The Pathology of the Disease in Comparison with the Cytopathogenic Effect of Bovine Virus Diarrhoea Virus on Tissue Culture A previous paper of this series described sporadic cases of an acute, noncomplicated mucosal disease occurring among heifers on a farm in southern Sweden. The post-mortem findings in one of the heifers from which the virus of bovine virus diarrhoea has been isolated, are described in the present report. The histological changes found in the epithelium of the skin, the mucosa of the gastrointestinal tract and the lymph nodes have been compared with the cytopathological changes seen in calf-kidney tissue cultures inoculated with different strains of virus diarrhoea virus. The major cell lesion encountered in both instances consisted of shrinking and karyopyknosis. The macroscopical changes are illustrated by Fig. 1—6, the histological by Fig. 7—12, and the cytopathological by Fig. 13—17. The results presented in both reports confirm the opinion of Gillespie et al. (1961) that “virus diarrhoea” and “mucosal disease” are synonymous and refer to a single disease caused by bovine virus diarrhoea virus and first described by Olafson et al. in 1946. At South Arne a highly repeatable time-lapse seismic survey (normalized root-mean-square error or NRMS of less than 0.1) allowed us to reliably monitor reservoir production processes during five years of reservoir depletion. Time-lapse AVO (amplitude v. offset) inversion and rock-physics analysis enables accurate monitoring of fluid pathways. On the crest of the field, water injection results in a heterogeneous sweep of the reservoir, whereby the majority of the injected water intrudes into a highly porous body. This is in contrast to a pre-existing reservoir simulation model predicting a homogeneous sweep. On the SW flank, time-lapse AVO inversion to changes in water saturation Δ S w reveals that the drainage pattern is fault controlled. Time-lapse seismic data furthermore explain the lack of production from the far end of a horizontal producer (as observed by production logging), by showing that the injected water does not result in the expected pressure support. On the highly porous crest of the reservoir compaction occurs. Time-lapse time shifts in the overburden are used as a measure for compaction and are compared with predictions of reservoir compaction from reservoir geomechanical modelling. In areas where compaction observations and predictions disagree, time-lapse seismic data give the necessary insight to validate, calibrate and update the reservoir geomechanical model. The information contained in time-lapse seismic data can only be fully extracted and used when the reservoir simulation model, the reservoir geomechanical model and the time-lapse seismic inversion models are co-visualized and available in the same software application with one set of coordinates. This allows for easy and reliable investigation of reservoir depletion and gives deeper insight than using reservoir simulation or time-lapse seismic individually.

The calcification in a totally calcified pineal tumour was studied. Transmission electron microscopy indicated that the tumour was a pinealoma. The type of calcification was investigated by bright-field and dark-field transmission electron microscopy in addition to electron diffraction and electron-induced x-ray fluorescence. The calcified material consisted predominantly of amorphous calcium phosphate. This type of calcification differs from the normal calcification present in pineal acervuli which consists of crystalline hydroxypatite.

The purpose of our research was to evaluate in vitro therapeutic efficacy of doxorubicin (DXR)-loaded immunoliposomes with Fab' fragments of the anti-CD74 antibody LL1 attached to the surface. LL1 is well suited for targeting purposes because it is internalized very fast by B-lymphoma cells. However, at in vivo application whole antibodies show fast clearance in circulation. Taking this fact into consideration, this study was initiated to elucidate the prospects of using Fab' fragments of LL1 in stead of the whole antibody for future targeting in vivo of DXR-loaded liposomes. The Fab' fragments were covalently attached to the surface of sterically stabilized liposomes by use of a PEG-based heterobifunctinal coupling agent. LL1 Fab' conjugated sterically stabilized DXR liposomes showed approximately six times faster accumulation of the drug in Raji human B-lymphoma cells than nontargeted liposomes. In vitro cytotoxicity, quantitated by a tetrazolium assay, against Raji cells gave IC(50) values of 0.13, 0.45, and 0.11 microM for DXR-loaded immunoliposomes, DXR-loaded liposomes and free drug, respectively. The results from this study suggest that DXR-loaded immunoliposomes targeted with Fab' fragments from the anti-CD74 antibody LL1 could be a useful system for future in vivo experiments.

Objective – To assess the presence of Epstein–Barr virus (EBV) and human herpesvirus 6B (HHV-6B) DNA in saliva and plasma from multiple sclerosis (MS) patients enrolled in a randomized, double-blind, placebo-controlled valacyclovir treatment study. Methods – DNA was prepared following ultracentrifugation of saliva and plasma. EBV and HHV-6B DNAs were determined by real-time polymerase chain reaction. Results – EBV and HHV-6B DNAs were detected in 41% and 65% of saliva samples, respectively. In patients treated with valacyclovir, the percentage of saliva samples with EBV was significantly reduced (9%; P = 0.000017), whereas the frequency of HHV-6B positive samples was unchanged (57%; P = 0.38). Longitudinal studies demonstrated a time-dependent reduction in the frequency of saliva samples containing EBV following valacyclovir treatment. In contrast, plasma contained EBV and HHV-6B DNAs in 17% and 25% of the samples, respectively, and these numbers were not significantly reduced following valacylovir treatment (13% and 16%, respectively), nor were they different from those of healthy controls (6% and 39%, respectively). Patients with high disease activity had a significantly higher frequency of EBV (P = 0.018) and HHV-6B (P = 0.023) positive samples than did patients with low disease activity. The human endogenous retrovirus HERV-H is associated with multiple sclerosis (MS). Previously performed reverse transcriptase-polymerase chain reactio ns (RT-PC R) on virion-RNA demonstrated sequence variants of the HERV-H family located in the particulate fraction of MS patient plasma samples and not in controls. In this study a significantly elevated level of antibodies towards peptides derived from HERV-H/RGH-2 DNA sequences in serum and cerebrospinal fluid (C SF) from MS patients is demonstrated. Further, Wistar rats immunized with purified virions develop a specific serologic response, indicating that some virion proteins are encoded by HERV-H-related sequences. A lso shown is that in RNA from blood cells, a HERV-H protease-env splice variant can be found together with an env splice variant in about 40% of MS patients but only in 10% of controls. The results substantiate the association between activated HERV-H and MS, but a causal relationship is yet to be demonstrated. HERV-H could represent a causal factor either by eliciting an autoimmune response or through the pathogenic potential of the retrovirus itself.

In a retrospective study, changes in baclofen dose and complication frequency were recorded in 79 patients with intrathecal baclofen administration and the effect on nightly muscle spasms was measured over a mean observation period of 34 months, during which time an increase in the daily dose of baclofen during the first 1--1-1/2 years is notified. On subsequent pump fillings the daily dose of baclofen remained stable in the group of non-multiple sclerosis patients. In contrast, the group of multiple sclerosis patients showed a steady increase in their daily dose of baclofen. We found a frequency of complications of 0.014 monthly often due to catheter problems. There was a significant decrease in numbers of nightly muscle spasms in an 8 h recording period from 77+/-20 preoperatively and 9+/-3 (P=0.02) 3 months after surgery. The steady increase in the daily dose of baclofen in order to obtain adequate reduction in spasticity and nightly muscle spasms in the first 1--1-1/2 years cannot fully be explained by caution and difficulties in achieving the correct dose, but also indicates that tolerance to baclofen occurs. Complications are often due to infection or catheter problems.

The paper describes the variation pattern of magnetic susceptibility of Lameta sediments and isotopic variation of organic13C from Chui Hill, Bergi, Kholdoda, Pisdura and Girad. The susceptibility pattern and a negative carbon isotopic anomaly allows fixation of the K/T boundary at these localities and they differ in these aspects from the inter-trappean sediments at Anjar.

The present study was carried out to investigate the long-latency soleus stretch reflexes M2 (peak latency of approximately 85 ms) and M3 (peak latency of approximately 115 ms) during walking in healthy and spastic multiple sclerosis (MS) patients. An 8° stretch was applied to the ankle extensors of the left leg in 8 healthy subjects during normal walking speed and 9 spastic MS patients and 10 age-matched healthy subjects during slow walking. When present in walking healthy subjects, M2 and M3 were modulated in a similar way and with the same amplitudes as previously described for the short latency soleus stretch reflex (M1). The spastic patients’ soleus M1 was significantly less modulated during walking. The patients’ M2 long-latency response was modulated in the same way as the age-matched healthy subjects. All patients’ M3 responses were absent or much suppressed during walking. The origin and functional importance of the short- and long-latency stretch reflexes in healthy and spastic persons are discussed in relation to the above findings and the behaviour of the stretch reflexes during matched isometric contractions. M3 is argued to be part of a transcortical reflex in healthy subjects.

The objective of this study was to evaluate the tendency for establishing spontaneously formed B-lymphoblastoid cell lines (LCL) from multiple sclerosis (MS) patients and healthy controls. We have previously established spontaneously formed B-lymphoblastoid cell lines from a patient with an MS-like disease and from another patient with MS who had a reactivated Epstein-Barr virus (EBV) infection. Both LCLs were found by transmission electron microscopy (EM) to produce retrovirus-like particles (RVLP) and EBV particles. Reverse transcriptase (RT) assays were positive in purified viral material from both LCLs. To substantiate these findings we initiated an intensified culturing procedure and were able to establish LCLs from 5 of 21 consecutive MS patients and 1 of 13 consecutive healthy controls. All LCLs were found to produce both RVLP and EBV particles by EM. Whether the putative new retrovirus (es) and EBV have any causal relationship to MS is still not known, but the findings support this possibility.

In April 1994, Vibrio metschnikovii was isolated from five infants with watery diarrhoea in Arequipa, Peru, as part of a passive cholera surveillance system. The children ranged in age from 11 to 20 months and had acute diarrhoea, with two cases showing moderate dehydration. Two children also had traces of blood in liquid stool. The children were seen at two different hospitals, and no evidence of a common source of infection was found. No additional V. metschnikovii isolates were identified in the remaining surveillance period that covered the rest of 1994 and 1995. However, stool samples were not screened for enteric pathogens other than vibrios. V. metschnikovii strains isolated from stool samples produced opaque and translucent colonies on agar plates, suggesting capsular material. All isolates were resistant to ampicillin, erythromycin and streptomycin. Plasmid analysis revealed a common 200-kb plasmid in isolates from all cases and an additional 2.7-kb plasmid in three of the isolates. Ribotyping of each isolate after restriction with BglI and HindIII endonucleases demonstrated identical ribotyping patterns. The cases reported suggest that V. metschnikovii may be associated with diarrhoea in man by mechanisms so far unknown.

The polar cusp and boundary layer are important in coupling magnetospheric energy sources to the high latitude ionosphere. ULF waves are one of the processes by which this coupling is realized. To study the source regions and propagation characteristics of discrete Pc1-2 (0.1–2 Hz) ULF wave packets, particularly unstructured emissions and Pc1 bursts at high latitudes, a triangular array of closely spaced induction magnetometers (∼150 km) was deployed beneath the average cusp projection during the 1992 Antarctic winter. From interstation time lags the wave velocity and direction of arrival were calculated with average uncertainties of ±60 kms−1 and ±8°. Wave sources were poleward of the array at low geomagnetic activity and equatorward at high activity. The sources also moved east to west with time, centred around local noon. These results are interpreted as indicative of the ionospheric signature of sources localized in the cusp, the low latitude boundary layer (LLBL) or the outer magnetosphere. Intercalibration of the results for typical events with extrapolations of PACE radar observations and DMSP satellite particle signatures support sources within these regions. Observed group velocities in the range 300–800 kms−1 with a mean of 450 kms−1 are consistent with wave propagation in the ionospheric waveguide. Signals above the waveguide lower cutoff frequency likely propagate away from the source in the ionospheric waveguide and across the magnetometer array. The results suggest a technique for monitoring the high latitude boundary regions and outer magnetosphere using local ULF wave measurements.

A natural N-linked glycosylation site (Asn-Val-Thr) at amino acid positions 18-20 (Kabat's numbering) was identified in the framework-1 (FR-1) region of the light chain variable (V kappa) domain of a murine anti-B cell lymphoma Ab, LL-2. Our earlier studies demonstrated that no contact between the V kappa-appended oligosaccharide and the Ag binding site was evident, because glycosylation at this site did not affect the Ag binding property of the Ab. By using the murine LL-2 F(ab')2 fragment (which is devoid of constant region-appended oligosaccharide) as substrate, as much as five bifunctional chelator molecules per F(ab')2 fragment could be site specifically conjugated at the V kappa-appended carbohydrate moiety with no reduction in immunoreactivity. The resulting conjugates labeled efficiently with both 90Y and 111In, with no significant effect on Ab affinity. In contrast, conjugation of less than five chelates/Ab fragment randomly at lysine residues resulted in a three- to fivefold reduction in affinity. By a single Arg to Asn mutation, an N-linked glycosylation site similar to that of LL-2 was introduced in the FR-1 segment of a nonglycosylated, humanized anti-carcinoembryonic Ag (CEA) Ab, MN-14 (hMN-14). Glycosylation at the engineered carbohydrate-addition site was demonstrated by SDS-PAGE analysis. Neither glycosylation nor site-specific conjugation of chelate at the V kappa-appended carbohydrate moiety resulted in the loss of immunoreactivity. The glycosylated hMN-14 conjugate labeled efficiently with 90Y.

Multiple sclerosis (MS) is associated with Epstein-Barr virus (EBV) infection, but impaired immune suppression may be part of the disease pathogenesis. CD8(+) T cells that are restricted by HLA-E exert an important immunoregulatory mechanism. To explore how EBV might interfere with immune regulation, we examined the expression of HLA-E and the frequency of CD8(+) cells recognizing HLA-E, presenting either an EBV peptide from the BZLF1 protein or a signal sequence peptide from HLA-A2, in relapsing remitting (MS-RR), primary progressive (MS-PP) MS patients, and healthy controls (HC). Reactions of 1,2,5,6,8,10-hexamethylheptalene (1) and its bond isomer, 1,4,5,6,8,10-hexamethylheptalene (2), with tricarbonylchromium complexes L{in3}Cr(CO){in3} (L=NH{in3} and Py) have been investigated. Thermodynamically less stable complex 1 exhibits higher reactivity with respect to Py{in3}Cr(CO){in3}/BF{in3} · Et{in2}O under the conditions of Ofele's reaction than complex2. At 10–30 °C, the Cr(CO){in3} group is coordinated to the asymmetrically substituted ring, which is accomplished by the shift of double bonds in the ligand, to afford tricarbonyl-[1,4,5,6,8,10-hexamethyl-η{su6}-(10a, 1–5)heptalene]-chromium (6) as the only mononuclear complex. Under more drastic conditions (Raush's reaction, 80 °C),1 ↔ 2 interconversion proceeds faster than the reaction of individual bond isomers with coordinatively unsaturated “hot” particles (solv){inn}Cr(CO){in3}. In this case, all of the four possible isomeric mononuclear complexes (6–9) and two binuclear complexes (10 and 11) are formed. The structures of complexes 6–11 have been studied by NMR and mass spectrometry, the structure of6 has been established by X-ray diffraction analysis. Heating a solution of6 in octane at 115 °C results in the isomerization of6 into complex7 through the intracycle 1,2-shift of the Cr(CO){in3} group and also in its conversion into complex8, which is the first example of interring η{su6}→η{su6}-haptotropic rearrangement in nonplanar seven-membered π-systems.

The characteristics of 1–20 mHz (Pc5) geomagnetic pulsations recorded during the daytime on the ground at cusp and boundary layer latitudes have been examined. On quiet and moderately disturbed days the major spectral contributions are due to three different mechanisms. Sustained oscillations whose properties are consistent with the Kelvin-Helmholtz instability at the low latitude boundary layer are the dominant mechanism at −70 to −75 geomagnetic latitude. Transient irregular pulsations are frequently seen at single stations at the foot of polar cap and boundary layer field lines. Occasionally similar transients occur essentially simultaneously at widely spaced stations accompanied by absorption spikes on riometer records. The latter signals are most likely due to solar wind pressure pulses on the magnetopause. At cusp latitudes the major spectral contribution arises from sustained irregular pulsations centred on magnetic noon. Although their occurrence is related to the proximity of the cusp's particle signature, it may be more appropriate to discuss these signals in terms of fluctuations in boundary layer or mantle currents.

Previous studies suggested that lesion of the spinothalamic tract is a necessary condition for development of CP. No previous study has in detail evaluated the association between the specific site of demyelinations and the presence of CP in MS. OBJECTIVE: The study aimed to evaluate the location of plaques in MS patients with CP including a group of MS patients without pain as a reference group. METHODS: All patients underwent a bedside sensory examination and MRI of the brain and spinal cord. MR imaging was acquired on an 1.5Tesla MR equipment. A trained neuroradiologist, blinded to pain status, evaluated the MRI. RESULTS: Thirteen MS patients with CP and 10 MS patients without pain were included. The ground magnetic and ionospheric signatures of reconnected flux tubes have been modeled using an expanding and rotating elliptical field-aligned current system. Assuming a constant total current of 2 × 105 A, initial ground field strengths of ∼100 nT are shown to decay rapidly with distance from the current system and with current system expansion. The associated ionospheric electric field estimates show that the flux tube footprint convects initially eastward or westward with velocity decreasing from 3 to 1 km s−1 and then poleward decreasing from 0.5 to 0.2 km s−1. The signature detected by a ground magnetometer array will be a 0.5 to 1-cycle oscillation with period of 8 min as the current system convects through the ionosphere above. These signatures are characterized by an initially rapid increase in field strength, with maximum amplitudes being reached within the first 5 min, followed by gradual decay. This modeling emphasizes that the temporal variation of the position of a recording magnetometer with respect to the ground image of the ionospheric footprint determines the form of the magnetic signature. The formalism should be useful in modeling flux tube behavior at high latitudes and may be relevant to dayside auroral observations.

In this study, we have measured the passive, the intrinsic, and the reflex-mediated mechanical response to stretch of the ankle extensors and flexors in 13 spastic multiple sclerosis patients and 10 healthy control subjects. In the ankle flexors, the patients had no reflex-mediated stiffness. The passive stiffness was increased by 138% (95% confidence interval: 26-91%) and the intrinsic stiffness by 79% (41-158%) when compared with the healthy subjects. In the ankle extensors, the reflex-mediated stiffness and the intrinsic stiffness of the patients were equal to the reflex-mediated and the intrinsic stiffness in healthy subjects. The passive stiffness was increased by 152% (41-352%). We conclude that spastic muscles in multiple sclerosis patients have an increased non-reflex stiffness (passive plus intrinsic stiffness), and that the reflex-mediated stiffness in the extensors during a sustained voluntary contraction does not differ significantly from healthy subjects.

Geomagnetic pulsations with frequencies in the range 0.1-2.5 Hz have been recorded with a six station Antarctic magnetometer array ranging in latitude from -62.3 to -80.6° invariant. The observed spectral characteristics were compared with the cusp and boundary layer locations for selected days in November and December, 1986, and July, 1990, in order to determine wave source regions. Specific pulsation features have been identified with different magnetospheric regions. The cusp is characterised by intense unstructured 0.15-0.45 Hz noise which diminishes rapidly away from this region. These signals are likely to result from a mixture of ion-cyclotron, Kelvin-Helmholtz and drift-wave instabilities. Narrowband unstructured Pc2 emissions typically around 0.2 Hz and short discrete Pc1 packets or bursts with frequencies below 0.4 Hz, occur in the local morning at boundary layer latitudes, accompanied by generally enhanced power levels between about 0.1 and 0.5 Hz. These emissions are probably due to ion-cyclotron waves generated near or below the He+ gyrofrequency on boundary layer field lines. Propagation of such signals into the polar cap via the ionospheric waveguide is uncommon, perhaps due to the existence of severe ionospheric irregularity features near the dayside auroral oval. Structured and quasi-structured emissions such as hydromagnetic chorus are recorded at the foot of plasmatrough field lines and are consistent with ion-cyclotron wave generation and propagation on closed field lines in the outer magnetosphere.

Fatigue, mood and quality of life were studied in patients with multiple sclerosis following progressive resistance training leading to improvement of muscular strength and functional capacity. Fatigue (Fatigue Severity Scale, FSS), mood (Major Depression Inventory, MDI) and quality of life (physical and mental component scores, PCS and MCS, of SF36) were scored at start, end and follow-up of a randomized controlled clinical trial of 12 weeks of progressive resistance training in moderately disabled (Expanded Disability Status Scale, EDSS: 3-5.5) multiple sclerosis patients including a Control group (n = 15) and an Exercise group (n = 16). Fatigue (FSS > 4) was present in all patients. Scores of FSS, MDI, PCS-SF36 and MCS-SF36 were comparable at start of study in the two groups. Fatigue improved during exercise by -0.6 (95% confidence interval (CI) -1.4 to 0.4) a.u. vs. 0.1 (95% CI -0.4 to 0.6) a.u. in controls (p = 0.04), mood improved by -2.4 (95% CI -4.1 to 0.7) a.u. vs. 1.1 (-1.2 to 3.4) a.u. in controls (p = 0.01) and quality of life (PCS-SF36) improved by 3.5 (95% CI 1.4-5.7) a.u. vs. -1.0 (95% CI -3.4-1.4) a.u. in controls (p = 0.01). The beneficial effect of progressive resistance training on all scores was maintained at follow-up after further 12 weeks. Fatigue, mood and quality of life all improved following progressive resistance training, the beneficial effect being maintained for at least 12 weeks after end of intervention.
