of cascaded and non-cascaded TEGs and TECs for future use them to develop sustainable self-cooling frameworks for cooling chip hotspots at different operating conditions. Finally, a case study is conducted in this paper for a sustainable self-cooling framework in order to address the question above. The results showed that the self-cooling framework can successfully cool down the hotspot at an acceptable temperature with not only no need for additional electrical power requirements but also for reducing the non-uniformity in the chip temperature distribution.

In this study, two highly thermotolerant and methanol-tolerant lipase-producing bacteria were isolated from cooking oil and they exhibited a high number of catalytic lipase activities recording 18.65 ± 0.68 U/mL and 13.14 ± 0.03 U/mL, respectively. Bacterial isolates were identified according to phenotypic and genotypic 16S rRNA characterization as Kocuria flava ASU5 (MT919305) and Bacillus circulans ASU11 (MT919306). Lipases produced from Kocuria flava ASU5 showed the highest methanol tolerance, recording 98.4% relative activity as well as exhibited high thermostability and alkaline stability. Under the optimum conditions obtained from 3D plots of response surface methodology design, the Kocuria flava ASU5 biocatalyst exhibited an 83.08% yield of biodiesel at optimized reaction variables of, 60 ○C, pH value 8 and 1:2 oil/alcohol molar ratios in the reaction mixture. As well as, the obtained results showed the interactions of temperature/methanol were significant effects, whereas this was not noted in the case of temperature/pH and pH/methanol interactions. The obtained amount of biodiesel from cooking oil was 83.08%, which was analyzed by a GC/Ms profile. The produced biodiesel was confirmed by Fourier-transform infrared spectroscopy (FTIR) approaches showing an absorption band at 1743 cm−1, which is recognized for its absorption in the carbonyl group (C=O) which is characteristic of ester absorption. The energy content generated from biodiesel synthesized was estimated as 12,628.5 kJ/mol. Consequently, Kocuria flava MT919305 may provide promising thermostable, methanol-tolerant lipases, which may improve the economic feasibility and biotechnology of enzyme biocatalysis in the synthesis of value-added green chemicals.

Cool roofs use reflective materials or coatings to reflect a portion of the incident solar radiation. This results in a lowering the surface temperature of the cool roof compared to black roofs, and thus helps reduce the cooling energy loads during the summer season. The research reported in this paper was conducted to assess experimentally and numerically the performance of cool and black roofs that were subjected to the hot, humid and dusty climate of Jubail Industrial City (JIC). This paper focused on characterizing one of the important properties of reflective coating material (RCM), which is its solar reflectivity. In this study, the effect of dust/dirt accumulation on the solar reflectivity of the RCM was investigated at different exposure times to the natural weathering conditions of JIC. The test results showed that dust and dirt can significantly contribute in reducing the solar reflectivity of the RCM. As such, a number of cleaning processes were conducted on the surface of the RCM so as to increase its solar reflectivity. The effect of each cleaning process on the solar reflectivity of the RCM was investigated. Finally, this paper provides a test protocol and procedure for characterizing the dust concentration/intensity on the surfaces of the RCM and cleaning this material after different exposure times to a natural and polluted climate.

HEV is the most causative agent of acute viral hepatitis globally. HEV causes acute, chronic, and extrahepatic manifestations. Chronic HEV infection develops in immunocompromised patients such as organ transplant patients, HIV-infected patients, and leukemic patients. The source of chronic HEV infection is not known. Also, the source of extrahepatic manifestations associated with HEV infection is still unclear. Hepatotropic viruses such as HCV and HBV replicate in peripheral blood mononuclear cells (PBMCs) and these cells become a source of chronic reactivation of the infections in allograft organ transplant patients. Herein, we reported that PBMCs and bone marrow-derived macrophages (BMDMs), isolated from healthy donors (n = 3), are susceptible to HEV in vitro. Human monocytes (HMOs), human macrophages (HMACs), and human BMDMs were challenged with HEV-1 and HEV-3 viruses. HEV RNA was measured by qPCR, the marker of the intermediate replicative form (ds-RNA) was assessed by immunofluorescence, and HEV capsid protein was assessed by flow cytometry and ELISA. HEV infection was successfully established in primary HMOs, HMACs, and human BMDMs, but not in the corresponding cells of murine origin. Intermediate replicative form (ds RNA) was detected in HMOs and HMACs challenged with HEV. The HEV load was increased over time, and the HEV capsid protein was detected intracellularly in the HEV-infected cells and accumulated extracellularly over time, confirming that HEV completes the life cycle inside these cells. The HEV particles produced from the infected BMDMs were infectious to naive HMOs in vitro. The HEV viral load was comparable in HEV-1- and HEV-3-infected cells, but HEV-1 induced more inflammatory responses. In conclusion, HMOs, HMACs, and human BMDMs are permissive to HEV infection and these cells could be the source of chronic and recurrent infection, especially in immunocompromised patients. Replication of HEV in human BMDMs could be related to hematological disorders associated with extrahepatic manifestations.

Explore the relationship between the recalled bond of pregnant women to their own mothers and the development of prenatal attachment.Women with a single-fetal pregnancy who consulted the outpatient service of the maternity "C" ward of the center of maternity and neonatology of Tunis and had perceived fetal movements were included. Maternal-fetal attachment was assessed by the Prenatal Attachment Inventory (PAI), and the Parental Bonding Instrument (PBI) was used to evaluate experienced bond to mother. Socio demographic and clinical data were collected on a pre-established form.Eighty respondents were retained from 95 recruited pregnant women. Their ages ranged from 19 to 44 with an average of 32 years. The rate of primiparity was of 28.7 % and half of pregnancies were identified at risk. The mean score of PAI was 55.3 (ET=10.79). Means scores of PBI "care" dimension and "overprotection" dimension were respectively of 26.26 (ET=5.82) and 17 (ET=6.38). The four types of maternal bonding according to the PBI scores were distributed as follow: affectionate constraint (31.3 %), affectionless control (35 %), optimal bonding (21.3 %) and weak bonding (12.5 %). The two PBI dimensions "care" and "overprotection" were significantly and negatively correlated (P0.05). The PAI scores were not correlated with the PBI "care" dimension scores but were significantly and negatively correlated with the "overprotection" dimension scores (P0.,01). In order to better explore this association, the "overprotection" dimension scores were studied according to the 21 items of the PAI. A negative and significant correlation was found only with three of the PAI items: item 12 (P0;01), 15, and 20 (P0;05).Our findings suggest that pregnant women whose mothers showed more overprotective and controlling behaviors could be at risk of developing a weak maternofetal attachment. Further research is needed.

Exosomes are nano-membrane vesicles that various cell types secrete during physiological and pathophysiological conditions. By shuttling bioactive molecules such as nucleic acids, proteins, and lipids to target cells, exosomes serve as key regulators for multiple cellular processes, including cancer metastasis. Recently, microvesicles have emerged as a challenge in the treatment of prostate cancer (PCa), encountered either when the number of vesicles increases or when the vesicles move into circulation, potentially with an ability to induce drug resistance, angiogenesis, and metastasis. Notably, the exosomal cargo can induce the desmoplastic response of PCa-associated cells in a tumor microenvironment (TME) to promote PCa metastasis. However, the crosstalk between PCa-derived exosomes and the TME remains only partially understood. In this review, we provide new insights into the metabolic and molecular signatures of PCa-associated exosomes in reprogramming the TME, and the subsequent promotion of aggressive phenotypes of PCa cells. Elucidating the molecular mechanisms of TME reprogramming by exosomes draws more practical and universal conclusions for the development of new therapeutic interventions when considering TME in the treatment of PCa patients.

Cisplatin (CISP) is one of the most widely used anti-cancer chemotherapeutic agents with remarkable efficacy against various types of cancers. However, it has been associated with nephrotoxicity amongst other undesirable side effects. Pomegranate (PE) is a potent antioxidant and anti-inflammatory agent effective against cancer, with a superior benefit of not being associated with the common toxicities related to the use of conventional chemotherapeutic agents. However, the application of PE is limited by its reduced solubility and decreased bioavailability. We investigated the potential of a novel nanoparticle (NP) enclosing PE to enhance its solubility and improve its bioavailability, and efficacy to prevent CISP-associated nephrotoxicity in a mice model of Ehrlich solid carcinoma (ESC). All mice were grouped into four cohorts: (I) control, (II) tumor, (III) CISP, and (IV) CISP + PE-NPs. The data obtained demonstrated that PE-NPs was beneficial in potently ameliorating CISP-induced nephrotoxicity in ESC mice. PE-NPs significantly attenuated CISP-induced oxidative stress and lipid peroxidation in the kidney via improving activities of antioxidants (SOD, GSH, and CAT). Additionally, PE-NPs considerably decreased CISP-induced inflammation in the kidney by decreasing the levels of NF-kB, IL-1β, and TNF-α. Notably, PE-NPs did not assuage the antitumor efficacy of CISP as revealed by histological assessment and tumor weight data. In summary, PE-NPs may be a potent alternative anticancer therapy devoid of nephrotoxicity.

This paper presents the isolation of single crystals of an anionic Cu(II) complex (NH4)(NHEt3)[Cu2(L)4Cl2]{HL = N-acetylanthranilic acid and NEt3= triethylamine}. The analyses revealed the complex packing in the triclinic "P-1" space group in addition to its dimeric structure and the octahedral geometry around the Cu(II)ions. In the complex, all the four carboxylate ligands are bridging the two Cu(II) centers and the charge on the anionic complex is compensated by an ammonium and protonated triethylamine ions. This Cu(II) complex is interestingly exerting highly significant cytotoxic effect on MDA-MB-231 breast cancer cells with IC50of 131.2 µg/ml and moderate anti-proliferative activity against both PC-3 prostatic cancer and T-24 transitional cell carcinoma cells with IC50 of 200.7 and 300.3 µg/ml, respectively.

Breast cancer is the most common neoplastic disorder diagnosed in women. The main goal of this study was to explore the effect of melatonin against breast cancer metastasis and compared this with the actions of taxol (a well-known chemotherapeutic drug), and the impact of their combination against breast cancer metastasis. Melatonin showed no cytotoxic effect while taxol showed antiproliferative and cytotoxic effects on MCF-7 and MDA-MB-231 cells. Furthermore, melatonin inhibited the generation of reactive oxygen species. Melatonin and taxol clearly decreased cell migration and invasion at low doses, especially those matching the normal physiological concentration at night. Melatonin and taxol markedly reduced DJ-1 and ID-1 and increased KLF17 messenger RNA and protein expression levels. The present results also showed that melatonin and taxol induced GSK3-β nuclear and Snail cytosolic localization. These changes were accompanied by a concurrent rise in E-cadherin expression. The above data show that normal levels of melatonin may help in preventing breast cancer metastasis through inhibiting DJ-1/KLF17/ID-1 signaling pathway. The combination of melatonin and taxol is a potent candidate against breast cancer metastasis, better than using melatonin or taxol as a single drug.

Paclitaxel (taxol) is an important agent against many tumours, including breast cancer. Ample data documents that paclitaxel inhibits breast cancer metastasis while others prove that paclitaxel enhances breast cancer metastasis. The mechanisms by which paclitaxel exerts its action are not well established. This study focuses on the effect of paclitaxel, particularly the low doses on breast cancer metastasis and the mechanisms that regulate it. Current results show that, paclitaxel exerts significant cytotoxicity even at low doses in both MCF-7 and MDA-MB-231 cells. Interestingly, paclitaxel significantly inhibits cell invasion and migration, decreases Snail and increases E-cadherin mRNA expression levels at the indicated low doses. Furthermore, paclitaxel-inhibiting breast cancer metastasis is associated with down-regulation of DJ-1 and ID-1 mRNA expression level with a concurrent increase in KLF17 expression. Under the same experimental conditions, paclitaxel induces KLF17 and concurrently represses ID-1 protein levels. Our results show for the first time that paclitaxel inhibits breast cancer metastasis through regulating DJ-1/KLF17/ID-1 signalling pathway; repressed DJ-1 and ID-1 and enhanced KLF17 expression.

Enclosed airspaces to reduce heat flow have been recognized for well over 100 years. Airspaces with one or more reflective surfaces define reflective insulation (RI) assemblies, a product type used in walls, roofs, windows with multiple panes, curtain walls and skylights. The thermal resistance (R value) of airspaces depends on the emittance of all surfaces, airspace dimensions and orientation, heat flow direction and surfaces temperatures. The modeling of RI now includes CFD coupled with radiation to quantify the total heat transfer. This study compares a validated model for airspace R values with existing methods such as ISO 6946 and hot-box results that provide the R values in the ASHRAE Handbook of Fundamentals. The existing methods do not include an airspace aspect ratio. This study showed that the aspect ratio can impact the R value by a factor of two. The impact of aspect ratio was calculated for double airspaces variation such as that for single airspaces. The present calculations are two-dimensional and also consider all the bounding airspace surfaces, while previous methods are one-dimensional and do not include surface temperature variations or detailed radiative transport.

The heat generation from recent advanced computer chips is increasing rapidly. This creates a challenge in cooling the chips while maintaining their temperatures below the threshold values. Another challenge is that the heat generation in the chip is not uniform where some chip components generate more heat than other components. This would create a large temperature gradient across the chip, resulting in inducing thermal stresses inside the chip that may lead to a high probability to damage the chip. The locations in the chip with heat rates that correspond to high heat fluxes are known as hotspots. This research study focuses on using thermoelectric modules (TEMs) for cooling chip hotspots of different heat fluxes. When a TEM is used for cooling a chip hotspot, it is called a thermoelectric cooler (TEC), which requires electrical power. Additionally, when a TEM is used for converting a chip’s wasted heat to electrical power, it is called a thermoelectric generator (TEG). In this study, the TEMs are used for cooling the hotspots of computer chips, and a TEC is attached to the hotspot to reduce its temperature to an acceptable value. On the other hand, the other cold surfaces of the chip are attached to TEGs for harvesting electrical power from the chip’s wasted heat. Thereafter, this harvested electrical power (HEP) is then used to run the TEC attached to the hotspot. Since no external electrical power is needed for cooling the hotspot to an acceptable temperature, this technique is called a sustainable self-cooling framework (SSCF). In this paper, the operation principles of the SSCF to cool the hotspot, subjected to different operating conditions, are discussed. As well, considerations are given to investigate the effect of the TEM geometrical parameters, such as the P-/N-leg height and spacing between the legs in both operations of the TEC mode and TEG mode on the SSCF performance.

The thermal resistance (R-value) of airspaces depends on the emittance of surfaces around the airspace, dimensions, heat-flow direction, and the temperatures of bounding surfaces. Assessing the energy performance of building envelope components and fenestration systems requires accurate results for the R-values of any enclosed spaces. The evaluation of reflective insulation R-values has evolved to include use of computational fluid dynamics and surface-to-surface radiation to quantify convective and radiation contributions to the heat transfer across airspaces of all types. This paper compares an advanced and validated computational tool for calculating enclosed airspace R-values with the widely-used ISO 6946 and airspace R-values in the ASHRAE Handbook-Fundamentals. The tool evaluates construction defects, air-infiltration impact, and dimensional aspect ratios that 1-D methods do not address. The differences between the methods that are currently being used to evaluate the R-value and the advantages of the advanced method for evaluation of reflective insulation applications are discussed.

Numerical simulations are conducted in this paper to investigate the hygrothermal performance of white and black roofing systems with and without Phase Change Materials (PCMs). The roofs considered in this paper are three roofing systems, namely, reference roof without drywall, reference roof with drywall but without PCM, and roofing system with PCM drywall of different melting temperatures. These roofing systems are subjected to the weather conditions of Kuwait city. In order to find out whether white roofs could lead to moisture-related problems, the numerical simulations are conducted for a period of 5 years. In case of the moisture accumulation is still increasing after 5 years on year-to-year basis of comparison of moisture, the period of simulation will be extended accordingly. The results show that no risk of condensation and mould growth occur in black and white roofing systems with and without PCM. In addition, the results show that white roofs experience an increase in the heating energy loads compared to black roofs. However, the decrease in the cooling energy loads for white roofs are typically much greater than the increase in the heating energy loads. As such, white roofs result in a net energy savings compared to black roofs. Furthermore, the results show that the months of the highest total energy loads for different roofing systems occur in July and August. The main outcome of this paper shows the capabilities of using reflective materials with different short-wave solar absorption coefficients and PCMs with different melting temperature for enhancing the energy performance of the roofing systems.
