Asthma is a heterogenous disease characterized by airway inflammation and variable expiratory airflow limitation resulting in variable respiratory symptoms. Characterization of airway inflammation is important to choose the optimal treatment for severe asthma patients eligible for biological treatment. However, counting cells in induced sputum samples are a time-consuming process, highly dependent on personal skills. Replacing eosinophil and neutrophil cell counting with qPCR for transcripts of selected mast cell, and basophil genes may provide more reproducible results.Aims The objective of this study was to compare qPCR with microscopy in asthma endotyping.Methods A qPCR method measuring five mast cell/basophil genes was applied on induced sputum samples from 30 severe asthma patients and compared with microscopy. Target gene Ct-values (CPA3, GATA2, HDC, MS4A2, TPSAB1/TPSB2) were referenced to household β-actin Ct values as a measure of relative mRNA abundance of the target in each sample. Target/β-actin-ratios in eosinophilic and non-eosinophilic groups determined by microscopy with an eosinophil threshold of 3% in 400 cells were compared using Mann–Whitney U Test. Spearman´s correlations were used to test for correlation between targets vs. FENO and targets vs. blood eosinophil counts.Results The study demonstrated a statistical difference in relative mRNA abundance for four mast cell/basophil specific genes. CPA3, GATA2, HDC and MS4A2 were elevated in eosinophilic asthma versus non-eosinophilic asthma patients. The study found that GATA2, CPA3, MS4A2 and TPSAB1/TPSB2 transcripts are positively correlated with FENO. Neither the five mast cell genes nor the five-gene signature correlated with blood eosinophils. The five-gene signature with a target/β-actin-ratio cut-off ≥2 generated sensitivity = 87%, specificity = 94%, NPV = 88% and PPV = 92% compared to microscopy.Conclusion This study confirms the contribution of mast cells in the pathogenesis of EA and suggests that mast cell mRNA markers could be one of the biomarkers used to identify EA

In Tanzania, the majority of the rural population still relies on fuelwood as their major source of cooking energy. The adaptation measures of small-scale farmers in response to increasing fuelwood scarcity play a key role in altering the course of nutrition insecurity, environmental degradation, and economic instability. This study delivers a classification of coping strategies that does not exist in the literature. Furthermore, it analyses the adaptation measures applied by small-scale farmers in the semi-arid region of Dodoma district in response to fuelwood scarcity. A comparison between two case study sites provides information on the choice of adaptation measures by households. Overall, 28 coping strategies from 24 studies are identified, then differentiated into preventive and acute measures that are arranged into eight clusters. The classification is then used as a codebook to identify applied coping strategies at two case study sites. In total, 23 adaptation measures, including two strategies not cited in the literature, were identified through 39 household interviews. This suggests that the majority of coping strategies applied are independently from regional and social conditions. The majority of the strategies applied at the case study sites and described in the literature are acute measures that do not tackle the underlying problem triggering forest degradation. It is observed that the adaptation measures across the case study sites are widely congruent, thus showing that acute strategies are not replaced by preventive strategies but rather co-exist.

In Tanzania, a majority of rural residents cook using firewood-based three-stone-fire stoves. In this study, quantitative performance differences between technologically advanced improved cooking stoves and three-stone-fire stoves are analysed. We test the performance of improved cooking stoves and three-stone-fire stoves using local cooks, foods, and fuels, in the semi-arid region of Dodoma in Tanzania. We used the cooking protocol of the Controlled Cooking Test following a two-pot test design. The findings of the study suggest that improved cooking stoves use less firewood and less time than three-stone-fire stoves to conduct a predefined cooking task. In total, 40 households were assessed and ask to complete two different cooking tasks: (1) a fast cooking meal (rice and vegetables) and (2) a slow cooking meal (beans and rice). For cooking task 1, the results show a significant reduction in firewood consumption of 37.1% by improved cooking stoves compared to traditional three-stone-fire stoves; for cooking task 2 a reduction of 15.6% is found. In addition, it was found that the time needed to conduct cooking tasks 1 and 2 was significantly reduced by 26.8% and 22.8% respectively, when improved cooking stoves were used instead of three-stone-fire-stoves. We observed that the villagers altered the initial improved cooking stove design, resulting in the so-called modified improved cooking stove. In an additional Controlled Cooking Test, we conducted cooking task 3: a very fast cooking meal (maize flour and vegetables) within 32 households. Significant changes between the initial and modified improved cooking stoves regarding firewood and time consumption were not detected. However, analyses show that both firewood and time consumption during cooking was reduced when large amounts (for 6–7 household members) of food were prepared instead of small amounts (for 2–3 household members).

Enhancing food security is one of the main goals of subsistence farmers in Sub-Saharan Africa. This study investigates the implementation of improved loam-made cooking stoves and its contribution to coping and livelihood strategies. Controlled combustion, air as well as smoke flue, and heat insulation facilitate the more efficient fuel consumption of improved cooking stoves compared to traditional stoves—namely three stone fires. Although the majority of small-scale farmers in Sub-Saharan Africa rely on the free public good of firewood, the increasing time needed for collecting firewood implies high opportunity costs for productive members of the family. The primary outcomes for users of improved stoves are reduced fuel consumption, greater safety, saved time, and reduced smoke in the kitchen. The paper illustrates part of the output, outcome, and impact of a participatory action research approach for implementing improved cooking stoves. Special emphasis was put on enabling the villagers to construct their stoves without external support, hence having locally manufactured stoves made of mud, bricks, and dried grass. The impact pathway of improved cooking stoves followed the training-of-trainers concept, where members of the initially established farmer groups were trained to construct stoves on their own. Special focus was given to knowledge exchange and knowledge transfer in order to increase firewood efficiency and overall satisfaction of users of improved cook stoves. Encouraging the members to further adapt the stoves enabled them to scale-up the construction of improved cooked stoves into a business model and increase dissemination while creating income. Although many important benefits, like time and knowledge gain, were identified by the farmers after adoption of the new technology, we found adoption rates differed significantly between regions.

Haloscopes aim at detecting axions by converting them into photons using high-quality resonant cavities, where the cavity resonance should be tuned with the unknown axion mass. Here, the authors improve exclusion limits using four phase-matched resonant cavities and a fast frequency scanning technique.

Abstract We present results of the Relic Axion Dark-Matter Exploratory Setup (RADES), a detector which is part of the CERN Axion Solar Telescope (CAST), searching for axion dark matter in the 34.67 μeV mass range. A radio frequency cavity consisting of 5 sub-cavities coupled by inductive irises took physics data inside the CAST dipole magnet for the first time using this filter-like haloscope geometry. An exclusion limit with a 95% credibility level on the axion-photon coupling constant of g aγ ≳ 4 × 10 −13 GeV −1 over a mass range of 34.6738 μeV < m a < 34.6771 μeV is set. This constitutes a significant improvement over the current strongest limit set by CAST at this mass and is at the same time one of the most sensitive direct searches for an axion dark matter candidate above the mass of 25 μeV. The results also demonstrate the feasibility of exploring a wider mass range around the value probed by CAST-RADES in this work using similar coherent resonant cavities.

The term open-air factor (OAF) was coined following microbiological research in the 1960s and 1970s which established that rural air had powerful germicidal properties and attributed this to Criegee intermediates formed in the reaction of ozone with alkenes. We have re-evaluated those early experiments applying the current state of knowledge of ozone–alkene reactions. Contrary to previous speculation, neither Criegee intermediates nor the HO radicals formed in their decomposition are directly responsible for the germicidal activity attributed to the OAF. We identify other potential candidates, which are formed in ozone–alkene reactions and have known (and likely) germicidal properties, but the compounds responsible for the OAF remain a mystery. There has been very little research into the OAF since the 1970s, and this effect seems to have been largely forgotten. In this opinion piece we remind the community of the germicidal open-air factor. Given the current global pandemic spread by an airborne pathogen, understanding the natural germicidal effects of ambient air, solving the mystery of the open-air factor and determining how this effect can be used to improve human welfare should be a high priority for the atmospheric science community.

In the marine environment, measurements of lipids as representative species within different lipid classes have been performed to characterize their oceanic sources and their transfer from the ocean into the atmosphere to marine aerosol particles. The set of lipid classes includes hydrocarbons (HC); fatty acid methyl esters (ME); free fatty acids (FFA); alcohols (ALC); 1,3-diacylglycerols (1,3 DG); 1,2-diacylglycerols (1,2 DG); monoacylglycerols (MG); wax esters (WE); triacylglycerols (TG); and phospholipids (PP) including phosphatidylglycerols (PG), phosphatidylethanolamine (PE), phosphatidylcholines (PC), as well as glycolipids (GL) which cover sulfoquinovosyldiacylglycerols (SQDG), monogalactosyl-diacylglycerols (MGDG), digalactosyldiacylglycerols (DGDG) and sterols (ST). These introduced lipid classes have been analyzed in the dissolved and particulate fraction of seawater, differentiating between underlying water (ULW) and the sea surface microlayer (SML) on the one hand. On the other hand, they have been examined on ambient submicrometer aerosol particle samples (PM1) which were collected at the Cape Verde Atmospheric Observatory (CVAO) by applying concerted measurements. These different lipids are found in all marine compartments but in different compositions. Along the campaign, certain variabilities are observed for the concentration of dissolved (∑DLULW: 39.8–128.5 µg L−1, ∑DLSML: 55.7–121.5 µg L−1) and particulate (∑PLULW: 36.4–93.5 µg L−1, ∑PLSML: 61.0–118.1 µg L−1) lipids in the seawater of the tropical North Atlantic Ocean. Only slight SML enrichments are observed for the lipids with an enrichment factor EFSML of 1.1–1.4 (DL) and 1.0–1.7 (PL). On PM1 aerosol particles, a total lipid concentration between 75.2–219.5 ng m−3 (averaged: 119.9 ng m−3) is measured. As also bacteria – besides phytoplankton sources – influence the lipid concentrations in seawater and on the aerosol particles, the lipid abundance cannot be exclusively explained by the phytoplankton tracer (chlorophyll a). The concentration and enrichment of lipids in the SML are not related to physicochemical properties which describe the surface activity. On the aerosol particles, an EFaer (the enrichment factor on the submicrometer aerosol particles compared to the SML) between 9×104–7×105 is observed. Regarding the individual lipid groups on the aerosol particles, a statistically significant correlation (R2=0.45, p=0.028) was found between EFaer and lipophilicity (expressed by the KOW value), which was not present for the SML. But simple physicochemical descriptors are overall not sufficient to fully explain the transfer of lipids. As our findings show that additional processes such as formation and degradation influence the ocean–atmosphere transfer of both OM in general and of lipids in particular, they have to be considered in OM transfer models. Moreover, our data suggest that the extent of the enrichment of the lipid class constituents on the aerosol particles might be related to the distribution of the lipid within the bubble–air–water interface. The lipids TG and ALC which are preferably arranged within the bubble interface are transferred to the aerosol particles to the highest extent. Finally, the connection between ice nucleation particles (INPs) in seawater, which are already active at higher temperatures (−10 to −15 ∘C), and the lipid classes PE and FFA suggests that lipids formed in the ocean have the potential to contribute to (biogenic) INP activity when transferred into the atmosphere.

A detailed understanding of particle stopping in matter is essential for nuclear fusion and high energy density science. Here, the authors report one order of magnitude enhancement of intense laser-accelerated proton beam stopping in dense ionized matter in comparison with currently used models describing ion stopping in matter.

A condensed multiphase halogen and dimethyl sulfide (DMS) chemistry mechanism for application in chemistry transport models is developed by reducing the CAPRAM DMS module 1.0 (CAPRAM-DM1.0) and the CAPRAM halogen module 3.0 (CAPRAM-HM3.0). The reduction is achieved by determining the main oxidation pathways from analysing the mass fluxes of complex multiphase chemistry simulations with the air parcel model SPACCIM (SPectral Aerosol Cloud Chemistry Interaction Model). These simulations are designed to cover both pristine and polluted marine boundary layer conditions. Overall, the reduced CAPRAM-DM1.0 contains 32 gas-phase reactions, 5 phase transfers, and 12 aqueous-phase reactions, of which two processes are described as equilibrium reactions. The reduced CAPRAM-HM3.0 contains 199 gas-phase reactions, 23 phase transfers, and 87 aqueous-phase reactions. For the aqueous-phase chemistry, 39 processes are described as chemical equilibrium reactions. A comparison of simulations using the complete CAPRAM-DM1.0 and CAPRAM-HM3.0 mechanisms against the reduced ones indicates that the relative deviations are below 5 % for important inorganic and organic air pollutants and key reactive species under pristine ocean and polluted conditions. The reduced mechanism has been implemented into the chemical transport model COSMO-MUSCAT and tested by performing 2D simulations under prescribed meteorological conditions that investigate the effect of stable (stratiform cloud) and more unstable meteorological conditions (convective clouds) on marine multiphase chemistry. The simulated maximum concentration of HCl is of the order of 109 molecules cm−3 and that of BrO is around 1×107 molecules cm−3, reproducing the range of ambient measurements. Afterwards, the oxidation pathways of DMS in a cloudy marine atmosphere have been investigated in detail. The simulations demonstrate that clouds have both a direct and an indirect photochemical effect on the multiphase processing of DMS and its oxidation products. The direct photochemical effect is related to in-cloud chemistry that leads to high dimethyl sulfoxide (DMSO) oxidation rates and a subsequently enhanced formation of methane sulfonic acid compared to aerosol chemistry. The indirect photochemical effect is characterized by cloud shading, which occurs particularly in the case of stratiform clouds. The lower photolysis rate affects the activation of Br atoms and consequently lowers the formation of BrO radicals. The corresponding DMS oxidation flux is lowered by up to 30 % under thick optical clouds. Moreover, high updraught velocities lead to a strong vertical mixing of DMS into the free troposphere predominately under cloudy conditions. The photolysis of hypohalous acids (HOX, X = Cl, Br, or I) is reduced as well, resulting in higher HOX-driven sulfite-to-sulfate oxidation in aerosol particles below stratiform clouds. Altogether, the present model simulations have demonstrated the ability of the reduced mechanism to be applied in studying marine aerosol–cloud processing effects in regional models such as COSMO-MUSCAT. The reduced mechanism can be used also by other regional models for more adequate interpretations of complex marine field measurement data.

Despite the high abundance of secondary aerosols in the atmosphere, their formation mechanisms remain poorly understood. In this study, the Master Chemical Mechanism (MCM) and the Chemical Aqueous-Phase Radical Mechanism (CAPRAM) are used to investigate the multiphase formation and processing of secondary aerosol constituents during the advection of air masses towards the measurement site of Mt. Tai in northern China. Trajectories with and without chemical–cloud interaction are modeled. Modeled radical and non-radical concentrations demonstrate that the summit of Mt. Tai, with an altitude of ∼1.5 km a.m.s.l., is characterized by a suburban oxidants budget. The modeled maximum gas-phase concentrations of the OH radical are 3.2×106 and 3.5×106 molec. cm−3 in simulations with and without cloud passages in the air parcel, respectively. In contrast with previous studies at Mt. Tai, this study has modeled chemical formation processes of secondary aerosol constituents under day vs. night and cloud vs. non-cloud cases along the trajectories towards Mt. Tai in detail. The model studies show that sulfate is mainly produced in simulations where the air parcel is influenced by cloud chemistry. Under the simulated conditions, the aqueous reaction of HSO3- with H2O2 is the major contributor to sulfate formation, contributing 67 % and 60 % in the simulations with cloud and non-cloud passages, respectively. The modeled nitrate formation is higher at nighttime than during daytime. The major pathway is aqueous-phase N2O5 hydrolysis, with a contribution of 72 % when cloud passages are considered and 70 % when they are not. Secondary organic aerosol (SOA) compounds, e.g., glyoxylic, oxalic, pyruvic and malonic acid, are found to be mostly produced from the aqueous oxidations of hydrated glyoxal, hydrated glyoxylic acid, nitro-2-oxopropanoate and hydrated 3-oxopropanoic acid, respectively. Sensitivity studies reveal that gaseous volatile organic compound (VOC) emissions have a huge impact on the concentrations of modeled secondary aerosol compounds. Increasing the VOC emissions by a factor of 2 leads to linearly increased concentrations of the corresponding SOA compounds. Studies using the relative incremental reactivity (RIR) method have identified isoprene, 1,3-butadiene and toluene as the key precursors for glyoxylic and oxalic acid, but only isoprene is found to be a key precursor for pyruvic acid. Additionally, the model investigations demonstrate that an increased aerosol partitioning of glyoxal can play an important role in the aqueous-phase formation of glyoxylic and oxalic acid. Overall, the present study is the first that provides more detailed insights in the formation pathways of secondary aerosol constituents at Mt. Tai and clearly emphasizes the importance of aqueous-phase chemical processes on the production of multifunctional carboxylic acids.

The project MarParCloud (Marine biological production, organic aerosol Particles and marine Clouds: a process chain) aims to improve our understanding of the genesis, modification and impact of marine organic matter (OM) from its biological production, to its export to marine aerosol particles and, finally, to its ability to act as ice-nucleating particles (INPs) and cloud condensation nuclei (CCN). A field campaign at the Cape Verde Atmospheric Observatory (CVAO) in the tropics in September–October 2017 formed the core of this project that was jointly performed with the project MARSU (MARine atmospheric Science Unravelled). A suite of chemical, physical, biological and meteorological techniques was applied, and comprehensive measurements of bulk water, the sea surface microlayer (SML), cloud water and ambient aerosol particles collected at a ground-based and a mountain station took place. Key variables comprised the chemical characterization of the atmospherically relevant OM components in the ocean and the atmosphere as well as measurements of INPs and CCN. Moreover, bacterial cell counts, mercury species and trace gases were analyzed. To interpret the results, the measurements were accompanied by various auxiliary parameters such as air mass back-trajectory analysis, vertical atmospheric profile analysis, cloud observations and pigment measurements in seawater. Additional modeling studies supported the experimental analysis. During the campaign, the CVAO exhibited marine air masses with low and partly moderate dust influences. The marine boundary layer was well mixed as indicated by an almost uniform particle number size distribution within the boundary layer. Lipid biomarkers were present in the aerosol particles in typical concentrations of marine background conditions. Accumulation- and coarse-mode particles served as CCN and were efficiently transferred to the cloud water. The ascent of ocean-derived compounds, such as sea salt and sugar-like compounds, to the cloud level, as derived from chemical analysis and atmospheric transfer modeling results, denotes an influence of marine emissions on cloud formation. Organic nitrogen compounds (free amino acids) were enriched by several orders of magnitude in submicron aerosol particles and in cloud water compared to seawater. However, INP measurements also indicated a significant contribution of other non-marine sources to the local INP concentration, as (biologically active) INPs were mainly present in supermicron aerosol particles that are not suggested to undergo strong enrichment during ocean–atmosphere transfer. In addition, the number of CCN at the supersaturation of 0.30 % was about 2.5 times higher during dust periods compared to marine periods. Lipids, sugar-like compounds, UV-absorbing (UV: ultraviolet) humic-like substances and low-molecular-weight neutral components were important organic compounds in the seawater, and highly surface-active lipids were enriched within the SML. The selective enrichment of specific organic compounds in the SML needs to be studied in further detail and implemented in an OM source function for emission modeling to better understand transfer patterns, the mechanisms of marine OM transformation in the atmosphere and the role of additional sources. In summary, when looking at particulate mass, we see oceanic compounds transferred to the atmospheric aerosol and to the cloud level, while from a perspective of particle number concentrations, sea spray aerosol (i.e., primary marine aerosol) contributions to both CCN and INPs are rather limited.

The multinomial ecological indicator values (EIV) categorize species according to their ecological optima. They are a widely used expert system in ecology and vegetation science, their biological causes are largely unknown. The root anatomy of Carex shows considerable quantitative and qualitative differences. It is hypothesized that root traits may be responsible for the niche differentiation of the species, which means they predict the ecological optima of Carex species in terms of the EIV. This relationship was tested for the 107 German Carex species over ecological gradients of soil moisture, nutrients, soil reaction, temperature and light. Generalized estimating equations were used to model the multinomial indicator values by six anatomical traits: radius of root and central cylinder (stele), sclerenchyma thickness, aerenchyma width, largest vessel diameter and vessel number. Sclerenchyma thickness and endoderm radius predict soil moisture optima; the endoderm radius also light optima. Species of dry soils have thicker sclerenchyma and a wider stele. Species of shady growth conditions have wider steles. The diameter of the largest vessel was positively related to soil nitrogen, species of nitrogen-rich soils possess wider vessels. The width of the aerenchyma was positively but marginally significant related to increasing soil moisture. Root anatomy predicts the ecological optima of species along several ecological gradients and may thus explain the ecological differentiation of the species. This hitherto unknown pattern could probably only be observed due to the extraordinary diversity of Carex, which occupy widely different habitats within a region.

Summary: Innovation in clean-energy technologies is central toward a net-zero energy system. One key determinant of technological innovation is the integration of external knowledge, i.e., knowledge spillovers. However, extant work does not explain how individual spillovers come about: the mechanisms and enablers of these spillovers. We ask how knowledge from other technologies, sectors, or scientific disciplines is integrated into the innovation process in an important technology for a net-zero future: lithium-ion batteries (LIBs), based on a qualitative case study using extant literature and an elite interview campaign with key inventors in the LIB field and R&D/industry experts. We identify the breakthrough innovations in LIBs, discuss the extent to which breakthrough innovations—plus a few others—have resulted from spillovers, and identify different mechanisms and enablers underlying these spillovers, which can be leveraged by policymakers and R&D managers who are interested in facilitating spillovers in LIBs and other clean-energy technologies.

Changes in chromosome number and structure, as well as alterations in genome size, are important cytological characters that often reflect speciation events. Due to a wide variety of previously reported chromosome numbers the species-rich arrowroot family (Marantaceae) is an ideal system to investigate the role of chromosomal changes in species diversification, especially of dysploidy and polyploidy. The mechanisms and direction of changes during evolution as well as ancestral states of chromosomal characters are largely unknown. This study provides a detailed survey of chromosomal and genome size variation in 43 Marantaceae accessions (37 species, 16 genera) from the whole distribution range. Phylogenetic tree mapping suggests that x = 13 is the ancestral basic chromosome number. Descending dysploidy to x = 9, 10, 11, and 12 occurred ten times in parallel within each of the five main clades. In contrast, ascending dysploidy to x = 14, occurred only once. Ploidy level variation was confined to basic numbers x = 9, 11, and 13 and was observed in four out of the five clades. The occurrence of triploids and pentaploids points towards heteroploid diploid-tetraploid and tetraploid-hexaploid hybridizations. Trends in genome evolution as exposed by regression analyses revealed a massive genome size increase after dysploidy - and a genome size decrease after polyploidy. A schematic illustration of possible origins and the modes of chromosomal changes in a biogeographical context is presented. Exemplified by the family Marantaceae, it is shown how variable chromosomal evolution can be and how it contributes to species richness and speciation in the plant kingdom.

Among ice core drilling sites in the European Alps, Colle Gnifetti (CG) is the only non-temperate glacier to offer climate records dating back at least 1000 years. This unique long-term archive is the result of an exceptionally low net accumulation driven by wind erosion and rapid annual layer thinning. However, the full exploitation of the CG time series has been hampered by considerable dating uncertainties and the seasonal summer bias in snow preservation. Using a new core drilled in 2013 we extend annual layer counting, for the first time at CG, over the last 1000 years and add additional constraints to the resulting age scale from radiocarbon dating. Based on this improved age scale, and using a multi-core approach with a neighbouring ice core, we explore the time series of stable water isotopes and the mineral dust proxies Ca2+ and insoluble particles. Also in our latest ice core we face the already known limitation to the quantitative use of the stable isotope variability based on a high and potentially non-stationary isotope/temperature sensitivity at CG. Decadal trends in Ca2+ reveal substantial agreement with instrumental temperature and are explored here as a potential site-specific supplement to the isotope-based temperature reconstruction. The observed coupling between temperature and Ca2+ trends likely results from snow preservation effects and the advection of dust-rich air masses coinciding with warm temperatures. We find that if calibrated against instrumental data, the Ca2+-based temperature reconstruction is in robust agreement with the latest proxy-based summer temperature reconstruction, including a Little Ice Age cold period as well as a medieval climate anomaly. Part of the medieval climate period around AD 1100–1200 clearly stands out through an increased occurrence of dust events, potentially resulting from a relative increase in meridional flow and/or dry conditions over the Mediterranean.

Cold glaciers at the highest locations of the European Alps have been investigated by drilling ice cores to retrieve their stratigraphic climate records. Findings like the Oetztal ice man have demonstrated that small ice bodies at summit locations of comparatively lower altitudes may also contain old ice if locally frozen to the underlying bedrock. In this case, constraining the maximum age of their lowermost ice part may help to identify past periods with minimum ice extent in the Alps. However, with recent warming and consequent glacier mass loss, these sites may not preserve their unique climate information for much longer. Here we utilized an existing ice cave at Chli Titlis (3030 m), central Switzerland, to perform a case study for investigating the maximum age of cold-based summit glaciers in the Alps. The cave offers direct access to the glacier stratigraphy without the logistical effort required in ice core drilling. In addition, a pioneering exploration had already demonstrated stagnant cold ice conditions at Chli Titlis, albeit more than 25 years ago. Our englacial temperature measurements and the analysis of the isotopic and physical properties of ice blocks sampled at three locations within the ice cave show that cold ice still exists fairly unchanged today. State-of-the-art micro-radiocarbon analysis constrains the maximum age of the ice at Chli Titlis to about 5000 years before present. By this means, the approach presented here will contribute to a future systematic investigation of cold-based summit glaciers, also in the Eastern Alps.

Only limited data on laboratory parameter dynamics and safety under prolonged biologic treatment in a “real-world” scenario are available for recommendations on screening and monitoring. This study is a retrospective analysis of routine parameter dynamics and laboratory adverse events (LAE) in psoriasis patients on long-term treatment (n = 199) with tumour necrosis factor (TNF)-α-antagonists (adalimumab, etanercept), and the interleukin (IL)12/23-antagonist ustekinumab. Overall, neutrophil (PMN) counts (–11%) and triglycerides (+9%) changed considerably. TNF-α-antagonists and ustekinumab differentially affected lymphocyte counts (+13% and ±0%, respectively). Dynamics were pronounced during the first 180 days of treatment. In 340 treatment-years, 15 Common Terminology Criteria for Adverse Events (CTCAE) III–IV° LAE were recorded (11 involved liver enzymes). They prompted alteration of the biologic regime in only 2 cases. Age, sex, previous systemic treatments, and psoriatic arthritis did not significantly predict LAE. Liver enzyme and triglyceride screening may be warranted in some instances. Our data suggest that unguided monitoring of other routine laboratory parameters is unnecessary under long-term biologic treatment.

Although its Holocene glacier history is still subject to debate, the ongoing iconic decline of Kilimanjaro's largest remaining ice body, the Northern Ice Field (NIF), has been documented extensively based on surface and photogrammetric measurements. The study presented here adds, for the first time, ground-penetrating radar (GPR) data at centre frequencies of 100 and 200 MHz to investigate bed topography, ice thickness and internal stratigraphy at NIF. The direct comparison of the GPR signal to the visible glacier stratigraphy at NIF's vertical walls is used to validate ice thickness and reveals that the major internal reflections seen by GPR can be associated with dust layers. Internal reflections can be traced consistently within our 200 MHz profiles, indicating an uninterrupted, spatially coherent internal layering within NIF's central flat area. We show that, at least for the upper 30 m, it is possible to follow isochrone layers between two former NIF ice core drilling sites and a sampling site on NIF's vertical wall. As a result, these isochrone layers provide constraints for future attempts at linking age–depth information obtained from multiple locations at NIF. The GPR profiles reveal an ice thickness ranging between (6.1 ± 0.5) and (53.5 ± 1.0) m. Combining these data with a very high resolution digital elevation model we spatially extrapolate ice thickness and give an estimate of the total ice volume remaining at NIF's southern portion as (12.0 ± 0.3) × 106 m3.

The phagocyte NADPH oxidase (the NOX2 complex) generates superoxide, the precursor to reactive oxygen species (ROS). ROS possess both antimicrobial and immunoregulatory function. Inactivating mutations in alleles of the NOX2 complex cause chronic granulomatous disease (CGD), characterized by an enhanced susceptibility to infections and autoimmune diseases such as Systemic lupus erythematosus (SLE). The latter is characterized by insufficient removal of dead cells, resulting in an autoimmune response against components of the cell's nucleus when non-cleared apoptotic cells lose their membrane integrity and present autoantigenic molecules in an inflammatory context. Here we aimed to shed light on the role of the NOX2 complex in handling of secondary necrotic cells (SNECs) and associated consequences for inflammation and autoimmunity during lupus.We show that individuals with SLE and CGD display accumulation of SNECs in blood monocytes and neutrophils. In a CGD phenotypic mouse strain (Ncf1** mice) build-up of SNECs in Ly6CHI blood monocytes was connected with a delayed degradation of the phagosomal cargo and accompanied by production of inflammatory mediators. Treatment with H2O2 or activators of ROS-formation reconstituted phagosomal abundance of SNECs to normal levels. Induction of experimental lupus further induced increased antibody-dependent uptake of SNECs into neutrophils. Lupus-primed Ncf1** neutrophils took up more SNECs than wild type neutrophils, whereas SNEC-accumulation in regulatory Ly6C−/LO monocytes was lower in Ncf1**mice. We deduce that the inflammatory rerouting of immune-stimulatory necrotic material into inflammatory phagocyte subsets contributes to the connection between low ROS production by the NOX2 complex and SLE.

A number of heavy-ion accelerators are either under construction (e.g., the Facility for Antiproton and Ion Research in Darmstadt and the High Intensity Accelerator Facility in China) or already in operation at many places worldwide. For these accelerators, activation of construction components due to beam loss, even during routine machine operation, is a serious issue, especially with the upcoming high-intensity facilities. Aluminum is one of the most commonly used construction materials in beam lines, collimators, and other components. Therefore, we report here on activation experiments on aluminum samples to verify and benchmark simulation codes. The analysis was performed by gamma spectroscopy of the irradiated targets. Our results on the induced activity measured in samples irradiated by uranium beams at 125 MeV/u and 200 MeV/u and a xenon beam at 300 MeV/u show activation levels significantly lower than those predicted by FLUKA simulations.

This study investigates whether a water deficit index (WDI) based on imagery from unmanned aerial vehicles (UAVs) can provide accurate crop water stress maps at different growth stages of barley and in differing weather situations. Data from both the early and late growing season are included to investigate whether the WDI has the unique potential to be applicable both when the land surface is partly composed of bare soil and when crops on the land surface are senescing. The WDI differs from the more commonly applied crop water stress index (CWSI) in that it uses both a spectral vegetation index (VI), to determine the degree of surface greenness, and the composite land surface temperature (LST) (not solely canopy temperature).Lightweight thermal and RGB (red–green–blue) cameras were mounted on a UAV on three occasions during the growing season 2014, and provided composite LST and color images, respectively. From the LST, maps of surface-air temperature differences were computed. From the color images, the normalized green–red difference index (NGRDI), constituting the indicator of surface greenness, was computed. Advantages of the WDI as an irrigation map, as compared with simpler maps of the surface-air temperature difference, are discussed, and the suitability of the NGRDI is assessed. Final WDI maps had a spatial resolution of 0.25 m.It was found that the UAV-based WDI is in agreement with measured stress values from an eddy covariance system. Further, the WDI is especially valuable in the late growing season because at this stage the remote sensing data represent crop water availability to a greater extent than they do in the early growing season, and because the WDI accounts for areas of ripe crops that no longer have the same need for irrigation. WDI maps can potentially serve as water stress maps, showing the farmer where irrigation is needed to ensure healthy growing plants, during entire growing season.

The importance of oxygen in supporting early animal ecosystems is unclear because most proxies are unable to distinguish well-oxygenated from intermediate waters. Here, the authors show that early skeletal animals were restricted to well-oxygenated habitats, suggesting they had a high metabolic oxygen demand.

Estimating evaporation is important when managing water resources and cultivating crops. Evaporation can be estimated using land surface heat flux models and remotely sensed land surface temperatures (LST), which have recently become obtainable in very high resolution using lightweight thermal cameras and Unmanned Aerial Vehicles (UAVs). In this study a thermal camera was mounted on a UAV and applied into the field of heat fluxes and hydrology by concatenating thermal images into mosaics of LST and using these as input for the two-source energy balance (TSEB) modelling scheme. Thermal images are obtained with a fixed-wing UAV overflying a barley field in western Denmark during the growing season of 2014 and a spatial resolution of 0.20 m is obtained in final LST mosaics. Two models are used: the original TSEB model (TSEB-PT) and a dual-temperature-difference (DTD) model. In contrast to the TSEB-PT model, the DTD model accounts for the bias that is likely present in remotely sensed LST. TSEB-PT and DTD have already been well tested, however only during sunny weather conditions and with satellite images serving as thermal input. The aim of this study is to assess whether a lightweight thermal camera mounted on a UAV is able to provide data of sufficient quality to constitute as model input and thus attain accurate and high spatial and temporal resolution surface energy heat fluxes, with special focus on latent heat flux (evaporation). Furthermore, this study evaluates the performance of the TSEB scheme during cloudy and overcast weather conditions, which is feasible due to the low data retrieval altitude (due to low UAV flying altitude) compared to satellite thermal data that are only available during clear-sky conditions. TSEB-PT and DTD fluxes are compared and validated against eddy covariance measurements and the comparison shows that both TSEB-PT and DTD simulations are in good agreement with eddy covariance measurements, with DTD obtaining the best results. The DTD model provides results comparable to studies estimating evaporation with similar experimental setups, but with LST retrieved from satellites instead of a UAV. Further, systematic irrigation patterns on the barley field provide confidence in the veracity of the spatially distributed evaporation revealed by model output maps. Lastly, this study outlines and discusses the thermal UAV image processing that results in mosaics suited for model input. This study shows that the UAV platform and the lightweight thermal camera provide high spatial and temporal resolution data valid for model input and for other potential applications requiring high-resolution and consistent LST.
